Morphological, anatomical, and transcriptomics analysis reveals the regulatory mechanisms of cassava plant height development

BackgroundCassava is one of three major potato crops and the sixth most important food crop globally. Improving yield remains a primary aim in cassava breeding. Notably, plant height significantly impacts the yield and quality of crops; however, the mechanisms underlying cassava plant height development are yet to be elucidated.ResultsIn this study, we investigated the mechanisms responsible for cassava plant height development using phenotypic, anatomical, and transcriptomic analyses. Phenotypic and anatomical analysis revealed that compared to the high-stem cassava cultivar, the dwarf-stem cassava cultivar exhibited a significant reduction in plant height and a notable increase in internode tissue xylem area. Meanwhile, physiological analysis demonstrated that the lignin content of dwarf cassava was significantly higher than that of high cassava. Notably, transcriptome analysis of internode tissues identified several differentially expressed genes involved in cell wall synthesis and expansion, plant hormone signal transduction, phenylpropanoid biosynthesis, and flavonoid biosynthesis between the two cassava cultivars.ConclusionsOur findings suggest that internode tissue cell division, secondary wall lignification, and hormone-related gene expression play important roles in cassava plant height development. Ultimately, this study provides new insights into the mechanisms of plant height morphogenesis in cassava and identifies candidate regulatory genes associated with plant height that can serve as valuable genetic resources for future crop dwarfing breeding.

PpGATA21 Enhances the Expression of PpGA2ox7 to Regulate the Mechanism of Cerasus humilis Rootstock-Mediated Dwarf in Peach Trees.

Dwarfing rootstocks enhance planting density, lower tree height, and reduce both labor in peach production. Cerasus humilis is distinguished by its dwarf stature, rapid growth, and robust fruiting capabilities, presenting substantial potential for further development. In this study, Ruipan 4 was used as the scion and grafted onto Amygdalus persica and Cerasus humilis, respectively. The results indicate that compared to grafting combination R/M (Ruipan 4/Amygdalus persica), grafting combination R/O (Ruipan 4/Cerasus humilis) plants show a significant reduction in height and a significant increase in flower buds. RNA-seq indicates that genes related to gibberellin (GA) and auxin metabolism are involved in the dwarfing process of scions mediated by C. humilis. The expression levels of the GA metabolism-related gene PpGA2ox7 significantly increased in R/O and are strongly correlated with plant height, branch length, and internode length. Furthermore, GA levels were significantly reduced in R/O. The transcription factor PpGATA21 was identified through yeast one-hybrid screening of the PpGA2ox7 promoter. Yeast one-hybrid (Y1H) and dual-luciferase reporter (DLR) demonstrate that PpGATA21 can bind to the promoter of PpGA2ox7 and activate its expression. Overall, PpGATA21 activates the expression of the GA-related gene PpGA2ox7, resulting in reduced GA levels and consequent dwarfing of plants mediated by C. humilis. This study provides new insights into the mechanisms of C. humilis and offers a scientific foundation for the dwarfing and high-density cultivation of peach trees.

Comparison of temporomandibular joint osseous components in unilateral and bilateral cleft lip and palate patients and normal controls: A CBCT study

ObjectiveThe objective of this study was to conduct a comparative analysis of the components of the temporomandibular joint in individuals with unilateral, bilateral cleft lip and palate (CLP), and in healthy individuals, utilizing cone beam computed tomography (CBCT) images. Method and materialThe present study employed a cross-sectional design and recruited participants aged 18 to 30 years. The participants were categorized into three groups: a control group consisting of 36 individuals without any cleft, a group of 35 patients with unilateral cleft lip and palate (UCLP), and a group of 15 patients with bilateral cleft lip and palate (BCLP). The analysis of CBCT images encompassed the examination of condylar height and angulation, glenoid fossa height and width, articular eminence inclination, as well as joint spaces across all three groups. The Kruskal-Wallis and Mann-Whitney tests were employed to ascertain the significant differences among the three groups. ResultsThe UCLP and BCLP groups demonstrated a statistically significant reduction in condylar height and articular eminence inclination in comparison to the control group. Furthermore, a significant difference in the width of the glenoid fossa was seen between the group with clefts and the control group. ConclusionThe CBCT images showed significant differences in several aspects of the temporomandibular joint, including condylar height, articular eminence inclination, and glenoid fossa width, in individuals with cleft palate. These abnormalities can contribute to the development of temporomandibular joint diseases. Therefore, recognizing these distinctions can help prevent further deterioration and progression of temporomandibular disorders (TMD) in CLP patients.

