The Anopheles coluzzii range extends into Kenya: detection, insecticide resistance profiles and population genetic structure in relation to conspecific populations in West and Central Africa

BackgroundAnopheles coluzzii is a primary vector of malaria found in West and Central Africa, but its presence has hitherto never been documented in Kenya. A thorough understanding of vector bionomics is important as it enables the implementation of targeted and effective vector control interventions. Malaria vector surveillance efforts in the country have tended to focus on historically known primary vectors. The current study sought to determine the taxonomic status of samples collected from five different malaria epidemiological zones in Kenya as well as describe the population genetic structure and insecticide resistance profiles in relation to other An. coluzzii populations.MethodsMosquitoes were sampled as larvae from Busia, Kwale, Turkana, Kirinyaga and Kiambu counties, representing the range of malaria endemicities in Kenya, in 2019 and 2021 and emergent adults analysed using Whole Genome Sequencing (WGS) data processed in accordance with the Anopheles gambiae 1000 Genomes Project phase 3. Where available, historical samples from the same sites were included for WGS. Comparisons were made with An. coluzzii cohorts from West and Central Africa.ResultsThis study reports the detection of An. coluzzii for the first time in Kenya. The species was detected in Turkana County across all three time points from which samples were analyzed and its presence confirmed through taxonomic analysis. Additionally, there was a lack of strong population genetic differentiation between An. coluzzii from Kenya and those from the more northerly regions of West and Central Africa, suggesting they represent a connected extension to the known species range. Mutations associated with target-site resistance to DDT and pyrethroids and metabolic resistance to DDT were found at high frequencies up to 64%. The profile and frequencies of the variants observed were similar to An. coluzzii from West and Central Africa but the ace-1 mutation linked to organophosphate and carbamate resistance present in An. coluzzii from coastal West Africa was absent in Kenya.ConclusionsThese findings emphasize the need for the incorporation of genomics in comprehensive and routine vector surveillance to inform on the range of malaria vector species, and their insecticide resistance status to inform the choice of effective vector control approaches.

CRITICAL REVIEW OF INSECTICIDE RESISTANCE IN US AEDES ALBOPICTUS: RESISTANCE STATUS, UNDERLYING MECHANISMS, AND DIRECTIONS FOR FUTURE RESEARCH

Aedes albopictus is a primary or secondary disease vector in Asia that invaded the United States around 1980. It is now present in more than half of US states and continues to expand in range. The willingness to bite in the daytime and the ability to colonize makes this species a target of control operations both to prevent nuisance biting and for public health reasons. As with other species, effective long-term control requires an integrated management strategy and information about efficacy of operational interventions. Studies from Asia, where this species is a primary vector, show that insecticide resistance is a developing concern that can compromise effective control. In this review, we summarize the status of insecticide resistance in US populations of Ae. albopictus, examine the current understanding of the mechanisms underlying resistance, and offer suggestions for future research directions.

Pyrethroid resistance status and co-occurrence of V1016G, F1534C and S989P mutations in the Aedes aegypti population from two dengue outbreak counties along the China-Myanmar border

