ASSESSMENT OF HIGH MOLECULAR WEIGHT PROTEINS IN SELECTED INDIGENOUS BREAD WHEAT VARIETIES OF PAKISTAN

Higher molecular weight gluten subunits (HMW-GS) articulate variation in allelic frequency, reflecting a strong correlation with bread quality. In order to determine this quality trait, 23 different Pakistani indigenous wheat varieties were screened that may be used as selection criteria in a breeding program. For detecting variations in HMW-GS, sodium dodecyl sulfate acrylamide gel electrophoresis was used. In all, 77 Glu-1 were identified including 25 at Glu-A1, 26 at Glu-B1 and 26 at Glu-D1. Alleles at Glu-1 were 2* with a frequency of 53.3% and N with 60% frequency at Glu-A1, 6+8 with 60%, 17+18 with 45 % at Glu-B1 and 5+10 with 60% and 72% at Glu-D1. The highest frequency of subunits 20 and 5+10 were found at Glu-B1-e and Glu-D1 (a), followed by a frequency of 2* and Null at Glu-A1 (b, c), which is then followed by 2+12 at GluD-1 (a). The lowest frequency of 13+16, 17+18 and 6+8 were observed at Glu-B1 (f, i, d), respectively. Glu-A1, Glu-B1, and Glu-D1 alleles showed genetic index (i.e. 0.587, 0.54), (0.587. 0.065 and 0.48, 0.39, respectively) with an average of 0.55 and 0.37. The glu-D1 locus has a lower genetic index comparatively. The composition N, 20, 5+10 was found in maximum number (4) in Fakhr-e-Sarhad, Pasban, Pirsabak-2004 and Tatara genotypes, followed by the composition of 2*, 20, 2+12 in one check (Marvi-2000) and two varieties i.e. Chakwal-97 and Bakhtawar-92. The results describing the allelic frequency and composition of bread wheat would benefit further breeding plans for wheat to select good quality parameters.

Potential of whey protein isolate-lactose conjugates in the in-vitro infant digestion

Protein base modification is a notable potential method to alter the molecular structure and physicochemical and functional properties of the protein, thus affecting protein digestibility. This study investigated the protein digestibility of whey protein isolate– lactose (WPI-Lac) conjugates using an in-vitro infant gastric digestion static model. WPI was conjugated with lactose by dry Maillard reaction under optimised conditions. The following conditions were studied, WPI-Lac heated at 40°C, water activity aw = 0.80 and incubation time of 0, 1, 3, 5 and 7 days. Based on the brown colour and the conjugation rates in ortho-phthaldehyde analysis, the incubation time of day 3 of conjugation promises the extent of conjugation and prevents the formation of advanced Maillard reaction products (MRPs). Functional properties of glycated protein were found to significantly higher (p<0.05) in antioxidant activity and solubility compared with native WPI. In addition, Fourier Transform Infrared (FTIR) analysis indicated that the WPI was modified by dry MR with lactose. WPI-Lac day 3 was evaluated using the in-vitro gastric infant digestion model which undergo simulated gastric infant condition at pH 3 with 19 µL of 0.625mg/mL of pepsin. Sodium dodecyl sulfate acrylamide gel electrophoresis (SDSPAGE) analysis of digesta revealed that MRPs increased susceptibility to be hydrolysed by pepsin. The digestion product affirms dry MR conjugation can potentially improve WPI digestibility. Herein, this study of WPI-Lac conjugates contributes to an understanding of how protein–disaccharide glycates affect in-vitro infant gastric digestion.

