First data on polybrominated diphenyl ethers and temporal trend of polychlorinated biphenyls in European brown bear as a bioindicator species

The contamination status on persistent organic pollutants (POPs) in the Croatian terrestrial ecosystem was investigated by analyzing two classes of contaminants in adipose tissue of male European brown bear (Ursus arctos, N = 27) collected in 2021 and 2022. In addition to seven indicator polychlorinated biphenyls (PCBs), for the first time seven polybrominated diphenyl ethers (PBDEs) were analyzed in terrestrial species from this region. Sum of mass fractions detected in adipose tissue ranged from 0.011 to 0.463 ng g−1 lipid weight (lw) for the PBDEs, and from 0.652 to 30.17 ng g−1 lw for PCBs, with two orders of magnitude difference in the median value for these two groups of POPs (PBDEs: 0.022 ng g−1 lw; PCBs: 2.307 ng g−1 lw). PCBs dominated over PBDEs with a share of over 94 % of the total mass fractions of both groups of analyzed POPs. Results of the comparison of the PCB levels in the adipose tissue of brown bears reported here and in previous investigation from this region showed decrease in PCB-28, but increase of PCB-118 and PCB-180, while other congeners seemed to have reached a steady-state level in the environment over a ten-year period. Among the compounds analyzed in this study, BDE-47, PCB-153 and PCB-180 were the dominant congeners. Somewhat higher sum of PCBs was measured in April compared to October, although not statistically significant, indicating possible impact of winter hibernation causing concentration of lipophilic compounds due to body fat loss. Revealed POP levels in brown bears are consistent with the data on the low contamination of the Croatian terrestrial ecosystem in general.

A comparison of stable isotopes and polychlorinated biphenyls among genetic strains of Lake Ontario lake trout (Salvelinus namaycush)

This study quantified stable carbon (δ13C) and nitrogen (δ15N) isotopes, polychlorinated biphenyl (PCB) concentrations and growth rates among multiple genetic strains of Lake Ontario lake trout (Salvelinus namaycush) to evaluate the potential role of genetics in these parameters. Fish ranging in age from 1 to 31 years (n = 72) and representing nine genetic strains including wild-recruits to hatchery fish derived from Lakes Ontario, Superior and Champlain watersheds, and individuals of unknown hatchery origin. Carbon (δ13C) and nitrogen (δ15N) stable isotope values averaged −22.2 ‰ and 17.4 ‰, respectively, but did not differ significantly among genetic strains. ΣPCB concentrations ranged from 42 to 1820 ng/g and varied significantly among individuals including those of similar age and genetic strain. For example, Sum PCB (ΣPCB) concentrations among 7-year-old fish (n = 16) ranged from 159 to 607 ng/g, which compares to growth rates of 3.5 – 32.9 %/yr for these same fish. Multivariate analysis of stable isotope and PCB profiles, however, provided considerable resolution among the strains. For example, fish of unknown hatchery origin ordinated most similar to Seneca Lake fish, the predominant strain stocked in Lake Ontario. Wild fish had a unique ordination with only Lake Superior Klondike strain fish overlapping into their ordination space. Lakes Champlain and Superior strain individuals had similar ordinations but did not overlap substantially with wild or Klondike strain fish. Combined, these differences agree with the ecologies described for these strains in their native ecosystems suggesting that insight can be gained from strain specific evaluations of ecological tracers and these pollutants among Great Lakes lake trout.

Risk factor determination and qualitative risk assessment of Mucormycosis in Harbor Porpoise, an emergent fungal disease in Salish Sea marine mammals

