Spiroannulations of β-Ketothioamides with Bromoenals via Selective C-Michael Addition and S-Michael Addition-Triggered Cascade Reactions.

A spiroannulation reaction of β-ketothioamides with aromatic β-bromoenals and aromatic α-bromoenals via selective C-Michael addition and S-Michael addition-triggered cascade reactions has been developed. This protocol provides a novel and rapid approach for the synthesis of substituted spirothiopyran and spirothiophene derivatives under mild conditions with moderate to good yields and a broad substrate scope.

Electrophilic aryl spiroannulation induced by diaryliodonium salts: efficient synthesis of [5,5]-spiro lactone-lactam scaffolds from α-cyanocarboxylates

Spirocyclic architectures represent one of the privileged three-dimensional scaffolds in pharmaceutical chemistry and drug discovery, while the new-skeletal synthesis of spiromolecules from simple starting materials remains a challenging task. Here, we report a first diaryliodonium salts mediated electrophilic aryl spiroannulation reaction from simple substrates α-cyanocarboxylate. With this strategy, a broad spectrum of new [5,5] spiro-fused skeletons could be efficiently constructed in one step via Cu-catalyzed cascaded N-arylation-acylation and etheral skeleton rearrangement reaction. The key point of this transformation was the intramolecular electrophilic cyclization due to the nucleophilicity of O-atom and electrophilicity of α-C induced by diaryliodonium salts. With this protocol, the more challenging activation of ether bonds was achieved via diaryliodonium salts-mediated. This method has the major advantages of a broad substrate scope and excellent functional group compatibility. Further synthesis elaboration was performed using substrate with steric hindrance to showcase the versatility of this methodology.

Trivalent Phosphine-Catalyzed [4+1] Spiro-annulation Reaction Using Allenyl Imide and Methylene Cyclocompounds.

The trivalent phosphine-catalyzed [4+1] spiro-annulation reaction of allenyl imide and activated methylene cyclocompounds has been developed for the construction of various spiro-2-cyclopenten-1-ones. Oxindoles, 3-isochromanones, and 2-indanones are selected as 1C synthons to capture the in situ-generated bis-electrophilic α,β-unsaturated ketenyl phosphonium intermediate, affording the corresponding monospiro- and bispiro-cyclopentenones in good to excellent yields (≤91%) under mild conditions. The primary attempt at asymmetric catalysis using monophosphine (R)-SITCP provides promising enantioselectivity (45% ee). A plausible reaction mechanism is also proposed.

Dearomative spiroannulation of indoles enabled by the diaza-[1,2]-Wittig rearrangement

Reported herein is the first diaza-[1,2]-Wittig rearrangement-enabled dearomative spiroannulation reaction of indoles. This protocol features metal-free conditions, a redox-neutral process, a broad substrate scope, and a good yield.

C-H activation-initiated spiroannulation reactions and their applications in the synthesis of spirocyclic compounds.

Spirocyclic skeletons are prevalent in natural products, pharmaceuticals and organic functional materials. Meanwhile, transition-metal-catalyzed C-H activation reactions have demonstrated unparalleled advantages such as high efficiency, excellent atom-economy, good chemoselectivity and regioselectivity for the formation of target organic molecules. In recent years, C-H activation reactions have been creatively utilized in the synthesis of spirocyclic compounds. This review summarizes the most recent progress made in C-H activation-initiated spiroannulation reactions and their applications in the construction of structurally diverse and biologically valuable spirocyclic scaffolds by using alkynes, diazo compounds, maleimides, alkenes, quinones and cyclopropenones as the coupling partners.

Acid-Catalyzed [4+1]-Dearomatization Spiroannulation of Hydroquinones and Naphthols

AbstractAcid-catalyzed [4+1]-dearomatization spiroannulation reactions of electron-rich hydroquinone and naphthol derivatives were demonstrated as convenient methods to access spirocyclic cyclohexadienone derivatives. The Lewis acid Bi(OTf)3 exhibited the best catalytic performance when a 1,2-dialkynylbenzene was used as the electrophile for the spiroannulation, whereas the Brønsted acid benzene-1,2-disulfonic acid was the best catalyst when 2-ethynylbenzylic esters were used as electrophiles. Most of the reaction conditions are mild, efficient, and simple to operate.

Rh(III)-Catalyzed Spiroannulation Reaction of N-Aryl Nitrones with 2-Diazo-1,3-indandiones: Synthesis of Spirocyclic Indole-N-oxides and Their 1,3-Dipolar Cycloaddition with Maleimides.

An efficient strategy for the preparation of spirocyclic indole-N-oxide compounds through a Rh(III)-catalyzed [4 + 1] spiroannulation reaction of N-aryl nitrones with 2-diazo-1,3-indandiones as C1 synthons under extremely mild conditions is presented. From this reaction, 40 spirocyclic indole-N-oxides were easily obtained in up to 98% yield. In addition, the title compounds could be successfully used for the construction of structurally intriguing maleimide-containing fused polycyclic scaffolds via a diastereoselective 1,3-dipolar cycloaddition reaction with maleimides.

