Biochemical analysis of Hyalomma dromedarii salivary glands and gut tissues using SR-FTIR micro-spectroscopy

Ticks are obligatory voracious blood feeders infesting diverse vertebrate hosts, that have a crucial role in the transmission of diverse pathogens that threaten human and animal health. The continuous emergence of tick-borne diseases due to combined worldwide climatic changes, human activities, and acaricide-resistant tick strains, necessitates the development of novel ameliorative tick control strategies such as vaccines. The synchrotron-based Fourier transform infrared micro-spectroscopy (SR-FTIR) is a bioanalytical microprobe capable of exploring the molecular chemistry within microstructures at a cellular or subcellular level and is considered as a nondestructive analytical approach for biological specimens. In this study, SR-FTIR analysis was able to explore a qualitative and semi-quantitative biochemical composition of gut and salivary glands of Hyalomma dromedarii (H. dromedarii) tick detecting differences in the biochemical composition of both tissues. A notable observation regarding Amide I secondary structure protein profile was the higher ratio of aggregated strands in salivary gland and beta turns in gut tissues. Regarding the lipid profile, there was a higher intensity of lipid regions in gut tissue when compared to salivary glands. This detailed information on the biochemical compositions of tick tissues could assist in selecting vaccine and/or control candidates. Altogether, these findings confirmed SR-FTIR spectroscopy as a tool for detecting differences in the biochemical composition of H. dromedarii salivary glands and gut tissues. This approach could potentially be extended to the analysis of other ticks that are vectors of important diseases such as babesiosis and theileriosis.

Multifunctionalized Carbon Dots as an Active Nanocarrier for Drug Delivery to the Glioblastoma Cell Line.

Nanoparticle-based nanocarriers represent a viable alternative to conventional direct administration in cancer cells. This advanced approach employs the use of nanotechnology to transport therapeutic agents directly to cancer cells, thereby reducing the risk of damage to healthy cells and enhancing the efficacy of treatment. By approving nanoparticle-based nanocarriers, the potential for targeted, effective treatment is greatly increased. The so-called carbon-based nanoparticles, or carbon dots, have been hydrothermally prepared and initiated by a polymerization process. We synthesized and characterized nanoparticles of 2-acrylamido-2-methylpropanesulfonic acid, which showed biocompatibility with glioblastoma cells, and further, we tested them as a carrier for the drug riluzole. The obtained nanoparticles have been extensively characterized by techniques to obtain the exact composition of their surface by using Fourier transform infrared (FTIR), X-ray photoelectron spectroscopy (XPS), and nuclear magnetic resonance (NMR) spectroscopy, as well as cryo-transmission electron microscopy. We found that the surface of the synthesized nanoparticles (NPs) is covered mainly by sulfonated, carboxylic, and substituted amide groups. These functional groups make them suitable as carriers for drug delivery in cancer cells. Specifically, we have successfully utilized the NPs as a delivery system for the drug riluzole, which has shown efficacy in treating glioblastoma cancer cells. The effect of nanoparticles as carriers for the riluzole system on glioblastoma cells was studied using live-cell synchrotron-based FTIR microspectroscopy to monitor in situ biochemical changes. After applying nanoparticles as nanocarriers, we have observed changes in all biomacromolecules, including the nucleic acids and protein conformation. These findings provide a strong foundation for further exploration into the development of targeted treatments for glioblastoma.

Biomolecular profiles of Arctic sea-ice diatoms highlight the role of under-ice light in cellular energy allocation.