Mucorales infections are increasing in frequency and are a One Health pathogen of concern. In humans and domestic animals, risk factors include being immunocompromised, elevated circulating serum iron, contaminated open wounds, or metabolic diseases such as ketoacidosis or uncontrolled diabetes. Mucormycosis was first identified in 2012 in Pacific Northwest marine mammals, predominantly in harbor porpoises. We performed an assessment to determine the overall qualitative risk, or risk score, of mucormycosis in harbor porpoises. Risk factors for this disease are unknown in aquatic mammals. In a separate risk factor analysis, potential risk factors such as pollutants, trace metals (e.g., iron), and co-infection with other pathogens (e.g., viruses and Brucella spp.) were examined in mucormycosis cases and noncases using a matched case-control study design, to determine the presence and strength of association of these factors with mucormycosis. Disease severity (gross and histopathology) and exposure scores were multiplied together to obtain the overall risk scores of 9 -16 which corresponded to moderate and severe, respectively. In the risk factor analysis, the factors most strongly associated with a mucormycosis case, relative to a control, were elevated liver iron, decreased blubber thickness, and the decreased ratio of the sum of PCB congeners/sum of PBDE congeners. The results of this study suggest that mucormycosis may pose an inordinately high risk to harbor porpoises (and potentially sympatric species in the Salish Sea such as southern resident killer whales) based on the detected prevalence and the severity of lesions observed at necropsy. However, the risk may be greater on an individual basis compared to the overall population, and is likely related to other factors such as increased POP and heavy metal burdens.

Follicular fluid and blood levels of persistent organic pollutants and reproductive outcomes among women undergoing assisted reproductive technologies

Persistent organic pollutants (POPs) are industrial chemicals resistant to degradation and have been shown to have adverse effects on reproductive health in wildlife and humans. Although regulations have reduced their levels, they are still ubiquitously present and pose a global concern. Here, we studied a cohort of 185 women aged 21–43 years with a median of 2 years of infertility who were seeking assisted reproductive technology (ART) treatment at the Carl von Linné Clinic in Uppsala, Sweden. We analyzed the levels of 9 organochlorine pesticides (OCPs), 10 polychlorinated biphenyls (PCBs), 3 polybrominated diphenyl ethers (PBDEs), and 8 perfluoroalkyl substances (PFASs) in the blood and follicular fluid (FF) samples collected during ovum pick-up. Impact of age on chemical transfer from blood to FF was analyzed. Associations of chemicals, both individually and as a mixture, to 10 ART endpoints were investigated using linear, logistic, and weighted quantile sum regression, adjusted for age, body mass index, parity, fatty fish intake and cause of infertility. Out of the 30 chemicals, 20 were detected in more than half of the blood samples and 15 in FF. Chemical transfer from blood to FF increased with age. Chemical groups in blood crossed the blood-follicle barrier at different rates: OCPs > PCBs > PFASs. Hexachlorobenzene, an OCP, was associated with lower anti-Müllerian hormone, clinical pregnancy, and live birth. PCBs and PFASs were associated with higher antral follicle count and ovarian response as measured by ovarian sensitivity index, but also with lower embryo quality. As a mixture, similar findings were seen for the sum of PCBs and PFASs. Our results suggest that age plays a role in the chemical transfer from blood to FF and that exposure to POPs significantly associates with ART outcomes. We strongly encourage further studies to elucidate the underlying mechanisms of reproductive effects of POPs in humans.

Accumulation of polychlorinated biphenyls (PCB) associated with bottom sediments in larvae of Chironomus riparius Meigen

Patterns of bioaccumulation of polychlorinated biphenyls (PCBs) by chironomid larvae (Chironomus riparius Meigen) were studied under laboratory conditions. Larvae were maintained for 20 days, separately in two batches of natural bottom sediments sampled from Rybinsk Reservoir (Russia), each differing in PCB content (425.6 μg / kg or 24.8 μg / kg dry weight, respectively). The study revealed that chironomid larvae accumulated PCBs at levels comparable to the PCB content measured in sediments from each exposure. The bioaccumulation coefficient (1.11 and 1.59, respectively, as per dry weight for compared sediment batches) depended directly on the sum PCB content in sediments. At high sediment PCB content, the ratios of homologous groups of PCBs in larvae and sediment were identical and corresponded to the Aroclor 1254 congeners profile; at lower PCB content, pronounced differences were revealed. In larvae, the ratios of 3, 4, 5, and 6CB homologues were evenly distributed at levels of 20–34 %, while the 4CB homologues prevailed (47 %) in sediments. The content of the latter compounds in larvae was lower than in sediments, and higher for the other homologous groups. The study indicates selective bioaccumulation of various homologous groups of PCBs under conditions of low sediment PCB content.

