The clinical application value of FAM19A4/mir124-2 methy lation test in hrHPV-positive women

Cervical cancer is one of the most common malignancies in women. The combination of human papillomavirus (HPV) genotyping with thinprep liquid-based cytology test (TCT) of cervical exfoliated cells and HPV vaccine helps greatly for the early diagnosis and prevention of cervical cancer. However, there is still a certain risk of missed diagnosis or over-diagnosis due to the limitations of its methodology. Therefore, women infected with high-risk HPV (hrHPV) need more reliable tests to identify whether the lesions will progress to cancer, which can guide early and accurate treatment and prevent over-treatment. Recently, studies have found that FAM19A4/mir124-2 methylation is closely related to cervical cancer, and its methylation degree increases with the severity of cervical lesions, which is expected to become a new biomarker of cervical cancer. Here, we summarized the clinical application value of FAM19A4/mir124-2 methylation test in hrHPV-positive women.

Comparison of clinicopathological characteristics of endogenous and exogenous cervical cancer with its clinical significance

<p class="abstract"><strong><span lang="EN-US">Background: </span></strong>Cervical cancer is the third leading cause of cancer death in women of less developed countries due to poor screening and decreased diagnostic approaches. We aimed to investigate and differentiate the distinct clinicopathological characteristics and prevalence of endogenous and exogenous cervical in hospitalized patients<span lang="EN-US">.</span></p><p class="abstract"><strong><span lang="EN-US">Methods: </span></strong>A study was performed contained 663 patients that were enrolled and underwent for screening of endogenous and exogenous cervical cancer in Qilu Hospital of Shandong University, from January 2010 to March 2015. </p><p class="abstract"><strong><span lang="EN-US">Results: </span></strong>Our results indicated that combined examination of thin-prep liquid-based cytology test and human papillomavirus (HPV)-DNA are effective for diagnosis of endogenous and exogenous cervical cancer. The clinicopathological characteristics based on tumor growth pattern, and high risk HPV-DNA incidence had no significant difference (p>0.05) in endogenous and exogenous cervical cancer patients. The higher ratio of lymph node metastasis in endogenous cervical cancer and exogenous cervical cancer during IB1-stage (24.3% vs 12.9%), and in IIA2-stage (36.4% vs 25%) was observed respectively. In addition, our data provide compelling evidence that the level of deep interstitial infiltration, and lymphatic vascular infiltration in endogenous cervical cancer was collectively higher (82.7% and 33%) compared to exogenous cervical cancer (62.4% and 18.3%) respectively during all stages<span lang="EN-US">.</span></p><p class="abstract"><strong><span lang="EN-US">Conclusions: </span></strong>The higher percentage of lymph node metastasis,<strong> </strong>deep interstitial infiltration, and lymphatic vascular infiltration was observed in endogenous cervical cancer which might be the biomarker and differential key points for the diagnosis of endogenous cervical cancer. Taken together, our study provides clinicopathological features to diagnose, and differentiate the endogenous and exogenous type cervical cancer with its prevalence.</p>

Diagnostic value of heat shock protein 90α and squamous cell carcinoma antigen in detection of cervical cancer.

ObjectiveTo ascertain plasma levels of heat shock protein 90α (HSP90α) and squamous cell carcinoma antigen (SCC-Ag) and their diagnostic potential in cervical cancer.MethodsIn a cross-sectional study, patients’ cervical tissue samples were screened for high risk (HR) human papilloma virus (HPV) DNA and underwent a thinprep-liquid based cytology test (TCT). Plasma samples were analysed by enzyme-linked immunosorbent assay (ELISA) for HSP90 α and SCC-Ag levels.ResultsOf the 295 women who underwent screening, 75 were healthy controls 75 (HR-HPV−ve TCT−ve), 110 were HR-HPV+ve, TCT−ve and 110 were HR-HPV+ve TCT+ve. There were significant differences between levels of HSP90α and SCC-Ag proteins across the patient groups with those positive for cervical cancer having the greatest levels of proteins compared with other groups. For patients with high grade SCC there was a significant correlation between levels of HSP90α and SCC-Ag. The area under the ROC curve for combined HSP90α*SCC-Ag was the largest compared with the single proteins. Using a cut-off value of 16.4 ng/ml to delineate cervical cancer diagnosis, the sensitivity and specificity of HSP90α*SCC-Ag were 90.3% and 95.1% respectively.ConclusionPlasma HSP90α protein levels correlated well with SCC-Ag levels in patients with cervical cancer and the combination of HSP90α*SCC-Ag may be a useful diagnostic biomarker.

