Platelet-rich plasma and ovarian quiescence: a bovine in vitro model for regeneration of the ovary.

Ovarian quiescence can be due to hormonal deficiency usually caused by apoptosis of granulosa cells responsible for oestrogen synthesis. This study evaluated the regenerative effect of platelet rich plasma (PRP) on bovine in vitro models to understand its effect on granulosa cells. Quiescent and healthy ovarian sections were cultured in the presence/absence of PRP for 72h and, at different times (0, 24, 48 and 72h), hematoxylin-eosin and immunohistochemical detection of Ki-67 were performed. Additionally, granulosa cells collected from healthy bovine ovaries were stressed with 100ng/mL of lipopolysaccharide (LPS) in presence/absence of PRP and evaluated at 0, 4, 8 and 24h for apoptosis by acridine orange and propidium iodide staining. Enzyme-linked immunosorbent assay tests were performed to evaluate oestrogen (E2) and anti-Müllerian hormone (AMH) concentrations on cultures of ovarian slices and granulosa cells. In slides of quiescent ovaries treated with PRP, a marked and widespread positivity to Ki-67 was expressed by 40-60% of the follicular wall cells at 48h of culture. Levels of E2 and AMH were significantly higher compared to untreated quiescent samples reaching the levels of healthy control samples. PRP counteracted the LPS effect and apoptosis (at 24h, there were 93.44±3.51% live cells with LPS+PRP compared to 37±1.32% with LPS) and significantly increased concentrations of E2 and AMH. PRP can stimulate granulosa cell proliferation and counteract inflammatory processes in vitro . This treatment could improve the reproductive ability of quiescent females.

Human BM-MSC secretome enhances human granulosa cell proliferation and steroidogenesis and restores ovarian function in primary ovarian insufficiency mouse model

Primary ovarian insufficiency (POI) is defined as the loss of ovarian function before 40 years of age. It clinically manifests as amenorrhea, infertility, and signs of estrogen insufficiency. POI is frequently induced by chemotherapy. Gonadotoxic chemotherapy reagents damage granulosa cells, which are essential for follicular function and development. Our recently published studies demonstrated that intraovarian transplantation of human mesenchymal stem cells (hMSCs) can restore fertility in a chemotherapy-induced POI mouse model. However, the regenerative mechanism underlying the hMSC effect in POI mice is not fully understood. Here, we report that the hMSC secretome increased the proliferation of human granulosa cells (HGrC1). We showed by FACS analysis that treatment of HGrC1 cells with hMSC-conditioned media (hMSC CM) stimulates cellular proliferation. We also demonstrated that the expression of steroidogenic enzymes involved in the production of estrogen, CYP19A1 and StAR, are significantly elevated in hMSC CM-treated HGrC1 cells. Our data suggest that hMSC CM stimulates granulosa cell proliferation and function, which may explain the therapeutic effect of hMSCs in our chemotherapy-induced POI animal model. Our findings indicate that the hMSC secretome may be a novel treatment approach for restoring granulosa cell and ovarian function in patients with POI.

The optimized research of the in vitro culture of preantral follicles in mice.

ObjectiveEfficiency of preantral follicle culture in vitro is low and is dependent on species, development stage, and follicle‐stimulating hormone (FSH) concentration. Here, we optimized the preantral follicle in vitro culture system in mice.MethodsThe primary follicles (PM follicles, 80‐100 μm diameter ) and early secondary follicles (ES follicles, 110‐130 μm diameter) isolated from 14‐day female mice were cultured in mediums containing 10 mIU/mL or 100 mIU/mL r‐FSH. The follicle growth and oocyte maturation were observed. Estradiol (E2) was detected by ELISA. FSH receptor (FSHR), Ki‐67, 3β‐HSD, CYP17, and CYP19 levels were detected by immunofluorescence and Western blot.ResultsThe antrum formation and oocyte maturation rates of ES follicles were significantly higher than those of PM follicles (P < .05). They were also significantly higher in ES follicles with 100 mIU/mL r‐FSH than with 10 mIU/mL r‐FSH (P < .05). A higher FSHR level was found in ES follicles. Meanwhile, with 10 mIU/mL r‐FSH, the ES follicles exhibited a pattern of flat growth, whereas a pattern of stereoscopic spatial growth was observed with 100 mIU/mL r‐FSH. The 100 mIU/mL r‐FSH stimulated granulosa cell proliferation more significantly than 10 mIU/mL r‐FSH. Moreover, FSH significantly promoted ES follicle granulosa cell proliferation compared to PM follicular granulosa cells. The secretion of E2 and the expressions of 3β‐HSD, CYP 17, and CYP 19 in ES follicles with 100 mIU/mL r‐FSH were significantly higher than those with 10 mIU/mL r‐FSH.ConclusionsThe 100 mIU/mL r‐FSH ideally promotes the development of ES follicles, whose growth pattern can more reasonably simulate the growth of follicles in vivo.

