Length of Overnight Fasting and 6-Y Weight Change in the Cancer Prevention Study-3

BackgroundLonger overnight fasting (ONF) is a potential strategy for weight control. Although promising, the evidence from large population-based studies is limited. ObjectivesTo examine the association of self-reported ONF duration with 3- and 6-y weight change in the American Cancer Society’s Cancer Prevention Study-3 prospective cohort. MethodsUnited States adult Cancer Prevention Study-3 participants completed a 24-h validated meal and snack timing and frequency grid (weekday and weekend) in 2015, from which weighted ONF hours were calculated. Participants reported body weight in 2015, 2018, and 2021. Three- and 6-y weight change (kg, and % body weight) were assessed. ResultsAmong 104,420 mostly female (78.5%) participants aged 52.7 ± 9.5 (standard deviation) y followed for 6 y, a 1-h increase in ONF length was associated with a small but statistically significant reduction in weight gain over 3- and 6-y periods [multivariable-adjusted mean difference in % body weight = −0.02, 95% confidence interval (CI): −0.05, −0.00, P = 0.03 and −0.04, 95% CI: −0.07, −0.01, P = 0.007, respectively]. The mean difference of 6-y % reduction in weight gain was slightly greater among individuals with overweight (−0.05, 95% CI: −0.10, 0.00, P = 0.05) and obesity (−0.06, 95% CI: −0.12, 0.01, P = 0.08) compared with those with healthy body mass index (−0.03, 95% CI:−0.07, 0.01, P = 0.13) or underweight (0.16, 95% CI: −0.04, 0.36, P = 0.13, Pinteraction < 0.0001). Stronger associations were observed among those ≤55 y than 56+ (Pinteraction = 0.01), and those with higher waist circumference (Pinteraction < 0.0001) but not by sex or earlier/later fasting period. ConclusionsLonger ONF was associated with slightly lower body weight in adult males and females over 6 y that was stronger among those with overweight or obesity, higher waist circumference, and those aged ≤55 y. The magnitude of weight change, although in the hypothesized direction, suggests that prolonged ONF may have modest impact on weight control over time.

Epidemiology Tools to Evaluate the Control of Proliferative Enteropathy in Commercial Pig Herds.

Proliferative enteropathy (PE) is characterized by diarrhea and reduced weight gains in growing pigs and intestinal hemorrhage in finishers. Vaccination, antibiotic medication, and improved hygiene can control PE, but their efficacy depends upon the epidemiology of PE. This study monitored the timing and severity of PE in 84 commercial pens across seven treatments, including disinfection, vaccination, no treatment, medication with olaquindox (50, 25 and 12.5 ppm), and combined disinfection and vaccination. Vaccination with or without lime disinfection suppressed clinical signs of PE and reduced the number of excreted L. intracellularis relative to untreated pigs housed in cleaned or cleaned and disinfected pens between 9 and 17 weeks of age. Continuous olaquindox mediation to 17 weeks of age prevented L. intracellularis infection, leaving finisher pigs naïve. These finisher pigs suffered an outbreak of hemorrhagic enteropathy with significant reductions in weight gain, feed intake, and mortalities of 4.6%. Over the 13 week grow/finish period, vaccinated pigs housed in disinfected pens showed significantly higher weight gain and feed intake relative to all other treatments, equating to a weight gain difference of between 3.6 and 3.9 kg per pig. Monitoring the immune response and fecal excretion of L. intracellularis in pens of pigs enabled effective PE control strategies to be evaluated on the farm.

Ameliorative effect of bofutsushosan (Fangfengtongshengsan) extract on the progression of aging-induced obesity.

This study aimed to compare fat accumulation in young and aged mice raised on a high-fat diet and to characterize the obesity-reducing effects of a Kampo medicine, bofutsushosan (BTS; fangfengtongshengsan in Chinese). Aged mice fed a high-fat diet containing 2% BTS extract for 28days exhibited a significant reduction in weight gain and accumulation of visceral and subcutaneous fat, which were greater degree of reduction than those of the young mice. When the treatment period was extended to two months, the serum aspartate aminotransferase and alanine aminotransferase levels and the accumulation of fat droplets in the hepatocytes decreased. The mRNA expression of mitochondrial uncoupling protein 1 (UCP1) in the brown adipose tissue was significantly reduced in the aged mice compared to the young mice but increased by 2% in the BTS-treated aged mice. Additionally, the effect of BTS extract on oleic acid-albumin-induced triglyceride accumulation in hepatoblastoma-derived HepG2 cells was significantly inhibited in a concentration-dependent manner. Evaluation of the single crude drug extracts revealed that Forsythia Fruit, Schizonepeta Spike, and Rhubarb were the active components in BTS extract. These results suggest that BTS extract is effective against visceral, subcutaneous, and ectopic fats in the liver, which tend to accumulate with aging. Thus, BTS extract is useful in preventing and ameliorating the development of obesity and metabolic syndrome.

