Environmentally driven changes in Atlantic salmon oxidative status interact with physiological performance

Farmed Atlantic salmon in sea cages experience natural seasonal changes in seawater temperature and photoperiod. Atlantic salmon have more oxidized tissues in spring and early summer, and this corresponds to the period of increasing day length, temperature, and growth rate. However, the molecular mechanisms underlying the redox metabolism of Atlantic salmon exposed to the interaction of seasonal cues remain unclear. This study aims to investigate the effect of seasonal changes in temperature and photoperiod on the redox metabolism at the molecular level of Atlantic salmon. Salmon reared in tanks were subjected to simulated natural (for farmed salmon in sea cages) photoperiod and temperature to mimic spring and summer (March to August). We found that the changing photoperiod and temperature, which were simulated to spring and summer conditions, influenced the redox metabolism of Atlantic salmon over the experimental period. We showed that increased utilization of tissue antioxidants such as vitamin C, vitamin E, and reduced glutathione in the spring was restored in August. The more reduced glutathione-based redox potentials and the low levels of malondialdehyde (MDA) in August in liver and muscle suggest lowered tissue oxidative activity at that time. We further revealed that the expression profiles of genes involved in growth hormone signaling and cell cycle regulation were correlated with oxidative stress patterns. Together, these promising results reveal the molecular mechanisms underlying the ecological implications of oxidative status. The study provides new insight into the possible role redox signaling plays in the growth regulation of fish to the seasonal shift in environmental factors.

Seasonal Changes in Photoperiod: Effects on Growth and Redox Signaling Patterns in Atlantic Salmon Postsmolts.

Farmed Atlantic salmon reared under natural seasonal changes in sea-cages had an elevated consumption of antioxidants during spring. It is, however, unclear if this response was caused by the increase in day length, temperature, or both. The present study examined redox processes in Atlantic salmon that were reared in indoor tanks at constant temperature (9 °C) under a simulated natural photoperiod. The experiment lasted for 6 months, from vernal to autumnal equinoxes, with the associated increase and subsequent decrease in day length. We found that intracellular antioxidants were depleted, and there was an increase in malondialdehyde (MDA) levels in the liver and muscle of Atlantic salmon with increasing day length. Antioxidant enzyme activity in liver and muscle and their related gene profiles was also affected, with a distinct upregulation of genes involved in maintaining redox homeostasis, such as peroxiredoxins in the brain in April. This study also revealed a nuclear factor-erythroid 2-related factor 2 (Nrf2)-mediated oxidative stress response in muscle and liver, suggesting that fish integrate environmental signals through redox signaling pathways. Furthermore, growth and expression profiles implicated in growth hormone (GH) signaling and cell cycle regulation coincided with stress patterns. The results demonstrate that a change in photoperiod without the concomitant increase in temperature is sufficient to stimulate growth and change the tissue oxidative state in Atlantic salmon during spring and early summer. These findings provide new insights into redox regulation mechanisms underlying the response to the changing photoperiod, and highlight a link between oxidative status and physiological function.

Environmental and Genetic (vgll3) Effects on the Prevalence of Male Maturation Phenotypes in Domesticated Atlantic Salmon

