Prediction of Degradation Profiles for Various Sartans under Solvent-Free Mechanochemical Conditions.

For the approval of a drug, the stability data must be submitted to regulatory authorities. Such analyses are often time-consuming and cost-intensive. Forced degradation studies are mainly carried out under harsh conditions in the dissolved state, often leading to extraneous degradation profiles for a solid drug. Oxidative mechanochemical degradation offers the possibility of generating realistic degradation profiles. In this study, a sustainable mechanochemical procedure is presented for the degradation of five active pharmaceutical ingredients (APIs) from the sartan family: losartan potassium, irbesartan, valsartan, olmesartan medoxomil, and telmisartan. High-resolution mass spectrometry enabled the detection of impurities already present in untreated APIs and allowed the elucidation of degradation products. Significant degradation profiles could already be obtained after 15-60 min of ball milling time. Many of the identified degradation products are described in the literature and pharmacopoeias, emphasizing the significance of our results and the applicability of this approach to predict degradation profiles for drugs in the solid state.

In situ-grown active transition metal-functionalized MOF via solvent-free mechanochemical conditions as effective catalyst for the preparation of carbamates from organic azides with amines and anilines

The solvent-free synthesis of MOF represents a promising and environmentally benign protocol. This paper outlines a green one-pot mechanochemical method for the synthesizing of UiO-66 in the absence of solvent, along with the implantation of three different metals (Cu, Ni, and Pd) into the defect sites. The materials have proven to be effective catalysts in the transformation between azidocarbonate and amines/anilines in moderate to excellent isolated yields (up to 94%), exhibiting better advantages compared with the exiting methodologies. More interestingly, Cu and Ni exhibited very good catalytic performance towards primary and secondary amines, respectively. It also should be noted that high yield was still observed after six cycles. Our work presents a novel and straightforward strategy for the preparation of secondary and tertiary carbamates in an unprecedented fashion, greatly enriched the toolbox for the synthetic community. Moreover, the combination of experimental studies and density functional theory (DFT) calculations gave more insight and evidence for the mechanism. Accompanied by the advantages of the effective and recyclable catalyst, the broad substrate scope, good functional group tolerance, and excellent reaction yields made the established method attractive in heterogeneous catalysis and the carbamates synthesis.

Using Mechanochemistry to Activate Commodity Plastics as Initiators for Radical Chain Reactions of Small Organic Molecules.

Radical initiators such as azo compounds and organic peroxides have been widely used to facilitate numerous transformations of free radicals, which enable the efficient synthesis of structurally complex molecules, natural products, polymers, and functional materials. However, these high-energy reagents are potentially explosive and thus often require special precautions or delicate operating conditions. We postulated that a more convenient and safer alternative for radical chain initiation could be developed by mechanical activation of thermodynamically stable covalent bonds. Here, we show that commodity plastics such as polyethylene and poly(vinyl acetate) are capable of acting as efficient initiators for radical chain reactions under solvent-free mechanochemical conditions. In this approach, polymeric mechanoradicals, which are generated by homolytic cleavage of the polymer chains in response to the applied mechanical energy provided by ball milling, react with tris(trimethylsilyl)silane to initiate radical chain dehalogenation of organic halides. Preliminary calculations support our proposed force-induced radical chain mechanism.

Asymmetric Organocatalysis Under Mechanochemical Conditions.

Asymmetric organocatalysis is a robust methodology providing access to numerous valuable compounds while having green chemistry principles in mind. The realization of organocatalytic transformation under solvent-free mechanochemical conditions brings additional benefits in terms of yields, selectivities, and, last but not least overall improved sustainability. This overview describes developments in the use of mechanochemistry as a vehicle for asymmetric organocatalytic transformations. The material is organized according to main catalytic activation modes, starting with covalent activation and proceeding to non-covalent activation modes. The advantages of mechanochemical organocatalytic reactions are particularly highlighted, but in some cases also, limitations are mentioned. Possibilities for target compound synthesis are also discussed.

Sustainable Production of Reduced Phosphorus Compounds: Mechanochemical Hydride Phosphorylation Using Condensed Phosphates as a Route to Phosphite.