Effect of day-old chick weight on the response of broilers to high doses of exogenous phytase

A total of 360 Ross 708 male broiler chicks were used in an 8 treatment and 9 replicate cage study to explore the influence of day-old chick weight on the efficacy of exogenous phytase. Treatments were arranged as a 2 × 4 factorial with the factors being diet (a positive (PC) and negative control (NC) varying in nutrient density fed without or with 2 concentrations of exogenous phytase) and chick weight (light; <38.5 g or heavy; >42 g). Chicks were sourced from the same breeder flock, with light and heavy chicks being selected from the naturally occurring heterogeneity in the population. The diets were corn-soybean meal based and the PC was formulated to meet the nutrient requirements of male broiler chicks. The NC was formulated to contain 120 kcal/kg, 0.5, 0.18, and 0.18% less apparent metabolizable energy (AME), crude protein (CP), calcium (Ca), and phosphorus (P), respectively, than the PC. Amino acid (AA) density in the NC was also reduced in line with the reduction in CP and the manufacturers' recommendations for the effect of phytase on amino acid digestibility. Phytase at either 1,000 FYT/kg or 3,000 FYT/kg was added only to the NC to create a total of 4 diets. Water and mash feed were available ad libitum and were offered to 8 replicate cages, each containing 5 chicks. The experiment was conducted over a period of 3 wk and diets were offered in 2 phases (starter from d 1 to 10 and grower from d 11 to 21). Growth performance was monitored at the end of each diet phase and on d 21 ileal digesta were collected for estimation of apparent digestibility of energy (DE), nitrogen (N), Ca, P, dry matter (DM), and AA. There were no statistically significant interactions between diet and day-old chick weight for any of the measured parameters. Light chicks had significantly lower weight gain (approx. 5%) at both d 10 and d 21 compared with heavy chicks. This effect was principally associated with reduced feed intake and there was no significant effect of chick weight on feed conversion ratio (FCR). Chick weight had no effect on ileal nutrient digestibility. The reduction in nutrient density from the PC to the NC generated a significant reduction in weight gain (around 12%) and a significant increase in FCR (1.68 vs. 1.83). This effect was associated with a significant reduction in ileal DE (approximately 150 kcal/kg) and in the digestibility of several AA. Exogenous phytase significantly increased weight gain, reduced FCR and generated a significant increase in the ileal digestibility of energy, N, P, and several AA. Although chick weight and diet did not interact statistically, heavy chicks benefited more than light chicks from high doses of exogenous phytase across almost all measured end points which was confirmed by regression analysis. In conclusion, light chicks have inferior performance outcomes than heavy chicks principally because of reduced feed intake, but putatively not in digestive capacity per se. Exogenous phytase is effective in improving performance and nutrient digestibility in nutrient deficient diets. The effect of chick weight per se, and also breeder flock age, on the utility of supra-nutritional inclusion concentrations of exogenous phytase warrants further study.

Effects of methylphenidate on femoral bone growth in male rats.

The use of Methylphenidate (MP) can have adverse effects on bone growth and mineralization. This study aimed to investigate the underlying pathophysiology of MP-induced skeletal deficits in growing rats using stereological and immunohistochemical methods. Male rats, aged 4weeks, were orally treated with MP through an 8-h/day water drinking protocol. The rats (n=30) were randomly divided into three groups: MP-High Dose (30/60mg/kg/day MP), MP-Low Dose (4/10mg/kg/day MP), and control (water only). After 13weeks, the femoral bones were assessed using calliper measurements, dual-energy X-ray absorptiometry, and biomechanical evaluation. The total femur volume, cartilage volume, growth zone volume, and volume fractions were determined using the Cavalieri method. Immunohistochemical analyses were conducted using alkaline phosphatase and anti-calpain antibody staining. Rats exposed to MP exhibited significant reductions in weight gain, femoral growth, bone mineralization, and biomechanical integrity compared to the control group. The total femoral volume of MP-treated rats was significantly lower than that of the control group. The MP-High Dose group showed significantly higher ratios of total cartilage volume/total femoral volume and total growth zone volume/total femoral volume than the other groups. Immunohistochemical evaluation of the growth plate revealed reduced osteoblastic activity and decreased intracellular calcium deposition with chronic MP exposure. The possible mechanism of MP-induced skeletal growth retardation may involve the inhibition of intracellular calcium deposition in chondrocytes of the hypertrophic zone in the growth plate. In this way, MP may hinder the differentiation of cartilage tissue from bone tissue, resulting in reduced bone growth and mineralization.