Arctic sea-ice diatoms fuel polar marine food webs as they emerge from winter darkness into spring. Through their photosynthetic activity they manufacture the nutrients and energy that underpin secondary production. Sea-ice diatom abundance and biomolecular composition vary in space and time. With climate change causing short-term extremes and long-term shifts in environmental conditions, understanding how and in what way diatoms adjust biomolecular stores with environmental perturbation is important to gain insight into future ecosystem energy production and nutrient transfer. Using synchrotron-based Fourier transform infrared microspectroscopy, we examined the biomolecular composition of five dominant sea-ice diatom taxa from landfast ice communities covering a range of under-ice light conditions during spring, in Svalbard, Norway. In all five taxa, we saw a doubling of lipid and fatty acid content when light transmitted to the ice-water interface was >5% but <15% (85%-95% attenuation through snow and ice). We determined a threshold around 15% light transmittance after which biomolecular synthesis plateaued, likely because of photoinhibitory effects, except for Navicula spp., which continued to accumulate lipids. Increasing under-ice light availability led to increased energy allocation towards carbohydrates, but this was secondary to lipid synthesis, whereas protein content remained stable. It is predicted that under-ice light availability will change in the Arctic, increasing because of sea-ice thinning and potentially decreasing with higher snowfall. Our findings show that the nutritional content of sea-ice diatoms is taxon-specific and linked to these changes, highlighting potential implications for future energy and nutrient supply for the polar marine food web.

Synchrotron-based FTIR microspectroscopy reveals DNA methylation profile in DNA-HALO structure

Fourier transform infrared (FTIR) spectroscopy is a rapid, non-destructive and label-free technique for identifying subtle changes in all bio-macromolecules, and has been used as a method of choice for studying DNA conformation, secondary DNA structure transition and DNA damage. In addition, the specific level of chromatin complexity is introduced via epigenetic modifications forcing the technological upgrade in the analysis of such an intricacy. As the most studied epigenetic mechanism, DNA methylation is a major regulator of transcriptional activity, involved in the suppression of a broad spectrum of genes and its deregulation is involved in all non-communicable diseases. The present study was designed to explore the use of synchrotron-based FTIR analysis to monitor the subtle changes in molecule bases regarding the DNA methylation status of cytosine in the whole genome. In order to reveal the conformation-related best sample for FTIR-based DNA methylation analysis in situ, we used methodology for nuclear HALO preparations and slightly modified it to isolated DNA in HALO formations. Nuclear DNA-HALOs represent samples with preserved higher-order chromatin structure liberated of any protein residues that are closer to native DNA conformation than genomic DNA (gDNA) isolated by the standard batch procedure. Using FTIR spectroscopy we analyzed the DNA methylation profile of isolated gDNA and compared it with the DNA-HALOs. This study demonstrated the potential of FTIR microspectroscopy to detect DNA methylation marks in analyzed DNA-HALO specimens more precisely in comparison with classical DNA extraction procedures that yield unstructured whole genomic DNA. In addition, we used different cell types to assess their global DNA methylation profile, as well as defined specific infrared peaks that can be used for screening DNA methylation.

Synchrotron-based infrared microspectroscopy of polymeric nanoparticles and skin: Unveiling molecular interactions to enhance permeation

The application of nanoparticles as permeation enhancers in skin drug delivery is a growing research field. However, the mechanisms of nanoparticles’ interaction with the skin structure are still unknown. Fucoidan/chitosan nanoparticles have demonstrated several physicochemical and biological advantages, among which is the enhancement of skin permeation. This study aims to elucidate permeation enhancement mechanisms using synchrotron-based Fourier Transform Infrared Microspectroscopy (SR-FTIRM) combined with multivariate analysis and in vitro skin permeation assay. Given the molecular weight influence on chitosan’s properties, the nanoparticles-skin interactions were evaluated with nanoparticles produced using low- and medium-molecular-weight chitosan. Chemical maps and spectral analysis revealed that fucoidan/chitosan nanoparticles induced changes in the lipids and protein regions. Inter-sample spectral differences were identified using principal component analysis. Low molecular weight fucoidan/ chitosan nanoparticles caused changes in the skin lipids’ lateral packing and structure at the stratum corneum layer towards a less ordered state and higher fluidity, and no evidence was found on proteins structure. The opposite was revealed for medium molecular weight fucoidan/chitosan nanoparticles, which induced changes in the secondary structure of keratin and altered lipid structure to an ordered and dense conformation. In vitro permeation assays with Franz diffusion cells correlate with the observed changes in the skin lipid and protein structure with enhanced skin permeation of a hydrophilic molecule incorporated within the fucoidan/chitosan nanoparticles. The findings of this study unveil molecular changes in the skin structure induced by the nanoparticles only possible with the application of the powerful and precise SR-FTIRM technique. This knowledge allows the design of nanoparticles towards an internalization pathway determining their fate within the skin structure.

