Investigation of microstructural, micromorphology, and surface plasmon resonance characteristics in Ni/Al, Ni/Cu, and Ni/SS thin films.

As the first boundary between the environment and the material, the surface plays an important role in their interaction with each other, therefore, the use of appropriate tools and analysis to examine the mechanical properties and morphology of surfaces has particular importance in industry and research. In this research, a thin film of nickel was deposited on metal substrates made of aluminum, copper, and steel by using the RF magnetic cathode. Then, using a non-contact atomic force microscope, the morphological properties of the nickel film with static parameters, Minkowski functionals (MF's), fractal, and multifractal were extracted to be analyzed and studied. After that, using parameters such as root mean square (RMS) roughness, skewness, and kurtosis, it was determined how the surface roughness, distribution, and probability density of particles on the film surface alters with the change of the substrate. Next, by examining and analyzing the Δα and Δf parameters obtained from the multifractal section, the morphology of the produced film on the metal substrates was investigated. Then, the change in the surface plasmon resonance (SPR) peak position is changed for the prepared film in the range of the absorption spectrum due to the substrate effect and the microstructural properties of the formed film. HIGHLIGHTS: Ni film has been deposited by Rf magnetron sputtering. The effect of metal substrates on the topography, fractality, and optical properties was studied. Minkowski functionals were used to investigate the surface morphology of the samples. Substrate's material and the topography of the formed film can changed the surface plasmon resonance position.

Surface plasmon resonance in gold-silver bilayer coated D-shaped multimode optical fiber: An approach to refractive index sensing

This paper presents the surface plasmon resonance (SPR) characteristics of D-shaped silica multimode optical fiber coated with bilayer silver-gold film. The effect of various coating thicknesses on the SPR characteristics was investigated through simulation using COMSOL Multiphysics. The inner layer Ag thickness varied from 20 nm to 60 nm, and the outer layer Au thickness ranged from 5 nm to 25 nm. Prior simulation results indicated that a single-layer silver (Ag) coating produces a narrower plasmonic curve than a single-layer gold (Au) coating. The confinement loss and the resonance wavelengths were simulated for the sensing medium with a refractive index (RI) range of 1.40–1.45. The simulation results show that the Ag/Aubilayers coated sensorshavebetter full-width-half-maximum (FWHM) and figure-of-merit (FOM) than the single-layer coated sensors. The Ag/Au with a 40 nm/5 nm thickness exhibits the highest confinement loss, with a wavelength sensitivity of 10 000 nm/RIU and a figure-of-merit (FOM) of 500 RIU−1. The proposed sensor using a D-shaped optical fiber coated with Ag/Au layers has a high potential for remote sensing applications involving chemical liquids, offering robust and accurate measurements within the tested refractive index range.

Microwave-assisted ultrafine silver nanoparticle synthesis using Mitragyna speciosa for antimalarial applications

Abstract This study investigates the antiplasmodial activity of ultrafine silver nanoparticles (AgNPs, 2–5 nm) synthesized using a green approach involving the Mitragyna speciosa extract and emphasizing the microwave-assisted irradiation technique. Various synthesis parameters were optimized, resulting in the successful production of spherical AgNPs, which exhibited a characteristic surface plasmon resonance peak at around 440 nm. The synthesized AgNPs demonstrated high stability, indicated by a zeta potential value of −28 mV. The antimalarial efficacy of the microwave-assisted AgNPs against the P. falciparum strain was evaluated, demonstrating a half-maximum inhibitory concentration (IC50) value of 1.56 µg·mL−1. Further enhancement in the antimalarial performance was observed when the AgNPs were conjugated with chloroquine (CQ), a traditional antimalarial drug, achieving an impressive IC50 value of 24 ng·mL−1. Additionally, all formulations exhibited low toxicity, with a cytotoxic concentration (CC50) exceeding 800 µg·mL−1 in Vero cells. Complementing these experimental findings, specific computational studies offered insights into the interactions between silver atoms and bioactive compounds in M. speciosa, as well as shedding light on the dynamics of CQ functionalization. These experimental and computational findings emphasize the potential of a sustainable, low-toxicity, and cost-effective AgNP synthesis process, showcasing significant promise in advancing green nanotechnology for the development of effective antimalarial medications.