Catechins prevent obesity-induced kidney damage by modulating PPARγ/CD36 pathway and gut-kidney axis in rats

Obesity is an epidemic and a growing public health concern worldwide. It is one of the significant risk factors for developing chronic kidney disease. In the present study, we evaluated the preventive effect of green tea catechins (GTC) against obesity-induced kidney damage and revealed the underlying molecular mechanism of action. Various green tea catechins were quantified in the catechins-rich fraction using HPLC. In vitro, the palmitic and oleic acid-treated NRK-52E cells showed reduced fat accumulation and modulated expressions of PPARγ, CD36, and TGFβ after GTC treatment. In vivo, rats were fed with a high-fat diet (HFD), and the effect of GTC was assessed at 150 and 300 mg/kg body weight doses. HFD-fed rats showed a significant reduction in weight gain and improved serum creatinine, urea, and urine microalbumin levels after GTC treatment. The improved adipokines and insulin levels in GTC treated groups indicated the insulin-sensitizing effect. Histopathology revealed reduced degenerative changes, fibrous tissue deposition, and mesangial matrix proliferation in GTC treated groups. GTC treatment also downregulated the gene expressions of lipogenic and inflammatory factors and improved the altered expressions of CD36 and PPARγ in the kidney tissue. Further, GTC prevented gut dysbiosis in rats by promoting healthy microbes like Akkermansia muciniphila and Lactobacillus reuteri. Faecal metabolome revealed reduced saturated fatty acids, and improved amino acid levels in the GTC treated groups, which help to maintain gut health and metabolism. Overall, GTC prevented obesity-induced kidney damage by modulating PPARγ/CD36 signaling and maintaining gut health in rats.

Taurochenodeoxycholic acid inhibits intestinal epithelial cell proliferation and induces apoptosis independent of the farnesoid X receptor.

Bile acids, such as taurochenodeoxycholic acid (TCDCA), are considered as functional small molecules involved in nutrition regulation or acting with adjuvant therapeutic effects against metabolic or immune diseases. The homeostasis of the intestinal epithelium depends on the conventional cellular proliferation and apoptosis of cells. Herein, mice and normal intestinal epithelial cells (IPEC-J2, a widely used normal intestinal epithelial cell line derived from porcine) were used as models to explore the regulatory effect of TCDCA on the proliferation of intestinal epithelial cells (IECs). In the mouse study, the oral gavage of TCDCA led to a significant reduction in weight gain, small intestinal weight, and the villus height of the intestinal epithelium while inhibiting the gene expression of Ki-67 in the intestinal epithelial crypts of mice (P < 0.05). TCDCA significantly downregulated the expression of the farnesoid X receptor (FXR) and upregulated the expression of caspase-9 in the jejunum (P < 0.05). The results of real-time quantitative PCR (RT-qPCR) suggested that TCDCA significantly inhibited the expression of tight junction proteins zonula occludens (ZO)-1, occludin, claudin-1, and mucin-2 (P < 0.05). In terms of apoptosis-related genes, TCDCA significantly inhibited the expression of Bcl2 and increased the expression of caspase-9 (P < 0.05). At the protein level, TCDCA decreased the expression of Ki-67 and PCNA, as well as FXR (P < 0.05). Caspase inhibitor Q-VD-OPh and guggulsterone, an FXR antagonist, significantly improved the inhibition of TCDCA-induced cell proliferation. Moreover, guggulsterone enhanced TCDCA-induced cell late apoptosis through flow cytometry and significantly lowered the TCDCA-induced up-regulated gene expression of caspase 9, despite both TCDCA and guggulsterone down-regulating the expression of FXR (P < 0.05). Overall, the effect of TCDCA on the induction of apoptosis is not dependent on FXR, whereas it would function via the activation of the caspase system. This provides a new perspective for the application of TCDCA or bile acid as functional small molecules in food, additives, and medicine.

