Derivation of site-specific environmental quality guideline values for fuel-contaminated soils on sub-Antarctic Macquarie Island.

Accidental fuel spills associated with the storage, transfer, and use of diesel fuel for power generation have occurred on sub-Antarctic Macquarie Island since the establishment of the island's research station in 1948. An extensive in situ remediation program was implemented by the Australian government from 2009 to 2016 that used nutrient addition and air sparging to enhance the microbial degradation of petroleum products. During this period, a range of ecotoxicological assessments were conducted to better understand the impacts of fuel in soils on native biota and their sensitivity. This study compiles this ecotoxicological data into a species sensitivity distribution (SSD) to establish environmental quality guideline values (EQGVs) for fuels in soils on Macquarie Island. The SSD model includes 13 critical effect concentrations (CECs) selected using an expert judgment approach. These include data from functional and community-based tests as well as traditional single-species toxicity tests using microbes, plants, and invertebrates and representing the range of carbon content (~3%-48%) and fuel composition at various stages of degradation (from fresh to 18 months aged) in soils as occurs at contaminated sites on the island. A protective concentration (PC80) of 97 mg/kg TPH C9-C40 (95% CI 24-283) was derived for special Antarctic blend diesel from the SSD and is recommended as an appropriate site-specific EQGV and potential remediation target for the immediate station area in the vicinity of infrastructure. More conservative PC values are also provided for areas with higher conservation values outside the station footprint. These EQGVs are the first to be produced for fuels in the sub-Antarctic and Antarctic regions. They will be used to inform ongoing environmental management on Macquarie Island and are likely suitable and recommended for use more broadly across the sub-Antarctic. Short-term insecticide exposure amid co-occurring stressors reduces diversity and densities in north-east Indian experimental aquatic invertebrate communities

Globally, river pesticide concentrations are associated with regional and local stream invertebrate diversity declines. Pesticides often co-occur with elevated nutrients (e.g. nitrogen and phosphorus) and sediments related to agriculture, making their individual effects difficult to disentangle. These effects are also less well studied in Asia, than in other geographic regions. Within Asia, India is one of the largest producers and users of pesticides and has approximately 60% of total land mass used for agriculture. Here we examine the responses of Indian river invertebrate communities subjected to malathion, nutrients, and sediment additions in a semi-orthogonal design, in three sequential (through time) short-term (120 h) mesocosm experiments. Additionally, a series of single-species toxicity tests were run that used 24 h exposure and 72 h recovery to examine the sensitivity of 13 local invertebrate taxa to malathion, and 9 taxa to cypermethrin, comparing these results to those from other biogeographic regions. Mesocosm results indicate that malathion exposure had a major effect compared to other stressors on communities, with a lesser effect of nutrients and/or sediments. In mesocosms, taxa richness, total abundance and the abundance of sensitive species all declined associated with malathion concentrations. Comparisons of organism sensitivities from other geographic locations and those in the current paper suggest taxa in India are relatively tolerant to malathion and cypermethrin. Our results further reinforce that the high observed aquatic pesticide concentrations known to occur in Asian freshwater ecosystems are likely to be negatively affecting biodiversity, homogenising biota towards those most stress tolerant.

Introducing the No-Significant-Effect Concentration.

The no-effect concentration (NEC) is the preferred threshold metric for single-species toxicity tests applied to derive safe concentration thresholds for contaminants in the environment for use in species sensitivity distributions. However, the NEC is only suitable when concentration-response (C-R) data exhibit a threshold response. We describe an alternative toxicity estimate, the no-significant-effect concentration (NSEC), which is better suited to C-R data for which the response is a monotonically decreasing function of concentration and no threshold effects are evident. We use a flexible, three-parameter sigmoidal function to describe the C-R relationship and detail both Bayesian and frequentist approaches to estimation and inference for the NSEC. While the NSEC is conceptually linked to the traditional no-observed-effect concentration (NOEC), it is a substantial improvement over the NOEC because it decouples the estimate from being directly dependent on the placement of treatment concentrations as well as admitting statements of precision of the resulting toxicity estimate. Mixture Toxicity of Three Unconventional Gas Fracking Chemicals, Barium, O-Cresol, and Sodium Chloride, to the Freshwater Shrimp Paratya australiensis.

