Effects of plastic film mulching on soil microbial carbon metabolic activity and functional diversity at different maize growth stages in cool, semi-arid regions

IntroductionPlastic film mulching has been widely used to enhance soil hydrothermal conditions and increase crop yields in cool, semi-arid areas. However, its impact on soil microbial carbon metabolic activity and functional diversity during plant growth remains unclear despite their important roles in nutrient cycling and soil quality evaluation.MethodsThis study used the Biolog EcoPlate technique to investigate the dynamics and driving factors of soil microbial carbon metabolic activity and functional diversity at different maize growth stages following plastic film mulching.Results and discussionThe results revealed that film mulching significantly increased microbial carbon metabolic activities [represented by average well color development (AWCD)] by 300% at the seedling stage and by 26.8% at maturity but decreased it by 47.4% at the flowering stage compared to the control (without mulching). A similar trend was observed for the microbial functional diversity index. Redundancy analysis identified soil moisture (SM), soil temperature (ST), dissolved organic carbon (DOC), microbial biomass carbon (MBC), and bacteria amounts as the primary factors influencing changes in soil microbial carbon source utilization. The mulch treatment significantly increased SM at all growth stages, while its warming effect disappeared at the flowering stage. Soil DOC, MBC, and bacterial populations were notably higher under mulching at the seedling and maturity stages but lower at the flowering stage. Pearson correlation analysis showed that changes in SM, ST, DOC, MBC, and bacterial populations positively correlated with the utilization of all carbon source classes, AWCD, and functional diversity indexes after film mulching. Furthermore, maize grain yield and water use efficiency increased by 142 and 129%, respectively, following film mulching. In conclusion, plastic film mulching enhanced soil microbial carbon metabolic activity and functional diversity at the seedling and maturity stages, improving crop yields in cool, semi-arid areas. Furthermore, the decrease in soil carbon metabolic capacity at flowering stage highlights that supplementing soil carbon sources should be considered after continuous film mulching to sustain or enhance farmland productivity and soil quality.

Impact of Bio-Organic Fertilizer Incorporation on Soil Nutrients, Enzymatic Activity, and Microbial Community in Wheat–Maize Rotation System

Excessive use of inorganic fertilizers disrupts soil nutrient balance and leads to soil degradation and a decrease in biodiversity. In contrast, bio-fertilizers enhance soil structure and fertility and promote plant growth and sustainable agriculture development. Therefore, this study focused on a rotation system of winter wheat and summer maize and aimed to explore the effects of applying chemical fertilizer (NPK) and bio-fertilizer (BF) in the winter wheat season on the sustainable soil development of current wheat and subsequent maize. Before sowing winter wheat four fertilization treatments were, respectively CK (100% NPK at 750 kg ha−1), A (60% NPK at 450 + 20% BF at 150 kg ha−1), B (60% NPK at 450 + 40% BF at 300 kg ha−1), and C (60% NPK at 450 + 60% BF at 450 kg ha−1), conducted. The results showed that treatment A (60% NPK + 20% BF) replacing the NPK at 300 kg ha−1 with BF at 150 kg ha−1 significantly soil nutrient contents, enzyme activity, and microbial metabolic activity. The study also found a positive correlation between soil parameters (total nitrogen, alkaline nitrogen, available phosphorus, organic matter, urease, and alkaline phosphatase in the winter wheat and maize cropping season). Furthermore, the soil microbial composition showed significant enrichment of Proteobacteria, Acidobacteria, Actinobacteria, and Firmicutes, and variations among treatments. Moreover, the application of biofertilizer enhanced the diversity of soil fungi species, particularly during the winter wheat season. This study highlights the importance of integrating biofertilizers with NPK fertilizer for agricultural system conversion and promoting agricultural production and sustainability.

