Weak Exchange Interactions in Multispin Systems: EPR Studies of Metalloporphyrins Decorated with {Cr7Ni} Rings.

Both metalloporphyrins and heterometallic {Cr7Ni} rings are of significant research interest due to their proposed roles in quantum information processing devices. In this study, we present a series of complexes in which [Cr7NiF3(Etglu)(O2CtBu)15] (N-EtgluH5 = N-ethyl-d-glucamine) heterometallic rings are coordinated to metalloporphyrin linkers: the symmetric [M(TPyP)] for M = Cu2+, VO2+, and H2TPyP = 5,10,15,20-tetra(4-pyridyl)porphyrin; and the asymmetric [{VO}(TrPPyP)] for H2(TrPPyP) = 5,10,15-(triphenyl)-20-(4-pyridyl)porphyrin. The magnetic interactions present in these complexes are unraveled using the continuous wave (CW) electron paramagnetic resonance (EPR) technique. The nature of the coupling between the {Cr7Ni} rings and the central metalloporphyrin is assessed by numerical simulations of CW EPR spectra and determined to be on the order of 0.01 cm-1, larger than the dipolar ones and suitable for individual spin addressability in multiqubit architectures.

Copper(II) coordination to the intrinsically disordered region of SARS-CoV-2 Nsp1

The intrinsically disordered C-terminal peptide region of severe acute respiratory syndrome coronavirus 2 nonstructural protein-1 (Nsp1-CT) inhibits host protein synthesis by blocking messenger RNA (mRNA) access to the 40S ribosome entrance tunnel. Aqueous copper(II) ions bind to the disordered peptide with micromolar affinity, creating a possible strategy to restore protein synthesis during host infection. Electron paramagnetic resonance (EPR) and tryptophan fluorescence measurements on a 10-residue model of the disordered protein region (Nsp1-CT10), combined with advanced quantum mechanics calculations, suggest that the peptide binds to copper(II) as a multidentate ligand. Two optimized computational models of the copper(II)-peptide complexes were derived: One corresponding to pH 6.5 and the other describing the complex at pH 7.5 to 8.5. Simulated EPR spectra based on the calculated model structures are in good agreement with experimental spectra.

Low-temperature trapping of N2 reduction reaction intermediates in nitrogenase MoFe protein-CdS quantum dot complexes.

The biological reduction of N2 to ammonia requires the ATP-dependent, sequential delivery of electrons from the Fe protein to the MoFe protein of nitrogenase. It has been demonstrated that CdS nanocrystals can replace the Fe protein to deliver photoexcited electrons to the MoFe protein. Herein, light-activated electron delivery within the CdS:MoFe protein complex was achieved in the frozen state, revealing that all the electron paramagnetic resonance (EPR) active E-state intermediates in the catalytic cycle can be trapped and characterized by EPR spectroscopy. Prior to illumination, the CdS:MoFe protein complex EPR spectrum was composed of a S = 3/2 rhombic signal (g = 4.33, 3.63, and 2.01) consistent with the FeMo-cofactor in the resting state, E0. Illumination for sequential 1-h periods at 233K under 1 atm of N2 led to a cumulative attenuation of E0 by 75%. This coincided with the appearance of S = 3/2 and S = 1/2 signals assigned to two-electron (E2) and four-electron (E4) reduced states of the FeMo-cofactor, together with additional S = 1/2 signals consistent with the formation of E6 and E8 states. Simulations of EPR spectra allowed quantification of the different E-state populations, along with mapping of these populations onto the Lowe-Thorneley kinetic scheme. The outcome of this work demonstrates that the photochemical delivery of electrons to the MoFe protein can be used to populate all of the EPR active E-state intermediates of the nitrogenase MoFe protein cycle.

