Methicillin-resistant staphylococci in clinical bovine mastitis: occurrence, molecular analysis, and biofilm production.

Staphylococcus aureus is an important pathogen that causes mastitis in cattle, and the emergence of methicillin-resistant S. aureus (MRSA) poses a threat to veterinary and human medicine. The aims of the study were to investigate the prevalence of MRSA and methicillin-resistant coagulase-negative staphylococci (MR-CoNS) isolated from clinical mastitis, their ability to form biofilms, and the antimicrobial susceptibility of S. aureus strains. In addition, the Staphylococcal Cassette Chromosome mec (SCCmec) type, spa type and the presence of Panton-Valentine Leucocidin in MRSA were evaluated. A total of 326 staphylococcal strains were screened by multiplex-PCR for S. aureus and Staphylococcus intermedius group (SIG) identification. The S. aureus strains (n = 163) were subjected to phenotypic testing for antimicrobial susceptibility and biofilm formation. Molecular analysis was performed on MRSA mecA-positive strains. Of 163 S. aureus isolates, 142 strains (87.1%) were resistant to at least one antibiotic, and all 19 MRSA strains were resistant to at least four out of five antibiotics tested. All S. aureus strains harboured the icaA gene and were biofilm producers. Nineteen MR-CoNS strains were also isolated. The most prevalent spa types among MRSA were t001 (57.9%) and t037 (31.6%), while one MRSA was type t008 and one was type t041. Most MRSA were SCCmec type I (63.2%) and III (31.6%) and only one strain was type IV. None of the MRSA isolates had the PVL gene. The prevalence of multidrug-resistant S. aureus in bovine mastitis is a serious concern. The finding of MRSA with spa types predominant in humans and infrequent in Italian cows and with SCCmec infrequently found in bovine milk or cheese suggest a human origin of these strains. The ability of MRSA and MR-CoNS involved in bovine mastitis to be transferred to humans and vice versa poses a public health concern.

ANTIMICROBIAL SUSCEPTIBILITY OF STAPHYLOCOCCUS SPP. ISOLATED FROM FELIDS AND CANIDS IN BELO HORIZONTE ZOO, BRAZIL.

The epidemiology of Staphylococcus spp. has become a major concern among humans and animals due to increasing antimicrobial resistance and frequent reports of infection. Despite the importance of animals as reservoirs for staphylococci, little is known about the epidemiology of Staphylococcus spp. in most nondomestic species, including canids and felids. This study evaluated the frequency, distribution, and patterns of antimicrobial resistance of staphylococcal species isolated from captive felids and canids from Belo Horizonte Zoo, Brazil. Rectal, oral, and nasal swabs from apparently healthy maned wolves (Chrysocyon brachyurus, n= 7), a lion (Panthera leo, n = 1), jaguars (Panthera onca, n = 3), and one swab of a cougar (Puma concolor) with an ear infection were streaked onto mannitol salt agar. Colonies identified by matrix-assisted laser desorption/ionization-time of flight mass spectrometry, polymerase chain reaction for the Staphylococcus intermedius group (SIG), and 16S rRNA gene sequencing. Isolates were subjected to antimicrobial susceptibility tests and Staphylococcus pseudintermedius strains were subjected to multilocus sequence typing. Staphylococcus species were isolated from 24 of the 34 samples (70.6%). Among the isolated strains, S. pseudintermedius and Staphylococcus felis were the most frequent species (41.7 and 25%, respectively). Five novel sequence types were identified among the S. pseudintermedius isolates. Resistance to tetracycline (7/24, 29.2%) or penicillin (6/23, 26.1%) was significantly higher than the other antimicrobial agents tested (P < 0.05). One isolate, Staphylococcus nepalensis, was positive for mecA and resistant to five antimicrobials, and was thus classified as multidrug-resistant. The present work suggests that maned wolves are natural hosts of SIG and also reports the isolation of S. felis in sick and healthy, captive, nondomestic carnivores. The isolated staphylococci were susceptible to most classes of antimicrobials tested. However, the multidrug-resistance capability of an S. nepalensis strain reinforces the hypothesis that felids and canids act as reservoirs of pathogens with antimicrobial resistance.

