Effect of Rhus coriaria L. on performance productive, physiological and carcass characteristics of broiler chicks

A study conducted to estimate effect of Rhus Coriaria L in performance productive, blood profile lipids, carcass characteristics and intestinal flora. A total of 135 chicks Ross308 that divided randomly for three treatments, each treatment divided to three replicates. T1 as control treatment (without supplementation), while T2, T3 received 1, 2 gm/kg diet from sumac powder respectively. The results showed significant superiority in body weight, weight gain and feed conversion in T2, T3 and significant decreased in feed intake in T2. The dressing percentage significantly increase T2, T3, also noted significant increase in breast weight in T3, drumstick weight and abdominal fat percentage in T2. As for the giblets the gizzard showed a significant increase in relative weight. The blood lipids showed significant decrease in TG, HDL, VLDL concentration in T2, T3. The microbiological study showed that number of E. Coli and clostridium significant decrease (P<0.05) in T3 and Staphylococcus decrease in T2, T3, conversely noted significant increase of lactic acid bacteria in T2, T3 compared to control. It can conclude from this study that sumac has a noticeable effect on the productive, physiological performance and intestinal flora of broilers.

Subacute hepatotoxicity of alkaloids extracts of Peganum harmala L. seeds in Wistar albino rats

Peganum harmala L. is a medicinal herbal plant widely used in Algerian traditional medicine. Some reports indicated the toxicity of this plant. In this context, the present study focused on investigate the subacute toxicity of alkaloids extract of P. harmala seeds on histo-function of the liver in rats. Sixteen adult Wistar albino rats were divided equally into four groups, and treated intraperitoneally for 30 days. Group I served as control, received water. Group II, group III, and Group IV received daily a single dose of 50, 100, and 200 mg/kg b.w of crude alkaloids of P. harmala respectively. Blood was collected for the determination of ALT, AST and ALP. Sections of the liver were prepared for histological studies. The results showed a significant decrease in body weights of animals and a significant increase in relative weights of liver in groups III and IV. Treated groups with alkaloids extract of seeds showed a significant increase in the concentration of ALT, AST, and ALP enzymes as compared to control group. These findings were supported with histopathological examination of liver of treated rats. Liver of groups III and IV suffered from severe tissue damage, which included inflammation, cell necrosis, and increase in the volume of some hepatocytes. Some cells contained more than one nucleus and cytoplasm contained micro vacuoles, indicating the onset of steatosis. In conclusion, these biochemical and pathological changes indicate dysfunction with hepatocyte damage. Therefore, the seeds of P. harmala plant must be used in herbal medicine with caution to avoid toxicity to the organism.

Evaluation of mycotoxin binder supplementation on production parameters and organ weights in Toulouse geese with experimental aflatoxicosis

The present study was undertaken to evaluate the beneficial effects of a mycotoxin binder (Mycotox NG 0.05%) in 40 day-old Toulouse geese from both sexes with experimental aflatoxicosis. The birds were reared from day one to 42 days of age on deep litter system and divided into four groups. Normal feed free of aflatoxin (AFB1), was given to the control (Group 1). The feed of Group 2 was supplemented with 0.5 g/kg Mycotox NG, aflatoxin (0.5 mg/kg feed) was supplemented to the feed of Group 3 and Mycotox NG (0.05%) + 0.5 mg/kg AFB1 - to the feed of Group 4. Production parameters (body weight gain, feed intake, feed conversion) and relative organ weights were recorded. The results showed that the total feed intake, final live weight of Mycotox NG + AFB1 treated birds (Group 4) at 6 weeks of age were significantly increased (P&lt;0.01) as compared to birds treated only with AFB1 (Group 3). The total feed conversion ratio of the group given AFB1 only (Group 3) at 6 weeks of age was significantly increased (P&lt;0.01) compared to controls while in Mycotox Ng + AFB1 treated birds (Group-4) it was significantly increased (P&lt;0.01) by post treatment week 1 vs controls, but not as compared to birds treated with AFB1 alone. There was a significant increase in relative weights of liver, kidneys, spleen, heart, pancreas, proventriculus and gizzard in birds fed only aflatoxin (Group 3). The co-administration of Mycotox NG (0.5 g/kg feed) with AFB1 (Group 4) reduced the relative weights of thymus and bursa of Fabricius. The study concluded that dietary supplementation of Mycotox NG could partially neutralise aflatoxicity in geese.