The 96-h acute toxicity of barium (Ba2+ ), o-cresol, and sodium chloride (NaCl) to Paratya australiensis was assessed in single, binary, and ternary combinations in addition to three biochemical assays: glutathione S-transferase, acetylcholinesterase, and sodium-potassium adenosine triphosphatase. The 96-h lethal concentrations that expressed 50% mortality (LC50) in the single-toxicant exposures were Ba2+ = 23.4 mg/L, o-cresol = 12.2 mg/L, and NaCl = 4198 mg/L. Mortality from o-cresol exposure occurred between 11 and 22 mg/L, whereas Ba2+ was more gradual across 10-105 mg/L, and most of the NaCl mortality occurred between 2050 and 4100 mg/L. Toxic units were used to assess the binary and ternary interactions of the toxicants. A more than additive effect was observed for most combinations in the binary chemical exposures, with the ternary combinations yielding highly synergistic interactions. Greater synergism was observed with the 96-h LC50 of o-cresol in combination with the three concentrations of NaCl (1025, 2050, and 3075 mg/L) compared with Ba2+ , with toxic units of 0.38, 0.48, and 0.10 (o-cresol) and 0.71, 0.67, and 0.50 (Ba2+ ). No notable enzyme activity trends were observed in the enzyme biomarker responses from both individual and mixture exposures. Although acute single-species toxicity tests tend to underestimate the effects of Ba2+ , o-cresol, and NaCl on populations, communities, and ecosystems in seminatural (e.g., mesocosms) and natural systems, there are currently no published acute toxicity data available for P. australiensis and the three toxicants used in the present study. Using Single-Species and Whole Community Stream Mesocosm Exposures for Identifying Major Ion Effects in Doses Mimicking Resource Extraction Wastewaters.

Wastewaters and leachates from various inland resource extraction activities contain high ionic concentrations and differ in ionic composition, which complicates the understanding and effective management of their relative risks to stream ecosystems. To this end, we conducted a stream mesocosm dose-response experiment using two dosing recipes prepared from industrial salts. One recipe was designed to generally reflect the major ion composition of deep well brines (DWB) produced from gas wells (primarily Na+, Ca2+, and Cl-) and the other, the major ion composition of mountaintop mining (MTM) leachates from coal extraction operations (using salts dissociating to Ca2+, Mg2+, Na+, SO42- and HCO3-)-both sources being extensive in the Central Appalachians of the USA. The recipes were dosed at environmentally relevant nominal concentrations of total dissolved solids (TDS) spanning 100 to 2000 mg/L for 43 d under continuous flow-through conditions. The colonizing native algal periphyton and benthic invertebrates comprising the mesocosm ecology were assessed with response sensitivity distributions (RSDs) and hazard concentrations (HCs) at the taxa, community (as assemblages), and system (as primary and secondary production) levels. Single-species toxicity tests were run with the same recipes. Dosing the MTM recipe resulted in a significant loss of secondary production and invertebrate taxa assemblages that diverged from the control at all concentrations tested. Comparatively, intermediate doses of the DWB recipe had little consequence or increased secondary production (for emergence only) and had assemblages less different from the control. Only the highest dose of the DWB recipe had a negative impact on certain ecologies. The MTM recipe appeared more toxic, but overall, for both types of resource extraction wastewaters, the mesocosm responses suggested significant changes in stream ecology would not be expected for specific conductivity below 300 µS/cm, a published aquatic life benchmark suggested for the region.

Species Sensitivity Distributions of Benthic Macroinvertebrates in Fludioxonil-Spiked Sediment Toxicity Tests.

The fungicide fludioxonil, given its physicochemical properties, potentially accumulates and persists in sediments. Fludioxonil has a widespread agricultural use to control various fungal diseases. Its residues may cause toxic effects to benthic aquatic fauna, thereby impacting ecosystem service functions of aquatic ecosystems. To assess the potential environmental effects of fludioxonil in the sediment compartment of edge-of-field surface waters, sediment-spiked single-species toxicity tests with benthic macroinvertebrates were performed. In all experiments artificial sediment was used with an organic carbon content of 2.43% on dry weight basis. The single-species tests were conducted with 8 benthic macroinvertebrates covering different taxonomic groups typical for the Yangtze River Delta, China. The 28d-EC10 and 28-LC10 values thus obtained were used to construct species sensitivity distributions (SSDs). In addition, our dataset was supplemented with 28d-EC10 and 28-LC10 values for 8 different benthic invertebrates from a study in the Netherlands that used field-collected sediment. Based on SSDs constructed with 28d-EC10 or 28d-LC10 values hazardous concentrations to 5% of the species tested (HC5's) were obtained. The HC5 values based on the toxicity tests from China were lower than those from the Netherlands, although 95% confidence bands overlapped. The HC5 values derived from the separate datasets from China and the Netherlands, as well as from the combined dataset, were compared to the Tier-3 Regulatory Acceptable Concentrations (RAC) for fludioxonil and the benthic invertebrate community derived from a sediment-spiked outdoor microcosm experiment conducted in the Netherlands. The HC5 values obtained appeared to be lower than this Tier-3 RAC when expressed in terms of total sediment concentration, but not always when expressed in terms of pore water concentrations.