Improving the enrichment of Cd and Zn in leaf mustard (Brassica juncea) by using metal-activating probiotics

Microbial-assisted phytoremediation is of great significance for the remediation of soil contaminated with heavy metals (HMs). Probiotics are beneficial microorganisms that can improve soil structure and fertility and promote plant growth. This study aimed to investigate the effect of two kinds of probiotics, including Lactobacillus casei (L) and Bacillus licheniformis (B), on activating the remediation potentiality of leaf mustard [Brassica juncea (L.) Czerniak.] for mining soil contaminated by Cd and Zn. The results showed that addition of two probiotics significantly reduced soil pH (with 0.05–0.32 units) and improved the available contents of soil HMs (16.0%–59.9% for Cd and 7.1%–23.9% for Zn) in the indoor-incubation experiment. After probiotic treatments, available Cd and Zn in potting soil treated with 1×109 cfu mL-1 of B were 1.65-fold and 1.66-fold of the control, respectively. Meanwhile, soil alkaline phosphatase, urease and sucrose activities were increased, indicating that soil microbial metabolic activities were also motivated. Addition of L and B significantly improved the biomass and chlorophyll contents of leaf mustard. The contents of Cd, Zn in shoot and root were significantly increased with 1×105 cfu mL-1 of L treatment (shoot: 75.2%, 71.9%; root: 36.8%, 61.2%). Furthermore, the activity of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) and the level of malondialdehyde (MDA) were increased or inhibited, indicating that the resistance of plants to HMs was enhanced. These results indicated that two kinds of probiotics could improve the activities of Cd and Zn directly in mining soil and promote the growth of leaf mustard, thus increasing the efficiency of phytoremediation for HMs. The study provides a reference value for probiotic-assisted phytoremediation of soil contaminated with HMs.

Recover of Soil Microbial Community Functions in Beech and Turkey Oak Forests After Coppicing Interventions

Forest management influences the occurrence of tree species, the organic matter input to the soil decomposer system, and hence, it can alter soil microbial community and key ecosystem functions it performs. In this study, we compared the potential effect of different forest management, coppice and high forest, on soil microbial functional diversity, enzyme activities and chemical-physical soil properties in two forests, turkey oak and beech, during summer and autumn. We hypothesized that coppicing influences soil microbial functional diversity with an overall decrease. Contrary to our hypothesis, in summer, the functional diversity of soil microbial community was higher in both coppice forests, suggesting a resilience response of the microbial communities in the soil after tree cutting, which occurred 15–20 years ago. In beech forest under coppice management, a higher content of soil organic matter (but also of soil recalcitrant and stable organic carbon) compared to high forest can explain the higher soil microbial functional diversity and metabolic activity. In turkey oak forest, although differences in functional diversity of soil microbial community between management were observed, for the other investigated parameters, the differences were mainly linked to seasonality. The findings highlight that the soil organic matter preservation depends on the type of forest, but the soil microbial community was able to recover after about 15 years from coppice intervention in both forest ecosystems. Thus, the type of management implemented in these forest ecosystems, not negatively affecting soil organic matter pool, preserving microbial community and potentially soil ecological functions, is sustainable in a scenario of climate change.

Comparison of microbial diversity and metabolic activities in organic and conventional rice farms in Thailand.

Studies of the soil microbial community have revealed that several factors, both biotic and abiotic, can influence the community structure, diversity, and function. Human agricultural practice has been one of the key factors that affect microbial structure in soil. In this study, we examined the effect of two rice farming practices (i.e., conventional and organic) on soil microbial diversity and their metabolic activities. We proposed that the use of herbicides and chemical fertilizers in the conventional rice farming may decrease the soil microbial diversity and that it may lead to a decline in soil quality. However, our results show that there is no significant difference in the soil chemical properties, microbial diversity, and microbial metabolic activities between the two agricultural management systems. The wetland water cycling regime of rice agriculture in Thailand perhaps prevents the accumulation of chemicals in the soil, thus subdue the effect of these chemicals on soil microbes. Furthermore, our results showed that the key determinant of soil chemical properties as well as microbial community structure and diversity is the soil physical property, while the farming activity influences the microbial metabolic functions. These findings indicate the complex nature of the soil, microbes, and human farming interactions.IMPORTANCERice is a major export commodity in Thailand. As such, an understanding of the effect of conventional or organic farming approaches on soil microbial community could enable a suitable farming management. In this study, microbial communities were surveyed and compared between the two rice farming practices for their diversity and metabolic activities. Results showed no significant differences in microbial community structure and diversity between the two rice farming practices, but significant differences were observed due to the soil type, namely, clay, silty clay, silty clay loam, loam, and silty loam. Interestingly, significant differences in metabolic functions were also observed in different soil farming activities, such as land rest, period of growth, and post-burning, but not due to conventional or organic practices. These findings showed that the soil physical type and the farming activity impact the microbial community more than whether it is conventional or organically farmed.

