Binary to ternary drug-drug molecular adducts of the antihypertensive drug ketanserin (KTS) with advanced physicochemical properties.

Focusing on a reliable supramolecular synthon approach, novel molecular salts of the antihypertensive medication ketanserin (KTS) with aromatic carboxylic acid derivatives (benzoic acid (BA), 2-hydroxybenzoic acid (2-HBA), and 2,5-dihydroxybenzoic acid (2,5-DHBA)) are reported. Binary salts of KTS with the respective salt former were obtained via solvent-assisted grinding followed by solution crystallization. Salt production was confirmed through crystal structure investigations that revealed proton transfer from the carboxylic acid group of the salt former to the piperidine nitrogen atom of KTS. A rigorous investigation of the crystal packing of novel binary salts of KTS inspired the construction of ternary adducts, and a ternary crystalline product was subsequently identified using milrinone (MLN), another cardiotonic drug. According to our knowledge, this is the first instance of a dual-drug ternary co-crystal combining both antihypertensive drugs. In order to evaluate the impacts of co-crystallization on the in vitro release behaviour of binary and ternary adducts, solubility tests for the cocrystal were carried out under a variety of physiological pH conditions. The results indicate that, in contrast to the parent drug and binary adducts, the solubility rate of the ternary adducts is significantly increased. Finally, the stability of the synthesised adduct was evaluated under a range of conditions, including temperature (40 ± 1 °C), humidity (90% ± 5% RH, 25 °C) and various solvents media, and it was established that they have good stability. We anticipate that the present findings will work with a wide range of medication combinations, providing a potential new approach to create multi-drug systems for cardiovascular disease.

Computational and Experimental Insights in Design and Development of Aceclofenac Co-Crystals

The present study is aimed at the design and development of aceclofenac co-crystals. Co-crystallization is one of the important techniques which helps to enhance the aqueous solubility of drugs by modifying the crystal structure using incorporation of the co-former into the formulation.. In the present study, supramolecular synthon approach and computational grid scan methods were used for the co-former selection. The different co-formers such as oxalic acid (OA), succinic acid (SA), maleic acid (MA), and benzoic acid (BA) were used. Aceclofenac (ACF) co-crystals were prepared by solvent evaporation and dry grinding method. Based on the computational results and experimental saturation solubility studies, maleic acid was considered as the suitable co-former for the preparation. The solid-state characterization showed partial conversion of ACF crystallinity to the amorphization as evident from the decrease in peak intensity and the number of peaks. The co-crystals showed increased aqueous solubility of ACF and higher drug release in basic pH. All the characterization parameters proved the co-crystal formation and the in vitro release studies showed that the solubility enhancement of the ACF by preparing ACF-MA co-crystals. Thus, it can be concluded that co-crystallization is one of the novel method for improving the bioavailability of aceclofenac.

Dual Functionality of Bile Acid: Physical Stabilization of Drugs in the Amorphous Form and Solubility Enhancement in Solution.

Drugs containing an amino aromatic nitrogen moiety were stabilized in the amorphous form by the surfactant cholic acid (CA). Coamorphous systems of lamotrigine (LAM), pyrimethamine (PYR), and trimethoprim (TRI) were each prepared with CA. Drug-CA interactions, investigated by IR and solid-sate NMR spectroscopy, revealed deprotonation of the carboxylic acid group in CA and the protonation of the most basic nitrogen of the drug. The coamorphous systems exhibited exceptional physical stability and resisted crystallization at (i) elevated temperatures (>100 °C) and (ii) accelerated storage conditions, 40 °C/75% relative humidity for 15 months. The dissolution performance of each coamorphous system was compared with the respective crystalline drug based on the area under the curve (AUC) of the concentration-time profiles. A 25-fold increase in AUC was observed in the PYR-CA coamorphous system. The solubility enhancement is attributed not only due to drug amorphization but also due to solubilization by CA. The supramolecular synthon approach, through a drug-CA interaction, yielded physically stable coamorphous systems with enhanced aqueous drug solubility.

