Effect of glycosylation on soy protein isolate–sodium carboxymethyl cellulose conjugates heat-induced gels and their applications as carriers of riboflavin

The objective of this study was to improve the rheological properties and riboflavin delivery ability of soy protein isolate (SPI) gels by incorporating sodium carboxymethyl cellulose (CMC) through glycosylation. The investigation focused on the structural and gel properties of SPI–CMC conjugates at different Maillard reaction times (30, 60, 90, 120, and 150 min). The results showed that the degree of grafting initially increased and then decreased as the Maillard reaction times increased, reaching its peak at 90 min (25.52%). The changes in the structure and conformation of the SPI–CMC conjugates were verified by Fourier transform infrared spectroscopy and surface hydrophobicity. An appropriate Maillard reaction reduced the content of highly ordered α-helix and β-sheet while increasing the content of disordered random coil and β-turn. This secondary structural change led to protein unfolding, exposure of surface hydrophobic groups, and orderly aggregation, forming a dense cross-linked gel network. When the Maillard reaction times increased, especially for 90 min, the gel strength and water-holding capacity of the SPI–CMC conjugate gels were significantly increased, along with a denser microstructure. Compared with the SPI and SPI–CMC mixture, SPI–CMC conjugate gels with 60 and 90 min reaction times exhibited notably increased viscosity and storage modulus (G′), as well as better structural recovery characteristics. The above physicochemical property improvements markedly enhanced the riboflavin encapsulation efficiency and riboflavin delivery ability of SPI–CMC conjugate gels, improving the ability to protect riboflavin from photodegradation and slowly releasing riboflavin in the simulated intestinal environment. Hence, SPI–CMC conjugate gels have significant potential for the delivery of nutrients.

Investigating the stability of vitamin D3 and Bifidobacterium lactis nanoparticles coated with polycaprolactone-polyethylene glycol-polycaprolactone triblock copolymer in Iranian white cheese and determining its physicochemical and sensory properties

ObjectiveTo investigate the stability and efficacy of vitamin D3 and Bifidobacterium lactis nanoparticles coated with polycaprolactone-polyethylene glycol-polycaprolactone (PCL-PEG-PCL) in enhancing the nutritional value of Iranian white cheese and to assess their impact on the cheese's physicochemical and sensory properties. MethodsBifidobacterium lactis and vitamin D3 were encapsulated using PCL-PEG-PCL. The particle size was analyzed using Dynamic Light Scattering (DLS). Five different cheese groups were produced and analyzed over two months. This included assessing microbial viability in simulated gastric and intestinal environments, quantifying vitamin D3 content, and evaluating physicochemical and sensory characteristics using a 9-point hedonic scale. ResultsThe Cheese with Bifidobacterium lactis coated with PCL-PEG-PCL and vitamin D3 nanoparticles (BCND3), showed a marked improvement in the stability of both Bifidobacterium lactis and vitamin D3. Specifically, the coated bacteria maintained a concentration of 106 cfu/g until day 45 in the intestinal model, demonstrating effective protection against the acidic gastric environment. The vitamin D3 nanoparticles displayed enhanced stability within the cheese matrix, contributing to the nutritional value and potentially extending the shelf life. Physicochemically, initial pH reduction due to lactic acid production stabilized by day 60, aligning with other groups. Sensory evaluation revealed a preference for the softer texture of cheese with vitamin D3 nanoparticles, suggesting a positive impact on consumer acceptability. ConclusionThe application of PCL-PEG-PCL coating significantly improved the stability of probiotics and vitamin D3 in cheese. This method proved effective in protecting Bifidobacterium lactis from harsh gastrointestinal conditions and in sustaining vitamin D3 content, with a notable impact on the cheese's sensory qualities. These findings highlight the potential of this coating technique in food fortification strategies.

Effects of Daidzein, Tempeh, and a Probiotic Digested in an Artificial Gastrointestinal Tract on Calcium Deposition in Human Osteoblast-like Saos-2 Cells.

