Microplastic coupled with soil dissolved organic matter mediated changes in the soil chemical and microbial characteristics.

The abundance of microplastics (MPs) in soil environments has attracted significant attentions, due to their impact on soil physico-chemical properties. However, limited information is available on the influences of MPs on soil carbon composition and microbial utilization characteristics. Therefore, a two-month incubation experiment was conducted to add polyethylene microplastics (PE-MPs) with different levels (1%, 10%) and sizes (150-300 μm and 75-150 μm) into different soils. After that, soil chemical properties including the dissolved organic carbon (DOC), spectral characteristics of dissolved organic matter (DOM) and soil microbial characteristics were analyzed. Results revealed that PE-MPs addition caused significant differences in soil chemical properties between farmland and woodland soils, particularly in soil pH, DOM composition, and soil phosphatase activity. Woodland soil always exhibited higher levels of DOC content, microbial diversity, and soil carbon source utilization compared to farmland soil, leading to increased humification in the DOM of woodland soil. PE-MPs with a larger particle size significantly increased both the soil DOC content and enzyme activity. Addition of PE-MPs altered the soil DOM composition, and the fluorescence parameters like the biological index (BIX) and humification degree. Moreover, the carbon source utilization intensity of microorganisms on PE MPs-contaminated soils is higher in woodland soils. Various analyses confirmed that compared to other soil properties, characteristics of soil DOM had a more significant impact on soil microbial community composition. Thus, PE-MPs in conjunction with soil DOM spectral characteristics regulated soil microbial diversity, which is crucial for understanding soil carbon sequestration.

Changes in Rhizosphere Soil Microorganisms and Metabolites during the Cultivation of Fritillaria cirrhosa

Fritillaria cirrhosa is an important cash crop, and its industrial development is being hampered by continuous cropping obstacles, but the composition and changes of rhizosphere soil microorganisms and metabolites in the cultivation process of Fritillaria cirrhosa have not been revealed. We used metagenomics sequencing to analyze the changes of the microbiome in rhizosphere soil during a three-year cultivation process, and combined it with LC-MS/MS to detect the changes of metabolites. Results indicate that during the cultivation of Fritillaria cirrhosa, the composition and structure of the rhizosphere soil microbial community changed significantly, especially regarding the relative abundance of some beneficial bacteria. The abundance of Bradyrhizobium decreased from 7.04% in the first year to about 5% in the second and third years; the relative abundance of Pseudomonas also decreased from 6.20% in the first year to 2.22% in the third year; and the relative abundance of Lysobacter decreased significantly from more than 4% in the first two years of cultivation to 1.01% in the third year of cultivation. However, the relative abundance of some harmful fungi has significantly increased, such as Botrytis, which increased significantly from less than 3% in the first two years to 7.93% in the third year, and Talaromyces fungi, which were almost non-existent in the first two years of cultivation, significantly increased to 3.43% in the third year of cultivation. The composition and structure of Fritillaria cirrhosa rhizosphere metabolites also changed significantly, the most important of which were carbohydrates represented by sucrose (48.00–9.36–10.07%) and some amino acid compounds related to continuous cropping obstacles. Co-occurrence analysis showed that there was a significant correlation between differential microorganisms and differential metabolites, but Procrustes analysis showed that the relationship between bacteria and metabolites was closer than that between fungi and metabolites. In general, in the process of Fritillaria cirrhosa cultivation, the beneficial bacteria in the rhizosphere decreased, the harmful bacteria increased, and the relative abundance of carbohydrate and amino acid compounds related to continuous cropping obstacles changed significantly. There is a significant correlation between microorganisms and metabolites, and the shaping of the Fritillaria cirrhosa rhizosphere’s microecology by bacteria is more relevant.