BackgroundOver the past two decades, dengue fever (DF) has emerged as a significant arboviral disease in Yunnan province, China, particularly in the China-Myanmar border area. Aedes aegypti, an invasive mosquito species, plays a crucial role in transmitting the dengue virus to the local population. Insecticide-based vector control has been the primary tool employed to combat DF, but the current susceptibility status of Ae. aegypti to commonly used insecticides is unknown. Assessment of Ae. aegypti resistance to pyrethroid insecticides and an understanding of the underlying mechanisms of this resistance in the China-Myanmar border region is of significant strategic importance for effectively controlling the DF epidemic in the area.MethodsAedes aegypti larvae collected from Ruili and Gengma counties in Yunnan Province were reared to adults in the laboratory and tested for susceptibility to three pyrethroid insecticides (3.20% permethrin, 0.08% lambda-cyhalothrin and 0.20% deltamethrin) by the standard WHO susceptibility bioassay. Genotyping of mutations in the knockdown gene (kdr), namely S989P, V1016G and F1534C, that are responsible for resistance to pyrethroid insecticides was performed using allele-specific PCR methods. A possible association between the observed resistant phenotype and mutations in the voltage-gated sodium channel gene (VGSC) was also studied.ResultsAedes aegypti mosquitoes collected from the two counties and reared in the laboratory were resistant to all of the pyrethroids tested, with the exception of Ae. aegypti from Gengma County, which showed sensitivity to 0.20% deltamethrin. The mortality rate of Ae. aegypti from Ruili county exposed to 3.20% permethrin did not differ significantly from that of Ae. aegypti from Gengma County (χ2 = 0.311, P = 0.577). By contrast, the mortality rate of Ae. aegypti from Ruili County exposed to 0.08% lambda-cyhalothrin and 0.20% deltamethrin, respectively, was significantly different from that of Ae. aegypti from Gengma. There was no significant difference in the observed KDT50 of Ae. aegypti from the two counties to various insecticides. Four mutation types and 12 genotypes were detected at three kdr mutation sites. Based on results from all tested Ae. aegypti, the V1016G mutation was the most prevalent kdr mutation (100% prevalence), followed by the S989P mutation (81.6%) and the F1534C mutation (78.9%). The constituent ratio of VGSC gene mutation types was significantly different in Ae. aegypti mosquitoes from Ruili and those Gengma. The triple mutant S989P + V1016G + F1534C was observed in 274 Ae. aegypti mosquitoes (60.8%), with the most common genotype being SP + GG + FC (31.4%). The prevalence of the F1534C mutation was significantly higher in resistant Ae. aegypti from Ruili (odds ratio [OR] 7.43; 95% confidence interval [CI] 1.71–32.29; P = 0.01) and Gengma (OR 9.29; 95% CI 3.38–25.50; P = 0.00) counties than in susceptible Ae. aegypti when exposed to 3.20% permethrin and 0.08% lambda-cyhalothrin, respectively. No significant association was observed in the triple mutation genotypes with the Ae. aegypti population exposed to 3.20% permethrin and 0.20% deltamethrin resistance (P > 0.05), except for Ae. aegypti from Gengma County when exposed to 0.08% lambda-cyhalothrin (OR 2.86; 95% CI 1.20–6.81; P = 0.02).ConclusionsAedes aegypti from Ruili and Gengma counties have developed resistance to various pyrethroid insecticides. The occurrence of multiple mutant sites in VGSC strongly correlated with the high levels of resistance to pyrethroids in the Ae. aegypti populations, highlighting the need for alternative strategies to manage the spread of resistance. A region-specific control strategy for dengue vectors needs to be implemented in the future based on the status of insecticide resistance and kdr mutations.Graphical

Nutritional Status, Sex, and Ambient Temperature Modulate the Wingbeat Frequency of the Diamondback Moth Plutella xylostella.

The diamondback moth, Plutella xylostella L. (Lepidoptera: Plutellidae), is a cosmopolitan horticultural pest that is undergoing a fast, climate-driven range expansion. Its wide geographic distribution, pest status, and high incidence of insecticide resistance are directly tied to long-distance migration. Wingbeat frequency (WBF) is a key aspect of P. xylostella migratory behavior, but has received limited scientific attention. Here, we investigated the effects of environmental parameters, age, adult nutrition, and sex on P. xylostella WBF. Across experimental regimes, WBF ranged from 31.39 Hz to 78.87 Hz. Over a 10-35 °C range, the WBF of both male and female moths increased with temperature up to 62.96 Hz. Though male WBF was unaffected by humidity, females exhibited the highest WBF at 15% relative humidity (RH). WBF was unaffected by adult age, but adult nutrition exerted important impacts. Specifically, the WBF of moths fed honey water (54.66 Hz) was higher than that of water-fed individuals (49.42 Hz). Lastly, males consistently exhibited a higher WBF than females. By uncovering the biological and (nutritional) ecological determinants of diamondback moth flight, our work provides invaluable guidance to radar-based monitoring, migration forecasting, and the targeted deployment of preventative mitigation tactics.

Insecticide resistance status of Aedes aegypti in border areas of Yunnan Province.