Adequate calcium intake is crucial for the prevention and treatment of bone-related issues. Developing a nutritional source of readily bioavailable calcium is particularly significant for individuals deficient in this essential element and at risk of developing osteoporosis. This research aimed to evaluate the impact of tempeh (T), daidzein (D), and Lactobacillus acidophilus (LA) within a simulated intestinal environment consisting of Caco-2 epithelial and Saos-2 cells, focusing on their implications for bone mineralization mechanisms. In the initial phase, calcium bioaccessibility from calcium citrate (CaCt), LA, D, the daidzein combination D-CaCt-LA (D1:1:1), and the tempeh combination T-CaCt-LA (T1:1:1) was assessed through digestion simulation. The calcium content of both untreated and digested samples was determined using atomic absorption spectrometry (AAS). In the subsequent stage, the digested samples were used to induce intestinal absorption in differentiated enterocyte-like Caco-2 cells. The permeable fractions were then evaluated in a culture of osteoblast-like Saos-2 cells. Preliminary cellular experiments employed the MTT assay to assess cytotoxicity. The results indicated that the analyzed products did not influence the deposition of extracellular calcium in Saos-2 cells cultured without mineralization stimulators. The combined formulations of permeable fractions of digested CaCt, LA, D, and T demonstrated the capacity to enhance the proliferation of Saos-2 cells. In Saos-2 cells, D, D1:1:1, and LA showed no discernible impact on intracellular calcium accumulation, whereas T and T1:1:1 reduced the calcium deposits. Additionally, mRNA transcripts and alkaline phosphatase (ALP) activity levels in Saos-2 cells cultured without mineralization induction were unaffected by the analyzed products. An examination of the products revealed no discernible effect on ALP activity or mRNA expression during Saos-2 cell differentiation. Our findings suggest that tempeh, daidzein, and L. acidophilus did not positively impact cellular calcium deposition in Saos-2 cells. However, tempeh, daidzein and its combination, and L. acidophilus might enhance the process of osteogenic differentiation in Saos-2 cells. Nevertheless, this study did not identify any synergistic impact on calcium deposition and the process of osteogenic differentiation in Saos-2 cells of isoflavones and probiotics.

Digestive characteristics of extracellular polysaccharide from Lactiplantibacillus plantarum T1 and its regulation of intestinal microbiota

This study assessed the potential prebiotic characteristics of the previously reported Lactiplantibacillus plantarum extracellular polysaccharide (EPS-T1) with immunological activity. EPS-T1 was a novel heteropolysaccharide composed of glucose and galactose (1.00:1.21), with a molecular weight of 1.41 × 106 Da. The monosaccharide composition, molecular weight, fourier transform infrared, and 1H NMR analysis showed that EPS-T1 was well tolerated in the simulated oral cavity, gastric fluid, and small intestinal fluid environments, and was not easily degraded. Meanwhile, EPS-T1 could effectively be used as a carbon source to promote the growth of beneficial Lactobacillus species (Lactobacillus delbrueckii ssp. Bulgaricus, Streptococcus thermophilus, Lacticaseibacillus rhamnose GG, Lactiplantibacillus plantarum, Lacticaseibacillus paracasei and Lactobacillus reuteri). After 24 h of fecal fermentation, EPS-T1(5 mg/mL) effectively reduced the relative abundance of harmful bacteria such as the Escherichia-Shigella, Citrobacter, Fusobacterium, Parasutterella, and Lachnoclostridium. While, the level content of beneficial flora (Bacteroides, Blautia, Phascolarctobacterium, Bifidobacterium, Parabacteroides, and Subdoligranulum) were significantly increased. In addition, EPS-T1 was able to significantly promote the enrichment of short-chain fatty acids such as acetic acid, propionic acid and butyric acid. These results provide some basis for the functional application of EPS-T1 as a potential prebiotic.