Structural and interfacial characterization of oil bodies extracted from Camellia oleifera under the neutral and alkaline condition

Camellia oleifera seed oil contains nearly 80% oleic acid, which accounts for health-benefits, and it is stored in the subcellular oil bodies (OBs) organelles. To utilize C. oleifera OBs as a dietary supplement of unsaturated fatty acid, the physical properties need to be investigated. The physical stabilities of extracted OBs against ionic strength (0.01–1M) and temperature (30–90 °C) were evaluated via Turbiscan analysis and confocal microscope. The results showed that OBs extracted at alkaline conditions (pH 9.0–11) exhibited uniform dispersion. The surface charge of the OBs decreased dramatically with increasing NaCl content, and OBs droplet aggregation occurred under a NaCl concentration of 0.1 mol/L. Moreover, high temperature (up to 90 °C) had a positive effect on OBs stability. Overall, OBs extracted under alkaline pH were more stable than those extracted under neutral pH. Differences in the interfacial proteins of OBs extracted at the neutral and alkaline pHs were studied via sodium dodecyl sulfate-acrylamide gel electrophoresis (SDS-PAGE) and liquid chromatography-tandem mass spectrometry (LC-MS/MS). Six types of oleosins, including Ole Ⅲ, the fragment of Ole Ⅱ, oleosin, Ole Ⅳ, Ole Ⅴ, and Ole Ⅰ, were found in the extracted membrane proteins. Oleosins and some coextracted endogenous proteins played a certain role in OBs stability.

Phenotypic and genotypic identification of lactic acid bacteria isolated from traditional pickles of the Çubuk region in Turkey.

A total of 152 lactic acid bacteria (LAB) were isolated from pickles produced in the Ankara-Çubuk region. These isolates were clustered into eight groups on the basis of their phenotypic characteristics including cell morphology, CO2 production from glucose, growth at 10 and 45 °C, growth in 6.5 % NaCl, and growth at pH 9.6. API 50 CH carbohydrate fermentation test, 16S ribosomal RNA (rRNA) sequence analysis, and sodium dodecyl sulfate-acrylamide gel electrophoresis (SDS-PAGE) whole-cell protein profile analysis were also performed for precise identification of the isolates at the species level. Molecular identification revealed that the most prevalent LAB species involved in pickle fermentation were Pediococcus ethanolidurans (46 isolates, 30.3 %), Lactobacillus brevis (37 isolates, 24.3 %), Lactobacillus plantarum (37 isolates, 24.3 %), and Lactobacillus buchneri (15 isolates, 9.9 %). Other LAB were found in minor frequencies such as Pediococcus parvulus (8 isolates, 5.3 %), Lactobacillus namurensis (6 isolates, 3.9 %), Lactobacillus diolivorans (1 isolate, 0.7 %), Lactobacillus parabrevis (1 isolate, 0.7 %), and Enterococcus casseliflavus (1 isolate, 0.7 %). When results of phenotypic and genotypic identification methods were compared, differences in the species distribution of LAB associated with pickles were defined between the API and the 16S rRNA sequencing. The API 50 CHL test coincided with the 16S rRNA results in 71 out of the 152 tested isolates, indicating that API gave unreliable identification results. A clear correlation could not be found between the results of whole-cell SDS profiles and 16S rRNA sequencing. Therefore, molecular characterization by 16S rRNA sequencing was considered to be the most reliable method for identifying isolates. The results presented in this work provide insight in to the LAB population associated with traditional Çubuk pickles and constitute a LAB strain resource for further studies involving the development of starter cultures.

In vitro HIV type 1 reverse transcriptase inhibitory activities of Thai medicinal plants and Canna indica L. rhizomes

Water and 80% ethanol extracts of 20 Thai medicinal plants used to treat AIDS were tested for their HIV type 1 reverse transcriptase inhibitory activity. The water extracts of Ipomoea carnea subsp. fistulosa (aerial parts), Vitex glabrata (branch), Vitex trifolia (aerial part), Vitex negundo (aerial part), Canna indica (rhizome), and Justicia gendarussa (aerial part) showed HIV-1 RT inhibition ratio (% IR) higher than 90% at a 200 μg/ml concentration. The water extract of Canna indica rhizomes (IC 50 22.56 μg/ml) was selected for further study, i.e. for its HIV-1 RT inhibition activity and the purification and characterization of the active proteins. Proteins in water extract were fractionated by ammonium sulfate precipitation and separated by sodium dodecyl sulfate acrylamide gel electrophoresis (SDS-PAGE), yielding two proteins, Cip31 (31 kDa) and Cip14 (14 kDa) with IC 50 of 17.41 and 19.25 μg/ml and isoelectric point (p I) of 3.5 and 6.35, respectively. Both proteins showed significant HIV-1 RT inhibition.

