Comparative Expression Studies of Fiber Related Genes in Cotton Spp.

Cotton fibers are the seed trichomes that are developed around the seed and are used to make clothes and yarn for the textile industry. Expression profiling of cotton fiber genes is very important to estimate the differential gene expression level at different fiber developmental stages. Expression analysis of fiber developing genes are very important to enhance the fiber length of cotton. The expression profiling of three gene families in five stages (0, 5, 10, 15 and 20 DPA) of cotton fiber tissues was carried out through real-time PCR. Expression analysis revealed that transcripts of GA-20 Oxidase, XTH, and PEPc were elevated from 5 to 20 days post-anthesis (DPA) fibers. Total RNA was extracted from various stages of cotton fiber development and was reverse transcribed to cDNA for PCR amplification. For data normalization, 18s rRNA was used as an internal control. The objective of this study was to explore the expression level of fiber developing genes at specific stages of fiber development. The results showed that most of the genes were expressed during the elongation phase in between 5 DPA to 15 DPA. Results obtained from this study may be helpful for the further identification of fiber genes and the improvement of fiber characteristics in cotton. PEPc and XTH genes that are expressed with a high rate during the fiber development may be used in breeding programmes for the improvement of fiber quality and quantity.

Cloning and functional characterization of auxin receptor TIR1 in Gossypium hirsutum

Transport inhibitor response 1 (TIR1), as an F-box family protein, is a nucleus receptor of auxin in plants. It is essential in the auxin signal transduction pathway and the expression of auxin-responsive genes. As the most important hormone in plants, auxin has a great influence on the ovule and fiber development of cotton (Gossypium hirsutum L.). Studies on the effect of auxin signal transduction pathway on cotton fiber development can fundamentally solve the problem of fiber yield and quality. The cDNA sequence of G. hirsutum TIR1 was obtained through homologous cloning and rapid-amplification of cDNA ends (RACE). The homology between GhTIR1 and that in Arabidopsis thaliana was 64.8%. To study the functions, subcellular localization, and the specific localization of GhTIR1 sequence, we constructed the fusion expression vector of GhTIR1 and its N-terminal or C-terminal deletion mutant genes with GFP. By transiently transforming Arabidopsis protoplasts and stably transforming Nicotiana tabacum BY-2 cell lines, GhTIR1 was localized in the nucleus through the N-terminal 1–197 amino acid sequence. The expression patterns of GhTIR1 in different organs and ovule and cotton fiber cells at different developmental stages showed that GhTIR1 was highly expressed in the initial and elongation stages of cotton fiber development. These results indicate that GhTIR1 plays an important role in the initiation and elongation of cotton fibers, especially the formation of primary fibers of cotton fiber cells.

Auxin Directly Upregulates GhRAC13 Expression to Promote the Onset of Secondary Cell Wall Deposition in Cotton Fibers.

Cotton fibers are single cells that show a relatively independent developmental process of cell differentiation, elongation, and secondary wall deposition. Auxin promotes fiber cell protrusion from the surface of the ovule. However, the role of auxin at other stages of cotton fiber development remains largely unknown. To gain a deeper insight into this aspect, we measured indoleacetic acid (IAA) content in developing fibers. Results showed an increase in IAA content at the transition stage from elongation to secondary cell wall deposition. Subsequently, we investigated the differences between two transgenic cottons that show upregulated and downregulated fiber auxin levels, respectively. In planta analysis revealed that, in addition to promoting cell elongation, auxin regulated the time of initiation of reactive oxygen species (ROS) production and secondary wall deposition in cotton fibers. This was closely correlated with the upregulated expression of GhRAC13, which regulates ROS-triggered cellulose synthesis. We found multiple putative auxin-responsive elements existed within the promoter region of GhRAC13, and IAA could induce proGhRAC13 activity. The dual-luciferase reporter assay further proved the activation of proGhRAC13 by GhARF5, an auxin-signaling activator. Altogether, our results suggest a role of auxin in promoting the onset of secondary growth by directly upregulating GhRAC13 expression in cotton fibers.