Gut Microbiome Analysis and Screening of Lactic Acid Bacteria with Probiotic Potential in Anhui Swine.

With the widespread promotion of the green feeding concept of "substitution and resistance", there is a pressing need for alternative products in feed and breeding industries. Employing lactic acid bacteria represents one of the most promising antimicrobial strategies to combat infections caused by pathogenic bacteria. As such, we analyzed the intestinal tract of Anhui local pig breeds, including LiuBai Pig, YueHei Pig, and HuoShou Pig, to determine the composition and diversity of intestinal microbiota using 16S rRNA. Further, the functionality of the pigs' intestinal microbiota was studied through metagenomic sequencing. This study revealed that lactic acid bacteria were the primary contributors to the functional composition, as determined through a species functional contribution analysis. More specifically, the functional contribution of lactic acid bacteria in the HuoShou Pig group was higher than that of the LiuBai Pig and YueHei Pig. Subsequently, the intestinal contents of the HuoShou Pig group were selected for the screening of the dominant lactic acid bacteria strains. Out of eight strains of lactic acid bacteria, the acid-production capacity, growth curve, and tolerance to a simulated intestinal environment were assessed. Additional assessments included surface hydrophobicity, the self-aggregation capability, co-agglutination of lactic acid bacteria with pathogenic bacteria, and an in vitro bacteriostatic activity assay. Lactobacillus johnsonii L5 and Lactobacillus reuteri L8 were identified as having a strong overall performance. These findings serve as a theoretical basis for the further development of pig-derived probiotics, thereby promoting the application of lactic acid bacteria to livestock production.

247 In Vitro Characterization of Equine Fore- and Midgut Digestion

Abstract Considerable advancements can be made in the field of equine nutrition and diet formulation, by improved feed characterization and the development of nutritional models, including in vitro models, to describe nutrient degradation. Methodologically, literature values for in vitro small intestine incubation times are widely variable ranging from 2 to 18 h. While some incubation times are selected to represent approximate mean retention times, conversely, insufficient in vitro incubation times and sampling time points could prohibit or result in inaccuracies characterizing nutrient degradability characteristics, ultimately affecting both degradability and digestion values and their use. The objective of this trial was to assess whether macronutrient degradability plateaus when digested for 2 h in a simulated gastric environment plus 8, 12, or 16 h in a simulated small intestinal environment. To replicate in vivo digestion, a pepsin-pancreatin approach was selected. The first step of this method replicates the gastric environment. Ten g of each test ingredient are weighed into beakers containing 60 mL hydrochloric acid (HCl; 0.1 M) and 0.4 g pepsin (~2,500 U/g) solution. Beakers are then covered and incubated for 2 h at 39 ℃ under constant stirring. One mL aliquots of the digestion liquid are sampled at 0, 5, 10, 20, 30 60, 90 and 120 min after the start of the gastric simulation. Before replicating intestinal digestion, samples are neutralized with sodium hydroxide and the pH is increased to 7. The second step of this method replicates the small intestinal environment. Fifty mL of pancreatin-phosphate buffer (350 U/g protease, 6,000 FIP U/g lipase, 7,500 U/g amylase) is added to the beakers, which are then covered and incubated for either 8, 12, or 16 h at 39 ℃ under constant stirring. One mL aliquots of the digestion liquid are sampled at 0, 5, 10, 20, 30 60, 90, 120, 150, 180 min and thereafter hourly up until the end of digestion (480, 720, or 960 min). Aliquot samples are analyzed for starch via a glucose assay, protein via o-phthalaldehyde assay, and lipids via gas chromatography. A model will is fit to the time point data to determine degradation rates and degradable fractions of the digested samples. Results from these analyses go beyond estimating gross nutrient and digestible nutrient values and inform on the rate of degradation of nutrients of equine feedstuffs and the impact of in vitro methodology. The resulting degradability values will be used in a mechanistic model of equine digestion which will inform precision feeding. Ultimately this will improve how we feed horses to reduce nutrient excretion into the environment and reduce the risk of metabolic conditions such as laminitis.

