Insecticidal Properties and Chemical Characterization of Laurus nobilis L. Essential Oils from Two Regions of Morocco against Callosobruchus maculatus (Coleoptera: Bruchinae)

Morocco is a significant botanical reservoir that boasts a wealth of raw materials with promising applications across various industrial sectors, notably in pharmaceuticals and food. The objective of this study was to assess the effectiveness of essential oils (EOs) derived from Laurus nobilis L. leaves originating from the Tanger (EOT) and Meknes (EOM) regions in combating Callosobruchus maculatus infection. The chemical compositions of these oils were examined using Fourier transform infrared (FTIR) spectroscopy and gas chromatography–mass spectrometry (GC-MS). The biological activity of the EOs was evaluated via repulsion and fumigation tests against C. maculatus at varying concentrations. FTIR analysis revealed distinct vibrational bands indicative of various chemical compounds. GC-MS analysis was used to delineate the major chemical constituents of the EOs. The three predominant compounds in the EOT were 1,8-cineole (37.64%), linalool (16.40%), and adamantane (12.00%), whereas 1,8-cineole (47.84%), toluene (17.60%), and α-phellandrene (8.44%) were the most abundant in the EOM. Notably, the EOs exhibited significant repellent activity against C. maculatus, with repulsion percentages ranging from 51.11 to 90.00% in Tanger and 67.78 to 93.33% in Meknes. Mortality rates varied from 0 to 100% depending on the treatment. However, the mean concentrations showed mortality rates ranging from 29.44 to 65.56% for the EOT and from 21.11 to 67.78% for the EOM, with LD50 values of 11.96 μL/L and 5.22 μL/L. Docking studies revealed that 1,8-cineole had the highest binding affinity for the active site of acetylcholinesterase, thus confirming its toxic activity against C. maculatus. The findings of this study highlight the ability of EOs extracted from L. nobilis in the Moroccan regions of Tanger and Meknes to act as effective insecticides and repellents against C. maculatus, thereby highlighting avenues for further exploration of pest management and agricultural practices.

Behavioral preferences of Solenopsis invicta Buren to essential oils and active compounds from amiaceae plants

The red imported fire ant (RIFA) (Solenopsis invicta Buren) has caused serious ecological and public safety problems in invasive areas. While extensive use of pesticides to control RIFA may negatively affect human health and environment, essential oils (EOs) of Lamiaceae possessing aromatic smell and multiple biological activities are potential alternatives of chemical pesticides. In this study, we evaluated the behavioral preferences of different castes of RIFA towards the EOs from five species of Lamiaceae plants (Salvia miltiorrhiza, Perilla frutescens, Pogostemon cablin, Schizonepeta tenusfolia, Mentha canadensis), and then investigated the active compounds in these EOs that trigger insect behavioral preferences by gas chromatography-electroantennographic detection (GC-EAD). The results of behavioral experiment indicated that three EOs strongly altered the behavior of RIFA. Notably, all concentrations of P. frutescens EO showed a significant repellent activity on virgin queen ants, with a repellent rate of 62.50% at the lowest concentration of 2 μg/ml. The repellent effect of S. tenusfolia EO against small worker ants reached a very significant difference between control and treatment groups, with the highest repellent rate of 85.00% observed at 1000 μg/ml. Save for the lowest concentration 2 μg/ml, other doses of M. canadensis EO showed a remarkable repellent effect on small worker ants, demonstrating a positive dose-effect relationship (with repellent rates of 77.50% and 65.00% at 1000 and 8 μg/ml, respectively). A total of 39 EAD-active compounds were detected in these EOs, and 12 of them were subsequently evaluated for behavioral preferences of RIFA. Our findings indicated that six compounds (menthone, menthol, caryophyllene oxide, pulegone, 3-methylcyclopentanol, eugenol) consistently triggered behavioral preferences in worker ants. Meanwhile, six compounds, including borneol, eugenol, phenylethyl alcohol, perilla ketone, 3-methylcyclopentanol, α-guaiene, were found to significantly alter the behavioral selectivity of virgin queen ants. These results may help to develop application value of selected Lamiaceae plants EOs and contribute to the exploration of new green control methods for invasive pests.