Once a Known Veterinary Pathogen, Now a Forgotten Zoonosis. Case Report of An Invasive Staphylococcus intermedius Group Infection

Introduction: Staphylococcus intermedius group (SIG), a known veterinary pathogen with the potential for zoonotic human infections, comprises S. intermedius, S. pseudintermedius, and S. delphini, which are not easily distinguishable. Without the proper equipment and procedures, it cannot be distinguished from Staphylococcus aureus (SAu), which causes underestimation of its true incidence. Case Presentation: A 52-year-old male with diabetes presented with complaints of fever and malaise. He developed respiratory failure and altered mental status; hence, intensive care was provided to him. Blood cultures and bronchoalveolar lavage culture developed methicillin-resistant SIG. Despite rapid adjustment of empiric antibiotic therapy, he died of multiple organ failure. Conclusions: Incorporating knowledge about this new pathogen and its aggressiveness into daily clinical practice can, through a high index of suspicion and detailed anamnesis, reduce misdiagnoses.

Determination of the frequency, species distribution and antimicrobial resistance of staphylococci isolated from dogs and their owners in Trinidad.

The close contact between humans and their dogs can lead to the commingling of staphylococci and the exchange of mobile genetic elements encoding antimicrobial resistance. The objectives of this study were to determine the species distribution and antimicrobial resistance patterns of staphylococci colonizing canine pets and their owners in Trinidad. Staphylococci were isolated from canine pets and their owners and identified using MALDI-TOF mass spectrometry. Antimicrobial susceptibilities were determined using the Kirby-Bauer disc diffusion method against seven classes of antimicrobial agents. A total of 440 staphylococci were isolated from 112 canine pets and their owners, 53.4% were from canine pets and 46.6% were from owners. Twenty-four species were detected, of which, most isolates (32.5%) belonged to the Staphylococcus intermedius group (SIG). S. sciuri was the most common species of coagulase-negative staphylococci (CoNS) comprising 22.3% of all isolates. Antimicrobial resistance was highest against commonly used antimicrobials, such as penicillin (51.4%), tetracycline (26.1%) and trimethoprim/sulfamethoxazole (18.6%). These antimicrobials also comprised the most common multidrug resistance (MDR) combination. Overall, 19.1% of isolates displayed multidrug resistance. No methicillin-resistant Staphylococcus aureus (MRSA) isolates were detected. However, methicillin resistance was detected in 13.3% and 15.1% of coagulase-positive staphylococci (CoPS) and the CoNS+CoVS (combined CoNS and coagulase-variable staphylococci) group respectively. The presence of methicillin-resistant staphylococci is worrisome because there is the potential for the transfer of these strains between dogs and humans. These strains may act as a reservoir of resistance genes.

Staphylococcal cassette chromosome mec elements from Staphylococcus intermedius group (SIG) isolates from dogs in a center for veterinary diagnostics in Brazil

ABSTRACT: Methicillin resistance in the Staphylococcus intermedius group (SIG) has emerged in small animal practice. Methicillin-resistant SIG (MRSIG) members have been implicated as causes of infections in both companion animals and humans. Staphylococcal cassette chromosome mec (SCCmec) elements carry the mecA/C genes, which encode for the transpeptidase PBP2a (PBP2’) responsible for β-lactam antibiotic resistance in staphylococci. This study examined the SCCmec types of MRSIG isolates from different clinical specimens of dogs that exhibited methicillin MIC ≥ 0.5 μg/mL by an automated identification and susceptibility system in a Center for Veterinary Diagnostics in São Paulo, Brazil. Susceptibility to methicillin was determined by broth microdilution testing, and Oxoid® M.I.C.Evaluator® strips. PBP2a production was detected using a latex agglutination assay. SCCmec typing was performed according to the International Working Group on the Classification of Staphylococcal Cassette Chromosome Elements (IWG-SCC) guidelines. SCCmec type II (2A), SCCmec type III (3A), composite SCC structures consisting of a class A mec gene complex in addition to multiple ccr gene complexes, and non-typable SCCmec elements were reported in these MRSIG isolates. SCCmec type variants differing from those so far acknowledged by IWG-SCC were found, indicating new rearrangements in the genetic context of mecA in these canine MRSIG isolates.