Effects of Zinc in an Outdoor Freshwater Microcosm System.

A long-term exposure outdoor microcosm study was conducted to evaluate the effects of zinc (Zn) on zooplankton, phytoplankton, and periphyton in a freshwater system. Five Zn treatment concentrations (nominal: 8, 20, 40, 80, and 160 μg/L Zn) and an untreated control with 3 replicates each were used. Various physical and chemical characteristics of the microcosms and biological assessment endpoints (e.g., total abundance, group abundance, species richness, chlorophyll a, etc.) were measured to determine the effects of Zn over time. In general, physical and chemical characteristics (e.g., total dissolved solids, total suspended solids, dissolved oxygen, pH, dissolved organic carbon) of water fluctuated over time, but they were not significantly different within treatments and controls during the study. Zinc significantly affected the population dynamics and community structure of plankton. The effects occurred 7 d after initial treatment exposures began and continued to the end of the treatment phase, especially at the high treatment concentrations. Total and group abundance, species richness, the Shannon index, and chlorophyll a concentrations for high Zn treatment concentrations were significantly lower than the controls during the treatment phase. The no-observed-effect, lowest-observed-effect, and median effect concentrations were generally lower than the literature-reported results from single-species toxicity tests for fish and invertebrates, suggesting that plankton are more sensitive to Zn than planktivores. Although primary producers play an important role in the ecosystem, they have not been consistently incorporated into numerical environmental quality criteria for freshwater organisms, at least in the United States. The results of the present study are useful for development of environmental quality guidelines for freshwater ecosystems and ecological risk assessment. On the balance between practical relevance and standardization - Testing the effects of zinc and pyrene on native nematode communities in soil microcosms

Soils are among the most densely inhabited and biodiverse habitats on our planet, and many important soil ecosystem services depend on the health condition of the native soil fauna. Anthropogenic stress such as chemical pollution acting on the native soil fauna might jeopardize these functions. Laboratory microcosm tests are an appropriate tool for assessing the risk of chemicals on the native soil fauna and can be regarded as intermediate tier tests, bridging the gap between single species toxicity tests and field testing. Nematodes are one of the most abundant and divers soil invertebrates, and as such native nematode communities might be suitable for ecotoxicological assessments in laboratory microcosm set ups. In order to test such a small-scale (30 g soil) microcosm system, two different chemicals (zinc and pyrene) were assessed in various soil types for their effects on the respective native nematode communities. Various community parameters such as total nematode density, genus richness and genus composition, as well as trait-related indices (e.g. maturity index) were monitored over a period of 8–10 weeks. The response of the nematode communities strongly varied between soil types, and these differences were more pronounced for Zn than for pyrene. Interestingly, the structure of the respective native nematode communities was shown to play a larger role for explaining the varying toxic effects than soil properties governing the bioavailability of the spiked chemicals. We demonstrated that exposure of natural nematode communities in their original soil matrix to the metal zinc and to pyrene under climatically highly controlled conditions resulted in quantitatively and qualitatively distinct responses. Upon comparison of various community indices, the maturity index was shown to be the most sensitive toxicity endpoint for all tested soils and chemicals.

Metal-doping of nanoplastics enables accurate assessment of uptake and effects on Gammarus pulex.

