Interplay between denitrifying and sulfate-reducing bacterial communities under acid mine drainage stress

Acid mine drainage (AMD) pollution leads to the enrichment of sulfate in paddy soil. The interplay between denitrifying and sulfate-reducing bacteria across varying soil depths, and their response to AMD pollution is not yet fully understood. The responses of denitrifying and sulfate-reducing bacteria(SRB) to AMD pollution were analyzed by collecting soil samples at four depths (0–10 cm, 10–20 cm, 20–40 cm, 40–60 cm) under two treatments (AMD-polluted and control soil). AMD pollution resulted in a significant decrease in pH value, elevated SO42- and metal concentrations. Compared to the control soil, denitrifying and sulfate-reducing bacteria in AMD-polluted soils were more sensitive to environmental fluctuations. Co-occurrence analysis revealed that there were more microbiological linkages and nodes in control soil than in polluted soil. Denitrifying and sulfate-reducing bacteria can maintain part of N and S cycle under long-term AMD pollution by increasing the negative correlation in ecological network. Desulfobacca exhibited a positive correlation with nitrate and held a significant position in the symbiotic network, suggesting its potential important role in inhibiting nitrogen loss. This study revealed the intricate ecological network of functional communities, which could have important implications for environmental management and the development of sustainable agricultural practices in areas affected by AMD pollution.

Contribution of sulfate-reducing bacteria in the formation of hydrogen sulfide in coal seams of Hancheng mining area, China

Coalbed hydrogen sulfide (H2S) is a toxic and corrosive gas, it’s very important for researching the origin of H2S in coalbed and related metabolic characteristics of microbes in the bioconversion of coal. The purpose of this study is to clarify the contribution and influencing factors of bacterial and sulfate-reducing bacterial (SRB) communities in the Taiyuan Formation (Nos. 5 and 11 coal seams) of the Hancheng mining area, by using gas composition testing, water quality measurements, 16S rRNA amplicon sequencing, and fluorescence real-time quantitative (qPCR) analysis. The results indicate that the coal seam gas is mainly composed of methane with a trace amount of H2S ranging from 4 to 56 ppm. The H2S content exhibits a positive correlation with the SRB gene copy number within the range of ∼30 to 200 copies/mL, indicating a strong association between the production of H2S and the contribution of SRB in the study area. The presence of Desulfobulbus, Desulfovibrio, Desulfomicrobium, Desulfotomaculum, Desulfobacca, Thermosinus, Desulfococcus, Desulfarculus, and Syntrophobacter genus from the coalbed-produced water further proves that H2S is reduced in dissimilatory sulfate reduction (DSR) pathway. Bacteria and SRB play a critical role in the conversion of coal organic matter through biodegradation. SRB community composition is more related to total organic carbon, total dissolved solids, and trace elements. This study gives an in-depth explanation of the formation of H2S in coalbed and provides novel views in toxic gas removal in future gas exploitation.

Active Sulfate-Reducing Bacterial Community in the Camel Gut

The diversity and activity of sulfate-reducing bacteria (SRB) in the camel gut remains largely unexplored. An abundant SRB community has been previously revealed in the feces of Bactrian camels (Camelus bactrianus). This study aims to combine the 16S rRNA gene profiling, sulfate reduction rate (SRR) measurement with a radioactive tracer, and targeted cultivation to shed light on SRB activity in the camel gut. Fresh feces of 55 domestic Bactrian camels grazing freely on semi-arid mountain pastures in the Kosh-Agach district of the Russian Altai area were analyzed. Feces were sampled in early winter at an ambient temperature of -15 °C, which prevented possible contamination. SRR values measured with a radioactive tracer in feces were relatively high and ranged from 0.018 to 0.168 nmol S cm-3 day-1. The 16S rRNA gene profiles revealed the presence of Gram-negative Desulfovibrionaceae and spore-forming Desulfotomaculaceae. Targeted isolation allowed us to obtain four pure culture isolates belonging to Desulfovibrio and Desulforamulus. An active SRB community may affect the iron and copper availability in the camel intestine due to metal ions precipitation in the form of sparingly soluble sulfides. The copper-iron sulfide, chalcopyrite (CuFeS2), was detected by X-ray diffraction in 36 out of 55 analyzed camel feces. In semi-arid areas, gypsum, like other evaporite sulfates, can be used as a solid-phase electron acceptor for sulfate reduction in the camel gastrointestinal tract.

