Polysaccharides from Polygonatum cyrtonema Hua Reduce Depression-Like Behavior in Mice by Inhibiting Oxidative Stress-Calpain-1-NLRP3 Signaling Axis.

Polysaccharides from Polygonatum cyrtonema Hua (PSP) exert antioxidant, anti-inflammatory, and antidepressant effects. Production of reactive oxygen species (ROS) and activation of the calpain system and the NOD-like receptor protein 3 (NLRP3) inflammasome are closely related to the pathogenesis of depression. However, the relationships among those pathways and the protective effects of PSP have not been characterized. In this study, lipopolysaccharide (LPS) and chronic unpredictable mild stress- (CUMS-) induced depression models were used to evaluate the protective mechanisms of PSP against depression. ROS levels were measured in HT-22 cells using flow cytometry. Brain tissues were collected to determine the levels of oxidation-related indicators and inflammatory cytokines. The protein levels of calpain-1, calpain-2, calpastatin, phosphatase and Tensin Homolog deleted on Chromosome 10 (PTEN), suprachiasmatic nucleus circadian oscillatory protein (SCOP), nuclear factor-erythroid factor 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), NLRP3, apoptosis-associated speck-like protein (ASC), caspase-1, cleaved-caspase-1, ionized calcium binding adapter molecule 1 (Iba1), phosphorylation of extracellular signal-regulated kinase (p-ERK), nuclear factor-kappa B (NF-κB), interleukin-1β (IL-1β), and glial fibrillary acidic protein (GFAP) were measured using western blotting or immunofluorescence. In cellular experiments, we showed that PSP attenuated LPS-induced production of ROS in HT-22 cells. In animal experiments, we found that LPS increased the expression of calpain-1, NLRP3, ASC, caspase-1, cleaved-caspase-1, Iba1, p-ERK, NF-κB, and GFAP and reduced the expression of calpastatin, PTEN, SCOP, and Nrf2. Administration of PSP reversed these changes. N-Acetyl-L-cysteine (NAC) administration also inhibited oxidative stress and activation of the calpain system and the NLRP3 inflammasome. Furthermore, PSP, calpeptin, MCC950 (a selective NLRP3 inflammasome inhibitor), and NAC reduced LPS-induced proinflammatory cytokine release. We also showed that PSP prevented CUMS-induced changes in the calpain system and the Nrf2 and NLRP3 signaling pathways and reduced depression-like behavior. These results indicate that PSP exerts antidepressant effects through regulation of the oxidative stress-calpain-1-NLRP3 signaling axis.

Calpain inhibition ameliorates depression-like behaviors by reducing inflammation and promoting synaptic protein expression in the hippocampus

Protease activity correlates with depressive or suicidal behaviors, with calpain activation being especially implicated in depression-like behaviors. However, the role of calpain in depression-like behaviors is currently unknown. In this study, the lipopolysaccharide (LPS) - and chronic unpredictable mild stress (CUMS)-induced depression models were used to evaluate the antidepressant effects of calpain inhibitors. Potential mechanisms were determined using pharmacological and biochemical methods. We found that i. p. injection of a calpain inhibitor, calpeptin, prevented LPS-induced depression-like behaviors, activation of astrocytes, inflammation, and reduction of synaptic protein expression levels. LPS injection (i.p.) promoted calpain activity, which degraded suprachiasmatic nucleus circadian oscillatory protein (SCOP). This led to the activation of ERK and nuclear translocation of nuclear factor kappa-B (NF-κB), the promotion of cytokine release, and the reduction of Arc, and PSD95 expression in the hippocampus. In contrast, i. p. injection of calpeptin blocked these changes. Furthermore, intraventricular injection of calpain inhibitor (PD150606) or an ERK inhibitor ameliorated the LPS-induced depression-like behaviors. Administration of calpeptin also remedied CUMS-induced depression-like behaviors, degradation of SCOP, activation of astrocytes, and reduction of synaptic protein expression levels. Finally, we also demonstrated that memantine, an N-methyl-d-aspartic acid (NMDA) receptor antagonist blocks LPS-induced degradation of SCOP. Together, our results show that calpain inhibition ameliorates depression-like behaviors, probably by reducing inflammation and promoting synaptic protein expression in the hippocampus.