Neurobehavioural and cognitive effects of prenatal exposure to organochlorine compounds in three year old children

BackgroundWe report data of a Belgian observational prospective cohort study regarding cognitive and behavioural development until the age of 36 months in relation to internal exposure to organochlorine pollutants [sum of polychlorinated biphenyls (sum PCB), dioxin-like activity, PCB118, PCB170, hexachlorobenzene (HCB) and p,p’-dichlorodiphenyldichloroethylene (DDE)] measured in cord blood.MethodsParticipants were recruited as part of an Flemish Environmental Health Survey (2002–2006). Two hundred and six mother-child pairs were recruited. Hundred twenty five toddlers [Reynell Taal Ontwikkelings Schalen (language development, RTOS), Snijders-Oomen Niet-verbale intelligentietest (non-verbal intelligence, SON), Bayley Scales, milestones, Infant Behaviour Questionnaire (IBQ), gender specific play behaviour, Neurobehavioral Evaluation System (NES)-attentional task] and their mothers [Home Observation Measurement of the Environment (HOME), Wechsler Abbreviated Scale of Intelligence (WASI), State-Trait Anxiety Inventory (STAI), general questionnaires] were tested. Statistical analysis was performed with the SPSS program. Much attention was paid to confounding factors.ResultsIn the first years of development, higher organochlorine pollutants were associated with less active children (delayed crawling: sum PCB*HCB (p < 0.05), sumPCB*DDE (p < 0.1); delayed first steps alone: sum PCB (p < 0.5), PCB118 (p < 0.01), PCB170 (p < 0.01), HCB (p < 0.01); less switching between toys: sum PCB (p < 0.01); less switching between toys in boys: PCB118 (p < 0.01), sum PCB(p < 0.01)). At 12 months children with higher dioxin-like activity tended to show less fear responses(p < 0.1) (IBQ 12 months). At 36 months, a slower development of language comprehension (RTOS) was related to all organochlorine exposure parameters(p < 0.1 or p < 0.05) except DDE. Lower nonverbal IQ scores (SON) were related to PCB118 in boys only(p < 0.05 or p < 0.01). Less masculine and more non-gender specific play behaviour was associated with sum PCB in boys and girls at 36 months(p < 0.1). Moreover, PCB118 (p < 0.05), PCB170 (p < 0.1), HCB(p < 0.05) and DDE(p < 0.05) were associated with diminished masculine play behaviour in boys.ConclusionOur data confirm the observations that neurobehavioral development of young children is adversely influenced by environmental concentrations of PCBs, especially in boys. In this context, observation of play behaviour seems to be a reliable, easy to perform and sensitive test to detect neurotoxic effects of chemicals like PCB’s and dioxin-like compounds in very young children. On the basis of our results, we hypothesize that an underarrousal pattern may play a role in the spectrum of effects measured in toddlers prenatally exposed to PCBs and dioxin-like compounds.

Basin-Specific Pollutant Bioaccumulation Patterns Define Lake Huron Forage Fish.

The Lake Huron ecosystem is unique among the Laurentian Great Lakes (USA/Canada) in that its surface area encompasses 3 distinct basins. This ecosystem recently experienced significant ecological restructuring characterized by changes in primary production, species dominance and abundances, and top predator energy dynamics. However, much of the evidence for this restructuring has been largely derived from biomonitoring data obtained from long-term sampling of the lake's Main Basin. We examined polychlorinated biphenyl (PCB) concentrations and the stable isotopes of carbon (δ13 C) and nitrogen (δ15 N) in rainbow smelt (Osmerus mordax), bloater (Coregonus hoyi), and round goby (Neogobius melanostomus) to determine spatial variability in these environmental markers as indicators of the ubiquity of trophic restructuring throughout Lake Huron. Stable isotopes indicated that North Channel fish occupied trophic positions between 0.5 and 1.0 lower relative to Main Basin and Georgian Bay conspecifics, respectively. Sum PCB concentrations for 41 congeners were highest for fish from the Main Basin (27.5 ± 3.0 ng g-1 wet wt) and Georgian Bay (26.3 ± 3.4 ng g-1 wet wt) relative to North Channel (13.6 ± 1.2 ng g-1 wet wt) fish. Discriminant functions analysis demonstrated basin-specific PCB congener profiles with individual species also having distinct profiles dependent on their basin of collection. These bioaccumulation patterns among Lake Huron forage fish mirror those reported for lake trout in this lake and indicate that the degree of food-web ecological restructuring in Lake Huron is not equivalent across the basins. Specifically, basin-specific PCB congener profiles demonstrated that differences among Lake Huron secondary and top predator consumer species are likely dictated by cross-basin differences in zooplankton community ecology and trophodynamics that can regulate the efficiencies of prey energy transfer and PCB congener bioaccumulation patterns in aquatic food webs. Lactational exposure to polychlorinated biphenyls is higher in overweight /obese women and associated with altered infant growth trajectory: A pilot study