Spectroscopic and magnetic investigations of the dilute magnetically doped semiconductors BaSn1-xMnxO3 (0.02 ≤ x ≤ 0.1)

Dilute magnetically doped semiconductors (DMSs) are key to fabricating novel spintronic devices with high functionalities, which could be utilized for the next-generation technological development. BaSnO3 is a transparent wide band gap (∼3.1 eV) semiconductor, in which magnetism can be induced through a subtle amount of magnetic impurity doping without deterring its optical transparency. In this work, we report an experimental investigation on the optical spectroscopic and magnetic properties of the DMSs BaSn1-xMnxO3 (0 ≤ x ≤ 0.1), synthesized using standard solid-state-reaction method at high-temperature and ambient pressure, through various spectroscopic, electron paramagnetic resonance (EPR) and temperature- and field-dependent magnetization measurements. All samples are found to be crystallize in the cubic structure with space group Pm3‾m, in which Mn ions are stabilized in the 4+ valence state. We find that the optical band gap monotonically decreases (from 3.1 eV to 2.08 eV) with increase in Mn doping concentration, which in turn leads to an increase in absorption efficiency in the visible region. An enhanced sp-d hybridization between localized d electrons of Mn ions and band electrons is likely the main reason for such band gap reduction. Interestingly, theoretical simulation of EPR spectra suggests that the signal is primarily arising from a combination of two spin systems, i.e., Mn4+ ions and singly ionized oxygen vacancies. The strength of magnetic interaction between Mn4+ ions is also increased for higher doped samples. Moreover, the nature of magnetic interaction is predominantly antiferromagnetic in higher doped samples, as corroborated by the negative value of Curie-Weiss temperature from the inverse susceptibility fitting.

Orientation Distribution of Molecules in a Smectic Liquid Crystal with a Distorted Director Geometry.

Quantitative determination of the molecular orientation distribution function in samples of liquid crystals with a complex director geometry was performed using the numerical simulation of electron paramagnetic resonance (EPR) spectra of the spin probes in a liquid-crystalline medium. To achieve the quantitative agreement of experimental and simulated EPR spectra, the hierarchy of the orientation order was explicitly taken into account, namely, the local ordering of liquid crystal molecules by the mean-field potential of surrounding molecules, and the partial disordering of local directors within the sample. The samples under study are planar and twist LC cells with liquid crystal 8CB cooled from the nematic into the smectic A phase in the magnetic field. The presence of the magnetic field perpendicular to the cell director leads to distortion of the orientation of the liquid crystal in the cell. The spin probe technique was successfully employed for the reliable measurement of orientation distribution functions of the low nonorthorhombic symmetry. Orientation order parameters up to 12th rank were measured, including nonaxial and nonorthorhombic order parameters. It is shown that the presence of several contradicting aligning forces leads to the tilt of the preferential director toward the direction, which is a compromise between the orienting forces.

Role of the paramagnetic donor-like defects in the high n-type conductivity of the hydrogenated ZnO microparticles

The magnetic and electronic properties of the hydrogenated highly conductive zinc oxide (ZnO) microparticles were investigated by electron paramagnetic resonance (EPR) and contactless microwave (MW) conductivity techniques in the wide temperature range. The EPR spectra simulation allowed us to resolve four overlapping EPR signals in ZnO microparticles. The Lorentzian EPR line with isotropic g-factor 1.9623(5) was related to the singly ionized oxygen vacancy. Another Lorentzian line with g|| = 1.9581(5), g⊥ = 1.9562(5) was attributed to the zinc interstitial shallow donor center, while EPR signal with g|| = 1.9567(5), g⊥ = 1.9556(5) and Gaussian lineshape was assigned to the hydrogen interstitial shallow effective-mass-like donor. The EPR signal with g|| = 1.9538(5), g⊥ = 1.9556(5) and Lorentzian lineshape was tentatively attributed to the shallow donor center. The charge transport properties in ZnO microparticles have been investigated by the contactless MW conductivity technique at T = 5–296 K. Two conduction mechanisms, including ionization of electrons from the shallow donors to the conduction band and hopping conduction process, have been distinguished. The hopping conduction process follows Mott’s variable-range hopping T−1/4 law at T = 10–100 K. The evaluated values of the average hopping distance (15.86 Å), and hopping energy (1.822 meV at 40 K) enable us to estimate the donor concentration in the investigated ZnO microparticles as ~ 1018 cm−3.