Genome-wide association study reveals the genetic basis of fiber quality traits in upland cotton (Gossypium hirsutum L.)

BackgroundFiber quality is an important economic trait of cotton, and its improvement is a major goal of cotton breeding. To better understand the genetic mechanisms responsible for fiber quality traits, we conducted a genome-wide association study to identify and mine fiber-quality-related quantitative trait loci (QTLs) and genes.ResultsIn total, 42 single nucleotide polymorphisms (SNPs) and 31 QTLs were identified as being significantly associated with five fiber quality traits. Twenty-five QTLs were identified in previous studies, and six novel QTLs were firstly identified in this study. In the QTL regions, 822 genes were identified and divided into four clusters based on their expression profiles. We also identified two pleiotropic SNPs. The SNP locus i52359Gb was associated with fiber elongation, strength, length and uniformity, while i11316Gh was associated with fiber strength and length. Moreover, these two SNPs were nonsynonymous and located in genes Gh_D09G2376 and Gh_D06G1908, respectively. RT-qPCR analysis revealed that these two genes were preferentially expressed at one or more stages of cotton fiber development, which was consistent with the RNA-seq data. Thus, Gh_D09G2376 and Gh_D06G1908 may be involved in fiber developmental processes.ConclusionsThe findings of this study provide insights into the genetic bases of fiber quality traits, and the identified QTLs or genes may be applicable in cotton breeding to improve fiber quality.

Investigation of the EIL/EIN3 Transcription Factor Gene Family Members and Their Expression Levels in the Early Stage of Cotton Fiber Development.

The ethylene-insensitive3-like/ethylene-insensitive3 (EIL/EIN3) protein family can serve as a crucial factor for plant growth and development under diverse environmental conditions. EIL/EIN3 protein is a form of a localized nuclear protein with DNA-binding activity that potentially contributes to the intricate network of primary and secondary metabolic pathways of plants. In light of recent research advances, next-generation sequencing (NGS) and novel bioinformatics tools have provided significant breakthroughs in the study of the EIL/EIN3 protein family in cotton. In turn, this paved the way to identifying and characterizing the EIL/EIN3 protein family. Hence, the high-throughput, rapid, and cost-effective meta sequence analyses have led to a remarkable understanding of protein families in addition to the discovery of novel genes, enzymes, metabolites, and other biomolecules of the higher plants. Therefore, this work highlights the recent advance in the genomic-sequencing analysis of higher plants, which has provided a plethora of function profiles of the EIL/EIN3 protein family. The regulatory role and crosstalk of different metabolic pathways, which are apparently affected by these transcription factor proteins in one way or another, are also discussed. The ethylene hormone plays an important role in the regulation of reactive oxygen species in plants under various environmental stress circumstances. EIL/EIN3 proteins are the key ethylene-signaling regulators and play important roles in promoting cotton fiber developmental stages. However, the function of EIL/EIN3 during initiation and early elongation stages of cotton fiber development has not yet been fully understood. The results provided valuable information on cotton EIL/EIN3 proteins, as well as a new vision into the evolutionary relationships of this gene family in cotton species.

Genome-wide analysis of cotton C2H2-zinc finger transcription factor family and their expression analysis during fiber development

