Acoustic-assisted centrifugal microfluidics for particle/cell separation

The process of microparticle manipulation using acoustic waves within microfluidic devices is called acoustophoresis. This research focuses on a new approach for concentrating and categorizing microparticles using an integrated acoustic centrifugal microfluidic system with standing surface acoustic waves (SSAWs) and centrifugal forces. The study employs a rectangular microchannel with multiple inlets and outlets placed on an electrified lab-on-a-disc (eLOD) device subjected to SSAWs, guiding microparticles radially. Various physical parameters such as distance from the center of rotation, lateral displacement, tilting angle, microchannel dimensions, microparticle size, disc rotation speed, and applied voltage to the piezoelectric were simulated to assess microparticle focusing. The results indicate that adjusting lateral displacement and tilting angle can direct microparticles toward desired outlets. Enhanced focusing is achievable at low rotation speeds with higher applied voltage and narrower microchannel widths. The system's potential for classifying circulating tumor cells (CTCs) and white blood cells (WBCs) based on volume and density differences was also demonstrated. This acoustic eLOD device offers an efficient means for manipulating micro- and bioparticles.

Research on particle separation microfluidic chip based on standing surface acoustic wave

Microparticle and cell separation is a key process in the study of cell properties, clinical diagnosis and disease treatment. In this paper, a particle separation method based on the combination of surface acoustic wave and microfluidic technology is proposed, which uses standing surface acoustic wave to achieve the separating of polystyrene microparticles of different sizes twice in a continuous flow. Through numerical simulation of the device model and particle trajectory simulation, the optimum particle separation parameters were determined. The particle separation microfluidic chip has successfully realized the particle separation. The separation yield of 5 μm particles is about 92.1%. The separation rate of 1 μm particles is about 83.3%.

Harnessing a Vibroacoustic Mode for Enabling Smart Functions on Surface Acoustic Wave Devices ‐ Application to Icing Monitoring and Deicing

AbstractMicroacoustic wave devices are essential components in the radio frequency (RF) electronics and microelectromechanical systems (MEMS) industry with increasing impact in various sensing and actuation applications. Reliable and smart operation of acoustic wave devices at low costs will cause a crucial advancement. Herein, this study presents the enablement of temperature and mechanical sensing capabilities in a Rayleigh‐mode standing surface acoustic wave (sSAW) chip device by harnessing an acoustic shear‐thickness dominant wave (SD) using the same set of electrodes. Most importantly, this mode is excited by switching the polarity of the sSAW transducer electrodes by simple electronics, allowing for direct and inexpensive compatibility with an existing setup. The method in the emergent topic of surface de‐icing is validated by continuously monitoring temperature and liquid–solid water phase changes using the SD mode, and on‐demand Rayleigh‐wave deicing with a negligible energy cost. The flexibility for adapting the system to different scenarios, and loads and the potential for scalability opens the path to impact in lab‐on‐a‐chip, internet of things (IoT) technology, and sectors requiring autonomous acoustic wave actuators.

Investigation on submicron particle separation and deflection using tilted-angle standing surface acoustic wave microfluidics

With the development of in vitro diagnostics, extracting submicron scale particles from mixed body fluids samples is crucial. In recent years, microfluidic separation has attracted much attention due to its high efficiency, label-free, and inexpensive nature. Among the microfluidic-based separation, the separation based on ultrasonic standing waves has gradually become a powerful tool. A microfluid environment containing a tilted-angle ultrasonic standing surface acoustic wave (taSSAW) field has been widely adapted and designed to separate submicron particles for biochemical applications. This paper investigated submicron particle defection in microfluidics using taSSAWs analytically. Particles with 0.1–1 μm diameters were analyzed under acoustic pressure, flow rate, tilted angle, and SSAW frequency. According to different acoustic radiation forces acting on the particles, the motion of large-diameter particles was more likely to deflect to the direction of the nodal lines. Decreasing the input flow rate or increasing acoustic pressure and acoustic wave frequency can improve particle deflection. The tilted angle can be optimized by analyzing the simulation results. Based on the simulation analysis, we experimentally showed the separation of polystyrene microspheres (100 nm) from the mixed particles and exosomes (30–150 nm) from human plasma. This research results can provide a certain reference for the practical design of bioparticle separation utilizing acoustofluidic devices.