Delineating Novel Therapeutic Drug and Vaccine Targets for Staphylococcus cornubiensis NW1T Through Computational Analysis

Staphylococcal multidrug resistance is an emerging future threat. Among staphylococci species, Staphylococcus intermedius group (SIG) comprises of coagulase-positive four staphylococci isolated from veterinary wounds and post-operative infections in humans and animals. Staphylococcus cornubiensis has been reported as a new member of this group and is associated with infections in a wide range of wild and domesticated animals as well as in humans. Cases of the antibiotic resistance in different members of the SIG are reported indicating its future threat for antibiotic therapy. The objective of this study was to determine the unique potential drug and vaccine targets for infectious S. cornubiensis NW1T. Genome-wide subtractive proteomic approach accompanied by immunoinformatics analysis was performed on the newly reported genome of S. cornubiensis NW1T. Applying a stringent comparative proteomic analyses, 35 proteins were identified as novel druggable targets based on their function in pathogen specific metabolic pathways as well as their druggablity potential. Using antigenicity and epitope prediction methods, the possible B and T-cells antigenic peptides of these pathogen-specific essential proteins were determined. As a result, six proteins were prioritized as potential vaccine candidates that fulfilled the basic criteria of effective vaccine targets. During analyses, candidate proteins of S. cornubiensis were found to carry T-cell and B-cell epitopes and predicted to interact with both class I and II MHC molecules at significantly lower IC50 values. Moreover, a leading antigenic peptide “LVDTLNAAG” from penicillin binding protein of this pathogenic bacteria was found to interact with multiple MHC alleles of class I and II with IC50 value of < 100 nM. Molecular docking showed favorable interactions between this lead antigenic peptide with human HLA-C*05:01 cell surface receptor. This peptide is predicted as a promising epitope capable of triggering the immune response against pathogenic S. cornubiensis NW1T. The candidate proteins and epitopes determined in this study may be promising novel drug and vaccine targets against infection-causing S. cornubiensis.

Characterization of Staphylococcus intermedius Group Isolates Associated with Animals from Antarctica and Emended Description of Staphylococcus delphini.

Members of the genus Staphylococcus are widespread in nature and occupy a variety of niches, however, staphylococcal colonization of animals in the Antarctic environment has not been adequately studied. Here, we describe the first isolation and characterization of two Staphylococcus intermedius group (SIG) members, Staphylococcus delphini and Staphylococcus pseudintermedius, in Antarctic wildlife. Staphylococcus delphini were found exclusively in Adélie penguins. The report of S. pseudintermedius from Weddell seals confirmed its occurrence in all families of the suborder Caniformia. Partial RNA polymerase beta-subunit (rpoB) gene sequencing, repetitive PCR fingerprinting with the (GTG)5 primer, and matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry gave consistent identification results and proved to be suitable for identifying SIG members. Comparative genomics of S. delphini isolates revealed variable genomic elements, including new prophages, a novel phage-inducible chromosomal island, and numerous putative virulence factors. Surface and extracellular protein distribution were compared between genomes and showed strain-specific profiles. The pathogenic potential of S. delphini was enhanced by a novel type of exfoliative toxin, trypsin-like serine protease cluster, and enterotoxin C. Detailed analysis of phenotypic characteristics performed on six Antarctic isolates of S. delphini and eight reference strains from different animal sources enabled us to emend the species description of S. delphini.

Comparing the treatment effect of narrow spectrum antimicrobial, probiotic and fluid with amoxicillin in mink kits (Neovison vison) with pre-weaning diarrhea

Pre-weaning diarrhea in mink kits (PWD), also known as “sticky kits” is a multifactorial syndrome of considerable concern in the mink production. Evidence based treatment protocols are not available, and treatment is therefore empirical and often based on the use of antimicrobials. The purpose of the study was to test the effect of 3 alternative treatments to a standard antibiotic treatment, to characterize the study groups microbiologically, and finally to compare the intestinal microbiota of the different treatment groups at the age of 42 days. In total, 226 one to three week old mink kits with PWD from 36 litters were treated with either 1) Lactobacillus reuteri, 2) benzylpenicillin, 3) Ringer lactate or 4) amoxicillin (controls). Effects of the treatments were measured as weight gain from day 0 to day 15 and mortality. Multivariable linear mixed model regression showed no significant difference in weight gain between probiotic-, penicillin or fluid-treated mink kits and the amoxicillin treated controls. There was also no significant difference in mortality risk between the treatment groups. Bacterial culture and next generation sequencing of the viral contents showed that the study groups were uniform with a high frequency of Staphylococcus intermedius group (SIG) bacteria, Escherichia coli, Enterococcus hirae, Mamastrovirus and Sapovirus which were representative for mink kits with PWD. 16S sequencing results of the bacterial microbiota, when the kits were 42 days old were dominated by clostridia in all groups and showed no clear differences in the bacterial composition between the different treatment groups.