Sulfate-reducing bacterial community shifts in response to acid mine drainage in the sediment of the Hengshi watershed, South China.

Sulfate-reducing bacteria (SRB) are an attractive option for treating acid mine drainage (AMD) and are considered to be of great significance in the natural attenuation of AMD, but the available information regarding the highly diverse SRB community in AMD sites is not comprehensive. The Hengshi River, which is continually contaminated by AMD from upstream mining areas, was selected as a study site for investigation of the distribution, diversity, and abundance of SRB. Overall, high-throughput sequencing of the 16S rRNA and dsrB genes revealed the high diversity, richness, and OTU numbers of SRB communities, suggesting the existence of active sulfate reduction in the study area. Further analysis demonstrated that AMD contamination decreased the richness and diversity of the microbial community and SRB community, and led to spatiotemporal shifts in the overall composition and structure of sediment microbial and SRB communities along the Hengshi watershed. However, the sulfate reduction activity was high in the midstream, even though AMD pollution remained heavy in this area. Spatial distributions of SRB community indicated that species of Clostridia may be more tolerant of AMD contamination than other species, because of their predominance in the SRB communities. In addition, the results of CCA revealed that environmental parameters, such as pH, TS content, and Fe content, can significantly influence total microbial and SRB community structure, and dissolved organic carbon was another important factor structuring the SRB community. This study extends our knowledge of the distribution of indigenous SRB communities and their potential roles in natural AMD attenuation.

Microbial sulfate reduction facilitates seasonal variation of arsenic concentration in groundwater of Jianghan Plain, Central China

Bacterial sulfate reduction (BSR) plays a vital but complex role in regulating groundwater arsenic concentration. A quarterly hydro-biogeochemical investigation was conducted to clarify how BSR participated in arsenic dynamics in the geogenic As-contaminated alluvial aquifers of the Jianghan Plain, central Yangtze River Basin. Anthropogenic input of sulfate was identified in the transitional season with higher Cl concentrations and Cl/Br molar ratios compared to the monsoon season. Seasonal increase of S(-II) and Fe(II) concentrations in monsoon season suggests the co-occurrence of iron and sulfate reduction. Quantitative analysis of dsrB gene abundance revealed the corresponding variations between dsrB gene abundance (up to 1.2 × 107 copies L−1) and Fe(II) in groundwater. High-throughput sequencing of the dsrB gene identified a considerable proportion of sequences in the sulfate-reducing bacterial community was affiliated with Desulfobulbus (22.7 ± 20.8%) and Desulfocapsa (11.5 ± 11.9%). Moreover, the relative abundance of Desulfocapsa increased with the Fe(II) in the groundwater (R = 0.78, P < 0.01). These results suggest that microbially-mediated sulfate reduction facilitated the abiotic reduction of As-bearing Fe-oxides in the monsoon season after anthropogenic input of sulfate in the transitional season under oscillating redox conditions in the groundwater systems. The present research provides new insights into the critical role of BSR in the seasonal redox cycling of iron and variation of As in the aquifer systems, which are not only applicable in the central Yangtze River basin, but also to other similar As-rich alluvial aquifers worldwide.

