A Nanographene-Porphyrin Hybrid for Near-Infrared-Ii Phototheranostics.

Photoacoustic imaging (PAI)-guided photothermal therapy (PTT) in the second near-infrared (NIR-II, 1000-1700nm) window has been attracting attention as a promising cancer theranostic platform. Here, it is reported that the π-extended porphyrins fused with one or two nanographene units (NGP-1 and NGP-2) can serve as a new class of NIR-responsive organic agents, displaying absorption extending to ≈1000 and ≈1400nm in the NIR-I and NIR-II windows, respectively. NGP-1 and NGP-2 are dispersed in water through encapsulation into self-assembled nanoparticles (NPs), achieving high photothermal conversion efficiency of 60% and 69%, respectively, under 808 and 1064nm laser irradiation. Moreover, the NIR-II-active NGP-2-NPs demonstrated promising photoacoustic responses, along with high photostability and biocompatibility, enabling PAI and efficient NIR-II PTT of cancer in vivo.

Virus-like nanoparticles as a theranostic platform for cancer.

Virus-like nanoparticles (VLPs) are natural polymer-based nanomaterials that mimic viral structures through the hierarchical assembly of viral coat proteins, while lacking viral genomes. VLPs have received enormous attention in a wide range of nanotechnology-based medical diagnostics and therapies, including cancer therapy, imaging, and theranostics. VLPs are biocompatible and biodegradable and have a uniform structure and controllable assembly. They can encapsulate a wide range of therapeutic and diagnostic agents, and can be genetically or chemically modified. These properties have led to sophisticated multifunctional theranostic platforms. This article reviews the current progress in developing and applying engineered VLPs for molecular imaging, drug delivery, and multifunctional theranostics in cancer research.

Biocompatible Polymer Nano-Constructs: A Potent Platform for Cancer Theranostics.

Nano-constructs of biocompatible polymers have drawn wide attention owing to their potential as theranostics for simultaneous therapy and detection of cancer. The present mini review summarizes various nano-architectures of polymers that have been developed as theranostic agents for the simultaneous treatment and diagnosis of cancer in a single platform. Additionally, research prospects of polymeric cancer theranostics for the future have been highlighted.

Smart nanomaterials as the foundation of a combination approach for efficient cancer theranostics

Despite numerous FDA-approved treatment and diagnostic approaches, cancer is one of the leading causes of death worldwide due to delayed diagnostics and the limited efficacy of treatments. The life quality of cancer patients is currently improving due to the development of nanotechnology-supported cancer diagnostics, detection, and nanomedicine for targeted drug delivery approaches investigated for cancer therapy. Considering these aspects, this comprehensive review presents the potential of smart nanomaterials (SMs) including carbon nanodots, metal-phenolic networks (MNPs), metal-organic frameworks (MOFs), quantum dots (QDs), carbon nanotubes (CNTs), metal/metal oxide nanoparticles, and silica nanoparticles, as well as polymeric nanoparticles, liposomes, amino acids, proteins, natural polysaccharides, and solid lipid nanoparticles, to manage cancer efficiently in a personalized manner. The outcomes of studies published in the last five years are critically analyzed in this review along with challenges and prospects in terms of a wide range of imaging technologies and therapeutic approaches, such as radiation responsive theranostics, magnetic responsive theranostics, drug delivery, photothermal/photodynamic therapy, and gene therapy. We believe that a novel multidisciplinary and combinational approach can be developed for early-stage cancer diagnostics and treat the diseases with efficacy using nano-based sensors, imaging agents, and pharmacologically relevant cargos. To promote research from the laboratory to clinical application, this review discusses viewpoints based on the tremendous potential of new and developing nanomaterials as a potential platform for cancer theranostics.

Recent Development of Gold Nanoparticles as Contrast Agents for Cancer Diagnosis.