Illuminating Host-Parasite Interaction at the Cellular and Subcellular Levels with Infrared Microspectroscopy.

Toxoplasma gondii (T. gondii) is an opportunistic protozoan that can cause brain infection and other serious health consequences in immuno-compromised individuals. This parasite has a remarkable ability to cross biological barriers and exploit the host cell microenvironment to support its own survival and growth. Recent advances in label-free spectroscopic imaging techniques have made it possible to study biological systems at a high spatial resolution. In this study, we used conventional Fourier-transform infrared (FTIR) microspectroscopy and synchrotron-based FTIR microspectroscopy to analyze the chemical changes that are associated with infection of human brain microvascular endothelial cells (hBMECs) by T. gondii (RH) tachyzoites. Both FTIR microspectroscopic methods showed utility in revealing the chemical alterations in the infected hBMECs. Using a ZnS hemisphere device, to increase the numerical aperture, and the synchrotron source to increase the brightness, we obtained spatially resolved spectra from within a single cell. The spectra extracted from the nucleus and cytosol containing the tachyzoites were clearly distinguished. RNA sequencing analysis of T. gondii-infected and uninfected hBMECs revealed significant changes in the expression of host cell genes and pathways in response to T. gondii infection. These FTIR spectroscopic and transcriptomic findings provide significant insight into the molecular changes that occur in hBMECs during T. gondii infection.

Live-Cell Synchrotron-Based FTIR Evaluation of Metabolic Compounds in Brain Glioblastoma Cell Lines after Riluzole Treatment.

Glioblastoma multiforme (GBM) is the most aggressive brain tumor, characterized by short median survival and an almost 100% tumor-related mortality. The standard of care treatment for newly diagnosed GBM includes surgical resection followed by concomitant radiochemotherapy. The prevention of disease progression fails due to the poor therapeutic effect caused by the great molecular heterogeneity of this tumor. Previously, we exploited synchrotron radiation-based soft X-ray tomography and hard X-ray fluorescence for elemental microimaging of the shock-frozen GBM cells. The present study focuses instead on the biochemical profiling of live GBM cells and provides new insight into tumor heterogenicity. We studied bio-macromolecular changes by exploring the live-cell synchrotron-based Fourier transform infrared (SR-FTIR) microspectroscopy in a set of three GBM cell lines, including the patient-derived glioblastoma cell line, before and after riluzole treatment, a medicament with potential anticancer properties. SR-FTIR microspectroscopy shows that GBM live cells of different origins recruit different organic compounds. The riluzole treatment of all GBM cell lines mainly affected carbohydrate metabolism and the DNA structure. Lipid structures and protein secondary conformation are affected as well by the riluzole treatment: cellular proteins assumed cross β-sheet conformation while parallel β-sheet conformation was less represented for all GBM cells. Moreover, we hope that a new live-cell approach for GBM simultaneous treatment and examination can be devised to target cancer cells more specifically, i.e., future therapies can develop more specific treatments according to the specific bio-macromolecular signature of each tumor type.

