Membrane Capacitance Measurements of Stimulus-Evoked Exocytosis in Adrenal Chromaffin Cells.

Research using membrane capacitance (Cm) measurements in adrenal chromaffin cells has transformed our understanding of the molecular mechanisms controlling regulated exocytosis. This is in part due to the exquisite temporal resolution of the technique, and the possibility of combining quantification of exo-/endocytosis at the whole-cell level, with the ability to simultaneously monitor and control the calcium signals triggering vesicle fusion. In this regard, experiments performed with Cm measurements complement amperometry experiments that give a measure of secreted transmitter and the behavior of the fusion pore, and fluorescent microscopy studies used to monitor vesicle and protein dynamics in imaged regions of the cell. In this chapter, we provide a detailed account of the methodology used to perform whole-cell patch clamp measurements of Cm in combination with voltage-clamp recordings of voltage-gated calcium channels to quantify stimulus-secretion coupling in chromaffin cells. Stimulus protocols developed for investigation of functionally distinct releasable vesicle pools are also described.

Permissive Modulation of Sphingosine-1-Phosphate-Enhanced Intracellular Calcium on BKCa Channel of Chromaffin Cells.

Sphingosine-1-phosphate (S1P), is a signaling sphingolipid which acts as a bioactive lipid mediator. We assessed whether S1P had multiplex effects in regulating the large-conductance Ca2+-activated K+ channel (BKCa) in catecholamine-secreting chromaffin cells. Using multiple patch-clamp modes, Ca2+ imaging, and computational modeling, we evaluated the effects of S1P on the Ca2+-activated K+ currents (IK(Ca)) in bovine adrenal chromaffin cells and in a pheochromocytoma cell line (PC12). In outside-out patches, the open probability of BKCa channel was reduced with a mean-closed time increment, but without a conductance change in response to a low-concentration S1P (1 µM). The intracellular Ca2+ concentration (Cai) was elevated in response to a high-dose (10 µM) but not low-dose of S1P. The single-channel activity of BKCa was also enhanced by S1P (10 µM) in the cell-attached recording of chromaffin cells. In the whole-cell voltage-clamp, a low-dose S1P (1 µM) suppressed IK(Ca), whereas a high-dose S1P (10 µM) produced a biphasic response in the amplitude of IK(Ca), i.e., an initial decrease followed by a sustained increase. The S1P-induced IK(Ca) enhancement was abolished by BAPTA. Current-clamp studies showed that S1P (1 µM) increased the action potential (AP) firing. Simulation data revealed that the decreased BKCa conductance leads to increased AP firings in a modeling chromaffin cell. Over a similar dosage range, S1P (1 µM) inhibited IK(Ca) and the permissive role of S1P on the BKCa activity was also effectively observed in the PC12 cell system. The S1P-mediated IK(Ca) stimulation may result from the elevated Cai, whereas the inhibition of BKCa activity by S1P appears to be direct. By the differentiated tailoring BKCa channel function, S1P can modulate stimulus-secretion coupling in chromaffin cells.

The Adrenal Medulla Modulates Mechanical Allodynia in a Rat Model of Neuropathic Pain.

We have investigated whether the stress response mediated by the adrenal medulla in rats subjected to chronic constriction injury of the sciatic nerve (CCI) modulates their nocifensive behavior. Treatment with SK29661 (300 mg/kg; intraperitoneal (I.P.)), a selective inhibitor of phenylethanolamine N-methyltransferase (PNMT) that converts noradrenaline (NA) into adrenaline (A), fully reverted mechanical allodynia in the injured hind paw without affecting mechanical sensitivity in the contralateral paw. The effect was fast and reversible and was associated with a decrease in the A to NA ratio (A/NA) in the adrenal gland and circulating blood, an A/NA that was elevated by CCI. 1,2,3,4-tetrahydroisoquinoline-7-sulfonamide (SKF29661) did not affect exocytosis evoked by Ca2+ entry as well as major ionic conductances (voltage-gated Na+, Ca2+, and K+ channels, nicotinic acetylcholine receptors) involved in stimulus-secretion coupling in chromaffin cells, suggesting that it acted by changing the relative content of the two adrenal catecholamines. Denervation of the adrenal medulla by surgical splanchnectomy attenuated mechanical allodynia in neuropathic animals, hence confirming the involvement of the adrenal medulla in the pathophysiology of the CCI model. Inhibition of PNMT appears to be an effective and probably safe way to modulate adrenal medulla activity and, in turn, to alleviate pain secondary to the injury of a peripheral nerve.