Because of the difficulty of measuring nanoplastics (NP), the use of NPs doped with trace metals has been proposed as a promising approach to detect NP in environmental media and biota. In the present study, the freshwater amphipod Gammarus pulex were exposed to palladium (Pd)-doped NP via natural sediment at six spiking concentrations (0, 0.3, 1, 3, 10 and 30 g plastic per kg of sediment dry weight) with the aim of assessing their uptake and chronic effects using 28 days standardized single species toxicity tests. NP concentrations were quantified based on Pd concentrations measured by ICP-MS on digests of the exposed organisms and faecal pellets excreted during a post-exposure 24 hour depuration period. Additionally, NP concentrations were measured in sediments and water to demonstrate accuracy of NP dosing and to quantify the resuspension of NP from the sediment caused by the organisms. A significant positive linear relationship between the uptake of NP by G. pulex and the concentration of NP in the sediments was observed, yet no statistically significant effects were found on the survival or growth of G. pulex. A biodynamic model fitted well to the data and suggested bioaccumulation would occur in two kinetic compartments, the major one being reversible with rapid depuration to clean medium. Model fitting yielded a mass based trophic transfer factor (TTF), conceptually similar to the traditional biota sediment accumulation factor, for NP in the gut of 0.031. This value is close to a TTF value of 0.025 that was obtained for much larger microplastic particles in a similar experiment performed previously. Mechanistically, this suggests that ingestion of plastic is limited by the total volume of ingested particles. We demonstrated that using metal-doped plastics provides opportunities for precise quantification of NP accumulation and exposure in fate and effect studies, which can be a clear benefit for NP risk assessment.

Species-specific effects of long-term microplastic exposure on the population growth of nematodes, with a focus on microplastic ingestion

Microplastics (MPs; <5 mm) released into freshwaters undergo changes in their density that make them highly bioavailable to the fauna in the sediments. In fine sediments, nematodes account for up to 90% of the meiobenthic organisms and have an important position in benthic food webs, by connecting lower (bacteria) and higher (e.g., macrofauna, fish) trophic levels. Due to their high ecological relevance, ubiquitous occurrence and very high individual densities, nematodes can serve as bioindicators of environmental pollution and especially MP pollution, since the ingested microscopic particles can be easily detected in the transparent bodies of these organisms. Single-species toxicity tests with Caenorhabditis elegans have revealed dose-dependent inhibitory effects on reproduction in nematodes exposed for 96 h to relatively high concentrations of polystyrene (PS) beads. Thus, in this study, we examined whether longer-term multigenerational tests of nematodes under continuous PS bead exposure are able to reveal more subtle impacts of MPs on population growth and whether the observed effects can be linked to the species-specific life history traits of the nematodes.We therefore tested three bacterial-feeding nematode species (C. elegans, Acrobeloides nanus, Plectus acuminatus) by exposing them to 1.0-µm PS beads at an exposure concentration of 107 beads ml−1. The experiment was conducted in semi-fluid medium for 21–49 days depending on the life-cycle characteristics of each nematode species. Ingested PS beads were quantified by fluorescence microscopy observations. Population growth rates, carrying capacities as well as doubling time and the time at which the maximum sustainable yield was reached were used to assess the effects of PS bead exposure on nematode population dynamics.All three nematode species readily ingested 1.0-µm PS beads in semi-fluid medium, but the number of ingested PS beads varied between species. PS bead exposure significantly decreased the carrying capacity of C. elegans, whereas A. nanus populations grew significantly faster in the presence of the beads. Long-term multigeneration tests revealed more subtle impacts of MP on C. elegans than occurred following short-term exposure scenarios. Our results show that MP-induced changes in nematodes population dynamics can alter nematode communities, which in term may impact the benthic food web.

Response of a nematode community to the fungicide fludioxonil in sediments of outdoor freshwater microcosms compared to a single species toxicity test

When entering aquatic ecosystems, hydrophobic organic chemicals like the fungicide fludioxonil partition to the sediment compartment where they pose potential risks to benthic invertebrates. To assess the ecological risk for sediment-dwelling invertebrates, nematodes are a suitable organism group, as they are abundantly present and possess key positions in the benthic food web. Therefore, the toxicity of the fungicide fludioxonil to nematodes was assessed in a standardized sediment toxicity test with Caenorhabditis elegans (ISO 10872), as well as in an outdoor sediment-spiked microcosm test system. In the microcosms, effects on the nematode species composition were studied, while exposure concentrations of fludioxonil were monitored in total sediment and pore water. Toxic effects on nematodes were better predicted using concentrations in pore water than total sediment concentrations. In laboratory single species tests, fludioxonil showed considerably lower toxicity in spiked field-collected sediment, compared to artificial ISO-sediments. Applying an assessment factor of 10 to the C. elegans 96-h EC10, a Tier-1 RACNematode of 7.99 mg kg−1 dry artificial sediment (corresponding to 69 μg l−1 in pore water) appeared to be protective for nematode communities in microcosms that showed no response in total abundance and species composition up to 39.9 mg fludioxonil kg−1 dry sediment (corresponding to 392 μg l−1 in pore water).