Sensitivity of acoustofluidic particle manipulation to microchannel height in standing surface acoustic wave-based microfluidic devices

In this paper, the flow and particle trajectories, induced by standing surface acoustic waves (SSAWs) in a poly-dimethylsiloxane microchannel, are investigated by establishing a two-dimensional cross-sectional model with the finite element method and improved boundary conditions. Extensive parametric studies are conducted regarding the channel height, ranging from 0.2 to 4.0 times the spacing of the repetitive vertical interference pattern, to investigate its influences on the flow field and microparticle aggregation. The first-order flow field is found to be related to the channel height, exhibiting a periodic spatial distribution and oscillatory variation in its amplitude as the height changes. We theoretically analyze the propagation mechanism of the acoustic waves in the vertical direction and thus determine the periodicity of the wave interference pattern. Furthermore, we find that the speed of the particle aggregation is a function of the channel height, so the channel height can be optimized to maximize the strength of the first-order flow field and thus minimize the time of particle aggregation. The optimum heights can reduce the aggregation time by up to 76%. In addition, the acoustophoretic motions of microparticles exhibit a spatially dependent pattern when the channel height becomes larger than a quarter of the wavelength of the SAW, which can be explained by the change in the ratio between the radiation force and the streaming drag force from position to position. Our findings provide guidelines to the design and optimization of SSAW-based acoustofluidic devices.

On the behavior of prolate spheroids in a standing surface acoustic wave field

The active manipulation of particle and cell trajectories in fluids by high-frequency standing surface acoustic waves (sSAW) allows to separate particles and cells systematically depending on their size and acoustic contrast. However, process technologies and biomedical applications usually operate with non-spherical particles, for which the prediction of acoustic forces is highly challenging and remains a subject of ongoing research. In this study, the dynamical behavior of prolate spheroids exposed to a three-dimensional acoustic field with multiple pressure nodes along the channel width is examined. Optical measurements reveal an alignment of the particles orthogonal to the pressure nodes of the sSAW, which has not been reported in literature so far. The dynamical behavior of the particles is analyzed under controlled initial conditions for various motion patterns by imposing a phase shift on the sSAW. To gain detailed understanding of the particle dynamics, a three-dimensional numerical model is developed to predict the acoustic force and torque acting on a prolate spheroid. Considering the acoustically induced streaming around the particle, the numerical results are in excellent agreement with experimental findings. Using the proposed numerical model, a dependence of the acoustic force on the particle shape is found in relation to the acoustic impedance of the channel ceiling. Hence, the numerical model presented herein promises high progress for the design of separation devices utilizing sSAW, exploiting an additional separation criterion based on the particle shape.

A Simulated Investigation of Lithium Niobate Orientation Effects on Standing Acoustic Waves.

The integration of high-frequency acoustic waves with microfluidics has been gaining popularity as a method of separating cells/particles. A standing surface acoustic wave (sSAW) device produces constructive interference of the stationary waves, demonstrating an increase in cell separating efficiency without damaging/altering the cell structure. The performance of an sSAW device depends on the applied input signal, design of the IDT, and piezoelectric properties of the substrate. This work analyzes the characteristics of a validated 3D finite element model (FEM) of LiNbO3 and the effect on the displacement components of the mechanical waves under the influence of sSAWs by considering XY-, YX-, and 1280 YX-cut LiNbO3 with varying electrode length design. We demonstrated that device performance can be enhanced by the interference of multiple waves under a combination of input signals. The results suggest that 1280 YX-cut LiNbO3 is suitable for generating higher-amplitude out-of-plane waves which can improve the effectiveness of acoustofluidics-based cell separation. Additionally, the findings showed that the length of the electrode impacts the formation of the wavefront significantly.

Acoustofluidic Engineering of Functional Vessel-on-a-Chip.

Construction of in vitro vascular models is of great significance to various biomedical research, such as pharmacokinetics and hemodynamics, and thus is an important direction in the tissue engineering field. In this work, a standing surface acoustic wave field was constructed to spatially arrange suspended endothelial cells into a designated acoustofluidic pattern. The cell patterning was maintained after the acoustic field was withdrawn within the solidified hydrogel. Then, interstitial flow was provided to activate vessel tube formation. In this way, a functional vessel network with specific vessel geometry was engineered on-chip. Vascular function, including perfusability and vascular barrier function, was characterized by microbead loading and dextran diffusion, respectively. A computational atomistic simulation model was proposed to illustrate how solutes cross the vascular membrane lipid bilayer. The reported acoustofluidic methodology is capable of facile and reproducible fabrication of the functional vessel network with specific geometry and high resolution. It is promising to facilitate the development of both fundamental research and regenerative therapy.