Follicle-stimulating hormone (FSHβ) gene polymorphisms and associations with reproductive traits in Rex rabbits

Follicle-stimulating hormone (FSH) stimulates granulosa cell proliferation and controls the development and maturation of oocytes. In this study with Rex rabbit, there was exploration of the relationships between reproductive traits and SNPs and haplotypes of FSHβ, and tested whether FSHβ SNPs would segregate between two lines, using 70 females from the White line and 100 females from the Beaver line. Three SNPs (FSH2, FSH30, FSH31) in exon1 and exon3 of FSHβ were strongly associated with reproductive traits in the combined population. For FSH2, the GG variant was associated with greater (P < 0.05) values for total number born (TNB) and number born alive (NBA), compared to those with the TT variant. For FSH30, for the AA variant there was greater (P < 0.05) values for TNB and NBA than with the GG and AG variants. For FSH31, the GG variant was associated with greater (P < 0.05) values for TNB, litter weight at birth (LWB) and litter size at 21 days of age (LS21), than the AG variant, and with greater (P < 0.01) values for NBA than the AG variant. When analyzed separately, FSH2 SNP was associated with LW21 in the Beaver line (P < 0.05), whereas FSH30 and FSH31 SNPs were associated with TNB and NBA in the White line (P < 0.05). It is concluded that genetic variation in FSHβ gene is associated with reproductive traits in the Rex rabbit, therefore, the FSH2, FSH30 and FSH31 SNPs can be used as molecular markers in genetic selection of rabbits.

Beta defensin 3 enhances ovarian granulosa cell proliferation and migration via ERK1/2 pathway in vitro†.

Antimicrobial peptides (AMPs) are regarded as host defense peptides that possess bactericidal activity as well as immunomodulatory function. However, the role of AMP in the mammalian ovary is unknown. In the present study, porcine granulosa cells were utilized in a cell model to study the role of porcine beta defensin 2 (pBD2; pDEFB4B) and 3 (pBD3; pDEFB103A) during ovarian follicular development. Granulosa cells were cultured in the absence and presence of 1, 10, and 50μg/ml of pDEFB4B or pDEFB103A. After 24 h of treatment, pDEFB103A but not pDEFB4B stimulated granulosa cell proliferation in a concentration-dependent manner (P<0.05). This effect was dependent on the stage of follicular development. In addition, transwell cell migration assay showed that in the presence of pDEFB103A (10μg/ml), a 2.5-fold increase in cell migration was achieved. Furthermore, further study revealed that pDEFB103A increased the mRNA levels of cyclin D1 (CCND1) and proliferating cell nuclear antigen (PCNA), both associated with cell proliferation. To study the potential pathway involved in pDEFB103A-induced cell proliferation and migration, western blots were performed. It was found that pDEFB103A significantly increased the phosphorylated-ERK1/2 to nonphosphorylated ratio. Moreover, pretreatment with the U0126, a specific ERK1/2 phosphorylation inhibitor, suppressed PDEFB103A inducing GCs ERK1/2 phosphorylation, as well as proliferation and migration, suggesting that PDEFB103A may act via activating the ERK1/2 pathway. Furthermore, using a signal transduction pathway Elk-1 trans-reporting system, the activation of ERK1/2 pathway by PDEFB103A was further confirmed. Our data suggest that AMP may play a physiological role in the mammalian ovary.