Functional analysis of three odorant receptors in Plutella xylostella response to repellent activity of 2,3-dimethyl-6-(1-hydroxy)-pyrazine

Plutella xylostella is an important pest showing resistance to various chemical pesticides, development of botanical pesticides is an effective strategy to resolve above problem and decrease utilization of chemical pesticides. Previous study showed that 2,3-dimethyl-6-(1-hydroxy)-pyrazine has significant repellent activity to P. xylostella adult which mainly effect to the olfactory system, however the molecular targets and mechanism are still unclear. Based on the RNA-Seq and RT-qPCR data, eight ORs (Odorant receptor) in P. xylostella were selected as candidate targets response to repellent activity of 2,3-dimethyl-6-(1-hydroxy)-pyrazine. Here, most of the ORs in P. xylostella were clustered into three branches, which showed similar functions such as recognition, feeding, and oviposition. PxylOR29, PxylOR31, and PxylOR46 were identified as the potential molecular targets based on the results of repellent activity and EAG response tests to the adults which have been injected with dsRNA, respectively. Additionally, the three ORs were higher expressed in antenna of P. xylostella, followed by those in the head segment. Furthermore, it was found that the bindings between these three ORs and 2,3-dimethyl-6-(1-hydroxy)-pyrazine mainly depend on the hydrophobic effect of active cavities, and the binding to PxylOR31 was more stabler and easier with an energy of −16.34 kcal/mol, together with the π-π T-shaped interaction at PHE195 site. These findings pave the way for the complete understanding of pyrazine repellent mechanisms.

Citrus Essential Oils as Potential Insecticides Against Thrips flavus

Background: Essential oils (EOs) are recognized for their potent insecticidal activity and are widely considered promising agents for pest control. Methods: The insecticidal activity of seven commercial citrus EOs against the global phytophagous pest Thrips flavus was evaluated using bioassays conducted under laboratory conditions. Moreover, the chemical composition of these EOs was analyzed using gas chromatography-mass spectrometry (GC-MS). Results: Among the 45 identified compounds, the major constituents were d-limonene (23.77%– 95.10%), methyl jasmonate (38.76%), and linalyl acetate (34.55%). Orange flower oil, sweet orange oil, tangerine peel oil, bergamot oil, lime oil, lemon oil, and grapefruit oil exhibited good insecticidal activity against T. flavus without demonstrating significant repellent activity. Orange flower oil and sweet orange oil showed higher insecticidal toxicity, with LC50 values of 0.20 g/L and 0.37 g/L, respectively. In pot experiments, the higher control efficacy against T. flavus was found after 7 days of treatment for orange flower oil (91.14±1.27% at 720.00 g a.i.·hm−2 and 100% at 900.00 g a.i.·hm−2), sweet orange oil (91.14±5.52% at 900.00 g a.i.·hm−2), tangerine peel oil (96.20±2.19% at 720 g a.i.·hm−2 and 100% at 900.00 g a.i.·hm−2), lemon oil (96.20±2.19% at 900 g a.i.·hm−2), and lime oil (97.47±2.53% at 900.00 g a.i.·hm−2). Conclusion: Taken together, orange flower oil and tangerine peel oil, which were found to contain d-limonene and methyl jasmonate, demonstrated rapid and more effective insecticidal activity compared to the other EOs tested, which makes these two EOs promising alternatives to chemical insecticides.

Development of terpenoid repellents against Aedes albopictus: a combined study of biological activity evaluation and computational modelling