Bevacizumab as Adjuvant Therapy in the Treatment of Keloid: A Randomized Clinical Trial

Background. Despite the availability of numerous therapies, keloid treatment remains a challenging clinical issue. Intralesional triamcinolone has been established as an effective corticosteroid treatment for keloids, while sporadic reports suggest the efficacy of intralesional verapamil. This study aimed to evaluate the safety and efficacy of bevacizumab as an adjuvant therapy for keloid treatment. Methods. This randomized controlled trial involved 38 patients diagnosed with keloid according to clinical criteria. The study compared the effects of intralesional triamcinolone combined with bevacizumab injections with intralesional triamcinolone alone. Patients were randomly assigned to either the combination treatment group, which received intralesional triamHEXAL® (20 mg/ml, every two weeks for three months) plus Avastin® (2.5 mg/ml, every two weeks for two months), or the single treatment group, which received intralesional triamHEXAL® alone. The Vancouver Scar Scale (VSS) was used for serial photographic records of scar evaluation, with differences in VSS scores considered the primary outcome, and changes in height and patient satisfaction visual analog score (VAS) were secondary outcomes. Results. A total of 38 patients participated, with a mean age (SD) of 35.32 (14.02) years and 50% male. No significant differences in age, BMI, disease duration, gender, causing, family history, or site were observed between the two groups. The single treatment group exhibited a mean reduction of 0.60 (95% CI: (−1.18, −0.01); P=0.045) in pigmentation score and a mean decrease of 1.37 (95% CI: (−2.68, −0.07); P=0.039) in total score compared to the combination treatment group after three months of treatment. There was a significant reduction in keloid height in the combination group after the end of the treatment (P=0.024). No significant differences in side effects were observed between the two groups. Conclusion. Our study demonstrates that bevacizumab can be considered an effective and safe adjuvant therapy option for keloid treatment, suggesting its potential as a promising treatment for the management of keloids. Molybdenum-Induced Oxidative Stress and Histopathological Alterations in the Epididymis

Molybednum is one of the trace elements required for proper functioning of the human body. The present study was designed to assess the effects of molybdenum on accessory reproductive organ i.e. epididymis. Male Wistar rats were administered with three different doses of molybdenum (50, 100 and 150 mg/kg body weight) for 60 days. A recovery study was also conducted in the highest dose group for which half the animals of the group were left untreated for the next 60 days. Exposure to molybdenum induced a significant decline in superoxide dismutase, ascorbic acid and glutathione while lipid peroxidation revealed a significant increase in epididymis in dose-dependent manner. Significant reduction in epididymal epithelial cells height, short sterocilia, increased intertubular stroma, decreased epididymal sperm reserve in the lumen and reduction in the diameter of cauda epididymal tubules with a relatively large amount of intertubular stroma was observed in epididymis of mice treated with molybdenum which might be correlated with enhanced oxidative stress. In the recovery group, significant improvement in antioxidant parameters and histoarchitecture of epididymis was observed indicating toxic effects of molybdenum can be reversed by cessation of exposure.

Deer grazing drove an assemblage‐level evolution of plant dwarfism in an insular system

Abstract Plant dwarfism, a syndrome characterised by a significant reduction in plant height and organ size, is a widely observed pattern of stress‐tolerant life‐form evolution that results from local adaptation to harsh environmental conditions. The drivers of assemblage‐level dwarfism have primarily been attributed to abiotic factors, such as low temperature, aridity, poor soil fertility or frequent fires. While biotic factors such as grazing pressure from herbivores can contribute to the establishment of plant dwarfism, these factors have rarely been tested at assemblage levels. Focusing on a dwarf plant assemblage comprising over 80 taxa on a small continental island in Japan with a high deer density, we hypothesised that historical deer grazing could also be a factor contributing to the large‐scale convergent evolution of dwarfism. To test this hypothesis, we measured the size of 1908 individual plants of 40 taxa pairs, comprising both palatable and unpalatable pairs from the island and their counterpart taxa from neighbouring regions, and sought to assess which factors (i.e. low solar radiation, estimated divergence time, low nutrient conditions and grazing pressure from deer) may have contributed to the formation of the dwarf plant assemblage on the island. We also performed genetic analysis to infer the time frames for the establishment of dwarf taxa. Statistical analyses revealed that plant size was significantly reduced mainly among the palatable taxa growing on the island, with preferential grazing by deer being identified as the most significant factor influencing plant size. Furthermore, genetic analyses revealed that dwarf ecotypes may have evolved over tens of thousands of years. Synthesis: To the best of our knowledge, this study is the first to demonstrate that interactions with herbivores can shape the assemblage‐level convergence of plant dwarfism. These findings enhance our current understanding of the formation of plant functional diversity.