Prevention of artifactual protein oxidation generated during sodium dodecyl sulfate-gel electrophoresis.

Prevention of artifactual protein oxidation occurring during sodium dodecyl sulfate (SDS) acrylamide gel electrophoresis is critical for identifying physiological protein oxidation implicated in human diseases due to the routine use of gel electrophoresis to separate the multiple proteins in proteomic studies. To develop a methodology that completely prevents artifactual protein oxidation in SDS acrylamide gel electrophoresis, cytochrome c was electrophoresed on polyacrylamide gels and subjected to trypsin in-gel digestion followed by tryptic peptide analysis by mass spectrometry. It was found that degassing the acrylamide solution to remove molecular oxygen prior to gel polymerization is a crucial process to protect the electrophoresed protein from reactive oxygen species generated during electrophoresis. However, significant artifactual protein oxidation remains that can only be eliminated entirely, if proteins are electrophoresed on an SDS gel photopolymerized with flavin as the photoinitiator and thioglycolate included in the cathode buffer as a reactive oxygen species scavenger. Using this combination of methodologies, cytochrome c isolated from adult rat heart mitochondria was purified and digested followed by mass spectrometric analysis, demonstrating the requisite high resolution of the polyacrylamide gel and the entire elimination of artifactual oxidation.

Characterization of P40, a cytadhesin of Mycoplasma agalactiae.

An immunodominant protein, P40, of Mycoplasma agalactiae was analyzed genetically and functionally. The gene encoding P40 was cloned from type strain PG2, sequenced, submitted to point mutagenesis in order to convert mycoplasma-specific TGA(Trp) codon to the universal TGG(Trp) codon, and subsequently expressed in Escherichia coli. Nucleotide sequence-derived amino acid sequence comparisons revealed a similarity of P40 to the adhesin P50 of Mycoplasma hominis and to protein P89 of Spiroplasma citri, which is expected to be involved in adhesion. The amino acid sequence of P40 revealed a recognition site for a signal peptidase and strong antigenic and hydrophilic motifs in the C-terminal domain. Triton X-114 phase partitioning confirmed that P40 is a membrane protein. Fab fragments of antibodies directed against recombinant purified P40 significantly inhibited adherence of M. agalactiae strains PG2 to lamb joint synovial cells LSM 192. Sera taken sequentially from sheep infected with PG2 revealed that P40 induced a strong and persistent immune response that gave strong signals on immunoblots containing recombinant P40 even 3 months after infection. The gene encoding P40 was present in a single copy in all of the 26 field strains of M. agalactiae analyzed and was not detected in closely related mycoplasma species. P40 was expressed as a protein with an apparent molecular mass of 37 kDa on sodium dodecyl sulfate-acrylamide gels by all M. agalactiae strains except for serotype C strains, which showed nonsense mutations in their p40 genes.