Pre-harvest male maturation is problematic for Atlantic salmon (Salmo salar) farmers and is regulated by the environment and genetics (e.g., vgll3). Five families of all-male salmon parr (produced using YY males crossed with XX females) with different vgll3 genotypes were split between three environmental regimes in January 2018. The “advanced maturation” regime used elevated temperature (16 °C) and continuous light from January 2018 with post-smolt maturation assessed in March 2018. The “extended freshwater” regime used ambient freshwater (1–16 °C) and simulated natural photoperiod (SNP) with post-smolt maturation assessed in November 2018. The “sea transfer” regime used ambient temperatures (1–14 °C) and SNP in freshwater until May 2018 when they were transferred to 9 °C seawater with natural photoperiod for 2.5 years (final mean weight of circa. 14 kg) and assessed for post-smolt maturation, 1 sea-winter (1 SW) maturation, and 2 sea-winter (2 SW) maturation in the autumn (November/December) of 2018, 2019, and 2020, respectively. Post-smolt maturation was highest in the advanced maturation and extended freshwater regimes (39–99% depending on family) and lowest in the sea transfer regime (0–95% depending on family). In the sea transfer regime, maturity incidence increased over time (0–95% post-smolt maturation, 1–100% 1 SW, and 50–90% 2 SW maturation, depending on family). In all regimes, those homozygous for the pre-designated vgll3 “early” maturing allele had the highest incidences of maturation whilst those homozygous for the “late” allele had the lowest. A low percentage of 2 SW phenotypic and genetic females were found (0–5% depending on family), one of which was successfully crossed with an XY male resulting in progeny with an approx. 50/50 sex ratio. These results show (i) post-smolt maturation varies dramatically depending on environment although genetic regulation by vgll3 was as expected, and (ii) crossing YY sperm with XX eggs can result in XX progeny which can themselves produce viable progeny with an equal sex ratio when crossed with an XY male.

Partial-year continuous light treatment reduces precocious maturation in age 1+ hatchery-reared male spring Chinook Salmon (Oncorhynchus tshawytscha).

Hatchery programs designed to conserve and increase the abundance of natural populations of spring Chinook Salmon Oncorhynchus tshawytscha have reported high proportions of males precociously maturing at age 2, called minijacks. High proportions of minijacks detract from hatchery supplementation, conservation and production goals. This study tested the effects of rearing juvenile Chinook Salmon under continuous light (LL) on minijack maturation in two trials. The controls were maintained on a simulated natural photoperiod for both trials. For trial 1, LL treatment began on the summer solstice 2019 or the autumn equinox 2019 and ended in late March 2020 (LL-Jun-Apr and LL-Sep-Apr, respectively). A significant reduction in the mean percent of minijacks (%MJ) was observed versus control (28.8%MJ) in both LL-Jun-Apr (5.4%MJ) and LL-Sep-Apr (9.3%MJ). Trial 2 was designed to evaluate whether stopping LL treatment sooner was still effective at reducing maturation proportions relative to controls. LL treatments began on the summer solstice 2020 and continued until the winter solstice (LL-Jun-Dec) or the final sampling in April 2021 (LL-June-Apr). LL-Jun-Dec tanks were returned to a simulated natural photoperiod after the winter solstice. Both photoperiod treatments showed a significant reduction in mean %MJ from the control (66%MJ): LL-Jun-Dec (11.6%MJ), LL-Jun-Apr (10.3%MJ). In both trials, minijacks had higher body weights, were longer and had increased condition factor when compared to females and immature males in all treatment groups at the final sampling. In both trials, there was little or no effect of LL treatment on fork length or body weight in immature males and females versus controls, but an increase in condition factor versus controls was observed. This study shows that continuous light treatment reduces minijack maturation in juvenile male spring Chinook Salmon and could provide an effective method for Spring Chinook Salmon hatcheries interested in reducing minijack production.

Evolution of sex ratio and egg production of gilthead seabream (Sparus aurata) over the course of five reproductive seasons

Spawning performance -relative fecundity and fertilization success- was studied in two hatchery-reared broodstocks of gilthead seabream (Sparus aurata) held under relatively constant well-water temperature (18–20 °C) and simulated natural photoperiod, for five consecutive spawning seasons, between 3 and 7 years of age. The spawning season lasted between 4 and 6 months each year, and the total number of eggs produced annually ranged between 1 480 000 and 3 100 000 eggs/kg female body weight, being the highest during the first and second spawning season. No difference was observed in monthly relative fecundity between years -although high variation existed within years, whereas fertilization success was the highest during the second and third reproductive season, and dropped significantly during the last year of the study. The male percentage of the broodstocks was 35% during the first spawning season of the females, and thereafter it decreased further and remained stable at around 15%–20% for the second and third spawning season. Substitution of older females with smaller males to readjust male percentage to 50% in the fourth spawning season, was followed by sex change of larger males to females and a drop of the male percentage to 18% in the following spawning season. The present study demonstrated the long spawning season of gilthead seabream -especially under constant water temperature, the high fecundity and fertilization success, and the stabilization of male percentage to ∼20% after the first spawning season or when a broodstock is modified to increase male percentage. These results are useful to the aquaculture industry, demonstrating that there is no need to add males to gilthead seabream broodstocks over the years, as the sex ratio is stabilized and egg production and fertilization success remain high with a relatively low male:female sex ratio.