Reduced rates of antimicrobial resistance in Staphylococcus intermedius group and Escherichia coli isolated from diseased companion animals in an animal hospital after restriction of antimicrobial use

The 2016 National Action Plan aims for reduction in antimicrobial resistance (AMR) to tetracyclines, third-generation cephalosporins, and fluoroquinolones in Escherichia coli isolates from livestock: to lower the tetracycline resistance of E. coli to 33% or less; to maintain the third-generation cephalosporin resistance of E. coli at the same level as in the other G7 countries as of 2020; and to maintain the fluoroquinolone resistance of E. coli at the same level as in the other G7 countries as of 2020. A relatively unexplored facet of reducing AMR is the impact of minimizing transmission of AMR strains by companion animals. In this study we compared AMR rates in Staphylococcus intermedius group (SIG) and E. coli isolated from diseased companion animals in an animal hospital before and after restriction of antimicrobial use. Our study spanned a 4.5-year period from 2014 to June 2018 during which antimicrobial use was restricted in 2016. During this period, abundance of methicillin-resistant SIG isolates from the hospital dropped from 41.5% to 9.3%, and that of extended-spectrum β-lactamase (ESBL)-producing E. coli isolates dropped from 29.5% to 9.5%. Tests for antimicrobial susceptibility revealed significantly reduced rates of AMR to enrofloxacin and levofloxacin in SIG isolates, and to cefazolin in E. coli isolates after antimicrobial use was restricted. Our observations suggest that restriction of antimicrobial use, especially that of third-generation cephalosporins and fluoroquinolones, is an effective method for reducing AMR rates. These findings will be relevant in guiding antimicrobial restriction approaches in other animal hospitals and clinics.

Staphylococcus cornubiensis sp. nov., a member of the Staphylococcus intermedius Group (SIG).

We here describe a novel species in the Staphylococcus intermedius Group (SIG) which is phenotypically similar to Staphylococcus pseudintermedius but is genomically distinct from it and other SIG members, with an average nucleotide identity of 90.2 % with its closest relative S. intermedius. The description of Staphylococcus cornubiensis sp. nov. is based on strain NW1T (=NCTC 13950T=DSM 105366T) isolated from a human skin infection in Cornwall, UK. Although pathogenic, NW1T carries no known virulence genes or mobilizable antibiotic resistance genes and further studies are required to assess the prevalence of this species in humans as well as its potential presence in companion animals.

Characterization, distribution, antimicrobial resistance and resistance risk factors in staphylococci isolated from cats from 2001 to 2014.

Relatively few studies have been published describing the patterns of staphylococcal isolation and antimicrobial resistance over time in cats. The objective of this retrospective study was to determine the frequency, location, characteristics and antimicrobial resistance profiles of staphylococci isolated by the Louisiana Animal Disease Diagnostic Laboratory between the years 2001 and 2014. All feline staphylococcal isolates were classified phenotypically. Isolates corresponding to known or possibly pathogenic species (Staphylococcus intermedius group (SIG) and Staphylococcus aureus (SA)) as well as Staphylococcus epidermidis (SE) and non‐speciated coagulase‐negative staphylococci (CNS) were further evaluated to determine antimicrobial resistance patterns. A total of 519 staphylococci were isolated. The largest percentage of isolates was CNS, representing 39.3% of the total, while SIG, SE, SA and non‐speciated coagulase positive staphylococci (CPS) represented 18.1%, 10.2%, 8.3% and 7.3%, respectively. Methicillin resistance (MR) was identified in 57.1% of SA and 20.5% of SIG. Resistance to 3 or more antimicrobial classes (multidrug resistance; MDR) was demonstrated in 54.5% of SA and 23.9% of SIG. The prevalence of MDR increased over time in both SIG and SA, while the prevalence of MR increased over time in SIG. An increase in mean antimicrobial resistance score over time was seen in SIG. This study demonstrates a high and increasing prevalence of MDR in SIG and SA, as well as increasing prevalence of MR in SIG isolated from cats.

