Novel Biological-Based Strategy for Synthesis of Green Nanochitosan and Copper-Chitosan Nanocomposites: Promising Antibacterial and Hematological Agents.

Two novel samples of nanoparticles based on chitosan were greenly synthesized using pomegranate peel extract. The extract served as a nanoparticle precursor, facilitating the precipitation of nanosized chitosan through the ionic gelation method. Additionally, by mixing the green chitosan nanoparticles with copper ions, a nanoscale composite of chitosan and copper oxide was also produced. Structural and morphological investigations (FTIR, XRD, SEM, EDX, and TGA analyses) were performed for greenly synthesized chitosan nanoparticles and their copper oxide composite to determine all the significant characteristics of those nanoparticles. In addition, both samples were tested using some biological investigations, such as antimicrobial activity and hematological effects. The antimicrobial tests yielded promising results for both the green chitosan nanoparticles and the CuO composite when tested using two bacterial strains and two fungal strains. Moreover, the results showed that using a similar concentration of both green-based chitosan samples resulted in a slightly larger inhibition zone and a lower minimum inhibition concentration (MIC) for the copper oxide chitosan composite compared to the chitosan nanoparticles for all microorganisms included in the test. The mean count of blood components (RBCs and platelets), clotting time, and cholesterol levels in three different blood samples were used to indicate the hematological activity of both greenly synthesized nanoparticles. The results verified a slight reduction in blood component count after the addition of green chitosan nanoparticles, but the chitosan copper oxide composite did not have a noticeable effect on the three blood samples. The chitosan nanoparticles were able to cause a considerable reduction in clotting time and cholesterol levels for all blood samples, thus acting as procoagulants. However, the mixing of CuO with chitosan nanoparticles prolonged the rate of clotting in blood samples from hypercholesteremic individuals, and thus, the mixture acted as an anticoagulant agent.

A rapid protocol for synthesis of chitosan nanoparticles with ideal physicochemical features

In this research, an innovative protocol is introduced to address crucial deficiencies in the formulation of chitosan nanoparticles (Cs NPs). While NPs show potential in drug delivery systems (DDSs), their application in the clinic is hindered by various drawbacks, such as toxicity, high material costs, and time-consuming and challenging preparation procedures. Within polymer-based NPs, Cs is a plentiful natural substance derived from the deacetylation of chitin, which can be sourced from the shells of shrimp or crab. Cs NPs can be formulated using the ionic gelation technique, which involves the use of a negatively charged agent, such as tripolyphosphate (TPP), as a crosslinking agent. Even though Cs is a cost-effective and biocompatible material, the formulation of Cs NPs with the correct size and surface electrical charge (zeta potential) presents a persistent challenge. In this study, various techniques were employed to analyze the prepared Cs NPs. The size and surface charge of the NPs were evaluated using dynamic light scattering (DLS). Morphological analysis was conducted using field emission-scanning electron microscopy (FE-SEM). The chemical composition and formation of Cs NPs were investigated using Fourier transform infrared (FTIR). The stability analysis was confirmed through X-ray diffraction (XRD) analysis. Lastly, the biocompatibility of the NPs was assessed through cell cytotoxicity evaluation using the MTT assay. Moreover, here, 11 formulations with different parameters such as reaction pH, Cs:TPP ratio, type of Cs/TPP, and ultrasonication procedure were prepared. Formulation 11 was chosen as the optimized formulation based on its high stability of more than three months, biocompatibility, nanosize of 75.6 ± 18.24 nm, and zeta potential of +26.7 mV. To conclude, the method described here is easy and reproducible and can be used for facile preparation of Cs NPs with desirable physicochemical characteristics and engineering ideal platforms for drug delivery purposes.

Green Synthesized Chitosan Nanoparticles for Controlling Multidrug-Resistant mecA- and blaZ-Positive Staphylococcus aureus and aadA1-Positive Escherichia coli.

