Nitrooxylative Dearomatization Reaction of β‐Naphthols with Hypervalent Iodine Reagent

AbstractA catalyst‐free intermolecular dearomatization reaction of β‐naphthols with hypervalent‐iodine‐based nitrooxylating reagent is reported. Various nitrooxylated β‐naphthalenones bearing a quaternary carbon stereogenic center were obtained smoothly in good to excellent yields (up to 87 %) under mild conditions. This method features mild conditions, broad substrate scope and excellent chemoselectivity.

Ni‐Catalyzed Regio‐ and Enantioselective Homoallylic Coupling: Synthesis of Chiral Branched 1,5‐Dienes Featuring a Quaternary Stereogenic Center and Mechanistic Analysis

AbstractAsymmetric synthesis of small molecules comprising quaternary stereogenic carbon centers represents a challenging objective. Here regio‐ and enantioselective synthesis of chiral 1,5‐dienes featuring quaternary stereocenters is reported via nickel‐promoted by reductive homoallylic coupling. The developed methodology features an atypical preference for the formation of unusual branched regioisomers (rr >20 : 1) in a sterically challenging allylic substitution event and furnishes the products with enantiomeric ratios of up to 98 : 2 and with high chemo‐ and E‐selectivity. A range of experimental evidences suggest that zinc plays a dual role to generate electrophilic and nucleophilic Ni(II)‐allyl intermediates empowering a unique formal bimetallic cross‐electrophile manifold in two separate kinetic regimes.

Asymmetric redox benzylation of enals enabled by NHC/Ru cooperative catalysis

The development of general methods for asymmetric benzylation of prochiral carbon nucleophiles remains a challenge in organic synthesis. The merging of ruthenium catalysis and N-heterocyclic carbene (NHC) catalysis for asymmetric redox benzylation of enals has been achieved, which opens up strategic opportunities for the asymmetric benzylation reactions. A wide range of 3,3'-disubstituted oxindoles with a stereogenic quaternary carbon center widely existing in natural products and biologically interesting molecules is successfully obtained with excellent enantioselectivities [up to 99% enantiomeric excess (ee)]. The generality of this catalytic strategy was further highlighted by its successful application in the late-stage functionalization of oxindole skeletons. Furthermore, the linear correlation between ee values of NHC precatalyst and the product elucidated the independent catalytic cycle of either the NHC catalyst or the ruthenium complex.

Interplay of diruthenium catalyst in controlling enantioselective propargylic substitution reactions with visible light-generated alkyl radicals

Transition metal-catalyzed enantioselective free radical substitution reactions have recently attracted attention as convenient and important building tools in synthetic chemistry, although construction of stereogenic carbon centers at the propargylic position of propargylic alcohols by reactions with free radicals remains unchallenged. Here we present a strategy to control enantioselective propargylic substitution reactions with alkyl radicals under photoredox conditions by applying dual photoredox and diruthenium catalytic system, where the photoredox catalyst generates alkyl radicals from 4-alkyl-1,4-dihydropyridines, and the diruthenium core with a chiral ligand traps propargylic alcohols and alkyl radicals to guide enantioselective alkylation at the propargylic position, leading to high yields of propargylic alkylated products containing a quaternary stereogenic carbon center at the propargylic position with a high enantioselectivity. The result described in this paper provides the successful example of transition metal-catalyzed enantioselective propargylic substitution reactions with free alkyl radicals.

Asymmetric Synthesis of a Quaternary Carbon Stereogenic Center by Organocatalysis Using a Primary Amino Acid and Its Salt.

In this personal account, our recent developments on the asymmetric synthesis of a quaternary carbon stereogenic center by organocatalysis using a primary amino acid and its salt as a catalyst are described in three chapters: (1) conjugate addition to nitroalkenes and vinyl ketones, (2) nucleophilic addition to π-allyl palladium complexes, and (3) nucleophilic substitution reactions with allyl and propargyl halides. By these methods, asymmetric α-allylation of α-branched aldehydes and ketones smoothly proceeded to give γ-nitroaldehydes, ketoaldehydes, α-allylated aldehydes, and α-allylated β-ketoesters possessing a quaternary carbon stereogenic center in good yields with high enantioselectivities.