Drought stress impacts soil microbial nutrient limitation more strongly than O3 pollution

Summer drought conditions generally coincide with exposure to high ground-level concentrations of ozone (O3). These two stressors compromise the availability of water, energy, and nutrients to vegetation and microbial communities present within the soil. However, relatively little is known regarding the responses of rhizosphere and non-rhizosphere soil microbial nutrient acquisition to conditions of drought and O3 exposure. Here, we used ecoenzymatic stoichiometry modeling and vector analyses to decipher the resource constraints on rhizosphere and non-rhizosphere soil microbial metabolic limitation within the soil of hybrid poplars exposed to two watering regimens (well-watered or reduced water of 40 %) and two chronic O3 treatments (charcoal-filtered air or ambient air +40 ppb of O3). Results show that, simulated drought stress resulted in stronger effects on the structure of soil microbial communities and associated enzymatic activity relative to exposure to elevated O3 levels. Exposure to these two stressors induced a microbial phosphorus (P) limitation response, with these limitation effects being more pronounced in the rhizosphere soil relative to non-rhizosphere soil. Structural equation modeling revealed that drought-associated declines in soil water availability and pH values were the primary factors influencing this microbial P limitation. Reduced soil water levels resulted in the exacerbation of microbial P limitation as a consequence of the inhibition of P mineralization and availability of C and N, whereas declining soil pH values had the opposite effect, alleviating microbial P limitations by reducing soil alkalinity and thereby aiding in the dissolution of the insoluble P present in alkaline soil. These results emphasize the complex responses of rhizosphere and non-rhizosphere soil microbial metabolic activity to elevated O3 levels and drought stress, providing invaluable insight into the importance of elemental stoichiometry-driven microbial metabolic variability of nutrient cycling activity at the root-soil interface in O3-polluted and drought-exposed ecosystems.

Soil microbial functional diversity is primarily affected by soil nitrogen, salinity and alkalinity in wetland ecosystem

Wetland ecosystems, recognized as important carbon sinks, face challenges due to climate change. Although wetland soil microorganisms play a major role in the wetland carbon cycle, the mechanisms influencing their spatial distribution remain elusive. Here, we investigate the impact of soil properties on the spatial distribution of soil microorganisms across five saline-alkali wetlands (Xianghai [XH], Niuxintaobao [NX], Momoge [MMG], Zhalong [ZL], and Halahai [HLH]) within the Songnen Plain, China. We utilized the Biolog-ECO method to assess the carbon metabolic activity, microbial diversity, and carbon source utilization preference of soil microorganisms. Using the structural equation model, we analyzed factors influencing microbial carbon metabolic activity and microbial diversity. Results indicate that southern wetlands harbor soil microbial communities exhibiting higher carbon metabolic activity, Shannon index, and substrate richness, compared to their northern counterparts. Specifically, soil microorganisms in NX and XH wetlands demonstrate a preference for labile organic carbon, whereas those in HLH and ZL wetlands preferred to use recalcitrant carbon. Furthermore, soil microbial carbon metabolic activity is primarily regulated by soil total nitrogen content, whereas soil microbial diversity is primarily regulated by soil salinity and alkalinity. Our findings revealed the distribution characteristics and influencing factors of soil microorganisms in the Songnen Plain wetland, contributing to understanding the maintenance mechanism of wetland carbon sink function.

Effect of stand age on soil microbial metabolic limitation and enzyme activities in Robinia pseudoacacia L. plantations in the loess hilly‐gully region, China