Prevalence of breast and gynecological diseases in female workers in the administrative office and workshop of an automobile part factory

Objective: To investigate the prevalence of breast and gynecological diseases in female workers in the administrative office and workshop of an automobile part factory, and to provide a basis for promoting the health of female workers. Methods: In March 2015, color ultrasound examination of the breast, uterus, and bilateral adnexa was performed for all female workers, and routine gynecological examination, routine leucorrhea examination, and thinprep liquid-based cytology test were performed for married female workers. The detection rates of breast and gynecological diseases in female workers in the administrative office and workshop were analyzed. Results: In the 314 female workers, the overall detection rate of breast and gynecological diseases was 86.31%; the detection rate of abnormal breast ultrasound results was 72.93%, mainly breast hyperplasia; the detection rate of abnormal gynecological ultrasound results was 12.14%, mainly hysteromyoma, pelvic effusion, and uterus-rectum fossa effusion. The overall detection rate of cervicitis or vaginitis in married female workers was 66.86%. The univariate analysis showed that compared with those in the administrative office, the female workers in the workshop had significantly higher detection rates of breast hyperplasia and grade 2 breast lesions on the Breast Imaging-Reporting and Data System (BI-RADS) and significantly lower detection rates of hysteromyoma and grade 3 breast lesions on the BI-RADS (all P <0.05) . The multivariate analysis showed that age was a risk factor for hysteromyoma (OR=1.261) , age and working in the workshop were risk factors for breast hyperplasia (OR=1.065 and 1.834) , married status was a protective factor against breast hyperplasia and grade 2 breast lesions on the BI-RADS (OR=0.334 and 0.450) , and working in workshop was a risk factor for grade 2 breast lesions on the BI-RADS (OR=1.890) and a protective factor against grade 3 breast lesions on the BI-RADS (OR=0.355) . Conclusion: Female workers in an automobile part factory have high prevalence rates of breast and gynecological diseases. Night shifts for female workers in the workshop should be reduced as much as possible, and female workers, especially those in the administrative office, should be guided to release pressure and take a balanced diet.

Value of high risk human papilloma virus detection in screening and diagnosing cervical lesion in littoral of Zhejiang province

To analyse the infection of high-risk human papiliomavirus (HR-HPV) in cervical lesion wome, and evaluate the significance of high-risk human pappilomavirus detection by hybrid capture II (HV-II) in screening and diagnosing cervical lesion, especially high grade cervical intraepithelial neoplasia (CIN). A series of 1130 patients of cervical lesion were preliminarily diagnosed by cervical cytological examination, HR-HPV detection by HC-II , colposcopy and biopsy under the colposcopy between June 2009 and December 2008, including 212 CIN I and (or) condyloma (CIN I/HPV I), 442 CIN II/III, 28 invasive cervical cancer. cervical cytological examination is by thin prep liquid-based cytology test(TCT),and HR-HPV detection is by HC-II. In 1130 cases the positive of HR-HPV was 65.84% (744/1130). Unusual cytology result were 862 cases, with 356 ASCUS, 84 ASCH, 216 LSIL, 184HSIL and 22 cancer. The number of biopsy > or = CINI/HPVI was 682, positive rate of HR-HPV was 78.59% (536/682). In screening CIN II or above, sensitivity, specificity, PPV and NPV of TCT were 88.94%, 32.73%, 48.49%, 80.60%, of HR-HPV DNA detectiort by HC-II were 90.21%, 51.82%, 57.14%, 88.14%, and of HR-HPV detection combined with cytology were 97.45%, 22.42%, 47.22%, 92.50%. The infection rate of HR-HPV in cervical lesions is higher in each age group. Infection rate of HR-HPV is ascending with serious degree of cervical lesion. HR-HPV detection by HC- II is an important method in screening cervical lesion. HR-HPV detection is a viable option in the management of women with ASCUS and LSIL of TCT, with higher sensitivity and NPV.

Genital human papillomavirus screening by gene chip in Chinese women of Guangdong province

Human papillomavirus (HPV) infections are associated with cervical cancer. There were only a few reports and detailed data about epidemiological research of HPV infection in general population of China. To determine the prevalence of genital HPV infection in Chinese women of Guangdong province. A total of 1705 women were screened by gene chip. All HPV-positive women were further examined by ThinPrep liquid-based cytology test (TCT), and the cervical biopsies of those women with positive HPV-DNA and abnormal TCT were collected for pathological diagnosis. The overall HPV prevalence was 9.03% (154 of 1705), and 72.3% (126 of 154) of total positive samples were high-risk types, with higher prevalence of types 52, 58, 16, 18 and CP8304. For women aged 51 years or older, the overall high-risk HPV prevalence was 12.2% (24 of 179), which was obviously higher than those of other age groups (P < 0.05). Our results showed that the HPV prevalence in Guangdong is very similar to the world level. Unlike most previous studies, our findings suggest that HPV prevalence increased with age, and that the predominant genotypes in this area were HPV 52 and 58.
