Nano-enabled pesticides: a comprehensive toxicity assessment of tebuconazole nanoformulations with nematodes at single species and community level

There is an increasing imperative to explore safer alternatives for pesticides due to their indiscriminate use and consequential health impacts on the environment and humans. Nanoformulations of pesticides are being developed as potential alternatives due to their beneficial properties, including enhanced solubility, targeted delivery to the site of action, improved stability and efficacy and reduced non-target effects. Nevertheless, a comprehensive assessment is necessary for these emerging nanopesticides compared to existing formulations, aiming to ascertain whether their "nano" characteristics exacerbate toxicity for non-target organisms. This study investigated the toxicity of tebuconazole (TBZ) in different formulations, including nanoformulations (poly-ε-caprolactone [PCL] and nanostructured lipid carrier [NLC] loaded with TBZ), as well as a commercial formulation, on the reproduction of the nematode Caenorhabditis elegans in both aqueous and soil matrices. Additionally, the impact of the correspondent nanocarriers without TBZ on C. elegans was examined. In water, TBZ in the form of nano and commercial formulations exhibited higher toxicity on the nematodes' reproduction than the TBZ (a.s.) attributable to higher freely dissolved concentrations of TBZ, which resulted in a toxicity order, ranging from the most to the least toxic as follows: NLC-TBZ > PCL-TBZ > commercial formulation > TBZ (a.s.). For NLC-TBZ, the excess toxicity could be clearly explained by combined toxicity of TBZ (a.s.) and nanocarriers, with the effect addition of the separate single compounds matching the observed effects of the nanoformulation. For PCL-TBZ, effects were stronger than expected from the effect addition of TBZ (a.s.) and PCL nanocarriers, potentially due to enhanced bioavailability of encapsulated TBZ in the gut of the nematodes. In soil, NLC with and without loaded TBZ showed higher toxicity than other tested compounds, while PCL nanocarriers without TBZ did not exhibit negative effects on the reproduction of C. elegans. Microcosm experiment, where long-term effects on native soil nematode fauna were tested, confirmed that TBZ-nanoformulations act via combined toxic effects of TBZ and nanocarriers. These findings contribute valuable insights to understanding nanopesticides' ecotoxicity and underscore the need for harmonized regulatory assessments to evaluate these novel formulations adequately.

Experimental investigation of the effect of tebuconazole on three biomarkers of innate immunity in the house sparrow (Passer domesticus).

Triazoles are among the most widely used fungicides in the world due to their efficacy against fungal crop diseases and their broad spectrum of action. Intensive use of triazoles has resulted in residual contamination in different compartments of agroecosystems and exposes non-target species to potential sublethal effects. Triazoles are known to be immunomodulators in medicine and therapeutic treatments, but very little data is available on their potential effect on immune parameters of non-target vertebrate species living in agroecosystems. In this study, we experimentally examined the impact of tebuconazole on three immune biomarkers (haemagglutination titre (HA), haemolysis titre (HL), and haptoglobin concentration (Hp)), as well as on the body condition of house sparrows (Passer domesticus). Our results suggest that tebuconazole had very little, if any, effect on the studied immune parameters. However, further studies are needed to better assess the effect of tebuconazole on bird immunity because (1) experimental individuals were kept under optimal conditions and the impact of tebuconazole on immunity may occur under suboptimal conditions, (2) only one concentration of tebuconazole was tested and its effect could be dose-dependent and (3) other complementary immunological biomarkers should be studied, given the complexity of the vertebrate immune system. Current knowledge on the potential effects of triazoles on the immunity of wild farmland vertebrates is still largely insufficient. Further physiological and immune studies should be conducted to better understand the effect of triazole fungicides on farmland birds.