AbstractSoil heterotrophic microorganisms play crucial roles in soil biogeochemical cycles by secreting extracellular enzymes, but their metabolism is often limited by resource availability. Identifying major resources that limit soil microbial metabolism could help to create reasonable management practices for the maintenance of ecosystem function. Robinia pseudoacacia plantations perform crucial functions in reconstituting the ecological integrity of disturbed ecosystems; however, soil microbial metabolic limitations in R. pseudoacacia plantations remain poorly understood. Herein, R. pseudoacacia plantations of four age classes (10, 22, 37, and 47 years old) were sampled in the loess hilly‐gully region of northern Shaanxi Province in China. This study analyzed the effects of stand age on soil extracellular enzyme activities and soil microbial metabolic limitation. It also determined the main factor driving soil microbial metabolic limitation and extracellular enzyme activities. The results of ecoenzymatic stoichiometry showed that nitrogen (N) limited soil microbial metabolism in R. pseudoacacia plantations. Soil microbial metabolism at 10 and 22 years old were more carbon (C)‐ and N‐limited than that at 37 and 47 years old. Except for N‐ and phosphorus (P)‐acquiring enzyme activities at 10–20 cm, soil C‐, N‐, and P‐acquiring enzyme activities (in 0–10 and 10–20 cm of soil) at 10 and 22 years old were significantly higher than those at 37 and 47 years old. The enzymatic ratio of C:N:phosphorus(P) acquisition in all R. pseudoacacia plantations, regardless of stand age and soil depth, deviated from 1:1:1. Soil nutrients explained most of the variation (90.80%) in soil extracellular enzyme activities based on variation‐partitioning analysis. The partial least squares path model showed that soil nutrients had the highest total negative effect on microbial C limitation and microbial N/P limitation. Our results confirmed that microbial metabolic limitation was induced by soil resource deficits. The findings provide another visual that increases our understanding of soil microbial metabolic limitation in R. pseudoacacia plantations.

Assessing the impact of biofertilizer on soil microbial dynamics and metabolic activity in a controlled maize pot-grown experiment

Biofertilizers, consisting of carefully selected microorganisms across various species and genera, exhibit distinct features that enhance soil fertility and promote plant growth. Embracing the principles of eco-friendly agriculture, the use of biofertilizers emerges as a pivotal strategy for sustainable farming, contributing to environmental preservation and the overall health and biodiversity of the soil. In this study, a commercially available biofertilizer, containing a specialized strain of Priestia megatherium with nitrogen-fixing capabilities, was employed alongside chemical fertilizers at two different doses (30 and 40 mg per kg of soil). The primary objective was to evaluate the impact of biofertilizer on the metabolic activity and structure of microbial communities in a short-term experiment involving potted maize plants, utilizing the BIOLOG® EcoPlates technique. Parameters such as average well-color development (AWCD) and substrate utilization across six guilds (SAWCD) were assessed to gauge microbial metabolic activity. Additionally, functional indexes, including Shannon diversity, Shannon evenness, and Simpson diversity, were calculated as indicators of soil microbial community functionality. While statistically significant differences in AWCD among the studied variants were not observed, all estimated functional indexes consistently revealed heightened microbial diversity and evenness following the application of biofertilizer. This noteworthy finding, achieved within a relatively short period of plant cultivation, underscores the necessity for further research to explore the biofertilizer's enduring effects on soil communities, both in controlled laboratory environments and under real-world field conditions.

Biochar application induces different responses of bacterial and fungal communities to metabolic limitation

AbstractBiochar, an environmentally friendly soil amendment, has the potential to alleviate soil degradation. However, the mechanisms and effects of biochar addition on soil microbes and metabolic limitation in degraded soils are still ambiguous because of the complex ecosystem and the varying conditions of planting. Therefore, a field experiment with six biochar application rates (0, 24, 60, 96, 132, and 168 Mg ha−1, applied in the 0–20 cm soil layer) was conducted in a loessial soil without planting to determine the response of metabolic limitation and the microbial community to the biochar application rate. Soil samples were collected in mid‐May and mid‐August (before and during the rainy season). The results showed that the soil microbial metabolic activity without biochar application was phosphorus (P) limited in mid‐May (vector angle = 81.37 ± 0.31°) and nitrogen (N) limited in mid‐August (vector angle = 27.36 ± 1.54°), and nutrient limitations generally decreased in the biochar treatments. Structural equation modeling indicated that microbial metabolic restriction was mainly influenced by biochar‐induced changes in element stoichiometric ratios and available phosphorus (p < 0.05). Under the influence of biochar content, only the carbon (C) limitation influenced bacterial composition by directly or indirectly affecting dissolved organic carbon and NO3−–N. However, the fungal composition was strongly shaped by both C limitation and P/N limitation. Our results indicated that the soil P status and its stoichiometric ratios play an important role in the biochar effects on metabolic limitation and that bacteria and fungi have different compositional responses to metabolic limitation in loessial soil.