Bacteria isolated from companion animals in Japan (2014–2016) by blood culture

We aimed to identify microorganisms isolated by blood culture (BC) from companion animals and to determine antimicrobial resistance of these isolates during 2014-2016 at veterinary laboratory, in comparison with those during 2010-2013, in Japan. Clinical data (animal species, visiting animals/hospitalized animals, and others except for disease type and clinical course including history of antimicrobial agent use)on ill animals at veterinary clinics or hospitals were obtained. We retrospectively analyzed animal-origin BC results extracted from the database in 2014-2016 and those obtained in 2010-2013. BC-positive samples were from most of dogs (n=174 in 2014-2016 and n=86 in 2010-2013). Escherichia coli (n=50, 25.1%) and Staphylococcus intermedius group (SIG) bacteria (n=23, 11.6%) were most prevalent in 2014-2016, while the percentages of E. coli (n=22, 25.3%) and SIG (n=9, 10.3%) in 2010-2013 were similar to those in 2014-2016. Percentages of extended-spectrum β-lactamase (ESBL)-producing E.coli andmethicillin-resistant staphylococci (MRS) rate of SIG bacteria isolated in 2014-2016 were 28.0% and 69.6% (vs. 22.7% and 44.4% in 2010-2013), respectively. Fourteen ESBL-producing E.coli in 2014-2016 were isolated from 7 visiting animals and 7 hospitalized ones, whereas the sixteen MRS of SIG were from 7 visiting animals and 9 hospitalized ones. Our observations support the prevalent microorganisms isolated by BC and their antimicrobial resistance patterns for two study periods.

New aspects upon the classification of Staphylococcus intermedins

In 1976, Staphylococcus intermedins was first described as being a new species isolated from pigeons, dogs, mink and horses. Until recently, the majority of coagulase-positive staphylococci originating from animals such as dogs and pigeons have been classified as S. intermedins. This species has been recognized to constitute normal skin flora of various animal species and to occasionally cause a variety of infections in dogs and cats. There have been six species of coagulase-positive staphylococci other than Staphylococcus aureus, namely S.intermedius, S.schleiferì subsp. coagulans, S.hyicus, S. lutrae, S.delphini and S. pseudintermedius. Sequence similarities of 16S rRNA genes among four of these species, except S. hyicus and S. lutrae, are 99% identical. In addition, it is very difficult to differentiate among S. intermedius, S. delphini and S. pseudintermedius phenotypically, and commercial kits are not available for the differentiation between of S. pseudintermedius and S. delphini. It has been shown that isolates phenotypically identified as Staphylococcus intermedius consist of four distinct species, including S. intermedius, Staphylococcus pseudintermedius and Staphylococcus delphini group A and B, which together represent the Staphylococcus intermedius group (SIG). Moreover, S. delphini group Β might be a novel species. In 2009, a new, rapid and inexpensive PCR-RFLP was developed and is capable of discriminating the SIG strains.

Ecological features, pathogenic properties, and role of Staphylococcus intermedius group representatives in animal and human infectious pathology

The paper reports changes in the species nomenclature in the genus Staphylococcus that involve the most pathogenic representatives of the genus, the coagulase-positive staphylococci cluster. To date, this cluster includes six species in addition to Staphylococcus aureus: S. intermedius, S. schleiferi spp. coagulans, S. lutrae, S. hyicus, S. pseudintermedius, and S. delphini. Special attention was given to the representatives of the Staphylococcus intermedius group (SIG), which encompasses three closely related species, S. intermedius, S. pseudintermedius, and S. delphini, whose hosts are mammals and birds living in close proximity to human beings. The current data on the pathogenicity factors and role of SIG representatives in animal and human infectious pathologies were analyzed. The approaches to species identification, together with ecological and epidemiological features, and antibiotic susceptibility were considered. Specific biological features of S. pseudintermedius, the species most similar to S. aureus, are considered from the perspective of the properties of newly emerging pathogens.