Effects of acute exposure to environmentally realistic tebuconazole concentrations on stress responses of kidney and digestive gland of Lymnaea stagnalis

This study aimed to investigate the effects of 24 and 72 h exposure to environmentally relevant concentrations of tebuconazole (TEB) (10, 100 and 500 µg/L) fungicide on the freshwater snail Lymnaea stagnalis. The focus was induction of oxidative stress, alteration of gene expressions and histopathological changes in the kidney and digestive gland. TEB treatment induced a time- and concentration-dependent increase in intracellular reactive oxygen species (ROS) and malondialdehyde (MDA) levels, while the total antioxidant capacity (TAC) was decreased. The activities of glutathione peroxidase (GPx), glutathione reductase (GR), and catalase (CAT) also increased in a time- and concentration-dependent manner in both tissues. TEB exposure significantly increased the mRNA levels of CAT, GPx, GR, heat shock proteins HSP40 and HSP70. Histological analysis revealed nephrocyte degeneration and disrupted digestive cells. The study concludes that acute exposure to TEB induces oxidative stress, alters antioxidant defense mechanisms, and leads to histopathological changes in L. stagnalis.

Fungicides from rice cultivation (tebuconazole and azoxystrobin) alters biochemical and histological markers of hammertoad tadpoles (Boanafaber).

Tebuconazole (TBZ) and azoxystrobin (AZX) are fungicides frequently used in rice cultivation. Despite protecting crops against fungal diseases, these compounds can contaminate the natural environments close to the crops, exerting negative effects on non-target organisms, the present study aimed to characterize the contamination by fungicides of a river that flows in an area dominated by rice cultivation in the north of the state of Santa Catarina, SC, Brazil. Concentrations of TBZ and AZX found in the field were used to evaluate their negative effects on development, biochemical biomarkers and histopatology of the liver of a native tadpole species, the hammerfrog (Boana faber). Tadpoles were exposed for 16 days to the lowest (1.20 μg/L) and highest (2.60 μg/L) concentration of TBZ, lowest (0.70 μg/L) and highest (1.60 μg/L) concentration of AZX, and the mix of both fungicides at lowest and highest concentration of each found in field analyses. Exposure to the lower TBZ concentration and both concentrations of the Mix accelerated the development of tadpoles. AZX caused an increase in the activities of glutathione S-transferase (GST), carboxylesterase (CbE) and glucose-6-phosphate dehydrogenase (G6PDH) in the liver, an increase in the levels of protein carbonyls (PC) in the liver and an increase in the activity of acetylcholinesterase (AChE) in muscle of tadpoles. TBZ, on the other hand, generated an increase in GST, G6PDH, PC and histopathological severity scores in liver and in muscle AChE activity. The effects were more intense in the groups exposed to the Mix of contaminants. No treatment altered brain AChE. The data showed that the fungicides from in rice cultivation found in natural aquatic environments around the crops pose risks to the health of the animals, compromising their metabolism and development.

Effects of Tebuconazole on the Earthworm Dendrobaena veneta: Full Life Cycle Approach

Tebuconazole (TEB), a widely used triazole fungicide, is effective against soil-borne and foliar fungal pathogens. Toxicants can exhibit varying effects depending on the life stage of organisms, although standard toxicity tests typically focus on adult individuals. This study aimed to assess TEB’s potential adverse effects on the earthworm Dendrobaena veneta throughout its life cycle. Effects were evaluated by exposing cocoons to varying TEB concentrations, monitoring hatching, newly hatched juvenile mass, and growth to adulthood. A reproduction test assessed impacts on adults, offering insights into how these results compared with cocoon exposure findings. Results revealed that TEB delayed hatching at concentrations of 25, 50, and 100 mg/kg by 6, 8, and 15.5 days, respectively. Newly hatched juveniles exhibited a 15.96% (50 mg/kg) and 27.37% (100 mg/kg) reduction in body mass compared to controls, with no subsequent compensation during growth. Results from the reproduction tests showed no adverse effects on adult survival, but the effects are observed on juveniles, indicating a higher sensitivity of this developmental stage. While several adverse effects were observed, it is important to note that these occurred at concentrations exceeding recommended application rates. TEB appears safe for earthworms when used correctly, but the presence of multiple contaminants and stressors warrants consideration.

Impact of technological processes on tebuconazole reduction in selected cereal species and the primary cereal product, and dietary exposure assessment.