Isolation and characterization of a new rat P450 (CYP3A18??) cDNA encoding P4506??-2 catalyzing testosterone 6??- and 16?? -???hydroxylations

A cytochrome P450 cDNA, encoding a new form of CYP3A protein, was isolated from a liver cDNA library of a male rat using anti-P450(6)beta-1 and anti-P450(6)beta-2 antibodies and the CYP3A2 cDNA. The cDNA (CYP3A18 cDNA) consisted of 1987 nucleotides, in which were contained an open reading frame of 1491 bp (corresponding to 497 amino acids), 5'-(59 bp) and 3'-noncoding regions (437 bp). The deduced amino acid sequence of CYP3A18 cDNA was completely identical in the first 27 N-terminal residues of P450(6)beta-2 previously isolated by us (Nagata et al. J Biochem 1990: 107, 718-725) from livers of rats treated with dexamethasone, and also shared higher extents of similarity with hamster CYP3A10 (78.5%) than with rat CYP3As previously sequenced (66.3-69.3%). Northern blot analyses indicated a male-dominant expression of this new CYP3A mRNA and enhanced expression in dexamethasone-or pregnenolone-16 alpha-carbonitrile (PCN)-treated, but not phenobarbital-or 3-methylcholanthrene-treated rats. Expressed CYP3A18 protein in COS-1 cells migrated at a position identical to that of purified P450(6)beta-2 on sodium dodecyl sulfate-acrylamide gel electrophoresis and catalyzed 16 beta- and 6 alpha-hydroxylations of testosterone. In contrast to CYP3A1 and CYP3A2, cytochrome b5 was not essential for maximal catalytic activities of recombinant CYP3A18 protein. These results, together with ontogenic profiles of CYP3A18 mRNA and P450(6)beta-2 protein, indicate that the newly isolated CYP3A18 cDNA encodes P450(6)beta-2 in rat liver.

A novel hepatocytic transcription factor that binds the alpha-fetoprotein promoter-linked coupling element.

We recently characterized a promoter-linked coupling element (PCE) in the rat alpha-fetoprotein (AFP) gene required for strong transcriptional stimulation by distant enhancers (P. Wen, N. Crawford, and J. Locker, Nucleic Acids Res. 21:1911-1918, 1993). In this study, oligonucleotide gel retardation and competition experiments defined the PCE as a 12-bp binding site, TGTCCTTGAACA, an imperfect inverted repeat from -166 to -155 near the AFP promoter. A factor that bound this site (PCF) was abundant in HepG2 nuclear extracts and detectable in extracts from several other AFP-producing hepatocarcinoma cell lines and fetal liver. Hepatocytic cell lines that did not express AFP, nonhepatocytic cell lines, adult liver, and fetal brain did not show the factor. Experiments excluded the possibility that PCF activity was due to binding of glucocorticoid receptor or an AP1-like factor that bound overlapping sites. Competition experiments with several mutant oligonucleotides determined that the optimum PCF binding site was TGTCCTTGAAC(A/T). Mutations decreased binding or totally abolished binding activity. In expression plasmids, PCE mutations strongly reduced gene expression. UV cross-linking to a PCE probe identified peptide bands near 34 kDa. PCF was purified by heparin-Sepharose chromatography followed by affinity binding to oligomerized PCE DNA. The product resolved as a complex of three peptides (PCF alpha 1, PCF alpha 2, and PCF beta, 32 to 34 kDa) on sodium dodecyl sulfate-acrylamide gels. The peptide sizes and gel patterns are unlike those of any of the well-described hepatic transcription factors, and the binding site has not been previously reported. PCF thus appears to be a novel transcription factor.

Glucosyltransferase Mutants of Leuconostoc mesenteroides NRRL B-1355

Leuconostoc mesenteroides NRRL B-1355 produces dextrans and alternan from sucrose. Alternan is an unusual dextran-like polymer containing alternating alpha(1-->6)/alpha(1-->3) glucosidic bonds. Cultures were mutagenized with UV and ethyl methanesulfonate, and colony morphology mutants were selected on 10% sucrose plates. Colony morphology variants exhibited changes from parent cultures in the production of one or more glucosyltransferases (GTFs) and glucans. Mutants were characterized by measuring resistance of glucan products to dextranase digestion, by electrophoresis, and by high-pressure liquid chromatography of maltose acceptor products generated from sucrose-maltose mixtures. Some mutants produced almost pure fraction L dextran, and cultures exhibited a single principal GTF band on sodium dodecyl sulfate-acrylamide gels. Other mutants produced glucans enriched for alternan. Colony morphology characteristics (size, smoothness, and opacity) and liquid culture properties (clumpiness, color, and viscosity in 10% sucrose medium) were explained on the basis of GTF production. Three principal GTF bands were detected.

