Diversity and activity of soil biota at a post-mining site highly contaminated with Zn and Cd are enhanced by metallicolous compared to non-metallicolous Arabidopsis halleri ecotypes.

Hyperaccumulators' ability to take up large quantities of harmful heavy metals from contaminated soils and store them in their foliage makes them promising organisms for bioremediation. Here we demonstrate that some ecotypes of the zinc hyperaccumulator Arabidopsis halleri are more suitable for bioremediation than others, because of their distinct influence on soil biota. In a field experiment, populations originating from metal-polluted and unpolluted soils were transplanted to a highly contaminated metalliferous site in Southern Poland. Effects of plant ecotypes on soil biota were assessed by measurements of feeding activity of soil fauna (bait-lamina test) and catabolic activity and functional diversity of soil bacteria underneath A. halleri plants (Biolog® ECO plates). Chemical soil properties, plant morphological parameters, and zinc concentration in shoots and roots were additionally evaluated. Higher soil fauna feeding activity and higher bacterial community functional diversity were found in soils affected by A. halleri plants originating from metallicolous compared to non-metallicolous ecotypes. Differences in community-level physiological profiles further evidenced changes in microbial communities in response to plant ecotype. These soil characteristics were positively correlated with plant size. No differences in zinc content in shoots and roots, zinc translocation ratio, and plant morphology were observed between metallicolous and non-metallicolous plants. Our results indicate strong associations between A. halleri ecotype and soil microbial community properties. In particular, the improvement of soil biological properties by metallicolous accessions should be further explored to optimize hyperaccumulator-based bioremediation technologies.

Changes in the composition of soil microbial communities and their carbon‐cycle genes following the conversion of primary broadleaf forests to plantations and secondary forests

AbstractThe soil organic carbon (C) cycle is primarily mediated by soil microorganisms and their genes that function in the C cycle (C‐cycle genes), both of which are strongly affected by land cover disturbance. However, the mechanism underlying microbially mediated soil C loss after conversion of primary natural broadleaf forests (BF) to plantation forests (PF) and secondary forests (SF) remains unknown. Here, we measured soil physicochemical properties and soil microbial community properties, and examined their linkages with microbial C‐cycle genes. Forest conversion dramatically decreased the richness of the soil fungal community but not of the bacterial community, and altered the composition of both communities. Analysis of C‐cycle genes revealed that the abundance of genes associated with C fixation, methane metabolism, and C degradation decreased by 51.3%, 57.9%, and 67.0%, respectively with the conversion of BF to PF; and by 6.3%, 4.1%, and 15.6%, respectively, with the conversion of BF to SF. The reductions in the abundance of C‐cycle genes, especially the reduction of hemicellulose‐ and lignin‐degradation genes, were primarily associated with the declines in the abundance of forest conversion‐sensitive microbes indexed by operational taxonomic units (fsOTUs, = 0.41). fsOTUs were taxonomically diverse and included members frequently co‐occurring with numerous other microbes in the microbial communities, indicating that the manipulation of fsOTUs by forest management could improve soil fertility and soil C sequestration. Forest conversion‐induced shifts in fsOTUs abundance were associated with changes in soil potassium permanganate oxidizable organic carbon (PXC) concentration, dissolved organic carbon (DOC) concentration, and soil pH. Our results indicate that alterations in soil substrate supply (e.g., DOC and PXC) and soil pH induced by forest conversion may strongly shape fsOTUs structure and decrease the abundance of hemicellulose and lignin degradation genes, and consequently increase C loss.