ABSTRACT To explore novel terpenoid repellents, 22 candidate terpenoid derivatives were synthesized and tested for their electroantennogram (EAG) responses and repellent activities against Aedes albopictus. The results from the EAG experiments revealed that 5-(2-hydroxypropan-2-yl)-2-methylcyclohex-2-en-1-yl formate (compound 1) induced distinct EAG responses in female Aedes albopictus. At concentrations of 0.1, 1, 10, 100, and 1000 mg/L, the EAG response values for compound 1 were 179.59, 183.99, 190.38, 193.80, and 196.66 mV, demonstrating comparable or superior effectiveness to DEET. Repellent activity analysis indicated significant repellent activity for compound 1, closest to the positive control DEET. The in silico assessment of the ADMET profile of compound 1 indicates that it successfully passed the ADMET evaluation. Molecular docking studies exhibited favourable binding of compound 1 to the active site of the odorant binding protein (OBP) of Aedes albopictus, involving hydrophobic forces and hydrogen bond interactions with residues in the OBP pocket. The QSAR model highlighted the influential role of hydrogen-bonding receptors, positively charged surface area of weighted atoms, polarity parameters of molecules, and maximum nuclear-nuclear repulsion force of carbon-carbon bonds on the relative EAG response values of the tested compounds. This study holds substantial significance for the advancement of new terpenoid repellents.

Identification and characterization of odorant receptors in Plutella xylostella antenna response to 2,3-dimethyl-6-(1-hydroxy)-pyrazine

Diamondback moth (Plutella xylostella), a worldwide migratory pest that is developing strong resistance to various chemical insecticides. It has been determined that four natural pyrazines isolated from Allium tuberosum showed significant repellent activity to P. xylostella, but the molecular target still unknown. In the present study, a novel synthetic route for 2,3-dimethyl-6-(1-hydroxy)-pyrazine which has the most significant repellent activity with a purity of 90.60% was established. Simultaneously, the bioassay result declared that the repellent grade was IV at a dosage of 0.01 mg which was the same as to the published data. Transcriptomics analysis detected 1643 upregulated and 3837 downregulated genes in P. xylostella antennae following this pyrazine exposure. Then, 2142 differentially expressed genes were annotated using Gene Ontology and 2757 genes were annotated by Kyoto Encyclopedia of Genes and Genomes. Moreover, this procedure identified 84 odour perception-related genes, 58 odorant receptor (OR) genes including 57 conventional ORs and the odorant receptor co-receptor (Orco, atypical odorant receptor) gene, and 26 odorant-binding protein (OBP) genes. Based on quantitative real time PCR (RT-qPCR) and differential expression results, 9 OR genes including the Orco were cloned and characterised. In summary, this study provides important basis for the utilization of pyrazines as the main active ingredients or lead compounds to developing new botanical pesticides, which will reduce application of chemical pesticides and postpone the development of resistance.

Synthesis of Carvacrol-Loaded Invasomes Nanoparticles Improved Acaricide Efficacy, Cuticle Invasion and Inhibition of Acetylcholinestrase against Hard Ticks.

Carvacrol is a monoterpenoid phenol found in many essential oils that has antibacterial, antifungal and antiparasitic activities. Drug loaded-invasome systems are used to deliver drugs utilizing nanoparticles to improve bioavailability, efficacy, and drug release duration. As a result, the present study developed carvacrol-loaded invasomes and evaluated their acaricidal effect against Rhipicephalus annulatus (cattle tick) and Rhipicephalus sanguineus (dog tick). Carvacrol loaded-invasome (CLI) was prepared and characterized using UV/Vis spectrophotometer, zeta potential measurements, Scanning Transmission Electron Microscopy (STEM), Fourier Transform Infrared (FT-IR) Spectroscopy, and Differential Scanning Calorimetry Analysis. CLI (5%) induced significant mortality (100%) in R. annulatus adult ticks with LC50 of 2.60%, whereas the LC50 of pure carvacrol was 4.30%. Carvacrol and CLI were shown to have a significant larvicidal action on both tick species, with LC50s of 0.24 and 0.21% against R. annulatus and 0.27 and 0.23% against R. sanguineus, respectively. Carvacrol and CLI (5%) induced significant repellent activities for 24 h against R. annulatus and R. sanguineus, as evidenced by the rod method and the petri-dish selective area choice method, respectively. High-performance liquid chromatography (HPLC) demonstrated that the CLI form had 3.86 times the permeability of pure carvacrol. Moreover, carvacrol and CLI inhibited acetylcholinesterase activity and decreased glutathione and malonedealdehyde levels in the treated ticks. In conclusion, invasomes significantly improved adulticidal and repellency activities of carvacrol against both tick species.