Pharmacological Effect of In Vitro Antioxidant Property and Green Synthesis of Silver Nanoparticles (AgNPs) Utilizing Murraya koenigii Antibacterial Application

ABSTRACT Background: Nonessential heavy metals pose a significant threat to human health due to their toxicity. Mercury, in particular, is identified as a hazardous metal. The study aims to detect mercury using colorimetric analysis with Murraya koenigii, emphasizing the eco-friendliness of the method. Aims and Objectives: The primary objective is to detect mercury using a colorimetric analysis method employing Murraya koenigii. Additionally, the study aims to investigate the eco-friendliness of this detection method. Materials and Methods: Colorimetric analysis was conducted using Murraya koenigii to detect mercury. Ultraviolet-visible (UV-vis) spectroscopy was employed to detect the formation of silver nanoparticles (AgNPs), with a characteristic surface plasmon resonance (SPR) band observed. X-ray diffraction (XRD) data analysis was performed to determine the crystalline nature and size of AgNPs. Scanning electron microscopy (SEM) was utilized to visualize the morphology of AgNPs. Fourier transform infrared (FTIR) spectroscopy was employed to identify functional groups involved in reducing silver ions. Antibacterial properties of synthesized AgNPs were tested against various microorganisms, including Escherichia coli, Staphylococcus aureus, Streptococcus mutans, and Enterococcus faecalis. Results: Mercury was successfully detected using colorimetric analysis with Murraya koenigii. Formation of AgNPs was confirmed by UV-vis spectroscopy, with a characteristic SPR band at 418 nm. AgNPs were found to be crystalline with an average size of 5.20 nm, as determined by XRD analysis. SEM images revealed spherical and polycrystalline AgNPs. FTIR spectra indicated the involvement of the -OH group of compounds in the extract in reducing silver ions. Synthesized AgNPs exhibited antibacterial properties against various microorganisms. Conclusion: A sustainable and eco-friendly method for synthesizing AgNPs using Murraya koenigii extract was successfully developed. This method not only detected mercury but also demonstrated antibacterial properties against various microorganisms. The study underscores the health implications of nonessential heavy metals, emphasizing the importance of eco-friendly detection and mitigation methods.

Efficient Bioelectrochemical Cell Generation and Green Synthesis of Silver Nanoparticles Using Pomegranate and Pineapple Peel Extracts: A Comprehensive Characterization Study

The present study investigates an environmentally conscious method for synthesizing silver nanoparticles (AgNPs) by employing extracts from pomegranate peel (PgP) and pineapple peel (PnP). This green synthesis approach offers a sustainable alternative to traditional chemical methods, thereby reducing the ecological footprint associated with nanoparticle production. The PgP and PnP extracts serve as both reducing and capping agents during the synthesis process, enhancing the biocompatibility of the resultant AgNPs. Various characterization techniques, including UV-Vis spectroscopy, Raman analysis, X-ray diffraction (XRD), dynamic light scattering (DLS), Fourier transform infrared (FTIR), and transmission electron microscopy (TEM), were utilized to analyze the synthesized AgNPs. UV-Vis spectroscopy confirmed the formation of AgNPs through characteristic surface plasmon resonance peaks, while FTIR examined the interaction between biomaterial components and the oxidation and coating of silver nanoparticles. Raman analysis elucidated the functional groups responsible for reducing and stabilizing AgNPs, while XRD provided insights into their crystalline structure. TEM images revealed the size and morphology of the nanoparticles, while DLS characterized their average size and morphology. In addition, the synthesized AgNPs were utilized in a bioelectrochemical cell to leverage their unique properties for enhanced electrochemical performance, showcasing their potential application in energy storage and conversion systems. Overall, this study demonstrates the feasibility of utilizing agricultural waste products such as PgP and PnP for sustainable AgNP synthesis, offering promising prospects for environmentally friendly nanotechnology advancement.