Assessment of cyto-nephrotoxicity and growth performance in Labeo rohita induced by fluorescein dye Y and B

The cyto-nephrotoxicity in fishes induced by dyes is a global issue. Fluorescein dye has a wide range of applications in industry and biological laboratories. In vivo experiment was planned to assess the sublethal effects of Fluorescein dye, Eosin Y, and Eosin B in Labeo rohita. This freshwater fish was exposed to increasing concentrations (0.0011, 0.0023, and 0.0047 mg/L) of both dyes in the aquarium for definite time intervals. A dye-free trial was set as a control. Fish was anesthetized and dissected by cutting on the ventral side to remove a kidney. Cyto-nephrotoxicity was detected through histopathology. The fish's significant mortality was observed when exposed to 0.0023 mg/L (LC50) and 0.0047 mg/L concentration of Eosin Y dye. These trial groups showed many kidney tissues alterations such as glomerular, tubular epithelial cell's degeneration, a reduced lumen of tubules, vacuolation, absence of bowmen's space, necrosis, glomerulonephritis with increased per tubular space, shrinking of the glomerulus, congestion, clogging, and degeneration of tubules. Eosin B showed less mortality and tissue damage, including glomerular contraction and increased space between glomerulus and capsule. ANOVA showed a highly significant reduction in weight gain of fish at different treatments levels of Eosin Y, but less or none in the case of Eosin B. Increase in fish weight was found to be 66.3 ± 0.38 g when exposed to Eosin Y at 0.0047 mg/L, but it was 69.05 ± 0.63 g when exposed to Eosin B while it was almost identical 69.2 ± 0.23 g for both dye-free trial groups. The pair-wise comparison showed that exposure of the Eosin Y at treatment level 0.0047 mg/L was found to be highly toxic for the kidneys of Labeo rohita. Although both classes of dye are cytotoxic at small concentrations, Eosin Y is acute cyto-nephrotoxic and was found to be involved in growth retardation. To prevent society from fatal consequences, usage of dyes having acute toxicity must be banned or switched to alternates having no effects.

Lipid Extract From a Vegetable (Sonchus Oleraceus) Attenuates Adipogenesis and High Fat Diet-Induced Obesity Associated With AMPK Activation.

Scope: Sonchus Oleraceus, named bitter vegetable (BV), has been known to have multiple health benefits such as anti-aging and anti-inflammation. However, the role of BV in the prevention of obesity is unclear. The aim of this study was to examine the effect of BV lipid extracts (BVL) on obesity development.Methods and Results: Following treatments of high fat diet-induced obese mice (C57BL/6J) with BVL (0.3 mg/g of BW per mouse) for a month, mice exhibited a significant reduction in weight gain, blood triglyceride, and fasting blood glucose compared to control mice. Intriguingly, phosphorylated AMPK, a key regulator of nutrient metabolism, was markedly increased in inguinal fat of BVL group. In 3T3-L1 cells, BVL-7 (100 μg/ml), an omega-3 fatty acid-rich fraction from BVL, lowered lipid accumulation, and down-regulated the gene expression of adipocyte markers. The inhibitory effect of BVL occurred at the early stage of adipocyte differentiation, leading to the delay of mitotic clonal expansion. AMPK knockdown by siRNA abolished the inhibitory effect of BVL-7 on adipogenesis, suggesting that AMPK is essential for BVL-regulated adipocyte differentiation.Conclusion: BVL can effectively inhibit adipogenesis through, at least in part, stimulating AMPK pathway and attenuate HFD-induced obesity. Our findings suggest that BVL can be a promising dietary supplement for protection against obesity, and the effective component of BVL can be potentially developed as anti-obesity drugs.

Genetic ablation of C-reactive protein gene confers resistance to obesity and insulin resistance in rats.