BackgroundC2H2-zinc finger protein family is commonly found in the plant, and it is known as the key actors in the regulation of transcription and vital component of chromatin structure. A large number of the C2H2-zinc finger gene members have not been well characterized based on their functions and structure in cotton. However, in other plants, only a few C2H2-zinc finger genes have been studied.ResultsIn this work, we performed a comprehensive analysis and identified 386, 196 and 195 C2H2-zinc finger genes in Gossypium hirsutum (upland cotton), Gossypium arboreum and Gossypium raimondii, respectively. Phylogenetic tree analysis of the C2H2-zinc finger proteins encoding the C2H2-zinc finger genes were classified into seven (7) subgroups. Moreover, the C2H2-zinc finger gene members were distributed in all cotton chromosomes though with asymmetrical distribution patterns. All the orthologous genes were detected between tetraploid and the diploid cotton, with 154 orthologous genes pair detected between upland cotton and Gossypium arboreum while 165 orthologous genes were found between upland cotton and Gossypium raimondii. Synonymous (Ks) and non-synonymous (Ka) nucleotide substitution rates (Ka/Ks) analysis indicated that the cotton C2H2-zinc finger genes were highly influenced mainly by negative selection, which maintained their protein levels after the duplication events. RNA-seq data and RT-qPCR validation of the RNA seq result revealed differential expression pattern of some the C2H2-zinc finger genes at different stages of cotton fiber development, an indication that the C2H2-zinc finger genes play an important role in initiating and regulating fiber development in cotton.ConclusionsThis study provides a strong foundation for future practical genome research on C2H2-zinc finger genes in upland cotton. The expression levels of C2H2-zinc finger genes family is a pointer of their involvement in various biochemical and physiological functions which are directly related to cotton fiber development during initiation and elongation stages. This work not only provides a basis for determining the nominal role of the C2H2-zinc finger genes in fiber development but also provide valuable information for characterization of potential candidate genes involved in regulation of cotton fiber development.

Genome-wide characterization, identification, and expression analysis of the WD40 protein family in cotton.

WD40 repeat proteins are largely distributed across the plant kingdom and play an important role in diverse biological activities. In this work, we performed genome-wide identification, characterization, and expression level analysis of WD40 genes in cotton. A total of 579, 318, and 313 WD40 genes were found in Gossypium hirsutum, G. arboreum, and G. raimondii, respectively. Based on phylogenetic tree analyses, WD40 genes were divided into 11 groups with high similarities in exon/intron features and protein domains within the group. Expression analysis of WD40 genes showed differential expression at different stages of cotton fiber development (0 and 8 DPA) and cotton stem. A number of miRNAs were identified to target WD40 genes that are significantly involved in cotton fiber development during the initiation and elongation stages. These include miR156, miR160, miR162, miR164, miR166, miR167, miR169, miR171, miR172, miR393, miR396, miR398, miR2950, and miR7505. The findings provide a stronger indication of WD40 gene function and their involvement in the regulation of cotton fiber development during the initiation and elongation stages.

Genome-wide comparative and evolutionary analysis of Calmodulin-binding Transcription Activator (CAMTA) family in Gossypium species

The CAMTA gene family is crucial in managing both biotic and abiotic stresses in plants. Our comprehensive analysis of this gene family in cotton resulted in the identification of 6, 7 and 9 CAMTAs in three sequenced cotton species, i.e., Gossypium arboreum, Gossypium raimondii, and Gossypium hirsutum, respectively. All cotton CAMTAs were localized in the nucleus and possessed calmodulin-binding domain (CaMBD) as identified computationally. Phylogenetically four significant groups of cotton CAMTAs were identified out of which, Group II CAMTAs experienced higher evolutionary pressure, leading to a faster evolution in diploid cotton. The expansion of cotton CAMTAs in the genome was mainly due to segmental duplication. Purifying selection played a significant role in the evolution of cotton CAMTAs. Expression profiles of GhCAMTAs revealed that GhCAMTA2A.2 and GhCAMTA7A express profoundly in different stages of cotton fiber development. Positive correlation between expression of these two CAMTAs and fiber strength confirmed their functional relevance in fiber development. The promoter region of co-expressing genes network of GhCAMTA2A.2 and GhCAMTA7A showed a higher frequency of occurrence of CAMTA binding motifs. Our present study thus contributes to broad probing into the structure and probable function of CAMTA genes in Gossypium species.