Investigation of the effect of amine and thiol as functional groups on gold Nanobipyramids properties

Metal nanoparticles are increasingly popular due to their unique properties, such as their vast surface area, tiny size, strong reactivity in live cells, and high-temperature stability. Gold nanoparticles, as one of the metal nanoparticle types, are frequently utilized in biosensor applications because of their adjustable Localized Surface Plasmon Resonance (LSPR) characteristics, less cytotoxic nature, and increased sensitivity to changes in the surrounding medium. The anisotropic metal nanoparticle of gold nanobipyramids (GNBPs) provides for higher Refractive Index Sensitivity (RIS) and Figure of Merit (FOM) value than others, allowing for higher sensitivity. This work lies in the comprehensive investigation of GNBPs functionalized with amine and thiol groups over varying shorter time durations (0.5 – 12 h) through the immediate change in optical, structural, morphological, electrical surfaces, and molecular properties. The findings show amine groups enable the red-shifted spectrum, whereas thiol groups impact the blue-shifted spectrum. By employing amine groups, it is able to produce side peaks in the XRD pattern made up of amine atoms. The use of thiol groups, meanwhile, has an impact on the growth increase in the (111) direction. Then, the highest surface density is obtained in 3 h, 59.78 % for amine-functionalized GNBPs (a-GNBPs) and 75.49 % for thiol-functionalized GNBPs (t-GNBPs). Next, a-GNBPs generate the highest angle slope compared to t-GNBPs, i.e., 140.422⁰. In addition, for the electrical surface effect, amine functionalization increases the zeta potential value from 0.3 to 2.5 mV, indicating the inhibition of nanoparticle agglomeration. On the other hand, thiol functionalization affects the decreasing of zeta potential value from 0.3 to -0.5 mV, allowing for nanoparticle agglomeration. Subsequent FTIR characterization analysis obtained shifts in the wave numbers, with the -OH bond exhibiting changes within the range of 3306.75 – 3269.95 cm-1, C=-CH bond providing for 2138.48 – 2136.56 cm-1 range, and the C = O bond showing alterations between 1637.59 – 1637.33 cm-1 following amine and thiol functionalization. Extended observation reveals that both a-GNBPs and t-GNBPs exhibit significant signal responses in glucose detection applications utilizing LSPR-based sensors, suggesting the feasibility of functionalizing GNBPs with amine and thiol groups for potential biosensor development.

Multi-interfacial bridging engineering of flexible MXene film for efficient electromagnetic shielding and energy conversion

Accompanied by the progressive development of electronic equipment, excellent electromagnetic interference (EMI) shielding materials display a satisfying prospect in protecting electronic devices against electromagnetic pollution/radiation, while integrating energy conversion. Heretofore, it remains a conundrum to availably construct thin films with multi-interfacial bridging engineering as multifunctional shielding devices. To effectively achieve electromagnetic wave attenuation and integrate energy conversion, a co-mixed vacuum-assisted filtration strategy is designed to synthesize Au@MXene/cellulose nanocrystal/dodecylbenzenesulfonic acid-doped polyaniline (AMCP) films. Profited from the interfacial engineering, the total EMI shielding effectiveness (SE) can be increased by 27 % with the highest value of 67.9 dB. MXene with localized surface plasmon resonance characteristics gives the composite films good energy conversion performance, that is, the composite film can be rapidly heated up to 100 °C under the irradiation of an infrared lamp, and its surface temperature remains stable after continuous irradiation. Additionally, the infrared emissivity is as low as 0.173 within the 8–14 μm, which is necessary to adapt various application scenarios. Therefore, it is reliable that the AMCP films constructed by multicomponent offer a facile strategy for MXene-based EMI shielding devices with integration characteristics.

Fibrillation of Human Serum Albumin Differentially Affected by Asp-, Arg-, and Tyr-Capped Gold Nanoparticles.