Contamination of cereals with tebuconazole (TEB) can affect the dietary risk assessment. This study investigates, for the first time, how mechanical, thermal, physical-chemical, and biochemical processes affect the TEB level in wheat, rye, and barley. The biochemical process of malting was the most effective for tebuconazole reduction (by 86%) in cereals. Thermal processes were also effective, i.e., boiling (70%) and baking (55%). These processes considerably decreased the concentration of tebuconazole, and Procesing Factors (PFs) were from 0.10 to 0.18 (malting), 0.56 to 0.89 (boiling), and 0.44 to 0.45 (baking), respectively. The concentration of TEB was not reduced after the application of mechanical processing. The risk was estimated in dietary exposure assessment on the basis of the highest reported levels of tebuconazole residues bread. At a high level of rye bread consumption, the potential exposure to tebuconazole reached only 3.5% and 2.7% in children and adults, respectively.

Chronic exposure to tebuconazole alters thyroid hormones and plumage quality in house sparrows (Passer domesticus).

Triazoles belong to a family of fungicides that are ubiquitous in agroecosystems due to their widespread use in crops. Despite their efficiency in controlling fungal diseases, triazoles are also suspected to affect non-target vertebrate species through the disruption of key physiological mechanisms. Most studies so far have focused on aquatic animal models, and the potential impact of triazoles on terrestrial vertebrates has been overlooked despite their relevance as sentinel species of contaminated agroecosystems. Here, we examined the impact of tebuconazole on the thyroid endocrine axis, associated phenotypic traits (plumage quality and body condition) and sperm quality in wild-caught house sparrows (Passer domesticus). We experimentally exposed house sparrows to realistic concentrations of tebuconazole under controlled conditions and tested the impact of this exposure on the levels of thyroid hormones (T3 and T4), feather quality (size and density), body condition and sperm morphology. We found that exposure to tebuconazole caused a significant decrease in T4 levels, suggesting that this azole affects the thyroid endocrine axis, although T3 levels did not differ between control and exposed sparrows. Importantly, we also found that exposed females had an altered plumage structure (larger but less dense feathers) relative to control females. The impact of tebuconazole on body condition was dependent on the duration of exposure and the sex of individuals. Finally, we did not show any effect of exposure to tebuconazole on sperm morphology. Our study demonstrates for the first time that exposure to tebuconazole can alter the thyroid axis of wild birds, impact their plumage quality and potentially affect their body condition. Further endocrine and transcriptomic studies are now needed not only to understand the underlying mechanistic effects of tebuconazole on these variables, but also to further investigate their ultimate consequences on performance (i.e. reproduction and survival).

Improvement of Akanthomyces lecanii resistance to tebuconazole through UV-C radiation and selective pressure on microbial evolution and growth arenas

Tebuconazole (TEB) is a fungicide widely used in agriculture; however, its constant application has increased the emergence of resistant plant pathogenic fungal strains and reduced the effectiveness of fungi as biological control agents; for instance, the entomopathogenic and hyperparasitic fungus Akanthomyces lecanii, suitable for simultaneous biological control of insect pest and plant pathogenic fungi, is highly sensitive to fungicides. We carried out the induction of resistance to TEB in two wild type strains of A. lecanii by UV radiation and selective pressure in increasing fungicide gradients using a modified Microbial Evolution and Growth Arena (MEGA), to produce A. lecanii strains that can be used as biological control agent in the presence of tebuconazole. Nine UV-induced and three naturally adapted A. lecanii strains were resistant to TEB at the agriculturally recommended dose, and three irradiated strains were resistant to TEB concentration ten times higher; moreover, growth, sporulation rates, production of hydrolytic enzymes, and virulence against the hemipteran Coccus viridis, a major pest of coffee crops, were not affected in the TEB-resistant strains. These A. lecanii TEB-resistant strains would have a greater opportunity to develop and to establish themselves in fields where the fungicide is present and can be used in a combined biological-chemical strategy to improve insect and plant pathogenic fungal control in agriculture. Also, the selective pressure through modified MEGA plate methodology can be used for the adaptation of entomopathogenic filamentous fungi to withstand other chemical or abiotic stresses that limits its effectiveness for pest control.