Effects of reduced snowpack due to climate warming on abiotic and biotic soil properties in alpine and boreal forest systems

Reduction in snow cover, depth, onset, and duration of seasonal snow in mid-latitude regions due to climate warming has multiple global and local scale ecosystem impacts. These effects include modulations of the hydrological cycles and increases in land surface solar radiation absorption due to decreased albedo. Changes in snow cover characteristics also affect underlying soils. Snow has an insulating effect on soils by decoupling air and soil temperatures, thus seasonal snow cover reduction leads to overall lower soil temperatures and an increase in freeze-thaw cycles. This is especially prominent during the fall and spring thaw seasons when the snow cover is not as extensive. This in turn has downstream impacts on soil physical, chemical, and biological properties. Among these impacts are soil moisture reduction, temperature, frost regimes, soil pH shifts, and alteration in nutrient flux dynamics during winter, snowmelt period and the following summer growing season. These changes in soil physicochemical properties due to snowpack reduction can then impact the biological soil properties via increased plant root mortality, reduced abundance and diversity of soil arthropods, and shifts in composition, abundance and activity of soil microbial communities. All these soil biotic factors can in turn alter the dynamics of soil nutrient fluxes and future greenhouse gas emissions. Here, we integrate data on the effects of snow cover reduction on abiotic and biotic soil properties, with focus on temperate alpine and forest ecosystems and with an outlook on future impacts.

Analysis of Salt Stress on Soil Microbial Community Composition and Its Correlation with Active Components in the Rhizosphere of Acanthopanax senticosus.

Salt stress can adversely affect plant seed germination, growth and development, and eventually lead to slow growth and even death of plants. The purpose of this study was to investigate the effects of different concentrations of NaCl and Na2SO4 stress on the physicochemical properties, enzyme activities, rhizosphere microbial community and seven active components (L-phenylalanine, Protocatechuic acid, Eleutheroside B, Chlorogenic acid, Caffeic acid, Eleutheroside E, Isofraxidin) of Acanthopanax senticosus rhizosphere soil. Statistical analysis was used to explore the correlation between the rhizosphere ecological factors of Acanthopanax senticosus and its active components. Compared with Acanthopanax senticosus under NaCl stress, Na2SO4 generally had a greater effect on Acanthopanax senticosus, which reduced the richness of fungi in rhizosphere soil and adversely affected the content of multiple active components. Pearson analysis showed that pH, organic matter, ammonium nitrogen, available phosphorus, available potassium, catalase and urease were significantly correlated with active components such as Caffeic acid and Isofraxidin. There were 11 known bacterial genera, 12 unknown bacterial genera, 9 known fungal genera and 1 unknown fungal genus significantly associated with the active ingredient. Salt stress had great changes in the physicochemical properties, enzyme activities and microorganisms of the rhizosphere soil of Acanthopanax senticosus. In conclusion, different types and concentrations of salts had different effects on Acanthopanax senticosus, and the active components of Acanthopanax senticosus were regulated by rhizosphere soil ecological factors.

Synthetic consortia of four strains promote Schisandra chinensis growth by regulating soil microbial community and improving soil fertility.

Synthetic consortia performed better in promoting Schisandra chinensis growth than individual strains, and this result provides valuable information for the development of synthetic microbial fertilizers. Schisandra chinensis is an herbal medicine that can treat numerous diseases. However, the excessive reliance on chemical fertilizers during the plantation of S. chinensis has severely restricted the development of the S. chinensis planting industry. Plant growth-promoting rhizobacteria (PGPR) can promote the growth of a wide range of crops, and synthetic consortia of them are frequently superior to those of a single strain. In this study, we compared the effects of four PGPR and their synthetic consortia on S. chinensis growth. The pot experiment showed that compared with the control, synthetic consortia significantly increased the plant height, biomass, and total chlorophyll contents of S. chinensis, and their combined effects were better than those of individual strains. In addition, they improved the rhizosphere soil fertility (e.g., TC and TN contents) and enzyme activities (e.g., soil urease activity) and affected the composition and structure of soil microbial community significantly, including promoting the enrichment of beneficial microorganisms (e.g., Actinobacteria and Verrucomicrobiota) and increasing the relative abundance of Proteobacteria, a dominant bacterial phylum. They also enhanced the synergistic effect between the soil microorganisms. The correlation analysis between soil physicochemical properties and microbiome revealed that soil microorganisms participated in regulating soil fertility and promoting S. chinensis growth. This study may provide a theoretical basis for the development of synthetic microbial fertilizers for S. chinensis.