Degradation of enamelins by proteinases found in porcine secretory enamel in vitro

The action of proteinases obtained from porcine secretory enamel on the porcine 89-kDa enamelin was examined in vitro. The results of sodium dodecyl sulphate-acrylamide gel electrophoresis of the reaction products indicated the following. (1) The 76- and 78-kDa proteinases localized in the outermost layer of the secretory enamel not only convert the 25-kDa amelogenin to the 20-kDa amelogenin, but also split the 89-kDa enamelin, which is the major enamelin component in the enamel in the very early secretory stage, into large fragments such as 25-, 41- and 56-kDa enamelins. (2) The serine proteinases localized in the inner layer of secretory enamel further degrade not only amelogenins but also enamelins.

Changes in myosin heavy chain isoform expression of overloaded rat skeletal muscles

1. 1. The effect of functional overload produced by tenotomy of synergistic gastrocnemius muscle on the expression of myosin heavy chain (MHC) isoforms in the plantaris and soleus muscles of the rat was studied using gradient sodium dodecyl sulfate-acrylamide gel electrophoresis. 2. 2. Five weeks tenotomy, the plantaris and soleus muscle weights induced by tenotomy of the gastrocnemius muscle were 44.3% ( P < 0.005) and 37.4% ( P < 0.005), respectively, heavier than the contralateral control muscles. 3. 3. Although four types of MHC isoforms were observed in both control and experimental plantaris, the percentage of MHC isoforms in the control and experimental muscles differed; the hypertrophied plantaris muscle contained more HCI ( P < 0.05), HCIIa and HCIId ( P < 0.05) and less HCIIb ( P < 0.05) than the control muscle. 4. 4. The control soleus muscle contained two MHC isofonns, HCI and HCIIa. However, there was only a single HCI isoform in the hypertrophied soleus muscle. 5. 5. These results indicate that overloading a skeletal muscle by removing its synergists produces not only the muscle hypertrophy but also the changes in the expression of MHC isofonns.

Leishmania amazonensis: Specific labeling of amastigote cysteine proteinases by radioiodinated N-benzyloxycarbonyl-tyrosyl-alanyl diazomethane

Living Leishmania amazonensis amastigotes were incubated with radioiodinated N-benzyloxycarbonyl- l-tyrosyl- l-alanyl diazomethane (Z-Tyr-AlaCHN 2), an irreversible inhibitor of mammalian cathepsins B and L. Parasite lysates were subjected to electrophoresis in gelatin-containing sodium dodecyl sulfate-acrylamide gels to detect regions of proteolytic activity, and the distribution of the inhibitor was ascertained by autoradiography. Of the three main bands of proteolysis associated with cysteine proteinases, two, with apparent molecular weights of 28 and 31 kDa, were shown to be labeled. The third enzyme activity, detected at the 35-kDa region in substrate gels, was only faintly labeled. The distribution of labeled bands was similar when lysates of untreated parasites were electrophoresed and the gels incubated with the radioiodinated inhibitor. Under reducing conditions, the inhibitor bound to polypeptides of 29, 31, 32, and 34 kDa, of which the first and the last were the most intensely labeled. Polypeptides with the same apparent molecular weights were labeled when amastigote lysates were incubated with the 125I inhibitor. Uptake of radioactivity by the parasites was time and concentration-dependent and more than 80% of the total counts could be precipitated with trichloroacetic acid. Radioactivity associated with the amastigotes was quite stable after they were pulsed with labeled inhibitor and chased for up to 24 hr in inhibitor-free medium. Both total uptake and labeling of cysteine proteinases were markedly reduced in parasites preincubated with Z-Phe-AlaCHN 2 prior to exposure to Z-Tyr( 125I)-AlaCHN 2. However, more radioiodinated inhibitor was taken up by parasites preincubated with cold inhibitor and chased in inhibitor-free medium, suggesting de novo synthesis or processing of inactive enzyme precursors.