Changes in circulating insulin-like growth factor-1 and its binding proteins in yearling rainbow trout during spring under natural and manipulated photoperiods and their relationships with gill Na+, K+-ATPase and body size

Smoltification in salmonids occurs during spring in response to increasing photoperiod to prepare for marine life. Smoltification is associated with increased hypo-osmoregulatory ability and enhanced growth potential, mediated by growth hormone and insulin-like growth factor (IGF)-1. Rainbow trout is uniquely insensitive to the induction of smoltification-associated changes by photoperiod, such as the activation of gill Na+,K+-ATPase (NKA). We measured the circulating IGF-1 and IGF-binding protein (IGFBP)-2b levels in yearling rainbow trout exposed to natural and manipulated photoperiods during spring and correlated these with gill NKA activity and body size. Although the effect of photoperiod manipulation on body size and circulating IGF-1 and IGFBP-2b was negligible, they were positively correlated with gill NKA activity in fish under simulated natural photoperiod. We next pit-tagged yearling rainbow trout and fed them a restricted ration or to satiation under a natural photoperiod. In April, gill NKA activity was higher in the satiation group than in the restricted group and positively correlated with body size and growth rate. In addition, circulating IGFBP-2b was positively correlated with gill NKA, size and growth, whereas circulating IGF-1 was correlated only with size and growth. The relationship between circulating IGF-1 and growth intensified from May to June, suggesting that the IGF-1–growth relationship was disrupted in April when gill NKA was activated. Two additional IGFBPs were related to growth parameters but not to gill NKA activity. The present study suggests that circulating IGFBP-2b and IGF-1 mediate the size-dependent activation of gill NKA in yearling rainbow trout during spring.

The importance of thermoperiod for proper gametogenesis and successful egg and sperm production in meagre (Argyrosomus regius) breeders in aquaculture

We examined the effect of constant water temperature throughout the year on gametogenesis, spawning success and egg/sperm/embryo quality in meagre (Argyrosomus regius). Two broodstocks were exposed to simulated natural photoperiod, andeither attenuated seasonal water temperature (SeasT, 16.4 to 19.6ºC) or relatively constant water temperature (CoT, 19.4 ± 0.6ºC).In the spawning period (May), 4 couples per group were induced to spawn with gonadotropin releasing hormone agonist (GnRHa). Gonadal stage of development, sperm quality parameters and plasma levels of sex steroids were evaluated prior to the GnRHa treatment. Spawning success and egg/sperm quality were examined over the following 4 weeks. Constant temperature did not prevent gametogenesis, but exposure to attenuated seasonal water temperature with the inclusion of winter low temperature was beneficial to both sexes. The mean (±SD) diameter of the largest vitellogenic oocytes prior to GnRHa administration was significantly higher in the SeasT compared to the CoT group (598 ± 27 vs 520 ± 17 μm). Testosterone plasma levels in the females were significantly higher in the SeasT group, but all other hormones were similar in both sexes. SeasT females spawned more consistently with higher relative fecundity, and 24-h embryo survival of the produced eggs. A more pronounced negative effect of constant water temperature was observed in males, since CoT males exhibited a spermiation index of 0 prior to GnRHa treatment, the latter clearly having a beneficial effect over the following 4 weeks. The study demonstrated that meagre do undergo gametogenesis to a significant extent even under constant water temperatures during the year. However, a seasonal thermal regime -even an attenuated one- was necessary for the proper development of the gametes, allowing for the successful spawning induction using the established GnRHa induction protocol.

A Short Period of Darkness after Mixing of Growing Pigs Intended for PDO Hams Production Reduces Skin Lesions.