Role of plant essential oil nanoemulsions on host colonization by the invasive ambrosia beetle Xylosandrus compactus

The development of effective control strategies targeting non-native ambrosia beetles (Coleoptera: Curculionidae: Scolytinae and Platypodinae) represents an emerging challenge. This is due both to their rapid spread into climatically suitable newly invaded regions rich in susceptible host plants and to the limited knowledge concerning sustainable management tools. In this context, the use of botanical insecticides, such as plant essential oils (EOs), is considered a promising option in Integrated Pest Management (IPM) strategies as an alternative to conventional insecticides for sustainable crop protection. Here, the impact of five EO nanoemulsions (Rosmarinus officinalis L. cultivar verbenone, Carlina acaulis L., Laurus nobilis L., Cupressus sempervirens L., and Betula alba L.) on the choice behavior, host colonization and progeny production by the invasive ambrosia beetle Xylosandrus compactus (Eichhoff) was evaluated under laboratory and semi-field conditions. The chemical composition of tested EOs was firstly evaluated by means of gas chromatography-mass spectrometry (GC-MS) analyses. Then, EO nanoemulsions were prepared and characterized through dynamic light scattering (DLS). Tested EO nanoemulsions showed single or bimodal droplet size distributions in the nanometric range. Results highlighted a significant repellent activity of R. officinalis cv. verbenone and C. acaulis EO nanoemulsions (at 3% EO concentration). However, the trend for repellence of the R. officinalis EO against the beetle was not confirmed at decreasing EO concentrations (i.e., 1% and 0.5%). Nanoemulsions based on C. acaulis EO significantly affected the beetle choices regardless of the tested EO concentration. Indeed, in no choice bioassays, a significantly lower number of individuals started infesting stems treated with these nanoemulsions. On the other hand, no effect of the EO treatment was found on the number of offspring produced by survived beetle females. The repellence by R. officinalis and C. acaulis EOs was confirmed in semi-field conditions. Overall, our findings provide new insights toward the use of R. officinalis and C. acaulis EOs as repellents of X. compactus, suggesting their future incorporation in non-chemical strategies, such as ‘push-pull’, as part of sustainable IPM packages targeting this invasive pest.

Essential Oil Composition of Seven Bulgarian Hypericum Species and Its Potential as a Biopesticide.

Hypericum species and especially H. perforatum L. are well known for their therapeutic applications. The present study assessed the essential oil (EO) composition, and antifungal and aphid suppression activity of seven Bulgarian Hypericum species. The EOs were analyzed by GC-MS-FID. Two experiments were conducted. In the first experiment, H. perforatum, H. maculatum, and H. hirsutum were used. Additionally, the EO composition of H. perforatum extracted via hydrodistillation (ClevA) and via commercial steam distillation (Com) were compared. The second experiment compared the EOs of H. perforatum, H. cerastoides, H. rumeliacum, H. montbretii, and H. calycinum (flowers and leaves) extracted via hydrodistillation and collected with n-hexane. Overall, the EO constituents belonged to four classes, namely alkanes, monoterpenes, sesquiterpenes, and fatty acids. The main class for compounds in H. maculatum and H. perforatum (section Hypericum) were sesquiterpenes for both experiments except for H. perforatum (Com). Hypericum montbretii (section Drosocarpium) EO had monoterpenes (38.09%) and sesquiterpenes (37.09%) as major groups, while H. hirsutum EO (section Taeniocarpium) contained predominately alkanes (67.19%). Hypericum hirsutum EO contained cedrol (5.04%), found for the first time in Hypericum species. Fatty acids were the main compounds in H. cerastoides (section Campylopus), while monoterpenes were the most abundant class in H. rumeliacum and H. calycinum EOs. α-Pinene and germacrene D were the major EO constituents of all analyzed Hypericum species except for H. hirsutum and H. cerastoides. Hypericum perforatum EO (Com) had significant repellent and insecticidal activity against two aphid species, Rhopalosiphum padi (Bird Cherry-oat aphid) and Sitobion avenae (English grain aphid) at concentrations of 0%, 1%, 2.5%, 3.5%, 4.5%, and 5%. The tested EOs did not show significant activity against selected economically important agricultural fungal pathogens Fusarium spp., Botrytis cinerea, Colletotrichum spp., Rhizoctonia solani, and Aspergillus sp. The EO of the Hypericum species found in the Bulgarian flora could be utilized for the development of new biopesticides for aphid control.