Fungal communities are passengers in community development of dune ecosystems, while bacteria are not.

An increasing number of studies of above-belowground interactions provide a fundamental basis for our understanding of the coexistence between plant and soil communities. However, we lack empirical evidence to understand the directionality of drivers of plant and soil communities under natural conditions: 'Are soil microorganisms driving plant community functioning or do they adapt to the plant community?' In a field experiment in an early successional dune ecosystem, we manipulated soil communities by adding living (i.e., natural microbial communities) and sterile soil inocula, originating from natural ecosystems, and examined the annual responses of soil and plant communities. The experimental manipulations had a persistent effect on the soil microbial community with divergent impacts for living and sterile soil inocula. The plant community was also affected by soil inoculation, but there was no difference between the impacts of living and sterile inocula. We also observed an increasing convergence of plant and soil microbial composition over time. Our results show that alterations in soil abiotic and biotic conditions have long-term effects on the composition of both plant and soil microbial communities. Importantly, our study provides direct evidence that soil microorganisms are not "drivers" of plant community dynamics. We found that soil fungi and bacteria manifest different community assemblies in response to treatments. Soil fungi act as "passengers," that is, soil microorganisms reflect plant community dynamics but do not alter it, whereas soil bacteria are neither "drivers" nor "passengers" of plant community dynamics in early successional ecosystems. These results are critical for understanding the community assembly of plant and soil microbial communities under natural conditions and are directly relevant for ecosystem management and restoration.

The Impact of Artificial Restoration of Alpine Grasslands in the Qilian Mountains on Vegetation, Soil Bacteria, and Soil Fungal Community Diversity

To understand how the soil microbial community structure responds to vegetation restoration in alpine mining areas, this study specifically examines the grassland ecosystem in the Qianmalong mining area of the Qilian Mountains after five years of artificial restoration. High-throughput sequencing methods were employed to analyze soil bacteria and fungi microbial characteristics in diverse grassland communities. Combined with modifications in vegetation diversity as well as soil physicochemical properties, the impact of vegetation restoration on soil microbiome diversity in this alpine mining area was investigated. The findings indicated that the dominant plants were Cyperus rotundus, Carex spp., and Elymus nutans. As the extent of the grassland’s restoration increased, the number of plant species, importance values, and plant community diversity showed an increasing trend. The plant functional groups were mainly dominated by Cyperaceae, followed by Poaceae. Plant height, density, plant cover, frequency, and aboveground biomass showed an increasing trend, and soil water content (SWC) increased. While soil pH and soil electrical conductivity (EC) exhibited a declining trend, available phosphorus (AP), total phosphorus (TP), total nitrogen (TN), nitrate nitrogen (NO3-N), soil organic carbon (SOC), and soil water content (SWC) showed an increasing trend. The dominant bacterial communities were Actinobacteriota, Proteobacteria, Acidobacteriota, Chloroflexi, Firmicutes, and Gemmatimonadota, while the dominant fungal communities were Ascomycota, Mortierellomycota, Basidiomycota, unclassified_k_Fungi, and Glomeromycota. Significant differences were detected within soil microbial community composition among different degrees of restoration grasslands, with bacteria generally dominating over fungi. SWC, TP, and TN were found to be the main soil physicochemical factors affecting the distribution of soil bacterial communities’ structure; however, SOC, TN, and NO3-N were the primary factors influencing the soil distribution of fungal communities. The results of this study indicate that different degrees of vegetation restoration in alpine mining areas can significantly affect soil bacterial and fungal communities, and the degree of restoration has varying effects on the soil bacteria and fungi community structure in alpine mining areas.