Persistent endocrine-disrupting chemicals found in human follicular fluid stimulate the proliferation of granulosa tumor spheroids via GPR30 and IGF1R but not via the classic estrogen receptors

Epidemiological studies have found that women have detectable levels of organic pollutants such as hexachlorobenzene (HCB), 2,2-dichlorodiphenyldichloroethylene (p,p’-DDE), polychlorinated biphenyl 153 (PCB153), perfluorooctanoate (PFOA), and perfluorooctane sulfonate (PFOS) in their follicular fluid. Thus, these compounds may directly affect the function of granulosa cells within the ovary and may promote granulosa cell tumor (GCT) progression. Two human GCT cell lines, COV434 and KGN, have been used as in vitro model systems to represent juvenile (JGCT) and adult (AGCT) GCT subtypes, respectively. In this study, we found that basal expression of estrogen receptor 1 (ESR1), estrogen receptor 2 (ESR2), and insulin-like growth factor 1 receptor (IGF1R) was higher in the AGCT subtype than in the JGCT subtype. All of the compounds acted as mitogenic factors at low nanomolar concentrations in the JGCT and AGCT forms of GCT. Interestingly, PFOA, PFOS, and HCB stimulated cell proliferation through IGF1R, whereas p,p’-DDE acted through GPR30. Moreover, a mixture of the five compounds also significantly stimulated granulosa cell proliferation; however, the observed effect was lower than predicted. Interestingly, the proliferative effect of a mixture of these compounds was dependent on IGF1R and GPR30 but independent of the classic estrogen receptors. Taken together, our results demonstrate for the first time that mixtures of persistent organic pollutants present in follicular fluids may induce granulosa tumor progression through IGF1R and GPR30 by acting as mitogenic factors in granulosa cells.

Resveratrol promotes in vitro activation of ovine primordial follicles by reducing DNA damage and enhancing granulosa cell proliferation via phosphatidylinositol 3-kinase pathway.

We aimed to study the effects of resveratrol on the morphology, DNA fragmentation, follicular activation and cell proliferation after in vitro culture of ovine ovarian tissue, and to verify if PI3K pathway is involved in resveratrol action in the sheep ovary. Ovaries were collected and divided into fragments. One fragment was fixed for histology (fresh control). The remaining fragments were cultured for 7days in control medium (α-MEM+ ) alone or with resveratrol (2, 10 or 30µM). After culture, ovarian tissue was destined to morphological analysis. TUNEL and proliferating cell nuclear antigen (PCNA) analyses were performed in the fresh control, α-MEM+ and 2µM resveratrol. Inhibition of PI3K activity was performed through pre-treatment with LY294002. The percentage of normal follicles was similar between α-MEM+ and 2µM resveratrol, and higher than those in other resveratrol treatments. An increase in follicular activation was observed in all treatments compared to fresh control. DNA fragmentation decreased in tissues cultured in 2µM resveratrol compared to α-MEM+ . Moreover, PCNA-positive cells were higher in 2µM resveratrol than in α-MEM+ . LY294002 inhibited follicular activation stimulated by α-MEM+ and 2µM resveratrol. In conclusion, 2µM resveratrol promotes primordial follicle activation compared to the fresh control by reducing DNA fragmentation and stimulating granulosa cell proliferation through activation of the PI3K pathway.

Transcriptome profiling to identify key mediators of granulosa cell proliferation upon FSH stimulation in the goose (Anser cygnoides)

ABSTRACT1. The low reproductive performance of geese has seriously hampered the development of the industry. Reproductive performance, particularly the egg laying rate mainly depends on the development of the follicle. Previous studies have shown that follicle-stimulating hormone (FSH) plays an important role in the process of follicular development, but the exact underlying mechanism remains unclear.2. This study showed that FSH stimulated granulosa cell proliferation in a dose-dependent manner. The effect of FSH treatment on granulosa cell proliferation was greatest at a dose of 100 mIU/ml FSH for 24 h.3. Secondly, the effect of different concentrations of FSH on goose granulosa cell proliferation was investigated, and de novo transcriptome assembly and gene expression analysis performed using short-read sequencing technology (Illumina). High-throughput sequencing results yielded 62.61 M reads and 7.8 G base pairs from granulosa cells treated with 100 mIU/ml FSH. These reads were assembled into 65,757 unigenes (mean length: 705 bp) with an N50 of 903 bp. A total of 110 upregulated and 510 downregulated differentially expressed genes (DEGs) were identified by RNA-seq.4. Functional analysis by gene ontology (GO) and KEGG pathway annotation indicated that hormone biosynthesis (GO:0042446), positive regulation of hormone secretion (GO:0046887), steroid biosynthesis, oxidative phosphorylation and carbon metabolism pathways were involved in FSH-mediated proliferation of goose granulosa cells.5. After screening, a group of key responsive genes including superoxide dismutase 1, fatty acyl-CoA reductase 1, transforming growth factor-beta receptor-associated protein 1 and follistatin were tested by real-time reverse transcription PCR to confirm differential expression in granulosa cells stimulated by FSH.6. FSH-stimulated goose granulosa cells and DEG profiling data provided comprehensive gene expression information at the transcriptional level that could promote better understanding of the molecular mechanisms underlying follicle development in response to FSH stimulation.