Leaf-Methanolic Extract of Pseudopanax arboreus (Araliaceae) (L. F. Phillipson) Reverses Amitriptyline-Induced Sexual Dysfunction in Male Rats

The people of the Bayangi tribe (Manyu Division) of Cameroon have used the leaves of Pseudopanax arboreus (Araliaceae) traditionally for decades as an aphrodisiac. In order to scientifically investigate this folk claim, we evaluated the effects of the leaf-aqueous extract of P. arboreus on the sexual behavior of normal male rats. The present study was designed to assess the effects of the leaf-methanolic extract of P. arboreus on amitriptyline-induced sexual dysfunction in male rats. Sexually impaired male rats were randomly divided into 4 groups of 8 rats each. Group 1 received 10 ml/kg distilled water, while group 2 was given 6 mg/kg Viagra. Groups 3 and 4 received 46.5 and 93 mg/kg of the leaf-methanolic extract, respectively. Female rats were made receptive by ovariectomy and subsequent hormonal treatment. Sexual behavior parameters were monitored on days 1, 7, 14, and 21 by pairing each male to a receptive female. The extract-treated rats registered significant decrease in mount latency (ML) and intromission latency (IL); nonsignificant increase in mount frequency (MF), intromission frequency (IF), and penile licking (PL); significant decrease in postejaculatory interval (PEI); contrasting effects in ejaculation latency (EL); and mean intromission interval (MII). Both doses of the extract also provoked a significant increase in relative weight of testes, but had no significant effect on the plasma hormonal profile. These findings are similar to those obtained with normal male rats and show that the leaf-methanolic extract of P. arboreus could constitute a potential solution to male sexual impairment.

Acute and Subacute Toxicity Evaluation of Alkaloids of Peganum harmala L. in Experimental Mice

The objective of the study was to evaluate the acute and subacute toxicity of total alkaloids seeds of Peganum harmala in female mice. All morphological, biochemical, hematological, histopathological changes, in addition to mortality and bodyweight changes were recorded. After acute intraperitoneal administration of dose118 mg/kg, there were no any remarkable changes in general appearance and mortality. After 24h, a significant increase in relative weights of heart and brain with no change in hematological, biochemical parameter compared to the normal group. For 5 days a significant reduction in the relative weights of the kidneys and increase of brain with significant change in hematological (MCHC, MCV) and biochemical parameter (AST, ALP and Urea). Subacute study of dose (18 mg/kg) for 28 days showed no remarkable changes in general appearance and deaths occurred in experimental group. A significant increase in relative weight of brain compared to the normal group was observed. In biochemical parameters, a significant increase was seen in both ALT and AST enzyme activities. There was no significant change in hematological parameters. Histopathological examination revealed a ground glass appearance of hepatocytes and the vascular congestion. Alkaloids seeds of P.harmala showed significant toxicity in female mice.

Acute and sub-chronic toxicity study of diaveridine in Wistar rats

Diaveridine, a developed dihydrofolate reductase inhibitor, has been widely used as anticoccidial drug and antibacterial synergist. However, few studies have been performed to investigate its toxicity. To provide detailed toxicity with a wide spectrum of doses for diaveridine, acute and sub-chronic toxicity studies were conducted. Calculated LD50 was 2330 mg/kg b.w. in females and 3100 mg/kg b.w. in males, and chromodacryorrhea was noted in some females before their death. In the sub-chronic study, diaveridine was fed to Wistar rats during 90 days at dietary levels of 0, 23, 230, 1150 and 2000 mg/kg, which were about 0, 2.0–2.3, 21.0–23.5, 115.2–126.9 and 212.4–217.9 mg/kg b.w., respectively. Significant decrease in body weights in both genders at 1150 and 2000 mg/kg groups and significant increases in relative weights of brain in both genders, liver in females, kidneys and testis in males, alkaline phosphatase and potassium in both genders at 2000 mg/kg diet were noted. Significant decrease in absolute weights of several organs, hemoglobin and red blood cell count in both genders, albumin and total protein in females were observed at 2000 mg/kg diet. Fibroblasts in the kidneys, cell swelling of the glomerular zone in the adrenals and inflammation in the liver were found at 2000 mg/kg group. The no-observed-adverse-effect level of diaveridine was 230 mg/kg diet (21.0–23.5 mg/kg b.w./day).