Impacts of o-cresol spill on composition and function of river sediment and soil microbial communities.

o-Cresol is a toxic substance with strong irritating and corrosive effects on skin and mucous membranes. To date, information on the effects of o-cresol on microbial communities in the natural environment is very limited. In the present study, 16S rRNA sequencing and metagenomic technique were carried out to elucidate the effects of the o-cresol spill on microbial communities in river sediments and nearby soils. o-Cresol spill induced the increase in the relative abundance of phyla Planctomycetes and Gemmatimonadetes, suggesting their resilience to o-cresol-induced stress. Uncultured Gemmatimonadetes genera and the MND1 genus exhibited enrichment, while the Pseudomonas genus dominated across all samples, indicating their potential pivotal roles in adapting to the o-cresol spill. Moreover, o-cresol spill impaired the metabolic functions of microbes but triggered their defense mechanisms. Under o-cresol pressure, microbial functions related to carbon fixation were upregulated and functions associated with sulfur metabolism were downregulated. In addition, the o-cresol spill led to an increase in functional genes related to the conversion of o-cresol to 3-methylcatechol. Several genes involved in the degradation of aromatic compounds were also identified, potentially contributing to the biodegradation of o-cresol. This study provides fresh insights into the repercussions of an abrupt o-cresol spill on microbial communities in natural environments, shedding light on their adaptability, defense mechanisms, and biodegradation potential.

The Diversity and Composition of Soil Microbial Communities Differ in Three Land Use Types of the Sanjiang Plain, Northeastern China.

In recent years, the Sanjiang Plain has experienced drastic human activities, which have dramatically changed its ecological environment. Soil microorganisms can sensitively respond to changes in soil quality as well as ecosystem function. In this study, we investigated the changes in soil microbial community diversity and composition of three typical land use types (forest, wetland and cropland) in the Sanjiang Plain using phospholipid fatty acid analysis (PLFA) technology, and 114 different PLFA compounds were identified. The results showed that the soil physicochemical properties changed significantly (p < 0.05) among the different land use types; the microbial diversity and abundance in cropland soil were lower than those of the other two land use types. Soil pH, soil water content, total organic carbon and available nitrogen were the main soil physico-chemical properties driving the composition of the soil microbial community. Our results indicate that the soil microbial community response to the three different habitats is complex, and provide ideas for the mechanism by which land use changes in the Sanjiang Plain affect the structure of soil microbial communities, as well as a theoretical basis for the future management and sustainable use of the Sanjiang plain, in the northeast of China.

Untargeted metabolomics to study changes in soil microbial community in response to tillage practices

Soil microbes are the main drivers of nutrient turnover in agro-ecosystems. The series of biochemical processes involved by these microorganisms are inseparable from the regulation of metabolites in the root-zone soils. Therefore, understanding the evolution of microbial communities driven by root-zone soil metabolites will help optimize agricultural management practices. Based on a field experiment, we explore the changes in soil properties, microbes, and metabolites under three tillage practices such as no-tillage with straw mulching (NTS), rotary tillage, and plough tillage with straw returning (RTS and PTS) based on untargeted metabolomics. We found that NTS helped to enhance nutrient contents and enzyme activities in the 0–20 cm soil layer. Specifically, compared with RTS and PTS, NTS increased grain yield by 6.4 % and 8.5 %, and SOC by 13.3 % and 5.7 %, respectively, but decreased pH by 2.8 % and 1.1 %. In addition, compared with NTS, RTS and PTS decreased the fungal richness by 16.5 % and 8.1 %, and decreased the fungal shannon index by 16.4 % and 9.2 %. However, tillage practices did not result in substantial differences in the alpha diversity of bacterial community. Both bacterial and fungal community composition were substantially affected by tillage practices and they were both significantly correlated with soil metabolites. In the symbiotic networks, NTS significantly increased the abundance of microorganisms with high connectivity, while RTS significantly increased the concentration of metabolites with high connectivity. Amino acids, as the main drivers, were significantly and positively associated with bacterial and fungal community composition, mainly enriched in the metabolic pathways of D − glutamine and D − glutamate metabolism and arginine and proline metabolism, and correlated with soil properties negatively and carbohydrates positively, directly or indirectly affecting soil microbial community composition. Our findings would provide new insight into metabolomics for the in-depth understanding of the metabolic mechanism for soil microbial community changes in dryland wheat fields under different tillage practices.