Antimicrobial resistance has recently been considered an emerging catastrophe globally. The public health and environmental threats were aggravated by the injudicious use of antibiotics in animal farming, aquaculture, and croup fields, etc. Consequently, failure of antibiotic therapies is common because of the emergence of multidrug-resistant (MDR) bacteria in the environment. Thus, the reduction in antibiotic spillage in the environment could be an important step for overcoming this situation. Bear in mind, this research was focused on the green synthesis of chitosan nanoparticles (ChiNPs) using Citrus lemon (Assam lemon) extract as a cross-linker and application in controlling MDR bacteria to reduce the antibiotic spillage in that sector. For evaluating antibacterial activity, Staphylococcus aureus and Escherichia coli were isolated from environmental specimens, and their multidrug-resistant pattern were identified both phenotypically by disk diffusion and genotypically by detecting methicillin- (mecA), penicillin- (blaZ), and streptomycin (aadA1)-resistance encoding genes. The inhibitory zone's diameter was employed as a parameter for determining the antibacterial effect against MDR bacteria revealing 30 ± 0.4 mm, 34 ± 0.2 mm, and 36 ± 0.8 mm zones of inhibition against methicillin- (mecA) and penicillin (blaZ)-resistant S. aureus, and streptomycin (aadA1)-resistant E. coli, respectively. The minimum inhibitory concentration at 0.31 mg/mL and minimum bactericidal concentration at 0.62 mg/mL of yielded ChiNPs were used as the broad-spectrum application against MDR bacteria. Finally, the biocompatibility of ChiNPs was confirmed by showing a negligible decrease in BHK-21 cell viability at doses less than 2 MIC, suggesting their potential for future application in antibiotic-free farming practices.

Green synthesis of chitosan nanoparticles using tea extract and its antimicrobial activity against economically important phytopathogens of rice

The aim of the present work is to biosynthesize Chitosan nanoparticles (CTNp) using tea (Camellia sinensis) extract, with potent antimicrobial properties towards phytopathogens of rice. Preliminary chemical analysis of the extract showed that they contain carbohydrate as major compound and uronic acid indicating the nature of acidic polysaccharide. The structure of the isolated polysaccharide was analyzed through FTIR and 1H NMR. The CTNp was prepared by the addition of isolated tea polysaccharides to chitosan solution. The structure and size of the CTNp was determined through FTIR and DLS analyses. The surface morphology and size of the CTNp was analysed by SEM and HRTEM. The crystalinity nature of the synthesized nanoparticle was identified by XRD analysis. The CTNp exhibited the antimicrobial properties against the most devastating pathogens of rice viz., Pyricularia grisea, Xanthomonas oryzae under in vitro condition. CTNp also suppressed the blast and blight disease of rice under the detached leaf assay. These results suggest that the biosynthesized CTNp can be used to control the most devastating pathogens of rice.

Resveratrol Nanoformulation Inhibits Invasive Breast Cancer Cell Growth through Autophagy Induction: An In Vitro Study.

The aim of this study was to synthesize chitosan nanoparticles (Cs NPs) for resveratrol (RSV) delivery and assess their effectiveness in inducing autophagy in MDA-MB 231 cells. In this experimental study, Pure and RSV-loaded Cs NPs (RSV. Cs NPs) were prepared via the ionic gelation method, and their physicochemical properties were characterized using standard techniques, and RSV release was measured in vitro. MDA-MB 231 cells were incubated with RSV, Cs NPs, and RSV. Cs NPs and Half-maximal inhibitory concentration (IC50) values were calculated following the MTT test. Cell viability was assessed by lactate dehydrogenase (LDH) assay, and autophagy was evaluated using the real-time polymerase chain reaction (PCR). NP formation was confirmed with the analysis of FTIR spectra. Pure and RSV. Cs NPs had 36.7 and 94.07 nm sizes with 18.3 and 27 mV zeta potentials, respectively. Above 60% of RSV entrapped within NPs was released in an initial burst manner followed by a gradual release till 72 hours. Cs and RSV. Cs NPs restrained cell proliferation at lower concentrations. RSV. Cs NPs showed the highest anticancer effect and stimulated autophagy, indicated by increased Beclin-1 ATG5, ATG7, LC3A, and P62 expression. RSV. Cs NPs show promising effects in inhibiting invasive breast cancer (BC) cells in vitro by inducing autophagy.