BackgroundInfant exposure to environmental chemicals, such as polychlorinated biphenyls (PCBs), may contribute to developmental programming of long-term metabolic disease risk. PCBs persist given their lipophilicity and long half-lives, allowing them to bio-accumulate in adipose tissue. These compounds can then be excreted into maternal milk resulting in infant exposure. ObjectiveTo determine the level of PCBs in milk from mothers with pre-pregnancy overweight and obese (OW/OB) versus normal weight status (NW) and evaluate the association of milk PCB levels with infant growth over the first 6 months of life. MethodsA pilot study of a subset of milk samples from mothers with NW (pre-pregnancy body mass index (BMI) < 25 kg/m2, n = 11) and OW/OB (pre-pregnancy BMI ≥ 25 kg/m2, n = 8) were examined approximately 2-weeks postpartum. PCB congeners 138 + 163, 132 + 153, 180, and the sum were measured using gas chromatography/mass spectrometry and adjusted for milk fat content. Infant growth was monitored from birth to 6 months for weight-for-age (WFA), length-for-age (LFA), weight-for-length (WFL), head circumference-for-age (HCA), and associations with milk PCB content determined using linear mixed modeling. ResultsTotal milk fat content did not differ by maternal weight status (p = 0.88). Milk from mothers with OW/OB had significantly higher PCB sum (p = 0.02) and PCB 138 + 163 (p = 0.03). PCB 132 + 153 (β −0.0008, p = 0.0218), PCB 180 (β −0.0010, p = 0.0279), and PCB sum (β −0.0006, p = 0.0138) were negatively associated with HCA Z-score growth to 6 months. PCB 180 was negatively associated with infant WFA (β −0.0015, p = 0.0058) and WFL Z-score (β −0.0016, p = 0.0263) to 6 months. There were no associations of PCB sum content with WFL, LFA, WFL Z-score over the first 6 months of life. ConclusionsMaternal overweight and obesity are associated with higher levels of total PCB congeners (132, 138, 153, 163, 180) in human milk. PCB congeners have negative associations with infant head circumference and weight trajectory over the first 6 months of life.

Association of plasma levels of lipid and polychlorinated biphenyls in Iranian adult

BackgroundExposure to Polychlorinated biphenyls (PCBs) continues over the world through seafood consumption and indoor exposure to building materials containing PCB. This study aimed to assess the relationship between plasma level of PCB congeners and lipid profile and Body Mass Index (BMI) as well. MethodsThe study population consisted of 181 Iranian adults. Data on BMI, plasma concentration of PCB congeners and serum level of lipid profile including Triglyceride, low-density lipoproteins and high-density lipoproteins, recruited from database of a project entitled “Occupational and environmental exposure to PCBs in Iran”. Multiple linear regression analysis of associations between different quartiles of PCB congeners and various lipid fractions and BMI have been conducted. ResultsA linear increase in average serum Triglyceride and low-density lipoproteins (LDL) levels of participants in first, second, third and fourth quartiles of some PCB congeners was obtained. Following adjustment for age, gender, diet and other variables, only the association between different quartiles of PCB 138, PCB 153, PCB 118 and PCB sum and TG remained statistically significant. ConclusionThe study showed a significant positive relation between plasma PCBs concentrations and serum level of TG in the study population with normal PCBs levels.