In vitro dissolution/permeation tools for amorphous solid dispersions bioavailability forecasting II: Comparison and mechanistic insights

Along with the increasing demand for complex formulations comes the need for appropriate in vitro methodologies capable of predicting their corresponding in vivo performance and the mechanisms controlling the drug release which can impact on in vivo drug absorption. In vitro dissolution-permeation (D/P) methodologies that can account for the effects of enabling formulations on the permeability of drugs are increasingly being used in performance ranking during early development stages. This work comprised the application of two different cell-free in vitro D/P setups: BioFLUX™ and PermeaLoop™ to evaluate the dissolution-permeation interplay upon drug release from itraconazole (ITZ)- HPMCAS amorphous solid dispersions (ASDs) of different drug loads. A solvent-shift approach was employed, from a simulated gastric environment to a simulated intestinal environment in the donor compartment. PermeaLoop™ was then combined with microdialysis sampling to separate the dissolved (free) drug from other species present in solution, like micelle-bound drug and drug-rich colloids, in real time. This setup was applied to clarify the mechanisms for drug release and permeation from these ASDs. In parallel, a pharmacokinetic study (dog model) was conducted to assess the drug absorption from these ASDs and to compare the in vivo results with the data obtained from each in vitro D/P setup, allowing to infer which would be the most adequate setup for ASD ranking. Even though both D/P systems resulted in the same qualitative ranking, BioFLUX™ overpredicted the difference between the in vivo AUC of two ASDs, whereas PermeaLoop™ permeation flux resulted in a good correlation with the AUC observed in pharmacokinetic studies (dog model) (R2 ≈ 0.98). Also, PermeaLoop™ combined with a microdialysis sampling probe clarified the mechanisms for drug release and permeation from these ASDs. It demonstrated that the free drug was the only driving force for permeation, while the drug-rich colloids kept permeation active for longer periods by acting as drug reservoirs and maintaining constant high levels of free drug in solution, which are then immediately able to permeate. Hence, the data obtained points BioFLUX™ and PermeaLoop™ applications to different momentums in the drug product development pipeline: while BioFLUX™, an automated standardized method, poses as a valuable tool for initial ASD ranking during the early development stages, PermeaLoop™ combined with microdialysis sampling allows to gain mechanistic understanding of the dissolution-permeation interplay, being crucial to fine tune and identify leading ASD candidates prior to in vivo testing.

Freestanding region-responsive bilayer for functional packaging of ingestible devices

Ingestible capsules have the potential to become an attractive alternative to traditional means of treating and detecting gastrointestinal (GI) disease. As device complexity increases, so too does the demand for more effective capsule packaging technologies to elegantly target specific GI locations. While pH-responsive coatings have been traditionally used for the passive targeting of specific GI regions, their application is limited due to the geometric restrictions imposed by standard coating methods. Dip, pan, and spray coating methods only enable the protection of microscale unsupported openings against the harsh GI environment. However, some emerging technologies have millimeter-scale components for performing functions such as sensing and drug delivery. To this end, we present the freestanding region-responsive bilayer (FRRB), a packaging technology for ingestible capsules that can be readily applied for various functional ingestible capsule components. The bilayer is composed of rigid polyethylene glycol (PEG) under a flexible pH-responsive Eudragit® FL 30 D 55, which protects the contents of the capsule until it arrives in the targeted intestinal environment. The FRRB can be fabricated in a multitude of shapes that facilitate various functional packaging mechanisms, some of which are demonstrated here. In this paper, we characterize and validate the use of this technology in a simulated intestinal environment, confirming that the FRRB can be tuned for small intestinal release. We also show a case example where the FRRB is used to protect and expose a thermomechanical actuator for targeted drug delivery.

Cellulose Nanofiber Platform for Pesticide Sequestration in the Gastrointestinal Tract.