Paramagnetic centers in graphite oxide according to EPR spectra and DFT calculations

The first attempt to determine experimentally based structures of paramagnetic centers in graphite oxide is presented. The attribution of satellite signals in electron paramagnetic resonance (EPR) spectra to forbidden transitions was proved by X- and Q-band experiments. Proton responsible for satellite lines is found to be able to undergo rapid isotopic exchange. Magnetic parameters of the paramagnetic centers were determined by numerical simulation of experimental EPR spectra. Density functional theory (DFT) was used for theoretical calculation of magnetic parameters for probable paramagnetic moieties. The comparison of DFT results with experimental values reveals the structures of oxygen-centered paramagnetic species in graphite oxide.

Analysis of radical formation by EPR in complex starch-protein-lipid model systems and corn extrudates.

The formation of short-lived and stable radicals was investigated using electron paramagnetic resonance (EPR) spectroscopy and compared with hydroperoxides and hexanal in complex starch-protein-lipid model systems, as well as in corn extrudates. Stable radicals were detected directly in ground samples. Short-lived lipid radicals were measured ex situ in ethyl acetate extracts of model systems and extrudates by the use of the spin trap PBN. Significant adduct formation was found after 30min at 50°C. During storage, lipid radicals (PBN adducts) increased in model systems. Simulation of EPR spectra from bulk oil demonstrated that mainly alkoxyl radical adducts were detected, to which rapidly decomposing peroxyl radical adducts also contributed. Stable radicals in extrudates were attributed to protein radicals based on g-value of 2.00467 compared with 2.00474 found in model system prepared with zein. The signal intensity of the stable radical remained constant during storage, but increased during extrusion.

Modeling of motional EPR spectra using hindered Brownian rotational diffusion and the stochastic Liouville equation.

Electron paramagnetic resonance (EPR) spectra of molecular spin centers undergoing reorientational motion are commonly simulated using the stochastic Liouville equation (SLE) with a rigid-body hindered Brownian diffusion model. Current SLE theory applies to specific spin systems such as nitroxides and to high-symmetry orientational potentials. In this work, we extend the SLE theory to arbitrary spin systems with any number of spins and any type of spin Hamiltonian interaction term, as well as to arbitrarily complex orientational potentials. We also examine the limited accuracy of the frequency-to-field conversion used to obtain field-swept EPR spectra and present a more accurate approach. The extensions allow for the simulation of EPR spectra of all types of spin labels (nitroxides, copper2+, and gadolinium3+) attached to proteins in low-symmetry environments.

Simulating Frequency-Domain Electron Paramagnetic Resonance: Bridging the Gap between Experiment and Magnetic Parameters for High-Spin Transition-Metal Ion Complexes.

We present a comparison of experimental and simulated frequency- and field-domain electron paramagnetic resonance (EPR) spectra of integer and half-integer high-spin transition-metal ion complexes. For the simulation of EPR spectra a new tool within the EPR simulation software EasySpin is introduced, which allows for field- and frequency-domain EPR simulations with the same theoretical model and the same set of spin Hamiltonian parameters. The utility of this approach is demonstrated on the integer-spin complexes NiBr2(PPh3)2 and [Tp2Mn]SbF6 (both S = 1) and the half-integer-spin Fe(III) porphyrins, hemin (Fe(PPIX)Cl) and Fe(TPP)Cl (both S = 5/2). We demonstrate that the combination of field- and frequency-domain EPR techniques allows the determination of spin Hamiltonian parameters, in particular large zero-field splittings, with high accuracy.

EMR Study and DFT-Assisted Identification of Transient Radicals in X-Irradiated Crystalline Sucrose.

Solid-state sucrose is a well-known dosimetric system, which is capable of reliable dose estimates only at a considerable time after exposure. Immediately after irradiation at room temperature, its electron paramagnetic resonance (EPR) spectrum is dominated by contributions from unstable radicals, which are studied here using continuous-wave EPR and electron-nuclear double resonance (ENDOR) spectroscopy. Four hyperfine tensors of proton couplings were determined, associated with two radical species, and subsequently compared to density functional theory calculation results, which led to the identification of the species with lower abundance (U2) as a radical formed by a H abstraction from C4. The more abundant center (U1) has not been definitively identified yet, but we present compelling evidence that it should be a C6 centered radical. Comparison of the simulated EPR spectra with all available data to the experimental ones suggests that the EPR spectrum of X-irradiated sucrose immediately after irradiation can now be almost entirely understood.