Fibrillation of proteins is associated with a number of debilitating diseases, including various neurodegenerative disorders. Prevention of the protein fibrillation process is therefore of immense importance. We investigated the effect of amino acid-capped AuNPs on the prevention of the fibrillation process of human serum albumin (HSA), a model protein. Amino acid-capped AuNPs of varying sizes and agglomeration extents were synthesized under physiological conditions. The AuNPs were characterized by their characteristic surface plasmon resonance (SPR), and their interactions with HSA were investigated through emission spectroscopy in addition to circular dichroism (CD) spectral analyses. Fluorescence lifetime imaging (FLIM) as well as transmission electron microscopy (TEM) were used to observe the fibrillar network. Thermodynamic and kinetic analyses from CD and fluorescence emission spectra provided insights into the fibrillation pathway adopted by HSA in the presence of capped AuNPs. Kinetics of the fibrillation pathway followed by ThT fluorescence emission confirmed the sigmoidal nature of the process. The highest cooperativity was observed in the case of Asp-AuNPs with HSA. This was in accordance with the ΔG value obtained from the CD spectral analyses, where Arg-AuNPs with HSA showed the highest positive ΔG value and Asp-AuNPs with HSA showed the most negative ΔG value. The study provides information about the potential use of conjugate AuNPs to monitor the fibrillation process in proteins.

PH sensor based on tilted fiber Bragg grating surface plasmon resonance with a polyaniline reaction deposition film layer.

In this paper, we propose a surface plasmon resonance (SPR) fiber-optic pH sensor combined with a tilted fiber Bragg grating (TFBG) by continuously coating gold and polyaniline (PANI) onto the surface of a TFBG. The micron-scale thickness polyaniline film provides the sensor with good sensitivity, and it achieves accurate measurement of pH values ranging from 2 to 12 by utilizing the pH-responsive mechanism of PANI and the surface plasmon resonance characteristics. Experimental results show that within the 2-12 pH range, the sensitivity of the TFBG surface plasmon resonance pH sensor based on PANI coating is 0.50335 nm/pH, and results demonstrate, a linear correlation coefficient between wavelength and pH value reaching 0.96614. This indicates significant potential for future engineering applications in real-world pH measurement using this sensor.

Selective Colorimetric Detection of Pb(II) Ions by Using Green Synthesized Gold Nanoparticles with Orange Peel Extract

Gold nanoparticles (AuNPs) were prepared by using a green approach that employed orange (citrus sinensis) peel water extract (OPE) as a reducing agent. In this case, the organic compounds present in orange peel were able to reduce Au(III) to Au(0) and, at the same time, to act as a capping agent, functionalizing the surface of the AuNPs, stabilizing them in a water solution. This “green” approach valorizes orange peel waste as a resourceful material and makes the synthetic process of AuNPs more environmentally sustainable, safe, and economically feasible than the traditional methods. The obtained gold nanoparticles (AuNPs@OPE) were characterized by FT–IR, DLS, SEM analysis, and UV–Vis spectroscopy; the latter showed a characteristic surface plasmon resonance (SPR) band at 530 nm, typical of spherical gold nanoparticles. The AuNPs@OPE were then tested as colorimetric sensors for heavy metals in water, showing an affinity and selectivity toward Pb2+. In fact, in the presence of Pb2+, the added cation favors the aggregation process, and, in this case, nanoparticles form clusters due to the interactions between Pb2+ and the carboxyl/hydroxyl groups on the surface of the AuNPs@OPE, increasing the size of the nanostructure. This process is accompanied by a change in color of the AuNPs@OPE from pink to violet, with a formation of a second, new SPR band, at a higher wavelength, relative to the aggregate formation. The colorimetric assay was tested at different times with the addition of Pb2+ ions showing different LOD values of 13.31 µM and 0.05 µM after 15 min and 90 min, respectively. The proposed colorimetric assay was also tested for analyzing Pb2+ in drinking water samples demonstrating the reliability to use AuNPs@OPE with real samples.