Supramolecular Synthon Approach in Designing Organic Sulfonates as Supramolecular Gelators: An Easily Accessible Topical Gel with Antibacterial Properties

The supramolecular synthon approach in the context of the crystal engineering rationale has been exploited to synthesize a new series of primary ammonium sulfonate salts derived from primary alkyl amines with varying alkyl chain lengths (An = CH3–(CH2)n–NH2; n = 2–11, 13–15, 17) and naphthalene-2-sulfonic acid (N2S) as potential supramolecular gelators. The sulfonate salts AnN2S with n ≥ 9 showed the ability to immobilize a number of polar and nonpolar solvents including dimethyl sulfoxide/water, resulting in supramolecular gels which were characterized by dynamic rheology and transmission electron microscopy. Single-crystal X-ray diffraction studies carried out on eight such salts confirmed the presence of gel-inducing hydrogen bonded supramolecular synthons. Anti-bacterial studies (zone inhibition, turbidity, and tetrazolium assays) revealed that the salt A14N2S had the ability to kill the Gram-positive bacterium Staphylococcus aureus. Laser scanning confocal microscopy and flow cytometry data taken under various staining conditions suggested reactive oxygen species-mediated RNA depletion as the plausible cause of bacterial cell death in the presence of the gelator salt. Shear thinning of the aqueous gel of A14N2S along with its anti-bacterial activity indicated that it could be a potential candidate for topical application.

Crystallographic and theoretical interpretation of supramolecular architecture in a new salt hydrate of DL-Tartaric acid and Dimethylamine (DLTA-DA)

Herein, DL-tartaric acid normally used as a coformer in cocrystal engineering is exploited to create the hybrid molecule with secondary amine utilizing the supramolecular synthon approach. The crystal engineering tactics are potentially applied to a wide range of crystalline materials and the nature of the intermolecular interactions, hydrogen bonds in particular which decide the supramolecular stabilization of the solid form can be studied. The mechanochemical mixing of DL-tartaric acid and aqueous dimethylamine showed salt-pseudopolymorphism, resulted in N-methylmethanaminium-3-carboxy-2,3-dihydroxypropanoate hydrate. The interactions present between the molecules were demonstrated by simulating the FTIR spectrum which showed O − H stretching of the carboxylic group that did not match with experimental value as only one supramolecular unit was considered; whereas the experimental spectrum displayed a broad band for O − H stretch at 3200 cm−1. The salt crystallized in the monoclinic system with space group I2/a. The complementary hydrogen bond functionalities present in the molecules instilled the formation of O–H···O, O–H···N, and N − H···O intermolecular contacts. From the graph set analysis, it was noted that the supramolecular architecture was maintained by the homosynthon dimer and one-dimensional O–H···O anionic chain built by R22(12)and C(7) motifs. Additionally, the cage type assembly was formed by additional hydrogen bonds present in between the chains with H2O as a linker. The 2D fingerprint plots confirmed the maximum percentage contribution of -O–H (59%) towards the Hirshfeld surface. Hence, a full analysis of crystal packing and discussions in the context of the supramolecular chemistry of the salt hydrate is presented.

Designing Supramolecular Gelators: Challenges, Frustrations, and Hopes.

This article is a personal account of the author, who serendipitously entered the field of supramolecular gels nearly two decades ago. A supramolecular synthon approach in the context of crystal engineering was utilized to develop a working hypothesis to design supramolecular gelators derived from simple organic salts. The activity not only provided a way to occasionally predict gelation, but also afforded clear understanding of the structural landscape of such supramolecular materials. Without waiting for an ab initio approach for designing a gel, a large number of supramolecular gelators derived from organic salts were designed following the working hypothesis thus developed. Organic salts possess a number of advantages in terms of their ease of synthesis, purification, high yield and stability and, therefore, are suitable for developing materials for various applications. Organic salt-based gel materials for containing oil spills, synthesizing inorganic nanostructures and metal nanoparticles, sensing hazardous gas and dissolved glucose, adsorbing dyes, and facilitating drug delivery in self-delivery fashion have been developed. The journey through the soft world of gelators which was started merely by serendipity turned out to be rewarding, despite the challenges and frustrations in the field.

A supramolecular synthon approach to design amorphous solid dispersions with exceptional physical stability.

A supramolecular synthon approach was exploited to design amorphous solid dispersions (ASDs) of drugs containing an amino aromatic nitrogen moiety and a polyacrylic acid polymer. The interaction between a drug and polymer was confirmed by differential scanning calorimetry, spectroscopy (IR and 15N NMR), and X-ray crystallography. The interaction decreased the molecular mobility, conferred exceptional physical stability and enhanced the drug dissolution.