Bisguanidinium-catalyzed formation of oxygen-containing quaternary stereogenic carbon centers

A bisguanidinium-catalyzed enantioconvergent SN2X reaction is described for the formation of C(sp3)–O bonds from racemic tertiary bromides and hydroxylamides/phenols, enabling efficient synthesis of bioactive compounds such as enantiopure bicalutamide and chiral fibrates.

Regio- and Diastereoselective Carbometalation Reaction of Cyclopropenes.

ConspectusThe various facets of the chemistry of cyclopropane derivatives, the smallest carbocycle, are amazingly diverse and continue to fascinate theoreticians, synthetic or structural chemists having interest in fundamental physical, medicinal chemistry, and natural product synthesis. The challenges generated by this intriguing cyclic arrangement of only three tetravalent carbons represent a wide area of the chemical spectrum. From fundamental aspects of bonding through the synthesis of highly strained molecules, the understanding of the mode of action in biological systems to the selective cleavage into acyclic substrates makes the chemistry of these small rings fascinating. Therefore, efficient routes to prepare differently polysubstituted cyclopropanes have always been of a primordial importance. In the past decade, we and others have expanded the scope of the carbometalation reaction of cyclopropenes as a broad and general method to the formation of stereodefined cyclopropane derivatives. Although cyclopropenes, with their even higher strain energy, easily undergo addition reactions of organometallic reagents, their carbometalation reactions generate new regio-, diastereo-, and enantioselectivity issues that needed to be addressed. These various stereochemical aspects accompanied our research from its origins to today, and we are proposing in this Account, a didactic overview of the different ways by which cyclopropenes can lead to the formation of polysubstituted cyclopropanes or open-products possessing several stereogenic centers as a single regio- and diastereomer.We initially launched our research campaign on the chemistry of these strained three-membered rings by the regio- and diastereoselective copper-catalyzed carbomagnesiation of enantiomerically enriched cyclopropenyl carbinols. The directing alcohol governed both the regio- and diastereoselectivity of the addition and also served as a good leaving group as it undergoes a selective 1,2-elimination reaction to provide enantioenriched alkylidenecyclopropanes in excellent yields and enantiomeric excesses. Then, we turned our attention to the regio- and stereoselective synthesis of stereodefined tri- and tetrasubstituted cyclopropanes through the diastereoselective addition to sp2- monosubstituted cyclopropenyl ester derivatives. With the aim to further expand this concept to the formation of penta- and hexa-substituted cyclopropanes as single isomer, we had first to design the preparation of the required 1,2-disubstituted cyclopropenes that would control the regioselective addition of the organometallic derivatives. The synthesis of penta- and hexa-substituted cyclopropanes was then reported for the first time as a single regio- and diastereomer. It should be noted that the in situ formed cyclopropyl-metal intermediate is configurationally stable and can be subsequently functionalized with pure retention of the configuration by addition of electrophiles. Then, the enantioselective-catalyzed carbometalation reaction of achiral cyclopropenes allowed the synthesis of several new classes of cyclopropane derivatives in high enantiomeric ratios. Finally, by combining the regio- and diastereoselective carbometalation reaction of a cyclopropene with a subsequent reaction of the resulting cyclopropylmetal species, a selective carbon–carbon bond cleavage was observed to lead to the preparation of acyclic substrates possessing several stereocenters including a quaternary carbon stereogenic center. Our original vision of using strain within an embedded double bond in a three-membered ring has provided new routes to the stereoselective synthesis of polysubstituted cyclopropanes and has been extremely successful, as it represents a current new tool for the synthesis of persubstituted cyclopropanes as a single diastereomer.

