Development of a Novel Assay Based on Plant-Produced Infectious Bursal Disease Virus VP3 for the Differentiation of Infected From Vaccinated Animals.

Infectious bursal disease virus is the causative agent of Gumboro disease, a severe infection that affects young chickens and is associated with lymphoid depletion in the bursa of Fabricius. Traditional containment strategies are based either on inactivated or live-attenuated vaccines. These approaches have several limitations such as residual virulence or low efficacy in the presence of maternally derived antibodies (MDA) but, most importantly, the impossibility to detect the occurrence of natural infections in vaccinated flocks. Therefore, the development of novel vaccination strategies allowing the differentiation of infected from vaccinated animals (DIVA) is a priority. Recently, commercial vectored and experimental subunit vaccines based on VP2 have been proved effective in protecting from clinical disease and posed the basis for the development of novel DIVA strategies. In this study, an engineered version of the VP3 protein of IBDV (His-VP3) was produced in plants, successfully purified from Nicotiana benthamiana leaves, and used to develop an enzyme-linked immunosorbent assay (ELISA) for the detection of anti-VP3 antibodies. The His-VP3 ELISA was validated with a panel of 180 reference sera and demonstrated to have 100% sensitivity (95% CI: 94.7–100.0) and 94.17% specificity (95% CI: 88.4–97.6). To evaluate the application of His-VP3 ELISA as a DIVA test, the novel assay was used to monitor, in combination with a commercial kit, detecting anti-VP2 antibodies, the immune response of chickens previously immunized with an inactivated IBDV vaccine, a recombinant Turkey herpes virus carrying the VP2 of IBDV (HVT-ND-IBD) or with plant-produced VP2 particles. The combined tests correctly identified the immune status of the vaccinated specific pathogen free white-leghorn chickens. Moreover, the His-VP3 ELISA correctly detected MDA against VP3 in commercial broiler chicks and showed that antibody titers fade with time, consistent with the natural decrease of maternally derived immunity. Finally, the novel assay, in combination with a VP2-specific ELISA, demonstrated its potential application as a DIVA test in chickens inoculated with VP2-based vaccines, being able to detect the seroconversion after challenge with a very virulent IBDV strain.

Coronavirus: proteomics analysis of chicken kidney tissue infected with variant 2 (IS-1494)-like avian infectious bronchitis virus.

Avian infectious bronchitis virus is one of the most important gammacoronaviruses, which causes a highly contagious disease. In this study, we investigated changes in the proteome of kidney tissue of specific-pathogen-free (SPF) chickens that were infected with an isolate of the nephrotropic variant 2 genotype (IS/1494/06) of avian coronavirus. Twenty 1-day-old SPF White Leghorn chickens were randomly divided into two groups, each comprising 10 chickens, which were kept in separate positive-pressure isolators. Chickens in group A served as a virus-free control group up to the end of the experiment, whereas chickens in group B were inoculated with 0.1 ml of 104.5 EID50 of the IBV/chicken/Iran/UTIVO-C/2014 isolate of IBV, and kidney tissue samples were collected at 2 and 7 days post-inoculation (dpi) from both groups. Sequencing of five protein spots at 2 dpi and 22 spots at 7 dpi that showed differential expression by two-dimensional electrophoresis (2DE) along with fold change greater than 2 was done by MS-MALDI/TOF/TOF. Furthermore, the corresponding protein-protein interaction (PPI) networks at 2 and 7 dpi were identified to develop a detailed understanding of the mechanism of molecular pathogenesis. Topological graph analysis of this undirected PPI network revealed the effect of 10 genes in the 2 dpi PPI network and nine genes in the 7 dpi PPI network during virus pathogenesis. Proteins that were found by 2DE analysis and MS/TOF-TOF mass spectrometry to be down- or upregulated were subjected to PPI network analysis to identify interactions with other cellular components. The results show that cellular metabolism was altered due to viral infection. Additionally, multifunctional heat shock proteins with a significant role in host cell survival may be employed circuitously by the virus to reach its target. The data from this study suggest that the process of pathogenesis that occurs during avian coronavirus infection involves the regulation of vital cellular processes and the gradual disruption of critical cellular functions.Electronic supplementary materialThe online version of this article (10.1007/s00705-020-04845-7) contains supplementary material, which is available to authorized users.

Comparison of the first Iranian native Ornithobacterium rhinotracheale vaccine with conventional vaccine: A challenge study.

