First report of Zaprionus indianus and Z. tuberculatus (Diptera: Drosophilidae) in Tunisia

In October 2019, unusual damage was observed on Carissa macrocarpa (Apocynaceae) fruit at the ornamental nursery of the Higher Agronomic Institute (ISA CM), Chott Mariem (Sousse, Tunisia). Samples of fruit were collected and brought to the laboratory for observation and rearing. After a few days, adult flies emerged from infested fruit. Zaprionus indianus Gupta, Z. tuberculatus Malloch (Diptera Drosophilidae) and Ceratitis capitata Wiedemann (Diptera, Tephritidae) were identified morphologically. Information on the morphology, distribution and ecology of the two Zaprionus species are provided. This is the first record of Z. indianus and Z. tuberculatus in Tunisia.

Ultrastructural features of spermatozoa and their phylogenetic application in Zaprionus (Diptera, Drosophilidae)

ABSTRACTThe genus Zaprionus consists of approximately 60 species of drosophilids that are native to the Afrotropical region. The phylogenetic position of Zaprionus within the Drosophilidae family is still unresolved. In the present study, ultrastructural features of spermatozoa of 6 species of Zaprionus as well as the species Drosophila willistoni and Scaptodrosophila latifasciaeformis were analyzed. The ultrastructure revealed that the species have the same flagellar ultrastructure. Two mitochondrial derivatives, one larger than the other, close to the axoneme were present, primarily in D. willistoni (subgenus Sophophora). Except for Z. davidi and Z. tuberculatus, the analyzed species had paracrystalline material in both mitochondrial derivatives. Moreover, the testes showed 64 spermatozoa per bundle in all of the species. In the cluster analysis, 6 Zaprionus species were grouped closely, but there were some incongruent positions in the cladogram. The results indicated that sperm ultrastructure is an important tool for elucidating the phylogeny and taxonomy of insects.

Detecting recent changes in the demographic parameters of drosophilid populations from western and central Africa

Previous genetic studies showing evidence of past demographic changes in African drosophilids suggested that these populations had strongly responded to Quaternary climate changes. We surveyed nine species of Zaprionus, a drosophilid genus mostly present in Africa, in forests located between southern Senegal and Gabon. The mitochondrial COI gene showed contrasted levels of sequence variation across species. Populations of the only cosmopolitan species of the genus, Z. indianus, and of its closely related sibling species, Z. africanus, are highly polymorphic and appear to have undergone a continuous population expansion beginning about 130,000years ago. Five less variable species probably underwent a population expansion beginning only about 20,000–30,000years ago. One of them, Z. taronus, was significantly structured between forest blocks. The last two species were nearly monomorphic, probably due to infection by Wolbachia. These results are similar to those obtained in three species from the melanogaster subgroup, and may be typical of the responses of African drosophilid populations to glacial cycles.

Taxonomic and evolutionary analysis of Zaprionus indianus and its colonization of Palearctic and Neotropical regions

Zaprionus indianus is a dipteran (Drosophilidae) with a wide distribution throughout the tropics and temperate Palearctic and Nearctic regions. There have been proposals to reclassify the genus Zaprionus as a subgenus or group of the genus Drosophila because various molecular markers have indicated a close relationship between Zaprionus species and the immigrans-Hirtodrosophila radiation within Drosophila. These markers, together with alloenzymes and quantitative traits, have been used to describe the probable scenario for the expansion of Zaprionus indianus from its center of dispersal (Africa) to regions of Asia (ancient dispersal) and the Americas (recent dispersal). The introduction of Z. indianus into Brazil was first reported in 1999 and the current consensus is that the introduced flies came from high-latitude African populations through the importation of fruit. Once in Brazil, Z. indianus spread rapidly throughout the Southeast and then to the rest of the country, in association with highway-based fruit commerce. These and other aspects of the evolutionary biology of Z. indianus are addressed in this review, including a description of a probable route for this species’ dispersal during its recent expansion.

Copia Retrotransposon in the Zaprionus Genus: Another Case of Transposable Element Sharing with the Drosophila melanogaster Subgroup

Copia is a retrotransposon that appears to be distributed widely among the Drosophilidae subfamily. Evolutionary analyses of regulatory regions have indicated that the Copia retrotransposon evolved through both positive and purifying selection, and that horizontal transfer (HT) could also explain its patchy distribution of the among the subfamilies of the melanogaster subgroup. Additionally, Copia elements could also have transferred between melanogaster subgroup and other species of Drosophilidae-D. willistoni and Z. tuberculatus. In this study, we surveyed seven species of the Zaprionus genus by sequencing the LTR-ULR and reverse transcriptase regions, and by using RT-PCR in order to understand the distribution and evolutionary history of Copia in the Zaprionus genus. The Copia element was detected, and was transcriptionally active, in all species investigated. Structural and selection analysis revealed Zaprionus elements to be closely related to the most ancient subfamily of the melanogaster subgroup, and they seem to be evolving mainly under relaxed purifying selection. Taken together, these results allowed us to classify the Zaprionus sequences as a new subfamily-ZapCopia, a member of the Copia retrotransposon family of the melanogaster subgroup. These findings indicate that the Copia retrotransposon is an ancient component of the genomes of the Zaprionus species and broaden our understanding of the diversity of retrotransposons in the Zaprionus genus.