Aedes aegypti is a main vector of arboviral diseases, principally dengue, chikungunya, and Zika. Insecticides remain the most effective vector control method. Pyrethroid is the main insecticide currently used, and the long-term use of insecticides can cause mosquitoes to develop knockdown resistance. Studying the mutation sites and genotypes of Ae. aegypti can reveal the mutation characteristics and regional distribution of the kdr gene in an Ae. aegypti population. Testing for a correlation between the mutation rate in various populations and pyrethrin resistance can clarify the resistance mechanism. The bioassay results showed that all 15 populations are resistant. In the study of the kdr gene, three non-synonymous mutations were identified in the DNA of first generation females from the wild Ae. aegypti population: S989P (TCC-CCC), V1016G (GTA-GGA), and F1534C (TTC-TGC). The mortality rate of the various populations was correlated with the mutation rate at the V1016G + F1534C locus, but not the S989P + V1016G locus. Aedes aegypti populations in border regions of Yunnan Province are resistant to permethrin and beta-cyfluthrin. The insecticidal effect of beta-cyfluthrin is stronger than that of permethrin. The mutation rate at sites V1016G + F1534C is negatively correlated with the mortality of Ae. aegypti based on bioassays. © 2024 The Authors. RESISTANCE STATUS IN ANOPHELES GAMBIAE AND PLASMODIUM FALCIPARUM TRANSMISSION IN TORI-BOSSITO DISTRICT, SOUTHERN BENIN, WEST-AFRICA

In prelude to a large scale trial aiming to assess the field efficacy of Fludora Fusion 562.5WP-SB, entomologic baseline data were collected in 14 villages from Tori-Bossito District in Southern Benin during the rainy season from April to May 2018. Malaria vectors aggressiveness and longevity, Plasmodium falciparum infection in malaria vectors, malaria entomologic transmission as well as insecticide resistance status in malaria vectors were determinate. Overall 2695 mosquitoes were collected during 504 persons*nights. Among mosquitoes collected, 408 were Anopheles gambiae s.l. and 43 Anopheles funestus s.l. The proportion of parous malaria vectors was 54.2% and 79.1% for An. gambiae s.l. and An. funestus s.l. respectively. A sample of 68 malaria vectors was tested for Plasmodium falciparum infection using quantitative Polymlerase Chain Reaction (qPCR). The infection rate was about 6% (4/68 vectors infected) corresponding to 1.58 infective bites per person per month. The mortality rate of An. gambiae s.l. exposed to deltamethrin 0.05%, pyrimiphos-methyl 0.25% and clothianidin at 9µg/ml was 38%, 98.86% and 100% respectively, indicating that An. gambiae s.l. was resistant to deltamethrin and susceptible to pyrimiphos-methyl and clothianidin.This study showed that An. gambiae s.l. and An. funestus s.l. are the two major vectors with a predominance of An. gambiae s.l.

Differential insecticide resistance in Anopheles arabiensis populations in the seaside area of Mbour and its suburbs in Senegal

Regular monitoring of insecticide resistance status is an important step in implementing appropriate and adapted insecticide-based strategies for vector control. In Senegal, Indoor Residual Spraying (IRS) and a national distribution campaign for long-lasting insecticide-treated net (LLIN) have been implemented since 2007 and 2009, respectively to prevent malaria transmission. To expand and ensure the sustainability of these strategies, we conducted a study on the status of insecticide resistance in malaria vectors in the seaside area of Mbour and its suburbs where no data were previously available.Anopheles larvae were sampled from four study sites (two in both coastal and inland areas) and reared to adulthood in the insectarium. Non-blood-fed females aged 3–5 days were then tested for susceptibility to permethrin, deltamethrin, lambdacyhalothrin, bendiocarb and pirimiphos-methyl. PCR amplification was used to identify sibling species of the An. gambiae complex and genotyping for the presence of resistance knockdown (kdr) L1014S, L1014F and Ace-1 G119S.Anopheles arabiensis was the only species present in the area. At all four sites, mosquitoes were resistant to deltamethrin, permethrin, and lambdacyhalothrin, and exhibited varying degrees of resistance to bendiocarb and pirimiphos-methyl. Overall, high levels of leucine-serine/phenylalanine substitutions at position 1014 (L1014S/L1014F) were observed, with frequencies ranging from 76.4 to 85.2 % for L1014F, and from 43.2 to 66.7 % for L1014S, compared to 8.1 to 28.3 for the Ace-1 G119S mutation.These results indicate a high level of phenotypic and genotypic resistance to insecticides, which is alarming, as it could have a significant impact on the operational effectiveness of current vector control tools that rely on pyrethroids. However, in the case of bendiocarb and pirimiphos-methyl, while some level of tolerance was observed, their potential use requires regular monitoring to prevent operational failure, as their deployment could potentially lead to an increase in resistance to them.