Effects of in ovo feeding of xylobiose and xylotriose on growth performance, carcass traits, ileal histomorphometry, and immune-related gene expression in broiler chickens

Xylooligosaccharides (XOS) are known as prebiotics which promote beneficial gut microbes, improve gut health and the performance of broilers. Xylobiose (XOS2) and xylotriose (XOS3) are two compounds in the XOS group. In ovo feeding could deliver these prebiotics to the embryo, and this is considered as an effective method for studying the effects of the prebiotics during embryonic development and the post-hatch period. This study investigated the effects of in ovo feeding of XOS2 and XOS3 on growth performance, carcass traits, ileal histomorphometry, and immune-related gene expression in broilers. A total of 144 fertilized eggs of Cobb 500 broilers were divided into three groups: (a) non-injected control (CON), (b) XOS2, and (c) XOS3. The eggs from XOS2 and XOS3 groups were injected with 0.5 ml of 0.85% normal saline solution containing 3 mg of XOS2 and 3 mg of XOS3, respectively, into their amniotic sac on the 17th embryonic day. A total of 131 hatched chicks were raised in 18 replicate pens (5 or more chicks per pen) following standard management practices and fed a corn-soybean meal-based diet for 21 days. The feed intake and body weight (BW) data were recorded weekly. The chickens were euthanized on day 7, day 14, and day 21 to collect ileum sections, and the carcass traits were recorded on only day 21. Total RNA was extracted from ileum tissues, and qPCR was performed to measure the gene expression. All the statistical analyses considered the significance level at a P value <0.05. The result showed that in ovo treatments did not affect the hatchability of the eggs. The XOS2 group showed a trend to increase the BW on day 0, compared to CON (P=0.052), and the XOS3 group showed better FCR improvement than the XOS2 group in the first week. There was no difference (P>0.05) among the groups for the weight of the breast muscle, drumsticks, gizzard, proventriculus, and carcass weight at the age of 21 days. The histomorphometry of the ileum on day 7 revealed a significant reduction (P<0.05) of villi height (VH), villi width (VW), and crypt depth (CD) in XOS2, compared to the CON group. On day 7, a lower CD and a higher VH to CD ratio were recorded in XOS3, compared to CON. On day 14, XOS3 showed an increase in VW and villi surface area (VSA) compared to XOS2 birds. On day 7, the expression of IL10 in XOS2 was higher compared to CON and XOS3, and CD56 was significantly higher (P<0.05) in the XOS2 compared to CON group. On day 21, the XOS2 and XOS3 significantly lower the expression of IFNγ. Overall, XOS2 and XOS3 significantly increased immunity until day 21. In conclusion, in ovo XOS feeding demonstrated the potential to modulate gut morphology and immunity of the chickens during the post-hatch days.

Amelioration of chromium toxicity in wheat plants through exogenous application of nano silicon

Chromium (Cr) contamination in agricultural soils poses a risk to crop productivity and quality. Emerging nano-enabled strategies show great promise in remediating soils contaminated with heavy metals and enhancing crop production. The present study was aimed to investigate the efficacy of nano silicon (nSi) in promoting wheat growth and mitigating adverse effects of Cr-induced toxicity. Wheat seedlings exposed to Cr (K2Cr2O7) at a concentration of 100 mg kg−1 showed significant reductions in plant height (29.56%), fresh weight (35.60%), and dry weight (38.92%) along with enhanced Cr accumulation in roots and shoots as compared to the control plants. However, the application of nSi at a concentration of 150 mg kg−1 showcased substantial mitigation of Cr toxicity, leading to a decrease in Cr accumulation by 27.30% in roots and 35.46% in shoots of wheat seedlings. Moreover, nSi exhibited the capability to scavenge oxidative stressors, such as hydrogen peroxide (H2O2), and malondialdehyde (MDA) and electrolyte leakage, while significantly enhancing gas exchange parameters, total chlorophyll content, and antioxidant activities (enzymatic and nonenzymatic) in plants grown in Cr-contaminated soil. This study further found that the reduced Cr uptake by nSi application was due to downregulating the expression of HMs transporter genes (TaHMA2 and TaHMA3), alongwith upregulating the expression of antioxidant-responsive genes (TaSOD and TaSOD). The findings of this investigation highlight the remarkable potential of nSi in ameliorating Cr toxicity. This enhanced efficacy could be ascribed to the distinctive size and structure of nSi, which augment its ability to counteract Cr stress. Thus, the application of nSi could serve as a viable solution for production of crops in metal contaminated soils, offering an effective alternative to time-consuming and costly remediation techniques.