In pursuit of a more sustainable production of phosphorous acid (H3PO3), a versatile chemical with phosphorus in the +3 oxidation state, we herein report that condensed phosphates can be employed to phosphorylate hydride reagents under solvent-free mechanochemical conditions to furnish phosphite (HPO32–). Using potassium hydride as the hydride source, sodium trimetaphosphate (Na3P3O9), triphosphate (Na5P3O10), pyrophosphate (Na4P2O7), fluorophosphate (Na2PO3F), and polyphosphate (“(NaPO3)n”) engendered phosphite in optimized yields of 44, 58, 44, 84, and 55% based on total P content, respectively. Formation of overreduced products including hypophosphite (H2PO2–) was identified as a competing process, and mechanistic investigations revealed that hydride attack on in-situ-generated phosphorylated phosphite species is a potent pathway for overreduction. The phosphite generated from our method was easily isolated in the form of barium phosphite, a useful intermediate for production of phosphorous acid. This method circumvents the need to pass through white phosphorus (P4) as a high-energy intermediate and mitigates involvement of environmentally hazardous chemicals. A bioproduced polyphosphate was found to be a viable starting material for the production of phosphite. These results demonstrate the possibility of accessing reduced phosphorus compounds in a more sustainable manner and, more importantly, a means to close the modern phosphorus cycle.

2,3-Dihydro-1,2,6-thiadiazine 1-Oxides by Biginelli-Type Reactions with Sulfonimidamides under Mechanochemical Conditions.

Biginelli-type multicomponent reactions (MCRs) with NH-free sulfonimidamides provide 2,3-dihydro-1,2,6-thiadiazine 1-oxides in high yields. The couplings are performed in a planetary ball mill under solvent-free mechanochemical conditions. Acetic acid or ytterbium triflate are used as catalysts. A representative product was characterized by X-ray single crystal structure analysis revealing molecular details of the highly functionalized three-dimensional heterocycle. Further product modifications lead to additional structural scaffolds.

A readily accessible porous organic polymer facilitates high-yielding Knoevenagel condensation at room temperature both in water and under solvent-free mechanochemical conditions

A novel nitrogen-rich amorphous porous organic polymer has been synthesized using a microwave-assisted process. Its high chemical stability, reusability and poor solubility enable the use of the porous polymer as a metal-free heterogeneous catalyst for CC bond formation at ambient temperature under environmentally benign conditions.

Synthesis of trifluoromethyl-substituted 1,2,6-thiadiazine 1-oxides from sulfonimidamides under mechanochemical conditions.

TBS-protected or NH-sulfonimidamides react with β-alkoxyvinyl trifluoromethylketones under solvent-free mechanochemical conditions to give 3-trifluoromethyl-substituted three-dimensional 1,2,6-thiadiazine 1-oxides. C4-Functionalized products can be obtained by starting from cyclic enones and brominations of the initially formed heterocycles. The stability of the products was investigated by varying the pH value and storage under aerobic conditions.

Developing benign syntheses using ion pairs via solvent-free mechanochemistry

Solvent-free mechanochemical conditions have been developed to investigate the significance of ion pairing and the use of weak bases for driving forward nucleophilic substitution reactions.

The mechanochemical synthesis of quinazolin-4(3H)-ones by controlling the reactivity of IBX.

Performing any synthesis using several arylamines and hypervalent iodine(V) reagents by direct mixing is unrealistic because of the high exothermic reaction or explosion. Herein we demonstrate, when anilines were substituted with an amide group at the ortho-position, successful chemical reactions could be performed due to intramolecular control. At maximum contact of the reacting substances, i.e., under solvent-free mechanochemical conditions, 2-aminobenzamides, aryl-, alkylaldehydes and the iodine(V) reagent o-iodoxybenzoic acid (IBX) led to substituted quinazolin-4(3H)-one derivatives in fair yields.

Insights into Mechanochemical Reactions at Targetable and Stable, Sub-ambient Temperatures.

We provide insights into the effects of stable and verifiable, low-temperature conditions on mechanochemical reactions. These are made possible by modifications made to a SPEX 8000 m Mixer/mill allowing reliable fine control of low-temperature mechanochemical reactions. Using the reduction of 4-tert-butylcyclohexanone as a model system we find the diastereomeric product distribution bore a strong dependence on the selected temperature. The same reduction in methanol at room temperature shows similar stereoselectivity trends. In both cases decreasing temperature favors increases in selectivity, although the effect is more pronounced in the solvent-free mechanochemical conditions. These results indicate that the cooled jar provides a heatsink to mitigate the exothermic character of the reaction. Stereoselectivity also showed a dependence on operating frequency, although the nature of this dependence remains unclear. Applications of our reactor extend far beyond what is presented herein.