Assessing the Effect of Rubber (Hevea brasiliensis (Willd. ex A. Juss.) Muell. Arg.) Leaf Chemical Composition on Some Soil Properties of Differently Aged Rubber Tree Plantations

Leaf litter plays a major role in carbon and nutrient cycling, as well as in fueling food webs. The chemical composition of a leaf may directly and indirectly influence decomposition rates by influencing rates of biological reactions and by influencing the accumulation of soil organic carbon content, respectively. This study aimed to assess the impact of the chemical composition of rubber (Hevea brasiliensis (Willd. ex A. Juss.) Muell. Arg.) leaves on various soil properties of different ages of rubber (4–5, 11–12, and 22–23 year-old). Synchrotron-based Fourier transform infrared microspectroscopy (Sr-FTIR) was utilized for analyzing the chemical composition of plant leaves. The Sr-FTIR bands illustrated that the epidermis of rubber leaves from 4–5-year-old trees was found to contain a high quantity of polysaccharides while mesophyll from 22–23-year-old trees had a large number of polysaccharides. The change in soil properties in the older rubber plantation could be attributed to its chemical composition. The change in soil properties across all tree ages, i.e., increased litter and organic carbon content, was a relatively strong driver of soil biota evolution. The aliphatic of C-H in the leaves showed high correlation with soil organic carbon (SOC) and permanganate-oxidizable C (POXC) from 22–23 year-old trees. This study shows the differences in the organic chemical composition of leaves that are consequential to soil organic carbon.

Synchrotron-Based Infrared Microscopy Studies of the Radiosensitization Effects of Nanoparticles used in Radiotherapy

Radiotherapy (RT) is the most important non-surgical modality for cancer and the recent technological advances brought a substantial gain in the balance between the probability of tumor control and the risk of normal tissue complications. However, the clinical management of some radioresistant tumors (such as brain gliomas) continues being a challenge since tumor doses are constrained by the surrounding healthy tissue tolerances. Within this context, new modalities such as radiosensitization in the presence of high atomic number nanoparticles (NP) are extensively evaluated, with the main goal of enhancing the differential effect between tumors and healthy tissues. In the present work, Synchrotron based Fourier Transform Infrared Microspectroscopy (SR-FTIRM), a vibrational spectroscopic technique for the biochemical cell analysis on a microscopic scale, provides relevant information on the treatment-induced modifications of the main biomolecules (nucleic acids, proteins and lipids). A series of treatments, including various NP sized, typed and chemical natured (3 nm gadolinium (GdNP), 1.9 nm gold (AuNP) nanoparticles), cell line used (F98 murine and U87-MG human glioma cells) and radiotherapy configuration (megavoltage X-ray photon energy (MV) normaly used in clinics, with respect to kilovoltage photons (kV)), were assessed for their induced cell macromolecular damages (Martinez-Rovira et al. (2019) Analyst 144, 5511 and DOI: 10.1039/c9an01109a). Additionally, alongside X-ray photon RT, the possible cell damages caused by several charged particle beams (proton, helium, carbon and oxygen) were also considered. Briefly, the biochemical information provided by the highly brilliant infrared light source produced at ALBA Synchrotron leads to a clear advantage in spectral quality at a cellular level and allow better characterization of NP induced damage in cells.

Fourier-transform spectra of metal salts of phytic acid in the mid- to far-infrared spectral range

Phytic acid (myo-inositol 1,2,3,4,5,6-hexakisphosphate; IP6) is the storage substance for phosphorus and many metals in plant seeds. Based on Synchrotron-based Fourier-transform infrared (FT-IR) spectra in the mid-infrared range from 1300 to 700cm−1, nine synthesized metal-IP6 salts, with monovalent to trivalent coordination, were classified into four groups: trivalent metal-IP6 salts, second-group divalent metal-IP6 salts, d-block and p-block divalent metal-IP6 salts, and monovalent metal-IP6 salts. Synchrotron FT-IR analysis in the far-infrared range, from 650 to 100cm−1, contained clear, broad bands specific to each metal-IP6 salt, including monovalent and divalent metal-IP6 salts. These broad bands are likely derived from vibrations of bonds between the metal and the oxygen of IP6. These unique far-infrared spectra may provide useful information for identifying metal-IP6 salts in biochemical fine-image analyses of seed tissues using synchrotron-based FT-IR microspectroscopy.