Effects of N-carbamylglutamate and L-arginine on steroidogenesis and gene expression in bovine granulosa cells

Feeding N-carbamylglutamate (NCG) and arginine (ARG) improves reproductive measures in pigs and reduces systemic steroid levels in pregnant ewes. We hypothesized that the effects of NCG and ARG on reproduction were due to direct effects on the ovary. Thus, the objectives of this study were to investigate the effects of NCG and ARG on granulosa cell (GC) steroidogenesis, gene expression, and cell proliferation in vitro. GC were collected from small (1–5mm) bovine follicles and treated in vitro with NCG or ARG in serum-free medium for 24h to 48h. Both NCG and ARG inhibited (P<0.05) IGF1- and FSH-induced GC estradiol production but only NCG inhibited (P<0.05) progesterone production. In contrast, NCG and ARG increased (P<0.05) GC numbers induced by IGF1 and FSH. NCG inhibited (P<0.05) StAR, CYP11A1 and CYP19A1 mRNA abundance in small-follicle GC, whereas ARG had no effect (P>0.10) on StAR, CYP11A1 or CYP19A1 mRNA abundance. We conclude that NCG and ARG may act directly on GC and therefore may regulate ovarian function by slowing follicular differentiation via inhibiting IGF1 action, and steroid synthesis while stimulating GC proliferation in cattle.

Stage-specific follicular extracellular vesicle uptake and regulation of bovine granulosa cell proliferation.

Follicular fluid within ovarian antral follicles contains numerous factors, which influence the development of a healthy oocyte including nucleic acids, steroids, proteins, and extracellular vesicles (EVs). Current evidence indicates that follicular EVs promote changes in cellular gene expression and support cumulus-oocyte complex expansion in vitro. In this study, we found EVs from different sized follicles differentially stimulate granulosa cell proliferation and this could be explained by both the differential contents associated, on or within the vesicles and by the preferential uptake of EVs dependent on follicle size from which they were isolated. Antibody array and inhibitor studies indicated that the Src, PI3K/Akt, and MAPK signaling pathways mediate the stimulatory effects of EVs on granulosa cell proliferation. This study demonstrates for the first time that EVs isolated from follicular fluid are capable of stimulating granulosa cell proliferation and that this stimulatory response is associated with the size of antral follicle from which the EVs originated. The study further also provides the first evidence that vesicles released by small antral follicles are preferentially taken up when compared to those isolated from large follicles, suggesting that vesicular surface proteins change during follicular maturation.

Simazine, a triazine herbicide, disrupts swine granulosa cell functions.

The triazine herbicide simazine is a pesticide commonly detected in surface and ground waters, although banned in most European countries since 2004. Concerns for humans and animal health result from its potential endocrine disrupting action, that can lead to reproductive disorders. The present in vitro study was undertaken to study simazine effects on swine granulosa cell function, namely cell viability, proliferation, steroidogenesis and NO production. Moreover, the ability of this substance to interfere with the angiogenetic process, a crucial event in reproductive function, was taken into account. Our data document that simazine treatment, at 0.1 or 10 μM concentration levels, stimulates granulosa cell proliferation and viability and impairs steroidogenesis, increasing in particular progesterone production. In addition, the in vitro angiogenesis bioassay revealed a significant simazine stimulatory effect on immortalized porcine Aortic Endothelial Cell proliferation. Collectively, these results show that simazine can display disruptive effects on ovarian cell functional parameters, possibly resulting in reproductive dysfunction. This hypothesis is also supported by the observed pro-angiogenetic properties of this herbicide, as already suggested for different endocrine disruptors.

Retinoic Acid Regulates Calcium Signaling to Promote Mouse Ovarian Granulosa Cell Proliferation.

