Spectrophotometric Determination of Norepinephrine in Industrial Equipment Cleaning Rinse Water by Its Own Absorbance and The Tollens’ Reagent

A literature review of the available spectrophotometric methods of determination of catecholamines is presented. Based on this review, two simple spectrophotometric methods for the determination of norepinephrine bitartrate in pharmaceutical formulations and industrial equipment cleaning rinse waters were chosen and validated. The first method for the determination of norepinephrine at ppm levels utilizes the absorbance band of the solution of norepinephrine bitartrate at 279 nm. The second method for the determination of norepinephrine at ppb levels is based on the reaction of norepinephrine with the Tollens’ reagent in the presence of the cationic surfactant cetylpyridinium chloride and the colorimetric determination of the formed colloidal silver nanoparticles. The calibration graphs are linear in the range from 1 to 150 mg/l of norepinephrine for the first method and 25 µg/l to 10 mg/l for the second method. The molar attenuation coefficients are 388 and 7560 m2/mol, respectively, the limits of detection are 1.1 mg/l and 3.1 µg/l, and the limits of quantification are 3.3 mg/l and 9.4 µg/l, respectively. Both methods are selective concerning the common excipients, show good accuracy (the relative uncertainties do not exceed 4 and 7%, respectively) and precision (the relative standard deviations do not exceed 4 and 7%, respectively), and do not require lengthy sample preparation and sophisticated laboratory equipment.

Quantification of morphine in exhaled breath condensate using a double network polymeric hybrid hydrogel functionalized with AuNPs

BackgroundMorphine serves as a foundation for creating other opioid derivatives, such as hydro/oxymorphine and heroin, which possess enhanced pain-relieving properties but are also prone to addiction and abuse. In cases of morphine overdose, it not only affects multiple immune functions but can also cause severe health complications. Given these concerns and the widespread use of morphine, it is crucial to develop efficient, uncomplicated, and precise methods for accurately detecting morphine in various biological and pharmaceutical samples.ResultsIn this investigation, a novel gold nanoparticle (AuNPs)-based double network hydrogel (DNH) nanoprobe has been fabricated for sensitive quantification of morphine in exhaled breath condensate samples. For that, gelatin/agarose DNH was fabricated through a one-step heating-cooling method in the presence of AuNPs, providing not only chemical stability but also prevent the AuNPs aggregation during synthesis process. In this method, the absorbance intensity of the nanoprobe gradually decreased with increasing morphine concentration due to the interaction morphine with AuNPs surface plasmon. The aggregation of AuNPs by addition of morphine was verified by UV-Vis spectrophotometry. The sensor displayed high sensitivity with detection limit of 0.006 µg.mL-1 in the linear range from 0.01 to 1.0 µg.mL-1. A reliable performance was attained for the spectrophotometric method for determination of morphine in the real samples.

Ascorbic acid and folic acid are frequently included together in multivitamin pharmaceutical products due to their complementary roles in maintaining human health. The quality and efficiency of supplements can vary depending on the manufacturer and the specific ingredients used, which are vital factors to consider. This research presents the development and validation of a simple, sensitive, accurate, and environmentally friendly derivative spectrophotometric technique utilizing the zero-crossing method for the simultaneous determination of ascorbic acid and folic acid, both in their pure form and in pharmaceutical formulations. The derivative spectrophotometry technique demonstrated measurable amplitude and substantial linearity for both analytes. At the zero-crossing points of folic acid (249.6 and 281 nm), ascorbic acid showed a measurable amplitude with a linear range of 0.17–12.0 μg/mL and 0.37–12 μg/mL, and detection limits of 0.057 μg/mL and 0.121 μg/mL, respectively. Conversely, folic acid displayed an observable amplitude at the zero-crossing point of ascorbic acid (265.6 nm) with a linear range from 0.26 to 15.0 μg/mL and a detection limit of 0.088 μg/mL.The greenness of the proposed derivative spectrophotometric method was evaluated using the White Analytical Chemistry (WAC) framework and the Green Analytical Procedure Index (GAPI). The method scored highly in both evaluations, confirming its minimal environmental impact and alignment with the principles of Green Analytical Chemistry. The GAPI assessment highlighted low environmental and health risks throughout the analytical process, making this method a green and sustainable choice for the simultaneous determination of ascorbic acid and folic acid. The method was successfully applied to synthetic laboratory mixtures and formulated tablets, showing excellent precision, accuracy, and recovery, while also being environmentally friendly.