Besides serving as a traditional inflammatory marker, C-reactive protein (CRP) is closely associated with the development of obesity, diabetes and cardiovascular diseases as a metabolic and inflammatory marker. We hypothesise that CRP protein directly participates in the regulation of energy and glucose metabolism rather than just being a surrogate marker, and that genetic deficiency of CRP will lead to resistance to obesity and insulin resistance. Crp gene deletion was achieved by transcription activator-like effector nuclease (TALEN) technology in rats. The Crp knockout animals were placed on either a standard chow diet or a high-fat diet. Phenotypic changes in body weight, glucose metabolism, insulin sensitivity, energy expenditure and inflammation condition were examined. The central impact of CRP deficiency on leptin and insulin hypothalamic signalling, as well as glucose homeostasis, were examined via intracerebral ventricular delivery of leptin and CRP plus glucose clamp studies in the wild-type and Crp knockout rats. CRP deficiency led to a significant reduction in weight gain and food intake, elevated energy expenditure and improved insulin sensitivity after exposure to high-fat diet. Glucose clamp studies revealed enhanced hepatic insulin signalling and actions. Deficiency of CRP enhanced and prolonged the weight-reducing effect of central injected leptin and promoted the central and peripheral roles of leptin. By contrast, reinstatement of CRP into the hypothalamus of the knockout rats attenuated the effects of central leptin signalling on insulin sensitivity and peripheral glucose metabolism. This study represents the first line of genetic evidence that CRP is not merely a surrogate blood marker for inflammation and metabolic syndromes but directly regulates energy balance, body weight, insulin sensitivity and glucose homeostasis through direct regulation of leptin's central effect and hypothalamic signalling.

CHARACTERIZATION OF ELECTRODEPOSITED ZIRCONIA MODIFIED NiCoCrAlY COMPOSITE COATINGS ISOTHERMALLY OXIDIZED AT 1000∘C

The objective of this study is to evaluate the electrodeposited zirconia (ZrO2)-modified NiCoCrAlY composite coatings subjected to isothermal oxidation at 1000∘C under atmospheric conditions for 24 h. NiCoCrAlY composite coatings with 0, 10, 40, and 60[Formula: see text]wt.% of cobalt contents have been compared. The oxidation studies have revealed that the doping of yttrium (Y) and ZrO2 results in a significant reduction in weight gain, thereby imparting improved oxidation resistance. The morphology of the oxides formed on the surface is analyzed using field emission scanning electron microscope and the phases are identified by X-ray diffraction studies. The major oxidation products formed on the surface of NiCrAlY and Ni10CoCrAlY coatings consist of NiO. In contrast, the surface of Ni40CoCrAlY coating comprises of predominantly CoO and small amount of NiCo2O4. Cobalt oxide (CoO) is formed as the major phase on the surface of the Ni60CoCrAlY coating. The grains on the surface of ZrO2-modified NiCoCrAlY composites are faceted, while such grains are not seen in the case of unmodified coatings. The texture analysis reveals that cooling rate is the deciding factor for the facet formation. The coatings have been characterized extensively using X-ray photoelectron spectroscopy to get in-depth understanding.

Potential amelioration of waterborne iron toxicity in channel catfish (Ictalurus punctatus) through dietary supplementation of vitamin C

Iron overload in water is a problem in many areas of the world, which could exert toxic effects on fish. To achieve maximum growth and overall fitness, iron induced toxicity must be alleviated. Therefore, this research was undertaken to investigate the potential mitigation of iron toxicity by dietary vitamin C supplementation in channel catfish (Ictalurus punctatus). Two doses of vitamin C (143 and 573 mg/kg diet) were tested against high environmental iron (HEI, 9.5 mg/L representing 25% of 96 h LC50). Fish were randomly divided into six groups with four replicated tanks. The groups were Control (vitamin C deficient feed), LVc (143 mg vitamin C supplemented per kg diet), HVc (573 mg vitamin C supplemented per kg diet), Con + Fe (control exposed to HEI), LVc + Fe (LVc exposed to HEI) and HVc + Fe (HVc exposed to HEI). Following an 8 week trial, there was a significant reduction in weight gain (WG%) in Con + Fe compared to the control, indicating a toxic effect of HEI on fish growth performance. Interestingly, WG% in both LVc + Fe and HVc + Fe groups were significantly higher than Cont + Fe, signifying that HEI inhibited growth, but this was alleviated by vitamin C. Both hemoglobin content and hematocrit were higher in LVc + Fe compared to the control and Con + Fe. In addition, exposure to HEI (Con + Fe) incited hepatic oxidative stress based on an over-accumulation of malondialdehyde (MDA) along with a significant inhibition in superoxide dismutase (SOD) and catalase (CAT) activities; whereas in LVc + Fe and HVc + Fe, the MDA content restored to basal level. A series of histopathological alterations were observed in the liver and gills, with the most severe lesions in Con + Fe, which was also complemented with a remarkable increase in hepatic iron accumulation. Vitamin C supplementations reduced the augmented concentrations of iron accumulation to that of the control. No effect, regardless of the treatments, was noted for fatty acid composition of muscle. Overall, our findings suggest that the vitamin C supplementation can be an effective therapeutic approach for boosting growth as well as alleviating iron toxicity in catfish.