Larvicidal and repellent potential of Ageratum houstonianum against Culex pipiens.

Mosquitoes are unquestionably the most medic arthropod vectors of disease.Culex pipiens, usually defined as a common house mosquito, is a well-known carrier of several virus diseases. Crude ethanol extracts of different organs ofAgratum houstonianumare tested withCulex pipiensLinnaeus (Diptera: Culicidae) to determine their larvicidal, antifeedant, and repellency effects. Alongside biochemical analysis, the activity of the AChE, ATPase, CarE, and CYP-450 is detected in the total hemolymph of theC. pipienslarvae to examine the enzymatic action on the way to explain their neurotoxic effect and mode of action. Through HPLC and GC-MS analysis of the phytochemical profile ofA. houstonianumaerial parts is identified. The larvicidal activity of aerial parts; flower (AF), leaf (AL), and stem (AS) ofA. houstonianumextracts are evaluated against the 3rd instar larvae ofC. pipiensat24-, 48- and 72-post-treatment.A. houstonianiumAF, AL, and AS extracts influenced the mortality of larvae with LC50 values 259.79, 266.85, and 306.86ppm, respectively after 24h of application. The potency of AF and AL extracts was 1.69- and 1.25-folds than that of AS extract, respectively. A high repellency percentage was obtained by AF extract 89.10% at a dose of 3.60mg/cm2.A. houstonianiumAF prevailed inhibition on acetylcholinesterase and decrease in carboxylesterase activity. Moreover, a significant increase in the ATPase levels and a decrease in cytochrome P-450 monooxegenase activity (- 36.60%) are detected. HPLC analysis prevailed chlorogenic and rosmarinic acid as the major phenolic acids in AL and AF, respectively. GC-MS analysis ofA. houstonianumresults in the identification of phytol as the major makeup. Precocene I and II were detected in AF. Linoleic, linolenic, and oleic acid were detected in comparable amounts in the studied organs. Overall, results suggest that the A. houstonianum flower extract (AF) exhibits significant repellent, antifeedant, and larvicidal activities.

Chemical characterisation, insecticidal and antioxidant activities of essential oils from four Citrus spp. fruit peel waste

Citrus peel is a major fruit waste generated during the processing of citrus fruits. We extracted and chemically characterised essential oil from four different species of citrus peel waste and evaluated its insecticidal, antioxidant and phytotoxic activities. GC-MS/MS characterisation revealed D-limonene as the major constituent of citrus peel waste essential oil. The essential oils from the four citrus peel wastes showed significant contact, fumigant and repellent activities against two key stored grain pests Tribolium castaneum (Herbst) and Callosobruchus chinensis (L.). DPPH, ABTS, and FRAP assays revealed the antioxidant potential of the citrus peel waste essential oil. The essential oil did not affect the germination of wheat seeds and revealed the safety of essential oil to germinating seeds. Hence, the essential oil extracted from citrus peel waste can be valorised and utilised as a botanical insecticide against T. castaneum and C. chinensis.

Odorant-Binding Protein 3-Oriented Rational Design and Discovery of Novel Jasmonate Derivatives as Potential Aphid-Repellent Agents.