Toxicological evaluation of an olive extract, H35: Subchronic toxicity in the rat

The safety of olive extract H35 containing 35% hydroxytyrosol (HT) was tested in a 90-day oral gavage study in Wistar rats. H35 was administered at 0, 345, 691 and 1381 mg/kg bw/day, equivalent to 0, 125, 250 and 500 mg HT/kg bw/day. Reductions in terminal body weight (9%), and a statistically significant reduction in body weight gain (17%, P < 0.05) at week 13 were observed in high dose males, as well as a statistically significant increase in relative weights of the liver, heart, and kidneys of high dose males and females. These changes were not accompanied by pathological or clinical observations and a trend towards reversal was observed in the recovery phase. H35 was well-tolerated and no toxicologically significant treatment-related changes were observed in condition and appearance of rats, neurobehavioral outcomes, motor activity assessments, functional observational battery (FOB), food intake, ophthalmoscopic examinations, hematology, clinical chemistry, urinalysis, necropsy findings, sperm parameters or estrus cycle. The lowest observed adverse effect level (LOAEL) was the 500 mg HT/kg bw/day based on statistically significant reductions in body weight gain and decreased body weight in males. The no observed adverse effect level (NOAEL) was 250 mg HT/kg bw/day, equivalent to 691 mg/kg bw/day of H35 extract.

Toxicity Studies of the Extracts of Parkia biglobosa Stem Bark in Rats

Extracts of Parkia biglobosa stem bark is used in Nigerian traditional medicine (NTM) to treat malaria, diarrhea and pains. To establish the toxicity profile of the medicine such parameters as the lethal dose (LD50) as well as effects on body functions and organs were evaluated in albino Wistar rats. The bioactive constituents of the water and methanol extracts were also evaluated as a link to toxicity. The LD50 was greater than 5000mg/kg per oral (p.o) for both extracts. No significant (P&lt;0.05) changes in body weights and vital organs of treated animals. However, at 5000mg/kg of water extract, a significant increase in relative weight of the kidneys and hyper -cholesterolemic effects were observed. The extract also elicited significant increase in blood glucose level. The kidneys and livers of animals treated with P. biglobosa water extract for 14 days revealed histopathological evidence of pathological lesions. The methanol extract did not show any changes in the levels of hepatic and hematological parameters, histopathological evidence of pathological lesions, and serum level of urea, uric acid, bilirubin, creatinine and total protein concentrations. Treatment elicited hypo -cholesterolemic effects and significant reduction in blood glucose level occurred in all the groups. The phytochemical screening revealed the presence of tannins, flavonoids, saponins, terpenes, cardiac glycosides, phenols and reducing sugars in the methanol extract, the water extract showed the presence of similar constituents with the absence of flavonoids and cardiac glycosides. This study has shown the toxicity characteristics of the methanol and water extracts of the stem bark P. biglobosa in short time treatment with the extracts.

Acute and sub-chronic oral toxicological evaluations of quinocetone in Wistar rats

To provide a detailed toxicity with wide spectrum of doses for quinocetone, a new antimicrobial growth promoting agent, acute and sub-chronic toxicological studies were conducted. For acute study, quinocetone was administered singly by oral gavage to Wistar rats and Kunming mice. Calculated LD 50 was 8687.31 mg/kg b.w./day in rats and 15848.93 mg/kg b.w./day in mice. In sub-chronic study, quinocetone was fed to Wistar rats at dietary levels of 0, 50, 300 and 1800 mg/kg or olaquindox (300 mg/kg), approximately equivalent to quinocetone 5, 30, 180 or olaquindox 30 mg/kg b.w./day. There was significant decrease in body weight in both genders, total protein and creatinine in females and alkaline phosphatase in males fed with 1800 mg/kg diet, while alkaline aminotransferase values decreased in all treated groups. Significant increase in relative weights of liver and kidneys in both genders and testis in male rats were noted at 1800 mg/kg diet. Histopathological observations revealed that 1800 mg/kg quinocetone diet and 300 mg/kg olaquindox diet could induce proliferation of bile canaliculi in the portal area. In conclusion, quinocetone can induce hepatic histological changes as well as leaking of different serum enzymes. The no-observed-adverse-effect level of quinocetone was considered to be 300 mg/kg diet.

Reproductive functions in the adult male rats after prolonged intraventricular administration of corticotropin-releasing factor (CRF).