The Impact and Regulatory Mechanisms of Nitrogen Fertilizer on Soil Carbon Cycling

Modern agriculture must increase production to meet the growing global demand for food. However, excessive use of nitrogen fertilizer has led to a series of environmental issues. The impact of nitrogen fertilizer on soil carbon cycling is of particular concern, as agriculture is a major global carbon reservoir, and soil carbon cycling is crucial for maintaining ecosystem functionality. Previous research indicates that nitrogen fertilizer not only affects soil carbon storage but also alters the structure and function of soil microbial communities, thereby influencing carbon decomposition rates. Nevertheless, the specific mechanisms underlying the effects of nitrogen fertilizer on soil carbon cycling remain elusive. This study aims to comprehensively and systematically analyze the impact of nitrogen fertilizer on soil carbon cycling and elucidate its regulatory mechanisms. Nitrogen fertilizer exerts complex effects on soil, potentially promoting organic carbon accumulation, yet under certain conditions, it may also lead to carbon losses, closely intertwined with its impact on soil microbial communities. Regional and seasonal variations in the response to nitrogen fertilizer highlight the intricate dynamics of soil carbon storage, emphasizing the need for a thorough spatiotemporal investigation. Nitrogen fertilizer also influences soil microbial diversity, resulting in the excessive enrichment of certain microbial populations, disrupting the balance of microbial diversity in the soil. Moderate use of nitrogen fertilizer stimulates soil microbial metabolic activity, whereas excessive use may lead to a decline in metabolic activity. The regulation of soil carbon decomposition rates involves soil enzyme activity and organic matter decomposition pathways, with moderate nitrogen fertilizer application promoting soil carbon cycling, while high concentrations may impede carbon decomposition pathways. In conclusion, nitrogen fertilizer management should be guided by scientific principles to avoid excessive use. Establishing a systematic monitoring system that considers soil nutrients, microbial communities, and other factors is essential for achieving sustainable nitrogen fertilizer use. This approach aims to promote sustainable agriculture, reduce the burden on ecosystems, and enhance soil health and carbon storage.

Effects of two apple rootstocks on the soil microecology of replanted apple orchard soil

Apple replant disease (ARD) seriously restricts the development of the apple industry. It is of great significance for the sustainable development of the apple industry to alleviate ARD by using rootstock resistance. Taking M9T337 and Malus robusta Rehd. as test materials, the differences in biomass, soil physical and chemical properties, soil enzyme activity, soil microbial metabolic activity and carbon source utilization characteristics, soil microbial quantity, and soil fungal community of two apple rootstocks in replanted soil. The results showed that the biomass, related soil enzyme activities, and AWCD metabolic activity of apple saplings with Malus robusta Rehd. as rootstock were better, the number of soil fungi and the copy number of four Fusarium genes were lower, and more beneficial bacteria were enriched. In conclusion, the apple saplings with Malus robusta Rehd. as rootstock have a better improvement effect on the replanted soil environment and are more suitable as rootstocks to prevent and control ARD than M9T337.

Enhancing the effect of biochar ageing on reducing cadmium accumulation in Medicago sativa L