Synthetic and Computational Study of the Enantioselective [3+2]-Cycloaddition of Chromones with MBH Carbonates.

Highly efficient and straightforward access to enantioenriched five-membered ring-fused chromanones is developed via [3+2]-cycloaddition of 3-cyanochromones with Morita-Baylis-Hillman carbonates. Densely functionalized chiral cyclopenta[b]chromanones with three continuous quaternary and tertiary stereogenic carbon centers were obtained in high yields with high ee and dr (≤97% yield, 97% ee, and >20:1 dr). Moreover, density functional theory calculations have been carried out to investigate the mechanism and regio- and diastereoselectivity of the reaction.

Enantioselective Palladium-Catalyzed Arylborylation/Cyclization of Alkenes to Access Boryl-Functionalized Heterocyclic Compounds Containing Quaternary Stereogenic Centers.

Asymmetric palladium-catalyzed arylboration/cyclization of both nonactivated and activated alkenes with B2pin2 was developed. A wide range of N-allyl-o-iodobenzamides and o-iodoacryanilides reacted with B2pin2 to afford borylated 3,4-dihydroisoquinolinones and oxindoles, respectively, in high yields with high enantioselectivities. The synthetic utility of this enantioselective protocol was highlighted by synthesizing various chiral 3,4-dihydroisoquinolinone and oxindole derivatives containing quaternary stereogenic carbon centers, including enantioenriched Roche anticancer agent (S)-RO4999200.

Enantioselective Synthesis of α-Aryl-β-Aminocyclopropane Carboxylic Acid Derivatives via Rh(II)-Catalyzed Cyclopropanation of Vinylsulfonamides with α-Aryldiazoesters.

The reaction of vinylsulfonamides with donor-acceptor carbenes derived from α-aryldiazoesters, catalyzed by the tert-butyl glycine-derived dirhodium complex Rh2(S-4-Br-NTTL)4, has been reported. This method provides a variety of α-aryl-β-aminocyclopropane carboxylic acid derivatives bearing one quaternary carbon stereogenic center vicinal to the amino-substituted carbon in high yields with excellent diastereo- and enantioselectivities. Vinylsulfonamides showed complementary advantages over the well-developed vinylamides or vinylcarbamates for this Rh(II)-catalyzed cyclopropanation strategy. Moreover, these conformationally restricted α-aryl-β-aminocyclopropyl carboxylic acid derivatives can be readily incorporated into dipeptides.

BF3·OEt2-Catalyzed Unexpected Stereoselective Formation of 2,4-trans-Diallyl-2-methyl-6-aryltetrahydro-2H-pyrans with Quaternary Stereocenters.

The present manuscript describes a convenient, mild, and highly stereoselective method for the allylation of δ-hydroxy-α,β-unsaturated ketones having a benzylic hydroxyl group at the δ-position using allyltrimethylsilane mediated by BF3·OEt2, leading to 2,4-diallyl-2-methyl-6-aryltetrahydro-2H-pyran ring systems with quaternary carbon stereogenic centers. This represents the first example of a tandem isomerization followed by one C-O and two C-C bond-forming reactions in one pot. The isolation of TMS-protected lactol as an intermediate from the reaction strongly supports the proposed mechanistic pathway.

Rh(III)-catalyzed selective C7-H functionalization of indolines with 1,3-enynes enables access to six-membered 1,7-fused indolines

We described herein a Rh(III)-catalyzed C7-selective CH activation/annulation of indolines with 1,3-enynes to efficiently access various privileged 1,7-fused indolines bearing an all-carbon quaternary stereogenic carbon center. Notably, the resulting products can be readily transformed into 1,7-fused indoles, further widening the C-7 derivatization of indoles and highlighting the synthetic utility of this methodology.

Oxidative Indole Dearomatization for Asymmetric Furoindoline Synthesis by a Flavin-Dependent Monooxygenase Involved in the Biosynthesis of Bicyclic Thiopeptide Thiostrepton.