Simple SummaryMixing unacquainted growing pigs is a common practice in commercial herds to adjust the group size to the pen dimensions and to balance the body weights of pigs within pens. Aggressive behavior following regrouping may include fights that can result in skin lesions and detrimental economic effects. Strategies aimed at limiting such issues can therefore improve animal welfare in practice. In the present study, we investigated the effects of darkness, maintained for 48 h after the formation of new social groups, on the expression of agonistic behavior and on the accumulation of skin lesions of growing pigs. The provision of 48 h of darkness significantly reduced the number of skin lesions on the mid- and rear thirds of pigs’ body. However, no corresponding reduction was observed in agonistic behavior, suggesting that darkness decreases the efficacy of aggressions, rather than how often or for how long they are expressed. Furthermore, an analysis of the location of lesions indicates that aggressions towards a fleeing companion, rather than reciprocal ones, were those mostly affected by darkness. The present results identify in the provision of darkness an easily applicable and relatively inexpensive intervention, that leads to the reduction of one of the most problematic consequences of agonistic interactions, i.e., skin lesions.Agonistic behavior after the regrouping of unfamiliar pigs has been recognized as one of the major welfare issues for pig husbandry, as it may result in lesions, lameness, and health problems. One scarcely investigated strategy to curb agonistic behavior is reducing the availability of visual stimuli potentially eliciting aggressions. In this study, we investigated the expression of agonistic behavior by growing pigs and the resulting accumulation of skin lesions over a period of 14 days following the formation of new social groups, which occurred in a condition of darkness maintained for 48 h. Compared to a simulated natural photoperiod (12 h light/day), darkness significantly reduced the number of skin lesions on the mid- and rear thirds of pigs’ body (p ≤ 0.01). A lack of corresponding decrease in frequency and duration of agonistic interactions suggests that darkness acts by decreasing the efficacy, not the expression, of aggressions. Furthermore, the location of lesions mostly affected by darkness indicates that the latter mostly acted by reducing the possibility of pigs to convey damage to a fleeing conspecific, rather than to one involved in a reciprocal fighting. The lighting regime provided did not affect growth performance traits of a 17-weeks feeding trial. The present results identify in the provision of darkness an easily applicable, and relatively inexpensive intervention, that leads to the reduction of skin lesions.

Physiological changes in off-season smolts induced by photoperiod manipulation in masu salmon (Oncorhynchus masou)

We induced season independent smolts in masu salmon (Oncorhynchus masou) by rearing juveniles under an advanced photoperiod (AP) in which LD15:9 was interrupted by LD9:15 from June for 4 months. AP fish showed higher degree of skin slivering from November than fish reared under a simulated natural photoperiod (SNP) and became larger in length and lower in condition factor in January. Gill Na+,K+-ATPase in AP fish started increasing in November and peaked in December to typical spring smolt levels while it remained low in SNP fish. In addition, AP fish had higher hypo-osmoregulatory ability than SNP fish, as judged by the lower serum sodium and chloride concentrations 48 h after seawater transfer. Serum levels of insulin-like growth factor (IGF)-I and IGF-binding protein (IGFBP)-1b, components of the somatotropic axis, in AP fish peaked in November and January, respectively. Serum IGFBP-1b levels were increased 48 h after transfer to 70% seawater in both groups, suggesting that it is not a quantitative index of hypo-osmoregulatory ability in the short-term but a marker of osmotic stress. The present study showed that off-season smolts could be induced in underyearling masu salmon by a square-wave photoperiod, and off-season smolts showed morphological and physiological changes comparable to yearling spring smolts. Increased levels of circulating IGF-I and IGFBP-1b in smolting fish suggest that these endocrine parameters are involved in the smoltification process.

Photoperiodic induction without light-mediated circadian entrainment in a High Arctic resident bird.