The influence of the duration of chronic unpredictable mild stress on the behavioural responses of C57BL/6J mice.

The chronic unpredictable mild stress (CUMS) model of depression in mice is a model commonly used to investigate stress-induced depressive-like behaviours. The duration of the stress-inducing procedure is variable, thus making it difficult to compare results and draw general conclusions from different protocols. Here, we decided to investigate how the duration of the CUMS procedure affects behavioural changes, body weight as well as the level of plasma corticosterone in stressed and nonstressed C57BL/6J mice subjected to CUMS for 18 or 36 days. We found that 18 days of CUMS induced a robust decrease in grooming time in the splash test and a significant increase in the immobility time in the tail suspension test (TST) and the forced swim test (FST). All of these stress-induced depression-related behavioural effects diminished or even disappeared after 36 days of CUMS. Plasma corticosterone levels were increased in the CUMS mice compared to those in the nonstressed mice. However, this effect was more pronounced in mice stressed for 18 days. On the other hand, a gradual decline in weight loss in the stressed animals was observed as the duration of the CUMS procedure increased. Altogether, the results indicate that 18 days of CUMS did not affect body weight but caused significant behavioural effects as well as a robust increase in corticosterone levels, while 36 days of CUMS induced significant reduction in weight gain but only slight or even non-significant behavioural effects. These results may indicate the presence of adaptive changes to the long-term CUMS procedure in C57BL/6J mice.

Time Restriction of Food Intake During the Circadian Cycle Is a Possible Regulator of Reproductive Function in Postadolescent Female Rats

ABSTRACTBackgroundWe previously reported that skipping breakfast is associated with menstrual disorders of female college students during postadolescent maturation.ObjectiveIn this study, we investigated the effects of meal timing during circadian cycle on the ovarian function using young female rats.MethodsConsidering that rats are nocturnally active, 8-wk-old female Wistar rats were classified into 3 groups: fed during the daytime only (nonactive phase), night-time only (active phase), or control group I (without time or calorie restriction, free access to a standard caloric diet, 20.0% protein, 62.9% carbohydrate, and 7.0% fat, 3.95 kcal/g) for 4 wk. The changes in body weight and frequency of ovulation in each group were evaluated by a weight scale and a vaginal smear, respectively. At the end of the period of dietary restriction, ovaries were removed, and the numbers of growing follicles (mean diameter >250 µm) and corpora lutea (>600 µm) were examined using hematoxylin-eosin-stained tissue sections. In addition, 8-wk-old female rats were fed only during the night-time for 4 wk under a 20%-reduced food supply of the control group II (without any restriction).ResultsIn the daytime-fed group, the frequency and number of ovulations were significantly decreased compared with those in the control group I (P < 0.05), with a reduced body weight gain concomitant with about 20% of reduction in the daily food intake. In contrast, in the night-time-fed group, even when a 20% reduction in the daily food intake was loaded, their estrus cyclicity did not change despite significant reductions in weight gain and food intake compared with control group II.ConclusionThese findings indicate that restricting food intake to the inactive phase impairs ovarian function in postadolescent female rats, suggesting that the timing of food intake during circadian cycle is one of the crucial factors interfering with the reproductive function.

Down syndrome (DS) in humans is associated with differences of the central nervous system and oromotor development. DS also increases risks for pediatric feeding challenges, which sometimes involve the use of altered food consistencies. Therefore, experimental food consistency paradigms are of interest to oromotor investigations in mouse models of Down syndrome (DS). The present work reports impacts of an altered food consistency paradigm on the Ts65Dn and Dp(16)1Yey mouse models of DS, and sibling control mice. At weaning, Ts65Dn, Dp(16)1Yey and respective controls were assigned to receive either a hard food or a soft food (eight experimental groups, n = 8–10 per group). Two weeks later, mice were assessed for mastication speeds and then euthanized for muscle analysis. Soft food conditions were associated with significantly smaller weight gain (p = .003), significantly less volitional water intake through licking (p = .0001), and significant reductions in size of anterior digastric myofibers positive for myosin heavy chain isoform (MyHC) 2b (p = .049). Genotype was associated with significant differences in weight gain (p = .004), significant differences in mastication rate (p = .001), significant differences in a measure of anterior digastric muscle size (p = .03), and significant reductions in size of anterior digastric myofibers positive for MyHC 2a (p = .04). In multiple measures, the Ts65Dn model of DS was more affected than other genotype groups. Findings indicate a soft food consistency condition in mice is associated with significant reductions in weight gain and oromotor activity, and may impact digastric muscle. This suggests extended periods of food consistency modifications may have impacts that extend beyond their immediate roles in facilitating deglutition.