Partial function prediction of sulfate-reducing bacterial community from the rhizospheres of two typical coastal wetland plants in China

Sulfate-reducing bacteria (SRB) are ubiquitous anaerobic microorganisms that play significant roles in the global biogeochemical cycle. Coastal wetlands, one of the major habitats of SRB, exhibit high sulfate-reducing activity and thus play significant roles in organic carbon remineralization, benthic geochemical action, and plant-microbe interactions. Recent studies have provided credible evidence that the functional rather than the taxonomic composition of microbes responds more closely to environmental factors. Therefore, in this study, functional gene prediction based on PacBio single molecular real-time sequencing of 16S rDNA was applied to determine the sulfate-reducing and organic substrate-decomposing activities of SRB in the rhizospheres of two typical coastal wetland plants in North and South China: Zostera japonica and Scirpus mariqueter. To this end, some physicochemical characteristics of the sediments as well as the phylogenetic structure, community composition, diversity, and proportions of several functional genes of the SRB in the two plant rhizospheres were analyzed. The Z. japonica meadow had a higher dissimilatory sulfate reduction capability than the S. mariqueter-comprising saltmarsh, owing to its larger proportion of SRB in the microbial community, larger proportions of functional genes involved in dissimilatory sulfate reduction, and the stronger ability of the SRB to degrade organic substrates completely. This study confirmed the feasibility of applying microbial community function prediction in research on the metabolic features of SRB, which will be helpful for gaining new knowledge of the biogeochemical and ecological roles of these bacteria in coastal wetlands.

The shift of sulfate-reducing bacterial communities from the upland to the paddy stage in a rapeseed-rice rotation system, and the effect from the long-term straw returning

Sulfate reduction is an essential process in the biogeochemical sulfur cycle in soils. It is mainly driven by dissimilatory sulfate-reducing bacteria (DSRB). The responses of the DSRB community in a rapeseed-rice rotation system to the environmental variability caused by the shift from the upland to the paddy stages, and the effects of the long-term straw returning on this shift remains unclear. We surveyed the sulfate reducing potential (SRP) and the structure of the DSRB community by high-throughput sequencing that targeted dsrB (and the xenologous dsrB) in soils. The SRP, ranging from 1.2 to 5.8μmold−1dwg−1, was increased in the paddy stage, and it was likely to be enhanced by the long-term straw returning. Highly abundant DSRB optimal taxonomic units (OTUs) were found to be affiliated with the Nitrospirae supercluster (including the uncultured DsrAB lineage WX, lineage 10, and an unknown clade with GU372064), the Environmental supercluster 1 (including the uncultured DsrAB lineage 8 and an unknown lineage related to EF065019), and the Firmicutes group (including the uncultured DsrAB lineage 6 and an unknown lineage between the uncultured DsrAB lineages 2 and 3). The environmental transformation from the upland to the paddy stages led to a decrease in the α-diversity and the number of detectable OTUs. The abundance of a few dominant DSRB OTUs was changed by either the environmental transformation from the upland to the paddy stages or the effects caused by the long-term straw returning. In this study, the changes in the DSRB community structure correlated with the decrease in soil pH and total phosphorus content, and the increase in available sulfate and moisture contents.