Evaluation of Pea Accessions Differing in Flower and Seed Coat Pigmentation for Resistance to Fusarium avenaceum Root Rot

ABSTRACTPea production across the world is significantly limited by root rot disease, which is caused by many fungal and oomycetes pathogens. In Canada, Fusarium avenaceum is the most devastating pathogen of the Fusarium root rot complex of pea. Host genetic resistance is the most effective control method for this disease. Evaluation of global pea accessions and Canadian varieties for F. avenaceum root rot resistance has not been reported to date. This study evaluated 20 pea accessions of different market classes with pigmented or nonpigmented seed coats and flowers for F. avenaceum resistance under controlled conditions. The pea accessions CDC Acer, CDC Vienna, PBA OURA, Morgan, CDC Blazer, CDC Dakota, and PI 280609, which have pigmented flowers and seed coats, were identified as resistant or partially resistant to F. avenaceum. This was based on their root rot severity scores and ability to tolerate F. avenaceum infection without significant (p &gt; 0.05) reductions in plant height, shoot dry weight, and root dry weight. Among the varieties with nonpigmented flowers and seed coats, only Cameor showed partial resistance to F. avenaceum when challenged with reduced conidial concentration. Root dry weight (R = −0.86), plant height (R = −0.82), and shoot dry weight (R = −0.78) had a strong negative correlation (p &lt; 0.001) with disease severity, suggesting that F. avenaceum root rot can negatively impact the growth and development of pea seedlings. F. avenaceum resistance identified in this study can be utilized to study the molecular basis of the resistance and develop disease‐resistant varieties. While our findings suggest a relationship between pigmentation and F. avenaceum resistance, future research with a larger, more diverse panel is warranted to validate these initial results.

Efficacy of Subthreshold Micropulse Laser for Central Serous Chorioretinopathy.

To evaluate the efficacy of a subthreshold micropulse laser (SML) in patients with central serous chorioretinopathy (CSCR). Retrospective clinical study conducted at the Departments of Ophthalmology at a university and a municipal hospital in Zurich, Switzerland. We enrolled acute and chronic CSCR patients with persistent subretinal fluid (SRF) treated with SML. Two treatment protocols (fluorescein/indocyanine green angiography or optical coherence tomography guided) were evaluated for efficacy after 3 and 6 months. The primary outcomes of the study were reduction and percentage of eyes with complete resolution of SRF 3 and 6 months after SML treatment. Secondary endpoints included changes in central subfield thickness (CST) and visual acuity (VA) after 3 and 6 months. The study involved 37 eyes (35 patients, 48.6% chronic). A statistically significant reduction in SRF height and CST could be shown, irrespective of SRF duration, type of CSCR, or chosen guidance after 3 and 6 months: SRF - 40 µm (p < 0.01), CST - 52 µm (p < 0.01). Percentage of eyes with complete resolution of fluid at 3 and 6 months after SML were 24.3 and 21.6%, respectively. No statistically significant functional improvement (VA) could be shown. Multivariable regression and linear mixed regression analyses did not identify statistically significant differences in SRF reduction, CMT change, or VA improvement with respect to the type of CSCR or the treatment plan used (p > 0.05). The effectiveness of SML in CSCR is under continuous debate. Our study findings demonstrate structural but only little functional changes with SML. In view of the shortage of verteporfin for photodynamic therapy, SML remains an important therapeutic option for CSCR patients.

Maxillary Sinus Augmentation with Anorganic Bovine Bone Mineral of Different Particle Sizes: A Split-Mouth Study with Histomorphometric, Radiographic, and Clinical Analyses.