Basolateral amygdala calpain is required for extinction of contextual fear-memory.

Extinction of fear-memory is essential for emotional and mental changes. However, the mechanisms underlying extinction of fear-memory are largely unknown. Calpain is a type of calcium-dependent protease that plays a critical role in memory consolidation and reconsolidation. Whether calpain functions in extinction of fear-memory is unknown, as are the molecular mechanisms. In this study, we investigated the pivotal role of calpain in extinction of fear-memory in mice, and assessed its mechanism. Conditioned stimulation/unconditioned stimulation-conditioned stimulation paradigms combined with pharmacological methods were employed to evaluate the action of calpain in memory extinction. Our data demonstrated that intraperitoneal or intra-basolateral amygdala (BLA) injection of calpain inhibitors could eliminate extinction of fear-memory in mice. Moreover, extinction of fear-memory paradigm-activated BLA calpain activity, which degraded suprachiasmatic nucleus circadian oscillatory protein (SCOP) and phosphatase and tensin homolog (PTEN), subsequently contributing to activation of a protein kinase B (AKT)-mammalian target of the rapamycin (mTor) signaling pathway. Additionally, cAMP-response element binding protein (CREB) phosphorylation was also augmented following extinction of fear-memory. Calpain inhibitor blocked the signaling pathway activation induced by extinction of fear-memory. Additionally, intra-BLA injection of rapamycin or cycloheximide also blocked the extinction of fear-memory. Conversely, intra-BLA injection of PTEN inhibitor, bpV, reversed the effect of calpeptin on extinction of fear-memory. Together, our data confirmed the function of BLA calpain in extinction of fear-memory, likely via degrading PTEN and activating AKT-mTor-dependent protein synthesis.

Acute Physiology and Neurologic Outcomes after Brain Injury in SCOP/PHLPP1 KO Mice

Suprachiasmatic nucleus circadian oscillatory protein (SCOP) (a.k.a. PHLPP1) regulates long-term memory consolidation in the brain. Using a mouse model of controlled cortical impact (CCI) we tested if (1) brain tissue levels of SCOP/PHLPP1 increase after a traumatic brain injury (TBI), and (2) if SCOP/PHLPP1 gene knockout (KO) mice have improved (or worse) neurologic outcomes. Blood chemistry (pH, pCO2, pO2, pSO2, base excess, sodium bicarbonate, and osmolarity) and arterial pressure (MAP) differed in isoflurane anesthetized WT vs. KOs at baseline and up to 1 h post-injury. CCI injury increased cortical/hippocampal SCOP/PHLPP1 levels in WTs 7d and 14d post-injury. Injured KOs had higher brain tissue levels of phosphorylated AKT (pAKT) in cortex (14d post-injury), and higher levels of phosphorylated MEK (pMEK) in hippocampus (7d and 14d post-injury) and in cortex (7d post-injury). Consistent with an important role of SCOP/PHLPP1 on memory function, injured-KOs had near normal performance on the probe trial of the Morris water maze, whereas injured-WTs were impaired. CA1/CA3 hippocampal survival was lower in KOs vs. WTs 24 h post-injury but equivalent by 7d. No difference in 21d cortical lesion volume was detected. SCOP/PHLPP1 overexpression in cultured rat cortical neurons had no effect on 24 h cell death after a mechanical stretch-injury.

Postnatal calpain inhibition elicits cerebellar cell death and motor dysfunction.

Calpain-1 deletion elicits neurodevelopmental disorders, such as ataxia. However, the function of calpain in postnatal neurodevelopment and its mechanisms remain unknown. In this study, we revealed that postnatal intraperitoneal injection of various calpain inhibitors attenuated cerebellar cytosolic calpain activity. Moreover, postnatal application of calpeptin (2 mg/kg) apparently reduced spectrin breakdown, promoted suprachiasmatic nucleus circadian oscillatory protein (SCOP) accumulation in cerebellar tissue. In addition, application of calpeptin decreased phosphorylated protein kinase B (p-AKT) level (p<0.05), as well as total AKT level (p<0.05). We also evidenced that administration of calpeptin obviously increased phosphorylation of mammalian target of rapamycin (p-mTor) (p<0.01). Apoptosis of granular cells and activation of caspase-3 (p<0.01) were facilitated after calpain inhibition. Importantly, cell numbers of granular cells were reduced and motor function was remarkably impaired in 4-month-old rats receiving postnatal calpain inhibition. Taken together, our data implicated that calpain activity in the postnatal period was critical for the cerebellar development. Postnatal calpain inhibition causes cerebellar granular cell apoptosis and motor dysfunction, likely through SCOP/AKT and p-mTor signaling pathways.