Toward an integrated genetic model for vent-distal SEDEX deposits

Although genetic models have been proposed for vent-proximal SEDEX deposits, an equivalent model for vent-distal deposits has not yet appeared. In view of this, it is the object of this paper to present a preliminary integrated vent-distal genetic model through exploration of four major components: (i) nature of the ore-forming fluid, (ii) role of density of the unconsolidated host sediments, (iii) dynamics of sulfate reduction and (iv) depositional environment. Two sub-groups of SEDEX Pb-Zn deposits, vent-proximal and vent-distal, are widely recognized today. Of the two, the latter is by far the largest in terms of metal content with each of the 13 largest containing in excess of 7.5 M (Zn+Pb) metal. In contrast, only one vent-proximal deposit (Sullivan) falls within this size range. Vent-proximal deposits are characteristically underlain by local networks of sulfide-filled veins (commonly regarded as feeder veins) surrounded by a discordant complex of host rock alteration. These attributes are missing in vent-distal deposits, which has led to the widespread view that vent-distal ore-forming fluids have migrated unknown distances away from their vent sites. Because of the characteristic fine grain size of ore minerals, critical fluid inclusion data are lacking for vent-distal ore-stage sulfides. Consequently, hypothetical fluids such as those which formed MVT deposits (120 °C, 20% NaCl equiv.) are considered to represent vent-distal fluids as well. Such high-salinity fluids are capable of carrying significant concentrations of Pb and Zn as chloride complexes while the relatively low temperatures preclude high Cu contents. Densities of such metalliferous brines result in bottom-hugging fluids that collect in shallow saucer-shaped depressions (collector basins). Lateral metal zoning in several deposits reveals the direction from which the brines came. Relative densities of the ore-forming fluid and sediment determine whether the ore-forming fluid stabilizes on top of the sediment column or sinks into it. Metal sulfide precipitation occurs when bacterially produced H2S, diffusing upward from anoxic conditions within the sediment, reacts with metal-bearing chloride complexes in the ore-forming fluid. Since H2S is produced by bacterial sulfate reduction within the first 2 m of the sediment column even where overlain by oxic water, sulfide precipitation will always occur within the anoxic sediment regardless of where the ore-forming fluid comes to rest. Because of the high porosity of the sediment, replacement is precluded as a mechanism of sulfide emplacement in favour of void filling. Detailed textural analyses of the HYC and Howards Pass deposits have demonstrated the abundance of pre-exhalative framboidal pyrite and provide evidence for sulfate-reducing bacteria operating in these basins under normal steady-state conditions before arrival of the ore-forming fluids. The sudden presence of ore-forming fluid, however, dramatically changes the formerly steady-state situation of the local bacterial environment. A major result of this new condition is recorded in the sulfur isotope compositions of the sulfides. Whereas pre-exhalative framboidal pyrite is isotopically light, ore-stage sulfides are significantly heavier and display a reduced fractionation relative to contemporaneous seawater sulfate. Much of the reduced fractionation is linked to the increase in H2S production by sulfate-reducing bacteria. The major factor contributing to this increase is the life-saving action of sulfate-reducing bacteria during which the metal toxicity is mitigated by removal of the toxic ions by precipitating them out as sulfides. Several scenarios representing hypothetical thermochemical sulfate reduction (TSR) conditions convincingly demonstrate the extreme improbability that TSR played a role in formation of vent-distal deposits. A wide range of depositional environments is suggested by host rocks which range from impure carbonate to calcareous or dolomitic siliciclastics to normal siltstones and greywackes to calcareous and siliceous siliciclastics to highly siliceous (cherty) shales. Using the analyses of Mo concentrations as a proxy indicator of euxinia in ancient bottom waters in three vent-distal deposits ranging from Late Cambrian to Early Silurian, euxinia was excluded in all three cases. Regardless of the redox condition of the water column, however, the overriding necessary condition for vent-distal deposits to form is that the water column be quiescent to permit the establishment of a pre-exhalative sulfate-reducing bacterial community. The paper concludes with a six-stage genetic model beginning with exhalation of a dense brine and concluding with sulfide preservation in anoxic sediment.

The development of new high-throughput cultivation methods aims to increase the isolation efficiency as compared to standard techniques that often require enrichment procedures to compensate the low microbial recovery. In the current study, estuarine sulfate-reducing bacteria were isolated using an anaerobic isolation procedure in 384-well microplates. Ninety-nine strains were recovered from initial sediments. Isolates were identified according to their partial 16S rRNA sequences and clustered into 13 phylotypes. Besides, the increase in species richness obtained through enrichments or resampling was investigated. Forty-four enrichment procedures were conducted and shifts in sulfate-reducing bacterial communities were investigated through dsrAB gene fingerprinting. Despite efforts in conducting numerous enrichment conditions only few of them were statistically different from initial sample. The cultural diversity obtained from 3 of the most divergent enrichments, as well as from resampled sediments equally contributed to raise the sulfate-reducing diversity up to 22 phylotypes. Enrichments (selection of metabolism) or resampling (transient populations and micro-heterogeneity) may still be helpful to assess new microbial phylotypes. Nevertheless, all the newly cultivated strains were all representatives of minor Operational Taxonomic Units and could eventually be recovered by maintaining high-throughput isolation effort from the initial sediments.