The aim of the present study was to compare the histomorphometrically evaluated new bone formation (NB), the radiographically measured graft stability, and the clinical implant outcome for maxillary sinus augmentation grafted with deproteinized bovine bone mineral (DBBM) with either small (Bio-Oss-S, Geistlich) or large (Bio-Oss-L, Geistlich) particles. Using a split-mouth study design, bilateral maxillary sinus augmentation was performed in 13 patients either with Bio-Oss-S particles (0.25 to 1 mm) or Bio-Oss-L particles (1 to 2 mm). After a healing period of 6 months, bone biopsies were axially retrieved in the molar region for histologic/histomorphometric analysis of NB, including subsequent staged implant placement. To determine graft stability, the maxillary sinus augmentation vertical graft heights were radiographically measured immediately after sinus augmentation, at implant placement, and at the 2- and 4-year post-augmentation follow-ups. In addition, the clinical implant-prosthodontic outcome (survival/ success/marginal bone loss) was assessed at 1 and 3 years post-loading. A total of 22 sinuses from 11 patients with split-mouth evaluation were ultimately available for data and statistical analysis. Histomorphometric analysis of the axially retrieved bone biopsies revealed the presence of NB (S: 25.5% ± 7.0% vs L: 23.6% ± 11.9%; P = .640), residual graft particles (S: 19.6% ± 9.2% vs L: 17.5% ± 6.3%; P = .365) as well as connective tissue (S: 54.9% ± 9.2% vs L: 58.9% ± 12.5%; P = .283), without significant differences between the use of small (Bio-Oss-S) and large (Bio-Oss-L) particles. However, there was significantly (P = .021) higher bone-to-graft contact (BGC) for the small-particle graft sites (27.9% ± 14.8%) compared to the large-particle graft sites (19.9% ± 12.9%), representing a significantly higher osteoconductivity. Both particle sizes showed significant (P < .01) vertical graft height reduction over time (4 years) of about 10%, with predominant graft reduction in the time period between sinus augmentation and implant placement compared to any follow-up periods after implant placement. At the 3-year post-loading implant evaluation, all implants and prostheses survived (100%), and the peri-implant marginal bone loss (S: 0.52 ± 0.19 mm; L: 0.48 ± 0.15 mm) as well as the peri-implant health conditions (S: 87.5%, L:81.2%) did not differ between implants inserted with the two different xenograft particles used. The use of small and large bovine xenograft particles for maxillary sinus augmentation provides for comparable bone formation, ensuring stable graft dimensions combined with high implant success and healthy peri-implant conditions. However, small particle size resulted in a higher BGC, providing for higher osteoconductivity than with the larger particle size.

A novel in-vitro model of intervertebral disc degeneration using hyperphysiological loading

Intervertebral disc (IVD) degeneration includes changes in tissue biomechanics, physical attributes, biochemical composition, disc microstructure, and cellularity, which can all affect the normal function of the IVD, and ultimately may lead to pain. The purpose of this research was to develop an in-vitro model of degeneration that includes the evaluation of physical, biomechanical, and structural parameters, and that does so over several load/recovery periods. Hyperphysiological loading was used as the degenerative initiator with three experimental groups employed using bovine coccygeal IVD specimens: Control; Single-Overload; and Double-Overload. An equilibrium stage comprising a static load followed by two load/recovery periods was followed by six further load/recovery periods. In the Control group all load/recovery periods were the same, comprising physiological cyclic loading. The overload groups differed in that hyperphysiological loading was applied during the 4th loading period (Single-Overload), or the 4th and 5th loading period (Double-Overload). Overloading led to a significant reduction in disc height compared to the Control group, which was not recovered in subsequent physiological load/recovery periods. However, there were no significant changes in stiffness. Overloading also led to significantly more microstructural damage compared to the Control group. Taking all outcome measures into account, the overload groups were evaluated as replicating clinically relevant aspects of moderate IVD degeneration. Further research into a potential dose–effect, and how more severe degeneration can be replicated would provide a model with the potential to evaluate new treatments and interventions for different stages of IVD degeneration.

Efficacy of acellular xenogeneic dermal matrix graft in the treatment of multiple gingival recessions: systematic review and meta-analysis.

This study aimed to compare the efficacy of acellular xenogeneic dermal matrix graft (AXDM) compared to connective tissue graft (CTG) in treating multiple gingival recessions. A systematic search of electronic databases was conducted to identify randomized clinical trials (RCTs) that compared AXDM and CTG. The selected studies were subjected to bias risk assessment, data extraction, and meta-analyses. Parameters such as gingival recession height, width, mean percentage of root coverage, and complete root coverage were analyzed. Seven RCTs involving 146 patients were included. The meta-analyses indicated that CTG was statistically superior to AXDM in reducing gingival recession height at the final follow-up (mean difference: -0.104mm, 95% confidence interval [CI]: -0.180-0.028, p = 0.008) and width at the final follow-up (mean difference: -0.285mm, 95% CI: -0.541-0.030, p = 0.029). CTG also demonstrated a significantly higher mean percentage of root coverage at the 6-month follow-up (difference in means: -2.761mm, 95% CI: -4.932-0.590, p = 0.013) and a higher percentage of complete root coverage at the 6-month follow-up (odds Ratio [OR]: 0.598, 95% CI: 0.4-0.892, p = 0.012) compared to AXDM. However, there was no significant difference in the number of teeth with complete root coverage between CTG and AXDM (OR: 1.610, 95% CI: 0.983-2.636, p = 0.058) and aesthetic outcomes (mean difference: 0.148, 95% CI: -0.277-0.573, p = 0.494). CTG is more effective than AXDM in treating multiple gingival recessions. This is evidenced by significant reductions in gingival recession height and width, a higher mean percentage of root coverage, and a greater percentage of complete root coverage at the 6-month follow-up. In some clinical situations an alternative to CTG is required for the treatment of multiple gingival recessions. AXDM, despite presenting clinical outcomes that are not as satisfactory as CTG, can be used for this purpose.