Simple SummaryThe development of nanotechnology has brought revolution to the diagnosis and therapy of diseases, with a high precision and efficacy. Because nanoparticles can integrate multifunctions together including imaging, targeting, and therapeutics, they are more efficient than the standalone diagnostic or therapeutic entities. Among which, gold nanoparticles are most extensively investigated due to their excellent biocompatibility, versatility and ease of functionalization. Excepting the using of gold nanoparticles as vehicles for therapeutics delivery, they are also good candidates as contrast agents for imaging diagnosis, from magnetic resonance imaging, CT and nuclear imaging, fluorescence imaging, photoacoustic imaging to X-ray fluorescence imaging. We summarize their recent applications in these imaging modalities and challenges for their clinical translation.The last decade has witnessed the booming of preclinical studies of gold nanoparticles (AuNPs) in biomedical applications, from therapeutics delivery, imaging diagnostics, to cancer therapies. The synthetic versatility, unique optical and electronic properties, and ease of functionalization make AuNPs an excellent platform for cancer theranostics. This review summarizes the development of AuNPs as contrast agents to image cancers. First, we briefly describe the AuNP synthesis, their physical characteristics, surface functionalization and related biomedical uses. Then we focus on the performances of AuNPs as contrast agents to diagnose cancers, from magnetic resonance imaging, CT and nuclear imaging, fluorescence imaging, photoacoustic imaging to X-ray fluorescence imaging. We compare these imaging modalities and highlight the roles of AuNPs as contrast agents in cancer diagnosis accordingly, and address the challenges for their clinical translation.

Liposomes as Versatile Platform for Cancer Theranostics: Therapy, Bio-imaging, and Toxicological Aspects.

Liposomes are nano-sized formulations having the benefits of site-specificity, biocompatibility, and biodegradability, which make them useful for the therapy and diagnosis of major diseases like cancer. In this review, various synthetic strategies of liposomes and their biomedical application in special concern to cancer are discussed. In context to the biomedical application, this article gives a detailed insight into subcellular targeted therapy and several therapeutic modifications like immunotherapy, receptor-based therapy, phototherapy, and combination therapy. The review also describes the liposome-based imaging platforms and the toxicity associated with liposomes. Owing to a significant amount of benefits of this carrier system, several products have been approved to be launched in the market and several others have already been marketed for clinical use.

Rational Design of All-Organic Nanoplatform for Highly Efficient MR/NIR-II Imaging-Guided Cancer Phototheranostics.

Organic theranostic nanomedicine has precision multimodel imaging capability and concurrent therapeutics under noninvasive imaging guidance. However, the rational design of desirable multifunctional organic theranostics for cancer remains challenging. Rational engineering of organic semiconducting nanomaterials has revealed great potential for cancer theranostics largely owing to their intrinsic diversified biophotonics, easy fabrication of multimodel imaging platform, and desirable biocompatibility. Herein, a novel all-organic nanotheranostic platform (TPATQ-PNP NPs) is developed by exploiting the self-assembly of a semiconducting small molecule (TPATQ) and a new synthetic high-density nitroxide radical-based amphiphilic polymer (PNP). The nitroxide radicals act as metal-free magnetic resonance imaging agent through shortened longitudinal relaxation times, and the semiconducting molecules enable ultralow background second near-infrared (NIR-II, 1000-1700nm) fluorescence imaging. The as-prepared TPATQ-PNP NPs can light up whole blood vessels of mice and show precision tumor-locating ability with synergistic (MR/NIR-II) imaging modalities. The semiconducting molecules also undergo highly effective photothermal conversion in the NIR region for cancer photothermal therapy guided by complementary tumor diagnosis. The designed multifunctional organic semiconducting self-assembly provides new insights into the development of a new platform for cancer theranostics.

Surface architectured black phosphorous nanoconstructs based smart and versatile platform for cancer theranostics

Black Phosphorous has recently gained the attention in the field of 2D nanomaterials owing to its exceptional structure and properties. Specifically, its features like drug loading efficiency, biocompatibility, optical, mechanical, electrical, thermal and phototherapeutic properties contribute to its rising demand as potential alternative to the graphene-based 2D nanomaterials in biomedical applications. Though the BP’s outlook appears promising, its practical applicability is highly challenging. In this review we have discussed the different strategies for synthesis of BP and then briefed its unique properties which renders it a potential platform for the biomedical application. We then discuss the importance of heterogeneous doping on improving the stability of BP against chemical degradation and enhancing its photoelectric properties. Meanwhile, BP-based nanoconjugates, stimuli responsive nanoplatforms, therapeutic, imaging and biosensing platforms are the domains of special and comprehensive interest for versatile biomedical applications of BP. The physicochemical interactions at the nano-bio interface like protein corona formation on the surface on nanoparticles due to interaction with the plasma proteins and others play a crucial role in the biological effects of BP. Hence, we have also discussed the recent studies on interactions between Black phosphorus based nanoconstructs with various biological molecules. Further, it is vital to consider the fact that, though biocompatible, BP-nanomaterials can induce inflammatory responses and exhibit toxicity in dose and time dependant manner. Therefore, we have briefed the biodegradation and toxicological aspects of BP to enlighten the readers about the safety and toxicity of black phosphorous. The future developments of this 2D nanomaterial will not only serve as a boon for oncology, but also functions as a potential nanoplatform for other biomedical applications.