Response of Soil Physicochemical Properties and Microbial Community Composition in Larix olgensis Plantations to Disturbance by a Large Outbreak of Bark Beetle

Forests are affected by a wide range of disturbances globally, resulting in the decline or death of large areas of them. There is a lack of comparative studies on how soil properties change in forests that die under the influence of disturbances, especially considering different levels of disturbance. For this study, we took Larix olgensis—a major plantation forest species in northeast China—as the research object, one in which a large outbreak of bark beetle led to large-scale forest death, and set up fixed sample plots characterized by different disturbance intensities. We investigated the responses of soil physicochemical properties and microbial community compositions to different disturbance intensities through the determination of soil nutrient indices and high-throughput sequencing. The results show that there were significant differences (p &lt; 0.05) in the effects of different disturbance intensities on soil physicochemical properties, where the soil moisture content, total nitrogen, total carbon, and total phosphorus in the control group were significantly higher than those in the disturbed groups. The soil pH was highest under low-intensity disturbance and the soil total potassium content was highest under high-intensity disturbance. At different disturbance intensities, the highest soil moisture content was found in the high-intensity group. Proteobacteria, Actinobacteria, Verrucomicrobia, Acidobacteria, Candidatus_Rokubacteria, Chloroflexi, Gemmatimonadetes, and Thaumarchaeota were the dominant populations with higher abundances; meanwhile, the relative abundance of Bacteroidetes, Tenericutes, and a tentatively unclassified fungus differed significantly (p &lt; 0.05) across disturbance intensities. Among the dominant microbial populations, Acidobacteria showed a significant negative correlation with soil pH and a significant positive correlation with total potassium content, Thaumarchaeota showed significant positive correlations with soil moisture content and total nitrogen content, and Firmicutes and Gemmatimonadetes showed significant negative correlations with total carbon content in the soil. Furthermore, soil total nitrogen content was the key factor driving changes in microbial communities. The results of this study provide a scientific basis for the study of the long-term effects of tree mortality caused by insect pests on soil microbial communities and their response mechanisms, which is of great theoretical value for the establishment of scientific and effective methods for woodland restoration.

Effects of pesticides on soil bacterial, fungal and protist communities, soil functions and grape quality in vineyards

Abstract Pesticides can have unintentional effects on non‐target organisms and change biotic communities. Such changes might be particularly important in soil microbial communities, which drive many ecosystem functions and may affect crop quality. Here, we investigated, in a three‐year study, how vegetation control (by herbicide application) and soil copper content (from long‐term copper‐based fungicide application) affect biodiversity and the community structure of soil bacteria, fungi and protists, and associated soil functions (respiration, decomposition) in Swiss vineyards. Furthermore, we determined the effects of these two management practices on grape quality as the most direct ecosystem service to farmers. Across all study years, the community composition of microorganisms was affected by herbicide application, however, a significant loss of operational taxonomic units (OTUs) was only observed in fungi and protists. Soil copper content reduced OTU richness of bacteria and protists in some years but had no significant effect on fungal richness. Copper changed the community composition in all three groups of soil microorganisms. While we found no effect of copper on soil functions, herbicide application reduced microbial respiration and biomass by about 39% and 45%, respectively. However, decomposition rates remained virtually unchanged by any pesticide. Yeast assimilable nitrogen (YAN) levels in grape were below the critical threshold of 140 mg/L in 40% of the vineyards without herbicide application and the variety Chasselas, whereas in vineyards with herbicide application, it was only 20%. Application of pesticides led to changes in richness and composition of soil microbial communities and directly reduced some soil functions (microbial biomass and respiration) but not all (decomposition). Some grape quality parameters can be indirectly enhanced by pesticide application, highlighting the trade‐off between the interests of nature conservation and the interests of the farmer. Balancing these two diverging interests requires the establishment of alternative vineyard management allowing reduced pesticide application.