Normal development of ovarian follicles is critical for female reproduction and endocrine function. We have identified retinoic acid (RA) and the RA-degrading enzyme CYP26B1 as regulators of ovarian follicle development and showed that RA and a CYP26 inhibitor stimulated ovarian granulosa cell proliferation. The mechanism underpinning RA-dependent proliferation, however, is not known. The current study was designed to examine the role of intracellular calcium (Ca2+) signaling in mediating the effects of RA on primary mouse granulosa cell proliferation. In single-cell Ca2+ imaging experiments, treatment of cultured granulosa cells with RA increased the steady-state Ca2+ content of the endoplasmic reticulum (ER) stores. This correlated with increased store-operated Ca2+ entry (SOCE) and enhanced inositol 1,4,5-trisphosphate receptor (IP3R)-dependent Ca2+ release. In proliferation assays, RA treatment or Cyp26b1 knockdown stimulated proliferation, whereas Cyp26b1 overexpression inhibited proliferation. When RA was given together with 2-aminoethoxydiphenylborane (2-APB), a blocker of IP3R-dependent ER Ca2+ release and SOCE, with xestospongin C, a selective IP3R- receptor antagonist, or with 3,5-bis (trifluoromethyl)pyrazole (BTP-2), a specific SOCE blocker, the stimulatory effect of RA on cell proliferation was abolished. Further investigation showed that treatment with 2-APB or BTP-2 inhibited RA induction of RA response element (RARE) activation in granulosa cells, confirming an important role for Ca2+ signaling in mediating RA actions. Overall, these data support a model in which RA regulates ovarian follicle development by stimulating granulosa cell proliferation and that this stimulatory effect is at least in part driven by the modulation of Ca2+ signaling.

Sphingosine-1-phosphate, regulated by FSH and VEGF, stimulates granulosa cell proliferation

Sphingosine-1-phosphate (S1P) is a bioactive polar sphingolipid which stimulates proliferation, growth and survival in various cell types. In the ovary S1P has been shown protect the granulosa cells and oocytes from insults such as oxidative stress and radiotherapy, and S1P concentrations are greater in healthy than atretic large follicles. Hence, we postulate that S1P is fundamental in follicle development and that it is activated in ovarian granulosa cells in response to FSH and VEGF. To test this hypothesis we set out: i) to evaluate the effect of FSH and VEGF on S1P synthesis in cultured bovine granulosa cells and ii) to analyse the effect of S1P on proliferation and survival of bovine granulosa cells in vitro. Seventy five thousand bovine granulosa cells from healthy medium-sized (4–7mm) follicles were cultured in 96-well plates in McCoy’s 5a medium containing 10ng/mL of insulin and 1ng/mL of LR-IGF-I at 37°C in a 5% CO2/air atmosphere at 37°C. Granulosa cell production of S1P was tested in response to treatment with FSH (0, 0.1, 1 and 10ng/mL) and VEGF (0, 0.01, 0.1, 1, 10 and 100ng/mL) and measured by HPLC. Granulosa cells produced S1P at 48 and 96h, with the maximum production observed with 1ng/mL of FSH. Likewise, 0.01ng/mL of VEGF stimulated S1P production at 48, but not 96h of culture. Further, the granulosa cell expression of sphingosine kinase-1 (SK1), responsible for S1P synthesis, was demonstrated by Western blot after 48h of culture. FSH increased the expression of phosphorylated SK1 (P<0.05) and the addition of a SK1 inhibitor reduced the constitutive and FSH-stimulated S1P synthesis (P<0.05). Sphingosine-1-phosphate had a biphasic effect on granulosa cell number after culture. At low concentration S1P (0.1μM) increased granulosa cell number after 48h of culture (P<0.05) and the proportion of cells in the G2 and M phase of the cell cycle (P<0.05), whereas higher concentrations decreased cell number (10μM; P<0.05) by an increase (P<0.05) in the proportion of cells in apoptosis (hypodiploid cells). In addition, treatment with SK-178 suppressed the FSH- and VEGF-stimulated rise of the granulosa cells number (P<0.05). Interestingly, the effect of 0.1μM S1P on granulosa cell number and their proportion in G2/M phases is similar to that observed with 1ng/mL FSH. The results of this study are the first to demonstrate sphingosine-1-phosphate (S1P) synthesis in granulosa cells under the control of FSH and VEGF. The later achieved through the regulation of sphingosine kinase 1 expression. This S1P augments the proportion of cells in the G2/M phase of the cell cycle that translates in increased granulosa cell proliferation.

Astragalin, a Flavonoid from Morus alba (Mulberry) Increases Endogenous Estrogen and Progesterone by Inhibiting Ovarian Granulosa Cell Apoptosis in an Aged Rat Model of Menopause.