Validation of the spectrophotometric method for the quantitative determination of nimesulide in solid dispersions systems obtained by centrifugal fiber formation

In order to increase the water solubility of the non-steroidal anti-inflammatory active pharmaceutical ingredient, nimesulide, solid dispersed systems were developed using the innovative technology of centrifugal fiber formation. Accordingly, in order to obtain correct results regarding the degree of increase in the solubility of nimesulide in solid dispersed systems, it is necessary to select a method for determining the quantitative content of the active pharmaceutical ingredient in the developed polymer composites. The aim of the study was to develop and validate a spectrophotometric method for the quantitative determination of nimesulide in solid dispersed systems obtained by centrifugal fiber formation. A small commercially available cotton candy machine «Cotton candy maker» (China) was used to produce solid dispersed systems of nimesulide by the method of centrifugal fiber formation. Spectrophotometric studies were performed using an OPTIZEN POP UV spectrophotometer (Meсasys, South Korea) in quartz glass cuvettes with an optical layer thickness of 1 cm (Agilent, USA). The developed spectrophotometric method for the quantitative determination of nimesulide content in centrifugally formed polymeric solid dispersed systems is based on the qualitative reaction of nimesulide with a 0.1 M sodium hydroxide solution, resulting in the formation of a yellow compound with a maximum optical absorption at 400 nm. The validation characteristics of the proposed method, in particular, specificity, linearity, precision, and robustness, were investigated. It was confirmed that the wavelength of 400 nm is specific for the analysis. It was proved that the method is linear (R2 = 0.999) in the range of nimesulide concentrations 80–120%. The accuracy of the method was confirmed at the level of convergence and reproducibility. When changing the wavelength by ± 2 nm, the standard deviation is 0.12%, which indicates the robustness of the developed method. Thus, it is proved that all the selected validation characteristics of the developed spectrophotometric method for determining the quantitative content of nimesulide in centrifugally formed polymeric solid dispersed systems meet the acceptance criteria, which confirms the validity of the method.

The use of a hypromellose based sorbent for a solid-phase extraction in the quantitative determination of procaine in combination drugs

Aim – to assess the selectivity and metrological characteristics of the spectrophotometric method for procaine determination in combination drugs using a solid-phase extraction sorbent based on hypromellose. Material and methods. The solid-phase extraction sorbent was obtained with hypromellose and ethyl cyanoacrylate; the procaine hydrochloride and sodium hydroxide formed the active centers. The sorbent was prepared using a standard method. The sorption capacity of the obtained sorbent samples for procaine was 10.2 ± 1.0 µg/g. For the study, we selected the combination drugs containing procaine and its salts, such as "Menovazin", "Sulfocamphocaine", and "Otolorin". The eluate optical density was registered using a SF-56 spectrophotometer (OKB SPECTR LLC) at an analytical wavelength of 292 nm in a cuvette with an optical layer thickness of 1 cm. The reference solution was a 0.1 M hydrochloric acid solution. To calculate the procaine content in the eluates, a calibration graph (dependence of eluate optical density on procaine concentration) was plotted in the concentration range of 1–20 mcg/ml. To evaluate the selectivity (specificity) of sample preparation in chromatographic separation of the combination drugs, the absorption spectra were registered in the wavelength range of 200–400 nm for eluates obtained using purified water and a 0.1 M hydrochloric acid solution. The procaine and benzocaine were identified in the eluate by the presence of absorption maxima at wavelengths of 222 and 286 nm for benzocaine, and 228 and 292 nm for procaine. For statistical processing of the experimental results and determination of metrological characteristics of the analysis method, we followed the GPM.1.1.0013 "Statistical processing of results of physical, physicochemical, and chemical tests". Results. We have demonstrated the selectivity of the spectrophotometric determination of procaine in certain combination drugs using a solid-phase extraction sorbent based on hypromellose. The selective binding of the analyzed substance with active centers in the sorbent and stepwise elution with purified water and a 0.1 M hydrochloric acid solution during solid-phase extraction eliminated the interfering effect of drug components during the UV-spectrophotometry of procaine. The use of a hypromellose based sorbent improved the metrological characteristics and reduced the systematic error of the quantitative determination method by an average of 14.4%. The proposed methodological approach using a selective sorbent might be recommended as the quality control for combination drugs containing procaine and its salts.