Combined metagenomics and metabolomic analysis of microbial community structure and metabolic function in continuous soybean cropping soils of Songnen Plain, China

Continuous cropping has a negative effect on soybean yield. In this study, a positioning experiment was conducted starting in 2015, with three treatments: maize–soybean rotation (SMR), 2-year maize, 2-year soybean rotation cropping (SC2), and 8-year soybean continuous cropping (SC8). We determined soybean yields (2015–2022) and analyzed soil microbial communities, functions, and metabolites composition in the 0–20 cm tillage layer using metagenomics technology and GC–MS technology during soybean flowering in 2022. Results indicated that continuous cropping (SC8) significantly reduced soybean yield compared to crop rotation (SMR) during the experimental period, while SC8 showed higher yield than SC2 in 2022. Compared to SMR, SC8 significantly increased soil N content and significantly decreased pH and TP, AP, and AK content. However, the pH and AK contents of SC8 were significantly higher than those of SC2. LeFSe analysis showed that Friedmanniella, Microlunatus, Nitrososphaera, Rubrobacter, Geodermatophilus, Nitriliruptor were enriched in SC8. Gaiella, Sphaerobacter, Methyloceanibacter were enriched in SC2. Sphingomonas, Cryobacterium, Marmoricola, Haliangium, Arthrobacter, Ramlibacter, Rhizobacter, Pseudolabrys, Methylibium, Variovorax were enriched in SMR. And the relative abundance of Cryobacterium, Marmoricola, Haliangium, Arthrobacter, Ramlibacter, Rhizobacter, Methylibium, Variovorax was significantly positively correlated with yield, while the relative abundance of Gaiella and Sphaerobacter was significantly negatively correlated with yield. SC8 significantly increased the abundance of genes in nitrogen metabolism and significantly decreased the abundance of genes related to phosphorus and potassium metabolism compared with SMR. However, the abundance of genes in potassium metabolism was significantly higher in SC8 than in SC2. Metabolomic analysis showed that compared to SMR, SC8 decreased the abundance of carbohydrates, ketones, and lipid. However, the abundance of carbohydrates, ketones, and lipid was significantly higher in SC8 than in SC2. Mantel test showed that soil pH and AK significantly affected soil microbial community, function, and metabolite composition. Soil C:N impacts on soil biological health and consequences on weed control in soybean and corn systems

Abstract Nitrogen availability has an important influence on agricultural weed growth since many weeds in annual cropping systems are more competitive in high-nitrogen soils. A potential method to control nitrogen availability is through soil carbon amendments, which stimulate soil microbial growth and immobilize nitrogen. Additionally, carbon amendments may alter soil microbial community composition, increase soil biological functioning, and improve soil health. In a two-year field experiment in corn (Zea mays L.) and soybean [Glycine max (L.) Merr.], we implemented five amendment treatments to test their ability to alter weed and crop growth through soil nitrogen availability and soil biological functioning. The treatments included: an untreated control, an unamended weed-free control, rye hay adding 3,600 kg C·ha-1 and 3,400 kg C·ha-1 each year, sawdust adding 5,000 kg C·ha-1 and 4,400 kg C·ha-1 each year, and a rye hay and sawdust combined treatment adding 8,600 kg C·ha-1 and 7,700 kg C·ha-1 each year. Each treatment was replicated 5 times in corn and 6 times in soybean. Each season, we explored correlations between crop and weed biomass and weed community composition and nitrogen immobilization measured through soil respiration and nitrogen availability. We also explored changes to the soil microbial community composition and soil health as a secondary result of the carbon amendment treatments. Nitrogen availability was lowest in plots treated with the highest C:N amendment. Increasing carbon improved soil health metrics, but the microbial community composition was most affected by the rye hay treatment. Amendments with high C:N reduced weed growth in both soybean and corn plots but only selected for specific weed communities in soybeans, leading to improved soybean competitiveness against weeds. In corn, crop growth and weed community composition remained consistent across amendment treatments. Targeted nitrogen immobilization may improve leguminous crop competition in some weed communities as part of an integrated weed management program.