Exploitation of nature-derived materials is an important approach to promote environmental sustainability. Among these materials, cellulose is of particular interest due to its abundance and relative ease of access. As a food ingredient, cellulose nanofibers (CNFs) have found interesting applications as emulsifiers and modulators of lipid digestion and absorption. In this report, we show that CNFs can also be modified to modulate the bioavailability of toxins, such as pesticides, in the gastrointestinal tract (GIT) by forming inclusion complexes and promoting interaction with surface hydroxyl groups. CNFs were successfully functionalized with (2-hydroxypropyl)-β-cyclodextrin (HPBCD) using citric acid as a crosslinker via esterification. Functionally, the potential for pristine and functionalized CNFs (FCNFs) to interact with a model pesticide, boscalid, was tested. Based on direct interaction studies, adsorption of boscalid saturated at around 3.09% on CNFs and at 12.62% on FCNFs. Using an in vitro GIT simulation platform, the adsorption of boscalid on CNFs/FCNFs was also studied. The presence of a high-fat food model was found to have a positive effect in binding boscalid in a simulated intestinal fluid environment. In addition, FCNFs were found to have a greater effect in retarding triglyceride digestion than CNFs (61% vs 30.6%). Overall, FCNFs were demonstrated to evoke synergistic effects of reducing fat absorption and pesticide bioavailability through inclusion complex formation and the additional binding of the pesticide onto surface hydroxyl groups on HPBCD. By adopting food-compatible materials and processes for production, FCNFs have the potential to be developed into a functional food ingredient for modulating food digestion and the uptake of toxins.

Effect of kidney bean extract on phytohaemagglutinin activity, functional properties, and the estimated glycaemic index in‐vitro

SummaryThis study aimed to decrease the activity of phytohaemagglutinins and kidney bean α‐amylase activity in some selected stable foods. Kidney bean α‐amylase inhibitors may inhibit mammalian α‐amylase, reducing the estimated glycaemic index (eGI). The effect of temperature, pH and protease pretreatment on PHA activity in the crude extract was investigated. Besides, the inhibitory effect of kidney bean extract on α‐amylase activity and properties was assessed. Results showed that the PHA activity was decreased to 6.97% from the aqueous extract of kidney beans, while the α‐AI activity was retained by 85.89% by acid protease treatment. The extract's α‐amylase inhibitory activity remained at 87.37% following simulation in a gastric environment, whereas no effect was observed in a simulated intestinal environment. Adding 3% of the extract decreased the eGI of bread, rice, steamed bread, and steamed corn bread from 65.00, 71.65, 68.17, and 63.41 to 30.69, 39.51, 22.30, and 19.69, respectively. The processing method of crude kidney bean extract examined in this study has significant reference value for the development of kidney bean α‐amylase inhibitors and can enhance the intensive processing level and industrial development value of kidney beans worldwide. This discovery suggests that kidney bean extracts can be used to develop foods with a low GI.

Characterization and evaluation of an oral vaccine via nano-carrier for surface immunogenic protein (Sip) delivery against Streptococcus agalactiae infection

Streptococcus agalactiae causes systemic disease in a variety of wild and farmed fish, resulting in high levels of morbidity and mortality, as well as serious economic losses to the Nile tilapia aquaculture industry. The development of economic and applicable oral vaccines is therefore urgently needed for the sustainable development of Nile tilapia aquaculture. In this study, mesoporous silica nanoparticles (MSNs) were fabricated using sol–gel synthesis technology, and the antigens of surface immunogenic protein (Sip) was loaded into MSNs to develop a nanovaccine MSNs-Sip@HP55. The results showed that the prepared nanovaccine exhibited pH-controlled release, which could survive in the simulated gastric environment (pH 1.5), and release antigens in the simulated intestinal environment at pH 7.4. The nanovaccine could induce innate and adaptive immune responses in Nile tilapia. When the challenge doses were 1.5 × 106, 1.18 × 106, and 0.88 × 106 CFU/mL, the relative protection rates in immunized Nile tilapia were 63.33 %, 64.23 %, and 76.31 %, respectively. Taken together, the nanovaccine exhibited a high antigen utilization rate and was easily administered orally via feeding, which could protect Nile tilapia against challenge with S. agalactiae in large-scale farms. Oral vaccine based on MSNs carriers is a potentially promising strategy for the development of fish vaccines.