Odorant-binding protein (OBP) is a potential target for developing insect behavior control agents due to its properties in transporting semiochemicals. In this study, 12 novel jasmonic acid (JA) derivatives were rationally designed and synthesized based on the binding features between Acyrthosiphon pisum OBP3 (ApisOBP3) and compound D1 [(E)-3,7-dimethylocta-2,6-dien-1-yl 2-(3-oxo-2-pentylcyclopentyl)acetate] with a binding affinity (Kd) of 26.79 μM. Most novel JA derivatives displayed better binding affinities than D1 (Kd = 1-26 μM). Among them, compound 6b [(E)-3,7-dimethylocta-2,6-dien-1-yl-2-((Z)-3-((acryloyloxy)imino)-2-pentylcyclopentyl)acetate] is the most promising compound with an excellent Kd of 1.33 μM and a significant repellent activity with repellent rates of 50-60% against A. pisum and Myzus persicae. Both hydrophobic and electrostatic interactions were found to contribute significantly to the binding of 6b to ApisOBP3. This study provides significant guidance for the rational design and efficient identification of novel aphid repellents based on aphid OBPs.

Development of a multilayer insect-repelling film with trans-anethole for the storage of almond flake cereal.

Trans-anethole (AN), which exhibits strong insect-repellent activity against Plodia interpunctella larvae, was applied on a polyethylene terephthalate (PET) film as an active packaging coating layer. All developed films at different concentrations (25%, 30%, 40%, and 50%) exhibited significant insect repellent activities. However, these films did not significantly differ from the control film in terms of color and transparency. In addition, the developed polypropylene (PP) and PET laminated films containing 25% AN (PP/AN25/PET) exhibited strong and continuous insect-repellent activity for up to 42 days. Finally, the developed film showed 2.86-fold stronger repellent activity than that of the control film when applied to the almond flake cereals packaging. These results suggest that PP/AN25/PET could be used as a potent insect-repelling packaging film in a realistic grain-packaging system. PRACTICAL APPLICATION: A PP/trans-anethole/PET film that exhibited good insect-repellent activity for 42 days was newly developed in this study. As it showed strong insect repellency, especially in almond flake cereals packaging, it is expected to have high potential as an insect-repelling grain-packaging film.

Biological activities and gene expression of detoxifying enzymes in Tribolium castaneum induced by Moutan cortex essential oil

ABSTRACT Tribolium castaneum is one of the most harmful storage pests in the world. The aim of this study was to determine the chemical composition, repellent, and contact activities of Moutan cortex essential oil against this insect pest. In addition, the effects of Moutan cortex were examined on the expressions of three major detoxifying enzyme genes in T. castaneum. Four components were identified in this essential oil by gas chromatography-mass spectrometry (GC-MS), which was predominantly paeonol (99.13%). Paeonol exerted significant repellent activity against T. castaneum, which was more potent than the positive control N.N-diethyl-meta-toluamide (DEET). The most significant contact toxicity was observed at 24 h after exposure to paeonol. Further, quantitative real-time PCR (qRT-PCR) was used to assess expression changes in three detoxification enzyme genes in T. castaneum, including carboxylesterase (CarE), glutathione S-transferase (Gst) and cytochrome P4506BQ8 (Cyp6bq8). Among these, Gst was most highly up-regulated after treatment with paeonol with the highest expression level of 4.9-fold (Rps18 as internal reference gene) greater than control at 24 h following treatment. Data indicated that Gst might play a critical role in metabolic detoxification of toxic xenobiotics. Taken together, our findings might lay a foundation for development of paeonol as a potential natural repellent or pesticide to control storage pests.

Chirality Effects on Repellent Properties of 4-Alkoxycoumarins Against Asian Tiger Mosquito (Diptera: Culicidae)

The use of semiochemicals as repellents and attractants has been proposed to complement insecticides used for the control of vector mosquito populations. In several studies, the optical purities of the molecules tested have been described as having little or no effect on repellent activity. However, these observations seem difficult to explain because of the chirality effect of molecules on the olfactory system of insects and humans. Thus, the purpose of this study is to assess the effects of chirality on the repellent properties of 4-alcoxycoumarins against Aedes albopictus Skuse, mosquito vector of arboviruses. We report here that the racemic (R/S)-4-sec-butoxycoumarin had the highest repellent effect (Repellent Index = 49.9%) followed by (R) enantiomer (Repellent Index = 24.2%) for the dose of 5 mg/mL. Contrary, no significant repellent activity was recorded for S-(+)-4-sec-butoxycoumarin. This experiment demonstrates the close relationship between the molecules' optical purities and the behavioral response of mosquitoes.