Investigating the Molecular Structural Features of Hulless Barley (Hordeum vulgare L.) in Relation to Metabolic Characteristics Using Synchrotron-Based Fourier Transform Infrared Microspectroscopy

The synchrotron-based Fourier transform infrared microspectroscopy (SR-FTIRM) technique was used to quantify molecular structural features of the four hulless barley lines with altered carbohydrate traits [amylose, 1-40% of dry matter (DM); β-glucan, 5-10% of DM] in relation to rumen degradation kinetics, intestinal nutrient digestion, and predicted protein supply. Spectral features of β-glucan (both area and heights) in hulless barley lines showed a negative correlation with protein availability in the small intestine, including truly digested protein in the small intestine (DVE) (r = -0.76, P < 0.01; r = -0.84, P < 0.01) and total metabolizable protein (MP) (r = -0.71, P < 0.05; r = -0.84, P < 0.01). Variation in absorption intensities of total carbohydrate (CHO) was observed with negative effects on protein degradation, digestion, and potential protein supply (P < 0.05). Molecular structural features of CHO in hulless barley have negative effects on the supply of true protein to ruminants. The results clearly indicated the impact of the carbohydrate-protein structure and matrix.

In‐depth study of the protein molecular structures of different types of dried distillers grains with solubles and their relationship to digestive characteristics

Dried distillers grains with solubles (DDGS) have been extensively utilised in ruminant rations in western Canada and USA, and it is important to ensure their consistent quality. Traditional chemical methods do not consider the inherent structural changes of feed ingredients. Synchrotron-based Fourier transform infrared microspectroscopy (SFTIRM) and diffuse reflectance infrared Fourier transform spectroscopy (DRIFT) have been utilised to detect the changes in molecular structure of several feedstuffs (e.g. barley, flaxseed and alfalfa). However, similar structural information is lacking for DDGS. The objectives of this study were to identify differences in protein molecular structures between different grains (wheat, triticale and corn) and DDGS (wheat DDGS, triticale DDGS, corn DDGS and wheat and corn blend DDGS) using SFTIRM and DRIFT and to reveal the relationship between changes in protein molecular structure and the digestive characteristics of the protein in DDGS when fed to dairy cattle. The protein molecular structure studies showed significant decreases (P < 0.01) in the amide I to amide II ratio and the α-helix to β-sheet ratio between grains and their DDGS. Protein digestive characteristics were correlated with protein molecular structures in grains and DDGS, and prediction equations were established to estimate protein digestive characteristics of DDGS using protein molecular structure parameters. For the DVE/OEB-1994 model, one of the best prediction equations was for truly absorbed protein in the small intestine (DVE) = 296.17 - 38.98 × the amide I to amide II ratio (R(2) = 0.72). For the NRC-2001 system, one of the best prediction equations was for metabolisable protein (MP) = 300.96 - 43.32 × the amide I to amide II ratio (R(2) = 0.76). Protein molecular structure varies between different DDGS and their original grains, and this variation is associated with the digestive characteristics of the proteins in the DDGS and their original grains.

Evidence of lipid peroxidation and protein phosphorylation in cells upon oxidative stress photo-generated by fullerols

An oxidative stress (OS) state is characterized by the generation of Reactive Oxygen Species (ROS) in a biological system above its capacity to counterbalance them [1]. Exposure to OS induces the accumulation of intracellular ROS, which in turn causes cell damage in the form of protein, lipid, and/or DNA oxidations. Such conditions are believed to be linked to numerous diseases or simply to the ageing of tissues. However, the controlled generation of ROS via photosensitizing drugs or photosensitizers (PS) is now widely used to treat various tumors and other infections [2,3]. Here we present a method to track the chemical changes in a cell after exposure to oxidative stress. OS is induced via fullerols, a custom made water soluble derivative of fullerene (C 60), under visible light illumination. Synchrotron-based Fourier Transform InfraRed Microspectroscopy (S-FTIRM) was used to assess the chemical makeup of single cells after OS exposure. Consequently, a chemical fingerprint of oxidative stress was probed in this study through an increase in the bands linked with lipid peroxidation (carbonyl ester group at 1740 cm −1) and protein phosphorylation (asymmetric phosphate stretching at 1240 cm −1).