Synthesis and Characterization of Temperature‐/pH‐Responsive Hydrogels for Drug Delivery

AbstractA temperature‐/pH‐responsive hydrogel was successfully prepared by in situ free radical polymerization of N‐isopropyl acrylamide (NIPAm), itaconic acid (IA), and poly (ethylene glycol) methyl ether acrylate (OEGA). The pore size, mechanical strength, and swelling ratio of the P(NIPAm‐IA‐OEGA) were strongly affected by the contents of IA. 5‐Fluorouracil (5‐Fu) was chosen as a model drug, and the maximum drug loading was 68 mg/g. P(NIPAm‐IA‐OEGA) had a much greater release rate in a simulated intestinal fluid environment (47.73 %, 16 h) than in gastric juice (33.11 %, 16 h). The hemolysis test and cytotoxicity test proved that prepared hydrogel had good biocompatibility. In summary, P(NIPAm‐IA‐OEGA) can be used for oral drug delivery systems in cancer therapy.

Theophylline-based hybrids as acetylcholinesterase inhibitors endowed with anti-inflammatory activity: synthesis, bioevaluation, in silico and preliminary kinetic studies.

In this study, we investigated the conjugation of theophylline with different compounds of natural origin hoping to construct new hybrids with dual activity against cholinergic and inflammatory pathways as potential agents for the treatment of Alzheimer's disease (AD). Out of 28 tested hybrids, two hybrids, acefylline-eugenol 6d and acefylline-isatin 19, were able to inhibit acetylcholinesterase (AChE) at low micromolar concentration displaying IC50 values of 1.8 and 3.3 μM, respectively, when compared to the galantamine standard AChE inhibitor. Moreover, the prepared hybrids exhibited a significant anti-inflammatory effect against lipopolysaccharide induced inflammation in RAW 264.7 and reduced nitric oxide (NO), tumor necrosis alpha (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) levels in a dose dependent manner. These hybrids demonstrated significant reductions in nitric oxide (NO), tumor necrosis alpha (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) levels in RAW 264.7 cells induced by lipopolysaccharide (LPS). The findings of this study were further explained in light of network pharmacology analysis which suggested that AChE and nitric oxide synthase were the main targets of the most active compounds. Molecular docking studies revealed their ability to bind to the heme binding site of nitric oxide synthase 3 (NOS-3) and effectively occupy the active site of AChE, interacting with both the peripheral aromatic subsite and catalytic triad. Finally, the compounds demonstrated stability in simulated gastric and intestinal environments, suggesting potential absorption into the bloodstream without significant hydrolysis. These findings highlight the possible therapeutic potential of acefylline-eugenol 6d and acefylline-isatin 19 hybrids in targeting multiple pathological mechanisms involved in AD, offering promising avenues for further development as potential treatments for this devastating disease.

Chain conformation, mucoadhesive properties of fucoidan in the gastrointestinal tract and its effects on the gut microbiota