Comparing the effects of fludioxonil on non-target soil invertebrates using ecotoxicological methods from single-species bioassays to model ecosystems

The lower tier toxicity tests used for risk assessment of plant protection products are conducted with single species, only regarding direct effects of the tested substances. However, it is not clear, if lower tier tests are able to protect in situ soil communities, as these tests are not able to account for direct and indirect effects of chemicals on multi-species systems in natural soil communities. This knowledge gap between single-species tests and field studies can be bridged using model ecosystems (microcosms), which allow for the assessment of direct and indirect effects of the compounds under evaluation. In the present study, single-species toxicity tests and soil-spiked microcosms were used to comparatively investigate the toxicity of the non-systemic fungicide fludioxonil (FDO) on non-target soil organisms, with nematodes being the test organisms of choice. The potential effects of FDO on nematodes were investigated in two different test systems: (i) standardized toxicity tests using Caenorhabditis elegans exposed to FDO-spiked soil (FDO concentrations 50–1207 mg/kg soil dry weight) and (ii) in situ nematode communities sampled from microcosms containing FDO-spiked soil (FDO concentrations 75–600 mg/kg soil dry weight). FDO dose-dependently inhibited the reproduction of C. elegans, with an effect concentration (EC50) of 209.9 mg FDO/kg soil dry weight and a no observed effect concentration (NOEC) of 63.0 mg FDO/kg soil dry weight. In the microcosms, FDO significantly affected trait-based indices, such as the Maturity Index (MI25) and the Enrichment Index (EI), which responded already at FDO concentrations of 14.3 and 62.4 mg/kg dry soil. Overall, this study provides new insights into the impact of the non-systemic fungicide FDO on non-target soil organisms and demonstrates the suitability of nematode-based tools, that allow for a quick and cost-effective lower and higher tier risk assessment of plant protection products.

Development and application of a multispecies toxicity test with tropical freshwater microalgae

Microalgae are commonly used in ecotoxicity testing due to their ease of culturing and rapid cell division rates. These tests generally utilise a single species of algae; however, microalgae occur in the environment as complex communities of multiple species. To date, routine multispecies toxicity tests using tropical microalgae have not been available. This study investigated four tropical freshwater microalgal species for use in a chronic multispecies toxicity test based on the population growth (cell division) rate: Pediastrum duplex, Monoraphidium arcuatum, Nannochloropsis-like sp. and Chlorella sp. 12. Flow cytometric analysis identified the different fluorescence and light scattering properties of each algal species and quantified each species within multispecies mixtures. Following optimisation of test media nutrients and pH, a toxicity testing protocol was developed with P. duplex, M. arcuatum and Nannochloropsis-like sp. There were no significant differences in growth rates of each alga when tested over 72 h as single species or in multispecies mixtures. Atrazine and imazapic, two herbicides with different modes of action, were used to assess the sensitivity of the multispecies toxicity test. Atrazine was toxic to all species with 72-h IC10 values of 7.2, 63 and 280 μg/L for P. duplex, M. arcuatum and Nannochloropsis-like sp. respectively, while imazapic was not toxic to any species at concentrations up to 1100 μg/L. The toxicity of atrazine and imazapic to each microalgal species in the multispecies toxicity test was the same as that determined from single-species toxicity tests indicating that the presence of these microalgae in a mixture did not affect the toxicity of these two herbicides. This study is the first to develop a multispecies tropical microalgal toxicity test for application in freshwaters. This time- and cost-effective tool can be utilised to generate data to assist environmental decision making and to undertake risk assessments of contaminants in tropical freshwater environments.