Surface acoustic wave-tuned plasmonic resonances in liquid crystal-covered gold nanostructures

In this work, we propose an acoustically tunable plasmonic device based on liquid crystal (LC)-covered gold nanostructures. By utilizing the standing surface acoustic waves (SSAWs) to realign LC molecules, one can effectively tune the effective refractive index of LCs, and subsequently tune localized surface plasmon resonances (LSPRs) of the gold nanostructures. A 15 nm blue shift of the LSPR peak with the driving voltage of 400 mVpp is experimentally observed. Upon removal of the applied SSAWs, the LC molecules can return to their original orientation, and so does the LSPR peak. Besides the excellent reversibility, this active plasmonic device also features low power consumption and easy integration, which could find many potential applications including switches, modulators, and couplers.

Surface Acoustic Wave-Based Microfluidic Device for Microparticles Manipulation: Effects of Microchannel Elasticity on the Device Performance.

Size sorting, line focusing, and isolation of microparticles or cells are fundamental ingredients in the improvement of disease diagnostic tools adopted in biology and biomedicine. Microfluidic devices are exploited as a solution to transport and manipulate (bio)particles via a liquid flow. Use of acoustic waves traveling through the fluid provides non-contact solutions to the handling goal, by exploiting the acoustophoretic phenomenon. In this paper, a finite element model of a microfluidic surface acoustic wave-based device for the manipulation of microparticles is reported. Counter-propagating waves are designed to interfere inside a PDMS microchannel and generate a standing surface acoustic wave which is transmitted to the fluid as a standing pressure field. A model of the cross-section of the device is considered to perform a sensitivity analysis of such a standing pressure field to uncertainties related to the geometry of the microchannel, especially in terms of thickness and width of the fluid domain. To also assess the effects caused by possible secondary waves traveling in the microchannel, the PDMS is modeled as an elastic solid material. Remarkable effects and possible issues in microparticle actuation, as related to the size of the microchannel, are discussed by way of exemplary results.

Acoustofluidic patterning in glass capillaries using travelling acoustic waves based on thin film flexible platform

Surface acoustic wave (SAW) technology has been widely used to manipulate microparticles and biological species, based on acoustic radiation force (ARF) and drag force induced by acoustic streaming, either by standing SAWs (SSAWs) or travelling SAWs (TSAWs). These acoustofluidic patterning functions can be achieved within a polymer chamber or a glass capillary with various cross-sections positioned along the wave propagating paths. In this paper, we demonstrated that microparticles can be aligned, patterned, and concentrated within both circular and rectangular glass capillaries using TSAWs based on a piezoelectric thin film acoustic wave platform. The glass capillary was placed at different angles along with the interdigital transducer directions. We systematically investigated effects of tilting angles and wave characteristics using numerical simulations in both circular and square shaped capillaries, and the patterning mechanisms were discussed and compared with those agitated under the SSAWs. We then experimentally verified the particle patterns within different glass capillaries using thin film ZnO SAW devices on aluminum (Al) sheets. Results show that the propagating SAWs can generate acoustic pressures and patterns in the fluid due to the diffractive effects, drag forces and ARF, as functions of the SAW device’s resonant frequency and tilting angle. We demonstrated potential applications using this multiplexing, integrated, and flexible thin film-based platform, including patterning particles (1) inside multiple and successively positioned circular tubes; (2) inside a solidified hydrogel in the glass capillary; and (3) by wrapping a flexible ZnO/Al SAW device around the glass capillary.

High-temperature characterization of interdigitated transducers on gallium arsenide and surface acoustic wave analysis via interdigitated transducer modeling

Interdigitated transducer devices may provide an advantageous platform to study stress-enhanced interfacial phenomena at elevated temperatures, and an appropriate device design requires a thorough understanding of temperature-dependent material properties. In this study, the scattering parameter response for a surface acoustic wave resonator is simulated using a frequency-domain finite element method from 20 to 177 °C. Experimental device measurements are taken for the interdigitated transducer device fabricated on semi-insulating GaAs 100 oriented in the 110 direction, and the results are in good agreement with the simulation. Surface acoustic wave analysis provides the magnitude of bulk stress values and surface displacement over the experimental temperature range produced by a standing surface acoustic wave. The computational analysis combined with experimental verification suggests that such devices, when optimized for the maximum magnitude, can produce strain levels that could influence chemical potential associated with crystalline growth, atomic diffusion, and catalytic reactions. The modeling results demonstrate an interdigitated transducer's potential as an experimental platform to study the impact of strain on temperature-sensitive surface and bulk phenomena in piezoelectric materials.