Integrated photodynamic Raman theranostic system for cancer diagnosis, treatment, and post-treatment molecular monitoring.

Theranostics, the combination of diagnosis and therapy, has long held promise as a means to achieving personalised precision cancer treatments. However, despite its potential, theranostics has yet to realise significant clinical translation, largely due the complexity and overriding toxicity concerns of existing theranostic nanoparticle strategies.Methods: Here, we present an alternative nanoparticle-free theranostic approach based on simultaneous Raman spectroscopy and photodynamic therapy (PDT) in an integrated clinical platform for cancer theranostics.Results: We detail the compatibility of Raman spectroscopy and PDT for cancer theranostics, whereby Raman spectroscopic diagnosis can be performed on PDT photosensitiser-positive cells and tissues without inadvertent photosensitiser activation/photobleaching or impaired diagnostic capacity. We further demonstrate that our theranostic platform enables in vivo tumour diagnosis, treatment, and post-treatment molecular monitoring in real-time.Conclusion: This system thus achieves effective theranostic performance, providing a promising new avenue towards the clinical realisation of theranostics.

PET imaging and pharmacological therapy targeting carbonic anhydrase-IX high-expressing tumors using US2 platform based on bivalent ureidosulfonamide.

Carbonic anhydrase-IX (CA-IX) is attracting much attention as a target molecule for cancer treatment since high expression of CA-IX can lead to a poor prognosis of patients. We previously reported low-molecular-weight 111In/90Y complexes with a bivalent ureidosulfonamide scaffold ([111In/90Y]In/Y-US2) as cancer radiotheranostic agents for single photon emission computed tomography and radionuclide-based therapy targeting CA-IX. Here, we applied the US2 platform to positron emission tomography (PET) imaging and pharmacological therapy targeting CA-IX high-expressing tumors by introducing 68Ga and natIn, respectively. In an in vitro cell binding assay, [67Ga]Ga-US2, an alternative complex of [68Ga]Ga-US2 with a longer half-life, markedly bound to CA-IX high-expressing (HT-29) cells compared with low-expressing (MDA-MB-231) cells. In a biodistribution study with HT-29 and MDA-MB-231 tumor-bearing mice, [67Ga]Ga-US2 showed accumulation in the HT-29 tumor (3.81% injected dose/g at 60 min postinjection) and clearance from the blood pool with time. PET with [68Ga]Ga-US2 clearly visualized the HT-29 tumor in model mice at 60 min postinjection. In addition, the administration of [natIn]In-US2 to HT-29 tumor-bearing mice led to tumor growth delay and prolonged mouse survival, while no critical toxicity was observed. These results indicate that [68Ga]Ga-US2 and [natIn]In-US2 may be useful imaging and therapeutic agents targeting CA-IX, respectively, and that US2 may serve as an effective cancer theranostic platform utilizing CA-IX.

PH/Reduction Dual Stimuli-Triggered Self-Assembly of NIR Theranostic Probes for Enhanced Dual-Modal Imaging and Photothermal Therapy of Tumors.

Tumor microenvironment plays a pivotal role in the growth and metastasis of tumors, and has become a promising target for precise diagnosis and treatment of tumors. Herein, a novel smart NIR theranostic probe Cy-1 that can simultaneously respond to low intracellular pH and reductive glutathione (GSH) is reported. This probe has demonstrated to be able to intermolecularly undergo a biologically compatible CBT-Cys condensation reaction to selectively form large nanoaggregates in the tumor microenvironment resulting in its enhanced accumulation and retention in the tumor, which as a consequence significantly improves the sensitivity of NIR/photoacoustic imaging and photothermal therapeutic efficacy of tumors in living mice. We thus believe that this dual stimuli-mediated self-assembly strategy may offer a promising and universal platform for cancer theranostics.