Characterization of the Microchemical Structure of Seed Endosperm within a Cellular Dimension among Six Barley Varieties with Distinct Degradation Kinetics, Using Ultraspatially Resolved Synchrotron-Based Infrared Microspectroscopy

Barley varieties have similar chemical composition but exhibit different rumen degradation kinetics and nutrient availability. These biological differences may be related to molecular, structural, and chemical makeup among the seed endosperm tissue. No detailed study was carried out. The objectives of this study were: (1) to use a molecular spectroscopy technique, synchrotron-based Fourier transform infrared microspectroscopy (SFTIRM), to determine the microchemical-structural features in seed endosperm tissue of six developed barley varieties; (2) to study the relationship among molecular-structural characteristics, degradation kinetics, and nutrient availability in six genotypes of barley. The results showed that inherent microchemical-structural differences in the endosperm among the six barley varieties were detected by the synchrotron-based analytical technique, SFTIRM, with the univariate molecular spectral analysis. The SFTIRM spectral profiles differed (P < 0.05) among the barley samples in terms of the peak ratio and peak area and height intensities of amides I (ca. 1650 cm(-1)) and II (ca. 1550 cm(-1)), cellulosic compounds (ca. 1240 cm(-1)), CHO component peaks (the first peak at the region ca. 1184-1132 cm(-1), the second peak at ca. 1132-1066 cm(-1), and the third peak at ca. 1066-950 cm(-1)). With the SFTIRM technique, the structural characteristics of the cereal seeds were illuminated among different cultivars at an ultraspatial resolution. The structural differences of barley seeds may be one reason for the various digestive behaviors and nutritive values in ruminants. The results show weak correlations between the functional groups' spectral data (peak area, height intensities, and ratios) and rumen biodegradation kinetics (rate and extent of nutrient degradation). Weak correlations may indicate that limited variations of these six barley varieties might not be sufficient to interpret the relationship between spectroscopic information and the nutrient value of barley grain, although significant differences in biodegradation kinetics were observed. In conclusion, the studies demonstrated the potential of ultraspatially resolved synchrotron based technology (SFTIRM) to reveal the structural and chemical makeup within cellular and subcellular dimensions without destruction of the inherent structure of cereal grain tissue.

Recent applications and current trends in Cultural Heritage Science using synchrotron-based Fourier transform infrared micro-spectroscopy

Abstract Synchrotron-based Fourier transform infrared micro-spectroscopy (SR-FTIR) is one of the emerging techniques increasingly employed for Cultural Heritage analytical science. Such a technique combines the assets of FTIR spectroscopy (namely, the identification of molecular groups in various environments: organic/inorganic, crystallized/amorphous, solid/liquid/gas), with the extra potential of chemical imaging (localization of components + easier data treatment thanks to geographical correlations) and the properties of the synchrotron source (namely, high brightness, offering high data quality even with reduced dwell time and reduced spot size). This technique can be applied to nearly all kind of materials found in museum objects, going from hard materials, like metals, to soft materials, like paper, and passing through hybrid materials such as paintings and bones. The purpose is usually the identification of complex compositions in tiny, heterogeneous samples. Recent applications are reviewed in this article, together with the fundamental aspects of the infrared synchrotron source which are leading to such improvements in analytical capabilities. A recent example from the ancient Buddhist paintings from Bamiyan is detailed. Emphasis is made on the true potential offered at such large scale facilities in combining SR-FTIR microscopy with other synchrotron-based micro-imaging techniques. To cite this article: M. Detecting molecular changes in Vimy flaxseed protein structure using synchrotron FTIRM and DRIFT spectroscopic techniques: Structural and biochemical characterization