Fucoidans are valuable marine polysaccharides with various bioactivities and physicochemical properties. However, its digestive properties, mucoadhesive properties, and bioactivity in the gastrointestinal tract are still unclear. In this study, simulated digestion, fecal fermentation in vitro, and rheology models were utilized to investigate the chain conformation, influence on gut microbiota, and mucin adhesive properties of fucoidan from the sea cucumber Thelenota ananas (Ta-FUC). The results showed that Ta-FUC was nondigestible with a temporary decrease in molecular weight in gastric conditions, accompanied by the chain conformation becoming more flexible. Moreover, Ta-FUC exhibited strong mucin adhesive function in the simulated intestinal environment, with supramolecular disulfide, hydrogen, and hydrophobic interactions in order of intensity. During fermentation, Ta-FUC was degraded by the intestinal flora to produce various short-chain fatty acids and promoted the relative abundance of Bacteroidota and Firmicutes, reducing the proportion of Proteobacteria. Therefore, these results indicate that Ta-FUC could be a potential prebiotic and ingredient for developing targeted delivery systems in the functional food and pharmaceutical industries.

Edible hydrogel from gelatin and alginate as functional low‐calorie noodle

AbstractThere is an urgent need to develop edible hydrogels that can be served as a main dish with low‐calorie content and high satiety value, yet are easy to digest. However, the development of such hydrogels remains challenging. In this study, low‐calorie edible hydrogels were prepared from gelatin and sodium alginate (gel/SA hydrogels) through a facile acid soaking‐drying method. These gel/SA hydrogels exhibited excellent mechanical properties with texture characteristics similar to that of Liangpi. The sensitivity of gelatin and acidified sodium alginate to pH resulted in a high swelling ratio of the hydrogels, contributing to weight retention in a simulated stomach environment and rapid degradation in a simulated intestinal environment. These properties indicate that these gel/SA hydrogels have a good prospect of being used as low‐calorie food with high satiety value and are easy to digest. Konjac glucomannan can also be incorporated into the gel/SA hydrogels to further improve their biological functionality.

Ultrasonic pretreatment improved the physicochemical properties and riboflavin delivery ability of transglutaminase-catalyzed soy protein isolate gel

Soy protein isolate (SPI) was pre-treated with ultrasonic (400 W, 30–120 min) and then formed gel using transglutaminase (TG) method. Effects of ultrasonic pretreatment on structural, rheological and thermal properties as well as riboflavin delivery ability of SPI gel were investigated. Ultrasonic treatment significantly decreased the particle size, increased the free sulfhydryl content, enhanced the surface hydrophobicity, unfolded the conformational structure of SPI, facilitated the disulfide bonds and ε-(γ-glutamyl) lysine isopeptide bonds formation in transglutaminase-catalyzed soy protein isolate gel (TCSG). Ultrasonic pretreatment, particular for 60 min, significantly enhanced the gel strength, storage modulus, loss modulus, consistency and thermal stability, but decreased the flow behavior index of TCSG. The TCSG without ultrasonic pretreatment showed a non-uniform gel network with large and irregular pores. When applied ultrasonic pretreatment, more dense, uniform and stable gel networks were developed. The above physicochemical properties improvements inhibited the swelling behavior and markedly enhanced the riboflavin encapsulation efficiency and riboflavin delivery ability of TCSG, leading to 81% of riboflavin released in simulated intestinal environment. Therefore, ultrasonic pretreatment could be an efficient approach to improving the gel properties and nutrients delivery ability of SPI gel. • Ultrasonic decreased the particle size and increased the free SH content of SPI. • Ultrasonic enhanced the strength, G′, G″, consistency and thermal stability of TCSG. • US-TCSG samples showed more dense, uniform and stable gel networks. • The gel swelling behavior was inhibited and riboflavin delivery ability was enhanced. • 81% of riboflavin was released in simulated intestinal environment.

Lecithin-Polysaccharide Self-Assembled Microspheres for Resveratrol Delivery.