The Choice Is Yours: Using Liquid-Assisted Grinding To Choose between Products in the Palladium-Catalyzed Dimerization of Terminal Alkynes

Herein, we report on the dimerization of terminal alkynes using various palladium catalysts under solvent-free mechanochemical conditions. When tetrakis(triphenylphosphine)palladium(0) was employed as the catalyst, we observed the 1,3-butadiyne as the major product. However, when we employed bis(triphenylphosphine)palladium(II) dichloride as the catalyst, we observed the trans-enyne as the major product. When we used a polymer-supported bis(triphenylphosphine)palladium(II) dichloride catalyst under liquid-assisted grinding conditions, we discovered the ability to tune the catalyst to generate either the diyne or trans-enyne as the major product, depending on the grinding medium.

Efficient, Scalable, and Solvent-free Mechanochemical Synthesis of the OLED Material Alq3 (q = 8-Hydroxyquinolinate)

The aluminum complex Alq3 (q = 8-hydroxyquinolinate), which has important applications in organic light-emitting diode materials, is shown to be readily synthesized as a pure phase under solvent-free mechanochemical conditions from Al(OAc)2OH and 8-hydroxyquinoline by ball milling. The initial product of the mechanochemical synthesis is a novel acetic acid solvate of Alq3, and the α polymorph of Alq3 is obtained on subsequent heating/desolvation of this phase. The structure of the mechanochemically prepared acetic acid solvate of Alq3 has been determined directly from powder X-ray diffraction data and is shown to be a different polymorph from the corresponding acetic acid solvate prepared by solution-state crystallization of Alq3 from acetic acid. Significantly, the mechanochemical synthesis of Alq3 is shown to be fully scalable across two orders of magnitude from 0.5 to 50 g scale. The Alq3 sample obtained from the solvent-free mechanochemical synthesis is analytically pure and exhibits identical photoluminescence behavior to that of a sample prepared by the conventional synthetic route.

An array-based study of reactivity under solvent-free mechanochemical conditions—insights and trends

An array-based approach is put forward to obtain insight into reactivity under mechanochemical solvent-free conditions. We describe a survey of sixty potential reactions between twelve metal salts MX2 {(M = Cu, X2 = (OAc)2, (HCO2)2, (F3CCO2)2, (acac)2, (F6acac)2, (NO3)2, SO4; M = Ni, X2 = (OAc)2, (NO3)2, SO4; M = Zn, X2 = (OAc)2, (NO3)2} and five bridging organic ligands {isonicotinic acid (HINA), 1,4-benzenedicarboxylic acid (H2BDC), acetylenedicarboxylic acid (H2ADC), 1,3,5-benzenetricarboxylic acid (H3BTC), 4,4′-bipyridyl (BIPY). Reaction conditions involved a ball mill, applied for 15 min at 30 Hz, without external heating. When examined by XRPD, forty of the combinations gave detectable reactions, thirty-eight with crystalline products. Of these, twenty-nine reactions were quantitative (consuming all of at least one reactant). Comparison of XRPD patterns with patterns simulated from single crystal X-ray diffraction data in the Cambridge Structural Database allowed structural identification of six products. Of particular interest are the microporous framework materials [Cu(INA)2] and [Cu3(BTC)2] (HKUST-1) obtained by reaction of the corresponding carboxylic acids with copper acetate. Other non-porous polymers with 3-dimensional connectivity, [Ni(ADC)(H2O)4], or 1-dimensional connectivity, [Cu(acac)2(BIPY)] and [Cu(F6acac)(BIPY)] were also obtained. Reaction between zinc acetate and H2ADC gave a new product which had not previously been characterised by single-crystal X-ray crystallography, but whose XRPD pattern suggests that it is isostructural with the known nickel polymer [Ni(ADC)(H2O)4]. Two further isostructural nickel and zinc products were obtained in reactions between HINA and nickel nitrate and zinc nitrate. Trends observed within the array are discussed. Copper acetate and copper formate were the most effective starting materials for reaction with carboxylic acids, potentially related to the basicity of their anions and the solvating effects of the formic and acetic acid byproducts. Amongst the ligands there was a general negative corelation between melting point and reactivity. The issue of pore templating in microporous phases and the generation of new structures is also discussed in relation to the Cu(INA)2, Cu3(BTC)2 and nickel nitrate-BIPY systems. Overall, the study suggests that mechanochemical reactivity between metal salts and organic ligands under solvent free conditions is remarkably general. Use of array-based approaches as demonstrated here is advocated a useful way to reveal underlying trends in reactivity under solvent free mechanochemical conditions and to highlight particular cases for more detailed study.