Antifungal Activity of Chitosan Polymeric Nanoparticles and Correlation with Their pH Against Mucor circinelloides Causing Mucormycosis, Along with Penicillium notatum and Aspergillus Species

Mucormycosis is uncommon, yet it is more prevalent among individuals with underlying health conditions and those who are immunocompromised. Chitosan is studied because of its appealing properties and diverse applications. The purpose of this work is to synthesize chitosan nanoparticles (CSNPs) by ionic gelation method at various pH levels and test them against Mucor and other filamentous fungus. Field Emission Scanning Electron Microscope, Zeta sizer, Zeta potential, and Fourier Transformed Infrared Spectroscopy were used to characterize CSNPs. Hydrodynamic size increased considerably with increasing pH. Our CSNPs were tested against fungal isolates of Aspergillus Flavus RCMB 02783, Aspergillus Fumigatus RCMB 02564, and Aspergillus Niger RCMB 02588, Penicillium Notatum (NCPF 2881) and (M. circinelloides CNRMA 03.894) causing mucromycosis. Antifungal activity was investigated using Minimum inhibitory concentration (MIC), Minimum Fungicidal concentration (MFC), Disc diffusion assay, and Antifungal inhibitory percentages methods. The best antifungal efficacy results were obtained through CSNPs prepared at pH = 4.4 at very low concentration for MIC (1.03 or 2.75 μg/mL) with 100% M. circinelloides inhibition followed by pH = 4.6 with MIC (73 or 208 μg/mL) and 93% M. cirecinelloides inhibition %. Future usage of these materials in masks or wound dressing to avoid fungal infections, including mucormycosis following COVID-19, penicillium, and aspergillosis toxicity and infections.Graphical

MiR126-targeted-nanoparticles combined with PI3K/AKT inhibitor as a new strategy to overcome melanoma resistance

Metastatic melanoma poses significant challenges as a highly lethal disease. Despite the success of molecular targeting using BRAFV600E inhibitors (BRAFis) and immunotherapy, the emergence of early recurrence remains an issue and there is the need for novel therapeutic approaches. This study aimed at creating a targeted delivery system for the oncosuppressor microRNA 126 (miR126) and testing its effectiveness in combination with a phosphatidylinositol 3-kinase (PI3K)/ protein kinase B (AKT) inhibitor for treating metastatic melanoma resistant to BRAFis. To achieve this, we synthesized chitosan nanoparticles containing a chemically modified miR126 sequence. These nanoparticles were further functionalized with an antibody specific to the chondroitin sulfate proteoglycan 4 (CSPG4) melanoma marker. After evaluation invitro, the efficacy of this treatment was evaluated through an invivo experiment using mice bearing resistant human melanoma. The co-administration of miR126 and the PI3K/AKT inhibitor in these experiments significantly reduced tumor growth and inhibited the formation of liver and lung metastases. These results provide evidence for a strategy to target an oncosuppressive nucleic acid sequence to tumor cells while simultaneously protecting it from plasma degradation. The system described in this study exhibits encouraging potential for the effective treatment of therapy-resistant metastatic melanoma while also presenting a prospective approach for other forms of cancer.

Green synthesis and antibacterial assessment of chitosan/silver nanocomposite conjugated with tobramycin against antibiotic resistant Pseudomonas aeruginosa