Response Characteristics between Sulfate-Reducing Bacteria and Environmental Factors in Petroleum Hydrocarbon-Contaminated Field of Northeast China

ABSTRACTLithology samples were collected from six sites of a petroleum hydrocarbon-contaminated site in northeast China along a contamination plume. The sulfate-reducing bacteria (SRB) diversity of all samples was analyzed by PCR-DGGE technology. The Shannon-Wiener indexes (1.004–3.665), Simpson indexes (0.516–0.907) and Pielou indexes (0.996–1.004) of all samples were used to characterize the abundances, advantages and evenness of the microbial communities. Additionally, Canoco for Windows 4.5 was utilized to analyze the correlation between dominant SRB and environmental factors. The results showed that the abundance, advantages and evenness of the sulfate-reducing bacterial community changed regularly along the contamination plume direction. The microbial homology of the samples was not high (0.25–0.80), and the dominant bacteria exhibited heteroplasmy. Additionally, the dominant bacteria were identified as uncultured bacteria. Canonical correspondence analysis (CCA) analysis showed that the distribution and structure of SRB communities were not obviously correlated with the concentration of total petroleum hydrocarbons (TPH), dissolved oxygen (DO) and other environmental factors. The results presented herein provide evidence of natural bioremediation in petroleum hydrocarbon-contaminated fields and information that will be useful to bioremediation of other contaminated fields.

Detection of anaerobic sulfate-reducing bacteria in oxygen-containing upper water layers of the Black and Baltic Seas

Fluorescent in situ hybridization (FISH) and PCR were used for analysis of phylogenetic structure of anaerobic sulfate-reducing bacterial communities in oxygen-containing upper water layers of meromictic basins: the Black Sea and the Gdansk Deep of the Baltic Sea. In the Black Sea (continental slope at depths 30–70 m), cells of sulfate-reducing bacteria (SRB) hybridizing with 16S rRNA-specific FISH-probes for Desulfotomaculum, Desulfobacter, and Desulfovibrio genera were revealed, whereas Desulfomicrobium-related bacteria were prevalent in the chemocline zone at a 150-m depth. Besides Desulfotomaculum (SRB subgroup 1), Desulfobacter (SRB subgroup 4), and Desulfovibrio-Desulfomicrobium (SRB subgroup 6), nested PCR with the use of 16S rRNA gene-specific primers detected the presence of Desulfococcus–Desulfonema–Desulfosarcina (SRB subgroup 5) in the oxygen-containing water column of the Black and Baltic seas. Active enrichment SRB culture that contained bacterium Desulfosporosinus sp. as a major component was obtained from the Black Sea water sample collected at a 70-m depth.

Decoupled distance-decay patterns between dsrA and 16S rRNA genes among salt marsh sulfate-reducing bacteria.

In many habitats, microorganisms exhibit significant distance-decay patterns as determined by analysis of the 16S rRNA gene and various other genetic elements. However, there have been few studies that examine how the similarities of both taxonomic and functional genes co-vary over geographic distance within a group of ecologically related microbes. Here, we determined the biogeographic patterns of the functional dissimilatory sulfite reductase gene (dsrA) and the 16S rRNA gene in sulfate-reducing bacterial communities of US East Coast salt marsh sediments. Distance-decay, ordination and statistical analyses revealed that the distribution of 16S rRNA genes is strongly influenced by geographic distance and environmental factors, whereas the dsrA gene is not. Together, our results indicate that 16S rRNA genes are likely dispersal limited and under environmental selection, whereas dsrA genes appear randomly distributed and not selected for by any expected environmental variables. Selection, drift, dispersal and mutation are all factors that may help explain the decoupled biogeographic patterns for the two genes. These data suggest that both the taxonomic and functional elements of microbial communities should be considered in future studies of microbial biogeography to aid in our understanding of the diversity, distribution and function of microorganisms in the environment.