Effects of Vegetation Succession on Soil Microbial Communities on Karst Mountain Peaks

Soil microorganisms are vital components of the karst terrestrial ecosystem. However, their responses to the vegetation succession on karst mountain peaks remain unclear as to whether soil microbial diversity and community compositions change with vegetation succession. We investigated the diversity and community compositions of soil bacteria and fungi and associated environmental factors along a vegetation succession from moss crusts (MC) to moss crusts with sparse grasses (MCG) to sparse grasses (G) on karst mountain peaks. The results indicated that soil organic carbon and total nitrogen generally increased, and soil pH changed in the range of 8.19–8.44 and slightly declined with vegetation succession. Overall, there was an increase in microbial biomass along the vegetation succession, with the dominant phyla of bacteria, including Proteobacteria, Acidobacteria, Actinobacteria, Choroflexi, Gemmatiomnadates, Bacteroidetes, and Planctomycetes, and the dominated phyla of fungi, including Basidiomycota and Ascomycota. Notably, both the bacterial and fungal community compositions were different among the three successional stages. Spearman’s correction analysis showed that soil organic carbon and total nitrogen had stronger and more significant influences on the soil microbial community compositions compared to soil water content, pH, and C:N ratio. Overall, our results provide evidence for the changes and influencing factors of the microbial community with the succession vegetation on karst mountain peaks.

Inoculation with Stutzerimonas stutzeri strains decreases N₂O emissions from vegetable soil by altering microbial community composition and diversity.

Plant growth-promoting rhizobacteria (PGPR) have been applied to mitigate nitrous oxide (N₂O) emissions from agricultural soils, but the microbial ecological mechanisms underlying N₂O mitigation are poorly understood. That is why only limited PGPR strains can mitigate N₂O emissions from agricultural soils. Therefore, it is of substantial significance to reveal soil ecological mechanisms of PGPR strains to achieve efficient and reliable N₂O-mitigating effect after inoculation. Inoculation with Stutzerimonas stutzeri strains decreased N₂O emissions from two soils with contrasting textures probably by altering soil microbial community composition and gene abundance involved in nitrification and denitrification. Our findings provide detailed insight into soil ecological mechanisms of PGPR strains to mitigate N₂O emissions from vegetable agricultural soils.

Intercropping changed the soil microbial community composition but no significant effect on alpha diversity.

Enhancing the planning of the forest-agricultural composite model and increasing the efficiency with which forest land is utilized could benefit from a thorough understanding of the impacts of intercropping between forests and agriculture on soil physicochemical properties and microbial communities. Populus cathayana × candansis cv. Xinlin No.1 and Glycine max intercrop soils, along with their corresponding monocrops, were used in this study's llumina high-throughput sequencing analysis to determine the composition and diversity of soil bacterial and fungal communities. The findings indicated that intercropping considerably raised the soil's total phosphorus content and significantly lowered the soil's carbon nitrogen ratio when compared to poplar single cropping. Furthermore, the total carbon and nitrogen content of soil was increased and the soil pH was decreased. The sequencing results showed that intercropping had no significant effect on soil alpha diversity. Intercropping could increase the composition of fungal community and decrease the composition of bacterial community in poplar soil. At the phylum level, intercropping significantly increased the relative abundance of four dominant phyla, i.e., Patescibacteria, Proteobacteria, Patescibacteria and Deinococcus-Thermus. And the relative abundances of only two dominant phyla were significantly increased. It was found that soil total phosphorus and available phosphorus content had the strongest correlation with soil bacterial community diversity, and soil pH had the strongest correlation with soil fungal community diversity. The results of this study were similar to those of previous studies. This study can serve as a theoretical foundation for the development of a poplar and black bean-based forest-agricultural complex management system in the future.