Background: To determine the mechanism by which the flavonoid glycoside astragalin (AST) reduces ovarian failure in an aged rat model of menopause. Methods: The in vivo effect of AST on granulosa cell (GC) apoptosis in aged female rats was determined using flow cytometry. In vitro, the effects of AST on cultured GCs were investigated using the MTT proliferation assay and western blot assays. Results: Aged rats had significantly higher GC apoptosis as compared with young female rats. Treatment of aged rats with AST (all three doses; p < 0.01) or Progynova (p < 0.01) significantly reduced GC apoptosis as compared with the aged controls. The proportions of total apoptotic GCs was 25.70%, 86.65%, 47.04%, 27.02%, 42.09% and 56.42% in the normal, aged, 17β-estradiol (E2), high dose AST, medium dose AST, and low dose AST-treated groups, respectively. Significant increases of serum E2 and P4 levels, as well as altered levels of serum follicle stimulating hormone (FSH) and luteinizing hormone (LH) levels. In cultured rat GCs, AST stimulated GC proliferation, E2 and progesterone (P4) secretion, reduced apoptosis, reduced the level of the pro-apoptotic protein Bcl-2 (p < 0.01), but had no effect on BAX. Conclusions: AST enhanced ovarian function in aged female rats by increasing E2 and P4 levels, and reducing ovarian GC apoptosis via a mechanism involving Bcl-2. These data demonstrate a new pharmacological activity for AST, as well as a novel mechanism of action, and further suggest that AST may be a new therapeutic agent for the management of menopausal symptoms.

The isoflavone daidzein directly affects porcine ovarian cell functions and modifies the effect of follicle-stimulating hormone.

The key biological active molecule of soya is the isoflavone daidzein, which possesses phytoestrogenic activity. The direct effect of soya and daidzein on ovarian cell functions is not known. This study examined the effect of daidzein on basic porcine ovarian granulosa cell functions and the response to follicle-stimulating hormone (FSH). We studied the effects of daidzein (0, 1, 10 and 100μm), FSH (0, 0.01, 0.1, 1IU/ml) and combinations of FSH (0, 0.01, 0.1, 1IU/ml)+daidzein (50μm) on proliferation, apoptosis and hormone release from cultured porcine ovarian granulosa cells and ovarian follicles. The expression of a proliferation-related peptide (PCNA) and an apoptosis-related peptide (Bax) was analysed using immunocytochemistry. The release of progesterone (P4) and testosterone (T) was detected using EIA. Leptin output was analysed using RIA. Daidzein administration increased granulosa cell proliferation, apoptosis and T and leptin release but inhibited P4 output. Daidzein also increased T release and decreased P4 release from cultured ovarian follicles. Follicle-stimulating hormone stimulated granulosa cell proliferation, apoptosis and P4, T and leptin release. The addition of daidzein promoted FSH-stimulated apoptosis (but not proliferation) but suppressed FSH-stimulated P4, T and leptin release. Our observations of FSH action confirm previous data on the stimulatory effect of FSH on ovarian cell proliferation, apoptosis and steroidogenesis and demonstrate for the first time the involvement of FSH in the upregulation of ovarian leptin release. Our observations of daidzein effects demonstrated for the first time that this soya isoflavone affected basic ovarian cell functions (proliferation, apoptosis and hormones release) and modified the effects of FSH. Daidzein promoted FSH action on ovarian cell proliferation and apoptosis and suppressed, and even inverted, FSH action on hormone release. The direct action of daidzein on basic ovarian cell functions and the ability of these cells to respond to FSH indicate the potential influence of soya-containing diets on female reproductive processes via direct action on the ovary.

Fibroblast growth factors (FGFs) have been shown to alter growth and differentiation of reproductive tissues in a variety of species. Within the female reproductive tract, the effects of FGFs have been focused on the ovary, and the most studied one is FGF2, which stimulates granulosa cell proliferation and decreases differentiation (decreased steroidogenesis). Other FGFs have also been implicated in ovarian function, and this review summarizes the effects of members of two subfamilies on ovarian function; the FGF7 subfamily that also contains FGF10, and the FGF8 subfamily that also contains FGF18. There are data to suggest that FGF8 and FGF18 have distinct actions on granulosa cells, despite their apparent similar receptor binding properties. Studies of non-reproductive developmental biology also indicate that FGF8 is distinct from FGF18, and that FGF7 is also distinct from FGF10 despite similar receptor binding properties. In this review, the potential mechanisms of differential action of FGF7/FGF10 and FGF8/FGF18 during organogenesis will be reviewed and placed in the context of follicle development. A model is proposed in which FGF8 and FGF18 differentially activate receptors depending on the properties of the extracellular matrix in the follicle.