Molecular weight and organization of cellulose at different stages of cotton fiber development

There is a continuous change in cell wall composition and organization during cotton fiber development. Cotton fiber strength correlates to the molecular weight (MW) and molecular weight distribution (MWD), and organization of cellulose chains in the secondary cell wall. These parameters change drastically during fiber development. This study reports on the MW, MWD, and organization of cellulose in cotton fibers harvested from two cotton cultivars of Gossypium hirsutum L., (Texas Marker-1 and TX55) at different levels of maturity. Fiber dissolution is necessary to estimate the molecular properties of cellulose. Cellulose in mature cotton fibers is larger in MW and highly crystalline and, therefore, poorly dissolves in common solvent systems. To facilitate the dissolution, fibers were first pretreated with 23% sodium hydroxide and then dissolved in a dimethylacetamide/lithium chloride solvent system. Gel permeation chromatography of dissolved fibers indicated that cellulose in both cultivars reaches its maximum MW around 30 days post anthesis. Fourier transform infrared microspectroscopy imaging in the transmission mode indicates changes in cellulose distribution in cotton fibers with fiber development. The distributions of infrared vibrations of cellulose at 897 (β-linkage of cellulose), 1161 (anti-symmetrical C-O-C stretching of cellulose), and 1429 cm−1 (CH2 scissoring of cellulose) provided information on cellulose deposition in intact cotton fibers.

Comparative Proteomic Analysis of Cotton Fiber Development and Protein Extraction Method Comparison in Late Stage Fibers.

The distinct stages of cotton fiber development and maturation serve as a single-celled model for studying the molecular mechanisms of plant cell elongation, cell wall development and cellulose biosynthesis. However, this model system of plant cell development is compromised for proteomic studies due to a lack of an efficient protein extraction method during the later stages of fiber development, because of a recalcitrant cell wall and the presence of abundant phenolic compounds. Here, we compared the quality and quantities of proteins extracted from 25 dpa (days post anthesis) fiber with multiple protein extraction methods and present a comprehensive quantitative proteomic study of fiber development from 10 dpa to 25 dpa. Comparative analysis using a label-free quantification method revealed 287 differentially-expressed proteins in the 10 dpa to 25 dpa fiber developmental period. Proteins involved in cell wall metabolism and regulation, cytoskeleton development and carbohydrate metabolism among other functional categories in four fiber developmental stages were identified. Our studies provide protocols for protein extraction from maturing fiber tissues for mass spectrometry analysis and expand knowledge of the proteomic profile of cotton fiber development.

Small RNA and degradome profiling reveals a role for miRNAs and their targets in the developing fibers of Gossypium barbadense

microRNAs (miRNAs) are 20-24 nucleotide non-coding small RNAs that play important roles in plant development. The stages of cotton fiber development include initiation, elongation, secondary wall thickening (SWT) and maturation. We constructed seven fiber RNA libraries representing the initiation, elongation and SWT stages. In total, 47 conserved miRNA families and seven candidate miRNAs were profiled using small RNA sequencing. Northern blotting and real-time polymerase chain reaction (PCR) analyses revealed the dynamic expression of miRNAs during fiber development. In addition, 140 targets of 30 conserved miRNAs and 38 targets of five candidate miRNAs were identified through degradome sequencing. Analysis of correlated expression between miRNAs and their targets demonstrated that specific miRNAs suppressed the expression of transcription factors, SBP and MYB, a leucine-rich receptor-like protein kinase, a pectate lyase, α-tubulin, a UDP-glucuronic acid decarboxylase and cytochrome C oxidase subunit 1 to affect fiber development. Histochemical analyses detected the biological activity of miRNA156/157 in ovule and fiber development. Suppressing miRNA156/157 function resulted in the reduction of mature fiber length, illustrating that miRNA156/157 plays an essential role in fiber elongation.