Solution combustion synthesis (SCS) of theranostic ions doped biphasic calcium phosphates; kinetic of ions release in simulated body fluid (SBF) and reactive oxygen species (ROS) generation

Theranostic ions offer suitable platforms for cancer theranostics; here, the effect of doping various amounts of theranostic ions (i.e., Sr2+, Fe2+, and Ti4+ ions) on the physicochemical properties and biological activities of calcium phosphates (CaPs) were investigated. The solution combustion synthesis (SCS) was conducted at different amounts of ions (i.e., = 0.1, 0.25, 0.5 mol). Desirable physicochemical properties were obtained in doped samples with 0.1 mol of ions. The particle size of the Sr, Fe, and Ti-doped samples was decreased from 68 to 39, 24, and 29 nm, respectively. The surface charge of the mentioned samples was changed from −20 to −24, −28, and −25 mV, respectively. Besides, the specific surface area of the mentioned samples was significantly increased from 38 to 79, 65, and 106 m2/g, respectively. It was found that bioactivity of doped CaPs improved ~95%, which relied on the high adsorption and desorption rate of Ca2+ ions in the simulated body fluid (SBF). The in vitro cell-based results demonstrate the potent effect of CaPs and theranostic ions doped CaPs on the reactive oxygen species (ROS) generation. In the presence of CaPs, the intracellular ROS generation is increased by about 60%. Besides, the intracellular ROS generation is improved in Sr2+, Fe2+, and Ti4+ ions doped CaPs by about 66, 64, and 68%. As a result of the high generation of ROS, the bone nodule formation of cell treated CaPs and theranostic ions doped CaPs is improved 25%–37%. Finally, it can be concluded that the use of the SCS approaches for doping of theranostic ions causes well-physicochemical properties and high biological activities.

Functional metal–organic framework-based nanocarriers for accurate magnetic resonance imaging and effective eradication of breast tumor and lung metastasis

The use of nanoscale metal–organic frameworks (MOFs) as drug delivery vehicles has attracted considerable attention in tumor therapy. In this study, novel biocompatible MOF-based nanocarriers were used as part of a facile and reproducible strategy for precision cancer theranostics. Both diagnostic (Mn2+) and therapeutic compounds (doxorubicin, DOX) were incorporated into the multifunctional MOF-based nanocarriers, which exhibited high colloidal stability and promoted T1-weighted proton relaxivity and low-pH-activated drug release. The obtained MOF-based nanocarriers exhibited significantly high cellular uptake and efficient intracellular drug delivery into cancer cells, which resulted in high apoptosis and cytotoxicity, in addition to effectively inhibiting the migration of 4T1 breast cancer cells. Moreover, the MOF-based nanocarriers could intensively deliver diagnostic and therapeutic agents to tumors to enable precise visualization of the nanocarrier accumulation and accurate tumor positioning, diagnosis, and imaging-guided therapy using magnetic resonance imaging (MRI). In addition, the functional MOF-based nanocarriers exhibited effective ablation of the primary breast cancer, as well as significant inhibition of lung metastasis with a high survival rate. Therefore, the developed nanocarriers represent a viable platform for cancer theranostics.

Virus-Inspired Deformable Mesoporous Nanocomposites for High Efficiency Drug Delivery.

Mesoporous nanoparticles as a versatile platform for cancer theranostics have been widely used, but their cellular delivery efficiency is still far from satisfactory. Although deformability is emerging as an important parameter influencing cellular uptake enhancement, the facile synthesis of deformable mesoporous nanocomposite with adjustable mechanical property is challenging but meaningful for a deeper understanding of cellular uptake mechanisms and significantly improving cancer therapy. In this work, yolk-shell structured eccentric mesoporous organosilica (YEMO) nanocomposites with adjustable mechanical property are successfully prepared by an organosilane-assisted anisotropic self-assembly approach. The feasibility to precisely control the mechanical property of the YEMO by manipulating the structural parameters, the crosslinking degree of mesoporous framework, and the rotation rate of the reaction is demonstrated. The study of the fabrication mechanism and mechanical properties of YEMO are discussed in detail. The Young's modulus (EY ) of YEMO can be adjusted from 2.4 to 65 MPa. Thereby, the continuous control of the cellular uptake from ≈15% to ≈80% under the same incubation time is achieved. To further prove the higher efficiency drug delivery of YEMO with soft characteristics, the higher toxicity of the "soft" YEMO loaded with the anticancer drug doxorubicin compared to the "stiff" one is demonstrated.