Signalling pathways involved in the synergistic effects of human growth differentiation factor 9 and bone morphogenetic protein 15.

Growth differentiation factor 9 (GDF9) and bone morphogenetic protein 15 (BMP15) act synergistically to regulate granulosa cell proliferation and steroid production in several species. Several non-Sma and mothers against decapentaplegic (SMAD) signalling pathways are involved in the action of murine and ovine GDF9 and BMP15 in combination, with the pathways utilised differing between the two species. The aims of this research were to determine if human GDF9 and BMP15 also act in a synergistic manner to stimulate granulosa cell proliferation and to identify which non-SMAD signalling pathways are activated. Human GDF9 with BMP15 (GDF9+BMP15) stimulated an increase in (3)H-thymidine incorporation (P<0.001), which was greater than the increase with BMP15 alone, while GDF9 alone had no effect. The stimulation of (3)H-thymidine incorporation by GDF9+BMP15 was reduced by the addition of inhibitors to the SMAD2/3, nuclear factor-KB (NF-KB) and c-Jun N-terminal kinase (JNK) signalling pathways. Inhibitors to the SMAD1/5/8, extracellular signal-regulated kinase mitogen-activated protein kinase (ERK-MAPK) or p38-MAPK pathways had no effect. The addition of the BMP receptor 2 (BMPR2) extracellular domain also inhibited stimulation of (3)H-thymidine incorporation by GDF9+BMP15. In conclusion, human GDF9 and BMP15 act synergistically to stimulate granulosa cell proliferation, a response that also involves species-specific non-SMAD signalling pathways.

Abstract A81: Estrogen-responsiveness of the TFIID subunit TAF4B and its potential function in ovarian cancer, epigenetic regulation and meiotic DNA repair

Abstract Female infertility affects approximately 10.9% of women ages 15-44 in the US, and the molecular mechanisms leading to this disorder are multifaceted and varied. We have previously demonstrated that the gonadal-enriched TFIID subunit TAF4B, a paralog of the general transcription factor TAF4A, is required for fertility in mice. Female mice deficient for TAF4B exhibit a phenotype resembling premature ovarian failure, including early oocyte loss, follicular atresia and severely reduced granulosa cell proliferation when treated with 17β-estradiol. The inability of estrogen to stimulate granulosa cell proliferation led us to hypothesize that TAF4B is involved in an estrogen signaling pathway within the ovary. A large percentage of Taf4b-knockout oocytes die by apoptosis immediately after birth, and this germ cell loss is attenuated by estrogen supplementation, further suggesting a connection between TAF4B and estrogen. Taf4b-knockout ovaries also display deregulated epigenetic marks, which can affect DNA repair during meiotic homologous recombination. Furthermore, estrogen is known to regulate epigenetic changes in the ovary, leading us to hypothesize that the estrogen rescue may occur via modulation of the epigenetic state and consequent repair of double-strand breaks during meiosis. Here, we show that Taf4b mRNA and TAF4B protein expression are upregulated by estrogens in whole ovaries and purified granulosa cells of the ovary and that this increase occurs via nuclear estrogen receptors. We observe significant increases of Taf4b mRNA in estrogen-exposed mouse ovarian tumors, and the mice exposed to estradiol had significantly diminished survival compared to those receiving a placebo pellet. Combined with the fact that epigenetic deregulation and DNA repair processes play a key role in tumorigenesis, these results suggest that in addition to fertility defects, TAF4B could also affect ovarian tumorigenesis later in life. Our preliminary data suggest that the loss of oocytes in Taf4b-knockout ovaries may occur due to deficiencies in epigenetic regulation and/or a deficiency in DNA repair during meiotic prophase, since DNA repair and meiosis related genes are significantly reduced in Taf4b-knockout ovaries. Future studies will determine if estrogen treatment of neonatal Taf4b-knockout ovaries ameliorates these epigenetic and DNA repair deficits, leading to the observed oocyte rescue, and will explore if ovarian tumorigenesis is altered in the absence of TAF4B. Activation of the PI3K/Akt/mTOR Pathway by Insulin Is Crucial for Amplification of FSH-Induced Differentiation, but Not Proliferation, of Ovarian Granulosa Cells.