SCOP/PHLPP1β in the basolateral amygdala regulates circadian expression of mouse anxiety-like behavior.

While disruption of the circadian clock triggers a spectrum of affective abnormalities, how the clock regulates mammalian emotionality remains unclear. Here, we characterized the time-of-day-dependent regulation of mouse anxiety-like behaviors. We show that anxiety-like behaviors are expressed in a circadian manner in mice and demonstrate that the clock machineries in the dorsal telencephalon (dTel) are required for the time-of-day-dependent regulation of anxiety-like behaviors. We identify suprachiasmatic nucleus circadian oscillatory protein (SCOP/PHLPP1β) as an essential intracellular signaling molecule mediating this temporal regulation downstream of the clock. Using viral-mediated, basolateral amygdala (BLA)-specific knockout of Scop, we demonstrate that deletion of SCOP in the BLA exerts anxiolytic effects on the elevated plus maze at early subjective night, thereby blunting the circadian variation in the anxiety-like behavior. We conclude that the circadian expression of SCOP in the BLA plays a key role in generating circadian rhythmicity in the anxiety-like behavior. Our results demonstrate SCOP as a regulator of anxiety-like behaviors and reveal its key roles in the anxiogenic functions of the BLA.

Different patterns of electrical activity lead to long-term potentiation by activating different intracellular pathways.

Deciphering and storing information coded in different firing patterns are important properties of neuronal networks, as they allow organisms to respond and adapt to external and internal events. Here we report that hippocampal CA1 pyramidal neurons respond to brief bursts of high-frequency stimulation (HFS) and θ burst stimulation (TBS) with long-lasting enhanced responses (long-term potentiation [LTP]), albeit by engaging different signaling pathways. TBS induces LTP through calpain-1-mediated suprachiasmatic nucleus circadian oscillatory protein degradation, ERK activation, and actin polymerization, whereas HFS requires adenosine A2 receptors, PKA, and actin polymerization. TBS- but not HFS-induced LTP is impaired in calpain-1 knock-out mice. However, TBS-induced LTP and learning impairment in knock-out mice are restored by activating the HFS pathway. Thus, different patterns of rhythmic activities trigger potentiation by activating different pathways, and cross talks between these can be used to restore LTP and learning when elements of the pathways are impaired.

A molecular brake controls the magnitude of long-term potentiation

Overexpression of suprachiasmatic nucleus circadian oscillatory protein (SCOP), a negative ERK regulator, blocks long-term memory encoding. Inhibition of calpain-mediated SCOP degradation also prevents the formation of long-term memory, suggesting rapid SCOP breakdown is necessary for memory encoding. However, whether SCOP levels also control the magnitude of long-term synaptic plasticity is unknown. Here we show that following synaptic activity-induced SCOP degradation, SCOP is rapidly replaced via mTOR-mediated protein synthesis. We further show that early SCOP degradation is specifically catalyzed by µ–calpain while late SCOP re-synthesis is mediated by m-calpain. We propose that µ–calpain promotes long-term potentiation induction by degrading SCOP and activating ERK, while m-calpain activation limits the magnitude of potentiation by terminating the ERK response via enhanced SCOP synthesis. This unique braking mechanism could account for the advantages of spaced vs. massed training in the formation of long-term memory.