Is Kragujevac city still a "hot spot" area, twenty years after the bombing?

After NATO bombing of Serbia in 1999, UNEP has identified Kragujevac as one of the four heavily polluted environmental "hot spots". Damaging of industrial and military targets caused the release of substantial amounts of hazardous chemical substances into the environment. This study was conducted in order to access the exposure of residents of Kragujevac city to persistent soil pollutants, twenty years after NATO air campaign. The paper reports the results of measuring radionuclides, polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs) and heavy metals (As, Cd, Co, Cr, Cu, Mn, Ni, Pb, Zn, and Hg) in soil samples collected from two depths (0-15cm and 15-30cm) at 30 locations along the riverbank of the Lepenica River. The average specific activities of 226Ra, 232Th, 40K and 137Cs were comparable to average worldwide values; excess lifetime cancer risk (ELCR) from natural radionuclides ranged from 1.1·10-4 to 3.3·10-4. The measured concentrations of As, Co, Cr, Cu, and Ni exceeded the limit values in most of the samples. Non-carcinogenic risk (hazard quotient and hazard index) and carcinogenic risk from heavy metals were assessed. Total hazard index was 0.257 and 2.16 for adults and children, respectively. Sum of measured PAHs ranged from 110 to 1026μgkg-1. Sum of PCBs exceeded the limit value of 20μgkg-1 in all samples (it ranged from 48.8 to 196.8μgkg-1), but it was still below the remediation level. The differences between two layers with respect to all measured variables were not statistically significant.

Predictors of plasma polychlorinated biphenyl concentrations among reproductive-aged black women.

Polychlorinated biphenyls (PCBs) are a class of lipophilic endocrine-disrupting chemicals with wide industrial use in the U.S. from the 1930s through 1977. Due to their environmental and biological persistence, low levels of PCBs remain detected in wildlife and humans. Although U.S. studies have shown higher serum PCB concentrations among Black women compared with White women, studies of correlates of PCB exposure among Black women are scarce. We examined predictors of plasma PCB concentrations in a cross-sectional analysis of baseline data from a prospective cohort study of 1693 premenopausal Black women aged 23-35 years from Detroit, Michigan (2010-2012). We collected demographic, behavioral, dietary, and medical data via self-administered questionnaires, telephone interviews, and in-person clinic visits, as well as non-fasting blood samples. We measured concentrations of 24 PCB congeners in baseline plasma from a subset of 762 participants. We used linear regression for log-transformed lipid-adjusted PCB concentrations to calculate percentage differences across levels of selected predictors. We did this separately for individual PCBs, sum of total PCBs, and sum of PCBs by degree of chlorination and hormonal activity. PCB concentrations were positively associated with age, duration of urban residence, cigarette smoking, heavy alcohol intake, and being breastfed in infancy, and inversely associated with body mass index (BMI) and lactation duration. The strength of some associations varied by degree of chlorination. For example, a 5-kg/m2 higher BMI corresponded to a 2.9% lower summed concentration of tri- and tetra-substituted PCBs (95% CI -4.6%, -1.2%), an 8.3% lower summed concentration of penta- and hexa-substituted PCBs (95% CI -10.0%, -6.5%), and a 12.1% lower summed concentration of hepta-, octa-, nona-, and deca-substituted PCBs (95% CI -13.7%, -10.4%). Likewise, associations for age and being breastfed in infancy were stronger for higher-chlorinated PCBs. Results agree with studies on predictors of PCB body burdens, few of which include large numbers of Black women.

Associations of prenatal exposure to polybrominated diphenyl ethers and polychlorinated biphenyls with long-term gut microbiome structure: a pilot study.