On-Chip Label-Free Sorting of Living and Dead Cells.

With the emergence of various cutting-edge micromachining technologies, lab on a chip is growing rapidly, but it is always a challenge to realize the on-chip separation of living cells from cell samples without affecting cell activity and function. Herein, we report a novel on-chip label-free method for sorting living and dead cells by integrating the hypertonic stimulus and tilted-angle standing surface acoustic wave (T-SSAW) technologies. On a self-designed microfluidic chip, the hypertonic stimulus is used to distinguish cells by producing volume differences between living and dead cells, while T-SSAW is used to separate living and dead cells according to the cell volume difference. Under the optimized operation conditions, for the sample containing 50% of living human umbilical vein endothelial cells (HUVECs) and 50% of dead HUVECs treated with paraformaldehyde, the purity of living cells after the first separation can reach approximately 80%, while after the second separation, it can be as high as 93%; furthermore, the purity of living cells after separation increases with the initial proportion of living cells. In addition, the chip we designed is safe for cells and can robustly handle cell samples with different cell types or different causes of cell death. This work provides a new design of a microfluidic chip for label-free sorting of living and dead cells, greatly promoting the multi-functionality of lab on a chip.

Investigation on a cascaded inertial and acoustic microfluidic device for sheathless and label-free separation of circulating tumor cells

High-precision and high-purity acquisition of tumor cells from whole blood is vital for early disease detection and diagnosis. Here, we investigated a cascaded inertial and acoustic microfluidic device for sheathless and label-free separation of circulating tumor cells (CTCs) from the blood through numerical methods. We introduced a spiral microfluidics channel in the first stage (1st) for cell focusing and rough sorting to improve chip integration and reduce the dependence on sheath flow and extra syringe pumps. In the 1st, we simulated the spiral microfluidic with a rectangular cross section to determine the key parameters affecting the migration kinetics of blood cells and tumor cells. Under the influence of Dean drag force and inertial lift force, blood cells migrate toward the inner side of the channel, while CTCs flow out close to the outer side. A flow rate of 400 μl/min was optimized for the operating flow rate. To improve and further enhance the 1st sorting efficiency and purity, we introduced tilted angle standing surface acoustic wave (SSAW) in the second stage (2st). Based on the parametric study, the SSAW with 33.3 MHz, tilted angle with 5°, and acoustic pressure amplitude with 0.7 MPa was selected as the operating parameter. The product of the 1st is used as input for the 2st acoustofluidic unit, enabling a more accurate separation process to obtain CTCs. The simulation results show that the inertial microfluidic units arranged in the first stage help to improve throughput and assist in 2st acoustofluidic separation, and the cascaded chip has accomplished a separation performance of nearly 100% in terms of purity and efficiency.

Velocity and Direction Adjustment of Actuated Droplets Using the Standing Wave Ratio of Surface Acoustic Waves (SAW)

Droplet actuation using surface acoustic wave (SAW) technology has recently been widely employed in “lab-on-a-chip” applications. In this article, SAWs generated by interdigital transducers (IDTs) were used to actuate microdroplets, where their velocity and direction could be adjusted by changing only the excitation phase shift, <italic xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">θ</i> , of the voltage applied to the two IDTs. Specifically, the equation of the vibration of the mixed traveling surface acoustic waves (TSAWs) and standing surface acoustic waves (SSAWs) formed on the substrate surface operating in the exciter–exciter mode has been reported by the authors. An analytical expression for the acoustic standing wave ratio has been derived and given as a function of <italic xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">θ</i> and the spatial phase difference. It can be seen from the expression that the directions of the TSAWs will be opposite to each other, in the range of <italic xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">θ</i> given by (0, π) and (π, 2π), and the component of the TSAWs can be adjusted by changing <italic xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">θ</i> in each direction. Following the theoretical analysis discussed here, two IDTs have been excited to generate a mixture of TSAWs and SSAWs. A series of experiments was carried out to control the velocity and direction of the actuated droplets, by changing only <italic xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">θ</i> . In addition, an experiment performed to compare the techniques shows that the upper limit of the velocity of the actuated droplets can be significantly increased using the exciter–exciter mode, showing that it has the potential to be an alternative method to the traditional exciter–absorber mode.