Insights into the impact of surfactants on the kinetics of selenium nanoparticles formation via lemon juice-assisted methodology, followed by their antibacterial and antioxidant assessment

Selenium nanoparticles (Se NPs) fabrication utilizing active species of natural lemon juice represents a promising pathway in the biosynthesis of nanoparticles. The reduction of Se4+ to Se(0) could be attributed to the presence of natural lemon juice, which was considered a supplier of vitamin C, polyphenolic compounds, flavonoids, and sugars. The synthesized spherical Se NPs exhibited a characteristic surface plasmon resonance band (SPR) at λ = 320 nm. The values of the rate constant for the reduction of Se (IV) to Se (0) by lemon juice were estimated to be kobs = 0.0011 s−1 for a Se concentration of 0.75 mmol.dm−3 and a lemon content of 0.001 mol.dm−3 at room temperature. Various conditions have been studied on the kinetics of Se NPs formation, such as the doses of SeO2 and lemon juice, and the presence of cationic and/or anionic surfactants like cetyltrimethyl ammonium bromide (CTABr) and sodium dodecyl sulfate (SDS), respectively. The synthesized Se particles were characterized by physicochemical techniques such as FT-IR, XRD, TEM, DLS, and zeta potential to investigate their main functional groups, crystallinity, chemical composition, particle size, and surface morphology. Moreover, the antioxidant power of Se NPs was tested in terms of their DPPH free radical scavenging activity. Finally, the antibacterial activity of the studied nanoparticles was confirmed against gram-positive and gram-negative bacteria (Staphylococcus aureus and Pseudomonas aeruginosa, respectively). In short, this study introduced the impact of surfactants on the formation of biocompatible Se NPs as an active antibacterial and antioxidant.

Size Dependence of Gold Nanorods for Efficient and Rapid Photothermal Therapy.

In recent years, gold nanomaterials have become a hot topic in photothermal tumor therapy due to their unique surface plasmon resonance characteristics. The effectiveness of photothermal therapy is highly dependent on the shape and size of gold nanoparticles. In this work, we investigate the photothermal therapeutic effects of four different sizes of gold nanorods (GNRs). The results show that the uptake of short GNRs with aspect ratios 3.3-3.5 by cells is higher than that of GNRs with aspect ratios 4-5.5. Using a laser with single pulse energy as low as 28 pJ laser for 20 s can induce the death of liver cancer cells co-cultured with short GNRs. Long GNRs required twice the energy to achieve the same therapeutic effect. The dual-temperature model is used to simulate the photothermal response of intracellular clusters irradiated by a laser. It is found that small GNRs are easier to compact because of their morphological characteristics, and the electromagnetic coupling between GNRs is better, which increases the internal field enhancement, resulting in higher local temperature. Compared with a single GNR, GNR clusters are less dependent on polarization and wavelength, which is more conducive to the flexible selection of excitation laser sources.

Core-shell structured lignin-stabilized silver nanoprisms for colorimetric detection of sulfur ions

Widespread occurrence of sulfides in domestic and industrial wastewater contributes to environmental pollution and poses risks to human health. Therefore, the development of highly selective, sensitive, and rapid sulfur ion (S2−) detection probes in aquatic ecosystems is of paramount importance. In this study, lignin-stabilized silver nanoprisms (EHL@AgNPRs) were prepared using the seed growth and self-assembly methods. Based on this, a novel, high-performance, and environmentally friendly S2− colorimetric detection method was proposed. Lignin is believed to coat the surface of AgNPRs through cation-π and electrostatic interactions, acting as an excellent dispersant and stabilizer to prevent aggregation and shape deformation. This allows AgNPRs to maintain localized surface plasmon resonance (LSPR) characteristics and superior colorimetric sensing sensitivity towards S2− even after 30 d. The EHL@AgNPRs exhibited remarkable selectivity towards S2− with a minimum detection limit of 41.3 nM. The conjugation of lignin with AgNPRs offers a highly promising approach for the rapid detection of S2− in natural aquatic environments and for the valorization of lignin.