Supramolecular Synthon Approach in Designing Molecular Gels for Advanced Therapeutics

AbstractLow‐molecular‐weight gelators (LMWGs) are an important class of soft materials that offer various potential applications including drug delivery. Structural diversities of the reported LMWGs and lack of molecular‐level understanding of the self‐assembly process of gelation make it difficult to design a gelator a priori. Most often gelators are discovered in a serendipitous manner and second‐generation gelators are designed by modifying known gelling scaffolds. Since gel network within which the solvent molecules are immobilized is often found to be crystalline, a supramolecular synthon approach in the context of crystal engineering is demonstrated to be quite effective in designing LMWGs for various applications including therapeutics. Self‐drug‐delivery systems, wherein the need for a delivery vehicle does not exist, are becoming an effective alternative to conventional drug delivery systems. In the form of a simple gel (for non‐invasive topical application) or injectable gel (for invasive subcutaneous applications), LMWGs derived from drugs provide an effective way to develop self‐drug‐delivery systems. This review article encompasses the early development of LMWG research, describes gradual transition from discovering just a gelator to a gelator having potential material applications including drug delivery, and highlights the merit of supramolecular synthon approach in designing LMWGs for self‐drug‐delivery applications.

Supramolecular Synthon Approach in Developing Anti-Inflammatory Topical Gels for In Vivo Self-Delivery.

A new series of tertiary-butyl ammonium (TBA) salts of various nonsteroidal anti-inflammatory drugs (NSAIDs) have been synthesized and characterized. Nearly 90 % of the NSAID-derived primary ammonium monocarboxylate (PAM) salts displayed remarkable gelation ability with various solvents including methyl salicylate. Single crystal X-ray diffraction data (SXRD) revealed the existence of 1D PAM synthon in the gelator salts. Structure-property correlation studies based on SXRD and powder X-ray diffraction (PXRD) data established the presence of the 1D PAM synthon in the bulk salts as well as in the corresponding xerogels. A parallel series of salts derived from TRIS (2-amino-2-(hydroxymethyl)-1,3-propanediol) and the same set of NSAIDs displayed poor gelation ability; only 33 % of the salts in the series displayed gelation ability. A few selected gelator salts of both TBA and TRIS were found to be biocompatible (MTT assay with RAW 264.7 cell line) and two of the selected salts (FLR.TBA and FLR.TRIS) possessed anti-inflammatory properties equal to the parent drug FLR (flurbiprofen). Finally a methyl salicylate topical gel derived from FLR.TRIS was successfully delivered in a self-delivery fashion to treat inflamed skin conditions in the mice model. Histological studies of the dorsal tissues of the untreated and treated mice clearly demonstrated the effect of topical gels in such treatment.

Supramolecular Gels by Design: Towards the Development of Topical Gels for Self-Delivery Application.

Following a supramolecular synthon approach, simple salt formation has been employed to gain access to a series of supramolecular gelators derived from the well-known non-steroidal anti-inflammatory drug (NSAID) ibuprofen. A well-studied gel-inducing supramolecular synthon, namely primary ammonium monocarboxylate (PAM), has been exploited to generate a series of PAM salts by reacting ibuprofen with various primary amines. Remarkably, all of the salts (S1-S7) thus synthesized proved to be good to moderate gelators of various polar and nonpolar solvents. Single-crystal and powder X-ray diffraction studies established the existence of the PAM synthons in the gel network, confirming the efficacy of the supramolecular synthon approach employed. Most importantly, the majority of the salts (S2, S3, S6, and S7) were capable of gelling methyl salicylate (MS), an important ingredient found in many commercial topical gels. In vitro experiments (MTT and PGE2 assays) revealed that all of the salts (except S3 and S7) were biocompatible (up to 0.5 mm concentration), and the most suited one, S6, displayed anti-inflammatory ability as good as that of the parent drug ibuprofen. A topical gel of S6 with methyl salicylate and menthol was found to be suitable for delivering the gelator drug in a self-delivery fashion in treating skin inflammation in mice. Histological studies, including immunohistology, were performed to further probe the role of the gelator drug S6 in treating inflammation. Cell imaging studies supported cellular uptake of the gelator drug in such biomedical application.

Experimental and computational crystal structure landscape study of nigerloxin: a fungal metabolite from Aspergillus niger

The crystal structure landscape for the bioactive fungal metabolite, nigerloxin, has been explored in the present study using a combined experimental and computational approach. Two new solvate forms of nigerloxin, have been identified from crystallization screening experiments. Additionally, a crystal structure prediction (CSP) study of nigerloxin anhydrate and monohydrate forms has been performed in the study. A comparison of the radical scavenging properties of nigerloxin and related β-hydroxy-γ-pyrone analogues have been performed with the help of bond dissociation enthalpy (BDE) and HOMO–LUMO energy gap computations, which indicate the lowest BDE and HOMO–LUMO gaps for nigerloxin. Various growth units present in the experimental and predicted crystal structures were identified using the supramolecular synthon approach and were utilized in the identification of the most probable crystal growth pathways available for nigerloxin. A total of twelve distinct crystal growth pathways were identified for nigerloxin anhydrate from the CSP based crystal structural landscape study. The 1-D nigerloxin water tape synthon (1HA) was identified as the most frequently observed growth unit in the predicted structures and was also found present in the two experimental solvate forms. The higher order supramolecular constructs derived from the synthon 1HA were identified as the most probable crystal growth pathways for the nigerloxin monohydrate.