Fungicides modify pest insect fitness depending on their genotype and population

Fungicides are the most sold pesticide group, with an 8% increase in sales in Europe within the last decade. While adverse short-term fungicide effects on non-target insect species have been reported, the long-term effects and their impact on fitness are unclear. As the effects may depend on both the fungicide and the genetic background of the species, we investigated the effects of the commonly used fungicide, fluazinam, on the Colorado potato beetle's life history traits, and whether the effects were dependent on a previously characterized insecticide resistance mutation (S291G in acetylcholinesterase-2 gene) in different populations. Our findings show that fungicide exposure can have both negative and positive, long-lasting effects on beetles, depending on the parental insecticide resistance status and population. In the Belchow population, individuals carrying resistance mutation had higher survival, but they produced offspring with lower egg-hatching rates. While, in the Vermont population, fungicide exposure increased the body mass and offspring quality in the beetles carrying resistance mutation but did not affect the beetles’ survival. Our results suggest that commonly used fungicides can have both negative and positive effects on pest insects’ life-history, however, their impact may differ depending on the population and parental genetic background.

Geographical survey of the mycobiome and microbiome of Southern California glassy-winged sharpshooters.

The glassy-winged sharpshooter, Homalodisca vitripennis Germar, is an invasive xylem-feeding leafhopper with a devastating economic impact on California agriculture through transmission of the plant pathogen, Xylella fastidiosa. While studies have focused on X. fastidiosa or known symbionts of H. vitripennis, little work has been done at the scale of the microbiome (the bacterial community) or mycobiome (the fungal community). Here, we characterize the mycobiome and the microbiome of H. vitripennis across Southern California and explore correlations with captivity and host insecticide resistance status. Using high-throughput sequencing of the ribosomal internal transcribed spacer 1 region and the 16S rRNA gene to profile the mycobiome and microbiome, respectively, we found that while the H. vitripennis mycobiome significantly varied across Southern California, the microbiome did not. We also observed a significant difference in both the mycobiome and microbiome between captive and wild H. vitripennis. Finally, we found that the mycobiome, but not the microbiome, was correlated with insecticide resistance status in wild H. vitripennis. This study serves as a foundational look at the H. vitripennis mycobiome and microbiome across Southern California. Future work should explore the putative link between microbes and insecticide resistance status and investigate whether microbial communities should be considered in H. vitripennis management practices. IMPORTANCE The glassy-winged sharpshooter is an invasive leafhopper that feeds on the xylem of plants and transmits the devastating pathogen, Xylella fastidiosa, resulting in significant economic damage to California's agricultural system. While studies have focused on this pathogen or obligate symbionts of the glassy-winged sharpshooter, there is limited knowledge of the bacterial and fungal communities that make up its microbiome and mycobiome. To address this knowledge gap, we explored the composition of the mycobiome and the microbiome of the glassy-winged sharpshooter across Southern California and identified differences associated with geography, captivity, and host insecticide resistance status. Understanding sources of variation in the microbial communities associated with the glassy-winged sharpshooter is an important consideration for developing management strategies to control this invasive insect. This study is a first step toward understanding the role microbes may play in the glassy-winged sharpshooter's resistance to insecticides.

Dynamic monitoring of the insecticide resistance status of Bemisia tabaci across China from 2019-2021.

Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is a major agricultural insect pest that causes severe economic losses worldwide. Several insecticides have been applied to effectively control this key pest. However, owing to the indiscriminate use of chemical insecticides, B. tabaci has developed resistance against these chemical compounds over the past several years. From 2019 to 2021, 23 field samples of B. tabaci were collected across China. Twenty species were identified as the Mediterranean 'Q' type (MED) and three were identified as MED/ Middle East-Asia Minor 1 mixtures. Subsequently, resistance of the selected populations to different insecticides was evaluated. The results showed that 13 populations developed low levels of resistance to abamectin. An overall upward trend in B. tabaci resistance toward spirotetramat, cyantraniliprole and pyriproxyfen was observed. In addition, resistance to thiamethoxam remained low-to-moderate in the 23 field populations. These findings suggest that the overall resistance of the field-collected B. tabaci populations has shown an upward trend over the years in China. We believe our study can provide basic data to support integrated pest management and insecticide resistance management of field B. tabaci in China. © 2023 Society of Chemical Industry.