Effect of dietary taurine supplementation on growth performance, digestive enzyme activities and antioxidant status of juvenile black carp (Mylopharyngodon piceus ) fed with low fish meal diet

An 8-week feeding trial was conducted to determine the effects of dietary taurine supplementation on growth performance, activities of digestive enzymes, antioxidant status and the target of rapamycin (TOR) gene expression in black carp (initial body weight 5.94 ± 0.02 g) fed with low fish meal diet. Six isonitrogen and isolipidic diets were formulated. High fish meal–based diet (HFM) contained 20% fish meal and 24% soybean meal as a positive control. Fifty per cent of fish meal in HFM was replaced by soybean meal and were supplemented with 0, 0.05%, 0.1%, 0.2% and 0.4% dietary taurine respectively (designated as T0.00 (a negative control), T0.05, T0.1, T0.2 and T0.4). The results showed that the partial replacement of fish meal by soybean meal without taurine supplementation resulted in a significant reduction in weight gain (WG), activities of amylase and lipase in intestine, superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) in serum, with a significant increase in feed conversion rate (FCR), the content of malonaldehyde (MDA), triglyceride (TG) and total cholesterol (TC) in serum. WG in groups supplemented with equal or above to 0.1% dietary taurine was significantly higher compared with T0 group. With increasing levels of dietary taurine, the activities of amylase, lipase, GSH-px and SOD and glutataione (GSH) content significantly increased (p < 0.05). FCR, the content of MDA, TG and TC in serum and crude lipid content in whole body were significantly reduced after taurine treatment (p < 0.05). In liver, TOR mRNA expression in groups with equal or above to 0.1% taurine was significantly higher than T0 group (p < 0.05).

Improving the high-temperature oxidation resistance of ASME-SA213-T11 boiler tube steel by plasma spraying with CNT-reinforced alumina coatings

PurposeThis paper aims to discuss that a conventional Al2O3, 1.5 Wt.% carbon nanotubes (CNTs)-Al2O3, 2 Wt.% CNTs-Al2O3 and 4 Wt.% CNTs-Al2O3 composite coatings were deposited with the help of Plasma spray process.Design/methodology/approachTo better understand the effect of CNT reinforcement on oxidation resistance, high-temperature oxidation behaviour of conventional Al2O3, 1.5 Wt.% CNTs-Al2O3, 2 Wt.% CNTs-Al2O3 and 4 Wt.% CNTs-Al2O3 composite coatings at 900°C was compared with the performance of the uncoated ASME-SA213-T11 boiler tube steel substrate.FindingsThe results showed that the CNT-reinforced alumina coatings exhibited better oxidation resistance and thermal stability than uncoated ASME-SA213-T11 boiler tube steel. The coated steel substrates had a lower mass gain rate than the substrate after different oxidation times.Originality/valueLimited literature is available where the CNT have been reinforced into the composite alloy powders and has been thermally spray-deposited for various surface engineering applications. This research showed that with the increase in the percentage of CNTs into the alloy powder mixture, there is a significant reduction in weight gain and hence higher resistance to oxidation.

Evaluation of a novel bacterial biomass as a substitution for soybean meal in plant‐based practical diets for Pacific white shrimp Litopenaeus vannamei