Biological Activity of Essential Oils of Four Juniper Species and Their Potential as Biopesticides.

The objective of this study was to assess the biological activity of essential oils (EOs) of four Juniperus species obtained via two different distillation methods and their potential as biopesticides. The studied factors were juniper species (Juniperus communis L., J. oxycedrus L., J. pygmaea C. Koch., and J. sibirica Burgsd), plant sex (male (M) and female (F)), and distillation method (hydrodistillation via a standard Clevenger apparatus (ClevA) and semi-commercial (SCom) steam distillation). The hypothesis was that the EO will have differential antioxidant, antimicrobial, and insecticidal activities as a function of plant species, plant sex, and distillation method. The two distillation methods resulted in similar EO composition within a given species. However, there were differences in the EO content (yield) due to the sex of the plant, and also differences in the proportions of some EO components. The concentration of α-pinene, β-caryophyllene, δ-cadinene and δ-cadinol was dissimilar between the EO of M and F plants within all four species. Additionally, M and F plants of J. pygmaea, and J. sibirica had significantly different concentrations of sabinene within the respective species. The EOs obtained via ClevA extraction showed higher antioxidant capacity within a species compared with those from SCom extraction. All of the tested EOs had significant repellent and insecticidal activity against the two aphid species Rhopalosiphum padi (bird cherry-oat aphid) and Sitobion avenae (English grain aphid) at concentrations of the EO in the solution of 1%, 2.5%, and 5%. The tested EOs demonstrated moderate activity against selected pathogens Fusarium spp., Botrytis cinerea, Colletotrichum spp., Rhizoctonia solani and Cylindrocarpon pauciseptatum. The results demonstrate that the standard ClevA would provide comparable EO content and composition in comparison with SCom steam distillation; however, even slight differences in the EO composition may translate into differential bioactivity.

New approach for evaluating the repellent activity of essential oils against storage pests using a miniaturized model of stored-commodity packaging and a wooden transport pallet

Essential oils (EOs) are promising compounds for repellent protection of packed commodities or processed food against storage insect pests. Currently, the efficacy of EOs has been tested only when applied to packages, whereas no tests are available for real-world situations in which products are placed on cardboard spacers and transport pallets. Therefore, the aim of this work was to test the effects of placing products on pallets and the treatment of packaging with bergamot essential oil as repellent of storage pests. Due to the size constraint posed by the size of pallets and packaging, miniaturized models were designed and constructed. In this work, we experimentally determined (i) the pest behavioral modification resulting from placing untreated packaging (i.e., without repellent treatment) containing untreated food on a miniaturized pallet vs. placing it on a simulated floor surface and (ii) the repellent effect of bergamot oil applied separately to carton packaging (containing untreated food), wooden pallets or cardboard pallet spacers located between the pallet and packaging on two key storage pests (Sitophilus granarius and Tribolium confusum). The beetle infestation was assessed as a number of beetles in the packaging, and the EO repellency was assessed as number of beetles in the packaging and in the cardboard. The placement of untreated packaging on untreated pallets and spacers decreased packaging infestation by both species in comparison with the placement of products directly on the floor surface. The treatment of various parts of the packaging set (i.e., food packaging, pallet, and cardboard spacer) with bergamot oil had significant repellent activity that was generally more apparent for S. granarius than for T. confusum. The wooden pallet treatment had a greater and longer (i.e., 24 h) repellent effect than the packaging or cardboard spacer treatment on the tested pests. The results showed that the treatment of some parts (such as pallets and cardboard spacers) of the packaging set with essential oils may make it unnecessary to treat primary food/commodity packaging. This may have profound implications for the use and acceptance of essential oils as insect repellents under industrial conditions, since producers may worry about the migration of EOs from the primary packaging to food. Moreover, the miniaturized packaging model presented in this study allowed effective EO repellent testing in large assays in a limited space (e.g., 360 miniaturized model pallets were used in this experiment, and testing this volume of actual Euro pallets would require an area of 346 m2).

Synthesis and Characterization of β-Cyclodextrin-Essential Oil Inclusion Complexes for Tick Repellent Development.