Insecticide resistance status of Aedes aegypti and Aedes albopictus in Malaysia (2010 to 2022): A review

This review aimed to determine the prevalence of the insecticide resistance status of the field-collected Aedes (Ae.) aegypti and Ae. albopictus in Malaysia from 2010 to 2022 towards carbamates, organochlorines, organophosphates and pyrethroids. Biological and environmental controls were summarized with an emphasis on the mosquito vector control strategies in Malaysia. The information in this review was extracted from several databases such as PubMed (MEDLINE), Science Direct and Scopus by using keywords including “insecticide resistance”, “carbamate resistance”, “organochlorine resistance”, “organophosphate resistance”, “pyrethroid resistance”, “Aedes” and “Malaysia”, between January 2022 and December 2022. Distribution of resistant Ae. aegypti and Ae. albopictus in Malaysia was mapped using QGIS software. Insecticide resistance in both Ae. aegypti and Ae. albopictus is widespread in Malaysia, although the rates vary by states. The most notable was the steep increase in permethrin resistance of Ae. aegypti in Selangor, Malaysia, over the past decade. Ae. albopictus also displayed moderate resistance to permethrin, though not as widespread as Ae. aegypti in Selangor, but showed sign of resistance in Sarawak, East Malaysia. Resistance towards four main classes of insecticides have been widely documented in Malaysia. The extensive resistance towards permethrin in Malaysia which is one of the current insecticides used in Malaysia suggested that policies supporting the widespread use of permethrin fogging needs further evaluation.

Insecticide Resistance and Metabolic Mechanisms in Aedes aegypti from Two Agrosystems (Vegetable and Cotton Crops) in Côte d'Ivoire.

Background: The emergence of insecticide resistance in Aedes vectors mosquitoes poses a real challenge for arboviral-borne disease control. In Côte d'Ivoire, data are available on phenotypic resistance and the presence of kdr mutations in Aedes aegypti. Therefore, information on metabolic resistance in Aedes populations is very scarce. Here, we assessed the insecticide resistance status of Ae. aegypti in periurban and rural areas of Côte d'Ivoire, and we investigated the role of detoxification enzymes as possible resistance mechanisms. Materials and Methods: Aedes mosquito eggs were collected between June 2019 to April 2021 in two agricultural sites. Adults of Ae. aegypti were tested using World Health Organization tube assays, with seven insecticides belonging to pyrethroids, organochlorines, carbamates, and organophosphates classes. We determined the knockdown times (KdT50, KdT95) and resistance ratios of pyrethroids in natural populations. The synergist piperonyl butoxide (PBO) was used to investigate the role of enzymes in resistance. Biochemical assays were performed to detect potential increased activities in mixed-function oxidase levels, nonspecific esterases (NSEs), and glutathione S-transferases. Results: The results showed that Ae. aegypti populations were resistant to five insecticides with mortality of 46% and 89% for 0.75% permethrin, 68% and 92% for 0.05% deltamethrin, 57% and 89% for lambda-cyhalothrin, 41% and 47% for dichlorodiphenyltrichloroethane (DDT), 82% and 91% for chlorpyrifos-methyl in Songon-Agban and Kaforo, respectively. Susceptibility to carbamates was observed in our study sites. After exposure to PBO, the susceptibility of Ae. aegypti to pyrethroids and DDT was partially restored in Songon-Agban. Whereas in Kaforo, none increase of the mortality rates of Ae. aegypti for these four insecticides was observed after exposure to PBO. Increased activity of NSE (α-esterases) was found in Songon-Agban compared with the reference susceptible strain. Conclusion: These findings provide valuable information to support decisions for vector control strategies in Cote d'Ivoire. Also, we highlight the need for the monitoring of insecticide resistance management in Aedes vectors.

Worldwide Status of Insecticide Resistance of Aedes aegypti and Ae. albopictus, Vectors of Arboviruses of Chikungunya, Dengue, Zika and Yellow Fever.