The gut microbiome is influenced by early-life exposures, but-despite potentially enormous implications for child health-is understudied in environmental epidemiology. This pilot study is one of the first to explore in utero exposures and long-term gut microbiome profiles. We examined the association between exposure to polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) during pregnancy and the mid-childhood gut microbiome. We measured levels of PBDE-47, -99, -100, and -153 and PCB-138, -153, and -180 in maternal plasma during early pregnancy (n=18) and at delivery (n=25) in women of European descent who breastfed the child participant of the Gestation and Environment cohort in Sherbrooke, Québec (recruited 2007-2009). Bacteria in the mid-childhood (6-8 years) fecal microbiome were detected with 16S rRNA sequencing. To test for differences at the taxon level, we used the Microbiome Comprehensive Association Mapping algorithm. Early pregnancy PCB-153, -180, and the sum of PCBs (Σ3PCB) concentrations were associated with a higher relative abundance of Propionibacteriales and Propionibacteriaceae in mid-childhood. Higher PCB-180 and Σ3PCB were associated with higher relative abundance of Bacillales Family XI. Higher PBDE-99 exposure was associated with a decrease in uncultured bacteria within the Ruminococcaceae NK4A214 group and PBDE-47 was associated with differences in Ruminococcus 2. These taxon-level changes did not result in differences in within- or between-subject diversity. Exposures at delivery were not associated with differences in taxa. Prenatal exposure to PCBs and PBDEs is associated with mid-childhood gut microbiome profiles. Larger studies are needed to confirm these results and explore health implications.

Mechanical Treatment of MSWI Fly Ash: A Way Forward to Inhibit PCB Reformation

ABSTRACT Fly ash from municipal solid waste incinerators has been classified as hazardous waste due to the presence of unintentionally-produced Persistent Organic Pollutants (POPs), with Polychlorinated biphenyls (PCB) being a major group among such POPs. Fly ash can be mechanically (MC) treated to destroy organic pollutants before final disposal or reuse. In this study, model fly ash (MFA) doped with 0.2% CuCl2 was artificially prepared and subjected to high intensity ball milling for 0 (original sample), 1, 2, 4, and 8 h of treatment. Furthermore, to explore PCB reformation in the MC treated fly ash, de novo tests were conducted at single reaction time, temperature, and air flow rate. It was found that the sum of PCB, dioxin like PCB, and corresponding International Toxic Equivalent concentrations (∑PCB, ∑dl-PCB and ∑I-TEQ, respectively) were greatly reduced both in gas and in solid phase of MFA after 8 h of treatment. Though chlorination degree successively decreased with prolonged milling, the homologue and isomer signatures of PCB were varied in concentration. Reformation of few isomers was also observed. Results from present study suggest that MC treatment is an effective method to control reformation of pollutants in the long term. However, few suggestions are made for further ongoing research work on MC treatment of fly ash, which are based on surprising results and experimental limitations obtained in this study.

Association of plasma PCB levels and HbA1c concentration in Iran

BackgroundThe rapid increase in prevalence of diabetes mellitus over the last decades warrants more attention to the effects of environmental and occupational exposures on glucose metabolism. Our study aimed to assess the association between the plasma levels of various congeners of polychlorinated biphenyls (PCBs) and the serum concentration of glycated haemoglobin (HbA1c).MethodsOur study population consisted of 140 Iranian adults from seven different occupational groups and a group of non-occupationally exposed female participants. The plasma concentration of PCBs were determined at the laboratory of occupational toxicology at RWTH Aachen University, Germany. We considered an HbA1c concentration of 5.7% and more as indicating a disturbed glucose metabolism. Logistic regression was used to assess the association between quartiles of concentrations of PCB congeners and serum HbA1c.ResultsParticipants with an increased HbA1c value had higher plasma levels of PCB 138, 153, 180 and the PCB sum, although this association was statistically not significant. There was no significant difference between the levels of PCB 138, 153, 180, the sum of these congeners, and PCB 118 in their quartiles when comparing with HbA1c concentrations.ConclusionsFor our cohort, we could not demonstrate a significant association between PCB and HbA1c concentrations indicating a disturbance of glucose metabolism.