Simulation of multi-frequency EPR spectra for a distribution of the zero-field splitting

We present a numerical procedure called ‘grid-of-errors’ to extract the distribution of magnetic interactions from continuous-wave electron-paramagnetic-resonance (EPR) spectra at multiple microwave frequencies. The approach is based on the analysis of the lineshape of the spectra and explicitly worked out for high-spin systems for which the lineshape is determined by a distribution of the zero-field splitting. Initial principal values of the zero-field splitting tensor are obtained from the EPR spectrum at a microwave frequency in the high-field limit, and the initial distribution is taken Gaussian. Subsequently, the grid-of-errors procedure optimizes this distribution, without any restriction to its shape, taking into account spectra at various microwave frequencies. The numerical procedure is illustrated for the Fe(III)–EDTA complex. An optimized distribution of the zero-field splitting is obtained, which provides a proper description of the EPR spectra at 9.5, 34, 94, and 275GHz. The proposed approach can be used as well for distributions of magnetic interactions other than the zero-field splitting.

Quantitative Interpretation of Multifrequency Multimode EPR Spectra of Metal Containing Proteins, Enzymes, and Biomimetic Complexes.

Electron paramagnetic resonance (EPR) spectroscopy has long been a primary method for characterization of paramagnetic centers in materials and biological complexes. Transition metals in biological complexes have valence d-orbitals that largely define the chemistry of the metal centers. EPR spectra are distinctive for metal type, oxidation state, protein environment, substrates, and inhibitors. The study of many metal centers in proteins, enzymes, and biomimetic complexes has led to the development of a systematic methodology for quantitative interpretation of EPR spectra from a wide array of metal containing complexes. The methodology is now contained in the computer program SpinCount. SpinCount allows simulation of EPR spectra from any sample containing multiple species composed of one or two metals in any spin state. The simulations are quantitative, thus allowing determination of all species concentrations in a sample directly from spectra. This chapter will focus on applications to transition metals in biological systems using EPR spectra from multiple microwave frequencies and modes.

Perifosine induced release of contents of trans cell-barrier transport efficient liposomes

Perifosine (OPP) containing liposomal formulation was previously found to deliver almost half of liposome encapsulated content through a tight cellular barrier in vitro. In order to understand the role of different liposome components, especially perifosine, in transendothelial transport the physical characteristics of liposome membranes composed of phosphatidylcholine, and cholesterol, as a main lipid constituents, and variable amount of helper lipids: dioleoyl phosphatidylethanolamine (DOPE), and alkylphospholipid perifosine.For this purpose, electron paramagnetic resonance (EPR) with computer aided EPR spectra simulation and fluorescence polarization spectroscopy were used to investigate how different membrane components influence membrane characteristics and the release of liposome entrapped substances. Beside methylester of palmitic acid with nitroxide group at different position on acyl chain usually used for such studies, the spin labeled and fluorescent labeled analog of perifosine were introduced.OPP increases membrane fluidity of liposomes as well as the release of liposome encapsulated content. The release of neutral molecules increases with OPP concentration, while the release of charged molecules is about an order of magnitude slower. Optimal OPP concentration, for release of charged molecules, is about 15mol%.These results are one step further toward the conclusion that the lysolipid-containing liposomes could be promising trans endothelial delivery system, since lysolipids, such as OPP, open tight cellular barriers, as was published before, and in the same time induce the release of liposome encapsulated content at physiological temperature, as shown here. Since many drug delivery systems are being developed, which mainly exploit the transcellular route of delivery through barrier-forming cells, we hope that the uniqueness of lysolipid-containing liposomes, exploiting the paracellular route, and thus avoiding efflux transporters, will foster further research in formulating other lysolipid-containing liposomes as drug delivery systems.