Controlling of Aedes aegypti and Ae. albopictus, vectors of five important mosquito-borne diseases, is known as the most effective method to prevent the transmission of arboviruses to humans, but the emergence of insecticide resistance is threat for control and prevention of vector borne diseases. A better understanding of mosquito resistance to insecticides will help to develop more effective methods to control insecticide resistance in mosquito vectors. Worldwide geographical distribution of insecticide resistance in Ae. aegypti and Ae. albopictus by the available papers and map of the data for carbamates, organochlorines, organophosphates, pyrethroids, microbial and insect growth regulator insecticides were reviewed. Article data published up to December 2022 were investigated by searching the following databases: "Google Scholar", "PubMed", "Scopus", "SID" and "Web of Knowledge". The results showed that the susceptibility and resistance status of Ae. aegypti and Ae. albopictus to insecticides in the world is very diverse. Due to the importance of Ae. aegypti and Ae. albopictus in the transmission of mosquito-borne arboviruses, resistance management should be given more attention worldwide to prevent insecticide resistance in the arbovirus vector and replace the new approach for vector control.

Monitoring of insecticide resistance for Apolygus lucorum populations in the apple orchard in China

Apolygus lucorum (Meyer-Dur) (Hemiptera: Miridae) is an important piercing-sucking pest that has been recorded on more than 200 host plants in China. In recent years, its damage to apple trees has become more and more serious, resulting in malformed fruit and even fruit shedding. It is primarily controlled with insecticides. However, the current status of insecticide resistance of A. lucorum in the apple orchard is rarely reported. In this study, we determined the susceptibility of one laboratory population and six geographic field populations of A. lucorum to ten chemical and six botanical insecticides by glass-vial and artificial diet bioassays. The results showed that among the ten chemical insecticides, bifenthrin exhibited the highest toxicity to this pest by glass-vial bioassays than other insecticides, followed by chlorpyrifos, and then one of the neonicotinoid insecticides nitenpyram. In the artificial diet bioassays, the LC50 values of the avermectin insecticides to A. lucorum populations were significantly lower than those of other classes of insecticides, followed by neonicotinoid insecticides. Among six botanical pesticides, veratramine and pyrethrins showed higher toxicity to A. lucorum populations, while rotenone and cnidiadin exhibited poor effects. The results indicated that chlorpyrifos, bifenthrin, avermectin, and emamectin benzoate could be recommended to mix or rotate with botanical pesticides veratramine and pyrethrins in controlling A. lucorum in the apple orchard. Our study would provide valuable information for choosing alternative insecticides and for insecticide resistance management of A. lucorum.

Insecticide resistance status of Aedes aegypti and Aedes albopictus mosquitoes in southern Benin, West Africa

BackgroundThe emergence of insecticide resistance in Aedes mosquitoes could undermine efforts to control arboviruses. The present study aims to assess in some communes of Southern Benin, the susceptibility level of Aedes aegypti (Linnaeus, 1762) and Aedes albopictus (Skuse, 1894) to insecticides commonly used in public health, as well as mechanisms involved.MethodsFemales Ae. albopictus and Ae. aegypti collected in Ifangni, Porto-Novo, Avrankou, Adjarra and Kétou from June 2021 to October 2022, were exposed to: deltamethrin 0.05%, permethrin 0.75%, alpha-cypermethrin 0.05%, pirimiphos methyl 0.25% and bendiocarb 0.1%, following the standard WHO susceptibility tube test protocol. In some sites, pre-exposure to the synergist PBO was used to verify if pyrethroid resistance of populations of Aedes was mediated by oxidases.ResultsFull susceptibility to deltamethrin and permethrin was observed in all tested populations of Ae. albopictus. However, with alphacypermethrin, a suspected resistance was observed in Adjarra (94.67%), Ifangni (93%) and Porto-Novo (94%), and a resistance in Avrankou (83%). The PBO-alphacypermethrin tests performed, led to a full susceptibility (100%) in all four sites, which confirms the full involvement of oxidases in resistance of all tested populations of Ae. albopictus to alphacypermethrin. At the opposite, Aedes aegypti was either resistant or suspected of being resistant to all tested pyrethroids in all four sites, except in Ifangni where a full susceptibility to alphacypermethrin was observed. The full susceptibility of Ae. aegypti to bendiocarb and pirimiphos-methyl in all communes suggests that these two insecticides can be good candidates for an effective control of pyrethroid-resistant Aedes vector populations. Use of permethrin and deltamethrin could also be considered for controlling populations of Ae. albopictus.ConclusionResults of the present study will help guide strategy to implement for an effective control of Aedes vector populations in Benin.