Synthesis of indene-fused spiro-dibenz(ox)azepines via Rh(III)-catalyzed cascade regioselective C-H activation/annulation.

We report an unprecedented atom-economic one-pot Cp*Rh(III)-catalyzed regioselective [3+2]-spiroannulation reaction between dibenz(ox)azepines and ynones, allowing the synthesis of biologically relevant novel spirocyclic dibenz(ox)azepines under operationally simple and mild reaction conditions. The reaction proceeds without any silver additive or external oxidant implementing a redox-neutral pathway. A broad substrate scope with diverse functional group tolerance permitted the regioselective synthesis of a wide spectrum of indene-containing spirocyclic dibenz(ox)azepines in good to excellent yields. Also, we showcased detailed mechanistic studies to justify the formation of spirocycles. In addition, the synthetic utility of this process was also demonstrated by the modular synthesis of various steroid conjugates.

Concise Synthesis of Spirocyclic Dihydrophthalazines through Spiroannulation Reactions of Aryl Azomethine Imines with Cyclic Diazo Compounds.

Spiroannulation reactions are fundamental and invaluable for the synthesis of spirocyclic compounds. Presented herein are novel cascade reactions of aryl azomethine imines with cyclic diazo compounds leading to the formation of spirocyclic dihydrophthalazine derivatives. Based on experimental mechanistic studies, the formation of the title products is believed to go through azomethine imine-assisted cylcometalation, Rh-carbene formation through dediazonization, and migratory insertion followed by reductive elimination and azomethine imine ring opening. Control experiments revealed that air acts as an effective and sustainable co-oxidant to facilitate the cascade reaction. In general, this concise synthesis of the unprecedented spirocyclic dihydrophthalazine derivatives has advantages such as easily accessible substrates, good functional group compatibility, mild reaction conditions, high efficiency and selectivity, and excellent atom-economy. In addition, the value of this protocol is underlined by its ready scalability and divergent derivation of products.

Regio- and Diastereoselective [3 + 2]-Spiroannulation of Benzoxazines with Chalcones: A Rh(III)-Catalyzed Redox-Neutral Approach to α-Aroyl Spiro-Indanamines

We report an atom-economic Rh(III)-catalyzed [3 + 2]-spiroannulation reaction between cyclic ketimines and α,β-unsaturated carbonyl compounds, allowing the synthesis of novel spirocycles with concomitant generation of three stereogenic centers in one pot. The reaction does not require any silver additives or external oxidants and is believed to proceed in a redox-neutral manner. A broad substrate scope with good functional group tolerance permitted the synthesis of a vast spectrum of spirocyclic 1,4-benzoxazine derivatives containing polysubstituted α-aroyl-indanamines in good to excellent yields with high diastereoselectivity.

Visible‐Light‐Mediated Synthesis of 1‐Oxa‐4‐aza‐spiro Oxazolines by Spiroannulation of Quinones with Vinyl Azides

AbstractA simple and efficient method has been developed to synthesize 1‐oxa‐4‐aza‐spirooxazolines. The reaction was carried out at room temperature using rose bengal as an organic photoredox catalyst and blue LED as a light source. It was observed that quinones underwent spiroannulation reaction with vinyl azide on C−O double bond instead of C−C double bond through which various corresponding 1‐oxa‐4‐aza‐spirooxazolines have been synthesized in good to excellent yields.

4-Dimethylaminopyridine-catalyzed [3 + 3] spiroannulation reactions of isatin-derived Morita-Baylis-Hillman carbonates with indoline-2-thiones

DMAP-catalyzed diastereoselective [3 + 3] spiroannulations of isatin-derived Morita-Baylis-Hillman carbonates with indoline-2-thiones were achieved. A series of bisindole-annulated thiopyran frameworks were synthesized in good yields with good diastereoselectivities. The strategy developed in this paper offered a novel way for the synthesis of valuable indole-annulated sulfur heterocycle derivatives which might be beneficial in organic synthesis and medicinal chemistry.

Diastereoselective access to [4,4]-carbospirocycles: governance of thermodynamic enolates with an organocatalyst in vinylogous cascade annulation.

A vinylogy concept driven annulation strategy is developed to access [4,4]-carbospirocycles from alkylidene malononitriles and cyclopentene-1,3-diones. The reaction is catalyzed by an inexpensive organocatalyst and products with three stereocenters were obtained as a single diastereomer in high yields. The spiro-selectivity originates from the reaction of the thermodynamic enolate intermediate which is fundamentally intriguing.

Asymmetric [3+2] spiroannulation of pyrazolone-derived Morita-Baylis-Hillman carbonates with alkynyl ketones: facile access to spiro[cyclopentadiene-pyrazolone] scaffolds.

A tertiary amine-catalyzed asymmetric [3+2] spiroannulation reaction of pyrazolone-derived Morita-Baylis-Hillman carbonates with alkynyl ketones was achieved under mild conditions. This protocol offers a facile approach to chiral spiro[cyclopentadiene-pyrazolone] scaffolds in good to high yields (up to 92%) with good degrees of enantiocontrol (up to 98% ee). In addition, scale-up reaction and transformation of the products were performed to show the synthetic utility of this protocol.