Essential oils (EOs) are used in several pest management applications. Due to their volatility, EOs may experience bioactivity reduction, thus requiring protection to extend their properties. In the present study, we investigated the inclusion complex formation (IC) of β-cyclodextrin (β-CD) with selected EOs with known tick repellent activity using two co-precipitation methods. ICs were characterized by evaluating EO mass concentration and inclusion efficiency (% IE) and other instrumental methods. Co-precipitation method 2 yielded the highest EO mass concentration (88 ± 6 μg/mg β-CD) for the 1:1 molar ratio geranium Egyptian EO IC. The EO volatile release over time from the ICs was investigated by headspace SPME/GC-MS analysis. ICs were also tested in tick repellency bioassays. ICs reported significant tick repellent activity, with lemongrass IC performing best overall. Method 1 showed the best combination of high mass concentration EO, controlled volatile release, and tick repellency with lemongrass EO. The results demonstrated that β-CD had selectively encapsulated different EOs. Moreover, the formation of ICs may improve EO tick repellent properties protecting the active ingredients and providing a better, long-lasting repellent action. These findings will allow the development of more effective naturally derived repellent products to protect individuals from tick bites and prevent tick-borne illnesses.

Isolation of Volatile Compounds with Repellent Properties against Aedes albopictus (Diptera: Culicidae) Using CPC Technology

The present work describes the use of Centrifugal Partition Chromatography (CPC) for the bio-guided isolation of repellent active volatile compounds from essential oils. Five essential oils (EOs) obtained from three Pinus and two Juniperus species were initially analyzed by gas chromatography–mass spectrometry (GC/MS) and evaluated for their repellent properties against Aedes albopictus. The essential oil from needles of P. pinea (PPI) presented the higher activity, showing 82.4% repellency at a dose of 0.2 μL/cm2. The above EO, together with the EO from the fruits of J. oxycedrus subsp. deltoides (JOX), were further analyzed by CPC using the biphasic system n-Heptane/ACN/BuOH in ratio 1.6/1.6/0.2 (v/v/v). The analysis of PPI essential oil resulted in the recovery of (−)-limonene, guaiol and simple mixtures of (−)-limonene/β-pheladrene, while the fractionation of JOX EO led to the recovery of β-myrcene, germacrene-D, and mixtures of α-pinene/β-pinene (ratio 70/30) and α-pinene/germacrene D (ratio 65/45). All isolated compounds and recovered mixtures were tested for their repellent activity. From them, (−)-limonene, guaiol, germacrene-D as well the mixtures of (−)-limonene/β-pheladrene presented significant repellent activity (>97% repellency) against Ae. albopictus. The present methodology could be a valuable tool in the effort to develop potent mosquito repellents which are environmentally friendly.

Fumigant and repellent potentials of Mentha pulegium L. and Citrus limon L. (Burm) essential oils against Tribolium confusum Duval. (Coleoptera: Tenebrionidae)

This study aims to investigate the chemical composition of essential oils (EOs) extracted from the aerial parts of pennyroyal (Mentha pulegium L.) and lemon peels (Citrus limon L.) and evaluate the fumigant toxicity and repellency against the most important and destructive stored product beetle in Algeria (T. confusum). Chemical analysis of the essential oil of M. pulegium showed that pulegone (62.24%), sabinene (5.51%) and menthol (16.5%) were the main essential components, while C. limon essential oil had limonene (66.93%) as the main compound, followed by β-pinene (4.52%), citral (3.09%) and β-Bisabolen (2.67%). The results show that the fumigant toxicity varies according to the concentration of essential oils, the median lethal concentration (LC50) values of Mentha pulegium and Citrus limon were 92.89 and 818.5 μl/Lair on T. confusum respectively. In addition, significant pest repellent activity was demonstrated with pennyroyal where repellent effects reached 80 and 90% (Class V) after 60 min exposure against the beetle at all concentrations (0.126, 0.314, 0.628 and 0.942 μL/cm2). The data obtained indicate that pennyroyal EOs show a higher toxicity and repellent activity against adults of T. confusum comparing to those of C. limon.