Growing degree days is the dominant factor associated with cellulose deposition in cotton fiber

Two field experiments were conducted to study the effect of temperatures on the cellulose content of cotton fiber at various stages of fiber development. In the first study, cotton was sown on three different dates so that temperatures were different during fiber development. In the second study, cotton was grown in semi-mobile chambers and night-time temperatures were controlled within the chambers. During the period from anthesis until the onset of rapid cellulose deposition, the average cellulose deposition rate was significantly correlated with growing degree days (GDD) and daily minimum temperature. The onset time of rapid cellulose deposition was significantly affected by GDD and daily maximum temperature. During the period of rapid cellulose deposition, the duration of rapid cellulose deposition and the average rate of cellulose deposition were significantly correlated with GDD. Therefore, GDD had the largest effect on cellulose deposition cotton fiber. The requisite number of GDD during cellulose synthesis must be reached during two stages of cotton fiber development in order to maximize cellulose content. The average cellulose deposition rate between anthesis and the onset of rapid cellulose deposition can be increased by warmer daily minimum. Warmer daily maximum temperatures advanced the onset of rapid cellulose deposition. The cellulose content of cotton fiber is also be affected by conditions during the period of rapid cellulose deposition. Cellulose contents are highest when cellulose accumulates at moderate rates during this period and when the duration of rapid cellulose deposition is long as possible.

Effects of Increased Night Temperature on Cellulose Synthesis and the Activity of Sucrose Metabolism Enzymes in Cotton Fiber

Temperature is one of the key factors that influence cotton fiber synthesis at the late growth stage of cotton. In this paper, using two early-maturing cotton varieties as experimental materials, night temperature increase was stimulated in the field using far-infrared quartz tubes set in semi-mobile incubators and compared with the normal night temperatures (control) in order to investigate the effects of night temperature on the cotton fiber cellulose synthesis during secondary wall thickening. The results showed that the activity of sucrose synthase (SuSy) and sucrose phosphate synthase (SPS) quickly increased and remained constant during the development of cotton fiber, while the activity of acid invertase (AI) and alkaline invertase (NI) decreased, increased night temperatures prompted the rapid transformation of sugar, and all the available sucrose fully converted into cellulose. With night temperature increasing treatment, an increase in SuSy activity and concentration of sucrose indicate more sucrose converted into UDPG (uridin diphosphate-glucose) during the early and late stages of cotton fiber development. Furthermore, SPS activity and the increased concentration of fructose accelerated fructose degradation and reduced the inhibition of fructose to SuSy; maintaining higher value of allocation proportion of invertase and sucrose during the early development stages of cotton fiber, which was propitious to supply a greater carbon source and energy for cellulose synthesis. Therefore, the minimum temperature in the nightime was a major factor correlated with the activity of sucrose metabolism enzymes in cotton fiber. Consequently, soluble sugar transformation and cellulose accumulation were closely associated with the minimum night temperature.

Identification and Analyses of miRNA Genes in Allotetraploid Gossypium hirsutum Fiber Cells Based on the Sequenced Diploid G. raimondii Genome

The plant genome possesses a large number of microRNAs (miRNAs) mainly 21-24 nucleotides in length. They play a vital role in regulation of target gene expression at various stages throughout the whole plant life cycle. Here we sequenced and analyzed ≈ 10 million non-coding RNAs (ncRNAs) derived from fiber tissue of the allotetraploid cotton (Gossypium hirsutum) 7 days post-anthesis using ncRNA-seq technology. In terms of distinct reads, 24 nt ncRNA is by far the dominant species, followed by 21 nt and 23 nt ncRNAs. Using ab initio prediction, we identified and characterized a total of 562 candidate miRNA gene loci on the recently assembled D(5) genome of the diploid cotton G. raimondii. Of all the 562 predicted miRNAs, 22 were previously discovered in cotton species and 187 had sequence conservation and homology to homologous miRNAs of other plant species. Nucleotide bias analysis showed that the 9th and 1st positions were significantly conserved among different types of miRNA genes. Among the 463 putative miRNA target genes, most significant up/down-regulation occurred in 10-20 days post-anthesis, indicating that miRNAs played an important role during the elongation and secondary cell wall synthesis stages of cotton fiber development. The discovery of new miRNA genes will help understand the mechanisms of miRNA generation and regulation in cotton.