Biochar (BC) application to farmland soil can reduce the mobility and bioavailability of Cd. Nevertheless, BC is prone to natural ageing in soil, which alters its structure, physicochemical properties, thereby affecting the immobilisation of Cd. We used dry–wet and freeze–thaw cycles to mimic the natural ageing of BC, and used adsorption experiments to explore the changes of Cd adsorption capacity of BC and aged BC (ABC). We conducted a pot experiment to investigate the effects of BC and ABC on soil biotic and abiotic factors, alfalfa growth, and Cd accumulation in agricultural soils with high and low Cd concentrations. The increase of specific surface area, pore size, oxygen containing functional groups and mineral composition leads to better adsorption capacity of ABC. The adsorption of Cd(II) by BC and ABC is mainly by monolayer adsorption and chemical adsorption. Applying BC and ABC to Cd-contaminated soil significantly increased the aboveground biomass and decreased the Cd accumulation by reducing the Cd bioconcentration factor in alfalfa. At high Cd levels, adding BC and ABC reduced the Cd content in alfalfa shoots by 32.8 % and 35.1 %, respectively; the fixing effect of ABC was better than that of BC. Adding BC and ABC significantly increased the microbial biomass and geometric mean of enzymes. BC addition increased soil pH by 0.32–0.36 units and cation exchange capacity (CEC) by 15.5 %. Adding BC and ABC significantly increased soil organic matter (SOM) by 5.7 % and 6.2 %, respectively. Random forest analysis showed that SOM, total organic carbon, and fluorescein diacetate hydrolase were important variables for Cd content in alfalfa shoots. Structural equation modelling showed that BC indirectly affected the Cd content in alfalfa shoots by affecting soil pH, CEC, SOM, microbial biomass, and microbial metabolic activity. BC has a long-term effect on alleviating Cd pollution in farmland.

Changes in soil microbial metabolic activity following long‐term forest succession on the central Loess Plateau, China

AbstractForest succession can alter terrestrial ecosystem processes and potentially impact subsurface carbon dynamics. However, the effects of long‐term forest succession on soil microbial metabolic activity remain unclear. This study investigated a long‐term forest succession approximately ~160 years after farmland abandonment on the central Loess Plateau, China, to explore the changes in soil microbial metabolism following the forest succession. Among them, the soil basal respiration (BR), microbial quotient (qMB), and metabolic quotient (qCO2) were studied. The results showed that the BR and qCO2 were increased following forest succession, whereas the qMB showed the opposite trend. Forest succession also increased the activities of β‐1,4‐glucosidase (BG) and β‐D‐cellobiosidase (CBH). The increase of BR and qCO2 and the decrease of qMB occurred in the early stage (<60 years). However, the increase of BG and CBH mainly occurred in the later period (>100 years). Soil microbial metabolic activity was closely related to litter biomass, dissolved organic carbon, fungal, and bacterial community composition. In conclusion, long‐term forest succession changes soil microbial community structure and promotes soil organic carbon accumulation, while the increase of soil respiration and metabolism promotes the release of carbon element following forest succession, thus reducing soil microbial carbon utilization efficiency.

Digestate Management and Processing Practices: A Review

The implementation of sustainable agro-energy systems that integrate crop, livestock, and bioenergy production is attracting increasing interest from farmers. Livestock produces large amounts of animal manure which can serve as organic fertilizer for crops and pasture growth. However, the nutrients contained in manure can adversely affect air, water, and soil quality and pose a public health risk if not handled properly. Existing manure management practices vary widely on a global scale. Researchers are striving to identify appropriate manure management practices with the aim of environmental protection. Anaerobic digestion of manure and subsequent digestate (DG) processing technologies have been proposed to stabilize manure so that it can be safely used for land applications. DG, which represents digested substrate removed from the anaerobic reactor after recovery of biogas, is a rich source of N, P, K, and S, various micronutrients, and organic matter, the addition of which to the soil can stimulate soil microbial biomass metabolic activities thus improving soil ecosystem function. However, the optimal fertilization properties of DG can be lost if it is neither fully stabilized nor contains biodegradable materials. To overcome these problems, various processing technologies can be used to convert DG into value-added by-products. Composting has been proposed as one such preferred post-treatment that can convert DG into mature, stable, safe, humus- and nutrient-rich compost. Other processing technologies such as thermal drying, gasification, hydrothermal carbonization, pyrolysis, membrane filtration, struvite precipitation, ammonia stripping, and evaporation have also been proposed for DG processing and nutrient recovery from DG. The objective of this review paper was to provide an overview of the current state of the art in DG management regulations and practices and to provide an update on the various processes that have been developed to meet DG stabilization requirements, with a focus on composting as one of the preferred solutions.