Organisms use changes in photoperiod to anticipate and exploit favourable conditions in a seasonal environment. While species living at temperate latitudes receive day length information as a year-round input, species living in the Arctic may spend as much as two-thirds of the year without experiencing dawn or dusk. This suggests that specialised mechanisms may be required to maintain seasonal synchrony in polar regions. Svalbard ptarmigan (Lagopus muta hyperborea) are resident at 74-81°N latitude. They spend winter in constant darkness (DD) and summer in constant light (LL); extreme photoperiodic conditions under which they do not display overt circadian rhythms. Here, we explored how Arctic adaptation in circadian biology affects photoperiodic time measurement in captive Svalbard ptarmigan. For this purpose, DD-adapted birds, showing no circadian behaviour, either remained in prolonged DD, were transferred into a simulated natural photoperiod (SNP) or were transferred directly into LL. Birds transferred from DD to LL exhibited a strong photoperiodic response in terms of activation of the hypothalamic thyrotropin-mediated photoperiodic response pathway. This was assayed through expression of the Eya3, Tshβ and deiodinase genes, as well as gonadal development. While transfer to SNP established synchronous diurnal activity patterns, activity in birds transferred from DD to LL showed no evidence of circadian rhythmicity. These data show that the Svalbard ptarmigan does not require circadian entrainment to develop a photoperiodic response involving conserved molecular elements found in temperate species. Further studies are required to define how exactly Arctic adaptation modifies seasonal timer mechanisms.

Effects of advanced and continuous photoperiod regimes on maturation control and profiles of sex steroids in brook trout (

Maturation of salmonid species reduces growth, flesh quality and immunocompetency and has inhibited, in part, the commercial production of brook trout in Tasmania. Photoperiod manipulation is routinely used to inhibit or reduce the incidence of maturation in salmonids, so an experimental adaptation of this approach was trialled experimentally for brook trout. Mixed-sex fish (age = 14 months) were subjected to simulated natural photoperiod (NP), advanced photoperiod (AP) and continuous photoperiod (CP) to investigate the response of endogenous circannual rhythm upon sexual maturity in this species. Light treatments commenced on the first day of the last month of the southern hemisphere’s winter and concluded towards the end of autumn when fish were ovulating. Maturation was observed in 100% of females and 96% of males held under NP. Fish exposed to AP corrected their maturation cycle by advanced phase shift of their endogenous rhythm for eight weeks and achieved final maturation during March–April. Plasma profiles of testosterone and oestradiol-17β were also adjusted according to advancement of photoperiod. Exposure to AP inhibited maturation by 6% and 8% in males and females, respectively. CP treatment for 10 consecutive months failed to inhibit gonadal development; however, this regime did delay or inhibit spermiation and ovulation.

Discontinuity in the molecular neuroendocrine response to increasing daylengths in Ile-de-France ewes: Is transient Dio2 induction a key feature of circannual timing?

In mammals, melatonin is responsible for the synchronisation of seasonal cycles to the solar year. Melatonin is secreted by the pineal gland with a profile reflecting the duration of the night and acts via the pituitary pars tuberalis (PT), which in turn modulates hypothalamic thyroid hormone status via seasonal changes in the production of locally-acting thyrotrophin. Recently, we demonstrated that, in the Soay sheep, photoperiodic induction of Tshb expression and consequent downstream hypothalamic changes occur over a narrow range of photoperiods between 12 and 14hours in duration. In the present study, we aimed to extend our molecular characterisation of this pathway, based on transcriptomic analysis of photoperiodic changes in the pituitary and hypothalamus of ovariectomised, oestradiol-implanted Ile-de-France ewes. We demonstrate that photoperiodic treatments applied before the winter solstice elicit two distinctive modes of accelerated reproductive switch off compared to ewes held on a simulated natural photoperiod, with shut-down occurring markedly faster on photoperiods of 13hours or more than on photoperiods of 12hours and less. This pattern of response was reflected in gene expression profiles of photoperiodically sensitive markers, both in the PT (Tshb, Fam150b, Vmo1, Ezh2 and Suv39H2) and in tanycytes (Tmem252 and Dct). Unexpectedly, the expression of Dio2 in tanycytes did not show any noticeable increase in expression with lengthening photoperiods. Finally, the expression of Kiss1, the key activator of gonadotrophin-releasing hormone release, was proportionately decreased by lengthening photoperiods, in a pattern that correlated strongly with gonadotrophin suppression. These data show that stepwise increases in photoperiod lead to graded molecular responses at the level of the PT, a progressive suppression of Kiss1 in the hypothalamic arcuate nucleus and luteinising hormone/follicle-stimulating hormone release by the pituitary, despite apparently unchanged Dio2 expression in tanycytes. We hypothesise that this apparent discontinuity in the seasonal neuroendocrine response illustrates the transient nature of the thyroid hormone-mediated response to long days in the control of circannual timing.