Sulfate-reducing bacterial communities in the water column of the Gdansk Deep (Baltic Sea)

Biodiversity of sulfate-reducing bacterial communities in the water column of the Gdansk Deep, Baltic Sea, where H2S had been detected in near-bottom layers, was analyzed by PCR with primers for the 16S rRNA genes of six major phylogenetic subgroups of sulfate-reducing bacteria (SRB). Using denaturing gradient gel electrophoresis followed by sequencing, the nucleotide sequences of reamplified dsrB gene fragments from investigated water samples were determined. For the first time the presence of nucleotide sequences of the dsrB gene was detected by PCR in the water samples from all hydrochemical layers, including subsurface oxic waters. The presence of the 16S rRNA genes of representatives of Desulfotomaculum, Desulfococcus-Desulfonema-Desulfosarcina, and Desulfovibrio-Desulfomicrobium SRB subgroups was also revealed throughout the water column of the Gdansk Deep. Analysis of translated amino acid sequences encoded by the dsrB gene demonstrated the highest homology with the relevant sequences of uncultured SRB from various marine habitats.

Relation between Sulfate Reducing Bacterium and Environmental Factors in Petroleum Hydrocarbon-Contaminated Site

The sulfate reducing bacterium (SRB) diversity of all samples were analyzed by PCR-DGGE technology and the Shannon-Wiener indexes (1.004-3.665) of all the samples were calculated. The results showed that the abundances, advantages and evenness of the sulfate-reducing bacterial community of all the samples showed certain change regularity along the pollution halo direction. The results of CCA analysis showed that the distribution and structure of sulfate reducing bacterium communities showed obvious correlation with the concentration of TPH, DO and other environmental factors.

Activity and structure of the sulfate-reducing bacterial community in the sediments of the southern part of Lake Baikal

The rates of sulfate reduction (SR) and the diversity of sulfate-reducing bacteria (SRB) were studied in the sediments of the Posol'skaya banka elevation in the southern part of Lake Baikal. SR rates varied from 1.2 to 1641 nmol/(dm3 day), with high rates (> 600 nmol/(dm3 day)) observed at both deep-water stations and in subsurface silts. Integral SR rates calculated for the uppermost 50 cm of the sediments were higher for gas-saturated and gas hydrate-bearing sediments than in those with low methane content. Enrichment SRB cultures were obtained in Widdel medium for freshwater SRB. Analysis of the 16S rRNA gene fragments from clone libraries obtained from the enrichments revealed the presence of SRB belonged to Desulfosporosinus genus, with D. lacus as the most closely related member (capable of sulfate, sulfite, and thiosulfate reduction), as well as members of the order Clostridiales.

The sulfate-reducing bacterial community of sulfide-rich water of the Ust’-Kachka resort spring, Perm Krai, Russia

Microbiological investigation of the highly mineralized, sulfide-rich cold spring of the Ust’-Kachka resort was carried out. The total number and biomass of microbial cells were 50 × 103 cell/mL and 15 μg/L, respectively. While the total microbial number was low, the sulfate reduction rate determined by the radioisotope method was relatively high (0.575 mg S L−1 day−1). An enrichment culture was obtained on Widdel medium. According to the results of cloning the 16S rRNA gene fragments with subsequent restriction analysis, the dominant organisms were group 6 sulfate-reducing bacteria (Desulfovibrio-Desulfomicrobium) and the microorganism exhibiting 99% similarity to the anaerobic haloalkaliphilic bacterium Halanaerobium hydrogeniformans.