Revision of the Afrotropical species of Zaprionus (Diptera, Drosophilidae), with descriptions of two new species and notes on the internal reproductive structures and immature stages

A new classification of the subgenus Zaprionus is proposed in light of recent phylogenetic findings. The boundaries of the armatus and inermis species groups are redefined. The vittiger subgroup is upgraded to the level of a species group. The tuberculatus subgroup is transferred from the armatus to the inermis group. A new monotypic group, neglectus, is erected. Full morphological descriptions of four species belonging to the vittiger group are given: Zaprionus lachaisei sp. n. from Tanzania and Zaprionus santomensis sp. n. from São Tomé and Principé, and two cryptic species of the indianus complex, Zaprionus africanus Yassin & David and Zaprionus gabonicus Yassin & David. Three nominal species are synonymised: Zaprionus beninensis Chassagnard & Tsacas, syn. n. with Zaprionus koroleu Burla, Zaprionus simplex Chassagnard & McEvey, syn. n. with Zaprionus neglectus Collart, and Zaprionus megalorchis Chassagnard & Tsacas, syn. n. with Zaprionus ornatus Séguy. Half of the 46 species of the subgenus are available as laboratory strains and this has allowed full descriptions of the internal structure of their reproductive systems and their immature stages.

On the Relationship between Zaprionus spinipilus Chassagnard & McEvey and Z. vittiger Coquillett, the Type Species of the Genus Zaprionus (Diptera: Drosophilidae)

Zaprionus vittiger Coquillett is the type species of the genus Zaprionus Coquillett. However, the species is only known from five old museum specimens collected from South Africa and Malawi. It has often been confused with many other Zaprionus species, especially with Z. spinipilus Chassagnard & McEvey, a widespread species in Africa known from Madagascar, Malawi, Ethiopia and Cameroon. We have recently collected flies from the type localities of both species (South Africa and Madagascar, respectively). This has prompted us to test the taxonomic boundaries of these two nominal species using molecular (the mitochondrial COII and the nuclear Amyrel genes), chromosomal, morphological (internal and external genitalia), and reproductive isolation analyses. The results suggest Z. spinipilus to be a junior synonym to Z. vittiger.

Variation in Malate Dehydrogenase Allozymes among Honeybee Populations in Kenya

SummaryPolyacrylamide gel electrophoresis was used to analyse phenoxyethanol extracts from honeybees (Apis mellifera) for malate dehydrogenase activity. Nine natural populations in Kenya consisting of the subspecies Apis mellifera litorea, Apis mellifera nubica, Apis mellifera monticola and Apis mellifera scutellata were sampled. Allozyme variation in the three Mdh alleles found was quite uniform in all the populations. There were small differences among the populations in allele frequencies and in levels of enzyme activity as indicated by intensity of bands. Genetic variation was lower in honeybees than in samples of five Zaprionus species (Diptera, Drosophilidae) taken earlier from the same localities.

An asymmetric species-isolating mechanism between two sympatric species of Zaprionus (Diptera, Drosophilidae)

The species Zaprionus tuberculatus and Z. sepsoides are prevented from mating with each other by behavioural mechanisms. Male Z. sepsoides can discriminate between females of the two species by chemosensory means, and rarely court female Z. tuberculatus. Male Z. tuberculatus may also be able to distinguish between females of the two species, but not infallibly. Females of both species can distinguish between the ‘Type-2’ male courtship songs of the two species. Female Z. sepsoides will not mate with males unless conspecific Type-2 song is produced during courtship. Female Z. tuberculatus, will, however, mate with males if Z. sepsoides Type-2 song is produced, albeit after a longer courtship than if conspecific Type-2 song is produced. The onus of species recognition in encounters between males and females of the two species, therefore, lies largely on Z. sepsoides. It is suggested that such a mechanism of species isolation could have evolved via a stage of partial asymmetric reproductive isolation, which could originally have arisen by the effect of genetic drift occurring in two temporarily isolated populations of an ancestral species.

Species and sex-specific songs and courtship behaviour in the genus Zaprionus (Diptera-Drosophilidae)

Courtship behaviour and songs of six Zaprionus species show differences not only between species but also between sexes. Courtship behaviour differs from that in the related genus Drosophila. In most species, males produce two song types which may alternate or be repeated. Singing occurs during courtship displays, mounting, after copulation and briefly during male-to-male interactions. Females produce a loud whine and body rocking movements in refusal but have a species-specific pulsed song produced during normal courtship which differs from the conspecific male song. One species produces an irregular male song but a regular female song. Sex-specific songs may have selective advantages but pose problems for the sensory template hypothesis.