Polychlorinated biphenyls, indicators of thyroid function and thyroid autoantibodies in the Anniston Community Health Survey I (ACHS-I)

In this study, we examined associations between serum concentrations of 35 polychlorinated biphenyl (PCB) congeners, pesticides, and indicators of thyroid function in participants of the Anniston Community Health Survey (ACHS). Study subjects lived in the vicinity of a former PCB production facility and had PCB concentrations 2 to 3 times higher than similar age and race groups from the general population. We investigated associations among serum levels of thyroid hormones (thyroxin [T4], free thyroxin [fT4], triiodothyronine [T3], thyroid stimulation hormone [TSH]) and auto-antibodies (thyroglobulin antibody [TgAb] or thyroperoxidase antibody [TPOAb]) and combined indicators of thyroid function with the sum of PCBs, varying PCB groups, individual PCB congeners, and 8 pesticides. Logistic and linear regression models were adjusted for log10 transformed total lipids, age, sex, ethnicity, BMI, smoking, and family history of thyroid disease. We also performed analyses stratified by ethnicity and sex.Linear regression showed inverse associations between TT3 and thyroid-like PCBs (sum of PCBs 28, 52, 74, 101, 105, and 118; p = .0004), two pesticides (hexachlorobenzene, and pp’-DDE), and individual congeners (PCBs 74, 105). Null associations were observed between PCBs, pesticides, TSH, TT4, and fT4. Logistic regression analyses did not provide support for TT3 findings and found no association with other thyroid hormones, antibodies, or combined indicator of thyroid function. These results suggest there is little evidence that these chemicals have any major clinical effect on thyroid function in this highly PCB exposed population.

Dynamics of persistent organic pollutants in obese adolescents during weight loss

We have investigated the dynamics of various persistent organic pollutants (POPs) in the serum of 94 obese adolescents (34 boys and 60 girls: age range 11–19years) before (0M) and after 5months (5M) of undergoing weight loss treatment. Six groups of POPs, such as polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethane and its metabolites (DDTs), chlordane compounds (CHLs), hexachlorocyclohexane isomers (HCHs), hexachlorobenzene (HCB) and polybrominated diphenyl ethers (PBDEs), were detected in all samples in the decreasing order of median levels: DDTs>PCBs>HCB>HCHs>CHLs>PBDEs. Levels and patterns of POPs between boys and girls at two time-points were similar. DDTs (0M/5M; median: 31/42ng/g lw) and PCBs (0M/5M; median: 17/28ng/g lw) were the major POPs. PCB 153 (0M/5M; 33/34% of the sum PCBs) was the most dominant PCB congener, followed by PCB 138 (0M/5M; 31/31%) and PCB 180 (0M/5M; 13/12%), respectively. The most important PBDE congeners were BDE 47 and 153, although total PBDE levels were low and ranged between 0.63 and 0.88ng/g lw. In general, levels of POPs in the obese adolescents were lower than previously reported in Belgian adolescents and adults. Due to weight loss, serum levels (except PBDEs) increased significantly thereafter combined with a body weight decrease (from 4 to 42kg). Serum concentrations increased by 1–3.5% per kilogram weight loss and 1–2.5% per BMI z-score loss for most POPs. To our knowledge, this is the first report on the dynamics of POPs in obese adolescents during weight loss. Lipid-soluble contaminants were released from adipose tissue into the blood leading to redistribution into the body. Whether the increase in the internal exposure to POPs may adversely influence health remains to be determined.

Serum polychlorinated biphenyls and leukocyte telomere length in a highly-exposed population: The Anniston Community Health Survey

BackgroundSerum polychlorinated biphenyls (PCBs) have previously been associated with longer leukocyte telomere length (LTL) in most, but not all, of the few previous studies. PCBs were produced in Anniston, Alabama from 1929 to 1971 and participants of the Anniston Community Health Survey (ACHS) were highly exposed. ObjectivesWe evaluated serum levels of 35 PCBs and relative telomere length in 559 ACHS participants. MethodsRelative LTL was measured in DNA extracted from blood clots. We assessed PCBs individually, grouped by chlorination, and summed PCBs. We used linear regression to assess the association between each PCB metric while adjusting for pertinent covariates. ResultsSerum PCBs were associated with longer LTL among white participants and the oldest age group of black participants. Among white participants, compared with those in the first quartile of sum PCBs those in the third quartile of sum PCBs had 8.09% longer relative LTL (95% CI: 1.99; 14.55) and those in the fourth had 7.58% longer relative LTL (95%CI: −0.01; 15.76) (p-quadratic=0.05). Among African American participants, serum PCBs were associated with longer relative LTL among those over age 64 only. Tests for interaction were not statistically significant. ConclusionsWe observed a non-linear positive association between serum PCBs and LTL among white participants. Serum PCBs were associated with longer LTL in the oldest age group of African Americans. This association may provide insight into the cancers previously associated with exposure to PCBs, melanoma and non-Hodgkin lymphoma, which have been associated with long LTL in previous studies.