Specific features of ecology, pathogenic properties, and role of the Staphylococcus intermedius group members in animal and human infectious pathology

The changes in the nomenclature of species in the genus Staphylococcus, including the most pathogenic cluster of the coagulase-positive staphylococci, are represented. Presently, besides S. aureus, this cluster consists of 6 species: S. intermedius, S. schleiferi ssp. coagulans, S. lutrae, S. hyicus, S. pseudintermedius, and S. delphini. A particular attention was paid to the Staphylococcus intermedius group (SIG), which includes three closely related coagulase-positive bacterial species: S. intermedius, S. pseudintermedius, and S. delphini. The hosts of SIG species are various mammals and birds, which live in a close contact with humans. The current knowledge about the virulence factors and pathogenicity for animals and humans are analyzed. The diffic6lties of the species identification, the features of ecology and epidemiology, antimicrobial resistance were reviewed. The biological features of S. pseudintermedius, which has the greatest similarity with S. aureus, are considered in the context of the properties of newly emerging pathogens.

Evaluation of Oxacillin and Cefoxitin Disk and MIC Breakpoints for Prediction of Methicillin Resistance in Human and Veterinary Isolates of Staphylococcus intermedius Group.

Staphylococcus pseudintermedius is a coagulase-positive species that colonizes the nares and anal mucosa of healthy dogs and cats. Human infections with S. pseudintermedius range in severity from bite wounds and rhinosinusitis to endocarditis; historically, these infections were thought to be uncommon, but new laboratory methods suggest that their true incidence is underreported. Oxacillin and cefoxitin disk and MIC tests were evaluated for the detection of mecA- or mecC-mediated methicillin resistance in 115 human and animal isolates of the Staphylococcus intermedius group (SIG), including 111 Staphylococcus pseudintermediusand 4 Staphylococcus delphini isolates, 37 of which were mecA positive. The disk and MIC breakpoints evaluated included the Clinical and Laboratory Standards Institute (CLSI) M100-S25 Staphylococcus aureus/Staphylococcus lugdunensis oxacillin MIC breakpoints and cefoxitin disk and MIC breakpoints, the CLSI M100-S25 coagulase-negative Staphylococcus (CoNS) oxacillin MIC breakpoint and cefoxitin disk breakpoint, the CLSI VET01-S2 S. pseudintermedius oxacillin MIC and disk breakpoints, and the European Committee on Antimicrobial Susceptibility Testing (EUCAST) S. pseudintermedius cefoxitin disk breakpoint. The oxacillin results interpreted by the VET01-S2 (disk and MIC) and M100-S25 CoNS (MIC) breakpoints agreed with the results of mecA/mecC PCR for all isolates, with the exception of one false-resistant result (1.3% of mecA/mecC PCR-negative isolates). In contrast, cefoxitin tests performed poorly, ranging from 3 to 89% false susceptibility (very major errors) and 0 to 48% false resistance (major errors). BD Phoenix, bioMérieux Vitek 2, and Beckman Coulter MicroScan commercial automated susceptibility test panel oxacillin MIC results were also evaluated and demonstrated >95% categorical agreement with mecA/mecC PCR results if interpreted by using the M100-S25 CoNS breakpoint. The Alere penicillin-binding protein 2a test accurately detected all mecA-positive isolates, although for four isolates, cefoxitin induction was required prior to testing. These data demonstrate that the cefoxitin surrogate test does not reliably detect the presence of mecA in S. pseudintermedius isolates and that laboratories should perform oxacillin disk or MIC tests of these isolates when they are encountered.

Staphylocccus intermedius group infections in humans: report of four cases and a literature review

Introduction: Staphylococcus intermedius group (SIG) comprises coagulase-positive staphylococci isolated from veterinary wounds, with anecdotal association of this pathogen and disease in humans. To date, 28 human cases have been described in the literature. Here, we described four cases of human infection caused by SIG members and, through a literature review, identified potential common characteristics and risk factors for this infection. Case presentations: Cases were obtained via electronic query of existing microbiology records for S. intermedius isolates at three teaching hospitals in Denver, CO, USA, between 2003 and 2008. Four cases were identified. Three cases were soft tissue and/or bone infection and one case was a urinary tract infection. Conclusion: Staphylococcus intermedius infections in humans are rarely identified. Dog ownership as well as diabetes or other causes of immunosuppression may place patients at higher risk. Misidentifying SIG infections could lead to delays in treatment as well as undertreatment of this zoonosis.