Insecticide resistance status of Aedes aegypti in southern and northern Ghana

BackgroundOutbreaks of Aedes-borne arboviral diseases are becoming rampant in Africa. In Ghana, there is no organized arboviral control programme with interventions restricted to mitigate outbreaks. Insecticide application is a crucial part of outbreak responses and future preventative control measures. Thus, knowledge of the resistance status and underlying mechanisms of Aedes populations is required to ensure optimal insecticide choices. The present study assessed the insecticide resistance status of Aedes aegypti populations from southern Ghana (Accra, Tema and Ada Foah) and northern Ghana (Navrongo) respectively.MethodsPhenotypic resistance was determined with WHO susceptibility tests using Ae. aegypti collected as larvae and reared into adults. Knockdown resistance (kdr) mutations were detected using allele-specific PCR. Synergist assays were performed with piperonyl butoxide (PBO) to investigate the possible involvement of metabolic mechanisms in resistance phenotypes.ResultsResistance to DDT was moderate to high across sites (11.3 to 75.8%) and, for the pyrethroids deltamethrin and permethrin, moderate resistance was detected (62.5 to 88.8%). The 1534C kdr and 1016I kdr alleles were common in all sites (0.65 to 1) and may be on a trajectory toward fixation. In addition, a third kdr mutant, V410L, was detected at lower frequencies (0.03 to 0.31). Pre-exposure to PBO significantly increased the susceptibility of Ae. aegypti to deltamethrin and permethrin (P < 0.001). This indicates that in addition to kdr mutants, metabolic enzymes (monooxygenases) may be involved in the resistance phenotypes observed in the Ae. aegypti populations in these sites.ConclusionInsecticide resistance underpinned by multiple mechanisms in Ae. aegypti indicates the need for surveillance to assist in developing appropriate vector control strategies for arboviral disease control in Ghana.Graphical

Lack of Known Target-Site Mutations in Field Populations of Ostrinia furnacalis in China from 2019 to 2021.

The Asian corn borer, Ostrinia furnacalis (Guenée) (Lepidoptera; Pyralidae), is one of the most destructive insect pests of corn, for which chemical insecticides have been the primary method of control, especially during outbreaks. Little information is currently available on the status of insecticide resistance and associated mechanisms in O. furnacalis field populations. Invasions and outbreaks of Spodoptera frugiperda in China in recent years have increased chemical application in corn fields, which adds to the selection pressure on O. furnacalis. This study was conducted to estimate the risk of insecticide resistance by investigating the frequency of insecticide resistant alleles associated with target site insensitivity in field populations of O. furnacalis. Using the individual-PCR genotype sequencing analysis, none of the six target-site insecticide resistant mutations were detected in O. furnacalis field populations collected from 2019 to 2021 in China. These investigated insecticide resistance alleles are common in resistant Lepidoptra pests and are responsible for resistance to pyrethroids, organophosphorus, carbamates, diamide, and Cry1Ab. Our results support the low insecticide resistance status in field O. furnacalis populations and betokens the unlikely development of high resistance mediated by the common target-site resistance alleles. Additionally, the findings would serve as references for further efforts toward the sustainable management of O. furnacalis.

Insecticide resistance monitoring and diagnostics of resistance mechanisms in cotton‐melon aphid, Aphis gossypii Glover in Central China

AbstractAphis gossypii Glover is one of the most serious pests in China, and the management of this pest mainly relies on insecticides. However, the resistance status of A. gossypii in Central China is still unknown. Therefore, in this study, we firstly determined the susceptibility of 15 A. gossypii field populations from Central China to six chemical insecticides using leaf‐dipping method during 2020 and 2021 and explored the potential resistance mechanisms of these populations. Most of the field populations of A. gossypii have evolved low to high levels of resistance to imidacloprid (RR = 5.98–2352.96‐fold), thiamethoxam (RR = 5.98–2194.96‐fold), flonicamid (RR = 7.18–306.07‐fold), and pirimicarb (RR = 6.90–283.00‐fold); however, 13 tested field populations remained susceptible to chlorpyrifos. For λ‐cyhalothrin, most populations have evolved low to moderate levels of resistance (RR = 5.72–47.38‐fold). The results of correlation analysis indicated that there was a prominent positive correlation between the enzyme activity of cytochrome P450 and the logarithm of LC50 to thiamethoxam and pirimicarb. Carboxylesterase (CarE) was positively correlated with λ‐cyhalothrin, while glutathione S‐transferase (GST) was negatively correlated with λ‐cyhalothrin. Furthermore, we found that multiple resistance associated target site mutations (V62I, R81T, A302S, S431F, M918L) can be detected in the field populations of A. gossypii. Aphis gossypii in Central China have developed different degrees of resistance to tested insecticides and enhanced detoxification of cytochrome P450 and carboxylesterase and co‐existed resistance mutations contribute to the multiple resistance of these field populations. Our results provide valuable information about the status of insecticide resistance in China and are helpful for designing sustainable management strategies of A. gossypii.