Sweet Theophylline Cocrystal with Two Tautomers of Acesulfame

A novel pharmaceutical cocrystal (THPAH12) of Theophylline (THP) was obtained with an artificial sweetener, Acesulfame (AH), in a molar ratio of 1:2. Solid state NMR spectra of the cocrystal indicate that the two AH molecules exist as keto and enol tautomers, which is further confirmed by the refined crystal structure. THPAH12 is the first keto form AH containing cocrystal. This highlights the fact that not only −OH of enol form of AH, but also the −NH–C═O group of the keto form of AH should be considered when designing new pharmaceutical cocrystals via the supramolecular synthon approach. Compared with pure THP, THPAH12 possesses enhanced solubility and hydration stability, which highlight its potential for further pharmaceutical applications.

Exploiting supramolecular synthons in designing gelators derived from multiple drugs.

A simple strategy for designing salt-based supramolecular gelators comprised of various nonsteroidal anti-inflammatory drugs (NSAIDs) and amantadine (AMN) (an antiviral drug) has been demonstrated using a supramolecular synthon approach. Single-crystal and powder X-ray diffraction established the existence of the well-studied gel-forming 1D supramolecular synthon, namely, primary ammonium monocarboxylate (PAM) synthon in all the salts. Remarkably five out of six salts were found to be capable of gelling methyl salicylate (MS)-an important ingredient in commercially available topical gels; one such selected biocompatible salt displayed an anti-inflammatory response in prostaglandin E2 (PGE2 ) assay, thereby indicating their plausible biomedical applications.

Designing a simple organic salt-based supramolecular topical gel capable of displaying in vivo self-delivery application

The supramolecular synthon approach has been exploited to design simple salt-based supramolecular gelators derived from a non-steroidal anti-inflammatory drug (NSAID) - naproxen; one such biocompatible anti-inflammatory gelator salt was converted into a topical gel that showed excellent in vivo self-delivery application in treating imiquimod (IMQ)-induced skin inflammation in mice.

Solid state manipulation of lornoxicam for cocrystals – physicochemical characterization

Context: Lornoxicam is an analgesic and anti-inflammatory drug of choice and belongs to Class II (low solubility) of BCS (Biopharmaceutical Classification System). Thus bioavailabilities problems are predominant.Objective: Through crystal engineering approach, a method was developed for obtaining multi-component cocrystals of lornoxicam using pharmaceutically acceptable compounds as guests.Materials and methods: Lornoxicam guest-free form was obtained from solution crystallization. Supramolecular synthon approach indicated that lornoxicam was in orthorhombic form. Further presence of intermolecular hydrogen bonding with layered structures was identified. Solvent drop grinding method permitted the formation of cocrystals of lornoxicam with catechol, resorcinol, benzoic acid, hydroxyquinone and 2,4 dihydroxy benzoic acid, all are capable of forming hydrogen bonding.Results and discussion: Lornoxicam cocrystals exhibited the difference in melting points and decomposition characteristics. The analysis of infrared (IR) indicated the shifting of characteristic bands of lornoxicam. The XPRD (X-Ray Powder Diffraction) pattern indicated the crystallinity of cocrystals and significant difference in 2θ value of intense peaks. Differential scanning calorimetry spectra of cocrystals denoted the changes in fusion endotherms, which are in agreements with melting points. The pH solubility profile of lornoxicam showed sigmoidal curve, which substantiated the pKa-dependent solubility. Lornoxicam cocrystals also exhibited a similar pH-solubility profile. Thus pairing of lornoxicam and coformers in the solution at high pH media was assumed. The in vitro dissolution studies of cocrystals were conducted at pH 4.0. The rapid rate of dissolution of cocrystals was observed in initial 10 min. The extent of dissolution was enhanced by 20% on account of cocrystallization.Conclusion: The lornoxicam cocrystals were obtained with improved physicochemical characteristics.

