This review deals with new optically active phosphine ligands which appeared in the literature during past eight years. Emphasis is laid on the utility of these ligands in transition metal-catalyzed asymmetric reactions. New methods for the synthesis of chiral phosphine ligands are also described.

Synthesis of chiral phosphine ligands with aromatic backbones and their applications in asymmetric catalysis

A general strategy for the synthesis of new chiral phosphine ligands has been discovered. A common feature of these ligands is that they contain rigid aromatic backbones which can be used to restrict conformational flexibility. The bite angle of P-M-P can be changed systematically depending on the aromatic backbone. Many asymmetric reactions can potentially be catalyzed by transition metal complexes of these ligands. Moderate selectivity has been realized in the reaction between 1,3-diphenyl-2-propenyl acetate and dimethyl malonate using palladium complexes of these ligands.

The combinatorial synthesis of chiral phosphine ligands

Chemistry is reported that allows for the synthesis and screening of phosphine ligands by standard combinatorial technology. To demonstrate the method a 63 member library of phosphine containing peptides was synthesized. Rhodium was complexed to the phosphine ligands while they were attached to the synthesis support. Each member of the library was screened for its ability to catalyze the asymmetric hydrogenation of enamide ( 3). Some correlation between specific substitutions in the primary sequence of the peptide and the highest enantiomeric excesses was observed.