Genotoxic effects induced for sub-cytotoxic concentrations of tebuconazole fungicide in HEp-2 cell line

Tebuconazole (TB) is an active ingredient in formulations applied to control of fungal diseases in plants and classified as a possible human carcinogen. In the present study, the sub-cytotoxic concentrations of TB were evaluated for genotoxicity response in HEp-2 cell line. The HEp-2 cell line was exposed to 20, 40, and 50 μg/mL of TB in the Cytokinesis-block micronucleus (CBMN) assay and 40, 60, and 80 μg/mL in the comet assay (CA). Both negative centromere micronuclei (MNC-) as DNA damage were induced by the concentrations of TB assayed, suggesting a mostly clastogenic effect. The results obtained from the comet assay suggest that part of the endogenous DNA damage in the HEp-2 cell line could be repaired in presence of low TB concentrations by promoting the damage response. In conclusion, exposition to sub-cytotoxic concentrations of TB induces genotoxicity in the HEp-2 cell line. Therefore, the safety of their application should be evaluated.

Multi-characteristic toxicity of enantioselective chiral fungicide tebuconazole to a model organism Mediterranean mussel Mytilus galloprovincialis Lamarck, 1819 (Bivalve: Mytilidae)

The survey of available scientific literature shows a lack of data on the chronic effects of tebuconazole (TEB) on non-target aquatic organisms. Therefore, this study evaluates toxicity (10 and 20 days) of two considered concentrations 2 ng/L (E1) and 2 μg/L (E2) of TEB to bioindicator species Mytilus galloprovincialis. To this end, the TEB concentrations measured in soft mussel tissues showed a time-dependent increasing trend. The viability of haemocyte and digestive gland (DG) cells was higher than 95 % during the experiment. However, DG cells lost the ability to regulate their volume in both groups after 20-d. The E1 treatment increased Cl− and Na+ levels, and E2 decreased Na+ levels in the haemolymph. In addition, levels of superoxide dismutase (SOD) activity and oxidatively modified protein (OMP) increased after 10- and 20-d in both treatments. Histopathological findings showed abnormalities in the E2, e.g., haemocyte infiltration, hypertrophy, and hyperplasia in gills and DG. This study reveals the potential risks of TEB usage in the model organism M. galloprovincialis, primarily via bioaccumulation of TEB in food web links, and improves knowledge about its comprehensive toxicity.

Evidence of environmental transfer of tebuconazole to the eggs in the house sparrow (Passer domesticus): An experimental study

Triazole compounds are among the most widely used fungicides in agroecosystems to protect crops from potential fungal diseases. Many farmland birds spend a significant part of their life cycle in agroecosystems, which may chronically expose them to pesticides. We experimentally tested whether exposure to environmental concentrations of tebuconazole could induce a contamination of the eggs in an agroecosystem sentinel species, the house sparrow (Passer domesticus). Wild-caught adult sparrows were maintained in captivity and exposed (exposed group) or not (control group) for seven months to tebuconazole through drinking water. Eggs were opportunistically collected for the determination of tebuconazole concentration by Liquid Chromatography coupled to tandem Mass Spectrometry in eggs. We found that eggs from exposed parents all contained tebuconazole with a mean concentration of 1.52 ng g−1 dry weight. In eggs from control parents, the tebuconazole concentration was below the limit of quantification (0.23 ng g−1 dry weight) for 11 out of 13 eggs. Thus, our study demonstrates for the first time that environmental exposure of female birds to tebuconazole can translate into egg contamination by this fungicide.

Effects of tebuconazole application at different growth stages on rice grain quality of rice-based untargeted metabolomics.

Tebuconazole (TEB) is a pesticide widely used in crops and has a strong control effect on fungal pathogens. TEB abuse has caused many food safety problems. In this study, the TEB residue in rice and the effect of TEB on white rice quality were investigated. The results showed that under two spraying concentrations, the TEB residue in rice was 11.21-19.05μg/kg and 24.45-31.12μg/kg, and there was no food safety risk of pesticide residue. When applying TEB according to the instructions, no significant effect was found. However, when 3 times the recommended TEB concentration was used at the filling stage, the protein content of white rice decreased significantly from 106.52mg/g to 80.72mg/g. At the jointing，heading and filling stage, the amylose content of white rice decreased to 53.95mg/g, 48.77mg/g and 49.04mg/g from the blank control group. Plant metabolic analysis using LC-QTOF/MS revealed that the amino acid-related metabolic pathways in white rice were significantly affected by TEB. This is closely related to the decrease in protein accumulation in white rice and the stress response of rice plants. The increase in pantothenic acid content in white rice indicated that the glycolysis pathway of white rice plants was affected, and the consumption of starch and sucrose increased, leading to the inhibition of amylose accumulation in white rice. The increase in soluble sugar content and decrease in phosphocholine content in white rice suggested that rice plants were affected by TEB exposure, which produced similar effects under drought stress.