Method of rapid spectrophotometric determination of iron in dithionite extracts of soils

Amethod for the determination of dithionite iron for soil samples and fractions of the order of 20–25 mg with a spectrophotometric termination has been developed and optimized.Unlike the previously described methods, phenanthroline was used as a chromogen, which made it possible to simplify the analysis by eliminating the combustion stage.The procedure for processing samples with citrate-bicarbonate-dithionite extractant has been optimized.The repeatability and intra-laboratory reproducibility of the technique are investigated.Verification of the proposed methodology by comparison with the standard one showed good convergence of the results.

Introduction of a spectrophotometric method for salivary iodine determination on microplate based on Sandell-Kolthoff reaction.

Iodine is an essential element for the synthesis of thyroid hormones. Therefore, a reliable marker of iodine supply is important. Iodine is predominantly excreted via kidneys, but also via salivary glands. Our aim was to introduce a new and simple method for determination of salivary iodine concentration (SLIC). Self-prepared chemicals and standards for Sandell-Kolthoff reaction on microplate with ammonium peroxydisulfate (AP) in the range 0-400 µg/L were used. Suitability of water-based standards (WBS) and artificial saliva-based standards (ASS) for standard curve were tested. We followed standards for method validation, defined concentration of used AP and compared our results with Inductively Coupled Plasma Mass Spectrometry (ICP-MS). WBS gave more reliable results than ASS as an underestimation of iodine concentration was found for ASS. LoB was 6.5 µg/L, LoD 12.0 µg/L, therefore analytical range was 12-400 µg/L. Intra- and inter-assay imprecisions at iodine concentrations, namely 20, 100, 165, and 350 µg/L were 18.4, 5.1, 5.7, and 2.8%, respectively, and 20.7, 6.7, 5.1, and 4.3%, respectively. Suitable molarity of AP was 1.0 mol/L and showed no difference to 1.5 mol/L (P values for samples with concentration 40, 100, and 150 µg/L, were 0.761, 0.085, and 0.275, respectively), whereas there was a significant change using 0.5 mol/L (P<0.001). Saliva samples could be diluted up to 1:8. There was no interference of thiocyanate and caffeine up to 193.5 mg/L. Our original method was comparable to ICP-MS. Spaerman coefficient was 0.989 (95% CI: 0.984-0.993). The new method for SLIC determination is in excellent agreement with ICP-MS and easy-to-use.

Simultaneous deep eutectic solvent based microextraction of Allura Red and Brilliant Blue in violet-purple foodstuffs

A vortex-assisted simultaneous deep eutectic solvent based microextraction and spectrophotometric determination method was established for the first time to simultaneously monitor Allura Red and Brilliant Blue in violet-purple foodstuffs. A ternary deep eutectic solvent was created using benzoic acid, tetra butyl ammonium bromide and benzyl alcohol at 1:1:3 mol ratio. The method variables including pH, volume of deep eutectic solvent and vortex time were optimized by Box-Behnken design of response surface methodology. The limits of detection were determined to be 25 and 9 µg/L for Allura Red and Brilliant Blue, respectively. The linear dynamic ranges of the method were obtained between 82 and 12,000 and 29–4200 µg/L for Allura Red and Brilliant Blue, respectively. Preconcentration factor of the method was obtained as 15. Greenness evaluation was performed using Analytical GREEnness tools. Extraction mechanisms affecting quantitative simultaneous microextraction of the target dyes were explained. The developed method was validated with high-performance liquid chromatography analysis. Analyte addition-recovery tests were also applied to confectionery samples to validate the proposed method and determine the dye contents of foodstuffs. Allura Red and Brilliant Blue contents of violet-purple confectioneries were determined between 3.85 and 74.59 and 4.02–26.11 µg/g concentrations with 94.5–98.6 % and 93.0–101.0 % recoveries, respectively.

LOW TEMPERATURE ANALYTICAL CLOUD POINT EXTRACTION. 2: PRECONCENTRATION AND SPECTROPHOTOMETRIC DETERMINATION OF GE(IV)