Background and Aim:The best strategy to prevent or control an Ornithobacterium rhinotracheale (ORT) infection is vaccination. The present study aimed to compare the efficacy of the first Iranian inactivated ORT vaccine (Razi, Iran), which had been prepared from a native strain, with the Nobilis ORT Inac (Intervet, The Netherlands) through a challenge trial.Materials and Methods:Seventy-two 1-day-old specific pathogen-free White Leghorn chickens were used in this study. The birds were divided randomly into four groups. Following the vaccination and challenge of the birds, the efficacy of the Razi and the Intervet ORT vaccines was evaluated by serological, bacteriological, and molecular methods.Results:The antibody titer in vaccinated groups was determined to be significantly higher than unvaccinated birds. In addition, the difference in postmortem lesion scores between the vaccinated and unvaccinated birds was significant. The differences in the means of the antibody titers and postmortem lesion scores in birds that were vaccinated by the Razi and Intervet ORT vaccines were not significant.Conclusion:Considering the results of this study, it can be concluded that the Iranian native ORT vaccine was comparable to the Intervet vaccine. The Razi ORT vaccine has effectively decreased the duration of the ORT infection and can effectively protect the chickens against an ORT infection.

Pathogenicity of a QX-like avian infectious bronchitis virus isolated in China

Avian infectious bronchitis is a serious and highly contagious disease caused by infectious bronchitis virus (IBV). We isolated a highly virulent IBV strain (CK/CH/JS/TAHY) from kidneys of diseased chickens. Phylogenetic analysis based on the S1 gene revealed that CK/CH/JS/TAHY clustered with the QX-like type. The S1 gene has 1,620 nucleotides and encoded a polypeptide of 540 amino acids with typical coronavirus cleavage recognition sites of HRRR. About 1-day-old specific pathogen-free White Leghorn chickens inoculated with CK/CH/JS/TAHY at 105.5 EID50 exhibited clinical signs including coughing, sneezing, nasal discharge, and tracheal vocalization accompanied by depression with 84% mortality and 100% morbidity. The kidneys of dead birds were swollen and pale and exhibited severe urate deposition. Histopathological examination revealed kidney hemorrhages, multifocal necrosis of the renal tubules and trachea with cilia loss, sloughing of epithelial cells, and edema of the lamina propria. IBV-specific antibodies appeared at 10 D post-infection. Chickens vaccinated with a CK/CH/JS/TAHY oil-emulsion vaccine showed 26.7% morbidity and 3% mortality indicating a protective effect. In conclusion, the IBV strain is a virulent avian IBV and that exhibited severe pathogenicity in chickens and is a vaccine candidate to prevent infection by Chinese QX-like nephropathogenic IBV strains.

Pathogenesis of Pancreatitis in Chickens after Experimental Infection with 9a5b Newcastle Disease Virus Mutant Isolate

The aim of this study was to investigate the effect of Newcastle disease virus (NDV) on the chicken pancreas. A virulent 9a5b mutant NDV isolate was inoculated intranasally into 32-day-old specific pathogen-free white Leghorn chickens. The pancreas was examined grossly and fixed for histopathological, immunohistochemical and electron microscopical investigations. Inflammatory changes were observed in the peripancreatic tissue at the early stage of infection (12h post infection) and became more prevalent towards the end of the experiment. Multifocal areas of necrotizing inflammation were detected in the exocrine portion of the pancreas by 5 days post infection (dpi) and became more severe at 10dpi. The endocrine islets were generally preserved, but slight degenerative changes were observed at 10 dpi. Immunohistochemically, NDV-nucleoprotein (NDV-NP) signals were detected in the peripancreatic tissues (associated with macrophages and other lymphoid cells) by 1 dpi. In the exocrine portion of the pancreas, NDV-NP signals were detected at 5 dpi and increased in intensity and distribution by 10 dpi. NDV particles were confirmed in the cytoplasm of exocrine acinar cells by transmission electron microscopy. CD3-positive cells were observed in the peripancreatic tissues earlier than in the pancreatic tissue. Moreover, in comparison with control chickens, insulin immunoexpression was unchanged, except on the last day of the experiment, when it was slightly reduced. The 9a5b NDV infection induced an inflammatory reaction and viral replication in the peripancreatic tissues earlier than in the pancreatic tissue. Furthermore, necrosis affected mainly the exocrine portion of the pancreas, while the endocrine portion was generally unaffected.