Residual Level, Histology, and Blood Biochemistry of Tebuconazole: A Repeated Dose 28-Day Oral Toxicity Study in Pigs.

In this study, we investigated the residual properties of tebuconazole-treated pigs. Twentypigs were treated with different concentrations (0.25, 1.25, 2.5, 12.5, and 25 mg/kg bw/d) of tebuconazole for 28 d. Blood biochemistry, histology, and residual levels were analyzed using the VetTest analyzer, Masson’s trichrome staining kit, and liquid chromatography-mass spectrometry, respectively. The final body weights were not significantly different between the control and treatment groups. Alkaline phosphatase, blood urea nitrogen, cholesterol, and gamma-glutamyl transpeptidase levels were significantly different from those of the control after exposure for 14 d. However, alanine aminotransferase levels showed changes only after exposure to pesticides for 28 d. The biochemical parameters were separated during the experimental period (14 d versus 28 d) by principal component analysis. Based on variable importance plots, blood urea nitrogen/creatinine ratio, blood urea nitrogen, glucose, and gamma-glutamyl transpeptidase are candidate biomarkers for tebuconazole exposure. The residual levels were observed at T4 (12.5 mg/kg bw/d) and T5 (25 mg/kg bw/d) in the liver and fat tissues, respectively. Fibrosis increased in the liver, kidney, and fat tissues, depending on the tebuconazole concentration. In conclusion, the residue limits of tebuconazole and the physiological changes caused by dietary tebuconazole in pigs provide important information for establishing maximum residue limits of pork and pork products.

Effect of short-term exposure to low concentration of tebuconazole: morphological, histometric and functional modifications in Danio rerio liver

Due to its high toxicity and bioaccumulation tendency, tebuconazole (TBZ) is one of the ten substances posing the highest risk of harmful effects in aquatic ecosystems. The liver, a key compartment for xenobiotics detoxification, is also the organ in which TBZ mainly accumulates in fish. Herein, we investigated for the first time the morpho-functional changes induced in zebrafish (Danio rerio) liver after a short-term exposure (48, 96, and 192 hours) to a low, environmentally relevant concentration of TBZ (5 µg/L) to disclose the early effects under a realistic exposure scenario. We revealed that pathological alterations with varying degrees of severity could be detected in all the examined samples. The injuries become intense and irreversible with increased exposure time involving both hepatocytes and vascular components based on the degree of tissue changes. The main morphological alterations were: parenchyma dyschromia, macrophages infiltration, congestion of blood vessels, and sinusoids. TBZ exposure also resulted in a significant decrease in glycogen contents and hepatocyte dimensions, and the modulation of superoxide dismutase, an early indicator of oxidative stress. We demonstrated that even a very low dose of TBZ affects hepatic morphology and function, disrupting liver homeostasis and physiology.

Multiple Mutations and Overexpression in the CYP51A and B Genes Lead to Decreased Sensitivity of Venturia effusa to Tebuconazole

Multiple demethylation-inhibiting (DMI) fungicides are used to control pecan scab, caused by Venturia effusa. To compare the efficacy of various DMI fungicides on V. effusa, field trials were conducted at multiple locations applying fungicides to individual pecan terminals. In vitro assays were conducted to test the sensitivity of V. effusa isolates from multiple locations to various concentrations of tebuconazole. Both studies confirmed high levels of resistance to tebuconazole. To investigate the mechanism of resistance, two copies of the CYP51 gene, CYP51A and CYP51B, of resistant and sensitive isolates were sequenced and scanned for mutations. In the CYP51A gene, mutation at codon 444 (G444D), and in the CYP51B gene, mutations at codon 357 (G357H) and 177 (I77T/I77L) were found in resistant isolates. Expression analysis of CYP51A and CYP51B revealed enhanced expression in the resistant isolates compared to the sensitive isolates. There were 3.0- and 1.9-fold increases in gene expression in the resistant isolates compared to the sensitive isolates for the CYP51A and CYP51B genes, respectively. Therefore, two potential mechanisms—multiple point mutations and gene over expression in the CYP51 gene of V. effusa isolates—were revealed as likely reasons for the observed resistance in isolates of V. effusa to tebuconazole.