The effect of intraventricular injection of synthetic oCRF on reproductive functions in the male rats were studied and changes were compared with intact and control rats (daily intraventricular injections of 10 microliter of 0.1% albumin solution in PBS). The long-term administration of oCRF into lateral ventricle of freely moving rats caused a significant increase in relative weights of adrenals, pituitary glands and a decrease in body weight. Plasma LH and testosterone levels were significantly depressed after 8 days of oCRF administration, while there were no alteration in studied parameters in the control rats. Control i.v.c. injections of 0.1% albumin solution in PBS were without effect on levels of any of the hormones or weights of testes, epidydimis, seminal vesicles and prostate. Obtained results indicate a central inhibitory action of oCRF on LH and subsequent testosterone release.

NINETY-DAY TOXICITY AND ONE-GENERATION REPRODUCTION STUDY IN RATS EXPOSED TO ALLETHRIN-BASED LIQUID MOSQUITO REPELLENT

Rats exposed to liquid mosquito repellent (LMR) containing allethrin (3.6% w/w) 8-hrs/ day for a period of ninety days did not produce any signs of toxicity or death. Significant increases in relative weight of liver and adrenal in males, brain and thyroid in females were observed. No significant changes were noticed in clinical enzyme profile, gonadal enzymes and histopathology of vital organs except mild changes in the activities of liver and serum alkaline phosphates (ALP), testicular glucose-6-phosphate dehydrogenase (G-6PDH) and epididymal sorbitol dehydrogenase (SDH). Rats exposed to LMR for one generation had not produced changes in their reproductive indices such as fertility index, gestation index, live pups/ dam and sex ratio. There was no change in the preweanling evaluation of pups such as survival and growth index on post-natal days (PND) 0, 4, 7,14 and 21. No significant pathomorphological changes were observed in liver, brain, kidney and gonads of PND 21 pups. Absences of any major adverse effects in the adult as well as weanling rats suggest the safe use of allethrin-based LMR.

Experimental Mycotoxicosis in Chickens Induced by Ochratoxin A and Penicillic Acid and Intervention with Natural Plant Extracts

The combined toxic effect of ochratoxin A (OTA) and penicillic acid (PA) on the body mass, the weight and pathomorphology of some internal organs was studied in 85 broiler chickens fed a mouldy diet containing 130, 300 or 800 ppb OTA and 1000-2000 ppb PA. The main pathomorphological changes were cloudy swelling and granular degeneration in the epithelium and mononuclear cell proliferation and activation of capillary endothelium in the kidney and liver; degenerative changes and depletion of lymphoid cells in lymphoid organs (bursa of Fabricius, thymus and spleen) were also seen. Protective effects of 5% total water extract of artichoke and a new natural phytosubstance Rosallsat against these pathomorphological changes were observed. A significant decrease in body mass and relative weight of lymphoid organs was found after 6 weeks of exposure and a greater decrease after 10 weeks of exposure to OTA and PA, and a protective effect of artichoke extract and a slight effect of Rosallsat against that decrease was observed. A significant increase in relative weight of liver and kidneys was also observed as well as a protective effect of artichoke extract against that increase. The quantity of OTA and the percentage of positive samples were significantly lower in tissues of chickens treated with artichoke extract or Rosallsat in addition to OTA than in those treated with only OTA.

The mechanism of lactitol (NS-4) in inducing adrenomedullary proliferative lesion in rats.

We used 13-week repeated oral administration of lactitol as part of a study to clarify the mechanism by which lactitol induces the proliferation of adrenomedullary chromaffin cells. There was a marked increase in urinary calcium (Ca) excretion even though the lactitol administration had no effect on the blood Ca level. A tendency for an increase in adrenal venous blood epinephrine (EPI) and norepinephrine (NE) concentrations was seen. Organ weight measurement of adrenal glands revealed a tendency for an increase in absolute weight and a significant increase in relative weight. Morphometric analysis of adrenomedullary chromaffin cells showed a tendency for an increased total cell volume and a decreased numerical density; but, there was no conspicuous change in the total cell number. Determinations of the anti-bromodeoxyuridine (BrdU) and antiproliferative cell nuclear antigen (PCNA) antibody-positive cell counts showed a tendency for an increased proliferation rate for adrenomedullary chromaffin cells. Electron microscopy showed a slight increase in the number of Golgi apparatuses in these cells. Because the marked increase in urinary Ca excretion was concomitant with morphological changes that suggested the hyperfunction of chromaffin cells in the adrenal medulla and a tendency for an increased cell proliferation rate, we assume that persistent hyperfunction of the adrenomedullary chromaffin cells, which was mediated by enhanced Ca absorption from the intestinal tract, may have induced proliferative lesion.