In this study, we employed aqueous ginger extract as an eco-friendly approach for the synthesis of chitosan nanoparticles (CsNPs), silver nanoparticles (AgNPs), and chitosan/silver nanocomposites (Cs/AgNCs). Using spectroscopic and microscopic techniques, we thoroughly characterized the physicochemical properties of these nanomaterials, confirming their desirable attributes. The AgNPs displayed a spherical morphology with an average diameter of 19 nm, while the Cs/AgNCs exhibited a size of approximately 28 nm. To assess their antibacterial efficacy, we investigated the activity of both AgNPs and Cs/AgNCs against Gram-negative pathogenic bacteria. Remarkably, these nanomaterials demonstrated significant antibacterial properties. To further enhance their antibacterial capabilities, we conjugated the nanocomposite with tobramycin using Sulfo NHS and EDC, resulting in a four-fold increase in antibacterial activity compared to tobramycin alone. Moreover, we explored the ability of AgNPs, CsNPs, Cs/AgNCs, and Cs/AgNCs/tobramycin (Cs/AgNCs/tbn) to degrade DNA and generate reactive oxygen species (ROS) in the presence of Pseudomonas aeruginosa. Encouraging results were obtained, highlighting their potential applications in antimicrobial strategies. Furthermore, we conducted a comprehensive toxicity assessment of AgNPs, CsNPs, Cs/AgNCs, and Cs/AgNCs/tob using zebrafish embryos. The findings revealed low to moderate levels of toxicity for these nanomaterials, with Cs/AgNCs/tbn demonstrating no observable toxic effects. Our study demonstrates the immense potential of utilizing aqueous ginger extract as a green synthesis method to produce AgNPs and Cs/AgNCs with robust antibacterial properties and minimal toxicity. These findings pave the way for the development of innovative nanomaterials for various biomedical applications.

Chitosan Nanoparticles Loaded with Capparis cartilaginea Decne Extract: Insights into Characterization and Antigenotoxicity In Vivo.

Plant-based foods may enhance the prevention of cancer. The present investigation aimed to assess the antigenotoxic effects of chitosan nanoparticles (CNPs) when loaded with the ethanol extract of C. cartilaginea (CNPs/Cc). Synthesis of CNPs and CNPs/Cc and their characterization were carried out using TEM, EDS, DSC, and Zeta potential. For in vivo experiments, animal groups were treated in the following groups: negative control, ethyl methanesulfonate (EMS) (240 mg/kg), CNPs (350 mg/kg), high and low doses of CNPs/Cc, CNPs plus EMS, high dose of CNPs/Cc plus EMS, and low dose of CNPs/Cc plus EMS. Bone marrow chromosomal aberrations and sperm shape abnormalities were examined. TEM results showed that CNPs and CNPs/Cc are spherical particles. CNPs' physical stability was observed to be lower than that of CNPs/Cc due to the presence of more positive charges on CNPs/Cc. EMS significantly enhanced chromosomal abnormalities and sperm shape abnormalities. CNPs showed powerful antigenotoxic properties. For the first time, it could be concluded that loading chitosan nanoparticles with C. cartilaginea extract significantly promotes its protective properties.

Ameliorated antimicrobial, antioxidant, and anticancer properties by Plectranthus vettiveroides root extract-mediated green synthesis of chitosan nanoparticles

Abstract In this investigation, using the biogenic approach, Plectranthus vettiveroides root extract was used to synthesize chitosan nanoparticles (P. vettiveroides CNPs). The produced nanoparticles (NPs) were characterized using UV-visible (UV/vis) absorption spectroscopy, Fourier transform infrared (FTIR) spectroscopy, and scanning electron microscopy (SEM). The typical absorption peaks in the UV/vis spectra were located around 253 nm. Functional groups were identified in P. vettiveroides CNPs by FTIR. As per SEM analysis, the NPs generated exhibited a spherical shape with an average diameter of 78.01 nm. In addition, the synthesized P. vettiveroides CNPs were examined for antioxidant and antibacterial properties and anticancer activities. They show a strong antioxidant activity compared to butylated hydroxytoluene as a standard antioxidant. P. vettiveroides root extract CNPs demonstrated the most significant zone of inhibition against Klebsiella pneumoniae (22 mm), followed by Escherichia coli (21 mm), Bacillus cereus (19 mm), and Staphylococcus aureus (17 mm). In addition, using MTT assay, anticancer efficacy against KB (oral cancer) cells was studied. The cytotoxic reaction was observed in a dosage-dependent manner. P. vettiveroides CNPs show bioefficacy because of their size and the existence of bioactive compounds, which can enhance antibacterial and anticancer activities by lysing bacterial and cancer cell walls.