Deep soil microbial carbon metabolic function is important but often neglected: a study on the Songnen Plain reed wetland, Northeast China

Soil microbial carbon metabolism is critical in wetland soil carbon cycling, and is also a research hotspot at present. However, most studies focus on the surface soil layer in the wetlands and the microorganisms associated with this layer. In this study, 0-75 cm soil profiles were collected from five widely separated reed wetlands in the Songnen Plain. which has a large number of middle-high latitude inland saline-sodic wetlands. The Biolog-ECO method was used to determine the carbon metabolic activity and functional diversity of soil microorganisms. The results showed that soil carbon metabolic activity decreased with increasing soil depth. The carbon metabolic activity of soil microorganisms in the 60-75 cm layer was approximately 57.41%-74.60% of that in the 0-15 cm layer. The soil microbial Shannon index and utilization rate of amines decreased with an increase in soil depth, while the Evenness index and utilization rate of polymers tended to increase with soil depth. Dissolved organic carbon (DOC) is the most important factor affecting microbial carbon source utilization preference, because microorganisms mainly obtain the carbon source from DOC. The result of the correlation analysis showed that the soil microbial carbon metabolic activity, Shannon index, and Evenness index significantly correlated with soil total carbon (TC), microbial biomass carbon (MBC), DOC, total nitrogen (TN), ammonium nitrogen (NH4+-N), nitrate nitrogen (NO3−-N) contents, and electrical conductivity (EC). This study emphasized the important role of microbial carbon metabolic function in deep soil.

MICROBIAL METABOLIC ACTIVITY AS LEGACY OF AGRICULTURAL MANAGEMENT IN MAIZE AGROECOSYSTEMS FROM MEXICO HIGHLANDS

<p><strong>Background</strong>. Soil ecological functions such as C mineralization, enzyme activity, and microbial biomass determine the maintenance of soil fertility in the short and long term. Microbial activity is a sensitive indicator of changes in soils under agricultural management. <strong>Objective</strong>. Evaluate the metabolic response of soil microbial communities in two temperate maize agroecosystems with different management intensities. <strong>Methodology</strong>. This study evaluated total soil nutrient concentrations, C mineralization, and microbial metabolic activity by comparing two agricultural regimes. The first one is an intensive regime (IR) characterized by the exclusive use of synthetic fertilizers in a maize monoculture. The second one is a traditional regime (TR) characterized by the use of mixtures of organic matter (maize and bean residues and manure) with synthetic fertilizers in a rotation system of maize and beans. Physical, chemical, and biological properties were tested in the laboratory, and the specific enzyme activity (SEA) and metabolic quotient (qCO<sub>2</sub>) were calculated. <strong>Results.</strong> Total soil C concentration was 19% higher in TR (26.6 mg g<sup>-1</sup>) than in IR (5.1 mg g<sup>-1</sup>); total C biomass was 30% higher in TR (279 mg C g<sup>-1</sup>) versus IR (83.9 mg C g<sup>-1</sup>), and potential C mineralization was 40% higher in TR (356 µg C g<sup>-1</sup> d<sup>-1</sup>) than IR (214 µg C g<sup>-1</sup> d<sup>-1</sup>); in contrast, SEA and qCO2 were lower in TR versus IR. These results support the hypothesis that the microbial community is more efficient under TR than IR because it produces extracellular and intracellular enzymes while growing in biomass. <strong>Implications</strong>. The present study provides new information about the effect of agricultural management on microbial activity, which is important for farmers not only in Mexico Highlands but also in any agricultural scenario exposed to changes in management practices. <strong>Conclusions</strong>. Assessment of biological soil properties is a sensitive indicator of changes in soil properties induced by management. Metabolic indices are suitable for the evaluation of ecological functions in cultivated soils.</p>

Responses of Soil Microbial Metabolic Activity and Community Structure to Different Degraded and Restored Grassland Gradients of the Tibetan Plateau.