Green synthesis, characterization, and antibacterial activity of silver nano particles against some bacterial isolates

Nanotechnology has emerged as a promising field for the development of novel antibacterial agents with reduced environmental impact. In this study, we present a novel green synthesis approach for the production of silver nanoparticles (AgNPs) using a plant-based extract. These AgNPs were subsequently characterized using various analytical techniques, including UV-Vis spectroscopy, X-ray diffraction (XRD), transmission electron microscopy (TEM), and Fourier-transform infrared spectroscopy (FTIR). UV-Vis spectroscopy confirmed the formation of AgNPs by exhibiting a characteristic surface plasmon resonance peak at around 401 and 420 nm. XRD analysis revealed the crystalline nature of the AgNPs, with distinct diffraction peaks corresponding to the face-centered cubic structure of silver. TEM analysis demonstrated that the synthesized AgNPs were predominantly spherical in shape and exhibited an average size within the nanoscale range. FTIR analysis was employed to elucidate the potential bioactive compounds present in the plant extract responsible for the reduction and stabilization of AgNPs. Furthermore, we evaluated the antibacterial activity of these synthesized AgNPs against a panel of bacterial isolates. All the bacterial isolates were sensitive to the silver nanoparticles. Staphylococcus aureus as found to be most resistant, while E. coli as found to be the most sensitive.

Study the Optical Properties of Noble Metal (Au, Ag, and Cu) Thin Film Biosensors Based on Surface Plasmon Resonance Performance by a Simulation Program

In this work, we studied the effect noble metal thicknesses, namely gold (Au), silver (Ag), and copper (Cu) on the surface plasmon resonance (SPR) characteristics for sensitive optical sensor and biosensor applications. SPR spectra for Au, Ag and Cu were widely studied in the Fresnel equation to calculate the total reflection, the wavelength ([Formula: see text] range from 500 nm to 700 nm with the angle of incidence ([Formula: see text] ranging from 0∘ to 90∘ and thickness of the film ([Formula: see text] reaching from 30 nm to 70 nm. Analysis simulation (in MATLAB) has been made for SPR of three metals placed on the N_LASF9 glass prism and the sensing medium is the air. The properties of the angle of resonance [Formula: see text] have been calculated from plotted reflectance via incident angle [Formula: see text]. The results give efficient detection in a change of reflection ([Formula: see text] and full half width maximum FWHM. The minimum reflection was obtained (0.003 a.u.) at wavelength 700 nm and thickness [Formula: see text]= 50 nm for gold Au at [Formula: see text] = 0.0. The best FWHM was recorded at the wavelength (500 nm) at thickness (70 nm) for silver at [Formula: see text] = 0.0, which was equal (0.1 nm).

Development of silver nanocubes created by pulsed laser ablation in liquid

This paper describes a simple approach for creating silver (Ag) nanocubes using pulsed laser ablation in a liquid medium. The development of nanocubical formations of Ag obtained by laser ablation using Nd: YAG laser was conducted for 5, 10, 15, and 20[Formula: see text]min. The surface morphological analysis was performed using field-emission scanning electron microscopy (FESEM) to show the formation of silver nanocubes with edge lengths ranging from 150[Formula: see text]nm to 250[Formula: see text]nm. The UV-visible spectroscopy demonstrates that the concentration of Ag nanostructures, evidenced by the characteristic localized surface plasmon resonance band near 400[Formula: see text]nm in the colloidal solution containing Ag nanoparticles, increased with the increasing laser ablation duration from 5[Formula: see text]min to 20[Formula: see text]min. The growth mechanism for Ag nanocubes can be easily understood with the change in laser ablation time from 5 to 10, 15, and then 20[Formula: see text]min. The Ag sheets with no specific shape start to develop after 5 min of laser ablation, and after 10[Formula: see text]min, larger particles form. Then, after 15[Formula: see text]min, a small number of cube-like nanostructures with rough and uneven edges was obtained. At the end of 20[Formula: see text]min, a complete cubic formed with fine and distinct edges and a very large amount of nanocubes. The elemental silver signal was present in Ag nanocubes, as revealed by the energy-dispersive X-ray spectroscopy (EDS) spectra. The produced Ag nanocubes may be used to construct two-dimensional nanocomposites with practical applications in the electrical, optoelectronic, electrochemical, and biological areas.

Recent advances in surface modified gold nanorods and their improved sensing performance.