Tubermycin B coated on Galactosylated Chitosan Nanoparticles synthesized by eco-friendly method ameliorates intracellular free radical production and reduces oxidative stress, reducing phosphorylation of IKKα/β/pIкBα/NF-кB pathway.

The objective of the current study was to investigate the cytotoxic potentials of Galactosylated Chitosan Nanoparticles. Specifically, the study aimed to develop Tubermycin B coated on Galactosylated Chitosan Nanoparticles using a new green method that replaces sodium borohydride in the reduction process. The study synthesized Tubermycin B coated on Galactosylated Chitosan Nanoparticles through a new green method. The cytotoxicity of these nanoparticles was evaluated in a mice intestinal tract model that had been induced with chlorpyrifos, which causes oxidative stress-related enterotoxicity. Multiple activities, including the apoptosis of intestinal macrophages and the activation of Ikappa α/β kinase (IKKα/β), were examined as indicators of the nanoparticles' efficacy. The stability of the synthesized Chitosan Nanoparticles was also assessed. Additionally, the encapsulation efficiency of Boscia angustifalia and Boscia senegalensis extracts within the nanoparticles was determined. The results of the study showed that Tubermycin B coated on Galactosylated Chitosan Nanoparticles effectively alleviated the oxidative stress-related enterotoxicity in the mice intestinal tract induced by chlorpyrifos. The nanoparticles prevented the apoptosis of intestinal macrophages and inhibited the activation of IKKα/β. The synthesized chitosan nanoparticles exhibited high stability. The encapsulation efficiency of Boscia angustifalia extract was recorded as 46.58%, whereas for Boscia senegalensis extract, it was 9.77%. The nanoparticles showed no cytotoxicity at all tested concentrations and demonstrated a medium-level anticancer effect. Based on the findings, it can be concluded that Tubermycin B coated on Galactosylated Chitosan Nanoparticles has the potential to alleviate oxidative stress-related enterotoxicity in the mice intestinal tract. The nanoparticles showed high stability and exhibited a medium-level anticancer effect. Furthermore, the study demonstrated that Boscia angustifalia extract exhibited higher anti-hepatitis C virus antibodies (anti-HCV) activity compared to Boscia senegalensis extract in an in-vitro system. Therefore, Boscia angustifalia could be considered a promising candidate for the development of an anti-HCV drug for future in-vivo studies.

The potential of suspended chitosan nanoparticles as a surgical irrigation fluid.

In this study, we sought to synthesize chitosan nanoparticles (CS-NPs) and characterize their morphology, efficacy in inhibiting bacterial attachment, and efficacy in eradicating bacteria established on implantable hardware. CS-NPs possess desirable properties, including antibacterial properties in biofilm-mediated infections. CS-NPs were produced using ionic gelation and characterized via scanning electron microscope imaging. Staphylococcus aureus was incubated with CS-NPs at various concentrations and compared to a 1% povidone-iodine with 1% H2 O2 control in 24-well plates. Stainless steel bone screws were placed in six-well plates and inoculated with S. aureus. After 24 h, the screws were transferred to one of three solutions (saline, 40 mg/mL CS-NP, or 1% povidone-iodine with 1% H2 O2 ). Four screws from each group were vortexed in saline and plated. The remaining screw from each group was prepped and imaged to map the location of persistent bacteria. Synthesized CS-NPs had a mean diameter of 0.39 ± 0.13 μm and circularity of 0.87 ± 0.05. The percent inhibition of bacterial attachment was 73% at 20 mg/mL, 73% at 30 mg/mL, 75% at 40 mg/mL, 79% at 50 mg/mL, and 78% at 60 mg/mL. When compared to saline, the 40 mg/mL CS-NP solution reduced bacteria on the screws by 76%. No bacteria were retrieved from the 1% povidone-iodine with 1% H2 O2 group. This study demonstrated that CS-NP solution effectively inhibited S. aureus bacterial attachment and was more effective than saline in eradicating bacteria from orthopedic hardware, suggesting that CS-NPs have the potential for prevention and treatment of musculoskeletal infections as a component of an intraoperative surgical irrigation solution.