Flesh quality of Atlantic salmon smolts reared at different temperatures and photoperiods

Possible interactive effects of temperature and photoperiod on flesh quality in Atlantic salmon post‐smolts were studied. Juvenile (initial mean weight 96.0 g ± 3.1 SEM) Atlantic salmon were reared at six different combinations of temperatures (4.3, 6.5 or 9.3°C) and photoperiods (continuous light or simulated natural photoperiod). At termination of the trial, the fish were slaughtered and flesh samples taken to investigate quality and textural properties in the different experimental groups. Final weight in the six experimental groups varied between 174 and 345 g. Softer texture was seen in the fast growing groups. Photoperiod has only minor effect on flesh quality and textural properties, whereas temperature had significant impact on most of the measured variables. Although positive for growth, higher temperatures might be less favourable in relation to softer muscle tissue.

Effects of different photoperiod regimes on the smoltification and seawater adaptation of seawater-farmed rainbow trout (Oncorhynchus mykiss): Insights from Na+, K+–ATPase activity and transcription of osmoregulation and growth regulation genes

Photoperiod is thought to be the main zeitgeber that induces smoltification in salmonids. However, its effects on the smoltification of rainbow trout (Oncorhynchus mykiss) are not fully understood and no published data documents the effects of the photoperiod regime currently used commercially, continuous light (LL). The present study compared the effect of four different photoperiod regimes (i.e. advanced phase photoperiod (APP), delayed phase photoperiod (DPP), LL and simulated natural photoperiod (SNP)) on the smoltification and growth of juvenile rainbow trout during their freshwater phase of winter-spring and the following summer post smolt phase. Smoltification was evaluated by monitoring gill Na+,K+–ATPase (NKA) activity and transcription of NKA α-subunit isoforms 1a and 1b, and Na+,K+,2Cl‐cotransporter 1a. Growth was measured as specific growth rate of both length and weight, and through molecular growth proxies such as the levels of circulating insulin-like growth factor 1 (IGF-I) in plasma and transcription of igf-I, igf binding protein 1b (igfbp1b), growth hormone receptor 1 (ghr1) and cathepsin L (ctsl) in the liver. Results indicate that APP induces a longer smolt window and higher levels of plasma IGF-I in both freshwater and seawater (two months post transfer), while DPP led to a shorter smolt window, lower plasma IGF-I levels in freshwater and seawater, an earlier decrease in liver igf-I and ctsl transcription in freshwater (as seen by modelling over time) and lower specific growth rate in freshwater. The transcription analysis of osmoregulatory genes complemented NKA activity and allowed for the detection of a transient response to light and of differences between the osmoregulatory capacity of parr and desmolted fish. Furthermore, an upregulation of the liver transcription of igf-I, ghr1 and ctsl was found in all treatments during the smolt window, which corresponded to the periods with highest growth. Finally, both plasma IGF-I and liver igf-I in seawater were found to be significantly correlated to fish growth in seawater. However, our data did not show that plasma IGF-I prior to seawater transfer could be used as a reliable predictor of growth in seawater. Overall, and especially when compared with other salmonid species, photoperiod seems to be a weaker inducer of smoltification in rainbow trout, according to the parameters that were tested, suggesting that other environmental cues might be more important drivers of this process.

Advanced puberty triggered by bi-weekly changes in reproductive factors during the photolabile period in a male teleost fish, Dicentrarchus labrax L.