The effect of microchannel height on the acoustophoretic motion of sub-micron particles

Acoustophoresis is an effective technique for particle manipulation. Acoustic radiation force scales with particle volume, enabling size separation. Yet, isolating sub-micron particles remains a challenge due to the acoustic streaming effect (ASE). While some studies confirmed the focusing ability of ASE, others reported continuous stirring effects. To investigate the parameters that influence ASE-induced particle motion in a microchannel, this study examined the effect of microchannel height and particle size. We employed standing surface acoustic wave (SSAW) to manipulate polystyrene particles suspended in the water-filled microchannel. The results show that ASE can direct particles as small as 0.31 µm in diameter to the centre of the streaming vortices, and increasing the channel height enhances the focusing effect. Smaller particles circulate in the streaming vortices continuously, with no movement towards the centres. We also discovered that when the channel height is at least 0.75 the fluid wavelength, particles transitioning from acoustic radiation-dominated to ASE-dominated share the same equilibrium position, which differs from the pressure nodes and the vortices’ centres. The spatial distance between particles in different categories can lead to particle separation. Therefore, ASE is a potential alternative mechanism for sub-micron particle sorting when the channel height is accurately adjusted.

Suppression of skyrmion Hall effect via standing surface acoustic waves in hybrid ferroelectric/ferromagnetic heterostructures

Magnetic skyrmion is a promising information carrier for its low critical driven current density, topological stability, and small size, which has been proposed for various devices such as racetrack memory and logic gates. However, the skyrmion Hall effect originating from Magnus force leads to transverse motion, which hinders the development of skyrmionic device applications. Here, we propose artificial tracks built by standing surface acoustic waves (SSAWs) to suppress the skyrmion Hall effect through micromagnetic simulations. We systematically study the dynamics of an isolated skyrmion under SSAWs and driven currents in a prototype of the ferromagnetic skyrmion system. The skyrmion Hall angle changes from 80° to 0°, where the skyrmion motion is along the driven current. An analytical model considering magnetoelastic energy induced by SSAWs is developed, and a linear relation between the current density and the critical SSAW amplitude to eliminate the skyrmion Hall effect is achieved. Furthermore, a reconfigurable multichannel skyrmion racetrack is constructed through the change of SSAW wavelengths. Our work opens a feasible route for the suppression of skyrmion Hall effect via SSAWs.

Acoustic prison for single live cell 3D multi-imaging enabled by light-sheet microscopy

Having a system capable of obtaining high resolution, quantitative, subcellular information of individual live cells through imaging, while minimizing cell damage, is of great importance in many fields of research. In recent years, optofluidic light-sheet fluorescent microscopy (LSFM) has emerged as a powerful tool providing a low photo-toxic imaging method utilising the fluidic environment offered by microfluidics. Here, the benefits of LSFM were integrated with an acoustic single cell prison for precise single cell handling and 3D multi-imaging in a semi-automated manner. The compact, monolithic, acousto-optofluidic platform employed standing surface acoustic waves (SSAWs) to sequentially trap an individual cell on either side of an imaging region, which gathered planar, cross-sectional images of the cell. A cytoplasmic stain was utilised to not only visualize the cell trajectory throughout the imaging process, but also to verify the cell viability post-acoustic exposure. A mitochondrial stain was also used to better demonstrate the resolution capabilities of the device. Through post-image processing, 3D volumetric images of the cell were reconstructed and the results between the first and second rounds of imaging were directly comparable. Furthermore, the acoustic prison advantageously positions the cells in the upper region of the channel, enabling the ability to accurately compare temporal changes in cell morphology; a capability that can lead to advancements in therapeutics and drug delivery to access responses of cells to stimuli over time.

Surface acoustic wave induced instability and pattern formation in a liquid polymer film

Recently, standing surface acoustic waves (SAWs) have been demonstrated to stimulate wrinkles on polymer films. Here, we theoretically investigate the underlying mechanism of the acoustic-induced micro-structures on a thin liquid film. The standing SAWs leak into the liquid polymer and generate radiation potential gradients. The strong potential gradients produce acoustic radiation pressure and induce an instability that features the wrinkling structure. Surface stability of the liquid film is firstly investigated by a linear instability analysis and the critical conditions of instability are analytically derived. Theoretical prediction of the film thickness that is favorable for surface instability well agrees with the optimal value found in previous experiments. Balance between the radiation and gravity-capillary pressure at the fluid surface forms the equilibrium equation of the surface profiles. Analytic solution reveals that the component of radiation pressure varying spatially makes the wavelength of the generated surface patterns equal to half of the SAW wavelength, while the constant component of radiation pressure absorbs the film onto the substrate. The interplay between the acoustic waves and the surface morphology is also investigated. Effects of the surface deformation on the radiation pressure is negligible except for that the mean thickness of the liquid approaches the resonance mode where singularity of the radiation pressure occurs.