Impact of weight reduction on structural and functional parameters of the optic nerve in idiopathic intracranial hypertension.

To evaluate the effect of weight reduction on the structural and functional parameters of the optic nerve in established cases of papilledema in Idiopathic Intracranial Hypertension (IIH). A prospective observational study in early and established cases of papilledema in IIH presenting from December 2019 to February 2021. Functional parameters (visual acuity, contrast sensitivity, mean deviation, VER), structural parameters (RNFL, GCL-IPL, and optic disc height), and clinical grading of papilledema were measured at baseline and every 6 weeks for 6 months. All patients underwent medical (oral acetazolamide) and diet therapy. Mean body mass index (BMI) at presentation was 26.32 ± 3.52 kg/m² and the mean change in BMI over 6 months was 1.27 ± 0.50 kg/m². 67% of eyes had papilledema of grades 2-3 at the presentation. At the end of 6 months, 75% of eyes had grade 1 papilledema while 13% showed complete resolution. A reduction of 5.32 ± 3.58 in mean deviation (r = 0.316; p-value 0.01) and gain in VER P100 amplitude of 4.2 ± 2.7 µV (r = 0.40; p-value 0.003) were noted over 6 months with reduction in BMI over 6 months. A statistically significant reduction in optic disc height (ODH) was noticed with BMI reduction over 6 months (p = 0.0007; r = 0.45).A reduction of 1.72 ± 0.90 kg/m² (equivalent to 6.53% weight loss) in BMI was associated with a four-grade change in clinical grading of papilledema. Reduction in BMI had a significant impact on both structural and functional parameters of the optic nerve in IIH. The involvement of a well-certified dietician should be an integral part of treatment in IIH.

Effect of Weight Bearing on Medial Longitudinal Arch of Foot in Healthy Adults

Background: The medial longitudinal arch (MLA) of the foot plays a pivotal role in biomechanical stability and mobility. Understanding its behavior under different loading conditions is crucial for both clinical and sports-related applications. Previous studies have offered insights into the arch's adaptability, yet the impact of gender on this dynamic has remained underexplored. Objective: This study aimed to investigate the differences in medial longitudinal arch height between genders in non-weight bearing (NWB) and full weight bearing (FWB) positions, to ascertain if gender influences arch behavior under various loading conditions. Methods: Conducted at the Biomechanics and Kinesiology Laboratory of Shifa Tameer e Millat University, this cross-sectional study enrolled volunteers aged 18 to 40 years with normal foot arches. Exclusion criteria included history of foot pain, foot anomalies, pregnancy, or menstruation at the time of data collection. Navicular height was measured using Kinovea software (Version 0.9.5), with participants in both NWB and FWB positions. The study received approval from the Institutional Review Board and Ethics Committee of Shifa International Hospital (IRB # 0213-23). Statistical analysis was performed using independent and paired sample t-tests. Results: The study involved equal numbers of male and female participants, with no significant gender-based differences observed in navicular height in NWB (Males: Left Foot 5.17 ± 0.865, Right Foot 5.19 ± 0.675; Females: Left Foot 5.24 ± 0.89, Right Foot 5.39 ± 0.867) and FWB positions (Males: Left Foot 4.73 ± 0.834, Right Foot 4.88 ± 0.846; Females: Left Foot 4.83 ± 0.898, Right Foot 4.80 ± 0.837). Significant reductions in navicular height from NWB to FWB were noted across both genders (p &lt; 0.01). Conclusion: The medial longitudinal arch height significantly changes under weight bearing, displaying a similar pattern of adaptability across genders. This indicates that gender does not significantly influence the biomechanical behavior of the MLA under the tested conditions.

Long-term effects of high-dose systemic corticosteroids on growth and bone mineral density in patients treated for childhood interstitial lung disease (chILD).