Organochlorine contaminants in wild mink from the lower Great Lakes basin, Canada, 1998-2006.

Organochlorine contaminants were measured in livers of wild mink (Neovison vison) trapped in the lower Great Lakes basin from 1998 to 2006. To assess exposure and potential risk in mink feeding on Great Lakes biota, concentrations of contaminants were compared in mink trapped within 7.8km of the shoreline as well as at inland sites (i.e., 8-40km). Overall, significant spatial variation in mean hepatic concentrations of sum PCBs and seven other organochlorines was found in mink from 13 Great Lakes sites, many of which are within the Great Lakes Areas of Concern. Mean sum PCB concentrations, on a lipid weight basis, ranged from 2μg/g in mink from inland Lake Ontario sites to 44μg/g in mink from western Lake Erie. Concentrations of other organochlorines in mink were generally low. Mink from western Lake Erie had the highest mean cumulative organochlorine burdens dominated largely by PCBs. A significant age effect was found with 1-year-old mink having significantly higher PCB burdens than mink less than 1year in age. With respect to published PCB threshold effect concentrations, some mink exceeded those associated with effects on reproduction and survival as well as the presence of jaw lesions. This was most consistently found in western Lake Erie where the health of populations of wild mink may be adversely affected and where no mink 2years of age or older were collected.

Distribution of serum levels of persistent organic pollutants, heterocyclic aromatic amine theoretical intake and nutritional cofactors in a semi-rural island population.

Persistent organic pollutant (POP) exposure is strongly associated with negative health effects in humans. Heterocyclic aromatic amines (HAAs) are formed during high temperature cooking of foods (i.e. meat and fish). Human exposure to HAA is through food consumption and from similar food groups to POPs. A study of serum samples for POPs in a non-occupational exposed population (n=149, age range 18-80years, recruited in 2012) and comparison with estimated HAA daily intake calculations based on food diaries were undertaken. Three different age groups (group 1, 18-29years; group 2, 30-44years; and group 3, 45-80years) were used to explore possible relationships between POP levels present in blood, HAA intake and nutritional cofactors. Significant differences (p<0.05) between groups (1 and 3) for POP levels were found for p,p'-DDE, polychlorinated biphenyl (PCB) 153, PCB 138 and the sum of PCBs. A similar trend was found between groups 2 and 3 for PCB 153 and sum of PCBs. Significant differences were found between groups 1 and 3 and groups 2 and 3 for HAA intake., i.e. HAA intake was lowest in those of middle age, which may well reflect a different pathway of human exposure between HAA and POPs through the diet preferences.

Bioaccumulation of Polychlorinated Biphenyls (PCBs) in Atlantic Sea Bream (Archosargus rhomboidalis) from Kingston Harbour, Jamaica.

Multiple sizes of Sea bream were collected from Kingston Harbour, Jamaica, to assess steady state bioaccumulation of polychlorinated biphenyls (PCBs) in a tropical fish. Sea beam fork lengths ranged from 7.3 to 21.5cm (n = 36 fish) and tissue lipids decreased with body length. Larger fish had lower δ13C isotopes compared to smaller fish, suggesting a change in diet. Linear regressions showed no differences in lipid equivalent sum PCB concentrations with size. However, differences in individual congener bioaccumulation trajectories occurred. Less hydrophobic PCBs decreased with increasing body length, intermediate PCBs showed no trend, whereas highly hydrophobic (above log KOW of 6.5) PCBs increased. The different congener patterns were interpreted to be a result of decreases in overall diet PCB concentrations with increased fish length coupled with differences in PCB toxicokinetics as a function of hydrophobicity yielding dilution, pseudo-steady state and non-steady state bioaccumulation patterns.