Mid-IR techniques were used to characterize any changes that occurred on a molecular level in flaxseed that had been heated using an autoclave. The objectives were to investigate the effects of autoclave heating on differences in diffuse reflectance infrared Fourier transform (DRIFT) and synchrotron-based Fourier transform infrared microspectroscopy (SFTIRM) based measurements of the protein α-helix to β-sheet ratio for flaxseed (Linum usitatissimum), cv. Vimy. Hierarchical cluster analysis (CLA) and principal components analysis (PCA) were also conducted to identify molecular differences in the DRIFT spectra. Flaxseed samples were kept raw for the control or autoclaved in batches at 120°C for 20, 40 or 60 min for treatments 1, 2 and 3, respectively. DRIFT analysis of protein secondary structure ratios showed a decrease (P 0.05). CLA and PCA were successfully used to make distinctions between the different treatment spectra and showed enhanced sensitivity upon selection of a smaller spectral window to include only the amide I and II portion of the IR spectrum. Our results indicated that autoclaving had a great enough effect on the mid-IR spectrum of flaxseed to identify the altered α-helix to β-sheet ratio and subsequently differentiated between the treatments using PCA and CLA suggesting greater sensitivity of mid-IR spectral methods in identifying the effect of heat treatment on protein secondary structure. Future study is needed to quantify the relationship between protein secondary structure and protein functionality.

Vimy. Hierarchical cluster analysis (CLA) and principal components analysis (PCA) were also conducted to identify molecular differences in the DRIFT spectra. Flaxseed samples were kept raw for the control or autoclaved in batches at 120°C for 20, 40 or 60 min for treatments 1, 2 and 3, respectively. DRIFT analysis of protein secondary structure ratios showed a decrease (P&lt; 0.05) in theα-helix toβ-sheet ratio for the whole seed while the results from synchrotron SFTIRM spot data from the endosperm tissue in flaxseed showed autoclaving had the opposite effect (P&gt; 0.05). CLA and PCA were successfully used to make distinctions between the different treatment spectra and showed enhanced sensitivity upon selection of a smaller spectral window to include only the amide I and II portion of the IR spectrum. Our results indicated that autoclaving had a great enough effect on the mid-IR spectrum of flaxseed to identify the alteredα-helix toβ-sheet ratio and subsequently differentiated between the treatments using PCA and CLA suggesting greater sensitivity of mid-IR spectral methods in identifying the effect of heat treatment on protein secondary structure. Future study is needed to quantify the relationship between protein secondary structure and protein functionality.

Ultra-spatial synchrotron radiation for imaging molecular chemical structure: Applications in plant and animal studies

Synchrotron-based Fourier transform infrared microspectroscopy (S-FTIR) has been developed as a rapid, direct, non-destructive, bioanalytical technique. This technique takes advantage of synchrotron light brightness and small effective source size and is capable of exploring the molecular chemical features and make-up within microstructures of a biological tissue without destruction of inherent structures at ultra-spatial resolutions within cellular dimension. To date there has been very little application of this advanced synchrotron technique to the study of plant and animal tissues' inherent structure at a cellular or subcellular level. In this article, a novel approach was introduced to show the potential of the newly developed, advanced synchrotron-based analytical technology, which can be used to reveal molecular structural-chemical features of various plant and animal tissues.