Children's interstitial lung disease (chILD) is a rare and potentially life-threatening condition. For many chILD conditions, systemic corticosteroids (sCCS) are considered the primary treatment despite a broad spectrum of potential side effects. We aimed to determine the long-term effects of sCCS treatment on growth, bone mineral density (BMD), and body composition after chILD. This descriptive cross-sectional single-center study included patients diagnosed with chILD before the age of 18 years treated with sCCS in the period 1998-2020. Dual-energy X-ray absorptiometry, anthropometric measurements, bone age determination, and blood tests were performed in 53 (55% males) of 89 eligible patients. Median (range) age was 19.3 (6.4;30.7 years). Participants received a median (range) cumulative sCCS dose of 1144 (135; 6178) mg over a 2.0 (0.1; 13.8) years period and latest dose was administered 11.7 (1.2; 19.6) years before follow-up. Mean delta height (height standard deviation scores[SDS] - target height SDS) was reduced at sCCS treatment initiation (mean: -0.55, 95% confidence interval [CI]: -0.91; -0.20, p < .005) and at sCCS treatment cessation (mean: -0.86, 95%CI:-1.22; -0.51, p < .001), but normalized in the majority at follow-up (mean: -0.29, 95%CI:-0.61; 0.03, p = .07). Mean (SD) BMD z-score for the spine and whole body was -0.34 (1.06) and 0.52 (1.13), with no significant correlation to sCCS dose. Excess body fat (>30% in females, >25% in males) was found in 58% of patients. Long-term treatment with sCCS did not cause significant long-term reduction of height but showed subtle effects on fat mass percentage and BMD. Given the severity of chILD, the observed long-term effects of sCCS on growth and BMD appear acceptable.

Effects of dietary Nisin on growth performance, immune function, and gut health of broilers challenged by Clostridium perfringens.

Nisin (Ni) is a polypeptide bacteriocin produced by lactic streptococci (probiotics) that can inhibit the majority of gram-positive bacteria, and improve the growth performance of broilers, and exert antioxidative and anti-inflammatory properties. The present study investigated the potential preventive effect of Nisin on necrotic enteritis induced by Clostridium perfringens (Cp) challenge. A total of 288 Arbor Acres broiler chickens of 1-d-olds were allocated using 2 × 2 factorial arrangement into four groups with six replicates (12 chickens per replicate), including: (1) control group (Con, basal diet), (2) Cp challenge group (Cp, basal diet + 1.0 × 108 CFU/mL Cp), (3) Ni group (Ni, basal diet + 100mg/kg Ni), and (4) Ni + Cp group (Ni + Cp, basal diet + 100mg/kg Ni + 1.0 × 108 CFU/mL Cp). The results showed that Cp challenge decreased the average daily gain (ADG) of days 15 to 21 (P<0.05) and increased interleukin-6 (IL-6) content in the serum (P < 0.05), as well as a significant reduction in villus height (VH) and the ratio of VH to crypt depth (VCR) (P<0.05) and a significant increase in crypt depth (CD) of jejunum (P<0.05). Furthermore, the mRNA expressions of Occludin and Claudin-1 were downregulated (P<0.05), while the mRNA expressions of Caspase3, Caspase9, Bax, and Bax/Bcl-2 were upregulated (P<0.05) in the jejunum. However, the inclusion of dietary Ni supplementation significantly improved body weight (BW) on days 21 and 28, ADG of days 15 to 21 (P<0.05), decreased CD in the jejunum, and reduced tumor necrosis factor-α (TNF-α) content in the serum (P<0.05). Ni addition upregulated the mRNA levels of Claudin-1 expression and downregulated the mRNA expression levels of Caspase9 in the jejunum (P<0.05). Moreover, Cp challenge and Ni altered the cecal microbiota composition, which manifested that Cp challenge decreased the relative abundance of phylum Fusobacteriota and increased Shannon index (P<0.05) and the trend of phylum Proteobacteria (0.05<P<0.10), while Ni addition increased the relative abundance of phylum Cyanobacteria (P<0.05). In conclusion, the results demonstrated that the addition of Ni effectively mitigated the negative impact of Cp challenge-induced the damage of growth performance and intestine in broilers via reducing inflammatory responses and intestinal apoptosis and modulating cecal microbial composition.

Comparative transcript abundance of gibberellin oxidases genes in two barley (Hordeum vulgare) genotypes with contrasting lodging resistance under different regimes of water deficit.