Climate change and land-use disturbances are supposed to have severely affected the degraded alpine grasslands on the Tibetan Plateau. Artificial grassland establishment has been implemented as a restoration tool against grassland degradation. However, the impact of such degradation and restoration processes on soil microbial communities and soil quality is not clearly understood. Here, we aim to investigate how the dynamics of microbial community and soil quality of alpine grasslands respond to a gradient of degradation and that of restoration, respectively. We conducted a randomised experiment with four degradation stages (light, moderate, heavy, and extreme degradation) and three restoration stages (artificial restoration for 1, 5, and 10 years). We analysed the abundance and diversity of soil bacteria and fungi, and measured soil nutrients, enzymatic activity and microbial biomass. The concentration of soil nitrogen (TN), soil organic matter (OM) in heavy degraded grassland decreased significantly by 37.4 and 45.08% compared with that in light degraded grassland. TN and OM in 10-years restored grassland also increased significantly by 33.10 and 30.42% compared to that in 1-year restored grassland. Four soil enzymatic activity indicators related to microbial biomass decreased with degradation gradient and increased with recovery time (i.e., restoration gradient). Both bacterial and fungal community structure was significantly different among grassland degradation or restoration successional stages. The LEfSe analysis revealed that 29 fungal clades and 9 bacterial clades were susceptible to degraded succession, while16 fungal clades and 5 bacterial clades were susceptible to restoration succession. We conclude that soil quality (TN, OM, and enzymatic activity) deteriorated significantly in heavy degraded alpine grassland. Soil microbial community structure of alpine is profoundly impacted by both degradation and restoration processes, fungal communities are more sensitive to grassland succession than bacterial communities. Artificial grasslands can be used as an effective method of restoring degraded grassland, but the soil functions of artificial grassland, even after 10 years of recovery, cannot be restored to the original state of alpine grassland.

Effects of intercropping and nitrogen application on soil microbial metabolic functional diversity in maize cropping soil.

Soil microorganism is an important indicator of soil health and plays a critical role in biogeochemical processes. We collected soil samples from a multi-year in-situ field experiment with two cropping modes (maize monocropping and maize-potato intercropping) subjected to four nitrogen (N) levels (N0, 0 kg·hm-2; N125, 125 kg·hm-2; N250, 250 kg·hm-2; N375, 375 kg·hm-2). By using the Biolog-ECO microplate method, soil microbial metabolic activity, diversity and utilization of six carbon groups were analyzed. The results showed that N application significantly increased the average well color development (AWCD) values, Simpson and Shannon indices, with the highest value at N250 in mono- and inter-cropped soils. Moreover, N application promoted the utilization of amino acids, amines and phenolic compounds, but decreased the utilization of polymers. Compared with monocropping at the same N level, intercropping improved the AWCD values, Simpson and Shannon indices, with a maximal improvement at N250. Intercropping and nitrogen application significantly affected the metabolic activities of the six carbon-source groups. In addition, intercropping improved the utilization of labile carbon sources, such as amino acids and carbohydrates. Results of the redundancy analysis and linear regression showed that intercropping and N application elevated AWCD values by increasing soil temperature, water content, and soil organic carbon content. Therefore, the changes in soil physicochemical properties after intercropping were the main reason for the enhancement of microbial metabolic activity under N application.

Response of Functional Diversity of Soil Microbial Community to Forest Cutting and Regeneration Methodology in a Chinese Fir Plantation

With the expansion of pure forest planting area and the increase in the number of rotations used, soil activity and plant productivity have significantly reduced. The functional diversity of soil microorganisms plays a vital role in forest health and the long-term maintenance of productivity. Though the optimization of forest cutting and regeneration methodologies is necessary to improve the functional diversity of soil microorganisms, the effects of harvest residual treatment on the functional diversity of soil microorganisms remain unclear. During the period 2018–2020, we designed four harvest residual treatments—reference (RF), residual burning (RB), crushing and mulching (MT), and no residuals (NR)—to determine soil physical and chemical properties. We also used microbial biomass (MB) to evaluate the diversity in carbon source metabolism of soil microorganisms through Biolog microplate technology, and discussed the response mechanism of microbial functional diversity to the different forest cutting and regeneration methodologies used in Chinese fir plantations. The results indicated that RB significantly increased the carbon metabolic capacity of the microbial community, the community richness, and its dominance compared to RF, MT, and NR; however, they also showed that it decreased the uniformity of the soil microbial community. NR showed a poor carbon utilization capacity for microorganisms compared to RF and MT, while MT significantly increased the utilization capacity of carbohydrate and amino acid carbon compared with RF. Soil nutrients were the main driving factors of soil microbial carbon metabolic activity, and the different responses of microbial functional diversity to various forest cutting and regeneration methodologies were mainly due to the variation in the nutrient inputs of harvest residues. This study provides a practical basis for enhancing the functional diversity of soil microorganisms in plantations through the management of harvest residues.