Transcriptome analysis reveals an activation of major histocompatibility complex 1 and 2 pathways in chicken trachea immunized with infectious laryngotracheitis virus vaccine

Infectious laryngotracheitis is an acute, contagious, upper respiratory disease of chickens caused by gallid herpes virus 1. Due to mortality rates that can reach up to 70% depending on the virulence of the virus, the disease is of great economic importance to the poultry industry. In this study, 15-d-old specific pathogen-free White Leghorn chickens were used to perform transcriptome analysis of chicken trachea immunized with infectious laryngotracheitis virus vaccine. Myosin and several collagen-related genes were downregulated in the immunized group, suggesting that normal function and structure may be compromised. In addition, we identified some cytokine receptors and several immune genes, such as Granzyme A (GZMA), CD4 molecule (CD4), CD8a molecule (CD8A), and CD8b molecule (CD8B), that were upregulated upon vaccination. The gene ontology analysis shows that genes included in the biological process cluster were related to antigen processing and presentation, positive regulation of immune system processes, T cell selection, and positive regulation of T cell activation. In conclusion, chicken embryo origin vaccine activation of the major histocompatibility complex 1 and 2 pathways provides insight for evaluation and design of infectious laryngotracheitis vaccines.

Comparative Study on the Pathogenesis of the Generated 9a5b Newcastle Disease Virus Mutant Isolate Between Chickens and Waterfowl

Lentogenic Newcastle disease viruses (NDVs), circulating among waterfowl, have the potential to become highly pathogenic by replication in chickens. The pathological studies that compare NDV infections in chickens and waterfowl are rare. The virulent 9a5b mutant NDV isolate was generated by passaging the lentogenic Goose/Alaska/415/91 NDV isolate in chickens. The pathogenesis of the virulent 9a5b mutant isolate is unknown in both chickens and waterfowl. In this study, the virulent 9a5b mutant NDV isolate was inoculated intranasally in 32-day-old specific pathogen-free white Leghorn chickens and Japanese commercial ducks. Unlike ducks, which remained clinically normal throughout the study, chickens had depression, gasping, oral discharges, and greenish-white soft feces. Gross and histologic lesion patterns as well as viral replication supported the differing clinical outcome. Ducks had slight inflammation mainly in respiratory and digestive tracts, whereas slight nonpurulent encephalitis, necrotizing pancreatitis, tubulointerstitial nephritis, and mild inflammation in respiratory and digestive tracts were detected in chickens. In agreement, interferon-beta (IFN-β)-immunopositive signals were more intense in lung tissue of ducks than that of chickens, and NDV replications were detected intensively in chicken tissues. These results suggest that the 9a5b mutant NDV isolate is more virulent in chickens, and slight histological lesions were induced in ducks even with virulent NDVs.

Pathogenesis of Ornithobacterium rhinotracheale in egg-laying hens with coexisting infectious bronchitis virus and Escherichia coli infections

Ornithobacterium rhinotracheale (ORT) is recognized as an important respiratory pathogen in turkeys and chickens. The severity of the disease is worsened when birds have coexisting infections with other respiratory pathogens. The objectives of this study were to investigate the pathogenesis of ORT infection with exposure to infectious bronchitis virus (IBV), Escherichia coli, or both in laying chickens. A total of 160 eight-week-old specific pathogen-free White Leghorn chickens were exposed experimentally to 1 of 7 treatments: ORT alone; E. coli alone; IBV alone; ORT + E. coli; ORT + IBV; IBV + E. coli; or ORT + IBV + E. coli. The clinical signs and pathological changes were evaluated in birds from all groups. Birds exposed to ORT or E. coli alone did not show any overt clinical signs. Birds exposed to IBV, ORT + IBV, IBV + E. coli, and ORT + IBV + E. coli exhibited clinical signs that varied from depression and nasal discharge to mortality. The ORT + IBV + E. coli-infected birds exhibited gross lesions, which included hemorrhagic tracheitis, fibrinous pneumonia, airsacculitis, pericarditis, and peritonitis. Histopathological studies of this group revealed distinct pathological changes. Immunohistochemistry staining demonstrated the presence of ORT antigens in the tracheas and lungs of the ORT + IBV + E. coli-infected birds. Mortality after ORT infection was noticed only in the ORT + IBV + E. coli group. We conclude that IBV and E. coli infections exacerbate ORT pathogenesis in adult laying hens. We also conclude that ORT persists in tissues such as the infraorbital sinuses for longer periods without causing significant respiratory disease by itself. This presence of ORT may predispose birds to the development of peritonitis and death with subsequent IBV and E. coli coinfections.