Biological effect of natural chitosan nanoparticles with transformed roots extract of Leonotis nepetifolia (L.) R.Br. in an in vitro model

Nanotechnology is a rapidly growing field of science that has a considerable impact on many branches of medicine. Nanoparticles are developing at a rapid pace due to their effect which is achieved by combining them with drugs as well as their increased importance in the drug delivery system. Synthesis of chitosan nanoparticles by ionic gelation is becoming increasingly important due to its low cost and environmentally friendly approach. This preliminary study was the first one to investigate the cytotoxic effect of chitosan nanoparticles with combination of Leonotis nepetifolia (L.) R.Br. transformed root extract against cervical cancer (HeLa), breast cancer (MCF-7), glioma cancer (MO59J) cell lines and normal human fibroblast (Hs68) cell line. Besides, a preliminary analysis of apoptosis induced by caspase 3/7, microscopic observations of morphology changes and apoptosis-related genes expression in cancer cells have been showed. According to MTT results, the synthesized chitosan nanoparticles in combination with Leonotis nepetifolia normal and transformed root extracts showed a stronger cytotoxic effect on all tested cancer lines (HeLa, MCF-7 and MO59J), compared with the nanoparticles and the extracts alone, with IC50 = 0.608, 0.306, 0.112 mg/mL, respectively. In contrast, no cytotoxic effect was shown against the human fibroblast (Hs68) line in the tested concentration range. Preliminary induction of apoptosis showed changes in cell morphology, genes expression and caspase 3/7 activation after treatment with chitosan nanoparticles in combination with normal and transformed from Leonotis nepetifolia root extracts. These discoveries have positive implications for further studies on development of nanoparticles in anti-cancer therapy. The key objective of this work is to test the anti-cancer potential of L. nepetifolia transformed root extract obtained by agrotransformation in combination with natural chitosan nanoparticles as a component capable of enhancing the effect of the extract against various in vitro models.

AKTIVITAS ANTIJAMUR PELAPUK KAYU NANOPARTIKEL KITOSAN-EKSTRAK N-HEKSAN DAGING BUAH KAYU GADIS (Cinnamomum parthenoxylon)

This study aims to synthesize chitosan nanoparticles - n-hexane extract of Cinnamomum parthenoxylon (CP) fruit using the ionic gelation method, to characterize them, and to determine their antifungal activity against the wood-rotting fungi Trametes versicolor and Fomitopsis palustris. Chitosan-CP extract nanoparticles were formulated with chitosan:Na-TPP:CP extract ratio of 1:1:0.5. Particle size and polydispersity index were determined using a particle size analyzer (PSA), and particles morphology was observed using a scanning electron microscope (SEM), and antifungal activity was measured using agar media. The results revealed that the average size of chitosan nanoparticles (base) was 276.9 nm, while the average size of chitosan-CP extract nanoparticles was 807.6 nm. The polydispersity index (PI) of both nanoparticles was greater than 0.7. Observations of the nanoparticles' morphology revealed that aggregation/agglomeration formed in the resulting particles. Both nanoparticles chitosan (base) and chitosan-CP extract inhibited the growth of white rot fungi (T. versicolor) more than brown rot fungi (F. palustris), as measured by the inhibitory zone.