An emerging method for rapid characterization of feed structures and feed component matrix at a cellular leveland relation to feed quality and nutritive value

Feed quality, feed characteristics, nutrient utilization and digestive behaviour are closely related to: (i) total feed composition, (ii) feed intrinsic structures, and (iii) biological component matrix (such as protein to starch matrix, protein to carbohydrate matrix). Conventional “wet” chemical analysis can determine total chemical composition, but fails to detect the feed intrinsic structures and biological component matrix due to destruction of feed samples during the processing for chemical analysis and the “wet” chemical analysis cannot link structural information to chemical information within intact feed tissue. Recently, advanced synchrotron-based Fourier transform infrared (FTIR) microspectroscopy has been developed as a non-destructive and non-invasive structural-chemical analytical technique. This technique can link chemical information to structural information of biological samples within intact tissue within cellular dimensions. It can provide four kinds of information simultaneously: tissue composition, tissue structure, tissue chemistry and tissue environment. However, this novel technique has been found mainly for medical science research, extremely rare for feed science and nutrition research. The objective of this review article was to illustrate synchrotron-based FTIR microspectroscopy as a novel research tool for rapid characterization of feed structures at a cellular level and for detection of chemical features and molecular chemical make-up of feed biological component matrix and nutrient interaction. The emphasis of this article was to show that feed structural-chemical features at a cellular level are closely related to feed characteristics, feed quality and nutritive value in animals. The synchrotron-based technology will provide us with a greater understanding of the plant-animal interface.

Multicomponent Peak Modeling of Protein Secondary Structures: Comparison of Gaussian with Lorentzian Analytical Methods for Plant Feed and Seed Molecular Biology and Chemistry Research

The objective of this study was to compare Gaussian and Lorentzian multicomponent peak modeling methods in quantification of protein secondary structures of various plant seed and feed tissues within intact tissue at a cellular and subcellular level using the advanced synchrotron light sourced Fourier transform infrared (FT-IR) microspectroscopy (S-FTIR). This experiment was performed at the beamline U10B at the National Synchrotron Light Source (NSLS) in Brookhaven National Laboratory (BNL), U.S. Dept of Energy (NSLS-BNL, NY). The results show that in the comparison of the Gaussian and Lorentzian multi-peak modeling methods, the Gaussian method is more accurate for fitting multi-peak curves of protein secondary structures than the Lorentzian method, with higher modeling R(2) values (0.92 versus 0.89, P < 0.05). There were no large differences (P > 0.05) in the quantification of the relative percentage alpha-helices, beta-sheets, and others in protein secondary structures of the plant seed tissues, with averages of 30.2%, 40.4%, and 29.4%, respectively. However, there are significant differences (P < 0.05) in the quantification of the ratios of sheet alpha-helix (1.42 versus 1.60; SEM = 0.058) in protein secondary structures of the plant seed tissues. With synchrotron FT-IR microspectroscopy, the ultrastructural-chemical makeup and nutritive characteristics could be revealed at a high spatial resolution. Synchrotron-based FT-IR microspectroscopy revealed that the secondary structure of protein differed between the plant seed tissues in terms of relative percentage and ratio of protein secondary structures (alpha-helix and beta-sheet) within cellular dimensions. The results also show that the flaxseed tissues contained higher (P < 0.05) percentage alpha-helix (38.6 versus 24.0%) beta-sheet (45.3 versus 36.9%), lower (P < 0.05) percentage of other secondary structures (16.1% versus 39.0%), and higher (P < 0.05) ratios alpha-helix beta-sheet (0.90 versus 0.69) than the winterfat seed tissues. It must be mentioned that the relative percentages of protein secondary structure may not reflect the true secondary structure. However, the purpose of modeling the relative percentage of secondary structure was to detect the variety of differences among seed/feed/plant tissues and their relation to nutritive value and digestive behavior. The results demonstrate the potential of highly spatially resolved synchrotron-based FT-IR microspectroscopy to reveal protein secondary structures of the plant seed/feed tissues. Further study is needed to quantify the relationship between protein secondary structures and nutrient availability and digestive behavior of various varieties of plant seed tissues. Information from the infrared probing of protein secondary structures can be valuable as a guide to maintaining protein nutritive value and quality for animal and human use.