Assessment of Reduced Vaccine Dose on Efficacy of an Inactivated Avian Influenza Vaccine Against an H5N1 High-Pathogenicity Avian Influenza Virus

Avian influenza (AI) vaccines are a viable emergency tool for use in a comprehensive strategy for dealing with high-pathogenicity AI in developed countries. However, the available doses of inactivated AI vaccine are limited to national vaccine banks and inventory stocks of some commercial biologics manufacturers. To determine if the available vaccine doses could be stretched by using reduced vaccine dose but maintain adequate efficacy, a study was conducted to determine if 3-wk-old specific pathogen-free White Leghorn chickens vaccinated with full, ½, ¼, and 1/10 doses of an inactivated H5N9 AI vaccine would be protected against a high-dose challenge of H5N1 highly pathogenic AI virus given 4 wk later. At all 4 AI vaccine doses, the AI-vaccinated chickens were protected from disease and death, but all the sham-vaccinated chickens developed clinical signs and died. There were no differences between the full, ½, and ¼dose AI vaccine groups for serological titers at 7 wk of age, or for cloacal and oropharyngeal titers of challenge virus shed at 2 d postchallenge. However, the 1/10 dose group had significantly reduced hemagglutination inhibition titers at 7 wk compared with ¼ dose, and the 1/10 dose group had more chickens shedding challenge virus from the oropharynx than the full dose group. Most importantly, the mean protective dose was 1/50 dose, and using the international regulatory standard of 50 mean protective doses for Newcastle disease vaccine as a guide, the full dose of the H5N9 AI vaccine would be the minimum dose acceptable for use in the field. Use of the full vaccine dose is especially important, because protection in commercial chickens in the field is typically less than seen in experimental studies in specific pathogen-free chickens in the laboratory.

A reovirus challenge model applicable in commercial broilers after live vaccination

The efficacy of live reovirus vaccines may be determined by challenge via the foot pad route 3 to 4 weeks after vaccination. Swelling and discoloration in the foot pad and shank are scored for a period of 14 days. The major disadvantages of this challenge model are the subjective judgement of gross foot pad and/or shank lesions, that it is very difficult to induce lesions in broilers, and that it causes animal suffering. Other reovirus challenge models are based on reisolation of the virus from different tissues or on scoring microscopic lesions in the tendons. Some disadvantages of these models are that they either cannot be used after vaccination with live reovirus because they cannot discriminate between vaccine and challenge virus or that the microscopic lesions scored need not necessarily be related to the challenge virus but may have been induced by other factors. Therefore, we have attempted to develop a reovirus challenge model that was an improvement on the existing ones, using isolation of reovirus from different organs followed by specific detection of the challenge virus with a monoclonal antibody that can discriminate between challenge and vaccine virus. The reovirus challenge model was examined in specific pathogen free (SPF) White Leghorn chickens and commercial broilers. In vivo studies were conducted to examine the efficacy of an attenuated reovirus vaccine in SPF White Leghorn chickens and commercial broilers with maternal immunity against reovirus. No challenge virus could be detected in any of the organs of the vaccinated chickens 3 and 10 days after challenge. In contrast, challenge virus could be isolated from the unvaccinated control group. At an increased challenge dose all unvaccinated challenge control birds were positive, while the vaccinated chickens were protected. It was shown that 1-day-old vaccination in the presence of maternal immunity was effective. It seemed that protection induced in broilers by the attenuated reovirus vaccine may not have been entirely humoral because in protected birds no antibodies against reovirus were detected by enzyme-linked immunosorbent at the time of challenge. Protection in these birds might therefore have been induced by cellular immunity.

Nonsuppurative myocarditis associated with so-called fowl glioma.

C/O specific pathogen-free White Leghorn chickens were intracerebrally inoculated at one day of age with a brain homogenate of Japanese bantams (Gallus gallus domesticus) affected with fowl glioma. Histologically, six of eight inoculated chickens developed nonsuppurative meningoencephalitis in cerebrum and two of them had the characteristic lesions of fowl glioma. Hyperplastic lymphoid foci concomitantly developed in many organs of these birds, especially in the heart. Apart from these lymphoid foci, lymphocytic myocarditis was observed in all inoculated birds. Matrix inclusions were also noted in myocardial cells. Immunohistochemically, avian leukosis virus antigens were detected in reticular cells in the lymphoid foci, mesangial cells of the kidney, smooth muscle cells of the blood vessels, and myocardial cells. Of these tissues, the myocardium of all inoculated birds consistently showed strong reactivity for this antigens. The matrix inclusions were also positive for the antigens. These results suggest that the causal virus of fowl glioma has a high propensity to replicate, especially in myocardium and nonsuppurative myocarditis occurs associated with so-called fowl glioma.