Large‐scale drivers of relationships between soil microbial properties and organic carbon across Europe

AbstractAimQuantify direct and indirect relationships between soil microbial community properties (potential basal respiration, microbial biomass) and abiotic factors (soil, climate) in three major land‐cover types.LocationEurope.Time period2018.Major taxa studiedMicrobial community (fungi and bacteria).MethodsWe collected 881 soil samples from across Europe in the framework of the Land Use/Land Cover Area Frame Survey (LUCAS). We measured potential soil basal respiration at 20 ºC and microbial biomass (substrate‐induced respiration) using an O2‐microcompensation apparatus. Soil and climate data were obtained from the same LUCAS survey and online databases. Structural equation models (SEMs) were used to quantify relationships between variables, and equations extracted from SEMs were used to create predictive maps. Fatty acid methyl esters were measured in a subset of samples to distinguish fungal from bacterial biomass.ResultsSoil microbial properties in croplands were more heavily affected by climate variables than those in forests. Potential soil basal respiration and microbial biomass were correlated in forests but decoupled in grasslands and croplands, where microbial biomass depended on soil carbon. Forests had a higher ratio of fungi to bacteria than grasslands or croplands.Main conclusionsSoil microbial communities in grasslands and croplands are likely carbon‐limited in comparison with those in forests, and forests have a higher dominance of fungi indicating differences in microbial community composition. Notably, the often already‐degraded soils of croplands could be more vulnerable to climate change than more natural soils. The provided maps show potentially vulnerable areas that should be explicitly accounted for in future management plans to protect soil carbon and slow the increasing vulnerability of European soils to climate change.

Microbial Features Indicating the Recovery of Soil Ecosystem Strongly Affected by Mining and Ore Processing.

Tailings-derived soils formed from waste materials produced during mineral processing often exhibit extremes of pH, low content of organic matter and limited nutrient availability. The success of site revitalization depends mostly on the ability to maintain natural soil functions. We analyzed technogenic sediments from four selected localities in Slovakia defined as environmental burdens: Slovinky (SLS, SLD), Markušovce (MAS, MAD), Lintich (LIS, LID), Horná Ves (HVS, HVD) in the presented research. None of these sites has long been used for its original purpose. In all localities, the concentrations of several risk elements (As, Ba, Cd, Co, Cr, Cu, Ni, Pb, Zn) still significantly exceed the statutory limit values. Besides the content of risk elements, the amounts of organic carbon, total nitrogen, pH value and moisture level in technogenic substrates were determined. We evaluated selected microbiological parameters, including microbial biomass carbon (MBC), microbial respiration and cellulolytic activity to determine how soil organisms tolerate long-term pollution. In general, the values of microbiological parameters were not as low as one would expect. The results confirmed a negative correlation between MBC content and concentrations of several toxic metals (Co, Cr, Cu, Ni, Zn). The values of assessed microbial indicators were in several cases comparable to those in natural soils. We noticed the lowest metabolic quotient values (qCO2) in the heavily polluted locality HVS. The microbial quotient (qMic) was low in every locality except HVS, where the substrate availability index (SAI) was highest. The soil microbial community properties have shown that, despite adverse conditions, these emerging soils allow the growth and development of microorganisms to such an extent that they can adequately use available (although limited) nutrients. The data obtained suggest that these severely impacted soil ecosystems can restore their original environmental functions in time.

Plant biomass, soil water content and soil N:P ratio regulating soil microbial functional diversity in a temperate steppe: A regional scale study

Soil microorganisms are influenced by various abiotic and biotic factors at the field plot scale. Little is known, however, about the factors that determine soil microbial community functional diversity at a larger spatial scale. Here we conducted a regional scale study to assess the driving forces governing soil microbial community functional diversity in a temperate steppe of Hulunbeir, Inner Mongolia, northern China. Redundancy analysis and regression analysis were used to examine the relationships between soil microbial community properties and environmental variables. The results showed that the functional diversity of soil microbial communities was correlated with aboveground plant biomass, root biomass, soil water content and soil N: P ratio, suggesting that plant biomass, soil water availability and soil N availability were major determinants of soil microbial community functional diversity. Since plant biomass can indicate resource availability, which is mainly constrained by soil water availability and N availability in temperate steppes, we consider that soil microbial community functional diversity was mainly controlled by resource availability in temperate steppes at a regional scale.