Three growth trials and a digestibility trial were designed to evaluate the efficacy of a novel bacterial biomass (BB) in commercial-type feed formulation for Pacific white shrimp, Litopenaeus vannamei. In trial 1, the basal diet was supplemented with 0, 60 and 120 g/kg BB to replace soybean meal (SBM). Significant improvement was observed in the survival when BB was incorporated in the diets. However, shrimp fed diets containing 120 g/kg BB exhibited significantly lower weight gain (WG) and higher feed conversion ratio (FCR). To confirm the results from trial 1 and explore the effects of BB supplementation at low levels, the basal diet was incorporated with 0, 10, 20, 40, 60 and 120 g/kg BB to replace SBM in trial 2. Significant reductions in WG, FCR, lipid content of whole body, protein retention efficiency and most amino acids retention efficiency were detected in shrimp fed with diet containing 120 g/kg BB. Trial 3 was designed to elucidate whether the digestible protein is the cause of reduced growth. No improvements in terms of growth performance and FCR were detected in the treatments balanced for digestible protein. Apparent digestibility coefficients of energy, protein and amino acid (AA) for BB were determined using chromic oxide as an inert marker and the 70:30 replacement technique. The energy, protein and individual amino acid digestibility coefficients of BB were significantly lower than those of fish meal (FM) and SBM that were given at the same time. Results of this study indicated that BB can be utilized up to 40 g/kg in shrimp feed without causing a decrease in growth. However, supplementations (≥60 g/kg) of BB can result in negative effects on growth response, FCR and protein as well as amino acids retention efficiency. At the lower levels of inclusion, shrimp performance was improved when BB was supplemented on a digestibility basis; however, at the higher level of inclusion, there was no improvement, indicating there may be other nutrients limiting. Based on enhanced survival in the treatment with BB supplementation in trial 1, further research regarding the immune effects of BB in practical shrimp feed will be necessary.

&lt;b&gt;Damage caused by exposure to propofol during gestation of mice

Literature shows that surgical procedure could be necessary at any stage of pregnancy and can cause adverse effects on the mother and fetus. One of the most used anesthetics in surgical centers is propofol however; the safety during pregnancy has not been completely established. The objective of this study was to investigate the possible toxic and teratogenic effects on the intrauterine and post-natal development of mice exposed to the dose of 15 mg kg-1 propofol on the caudal vein fifth, tenth and fifteenth day of gestation. A significant reduction in weight gain was observed in female mice who received a 15 mg kg-1 dose of propofol on the fifth gestational day. A higher rate of embryonic loss post implantation and resorption was also observed in this group. In regards to physical development, the anesthetic increased significantly the offspring weight gain, the time in which pinna detachment occurred, and the anogenital distance of pups whose females received propofol on the fifteenth day of gestation. Based on these results, we concluded that administration of propofol in the beginning stages of gestation increases the number of abortions and promotes alterations in the physical development of pups whose mothers were anesthetized in the final stages of gestation.

Evaluation of dried fermented biomass as a feed ingredient in plant-based practical diets for juvenile Pacific white shrimp Litopenaeus vannamei

Two growth trials were designed to evaluate the utilization of dried fermented biomass (DFB) in commercial type feed formulation for Pacific white shrimp, Litopenaeus vannamei. In trial 1, four experimental diets were formulated to utilize increasing levels (0, 25, 50 and 100 g/kg) of spray-dried fermented biomass (SDFB) as a replacement of fish meal (FM). Results indicated that SDFB can be utilized up to 50 g/kg as a substitution for FM without causing growth depression in shrimp. However, dietary SDFB supplementation at 100 g/kg significantly reduced the weight gain (WG) of shrimp and increased feed conversion ratio (FCR). This reduction in performance is likely due to palatability or nutrient imbalances of the feed. In trial 2, nine experimental diets were formulated with increasing levels (0, 20, 40, 60 and 120 g/kg) of spray-dried (S) or granular (G) DFB to replace soy protein concentrate (SPC) or SPC + corn protein concentrate (CPC). This allowed the comparison between spray-dried and ring-dried products. Ring drying produced a granular product, reducing dust and increasing product particle size. Shrimp fed with diet containing 20 g/kg GDFB performed the best in terms of final mean weight, WG and FCR. Significantly reduced growth and increased FCR were observed in shrimp fed diets containing 60 and 120 g/kg SDFB. Lipid content of whole body was significantly reduced when GDFB was incorporated at 120 g/kg. No significant differences were detected in survival, protein retention efficiency as well as protein and ash contents of the whole shrimp. Results from analysis of covariance indicated that the processing method (covariant) had a significant effect on final mean weight, WG and FCR. In general, shrimp fed with diet containing granular product performed better as compared to those fed with diets utilizing spray-dried product. GDFB can be utilized in the diets up to 120 g/kg in practical shrimp feeds as a substitute for SPC and CPC without compromising the growth of shrimp. However, a significant reduction in WG was observed in the diets containing 60 and 120 g/kg SDFB. The results in the current study demonstrate that processing changes to produce a granular product produced an improved feed ingredient for shrimp.