Pathogenicity of Serotype 1 Fowl Adenovirus in Commercial Broiler Chickens

The pathogenicity of a serotype 1 fowl adenovirus (FAV-99ZH), isolated from broiler chickens exhibiting gizzard erosion, was investigated in commercial broiler chickens. Five-, 3-, and 1-wk-old commercial broiler chickens were inoculated with FAV-99ZH by both oral and ocular routes. In the 5-wk-old chickens (trial 1), none of which had the maternal antibody to FAV-99ZH, severe gizzard erosions were observed on days 5, 7, and 10 postinoculation (PI). Among the 3-wk-old chickens (trial 2), which were separated into a control group and three treatment groups according to their maternal antibody titer levels, some chickens showed clinical signs such as depression and anorexia. Compared with the control group, all the treatment groups showed decreased weight gain. One treatment group, moreover, showed significantly decreased (P < 0.05) weight gain on day 10 PI. Severe gizzard lesions, such as erosion or ulcers, were observed from day 4 PI in all treatment groups regardless of their maternal antibody levels. The 1-wk-old chickens (trial 3) were separated into a control group and two treatment groups according to their titer levels of the inoculated virus. In spite of high maternal antibody levels, severe gizzard lesions were observed in both treatment groups, which also showed decreased weight gain. One treatment group, inoculated with the higher dose, showed significantly decreased (P < 0.05) weight gain on days 10 and 14 PI compared with the control group. Fowl adenovirus was recovered mainly from gizzard and rectal (including feces) samples from inoculated chickens but was not recovered from liver samples in any of the trials or in any of the control chickens. Although the reproduced disease was similar to that described in a previous report of experimental infection of specific-pathogen-free (SPF) white leghorn chickens with fowl adenovirus, the pathogenicity of FAV-99ZH in commercial broiler chickens was more severe than that in the SPF white leghorn chickens. The results of the present study indicate that FAV-99ZH isolated from gizzard erosion had pathogenicity and produced severe lesions in the gizzards of broiler chickens and that FAV-99ZH could infect and produce illness in broiler chickens with maternal antibodies against this virus.

Experimental Infection of Specific-Pathogen-Free Chickens with Serotype-1 Fowl Adenovirus Isolated from a Broiler Chicken with Gizzard Erosions

Gizzard lesions were formed in specific-pathogen-free (SPF) white leghorn chickens inoculated with fowl adenovirus (FAV). The virus, serotype 1 FAV 99ZH strain (FAV-99ZH), was originally isolated from the gizzard mucosa of commercial broiler chickens exhibiting gizzard erosion with intranuclear inclusion bodies. Five-day-old and 53-day-old SPF white leghorn chickens were inoculated with FAV-99ZH by both oral and ocular routes and then examined at necropsy on days 3, 5, 7, 10, 14, and 21 postinoculation (PI). There were no clinical signs in any of the chickens after the inoculation. Focal gizzard lesions occurred macroscopically, however, in inoculated chickens at several experimental periods. FAV was recovered from tissue samples of the proventriculus, gizzard, pancreas, and rectum by day 10 or 7 PI but was not recovered from liver samples of any of the chickens. These results indicate that FAV isolated from gizzard erosion is able to reproduce gizzard lesions as necrosis and erosion in SPF white leghorn chickens and that it may have a greater degree of tissue tropism in gizzards and other digestive organs than in the liver.

Infectious Bronchitis Virus Antibodies in Tears and Their Relationship to Immunity

Antibodies to infectious bronchitis virus (IBV) in chicken tears were investigated to determine if they could be used as an indicator of protective immunity. Antibody production in tears and serum was measured by enzyme-linked immunosorbent assay (ELISA) in specific-pathogen-free (SPF) white leghorn and broiler chickens vaccinated with a live attenuated vaccine containing the Massachusetts (Mass) Connaught strain of IBV. The effect of virulent infectious bursal disease virus (IBDV) infection on antibody production in tears was also evaluated. Immunity was assessed by challenging the chickens with Mass 41 and performing tracheal swabbings 5 days later. In addition, tears were also evaluated for virus-neutralizing (VN) antibodies to IBV. Following eyedrop vaccination, anti-IBV antibodies were consistently detected by ELISA in tears prior to and in higher concentrations than in the sera of SPF white leghorn and broiler chickens. Maternal IBV antibodies were present in the tear secretions of broiler chickens but in lower concentrations than in sera. Infection of SPF chicks with a virulent and immunosuppressive strain of IBDV at 1 day of age greatly reduced IBV ELISA antibody production in tears as well as serum compared with infection of chickens with IBDV at 14 days of age. IBV ELISA and VN antibody levels in tears were not accurate indicators of IBV immunity as determined by challenge with Mass 41. High tear IBV antibody titers were observed in some chickens determined to be susceptible to IBV challenge and low tear titers were detected in some protected chickens. This finding suggests that mechanisms other than antibody-mediated immunity in tears are important in viral clearance following challenge.