Calpain Cleaves a Memory Inhibitor

Calpain, a calcium-dependent protease, has been implicated in neuronal responses to various stimuli, and Shimizu et al . provide evidence that calpain contributes to certain types of learning and memory. Shimizu et al . (see commentary by Bolshakov) found that suprachiasmatic nucleus circadian oscillatory protein (SCOP) was abundant in the hippocampus of mice and rats. Overexpression of SCOP in cultured hippocampal neurons decreased the expression of an adenosine 3′,5′-monophosphate (cAMP) response element (CRE)-regulated reporter gene stimulated by brain-derived neurotrophic factor (BDNF), whereas knockdown of the endogenous protein using RNA interference enhanced reporter gene expression in the presence or absence of BDNF. CRE-mediated gene expression is downstream of activation of mitogen-activated protein kinases (MAPKs) and contributes to synaptic changes involved in learning and memory. SCOP was previously reported to inhibit K-Ras by sequestering the nucleotide-free form, thereby inhibiting MAPK activation. The authors found that SCOP abundance was decreased in cultured hippocampal neurons stimulated with BDNF, the glutamate receptor agonist NMDA, or KCl. Assays for SCOP cleavage in cell-free extracts in the presence of various protease pharmacological inhibitors indicated that calpains, not caspase 3 or the proteasome, cleaved SCOP and that SCOP cleavage required calcium. In cultured hippocampal neurons, calpain inhibition blocked SCOP degradation and decreased the phosphorylation (and activation) of the MAPK ERK-2 stimulated by BDNF. In mice, memory for new objects was associated with a decrease in the abundance of SCOP (in whole hippocampal extracts). Mice overexpressing SCOP using an inducible promoter showed decreased memory for new objects 24 hours after object training but no deficiency in short-term memory. Consistent with a role for calpain in memory, administration of a calpain inhibitor into the hippocampus blocked new object memory after 24 hours but had no effect on short-term memory measured after 8 minutes. Proteolytic Degradation of SCOP in the Hippocampus Contributes to Activation of MAP Kinase and Memory

Because activation of ERK1/2 MAP kinase (MAPK) is critical for hippocampus-dependent memory, there is considerable interest in mechanisms for regulation of MAPK during memory formation. Here we report that MAPK and CREB-mediated transcription are negatively regulated by SCOP (suprachiasmatic nucleus [SCN] circadian oscillatory protein) and that SCOP is proteolyzed by calpain when hippocampal neurons are stimulated by brain-derived neurotrophic factor (BDNF), KCl depolarization, or NMDA. Moreover, training for novel object memory decreases SCOP in the hippocampus. To determine if hippocampus-dependent memory is influenced by SCOP in vivo, we generated a transgenic mouse strain for the inducible overexpression of SCOP in the forebrain. Overexpression of SCOP completely blocked memory for novel objects. We conclude that degradation of SCOP by calpain contributes to activation of MAPK during memory formation.

SCOPing out Proteases in Long-Term Memory

The ERK1/2 MAP kinase (MAPK) signaling pathway is important for the formation of long-term memories. In this issue, Shimizu et al. (2007) provide new insight into the regulation of this pathway by demonstrating that activation of MAP kinase occurs through the calcium-dependent degradation of SCOP (suprachiasmatic nucleus circadian oscillatory protein) by the protease calpain.

Suprachiasmatic Nucleus Circadian Oscillatory Protein, a Novel Binding Partner of K-Ras in the Membrane Rafts, Negatively Regulates MAPK Pathway

Suprachiasmatic nucleus circadian oscillatory protein (SCOP) is a member of the leucine-rich repeat (LRR)-containing protein family. In addition to circadian expression in the rat hypothalamic suprachiasmatic nucleus, SCOP is constitutively expressed in neurons throughout the rat brain. Here we found that a substantial amount of SCOP was localized in the brain membrane rafts, in which only K-Ras was abundant among Ras isoforms. SCOP interacted directly through its LRR domain with a subset of K-Ras in the guanine nucleotide-free form that was present in the raft fraction. This interaction interfered with the binding of added guanine nucleotide to K-Ras in vitro. A negative regulatory role of SCOP for K-Ras function was examined in PC12 cell lines stably overexpressing SCOP or its deletion mutants. Overexpression of full-length SCOP markedly down-regulated ERK1/ERK2 activation induced by depolarization or phorbol ester stimulation, and this inhibitory effect of overexpressed SCOP was dependent on its LRR domain. These results strongly suggest that SCOP negatively regulates K-Ras signaling in the membrane rafts, identifying a novel mechanism for regulation of the Ras-MAPK pathway.