Compositional changes and apoprotein A‐I metabolism of plasma high density lipoprotein in estrogenized chicks

The effect of estrogen on compositional changes, apolipoprotein (apo) A-I metabolism and the morphology of plasma high density lipoprotein (HDL) were investigated in chicks. The administration of 17 beta-estradiol (25 mg/kg body weight) to growing male chicks (8-week-old) markedly reduced the concentrations of plasma HDL components, except for triglyceride (TG). At the same time, levels of TG, total cholesterol (TC) and phospholipid (PL) in plasma were greatly elevated. The respective values for TG, TC, PL and protein in HDL were 13.9, 89.3, 154.1 and 231.7 (mg/dL) in the control, and 39.0, 35.1, 113.8 and 160.0 (mg/dL) in chicks upon estrogen treatment for one day. In vivo kinetic studies showed that the fractional catabolic rate of HDL apo A-I was significantly higher (p less than 0.05) in estrogen-treated chicks than in control birds, indicating an increased efficiency of HDL removal in the former. The production rate of HDL apo A-I also was significantly lower (p less than 0.05) in estrogen-treated chicks. Sodium dodecyl sulfate-acrylamide gel electrophoresis followed by laser scanning densitometry of HDL apolipoproteins in estrogen-treated chicks revealed a reduction of apo A-I and the occurrence of new apolipoproteins which had been absent in HDL of untreated birds. The HDL particles showed that the mean particle size of HDL became larger upon estrogen treatment. Particles with diameters between 70 and 123 A were predominant in HDL of control chicks, while particles with diameters between 97 and 143 A were most abundant in HDL of estrogen-treated chicks.

Cut-1 a Caenorhabditis elegans gene coding for a dauer-specific noncollagenous component of the cuticle

We have molecularly identified a new gene of Caenorhabditis elegans that codes for a component of the cuticle. The gene has been physically mapped on LGII near the locus sqt-1. The structure and the sequence of the gene have been determined and antisera have been raised against parts of the protein produced as fusions in Escherichia coli. By transcription analysis, and by the use of specific antisera, we have determined that this gene is expressed specifically during dauer larva formation. In extracts of worms completing the dauer transformation the product of this gene migrates in sodium dodecyl sulfate acrylamide gels with an apparent molecular mass of 40 kDa. By immunofluorescence we have determined that it is a component of the cuticles of dauer larvae. It forms a ribbon ∼ 2 μm wide running along the lateral lines underneath the alae. Once it is assembled in the cuticle the protein becomes insoluble even in the presence of strong detergents and reducing agents in a manner that is similar to that described for the noncollagenous, insoluble residue of nematode cuticles called cuticlins; therefore, we have named the gene cut-1 for cuticlin 1. cut-1 represents the first gene for a noncollagenous component of C. elegans cuticle that has been characterized molecularly.