This study evaluated the impact of continuous light (LL) within the photolabile period on advanced puberty in juvenile male European sea bass. The exposure to an LL regime for 1 month, from August 15 to September 15 (LLa/s), was compared to a constant simulated natural photoperiod (NP) and constant continuous light conditions year-round (LLy). Somatic growth, hormone plasma levels, rates of testicular maturation and spermiation, as well as the mRNA levels of some reproductive genes were analyzed. Our results demonstrated that both LLa/s and LLy treatments, which include LL exposure during the photolabile period, were highly effective in inhibiting the gametogenesis process that affects testicular development, and clearly reduced the early sexual maturation of males. Exposure to an LL photoperiod affected body weight and length of juvenile fish during early gametogenesis and throughout the first year of life. Interestingly, LL induced bi-weekly changes in some reproductive factors affecting Gnrh1 and Gnrh2 content in the brain, and also reduced pituitary fshβ expression and plasmatic levels of 11-KT, E2, Fsh throughout early gametogenesis. We suggest that low levels of E2 in early September in the LL groups, which would be concomitant with the reduced number of spermatogonial mitoses in these groups, might indicate a putative role for estrogens in spermatogonial proliferation during the early gonadal development of this species. Furthermore, a significant decrease in amh expression was observed, coinciding with low plasma levels of 11-KT under LL regimes, which is consistent with the idea that this growth factor may be crucial for the progress of spermatogenesis in male sea bass.

The use of photoperiods to provide year round spawning in lumpfish Cyclopterus lumpus

In order to provide year round spawning broodstock, lumpfish (initial size 746 g and 24.9 cm) were reared under four different photoperiod regimes from January 2017 to July 2018. One group was reared under simulated natural photoperiod (LDN, control group) for Tromsø (70°N). The second group was transferred to continuous light (LD240) on 30 January 2017 and reared at LD24:0 throughout the trial period. Two compressed and phase advanced photoperiods were also established. Both groups were moved from LDN to LD24:0 on 30 January 2017, and after that reared at compressed natural photoperiods where the annual photoperiod was compressed down to six months (L6) or nine months (L9) for the duration of the study. Spawning time was shifted in both compressed groups during both years of the study. Spawning activity in the second year of the study was higher and followed more closely the expected spawning period in the compressed and the LDN groups. Spawning in the LD240 group was spread out over the experimental period with no distinct peak in spawning. A seasonal and pronounced drop in condition factor was found for females in the L9, L6 and the LDN groups. This post-spawning loss in condition was closely related to the spawning activity of each group. The current findings suggest that photoperiod has a strong influence on the timing of lumpfish maturation and can be used as an efficient and inexpensive tool to secure lumpfish reproduction operations i.e. year-round supply of egg and milt and/or timing with optimal temperature regimes.

Experimental evidence of masculinization by continuous illumination in a temperature sex determination teleost (Atherinopsidae) model: is oxidative stress involved?

The present study evaluates the influence of continuous light on phenotypic sex ratios in Chirostoma estor, a temperature sex determination animal model. Relative gene expression levels of 5 day old larvae were performed on two early gonad differentiation genes (sox9 and foxl2), two stress axis activation genes (gcr1 and crf) and four reactive oxygen species (ROS) antagonist effector genes (sod2, ucp2, gsr and cat). Two light treatments were applied from fertilization; control (12L:12D) simulated natural photoperiod and a continuous illumination photoperiod. By the end of the trial (12 weeks after hatching), differentiated and normal gonads were clearly identifiable in both treatments by histological observations. Regarding sex ratio, 73% of phenotypic males were found in continuous illumination compared with 40% in controls. Consistently, the sox9 gene (involved in early testis differentiation) showed an over expression in 64% of the individual larvae analysed compared with foxl2 (ovarian differentiation) suggesting a masculinization tendency in continuous illumination. On the other hand, only 36% of individuals showed the same tendency in the control treatment consistent with phenotypic sex ratios found under normal culture conditions. Relative gene expression results did not show significant difference in sod2, ucp2 and gcr1 levels, but cat, gsr and crf showed significantly higher expression levels in the continuous illumination treatment suggesting that both, the stress axis and ROS response mechanisms were activated at this time. This study suggests, a link between continuous light, oxidative stress and environmental sex determination in vertebrates. However, further research is necessary to describe this possible upstream mechanism that may drive some aspects of sexual plasticity in vertebrates.