Genus-specific and phase-dependent effects of nitrate on a sulfate-reducing bacterial community as revealed by dsrB-based DGGE analyses of wastewater reactors

The biogenic production of hydrogen sulfide is a serious problem associated with wastewater treatment. The aim of this study was to investigate the inhibitory effect of nitrate on the dynamics of sulfate-reducing bacteria (SRB) community in a laboratory-scale wastewater reactor, originating from a denitrifying plant using activated sludge. For this purpose, denaturing gradient gel electrophoresis (DGGE) analysis targeting the dsrB (dissimilatory sulfite reductase) gene was used in combination with chemical analyses and measurement of oxidation and reduction potential (ORP). The reactors were initially dosed with 1.0 and 4.0g/L potassium nitrate and anaerobically incubated for 490h. Addition of 4.0g/L nitrate to the reactor was associated with a prolonged inhibition (over 300h, i.e., 12.5days) of sulfate reduction and this was consistent with a rapid decrease in ORP associated with nitrate depletion. The DGGE analysis revealed that nitrate addition remarkably attenuated a distinct group of dsrB related to Desulfovibrio, whereas other dsrB groups were not influenced. Furthermore, another sulfate reduction by Syntrophobacter in the later stages of the incubation period occurred in both reactors (regardless of the nitrate concentration), suggesting that different SRB groups are associated with sulfate reduction at different stages of the wastewater treatment process.

Investigation of the sulfate-reducing bacterial community in the aerobic water and chemocline zone of the Black Sea by the fish technique

Fluorescent in situ hybridization (FISH) was used to analyze the abundance and phylogenetic composition of sulfate-reducing bacteria in the aerobic waters and in the oxic/anoxic transitional zone (chemocline) of the Black Sea, where biogenic formation of reduced sulfur compounds was detected by radioisotope techniques. Numerous sulfate-reducing bacteria of the genera Desulfotomaculum (30.5% of detected bacterial cells), Desulfovibrio (29.6%), and Desulfobacter (6.7%) were revealed in the aerobic zone at a depth of 30 m, while Desulfomicrobium-related bacteria (33.5%) were prevalent in the upper chemocline waters at 150-m depth. Active cells of sulfate-reducing bacteria were much more abundant in the samples collected in summer than in the winter samples from the deep-sea zone. The presence of physiologically active sulfate reducers in oxic and chemocline waters of the Black Sea correlates with the hydrochemical data on the presence of reduced sulfur compounds in the aerobic water column.

Sulfate reduction, molecular diversity, and copper amendment effects in bacterial communities enriched from sediments exposed to copper mining residues

Sulfate-reducing bacterial communities from coastal sediments with a long-term exposure to copper (Cu)-mining residues were studied in lactate enrichments. The toxicity of excess copper may affect sulfate-reducing bacterial communities. Sulfate reduction was monitored by sulfate and organic acid measurements. Molecular diversity was analyzed by 16S rRNA, dissimilatory sulfate reduction dsrAB, and Cu translocating phospho-type adenosine triphosphatases (P-ATPases) cop-like gene sequence profiling. The influence of Cu amendment was tested in these enrichments. Results showed fast sulfate reduction mostly coupled to incomplete organic carbon oxidation and partial sulfate reduction inhibition due to copper amendment. The 16S rRNA clonal libraries analysis indicated that delta- and gamma-Proteobacteria and Cytophaga-Flexibacter-Bacteroides dominated the enrichments. The dsrAB libraries revealed the presence of Desulfobacteraceae and Desulfovibrionaceae families-related sequences. Copper produced significant shifts (i.e., a decrease in the relative abundance of sulfate-reducing microorganisms) in the enriched bacterial community structure as determined by terminal-restriction fragment length polymorphism (T-RFLP) profiling and multivariate analyses. Clonal libraries of cop-like sequences showed low richness in the enriched microbial communities, and a strong effect of copper on its relative abundance. Novel Cu-P(IB)-ATPase sequences encoding Cu resistance were detected. The present study indicates that Cu does not significantly affect sulfate reduction and genetic diversity of taxonomic and dissimilatory sulfate-reduction molecular markers. However, the diversity of Cu resistance genetic determinants was strongly modified by this toxic metal.