Poor water solubility and low chemical stability, seriously limit the efficient bioavailability of resveratrol. Here, we propose encapsulating resveratrol in lecithin-polysaccharide self-assembled microspheres (LPSM). An LPSM was designed with a lecithin core, and alginate-carboxymethyl chitosan biolayer shell. The LPSM had a spherical shape with 12.171 ± 0.960 μm of particle size and −30.86 ± 1.37 mV of zeta potential. The introduce of lecithin remarkably increased the encapsulation efficiency of resveratrol to 92.78 ± 0.82%. The LPSM elevated the antioxidant capacity and ultraviolet resistance of resveratrol. Moreover, LPSM inhibited release in a simulated gastric environment, promoted sustained release in simulated intestinal environment, and elevated the bioavailability of resveratrol during in vitro simulated digestion. Results indicate that LPSM is promising as a carrier for resveratrol delivery to enhance stability and bioaccessibility.

One-step emulsification for controllable preparation of ethyl cellulose microcapsules and their sustained release performance

A simple and versatile strategy for controlled production of monodisperse ethyl cellulose (EC) microcapsules by a single-stage emulsification method has been developed. Monodisperse oil-in-water emulsions, obtained by a microfluidic device, are used as templates for preparing EC microcapsules. Oil-soluble ethyl acetate (EA) is miscible with water, so the interfacial mass transfer between EA and water occurs sufficiently, which leads to water molecules pass through the phase interface and diffuse into emulsion interior. Water molecules aggregate at the interface, and some merge into a large water drop in the central position of the emulsion. After evaporation of EA solvent, monodisperse EC microcapsules create large numbers of pits on the surface with a hollow structure. Curcumin is used as a model drug and embedded in the hollow structure. EC microcapsules have good, sustained drug release efficacy in a simulated intestinal environment, and the release process of EC microcapsules containing 6.14% drug-loaded capacity is fully consistent with the vitro drug release model. Such simple techniques for making EC microcapsules may open a window to the controlled preparation of other multifunctional microcapsules. Besides, it offers theoretical guidance for the study of EC microcapsules as drug carriers and expanding clinical application of curcumin.

In vitro Cholesterol Assimilation by Bifidobacterium animalis subsp. lactis (BPL1) Probiotic Bacteria Under Intestinal Conditions.

Hypercholesterolemia is one of the principal causes of the development of cardiovascular diseases. Recently, probiotics consumption has also been proposed as a non-pharmacological intervention to control cholesterol concentrations. To evaluate in vitro assimilation of cholesterol by Bifidobacterium animalis subsp. lactis (BPL1) under simulated intestinal environment in anaerobic conditions and to review and discuss potential physiological mechanisms in this context. Bacterial viability and cholesterol assimilation were evaluated in both standard MRS and Stimulated Intestinal Fluid (SIF) medium under anaerobic conditions and in the presence or absence of cholesterol. For assimilation assays, cholesterol concentrations in the different suspensions, containing the probiotic or not, were determined by chromatography coupled to mass spectrometry. The results showed that the growth of B. lactis BPL1 under intestinal conditions is favored when cholesterol is present in the culture medium. In addition, cholesterol assimilation of up to 44.4% under intestinal and anaerobic conditions was observed. Taking into account the revised literature and the experimental results presented herein, the administration of functional foodstuffs together with probiotic bacteria, such as B. lactis BPL1, could be a potentially effective option to decrease hypercholesterolemia, thus preventing the development of cardiovascular diseases. Nevertheless, further studies on mechanisms of effectiveness in animals and clinical trials are still needed.

The Effects of Various Food Products on Bisphosphonate's Availability.

The bioavailability of orally administered bisphosphonates is very low (<1%) due to their short absorption window in the proximal duodenum and high affinity for food. Food ingredients are able to bind the drug, but the presence of food extends the residence time of bisphosphonates in the absorption window. Therefore, the main goal of this study is to select a group of food products that are characterized by low binding affinity to bisphosphonates and thus will not reduce their availability upon concomitant administration. For this purpose, a combination of three methods was applied: (1) evaluation of sorption capacity for rows of digested food samples in a simulated intestinal environment; (2) evaluation of drug availability in simulated chyme; and (3) evaluation of drug availability using a simulating needle device. The results indicate that food products such as egg white and white bread are most suitable for consumption during oral bisphosphonate intake.