The interest in indole dearomatization, which serves as a useful tool in the total synthesis of related alkaloid natural products, has recently been renewed with the intention of developing new methods efficient in both yield and stereoselective control. Here, we report an enzymatic approach for the oxidative dearomatization of indoles in the asymmetric synthesis of a variety of furoindolines with a vicinal quaternary carbon stereogenic center. This approach depends on the activity of a flavin-dependent monooxygenase, TsrE, which is involved in the biosynthesis of bicyclic thiopeptide antibiotic thiostrepton. TsrE catalyzes 2,3-epoxidation and subsequent epoxide opening in a highly enantioselective manner during the conversion of 2-methyl-indole-3-acetic acid or 2-methyl-tryptophol to furoindoline, with up to >99 % conversion and >99 % ee under mild reaction conditions. Complementing current chemical methods for oxidative indole dearomatization, the TsrE activity-based approach enriches the toolbox in the asymmetric synthesis of products possessing a furoindoline skeleton.

Bismuth(iii)-catalyzed regioselective alkylation of tetrahydroquinolines and indolines towards the synthesis of bioactive core-biaryl oxindoles and CYP19 inhibitors.

Bismuth(iii)-catalyzed regioselective functionalization at the C-6 position of tetrahydroquinolines and the C-5 position of indolines has been demonstrated. For the first time, one pot symmetrical and unsymmetrical arylation of isatins with tetrahydroquinolines was accomplished giving a completely new product skeleton in good to excellent yields. Most importantly, this protocol leads to the formation of a highly strained quaternary carbon stereogenic center, which is a challenging task. Benzhydryl and 1-phenylethyl trichloroacetimidates have been used as the alkylating partners to functionalize the C-6 and C-5 positions of tetrahydroquinolines and indolines, respectively. The scope of the developed methodology has been extended for the synthesis of the bioactive CYP19-inhibitor and its analogue.

Dearomatization reaction of β-naphthols with disulfurating reagents.

p-TsOH-catalyzed intermolecular dearomatization reactions of β-naphthols with disulfurating reagents were developed. Various β-naphthalenones bearing a quaternary carbon stereogenic center were obtained smoothly in good to excellent yields with high chemoselectivity in the presence of 5 mol% p-TsOH. This reaction features mild reaction conditions and excellent functional group tolerance.

Asymmetric O-to-C Aryloxycarbonyl Migration of Indolyl Carbonates Using Single-Handed Dynamic Helical Polyquinoxalines Bearing 4-Aminopyridyl Groups as Chiral Nucleophilic Catalysts

Abstract Use of single-handed dynamic helical macromolecules as nucleophilic catalysts in asymmetric Steglich-type O-to-C aryloxycarbonyl rearrangement of 3-substituted indol-2-yl aryl carbonates is demonstrated. Among several single-handed poly(quinoxaline-2,3-diyl) copolymers (PQXap) bearing achiral 4-aminopyridin-3-yl groups at the 5-position of the quinoxaline rings, PQXmdpp and PQXapy, containing N-methylpyrrolidine-fused pyridin-3-yl and 4-(1-azetidinyl)pyridin-3-yl groups, respectively, showed higher enantioselectivity and catalytic activity than PQXdmap, bearing 4-dimethylaminopyridine-3-yl groups. Substrates bearing p-(trifluoromethyl)phenyloxycarbonyl groups on both the nitrogen and oxygen atoms showed high reactivity, giving oxindoles with a quaternary stereogenic carbon center at their 3-positions in up to 97:3 enantiomeric ratio in THF. The macromolecular catalysts underwent inversion of their helix sense by solvent effect, allowing the same catalyst to give the opposite enantiomer in a mixture of methyl t-butyl ether and 1,1,2-trichloroethane (3:1). The macromolecular catalysts could be easily recovered by adding acetonitrile to the reaction mixture and were reused four times without reduction in enantioselectivity.