Preparation, characterization and protective effect of chitosan - Tripolyphosphate encapsulated dihydromyricetin nanoparticles on acute kidney injury caused by cisplatin

Dihydromyricetin (DMY) is a natural dihydroflavonol compound known for its diverse pharmacological benefits. However, its limited stability and bioavailability posed significant challenges for further applications. To address these issues, in this study, an ion crosslinking method was utilized to prepare chitosan nanoparticles that were loaded with DMY. The synthesized chitosan nanoparticles (CS-DMY-NPs) were spherical in shape with particle size and ζ potential of 198.7 nm and 45.05 mV, respectively. Furthermore, in vitro release experiments demonstrated that CS-DMY-NPs had sustained release and protective effects in simulated gastric and intestinal fluids. CS-DMY-NPs exhibited better antioxidant activity by ABTS and DPPH radical scavenging activity than free DMY. In vivo study showed that CS-DMY-NPs alleviated cisplatin-induced kidney damage by inhibiting oxidative stress and proinflammatory cytokines, and had better activity compared to DMY (free). Immunofluorescence data showed that CS-DMY-NPs activated the Nrf2 signaling pathways in a dose-dependent manner to combat cisplatin-induced kidney damage. Our results demonstrate that CS-TPP has good compatibility with DMY, and CS-DMY-NPs exhibited better protective effects against cisplatin-induced acute kidney injury (AKI) than free DMY.

Investigation of novel HCV therapies: Boscia angustifalia &Boscia senegalensis extracts loaded on galactosylated chitosan nanoparticles synthesized by eco-friendly method for HCV treatment

Hepatitis C virus (HCV) is a major causative agent of chronic liver diseases including chronic hepatitis, liver cirrhosis, and hepatocellular carcinoma worldwide. Treatment of HCV has evolved from early interferon monotherapy to the current all-oral regimens using direct-acting antivirals. However, antiviral resistance has become a critical issue in the treatment of chronic hepatitis C after receiving therapy with direct-acting antivirals (DAA) with a 0.5 % chance of the hepatitis C virus recurrence, similar to other chronic viral infections. So, retreatment options following treatment failure have become crucial issues. Hence, this study aims to investigate a new promising therapy for HCV. In the field of nanomedicine, chitosan nanoparticles are well-known delivery systems that are frequently used as polymeric carriers. Galactosylated chitosan nanoparticles have been widely applied in HCV treatment. In this study, we have modified galactosylation by an eco-friendly method using l-ascorbic instead of hazardous reagents and we have loaded it with newly tested two Boscia extracts each in three different concentrations. The synthesized chitosan nanoparticles showed two dispersion peaks, at 196 ± 29 nm and 1.33 ± 0.36 μm, with a zeta potential of +3.3 ± 0.4mV with high stability in a range of 40.7 mV. The percentage of encapsulation of Boscia angustifalia extract was found to be 46.58 ± 1.33 % and for Boscia senegalensis extract was 9.77 ± 0.33 %. The release of Boscia angustifalia extract from the nanoparticles was about 40 % in acidic media after 180 min and about 60 % in normal pH. However, the release of Boscia senegalensis extract was 20 % in acidic media and 56 % in normal media after 24 h. Testing of these two newly developed composites against HCV was carried out using an in vitro system for the production of hepatitis C virus (HCV) which was established by infection of human hepatoma cells. Evidence for persistent virus production was monitored by the ELISA technique using an anti-HCV-specific antibody. Results obtained showed that all samples had an anti-HCV activity that increased by increasing concentration, and Boscia angustifalia had remarkable anti-HCV activity compared to Boscia senegalensis.

Synthesis of chitosan nanoparticles from Tasar silkworm, Antheraea mylitta pupae and its antioxidant efficacy and antibacterial activity against fish pathogens

Aim: To synthesize and characterize chitosan nanoparticles (CNPs) from the pupae of tasar silkworm, Antheraea mylitta. The antioxidant and antibacterial activities of CNPs against different strains of selected bacterial fish pathogens were also evaluated. Methodology: CNPs were prepared using sodium tripolyphosphate anions. Characterization of nanoparticles was done by FTIR, Zeta potential analysis and Particle size distribution analysis. The antibacterial activity of chitosan and CNPs against Aeromonas hydrophila (ATCC-7966), Escherichia coli (ATCC-25922), and Staphylococcus aureus (ATCC-25923) was evaluated by disc diffusion method. Results: CNPs inhibited the growth of selected bacteria and the maximum zone of inhibition (14 mm) was observed against E. coli and A. hydrophila. Chitosan and CNPs had free radical scavenging effects on hydrogen peroxide (H2O2) and 2,2-diphenyl-1-picrylhydrazyl (DPPH). Interpretation: CNPs obtained from silkworm pupae have potential antibacterial as well as antioxidant activities and can be incorporated in aquafeed to prevent fish diseases. A Simple Method for Synthesis of Chitosan Nanoparticles with Ionic Gelation and Homogenization.