Barley (Hordeum vulgare ) is the world's fourth most important cereal crop, and is particularly well adapted to harsh environments. However, lodging is a major productivity constraint causing 13-65% yield losses. Gibberellic acid (GA) homeostatic genes such as HvGA20ox, HvGA3ox and HvGA2ox are responsible for changes in plant phenotype for height and internodal length that contribute towards lodging resistance. This study explored the expression of different HvGAox transcripts in two contrasting barley genotypes (5-GSBON-18, lodging resistant; and 5-GSBON-70, lodging sensitive), which were sown both under controlled (hydroponic, completely randomised factorial design) and field conditions (split-plot, completely randomised block design) with two irrigation treatments (normal with three irrigation events; and water deficit with one irrigation event). In the hydroponic experiment, expression analysis was performed on seedlings at 0, ¾, 1½, 3 and 6h after application of treatment. In the field experiment, leaf, shoot nodes and internodes were sampled. Downregulation of HvGA20ox.1 transcript and 2-fold upregulation of HvGA2ox.2 transcript were observed in 5-GSBON-18 under water deficit conditions. This genotype also showed a significant reduction in plant height (18-20%), lodging (<10%), and increased grain yield (15-18%) under stress. Utilisation of these transcripts in barley breeding has the potential to reduce plant height, lodging and increased grain yield.

Identification of a novel SNP in the miR172 binding site of Q homoeolog AP2L-D5 is associated with spike compactness and agronomic traits in wheat (Triticum aestivum L.)

Key messageThis study found that the compact spike locus of ANK-15 is on chromosome 5D instead of 2B. We have identified a new allele of AP2L-D5 as the candidate causal polymorphism.Spike architecture is a key determinant of wheat yield, a crop which supports much of the human diet but whose yield gains are stagnating. Spike architecture mutants offer opportunities to identify genetic factors contributing to inflorescence development. Here, we investigate the locus underlying the compact spike phenotype of mutant line ANK-15 by conducting mRNA-sequencing and genetic mapping using ANK-15 and its non-compact spike near-isogenic line Novosibirskaya 67 (N67). Previous literature has placed the compact spike locus of ANK-15 to chromosome 2B. However, based on the single nucleotide polymorphisms (SNPs) identified using mRNA-seq data, we were unable to detect polymorphisms between N67 and ANK-15 in the putative chromosome 2B region. We performed differential expression analysis of developing rachis and found that AP2L-D5, the D homoeolog of the domestication Q gene, is upregulated in ANK-15 in comparison to N67. ANK-15 carries a SNP in the microRNA172 binding site of AP2L-D5, which is predicted to lead to higher expression of AP2L-D5 due to decreased miRNA172-mediated degradation. Furthermore, we performed genetic mapping using an ANK-15 × N67 F2 population and found a single quantitative trait locus on chromosome 5D coinciding with the position of AP2L-D5. This result suggests that AP2L-D5 is likely the underlying causal gene for the compact spike phenotype in ANK-15. We performed a field trial to investigate the effect of the AP2L-D5 allele on agronomic traits and found that the AP2L-D5 allele from ANK-15 is associated with a significant reduction in height, increased thousand grain weight (TGW), and increased grain width.

Flood Tolerance of Hybrid Napier (Pennisetum purpureum x Pennisetum glaucum) Cultivars

A pot culture study was conducted from February to May 2020 at the Agronomy Farm, Department of Agronomy, College of Agriculture, Vellanikkara, Kerala to study the effects of short-term flooding on the growth, physiology, and yield of four high-yielding cultivars of hybrid napier (Pennisetum purpureum x Pennisetum glaucum) cultivated in Kerala. The experiment followed a 4 x 2 factorial Completely Randomized Design with three replications. The treatments comprised two factors: varieties and flooding treatments. The four hybrid napier varieties used in the study were CO-3 (V1), CO-5 (V2), IGFRI -3 (V3), and Suguna (V4). The flooding treatments included no flooding (F0) as a control and short-term flooding stress treatment (F1) at three different growth stages: 30 days after planting (DAP), 60 DAP and just after the first harvest (75 DAP). Flooding stress was imposed for seven days by inundating water in the pots up to 3-4 cm above the soil level. Flooding at all three stages resulted in a significant reduction in plant height, tiller number, leaf number and above-ground biomass for all varieties. A severe reduction in growth and yield parameters was observed when flooding stress was imposed immediately after the first harvest. Among the varieties, CO-5 had the tallest plants, followed by Suguna, CO-3, and IGFRI-3. When exposed to flooding stress, variety CO-5 was also found to be significantly superior in terms of above-ground biomass production, followed by CO-3. Superoxide dismutase (SOD) activity increased in all varieties due to flooding stress, with a pronounced increase observed in Suguna. Variety CO-5, which exhibited greater plant height and above-ground biomass production, was found to be more tolerant to flooding stress, while variety IGFRI-3, with a dwarf plant stature and the lowest biomass yield, was identified as sensitive to short-term flooding stress.