Effect of algal surface area and species interactions in toxicity testing bioassays

Single and multispecies algal bioassays were assessed using copper toxicity with three green algae (Scenedesmus subspicatus, Scenedesmus quadricauda and Ankistrodesmus angustus) and one blue-green algae species (Oscillatoria prolifera). Single and multispecies toxicity tests were conducted based on cell density as per standard toxicity testing, and on equivalent surface area. A higher copper sulfate toxicity was registered for O. prolifera, followed by S. subspicatus, S. quadricauda, and A. angustus in single-species toxicity tests based on cell density. Single species toxicity tests based on surface area showed increased copper toxicity with increasing algal surface area except for A. angustus. In multispecies control bioassays, the growth of A. angustus was inhibited in the presence of other species in surface area-based tests. As compared to single species bioassays, O. prolifera, and S. quadricauda showed a decreased sensitivity to copper sulfate in both cell density and surface area based multispecies tests. However, for the algae species with the smallest surface area, S. subspicatus, 96h-EC50 value decreased in multispecies bioassays based on surface area as compared to the single species test, while it increased in multispecies bioassays based on cell density. The difference in S. subspicatus sensitivity to copper between tests based on cell density and surface area supports the need to adopt multispecies toxicity testing based on surface area to avoid the confounding effect on copper toxicity of increased biomass for metal binding. 96h-EC50 values for all species combined in the multispecies test based on cell density and on surface area were significantly different from 96h-EC50 values obtained in single species bioassays. These results demonstrate that single-species bioassays may over- or underestimate metal toxicity in natural waters.

Chronic Toxicity of Ferric Iron for North American Aquatic Organisms: Derivation of a Chronic Water Quality Criterion Using Single Species and Mesocosm Data

Iron is a common pollutant in waters near coal and hard rock mine disturbances. The current 1000 µg/L total recoverable chronic criterion for iron (Fe) for protection of aquatic life in the United States was developed using very limited data in 1976 and has not been revised since. To develop a more scientifically based criterion, several chronic laboratory toxicity experiments (> 30 days) were conducted with ferric Fe at circumneutral pH on a taxonomically diverse group of organisms including brown trout (Salmo trutta), mountain whitefish (Prosopium williamsoni), boreal toad tadpoles (Bufo boreas), the oligochaete worm Lumbriculus variegatus, the mayfly Hexagenia limbata, and the planarian Dugesia dorotocephala. Results of these tests and those of previously published toxicity data were used to derive a Final Chronic Value (FCV) of 499 µg/L by using the US Environmental Protection Agency’s recommended methods based on single species toxicity tests. In addition to single species toxicity tests, ferric Fe toxicity experiments (10 days) were performed on mesocosms containing naturally colonized communities of benthic macroinvertebrates. Fourteen genera in the mesocosms occurred at sufficient densities to estimate an iron concentration resulting in 20% reduction in abundance (EC20). Three of these taxa had EC20s less than the FCV of 499 µg/L derived from single species tests: the mayfly Epeorus sp. (335 µg/L), the caddisfly Micrasema sp. (356 µg/L), and midge Tanytarsini (234 µg/L). When mesocosm results were included, the FCV was lowered to 251 µg/L. These findings support the suggestion that modernization of water quality criteria should include data generated from mesocosm experiments and other lines of evidence.

Application of microcosm and species sensitivity distribution approaches in the ecological hazard assessment of 4-tert-butylphenol

ABSTRACTIn order to evaluate the ecological hazard of 4-tert-butylphenol (4-TBP), species abundances and physicochemical properties were monitored for 63 days in a microcosm system containing planktonic algae, rotifers, shrimps and Daphnia. The 63 days-NOEC (no observed effect concentration) of 4-TBP for this system was calculated. At the same time, species sensitivity distribution curves were constructed based on the toxicity data of EC10, EC50 and NOEC derived from laboratory single-species toxicity tests, which further gives no obvious hazard concentrations for 95% species in the ecosystem (HC5). In both the microcosm and the single-species tests, the shrimp (Neocaridina sp.) was the most sensitive species to 4-TBP exposure, while most algae species were relatively insensitive compared to the zooplankton. For the microcosm exposed to 4-TBP, significant changes occurred to the community structure instead of the community function, resulting in a NOECmicrocosm of 265.38 μg L–1 which approached the EC10-based HC5 (0.376 mg L–1). Nevertheless, EC50-based HC5 was higher than the NOECmicrocosm, and the NOEC-based HC5 was lower. The data showed that the EC10-based HC5 was protective at the similar level with the result of microcosm for 4-TBP, providing supportive data for chemical risk assessment.