Characterization of PROFILIN genes from allotetraploid (Gossypium hirsutum) cotton and its diploid progenitors and expression analysis in cotton genotypes differing in fiber characteristics

The actin-binding protein profilin (PRF) plays an important role in cell growth and expansion by regulating the organization of the actin filaments. Recent studies have reported association between fiber elongation in cultivated cotton (Gossypium hirsutum) and PRF expression. In the present study, we cloned four genomic clones from allotetraploid cotton (G. hirsutum) and its putative diploid progenitors (G. arboreum and G. raimondii) designated GhPRF1_A, GhPRF1_D, GaPRF1, and GrPRF1 encoding cotton PRF and characterized their genomic structure, phylogenetic relationships and promoter structure. Sequence analysis of the coding regions of all clones resulted in a single protein product which revealed more than 80% similarity to most plant PRFs and a typical organization with an actin-binding and a polybasic phospholipid binding motif at the carboxy terminus. DNA blot hybridization suggested that PRF gene is present with more than one copy in the allotetraploid species G. hirsutum. Expression analysis performed in various organs of cultivated cotton revealed that the PRF gene was preferentially expressed in cotton fibers. Very low levels of expression were observed in whole flowers, while PRF transcripts were not detected in other organs examined. Furthermore, higher levels of expression were observed at the early stages of cotton fiber development (at 10days post anthesis), indicative that this gene may play a major role in the early stages of cotton fiber development. Quantitation of the expression by real-time PCR revealed higher expression levels in a G. hirsutum variety with higher fiber percentage compared to a variety with lower percentage. In addition, higher levels of expression were found in cultivated allotetraploid G. barbadense cotton species with higher fiber length in comparison to cultivated allotetraploid G. hirsutum.

Characterization and Promoter Analysis of a Cotton RING-Type Ubiquitin Ligase (E3) Gene

A cotton fiber cDNA, GhRING1, and its corresponding gene have been cloned and characterized. The GhRING1 gene encodes a RING-type ubiquitin ligase (E3) containing 338 amino acids (aa). The GhRING1 protein contains a RING finger motif with conserved cysteine and histine residues at the C-terminus, and is classified as a C(3)H(2)C(3)-type RING protein. Blast searches show that GhRING1 has the highest homology to At3g19950, a zinc finger family protein from Arabidopsis. Real time RT-PCR analysis indicates that the GhRING1 gene is expressed in cotton fibers in a developmental manner. The transcript level of GhRING1 gene reaches a maximum in elongating fibers at 15 days post-anthesis (DPA). In vitro auto-ubiquitination assays using wheat germ extract and a reconstitution system demonstrate that GhRING1 has the ubiquitin E3 ligase activity. The histochemical GUS assay was performed to analyze tissue specificity of the GhRING1 and At3g19950 promoters in transgenic Arabidopsis plants. The GUS assay shows that the promoter of At3g19950 is highly activated in leaves, roots, trichomes, and also in anthers and stigma of flowers. In contrast, the GUS expression directed by the GhRING1 promoter is only located at stipules and anthers. The expression pattern of GhRING1 suggests that protein ubiquitination and turnover may be involved in transition to different stages of cotton fiber development.

Phylogenetically Distinct Cellulose Synthase Genes Support Secondary Wall Thickening in Arabidopsis Shoot Trichomes and Cotton Fiber

Abstract Through exploring potential analogies between cotton seed trichomes (or cotton fiber) and arabidopsis shoot trichomes we discovered that CesAs from either the primary or secondary wall phylogenetic clades can support secondary wall thickening. CesA genes that typically support primary wall synthesis, AtCesA1,2,3,5, and 6, underpin expansion and secondary wall thickening of arabidopsis shoot trichomes. In contrast, apparent orthologs of CesA genes that support secondary wall synthesis in arabidopsis xylem, AtCesA4,7, and 8, are up-regulated for cotton fiber secondary wall deposition. These conclusions arose from: (a) analyzing the expression of CesA genes in arabidopsis shoot trichomes; (b) observing birefringent secondary walls in arabidopsis shoot trichomes with mutations in AtCesA4, 7, or 8; (c) assaying up-regulated genes during different stages of cotton fiber development; and (d) comparing genes that were co-expressed with primary or secondary wall CesAs in arabidopsis with genes up-regulated in arabidopsis trichomes, arabidopsis secondary xylem, or cotton fiber during primary or secondary wall deposition. Cumulatively, the data show that: (a) the xylem of arabidopsis provides the best model for secondary wall cellulose synthesis in cotton fiber; and (b) CesA genes within a "cell wall toolbox" are used in diverse ways for the construction of particular specialized cell walls.