A supramolecular synthon approach to aid the discovery of architectures sustained by C–H⋯M hydrogen bonds

Hydrogen bonding in the form of eight-membered {···HCNM}2 synthons is identified in the crystal structures of nickel and copper complexes. These C-H···M interactions lead to the formation of well-defined zero-, one- and even two-dimensional supramolecular aggregation patterns.

Coformer Selection in Pharmaceutical Cocrystal Development: a Case Study of a Meloxicam Aspirin Cocrystal That Exhibits Enhanced Solubility and Pharmacokinetics

Meloxicam is a nonsteroidal anti-inflammatory drug with low aqueous solubility and high permeability. Because of its low solubility under acidic conditions (e.g., pH 1-5), it can take more than 2 h for meloxicam to reach its therapeutic concentration in humans. Although the slow onset of meloxicam does not necessarily impact the current label indications, the slow onset does prevent meloxicam from its potential application for the relief of mild-to-medium-level acute pain. Pharmaceutical cocrystallization of meloxicam, which represents a promising approach to generate diverse novel crystal forms, could be used to improve the aqueous solubility and accelerate the onset of action. In this contribution, we describe how a novel method can be used for coformer selection to enable the efficient and effective development of a pharmaceutical cocrystal with desired physicochemical and pharmacokinetic properties. Aspirin was selected as the coformer for meloxicam based upon this alternative route, which combines the supramolecular synthon approach with findings in the previous pharmacological and toxicological studies of meloxicam. The resulting cocrystal of meloxicam and aspirin exhibited superior kinetic solubility and possessed the potential to significantly decrease the time required to reach the human therapeutic concentration compared with the parent drug, meloxicam.

Comparison of the polymorphs and solvates of two analogous fungicides—a case study of the applicability of a supramolecular synthon approach in crystal engineering

The polymorphism and solvate formation of thiophanate-ethyl (TE), a fungicidal active, were investigated by solvent crystallization and compared to a close analogue, thiophanate-methyl (TM). Four polymorphs and seven solvates of TE were found and structurally compared with the previously found two polymorphs and fourteen solvates of TM by analyzing the hydrogen bonding patterns and using fingerprint plots, packing coefficients and lattice energies. TE and TM have the same functional groups that can build identical supramolecular synthons. Despite the strong similarities, the polymorphs and solvates of the two actives show significant differences in hydrogen bonding and packing. The results demonstrate the challenges in using a supramolecular synthon approach, and promote the importance in finding methods to also make use of packing effects and lipophilic interactions in crystal engineering.

Ferrocene based organometallic gelators: a supramolecular synthon approach

The supramolecular synthon namely primary ammonium dicarboxylate (PAD) synthon has been exploited to generate a new series of salt based low molecular weight gelators (LMWGs) derived from ferrocenedicarboxylic acid (FDCA) and primary amines Me–(CH2)n–NH2 (n = 3–15). While most of the salts are capable of forming gels with DMSO and DMF, nearly all of them show a tendency to form gels with at least one of the solvents studied. Structure property correlation based on single crystal and powder X-ray diffraction data in combination with optical-, scanning-, and transmission-electron microscopy reveals that the supramolecular synthon approach adopted herein for designing LMWGs is indeed useful and allows one to get an easy access to a new series of organometallic gelators.

Supramolecular Synthons in Designing Low Molecular Mass Gelling Agents: L‐Amino Acid Methyl Ester Cinnamate Salts and their Anti‐Solvent‐Induced Instant Gelation

Easy access to a class of chiral gelators has been achieved by exploiting primary ammonium monocarboxylate (PAM), a supramolecular synthon. A combinatorial library comprising of 16 salts, derived from 5 L-amino acid methyl esters and 4 cinnamic acid derivatives, has been prepared and scanned for gelation. Remarkably, 14 out of 16 salts prepared (87.5 % of the salts) show moderate to good gelation abilities with various solvents, including commercial fuels, such as petrol. Anti-solvent induced instant gelation at room temperature has been achieved in all the gelator salts, indicating that the gelation process is indeed an aborted crystallization phenomenon. Rheology, optical and scanning electron microscopy, small angle neutron scattering, and X-ray powder diffraction have been used to characterize the gels. A structure-property correlation has been attempted, based on these data, in addition to the single-crystal structures of 5 gelator salts. Analysis of the FT-IR and (1)H NMR spectroscopy data reveals that some of these salts can be used as supramolecular containers for the slow release of certain pest sex pheromones. The present study clearly demonstrates the merit of crystal engineering and the supramolecular synthon approach in designing new materials with multiple properties.