Species composition and insecticide resistance in malaria vectors in Ellibou, southern Côte d’Ivoire and first finding of Anopheles arabiensis in Côte d’Ivoire

BackgroundKnowing the species composition and insecticide resistance status of the target vector population is important to guide malaria vector control. The aim of this study was to characterize the malaria vector population in terms of species composition, insecticide susceptibility status and potential underlying resistance mechanisms in Ellibou, southern Côte d’Ivoire.MethodsA 1-year longitudinal entomological survey was conducted using light traps and pyrethroid spray catches to sample adult mosquitoes in combination with larval sampling. The susceptibility status of Anopheles gambiae sensu lato (s.l.) to bendiocarb, deltamethrin, DDT and malathion was assessed using the World Health Organization insecticide susceptibility test. Additionally, An. gambiae specimens were screened for knockdown (kdr) and acetylcholineesterase (ace1) target site resistance alleles, and the expression levels of eight metabolic resistance genes, including seven cytochrome P450 monooxygenases (P450s) and one glutathione S-transferase (GST), measured with reverse transcription quantitative real-time polymerase chain reaction (qPCR).ResultsOverall, 2383 adult mosquitoes from 12 different taxa were collected with Culex quinquefasciatus and An. gambiae being the predominant taxa. Molecular identification of An. gambiae s.l. revealed the presence of Anopheles arabiensis, Anopheles coluzzii, An. gambiae sensu stricto (s.s.) and Anopheles coluzzii/An. gambiae s.s. hybrids. Anopheles gambiae mosquitoes were resistant to all insecticides except malathion. PCR diagnostics revealed the presence of ace1-G280S and the kdr L995F, L995S and N1570Y target-site mutations. Additionally, several genes were upregulated, including five P450s (i.e., CYP6P3, CYP6M2, CYP9K1, CYP6Z1, CYP6P1) and GSTE2.ConclusionThis is the first documented presence of An. arabiensis in Côte d’Ivoire. Its detection – together with a recent finding further north of the country – confirms its existence in the country, which is an early warning sign, as An. arabiensis shows a different biology than the currently documented malaria vectors. Because the local An. gambiae population was still susceptible to malathion, upregulation of P450s, conferring insecticide resistance to pyrethroids, together with the presence of ace1, suggest negative cross-resistance. Therefore, organophosphates could be an alternative insecticide class for indoor residual spraying in the Ellibou area, while additional tools against the outdoor biting An. arabiensis will have to be considered.

Insecticide Resistance Status of Aedes albopictus (Diptera: Culicidae) Populations from Cuba.

Aedes albopictus (Skuse, 1894) is one of the major vectors for arboviruses such as dengue, Zika, and chikungunya. Originally from Southeast Asia, this species has spread to Africa, Europe, and the Americas, including Cuba. This spread has been favored by its great adaptability to variable temperatures and to the resistance of its eggs to desiccation. Chemical control of mosquitoes is an essential alternative to stop arbovirus transmission, but insecticide resistance status of the Cuban Ae. albopictus populations is unknown. For this study, Ae. albopictus larvae and adults were collected from two municipalities in Havana, Cuba in 2019. Adult bioassays for deltamethrin, cypermethrin, lambda-cyhalothrin, chlorpyrifos, propoxur, and bendiocarb susceptibility were conducted according to CDC methodology. Larval bioassays for temephos susceptibility were performed following WHO protocols. Resistance profiles for α and β-esterases, glutathione S-transferase (GST), and multifunction oxidases (MFO) pathways were constructed and analyzed. Resistance to temephos and deltamethrin was detected in Mulgoba and Plaza field populations, but resistance to lambda-cyhalothrin was only found in the Plaza colony. Plaza colony exhibited a higher expression level to all four metabolic enzymes and α-esterases and GTS were over-expressed in Mulgoba. The development of insecticide resistance in Cuban Ae. albopictus populations makes it imperative that we develop integrated control strategies to minimize the development of resistance and provide effective vector control that prevents the onset of arbovirus epidemics.