Cloning and Characterization of a CAP Gene Expressed in Gossypium arboreum Fuzzless Mutant

The adenylyl cyclase‐associated protein (CAP) was first identified in the budding yeast as a protein that functions in cytoskeletal organization and signal transduction. We isolated two CAP homologs from wild‐type cotton Gossypium arboreum L. (DPL971) and its natural fuzzless mutant (DPL972). Alignments of 3998‐bp genomic and 1425‐bp cDNA sequences revealed that the cap genes contain nine introns and an open reading frame (ORF) encoding a 471 amino acid protein. The predicted amino acid sequence shares high identity (∼70%) with Arabidopsis thaliana, Medicago truncatula, and Oryza sativa Expression profiles of the gene were studied from ovule, leaf, hypocotyl, and root tissues, through reverse transcriptase polymerase chain reaction and Northern blotting analysis. The results revealed that the gene was expressed in all tissues tested in wild‐type and fuzzless plants. The expression of GaCAP reached the highest level in both wild‐type and mutant fibers during 1 to 4 d postanthesis (DPA), then gradually declined in the mutant after 4 DPA. By comparing the predicted amino acid sequences, a polar substitution was observed at a conserved position between wild‐type and mutant. At this position, the conserved uncharged unpolar alanine (amino acid 44) of GaCAP was replaced by a polar threonine in GaCAPm. Therefore, our results suggest that GaCAP may play a functional role during early stages of cotton fiber development.

Cotton fiber germin-like protein. I. Molecular cloning and gene expression.

The presence of cotton ( Gossypium hirsutum L.) fiber transcripts coding for a germin-like protein (GLP) was revealed by differential display analysis in which early stages of cotton fiber development between a wild type line, Texas Marker-1 (TM1) and a near isogenic mutant, Naked Seed (N1) were compared. Transcripts of the cotton GLP ( GhGLP1) accumulated specifically in TM1, but did not accumulate in the mutant although the GhGLP1 gene was present in both lines. The deduced protein sequence of GhGLP1 is similar to Prunus persica auxin-binding proteins, a barley ADP-glucose pyrophosphatase/phosphodiesterase and two different classes of hydrogen peroxide-producing enzymes: wheat germin oxalate oxidase and moss extracellular Mn-superoxide dismutase. Cotton GLPs constitute a multigene family like those of Arabidopsis, rice, soybean, and barley. GhGLP1 transcripts accumulated to their highest levels during the period of fiber expansion, followed by a sharp decline when the rate of cell expansion decreased. While germins and GLPs appear to be involved in defense mechanisms in some plants, both biotic and abiotic stress down-regulated the expression of GhGLP1. Numerous functions have been proposed for dicot GLPs. However, to date, there is little direct evidence for how these proteins function in vivo. The association of maximal GhGLP1 expression with stages of maximal cotton fiber elongation suggests that some GLPs may be important for cell wall expansion.

Nucleolar size at early stages of cotton fiber development in relation to final fiber dimension

Changes in nucleolar size and nucleolar vacuolation at early stages of fiber development and final fiber dimensions were determined for cotton of different species: Gossypium hirsutum L. cv. B49, Gossypium barbadense L. cv. Menoufi and Gossypium arboreum L. cv. Virnar. Size of the nucleolus in combination with its vacuolation at an early stage of development was found to be clearly associated with the final result of fiber development.