A structurally precise AgxAu25-x nanocluster based cancer theranostic platform with tri-targeting/in situ O2-generation/aggregation enhanced fluorescence imaging/photothermal-photodynamic therapies.

The photothermal and photodynamic performances of structurally precise oil-soluble AgxAu25-x (x ≤ 13) nanoclusters were first explored and they were solubilized into new assemblies to form a versatile cancer theranostic platform with tri-targeting/in situ O2-generation/aggregation enhanced fluorescence imaging/photothermal-photodynamic therapy effects, which will provide an important reference for precision theranostics at the atomic level in future.

Biotinylated platinum(ii) metallacage towards targeted cancer theranostics.

A biotinylated metallacage was prepared via metal-coordination-driven self-assembly, combining fluorescence, anti-cancer and targeted properties into a single entity. The metallacage was successfully employed for targeted cell imaging and therapy, offering a platform for cancer theranostics.

Multifunctional magnetic iron oxide nanoparticles: an advanced platform for cancer theranostics.

Multifunctional magnetic nanoparticles and derivative nanocomposites have aroused great concern for multimode imaging and cancer synergistic therapies in recent years. Among the rest, functional magnetic iron oxide nanoparticles (Fe3O4 NPs) have shown great potential as an advanced platform because of their inherent magnetic resonance imaging (MRI), biocatalytic activity (nanozyme), magnetic hyperthermia treatment (MHT), photo-responsive therapy and drug delivery for chemotherapy and gene therapy. Magnetic Fe3O4 NPs can be synthesized through several methods and easily surface modified with biocompatible materials or active targeting moieties. The MRI capacity could be appropriately modulated to induce response between T1 and T2 modes by controlling the size distribution of Fe3O4 NPs. Besides, small-size nanoparticles are also desired due to the enhanced permeation and retention (EPR) effect, thus the imaging and therapeutic efficiency of Fe3O4 NP-based platforms can be further improved. Here, we firstly retrospect the typical synthesis and surface modification methods of magnetic Fe3O4 NPs. Then, the latest biomedical application including responsive MRI, multimodal imaging, nanozyme, MHT, photo-responsive therapy and drug delivery, the mechanism of corresponding treatments and cooperation therapeutics of multifunctional Fe3O4 NPs are also be explained. Finally, we also outline a brief discussion and perspective on the possibility of further clinical translations of these multifunctional nanomaterials. This review would provide a comprehensive reference for readers to understand the multifunctional Fe3O4 NPs in cancer diagnosis and treatment.

Static DNA Nanostructures For Cancer Theranostics: Recent Progress In Design And Applications.

Among the various nano/biomaterials used in cancer treatment, the beauty and benefits of DNA nanocomposites are outstanding. The specificity and programmability of the base pairing of DNA strands, together with their ability to conjugate with different types of functionalities have realized unsurpassed potential for the production of two- and three-dimensional nano-sized structures in any shape, size, surface chemistry and functionality. This review aims to provide an insight into the diversity of static DNA nanodevices, including DNA origami, DNA polyhedra, DNA origami arrays and bioreactors, DNA nanoswitch, DNA nanoflower, hydrogel and dendrimer as young but promising platforms for cancer theranostics. The utility and potential of the individual formats in biomedical science and especially in cancer therapy will be discussed.

Two-dimensional nanosheets with high curcumin loading content for multimodal imaging-guided combined chemo-photothermal therapy

Nowadays, two-dimensional (2D) nanomaterials with many fascinating physicochemical properties have drawn extensive attention as drug delivery platforms for cancer theranostics. Nevertheless, current existing 2D nanomaterial-based drug delivery systems normally undergo the bottlenecks of hash preparation process, low drug loading content and unsatisfactory therapeutic outcome. Herein, we developed a novel nanoparticles-induced assemble strategy to construct 2D nanosheets with ultra-high curcumin loading content of 59.6 % and excellent stability in water. Furthermore, a distinct photothermal effect and multimodal imaging property after polydopamine coating could be obtained, thereby leading to precise and efficient ablation of tumor in combination of curcumin-induced chemotherapy. More importantly, the design principle of our work offers novel facile strategy to assemble metal-binding drugs into 2D nanomedicine with high drug content and well-defined shapes.