Mechanism of the inhibitory effect of thiopentone on stimulus-secretion coupling in chromaffin cells

This study was undertaken to clarify the mechanism of action of thiopentone on stimulus-secretion coupling in cholinergic postsynaptic cells using cultured bovine adrenal chromaffin cells. Thiopentone (20-100 microns) inhibited carbachol-induced Ca2+ uptake into, and catecholamine release from the cells in a concentration-dependent manner. The inhibition of catecholamine release was almost parallel to the inhibition of Ca2+ uptake. The inhibitory effect of thiopentone was not overcome by an increase in the concentration of carbachol, whereas that of alcuronium was. Contrary to the inhibition by diltiazem, which was reversed by increasing the Ca2+ concentration, the inhibition by thiopentone was not overcome by a high Ca2+ concentration. Compared with the inhibition of carbachol-induced catecholamine release, a higher concentration of thiopentone was required to inhibit high K+ (56 mM KCl)-induced catecholamine release (200 microns thiopentone exhibited 32% inhibition). The results suggest that thiopentone blocks the stimulus-secretion coupling in the chromaffin cells as a result of inhibiting Ca2+ uptake through nicotinic receptor-linked channels. The linkage between receptor stimulation and Ca2+ channel activation seems to be the process most susceptible to inhibition by the barbiturate.

Linopirdine Modulates Calcium Signaling and Stimulus-Secretion Coupling in Adrenal Chromaffin Cells by Targeting M-Type K+Channels and Nicotinic Acetylcholine Receptors

Adrenal chromaffin cells synthesize and release catecholamines and several other transmitters that play important physiological roles in the coordinated response to stress or danger. The main trigger for secretion is acetylcholine (ACh) released from splanchnic nerve terminals that activates nicotinic ACh receptors (nAChRs) on the chromaffin cells, causing membrane depolarization and Ca2+ entry primarily through voltage-gated Ca2+ channels (Ca-channels). G protein-coupled receptors (GPCRs) can also trigger secretion, and it has been suggested that closure of M-type K+ channels might contribute to this process. However, GPCRs have multiple effects on calcium signaling and secretion, including release of intracellular Ca2+ stores, activation of second messenger pathways and kinases, and Ca2+ entry through store/receptor-operated channels. Hence, the effects of M-channel closure on [Ca2+]i signaling and transmitter release remain unclear. We have investigated the effects of linopirdine, a relatively selective blocker of M-channels, on stimulus-secretion coupling in chromaffin cells. Linopirdine produced a small increase in [Ca2+]i in approximately 63% of cells because of influx through Ca-channels. However, this was not sufficient to promote catecholamine release. We also show that linopirdine reduced cholinergic-stimulated increases in [Ca2+]i and secretion, primarily through potent block of nAChRs and a subtle effect on Ca2+ entry via Ca-channels. Hence, our data support the hypothesis that M-channels help control the excitability of chromaffin cells, but additional pathways need to be recruited by GPCRs to trigger catecholamine release. Furthermore, linopirdine potently targets nAChRs to modulate stimulus-secretion coupling in adrenal chromaffin cells.

Destabilization of actin filaments as a requirement for the secretion of catecholamines from permeabilized chromaffin cells

In the search for a functional role of cytoskeletal proteins in the mechanism(s) of stimulus-secretion coupling, we have previously demonstrated that the actomyosin system might be involved in the transport of cations across the plasma membrane of bovine adrenal chromaffin cells [(1986) J. Biol. Chem. 261, 5745-5750]. To establish whether actin and myosin might also be involved in later stages of the cellular response, we have examined the possible effects of various actin-specific reagents on the calcium-mediated secretion of catecholamines from digitonin-permeabilized cells. F-Actin-destabilizing agents, such as cytochalasin D or DNase 1, were found to promote Ca 2+-stimulated (as well as basal) secretion. By contrast, stabilizers, like phalloidin, produced the opposite effect. It is concluded that stimulus-secretion coupling in chromaffin cells might require the reorganization of actin for modulating both ion transport across the plasma membrane and exocytotic secretion per se.

Control of stimulus-secretion coupling in adrenal medullary chromaffin cells by microfilament-specific macromolecules.

We have incorporated the myosin fragment heavy meromyosin (HMM), which is known to interact mechanochemically and enzymatically with actin filaments, into intact chromaffin cells of the bovine adrenal medulla, in order to study the possible involvement of actin and myosin in stimulus-secretion coupling. HMM was found to stimulate secretion of catecholamines, to cause depolarization of the plasma membrane, and to enhance 22Na+ uptake. HMM-stimulated catecholamine secretion was dependent on the presence of extracellular Na+. The Na+ uptake caused by HMM was inhibited by 10 microM amiloride. Acetylcholine-stimulated catecholamine secretion and 22Na+ uptake were both enhanced by HMM incorporation. A Na+/H+ antiporter, activated by the interaction of HMM with the cells' microfilaments, seems to be involved in HMM action and could possibly also be a component of stimulus-secretion coupling in chromaffin cells, induced by regular agonists.

The possible implication of membrane-associated actin in stimulus-secretion coupling in adrenal chromaffin cells

Chromaffin cells of the bovine adrenal medulla were fused with liposomes containing DNAaseI. Resting membrane potential measurements, obtained by the use of the cyanine dye diS-C 3 (5), showed that DNAaseI incorporation causes depolarization from −56 to −31 mV, which is similar to that induced by ouabain. The level of basal secretion which occurs after the introduction of DNAaseI is increased several fold. Actin filaments, which can be depolymerized by DNAaseI, thus appear to be involved in stimulus-secretion coupling in chromaffin cells. It is suggested that plasma membrane-associated microfilaments control the Na +-K +-pump and/or the permeability of Ca 2+ ions thus affecting the membrane potential as well as secretion.