Histopathological alterations in Van fish (Alburnus tarichi Güldenstädt, 1814) exposed to tebuconazole

ABSTRACT Tebuconazole (TEB) is one of the most commonly used fungicides in the Lake Van Basin in the Eastern Anatolia Region, Turkey, which is one of the largest alkaline lakes in the world. In this study, the effects of TEB on the Van fish, the only vertebrate species living in Lake Van were investigated. The TEB was administered at a dose of 2.5 mg/L to the Van fish, which were examined histologically via the gill, liver, and kidney tissues after 24, 48, 72 and 96 h of exposure to TEB. Normal histological structure was observed in the gill, liver, and kidney tissues in the control group. Epithelial hyperplasia, epithelial necrosis, epithelial lifting, lamellar fusion, and an increase in mucus cell counts were observed in the gill tissues of the fish exposed to TEB. Dilatation and congestion of the sinusoids, melano-macrophages aggregation, and, focal necrosis were detected in the liver tissues of the fish exposed to TEB. Bleeding in the kidney tissues, hyaline droplet degeneration, contraction of the glomerulus and enlargement of the space within the Bowman’s capsule were observed. The findings of the present study indicated that sublethal concentrations of TEB result in histopathologic changes in the gill, liver, and kidney.

Polymerase Chain Reaction-Assisted Evaluation of the Efficacy of Seed-Treatment Prevention of Sporisorium reilianum Infection in Sorghum Seedlings.

Head smut, caused by Sporisorium reilianum [(Kuhn) Langdon and Fullerton], is a major disease of sorghum. Seed treatment is considered to be the most effective way to control the disease; however, the pathogen can infect at the seedling stage and the infected plant will not display symptoms until the reproductive stage is reached. The evaluation of the efficacy of seed treatments is time consuming and is dependent upon visible symptoms. Polymerase chain reaction (PCR) methods have the ability to identify pathogens and diagnose their presence at an early stage of infection. In this study, the S. reilianum-specific primer SR3 was used for PCR detection pathogen. We optimized temperature, humidity, and spore quantity test conditions and were able to achieve >88% infection incidence in sorghum seedlings. Sorghum seeds were soaked in various concentrations of tebuconazole and planted for 7 days in soil containing 0.2% teliospores. The efficacy of tebuconazole against S. reilianum was evaluated by PCR and recorded as disease incidence. Results indicated that the reduction in disease incidence after exposure to 0.15, 0.30, 0.45, 0.60, and 0.75 μg/mL tebuconazole was 6.24, 37.48, 67.74, 81.24, and 93.74%, respectively. Significant differences between the concentrations of tebuconazole were observed. The PCR assay represents a valuable tool for evaluating the efficacy of fungicide seed treatments for the control of S. reilianum in sorghum under laboratory conditions.

Effect of tebuconazole and trifloxystrobin on Ceratocystis fimbriata to control black rot of sweet potato: processes of reactive oxygen species generation and antioxidant defense responses.

Black rot, caused by Ceratocystis fimbriata, is one of the most destructive disease of sweet potato worldwide, resulting in significant yield losses. However, a proper management system can increase resistance to this disease. Therefore, this study investigated the potential of using tebuconazole (TEB) and trifloxystrobin (TRI) to improve the antioxidant defense systems in sweet potato as well as the inhibitory effects on the growth of and antioxidant activity in C. fimbriata. Four days after inoculating cut surfaces of sweet potato disks with C. fimbriata, disease development was reduced by different concentrations of TEB + TRI. Infection by C. fimbriata increased the levels of hydrogen peroxide (H2O2), malondialdehyde (MDA), and electrolyte leakage (EL), and the activity of lipoxygenase (LOX) by 138, 152, 73, and 282%, respectively, in sweet potato disks, relative to control. In the sweet potato disks, C. fimbriata reduced the antioxidant enzyme activities as well as the contents of ascorbate (AsA) and reduced glutathione (GSH) by 82 and 91%, respectively, compared with control. However, TEB + TRI reduced the oxidative damage in the C. fimbriata-inoculated sweet potato disks by enhancing the antioxidant defense systems. On the other hand, applying TEB + TRI increased the levels of H2O2, MDA, and EL, and increased the activity of LOX in C. fimbriata, in which the contents of AsA and GSH decreased, and therefore, inhibited the growth of C. fimbriata. These results suggest that TEB + TRI can significantly control black rot disease in sweet potato by inhibiting the growth of C. fimbriata.