Afforestation alters soil microbial community composition and reduces microbial network complexity in a karst region of Southwest China

AbstractSoil microbial community plays important roles in altering ecological processes and biogeochemical cycles in ecosystems. However, little is known about how afforestation influences the diversity, composition, and ecological network of soil microbial community in karst regions. In this study, soil samples were collected from one farmland (FL) and three afforestation lands including one bamboo forest (BA), one landscape tree planting forest (LAT), and one orange orchard (ORO) in a karst region of Southwest China. The bacterial and fungal communities were characterized using the high‐throughput sequencing approach, and soil properties including soil organic carbon, pH, soil water content, and soil total and available nutrient contents were measured under different land use treatments. Results showed that conversion from FL to BA and LAT significantly reduced the Shannon diversity of bacterial community. At the phylum level (top 10), genus level (top 30), and operational taxonomic units (OTUs) level, afforestation from FL resulted in significant changes in nine phyla, 24 genera, and 31.32% OTUs of bacterial community, and in three phyla, 13 genera, and 11.62% OTUs of fungal community. The number of nodes, number of negative edges, connectivity, average degree, and relative modularity of the microbial network under afforestation lands decreased by 9.33%–18.66%, 47.98%–72.75%, 0.45%–5.93%, 14.73%–22.29%, and 6.46%–23.50%, respectively, compared with FL. The soil organic carbon, total potassium and total phosphorus were identified as the key soil properties affecting the microbial community. Compared with LAT and ORO, the changes in bacterial and fungal communities under BA were more obvious because of the higher contents in soil organic carbon (13.48%) and total potassium (27.18%). In conclusion, afforestation significantly changed compositions and ecological network complexity of soil microbial community in karst regions, and conversion from FL to BA had stronger influences on the changes of soil microbial community than other afforestation lands in Southwest China. These findings provide the context necessary to evaluate the responses of soil microbial community to land use changes in karst regions.

Characteristics of soil nitrogen and phosphorus fractions and microbial traits with increasing stand age in two-layered Cunninghumia lanceolata + Phoebe bournei plantations.

Soil nitrogen and phosphorus are two key elements limiting tree growth in subtropical areas. Understanding the regulation of soil microorganisms on nitrogen and phosphorus nutrition is beneficial to reveal maintenance mechanism of soil fertility in plantations. We analyzed the characteristics of soil nitrogen and phosphorus fractions, soil microbial community composition and function, and their relationship across three stands of two-layered Cunninghumia lanceolata + Phoebe bournei with different ages (4, 7 and 11 a) and the pure C. lanceolata plantation. The results showed that the contents of most soil phosphorus fractions increased with increasing two-layered stand age. The increase in active phosphorus fractions with increasing stand age was dominated by the inorganic phosphorus (9.9%-159.0%), while the stable phosphorus was dominated by the organic phosphorus (7.1%-328.4%). The content of soil inorganic and organic nitrogen also increased with increasing two-layered stand age, with NH4+-N and acid hydrolyzed ammonium N contents showing the strongest enhancement, by 152.9% and 80.2%, respectively. With the increase of stand age, the composition and functional groups of bacterial and fungal communities were significantly different, and the relative abundance of some dominant microbial genera (such as Acidothermus, Saitozyma and Mortierella) increased. The relative abundance of phosphorus solubilization and mineralization function genes, nitrogen nitrification function and aerobic ammonia oxidation function genes tended to increase. The functional taxa of fungi explained 48.9% variation of different phosphorus fractions. The conversion of pure plantations to two-layered mixed plantation affected soil phosphorus fractions transformation via changing the functional groups of saprophytes (litter saprophytes and soil saprophytes). Changes in fungal community composition explained 45.0% variation of different nitrogen fractions. Some key genera (e.g., Saitozyma and Mortierella) play a key role in promoting soil nitrogen transformation and accumulation. Therefore, the conversion of pure C. lanceolata plantation to two-layered C. lanceolata + P. bournei plantation was conducive to improving soil nitrogen and phosphorus availability. Bacteria and fungi played important roles in the transformation process of soil nitrogen and phosphorus forms, with greater contribution of soil fungi.