Pollution-induced community tolerance in benthic macroinvertebrates of a mildly lead-contaminated lake

Pollution-induced community tolerance (PICT) has been used to demonstrate effects of sediment contamination on microbes and meiofauna. Our study explored the potential to detect PICT in benthic macroinvertebrates of a lake with long-term mild lead (Pb) contamination. We collected macrobenthos from two areas in Caddo Lake, Texas, a control area (CO) with a mean sediment Pb level of 11μg/g and Goose Prairie (GP) where sediment Pb levels averaged 74μg/g. Upon return to the laboratory, we exposed macroinvertebrates to a lethal lead concentration and assessed 48-h mortality. Mortality of CO macrobenthos was significantly higher than that of GP macrobenthos, providing evidence that these communities differed in their tolerance to lead. A comparison of macrobenthos community composition between the areas showed that the GP macrobenthos lacked metal-sensitive taxa such as gastropods and amphipods (which were present at CO). Similarly, a higher proportion of the GP benthos belonged to metal-tolerant taxa such as isopods and chironomids. Thus, changes in community composition appeared to be at least partly responsible for differences in community tolerance. Our results showed that a sediment Pb concentration below effect-based sediment quality guidelines had a measurable impact on macrobenthos, thus demonstrating that results from single-species toxicity tests may underestimate impacts on communities. This study also confirms that the PICT approach with macroinvertebrates is a feasible and potentially powerful approach for detecting contaminant impacts.

Larval aquatic insect responses to cadmium and zinc in experimental streams.

To evaluate the risks of metal mixture effects to natural stream communities under ecologically relevant conditions, the authors conducted 30-d tests with benthic macroinvertebrates exposed to cadmium (Cd) and zinc (Zn) in experimental streams. The simultaneous exposures were with Cd and Zn singly and with Cd+Zn mixtures at environmentally relevant ratios. The tests produced concentration-response patterns that for individual taxa were interpreted in the same manner as classic single-species toxicity tests and for community metrics such as taxa richness and mayfly (Ephemeroptera) abundance were interpreted in the same manner as with stream survey data. Effect concentrations from the experimental stream exposures were usually 2 to 3 orders of magnitude lower than those from classic single-species tests. Relative to a response addition model, which assumes that the joint toxicity of the mixtures can be predicted from the product of their responses to individual toxicants, the Cd+Zn mixtures generally showed slightly less than additive toxicity. The authors applied a modeling approach called Tox to explore the mixture toxicity results and to relate the experimental stream results to field data. The approach predicts the accumulation of toxicants (hydrogen, Cd, and Zn) on organisms using a 2-pKa bidentate model that defines interactions between dissolved cations and biological receptors (biotic ligands) and relates that accumulation through a logistic equation to biological response. The Tox modeling was able to predict Cd+Zn mixture responses from the single-metal exposures as well as responses from field data. The similarity of response patterns between the 30-d experimental stream tests and field data supports the environmental relevance of testing aquatic insects in experimental streams. Alkyl polyglucoside compound influences freshwater plankton community structure in floating field mesocosms

Synthetic surfactants in cleaners and detergents commonly contaminate freshwater systems, therefore use of low-toxicity alternatives is becoming increasingly important. Alkyl polyglucosides (APGs) derived from natural products are less toxic than synthetic surfactants, and degrade rapidly reducing chemical exposure time. However, single species toxicity tests showed APGs have toxic effects on aquatic primary producers and zooplankton, and that species demonstrate different sensitivities to APGs. Furthermore, species unaffected by APGs directly may be indirectly affected by removal of a food source or changes in predator densities, thereby changing plankton community structure. To determine the effects of APGs on plankton communities under environmental conditions, floating mesocosms were deployed in a shallow pond in southeast Georgia, USA and dosed with 0, 0.01, 2.5, 5, or 10mgL-1 APG. Zooplankton community composition and abundance, phytoplankton abundance (as chlorophyll a), and water column dissolved oxygen concentration were determined weekly for 1month. Zooplankton abundance decreased primarily due to loss of copepods, and community composition shifted toward small-bodied cladocerans (Bosmina sp.), and chlorophyll a concentrations declined by up to 81% following exposure to APG concentrations of 2.5mgL-1 or greater. Concentrations of dissolved oxygen never dropped below 5.70mgL-1, but the observed declines of~2mgL-1 could become stressful during periods of high water temperatures. Nevertheless, the APG-induced shift from copepod to cladoceran dominated communities and decrease in autochthonous carbon availability has important implications for food availability and quality to higher trophic levels such as planktivorous fishes.