Personal exposure assessment of pesticides in residents: The association between hand wipes and urinary biomarkers

BackgroundResidential exposure to pesticides may occur via inhalation of airborne pesticides, direct skin contacts with pesticide-contaminated surfaces, and consumption of food containing pesticide residues. The aim was to study the association of dermal exposure to pesticides between the use and non-use periods, between farmer and non-farmer families and between dermal exposure and the excretion of metabolites from urine in residents living close to treated agricultural fields. MethodsIn total, 112 hand wipes and 206 spot urine samples were collected from 16 farmer and 38 non-farmer participants living within 50 m from an agricultural field in the Netherlands. The study took place from May 2016 to December 2017 during the use as well as the non-use periods of pesticides. Hand wipes were analysed for the parent compound and urines samples for the corresponding urinary metabolite of five applied pesticides: asulam, carbendazim (applied as thiophanate-methyl), chlorpropham, prochloraz and tebuconazole. Questionnaire data was used to study potential determinants of occurrence and levels of pesticides in hand wipes according to univariate and multivariate analysis. ResultsCarbendazim and tebuconazole concentrations in hand wipes were statistically significantly higher in the pesticide-use period compared to the non-use period. In addition, especially during the use periods, concentrations were statistically significantly higher in farmer families compared to non-farmer families. For asulam, chlorpropham and prochloraz, the frequency of non-detects was too high (57–85%) to be included in this analysis. The carbendazim contents in urine samples and hand wipes were correlated on the first and second day after taking the hand wipe, whereas chlorpropham was only observed to be related on the second day following the spray event. ConclusionsConcentrations in hand wipes were overall higher in pesticide use periods compared to non-use periods and higher in farmer families compared to non-farmer families. Only for carbendazim a strong correlation between concentrations in hand wipes and its main metabolite in urine was observed, indicating dermal exposure via contaminated indoor surfaces. We expect this to be related to the lower vapour pressure and longer environmental lifetime of carbendazim compared to the other pesticides studies.

Toxicogenetic of tebuconazole based fungicide through Lactuca sativa bioassays

The rampant use of pesticides can cause serious environmental problems. They can be contaminating surface water and groundwater, affecting the surrounding micro and macro biota. In this sense, this work aimed to evaluate the effects of a tebuconazole-based fungicide through endpoints accessed in Lactuca sativa bioassays. Germinated-seeds with roots upon 2 mm were treated with a fungicide containing Tebuconazole (TBZ) as active compound. The final concentration of TBZ in the tested solutions were 0.025 (C1); 0.05 (C2); 0.1 (C3); 0.2 (C4) and 0.4 g/L (C5). L. sativa roots were exposed for 24 h to these solutions and Petri dishes containing the treated seeds were kept in incubation chamber at 24 °C. Two positive controls (PC,) the herbicide trifluralin (0.84 mg/L) and Methanesulfonate (4 ×10−4 mol/L), were applied. Distilled water was negative control (NC). The following endpoints were analyzed: root growth (RG), cytogenotoxic potential by cell cycle analysis, induction of DNA damage through TUNEL and comet assays. The obtained data were submitted to one-way variance analysis (ANOVA) and then to Tukey or Kruskal Wallis (P < 0.05) tests. The concentrations (C1, C2, C4 and C5) affected negatively the RG of L. sativa, in comparison with the NC. The mitotic index was reduced by 25% from NC to C1 and in the rest of treatments it did not present significant modifications. However, from C3 to C5 great amount of chromosome alterations were observed, in comparison with the NC. TBZ-based fungicide also induced DNA fragmentation as measured by TUNEL and comet assays. Thus, TBZ-based fungicide in some concentrations can have phytotoxic, cytotoxic and genotoxic effects in roots and meristematic cells of L. sativa.