Cloud point extraction is a powerful tool for the analytical preconcentration of trace amounts of analytes. The standard cloud point extraction procedure requires prolonged heating of the solution, which is not always appropriate in the case of thermally unstable analytes or analytical forms and significantly limits the capabilities of the method. One of the ways to modify cloud point extraction is to find approaches to intensifying the process of analytical concentration. The main disadvantage of cloud point extraction is the need for thermal, ultrasonic, infrared or any other initiation of the formation of the surfactant rich phase. It has been shown that salts of aromatic carboxylic acids can cause instantaneous formation of a surfactant-rich phase. In this work, the conditions of micellar extraction concentration of Ge(IV) in the form of a complex with 6,7-dihydroxy-2-phenyl-4-methylbenzopyrylium bromide into the micellar phase of the nonionic surfactant triton X-100 were studied and optimized. It is shown that the introduction of ammonium benzoate into the system at a pH of 1.0 and a triton X-100 concentration of 0.5 vol.% leads to the initiation of the formation of a micellar phase at room temperature. The method of spectrophotometric determination of Ge(IV) with 6,7-dihydroxy-2-phenyl-4-methylbenzopyrylium bromide after its micellar- extraction concentration has been developed. The calibration graph is linear in the concentration range of 4.36–472 μg/L, and the limits of detection and determination are 1.31 and 4.36 μg/L, respectively. The proposed method was tested in the analysis of model solutions and biologically active additives, and the relative standard deviation does not exceed 5.1%.

Green and Versatile High-Throughput Microwell Oxidation-Based Spectrophotometric Methods for Determination of Galidesivir in Capsules.

Galidesivir (GDV) is a promising new antiviral drug for the potent and safe treatment of a broad spectrum of viral diseases, including COVID-19. In the literature, no analytical method exists for the determination of GDV in bulk and dosage form. The aim of this study was the development of versatile green and simple microwell spectrophotometric methods (MW-SPMs) for the determination of GDV in its bulk form and capsules. Three MW-SPMs were developed involving the oxidation of GDV by ammonium metavanadate (AMV), chromium trioxide (CTO), and potassium iodate (PIO) in an acid medium. The reactions were carried out in 96-well plates at room temperature and the absorbances of chromogenic reaction products were measured by an absorbance microplate reader at 780, 595, and 475 nm for AMV, CTO, and PIO, respectively. Variables influencing the reactions were carefully investigated and optimized. Linear relations with excellent correlation coefficients (0.9991-0.9997) were found between the absorbances and GDV concentrations in the range of 25-500 µg/mL. The LODs were ≥8.3 µg/mL. The accuracy and precision of the three MW-SPMs were confirmed by recovery and replicate analysis, respectively. The recovery values were 98.6-101.2% and the RSDs were ≤1.02%. The proposed MW-SPMs were successfully applied to the analysis of GDV in bulk drug and capsules with high accuracy and precision. The greenness of the MW-SPMs was confirmed by three comprehensive metric tools. The proposed MW-SPMs combined the inherent advantages of microwell-based analysis and the use of common laboratory reagents for the reactions involved. These advantages include high-throughput, ready automation, reduced sample/reagent volume, precise measurements, and versatility. The advantages of the use of common laboratory reagents include availability, consistency, compatibility, safety, and cost-effectiveness. Overall, the proposed MW-SPMs are versatile valuable tools for the quantitation of GDV during its pharmaceutical manufacturing.

A NOVEL SPECTROPHOTOMETRIC METHOD FOR THE ESTIMATION OF RANOLAZINE IN BULK DRUG AND IT’S FORMULATION

The simple, rapid, precise, accurate, robust and economical analytical method has been developed for estimation of Ranolazine in bulk and pharmaceutical dosage form. This study describes the validation of an UV Spectrophotometric method for quantitative determination of Ranolazine in pharmaceutical dosage form using acetate buffer (pH 4.4) as solvent. The maximum absorbance was found at 272 nm. The Linearity was found to be in range of 5 to 40µg/ml. The %RSD for Intraday and Interday precision studies was found to be 0.793 and 0.793 respectively. The % assay was found to be 99.6%±0.05. The average % recovery of ranoalazine was found to be in between 98.5-101.7%. LOD and LOQ was found to be 0.58 µg/ml and 1.75 µg/ml respectively. The results suggest that this method can be employed for routine analysis of Ranolazine in bulk and commercial pharmaceutical formulations. Development And Validation of UV-VIS Spectrophotometric Method for Determination of Triamcinolone from Dissolution Media

A simple, accurate UV-VIS Spectrophotometric method has been developed using a dissolution media, Tween 20 (0.15%) as a solvent to determine the Triamcinolone. At predetermined λmax of 240 nm, method was proved linear in the range of 10-50 µg/ml with good correlation coefficient (R2= 0.998) and excellent recovery (between 98 and 101%). This method was successfully applied to the determination of Triamcinolone from dissolution media, 0.15% Tween 20 aqueous solution. This method can be employed for the dissolution study of Triamcinolone in injectable suspension.