Speciation of Cr(iii) in intermediate phases during the sol–gel processing of Cr-doped SrTiO3powders

The local environment of Cr3+ during the sol–gel synthesis of 1% and 5% Cr3+-doped SrTiO3 (Cr3+:SrTiO3) bulk powders was systematically studied by structural characterization techniques and dopant-specific spectroscopies. After calcination at 800 °C, the precursors were annealed between 850 °C and 1050 °C for up to 6 h. We observe the formation of numerous phases in addition to the final product of SrTiO3. One of these is a metastable Ruddelsden–Popper phase (Sr2TiO4) that forms when the precursor is annealed at 1050 °C for less than 2 h. Electron paramagnetic resonance (EPR) spectroscopy reveals a new signal that correlates with appearance of the Sr2TiO4 phase and is consistent with Cr3+ substitution into the axially-compressed Ti4+ site in Sr2TiO4. The best agreement between experiment and the simulated EPR spectra of Cr3+:Sr2TiO4 is when |D| = 0.0207 cm−1, g∥ = 1.9803 and g⊥ = 1.9793. The majority of the sample is converted to Cr3+:SrTiO3 with increasing annealing times at 1050 °C as detected by low-temperature emission and EPR spectroscopies, and powder X-ray diffraction.

Simulation of electron paramagnetic resonance spectra of spin-labeled molecules from replica-exchange molecular dynamics

We report a general approach for the simulation of the electron paramagnetic resonance (EPR) spectra of spin labels attached to peptides and proteins directly from replica-exchange molecular dynamics (REMD) trajectories. Conventional MD trajectories are generally inadequate for the prediction of EPR line shapes since the label can become trapped in one or more of a set of rotameric states, thus preventing both conformational sampling and accurate estimates of the exchange rates between different rotamers. The advantage of using REMD is its ability to provide both efficient conformational sampling and kinetic information for spin-label dynamics. Our approach is illustrated with spin-labeled peptide. This approach to REMD-EPR simulation paves the way for the wider application of MD modeling to the simulation and interpretation of EPR spectra of spin-labeled molecules.

Interaction of Antidepressant Drug, Clomipramine, with Model and Biological Stratum Corneum Membrane as Studied by Electron Paramagnetic Resonance

The interactions of tricyclic antidepressant drug, clomipramine (CLO), with pig ear stratum corneum (SC) and model membranes were investigated by electron paramagnetic resonance (EPR) spin labeling to get some insight into the possible application of this drug in transdermal delivery. The changes in membrane characteristics caused by CLO in the regions that are close to the water-lipid interfaces and the central parts of the membranes were searched. The experimental results were supported by computer simulation of EPR spectra, which showed heterogeneity of the membranes composed of regions with different fluidity characteristics. CLO was effective in both parts of the layers, indicating intercalation of the drug into model membranes as well as into the pig ear SC. Introduction of various molar ratios of CLO caused a decrease in the order parameter and an increase in the rotational diffusion of nitroxide moiety in different membrane regions as well as an increase in the polarity of spin probe environment. It also changed the number of resolved spectral components, which reflects the heterogeneity of the membrane. The fluidizing effect of CLO on pig ear SC throughout the whole membrane layers indicates that CLO penetrates into the SC, which is important for its transdermal delivery.

Coordination and RedOx ratio of iron in sodium‐silicate glasses

Mössbauer and Electron paramagnetic resonance (EPR) spectroscopy studies of iron status in doped sodium-silicate glasses (100−x, mol%)Na2O(x, mol%)SiO2(y, wt.%) Fe2O3 (x=75; 81; y=0.05–13) were made as a function of glass matrix and total iron content.55Total iron content is expressed as Fe2O3 in the paper. Mössbauer spectroscopy identified three signals: Fe3+ doublet, Fe2+ doublet and Fe3+ broad hyperfine splitting (HFS) at room temperature dealing with slow relaxation effect of ferric ions. X-band EPR spectra exhibits resonance signals at g≈2.0, g≈4.3 and a shoulder in the region of g≈6.0. EPR spectra were simulated on the basis of rhombic spin-Hamiltonian with Zeeman and fine-structure terms. The nature of EPR absorption signals was interpreted as a combination of five types of Fe3+ complexes. Three of them were clustered ferric ions and two others were isolated paramagnetic ferric ions in distorted octahedral sites. The iron RedOx ratio (Fe2+/Fe3+) has been estimated from Mössbauer spectroscopy, while proportion of Fe3+ ions in adjacent sites (Fe3+isolated)/Fe3+clustered) has been obtained from Mössbauer spectra and simulated EPR spectra.