Chitosan nanoparticles (CNPs) are known to have great utility in many fields (pharmaceutical, agricultural, food industry, wastewater treatment, etc.). In this study we aimed to synthesize sub-100 nm CNPs as a precursor of new biopolymer-based virus surrogates for water applications. We present a simple yet efficient synthesis procedure for obtaining high yield, monodisperse CNPs with size 68-77 nm. The CNPs were synthesized by ionic gelation using low molecular weight chitosan (deacetylation 75-85%) and tripolyphosphate as crosslinker, under rigorous homogenization to decrease size and increase uniformity, and purified by passing through 0.1 μm polyethersulfone syringe filters. The CNPs were characterized using dynamic light scattering, tunable resistive pulse sensing, and scanning electron microscopy. We demonstrate reproducibility of this method at two separate facilities. The effects of pH, ionic strength and three different purification methods on the size and polydispersity of CNP formation were examined. Larger CNPs (95-219) were produced under ionic strength and pH controls, and when purified using ultracentrifugation or size exclusion chromatography. Smaller CNPs (68-77 nm) were formulated using homogenization and filtration, and could readily interact with negatively charge proteins and DNA, making them an ideal precursor for the development of DNA-labelled, protein-coated virus surrogates for environmental water applications.

In Vivo Wound-Healing Effect of Chemical and Green Synthesized Chitosan Nanoparticles Using Lawsonia inermis Ethanolic Extract.

Wounds can be a result of surgery, an accident, or other factors. There is still a challenge to find effective topical wound-healing agents. This study aims to investigate the wound-healing activity of chemical and green synthesized chitosan nanoparticles (Ch-NPs) using Lawsonia inermis leaves extract. The nanoparticles were morphologically and chemically characterized using X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), and high-resolution transmission electron microscopy (HRTEM). Forty-five adult female albino rats were randomly divided into three groups. The cutaneous surgical wounds were topically treated with 0.9% normal saline (control group), green Ch-NPs (second group), and chemical Ch-NPs gels (third group), respectively. The clinical picture of wounds and histopathological changes were assessed on the 3rd, 7th, 14th, and 21st days post-treatment. X-ray diffraction analysis revealed great crystallinity and purity of nanoparticles. The studied nanoparticles increased the wound contraction percent (WC%), reduced healing time and wound surface area (WSA), and these results were backed up by histological findings that indicated improved epithelialization, dermal differentiation, collagen deposition, and angiogenesis in treated rats compared with control rats (p < 0.05). We concluded that the wound-healing effects of the studied nanoparticles are encouraging, and further studies for complete assessment are still needed.

Green Synthesis of Chitosan Nano Particles at Different Temperature for Bio-Medical Applications

Chitosan is a semi biopolymer obtained from natural biopolymer Chitin by depolymerisation and de-acetylation. It is bio-degradable, non-toxic and an ideal candidate for pharmaceutical uses. But major bottle-neck of biomedical application of chitosan is that it is insoluble in water therefore we have reported here preparation of ionically cross linked chitosan nano material using tartarate ion as ionic cross linker by greener hydrothermal technique at different temperature and pressure to regulate particle sizes. To the best of our knowledge this is the first time we are reporting hydrothermal synthesis of chitosan nanoparticles at high pressure to regulate particle sizes. Hydro- dynamic particle sizes have been determined by DLS measurement. We have also studied fluorescence behaviour of these chitosan nanoparticles and studied their anti-bacterial activities by Alamar assay on E Coli and S Aureus.