Photoperiod manipulation for the reproductive management of captive wolffish populations: Anarhichas minor and A. lupus

This study provides new and practical information to implement the use of photoperiodic manipulation for the control of the sexual maturation of Canadian 6+ Atlantic wolffish (Anarhichas lupus) and 3+ first-time spawners spotted wolffish (Anarhichas minor) from Canadian and Norwegian populations. Wolffish reproductive cycle (gametogenesis and oocyte maturation and spermiation) was monitored for 23 consecutive months. Control groups were held under a 12-month simulated natural photoperiod and treatment groups under an 8-month compressed photoperiod. Plasma sex steroid concentrations (estradiol-17β and 11-ketotestosterone), oocyte diameter growth, and milt production were assessed monthly. For all groups under study, fish subjected to the compressed photoperiod spawned 2–6 months earlier than the controls. Complete out-of-shift cycle was not achieved based on the completion of two reproduction cycles, and this is briefly discussed. Photoperiod treatment induced temporal shifts in sex steroid profiles, which are the likely mediators of altered timing of ovulation/final maturation. Photoperiod has a strong influence on the timing of wolffish maturation and could be used as an efficient and inexpensive tool to secure wolffish reproduction operations (year-round supply of egg and milt and/or timing with optimal temperature regimes).

The effect of continuous light and compressed photoperiods on growth and maturation in lumpfish Cyclopterus lumpus

From an aquaculture point of view, control of the sexual maturation cycle is critical for a sustainable production of the species. For year-round reliable production of juvenile lumpfish of the appropriate size for stocking salmon cages, there is a need for basic and applied knowledge on the control of sexual maturation in cultured lumpfish broodstock. Lumpfish (initial size 219g and 16.9cm) were reared under advanced simulated natural photoperiod (SLDN, control group) for Tromsø (70°N). In addition there were two groups exposed to continuous light from April 2014 to January 2015 (PP3) and from April 2014 to April 2015 (PP6) followed by 8week decline in hours of light from 24 to 4h (autumn signal) and subsequent 8week rise from 4 to 24h (spring signal). Exposure of lumpfish to continuous light followed by an autumn-spring signal stimulated somatic growth and affected age at first maturity in females. The growth enhancing effect of continuous light lasting for approximately six months in females compared to one month in the males. Spawning colouration and running milt was seen in males in all three photoperiod groups from September 2014 onwards indicating that maturation started prior to the experimental treatment. In the females spawning time, egg volume and hatching success varied between the photoperiod groups. No spawning occurred in the SLND group, whereas spawning was seen in both PP female groups 3–6months after the onset of short autumn-short spring photoperiodic signal. Hatching success was higher in the PP3 group (83.4%) compared to the PP6 group (72.3%). The current findings are the first step in the development of photoperiod regimes which may provide simple and effective off-season maturation in lumpfish.

Efficacy of 24 h light to reduce maturation in Arctic charr (Salvelinus alpinus) is dependent on both the start date and duration

Two lab-based trials quantified the effect of a range of continuous light (LL) treatments overwinter on the somatic growth and incidence of sexual maturation in the fall age 2 of individually identified under-yearling Arctic charr (Labrador strain) at constant 10°C. Controls were maintained under simulated natural photoperiod (LDN; Latitude 45°N). In trial 1 (n=120/trt, mean 54g initial body weight), LL starting the 1st day of either October, November, December, January or February (all returned to LDN on April l) resulted in a step-wise reduction in maturity of 43, 55, 59, 67 73% compared with 77% in the control (sexes pooled). In trial 2 (n=90/trt, mean 24g initial body weight), LL starting either August 13 or October 1 and ending April 1 reduced maturity to <10%, compared to 33% (LL Dec1-Apr1) and 50% in the LDN controls. LL starting October 1 and ending either February 1, April 1 or June 1 were equally effective, resulting in <15% maturity. The importance of returning from LL to LDN in spring to reduce maturity was demonstrated by the LL constant (Oct–Oct) treatment which resulted in relatively high maturity, 31%. The reduction of maturity rate was independent of both somatic growth and condition factor during winter in all treatment groups. Among fish that matured, the timing of completion of sexual maturation was affected by both the LL start and end date.