On the interaction of bovine serum albumin with ionic surfactants: Temperature induced EPR changes of a maleimide nitroxide reflect local protein dynamics and probe solvent accessibility

The interaction of bovine serum albumin (BSA) with the ionic surfactants sodium dodecylsulfate (SDS, anionic), cetyltrimethylammonium chloride (CTAC, cationic) and N-hexadecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (HPS, zwitterionic) was studied by electron paramagnetic resonance (EPR) spectroscopy of spin label covalently bound to the single free thiol group of the protein. EPR spectra simulation allows to monitor the protein dynamics at the labeling site and to estimate the changes in standard Gibbs free energy, enthalpy and entropy for transferring the nitroxide side chain from the more motionally restricted to the less restricted component. Whereas SDS and CTAC showed similar increases in the dynamics of the protein backbone for all measured concentrations, HPS presented a smaller effect at concentrations above 1.5 mM. At 10 mM of surfactants and 0.15 mM BSA, the standard Gibbs free energy change was consistent with protein backbone conformations more expanded and exposed to the solvent as compared to the native protein, but with a less pronounced effect for HPS. In the presence of the surfactants, the enthalpy change, related to the energy required to dissociate the nitroxide side chain from the protein, was greater, suggesting a lower water activity. The nitroxide side chain also detected a higher viscosity environment in the vicinity of the paramagnetic probe induced by the addition of the surfactants. The results suggest that the surfactant–BSA interaction, at higher surfactant concentration, is affected by the affinities of the surfactant to its own micelles and micelle-like aggregates. Complementary DLS data suggests that the temperature induced changes monitored by the nitroxide probe reflects local changes in the vicinity of the single thiol group of Cys-34 BSA residue.

Paramagnetic Oxotungsten(V) Complexes Containing the Hydrotris(3,5-dimethylpyrazol-1-yl)borate Ligand

Sky-blue Tp*WOCl(2) has been synthesized from the high-yielding reaction of Tp*WO(2)Cl with boron trichloride in refluxing toluene. Dark-red Tp*WOI(2) was prepared via thermal decarbonylation followed by aerial oxidation of Tp*WI(CO)(3) in acetonitrile. From these precursors, an extensive series of mononuclear tungstenyl complexes, Tp*WOXY [X = Cl(-), Y = OPh(-), SPh(-); X = Y = OPh(-), 2-(n-propyl)phenolate (PP(-)), SPh(-), SePh(-); XY = toluene-3,4-dithiolate (tdt(2-)), quinoxaline-2,3-dithiolate (qdt(2-)), benzene-1,2-diselenolate (bds(2-)); Tp* = hydrotris(3,5-dimethylpyrazol-1-yl)borate], was prepared by metathesis with the respective alkali-metal salt of X(-)/XY(2-) or (NHEt(3))(2)(qdt). The complexes were characterized by microanalysis, mass spectrometry, electrochemistry, IR, electron paramagnetic resonance (EPR), and electronic absorption spectroscopies, and X-ray crystallography (for X = Y = OPh(-), PP(-), SPh(-); XY = bds(2-)). The six-coordinate, distorted-octahedral tungsten centers are coordinated by terminal oxo [W≡O = 1.689(6)-1.704(3) Å], tridentate Tp*, and monodentate or bidentate O/S/Se-donor ligands. Spin Hamiltonian parameters derived from the simulation of fluid-solution X-band EPR spectra revealed that the soft-donor S/Se ligand complexes had larger g values and smaller (183)W hyperfine coupling constants than the less covalent hard-donor O/Cl species. The former showed low-energy ligand-to-metal charge-transfer bands in the near-IR region of their electronic absorption spectra. These oxotungsten(V) complexes display lower reduction potentials than their molybdenum counterparts, underscoring the preference of tungsten for higher oxidation states. Furthermore, the protonation of the pyrazine nitrogen atoms of the qdt(2-) ligand has been examined by spectroelectrochemistry; the product of the one-electron reduction of [Tp*WO(qdtH)](+) revealed usually intense low-energy bands.