Gold nanorods (AuNRs) have received tremendous attention recently in the fields of sensing and detection applications due to their unique characteristic of surface plasmon resonance. Surface modification of the AuNRs is a necessary path to effectively utilize their properties for these applications. In this Article, we have focused both on demonstrating the recent advances in methods for surface functionalization of AuNRs as well as their use for improved sensing performance using various techniques. The main surface modification methods discussed include ligand exchange with the assistance of a thiol-group, the layer by layer assembly method, and depositing inorganic materials with the desired surface and morphology. Covered techniques that can then be applied for using these functionalized AuNRs include colourimetric sensing, refractive index sensing and surface enhance Raman scattering sensing. Finally, the outlook on the future development of surface modified AuNRs for improved sensing performance is considered.

Rapid colorimetric sensing of chlorpromazine HCl antipsychotic through in situ growth of gold nanoparticles.

Antipsychotic drugs like chlorpromazine hydrochloride (CPZ) are widely used to treat mental illnesses but can accumulate in the environment if not properly disposed of. Long-term exposure to trace levels of such pharmaceuticals may pose health risks. This study reports a colorimetric assay for detection of the antipsychotic drug chlorpromazine hydrochloride (CPZ) based on its ability to reduce gold ions and form gold nanoparticles (AuNPs). Optimization of reaction conditions such as pH, temperature and reagent concentrations enabled quantitative analysis of CPZ concentrations from 0.1-30 μg mL-1, with a detection limit of 0.06 μg mL-1, 0.23 μg mL-1 quantification limit and less than 3.5% RSD. The AuNPs exhibited a characteristic surface plasmon resonance band at 527 nm detectable by UV-vis spectrophotometry. Method validation with spiked serum, urine and environmental water samples demonstrated acceptable accuracy and precision. Interfering substances showed minimal impact, indicating resilience and specificity. This rapid, inexpensive colorimetric assay could facilitate environmental monitoring and biomedical analysis of antipsychotic drugs.

Green synthesis of silver nanoparticles using Justicia adhatoda leaves extract and its anticancer effect on human lung carcinoma via induced apoptosis mechanism

In this study, Justicia adhatoda leaves extract was used to synthesize silver nanoparticles (AgNPs) and assess its anticancer attributes against human lung carcinoma (A549) cell line via induction of apoptosis mechanism. Initially, the biosynthesis of AgNPs was evident by observing the appearance of dark brown colour solution. UV–Vis spectrum showed characteristic surface plasmon resonance peak at 420 nm. FT-IR spectroscopy demonstrated the presence of distinct functional groups in both the extract and AgNPs. The peaks in XRD pattern corresponded to 101, 111, 200, and 220 crystal planes, thereby representing face-centred cubic silver. The EDX analysis confirmed the reduction of silver ions. The HR-SEM revealed spherical-shaped nanoparticles with an average size of 8–13 nm. In vitro cytotoxicity of extract and AgNPs was determined against A549 cell lines using MTT assay. The AgNPs revealed higher cytotoxity (59.25 ± 1.3–5.55 ± 1.3 %) than the extract (81.48 ± 1.3–16.66 ± 1.6 %) with reduced viabilities of cells at different concentrations. The IC50 values of the extract and AgNPs were calculated as 95.36 and 15.1 µg/mL, respectively. Apoptosis of AgNPs was analyzed using varied techniques as per the standard protocols. Acridine orange staining showed reduced green fluorescence of A549 cells due to the action of AgNPs. DNA fragmentation assay revealed the breakage of DNA double strands of cells and yielded laddered pattern. Flow cytometry analysis depicted increment in the cells count in S phase and concomitant decrement in Go/G1 phase. Quantitative real-time PCR demonstrated the upregulation and downregulation of Bax and Bcl-2 genes, respectively. Additionally, the exposure of A549 cells with AgNPs induced cytochrome C production from depolarized mitochondrial membrane into cytosol, which further upregulated caspase-9 and caspase-3 genes. In conclusion, J. adhatoda leaves-mediated AgNPs induced the apoptosis mechanism in A549 lung adenocarcinoma cells and indicated its paramount role as an effectual anticancer agent in future nanomedicine.