Human Neutrophils Express the αl-Antitrypsin Gene and Produce αl-Antitrypsin

The potent serine protease, neutrophil elastase (NE), is stored in neutrophil azurophilic granules, where it is available to degrade phagocytosed material and can be released by the cell to assist in tissue migration and help clear tissue debris. While neutrophils carry NE, they cannot produce it; the NE gene is expressed only in bone marrow granulocyte precursor cells. Protection of normal tissues from the destructive capacity of NE is provided by alpha 1-antitrypsin (alpha 1 AT), a 52-Kd serine antiprotease produced by hepatocytes and mononuclear phagocytes. In the context of the broad destructive capacity of NE, we evaluated the concept that human neutrophils may be able to modulate the extracellular activity of NE by synthesizing and secreting alpha 1AT. Immunocytochemical analysis demonstrated that the neutrophil contains alpha 1AT. Northern analysis and in situ hybridization with alpha 1AT-specific probes demonstrated the presence of alpha 1AT messenger RNA transcripts within neutrophils. [35S]methionine-labeling of neutrophils followed by immunoprecipitation of the supernatant with an anti-alpha 1AT antibody and sodium dodecyl sulfate-acrylamide gel analysis demonstrated that neutrophils can synthesize alpha 1AT de novo and secrete the synthesized molecule. In the presence of major neutrophil degranulation, the antiprotease effect of neutrophil alpha 1AT is overwhelmed, allowing the NE to act unopposed in the extracellular microenvironment. However, in conditions where small amounts of NE are released by neutrophils, at least some of the secreted newly synthesized alpha 1AT was capable of complexing with NE. Thus, despite the fact that the neutrophil cannot synthesize NE, it can synthesize and secrete alpha 1AT, the inhibitor of NE, ie, the neutrophil is capable, to some extent, of modulating NE activity in the local milieu without the help of antiproteases produced by other cells.

Human neutrophils express the alpha 1-antitrypsin gene and produce alpha 1-antitrypsin

The potent serine protease, neutrophil elastase (NE), is stored in neutrophil azurophilic granules, where it is available to degrade phagocytosed material and can be released by the cell to assist in tissue migration and help clear tissue debris. While neutrophils carry NE, they cannot produce it; the NE gene is expressed only in bone marrow granulocyte precursor cells. Protection of normal tissues from the destructive capacity of NE is provided by alpha 1-antitrypsin (alpha 1 AT), a 52-Kd serine antiprotease produced by hepatocytes and mononuclear phagocytes. In the context of the broad destructive capacity of NE, we evaluated the concept that human neutrophils may be able to modulate the extracellular activity of NE by synthesizing and secreting alpha 1AT. Immunocytochemical analysis demonstrated that the neutrophil contains alpha 1AT. UHF-1, a factor required for maximal transcription of early and late sea urchin histone H4 genes: analysis of promoter-binding sites.

A protein, denoted UHF-1, was found to bind upstream of the transcriptional start site of both the early and late H4 (EH4 and LH4) histone genes of the sea urchin Strongylocentrotus purpuratus. A nuclear extract from hatching blastulae contained proteins that bind to EH4 and LH4 promoter fragments in a band shift assay and produced sharp DNase I footprints upstream of the EH4 gene (from -133 to -106) and the LH4 gene (from -94 to -66). DNase I footprinting performed in the presence of EH4 and LH4 promoter competitor DNAs indicated that UHF-1 binds more strongly to the EH4 site. A sequence match of 11 of 13 nucleotides was found within the two footprinted regions: [sequence: see text]. Methylation interference and footprinting experiments showed that UHF-1 bound to the two sites somewhat differently. DNA-protein UV cross-linking studies indicated that UHF-1 has an electrophoretic mobility on sodium dodecyl sulfate-acrylamide gels of approximately 85 kDa and suggested that additional proteins, specific to each promoter, bind to each site. In vitro and in vivo assays were used to demonstrate that the UHF-1-binding site is essential for maximal transcription of the H4 genes. Deletion of the EH4 footprinted region resulted in a 3-fold decrease in transcription in a nuclear extract and a 2.6-fold decrease in expression in morulae from templates that had been injected into eggs. In the latter case, deletion of the binding site did not grossly disrupt the temporal program of expression from the injected EH4 genes. LH4 templates containing a 10-bp deletion in the consensus region or base substitutions in the footprinted region were transcribed at 14 to 58% of the level of the wild-type LH4 template. UHF-1 is therefore essential for maximal expression of the early and late H4 genes.