Follicle stimulating hormone (FSH) controls both proliferation and differentiation of granulosa cells. FSH induced differentiation and in particular steroidogenesis are synergized by insulin. Whether insulin potentiates the proliferative effect of FSH is not known. Moreover, the crosstalk between insulin and FSH intracellular signaling pathways in granulosa cells has not been examined. The aim of this study was to investigate whether insulin affects granulosa cell proliferation and to explore the mechanisms that are cooperatively regulated by insulin and FSH during granulosa cell proliferation and differentiation. Primary granulosa cells obtained from estradiol-treated immature rats were cultured in defined medium in the absence of serum. Proliferation was stimulated by treatment with FSH and activin, both at a 50 ng/ml final concentration. As expected, FSH/activin treatments stimulated granulosa cell proliferation as determined by MTT assays. However, FSH/activin-induced proliferation was not affected by co-treatment with increasing doses of insulin. Moreover, insulin itself did not stimulate granulosa cell proliferation. On the other hand, stimulation of P450aromatase (Cyp19) and P450 side chain cleavage (P450scc) expression by FSH was potentiated by the presence of insulin in the culture medium. Interestingly, dose-response experiments revealed a characteristic bell-shaped curve in which concentrations of 1, 10 and 50 μg/ml of insulin progressively potentiated the effect FSH on Cyp19 and P450scc. However, concentrations higher than 100 μg/ml led to a decrease of Cyp19 and P450scc stimulation. A similar response was observed in the stimulation of liver receptor homolog-1 by FSH. In the following experiments, we aimed to determine the intracellular signaling pathway mediating insulin effects, utilizing a dose of 10 μg/ml of insulin. For this purpose, granulosa cells were pretreated with PI3k inhibitors (wortmannin and LY294002), a MAPK inhibitor (UO126), or a SRC kinase inhibitor (PP2) prior to stimulation with FSH plus insulin. The potentiating effect of insulin on the FSH-induced expression of Cyp19 and P450scc was prevented by the two PI3k inhibitors. However, these inhibitors did not affect FSH-induced stimulation of gene expression. In contrast, the MAPK inhibitor decreased the expression of Cyp19 and P450scc expression to values significantly lower than those found in the presence of FSH alone. This suggests that MAPK inhibition prevented FSH stimulation, precluding any analysis of the effects of this pathway on insulin action. Finally, treatment with a SRC kinase inhibitor had no effect on the super-induction of Cyp19 or P450scc expression observed in the presence of FSH and insulin. To confirm these findings and to explore the downstream targets of PI3k/Akt, granulosa cells were treated with rapamycin, an inhibitor of mTOR kinase which is activated by Akt. Similar to the results observed by inhibiting PI3K, rapamycin treatment prevented the insulin-induced potentiation of FSH actions. Taken together, this evidence indicates that the effects of insulin in granulosa cell steroidogenesis are not only dose-related but they also follow a bell-shaped dose-response curve. These results also demonstrate that activation of the PI3K/Akt/mTOR pathway by insulin is crucial for the amplification of FSH-induced differentiation, but not proliferation, of ovarian granulosa cells. Oocyte-thecal Cell Regulatory Loop in the Control of Preantral Follicle Development

Oocyte-somatic cell interaction plays a crucial role in preantral folliculogenesis. Although topical research has focused on oocyte-granulosa cell interaction over the past decade, an oocyte-thecal cell regulatory loop may also play an important role during the preantral stage. Formation of the thecal cell layer is a key event that occurs during preantral folliculogenesis. Granulosal factors (e.g. IGF-I and KL) appear to stimulate the recruitment of thecal cells from stromal cells. Oocyte-derived GDF9 appears to indirectly modulate thecal cell differentiation, perhaps through regulating the granulosa/IGF-I and KL expression. Theca-produced androgens stimulate granulosa cell proliferation and preantral follicle growth. GDF9 enhances preantral follicle growth by up-regulating thecal androgen production, suggesting that a threshold level of androgens derived from thecal cells is necessary for preantral follicle growth, and that the androgen production may be controlled by the oocyte-derived GDF9. The challenge ahead is not to only understand the precise nature of these interactions, and to elucidate how dysregulation in these interactions may lead to ovarian pathologies such as polycystic ovary syndrome and gonadotropin poor-responsiveness.
