Exercise May Promote Skeletal Muscle Hypertrophy via Enhancing Leucine-Sensing: Preliminary Evidence.

Several studies have indicated a positive effect of exercise (especially resistance exercise) on the mTOR signaling that control muscle protein synthesis and muscle remodeling. However, the relationship between exercise, mTOR activation and leucine-sensing requires further clarification. Two month old Sprague-Dawley rats were subjected to aerobic exercise (treadmill running at 20 m/min, 6° incline for 60 min) and resistance exercise (incremental ladder climbing) for 4 weeks. The gastrocnemius muscles were removed for determination of muscle fibers diameter, cross-sectional area (CSA), protein concentration and proteins involved in muscle leucine-sensing and protein synthesis. The results show that 4 weeks of resistance exercise increased the diameter and CSA of gastrocnemius muscle fibers, protein concentration, the phosphorylation of mTOR (Ser2448), 4E-BP1(Thr37/46), p70S6K (Thr389), and the expression of LeuRS, while aerobic exercise just led to a significant increase in protein concentration and the phosphorylation of 4E-BP1(Thr37/46). Moreover, no difference was found for Sestrin2 expression between groups. The current study shows resistance exercise, but not aerobic exercise, may increase muscle protein synthesis and protein deposition, and induces muscle hypertrophy through LeuRS/mTOR signaling pathway. However, further studies are still warranted to clarify the exact effects of vary intensities and durations of aerobic exercise training.

Study of the functional product’s protein compounds digestion features

The aim of the study was to investigate the transformation of meat product’s proteins from pig hearts and aortas during enzymatic hydrolysis in an in vitro model of the gastrointestinal tract. The model consisted of three phases simulating digestion processes: “oral cavity” phase (a-amylase, pH 7.0; 2 min), “stomach” phase (pork pepsin, pH 3.0; 120 min), “intestine” phase (pork pancreatin, pH 7.0; 130 min). The product was sequentially subjected to hydrolysis, at the end of each phase, samples were taken to determine the protein concentration (biuret method) and visualize the protein fractions (one-dimensional electrophoresis). A significant increase in protein concentration at the “stomach” phase was revealed by 3.2 times, and the absolute content by 4.6 times. At the “intestine” phase, a decrease in the number of peptide complexes with copper ions by 1.8 times, the absolute protein content by 8.5% was re‑ vealed. The noted tendency was confirmed by electrophoretic studies — at the stage, simulating digestion in the stomach, the prod‑ ucts of meat product’s proteins hydrolysis were visualized; at the “intestine” phase, a low expression of protein fractions in the range of more than 10 kDa is shown. The maximum hydrolysis of protein compounds at the “stomach” phase to poly- and oligopeptides was confirmed, continuing at the “intestine” stage with the accumulation of free amino acids. This methodology makes it possible to visualize the products of hydrolysis of proteins in a meat product at all stages of the model and to monitor changes in protein concentration in the system.

Concentration of fermented tilapia feces suspension by decomposer bacteria as a feed to induce moina macrocopa sexual reproduction

Sexual induction of M. macrocopa can be induced by setting a density of feed concentration. To optimize the production of ephippia, sexual females must be given sufficient quality feed. Fermentation of tilapia fish from the results of preliminary studies showed a significant increase in protein concentration but fat concentration decreased. This research to determine the best concentration of fermented tilapia feces feed to produce ephippia M. macrocopa. This research is experimental by using a completely randomized design (CRD). This research consisted of 4 treatments of feed concentration is fermented feces suspension concentration of 33,30 mg/L (P1), 37,00 mg/L (P2) and 40,70 mg/L (P3) and control using rice bran suspension feed concentration of 37,00 mg/L(P0), with each using 5 replications. Induction sexual offspring is maintained at a density of 1000 ind/L for 6 days. During cultivation is calculated survival rate, M. macrocopa ephippia production, and some water quality parameters as support. Data were analyzed using ANOVA and Duncan test. This research showed that M. macrocopa cultivation using several concentrations of suspension feed of fermented tilapia feces at concentrations of 40.70 mg/L can induce sexual females and produce ephippia and showed the highest survival rate (1186 ± 26,45 grains/L) and 88,13%.

Comparative pulmonary toxicity of a DWCNT and MWCNT-7 in rats

Very little is known about the in vivo toxicity of inhaled double-walled carbon nanotubes (DWCNTs). In the present study, we compared the pulmonary toxicity of DWCNT to MWCNT-7, a well-known multi-walled carbon nanotube. Rats were divided into six groups: untreated, vehicle, low-dose DWCNT, high-dose DWCNT, low-dose MWCNT-7, and high-dose MWCNT-7. The test materials were administered by intra-tracheal intra-pulmonary spraying (TIPS) every other day for 15 days: the low-dose and high-dose groups were administered final total doses of 0.25 and 0.50 mg/rat of the test material. The animals were sacrificed 1 and 6 weeks after the final TIPS administration. Six weeks after the final TIPS administration, rats administered MWCNT-7 had high levels of macrophage infiltration into the lung with dense alveolar wall fibrous thickening throughout the lung; significant elevation of lactate dehydrogenase activity, alkaline phosphatase activity, and total protein concentration in the bronchioalveolar lavage fluid; an increase in the pulmonary cell PCNA index; slightly elevated levels of 8-OHdG DNA adducts in lung tissue DNA; a small but significant increase in protein concentration in the pleural cavity lavage fluid and an increase in the visceral mesothelial cell PCNA index. None of these parameters was increased in rats administered DWCNT. The primary lesion in rats administered DWCNT was scattered formation of granulation tissue containing internalized DWCNT fibers. Our data indicate that DWCNT has lower pulmonary and pleural toxicity than MWCNT-7.

Beneficial effects of exogenous selenium, glycine betaine and seaweed extract on salt stressed cowpea plant

Pot experiments in sandy soil were carried out to investigate foliar applications of [selenium (5 & 10μM Se), glycine betaine (5 & 10mMGB) and seaweed extract (2 & 4% SWD)] on cowpea plant, under salt stress (0 & 50mM NaCl). Growth, yield parameters and biochemical constituents were determined. The obtained results indicated that all foliar treatments under both salinity concentrations achieved an increment in most of the growth, yield parameters and biochemical constituents. As for the effect of single treatment on growth and yield parameters, 5μM Se without salinity revealed higher significant values than all treatments followed by 10μM Se under 50mM NaCl. Meanwhile, 4% SWE under 50mM NaCl showed the highest values of photosynthetic pigments, proline and Phenylalanine ammonia lyase (PAL). On the other hand, 10mMGB 50mM under NaCl recorded the highest value of total soluble sugars (TSS), while a significant increase in protein concentrations was observed by 5μM Se without salinity.

ASK1 deficiency attenuates hyperoxia‐induced inflammation and cell apoptosis in the lung

RationaleApoptosis signal‐regulating kinase 1 (ASK1), a member of the MAPK kinase (MAP3K) family, is activated by various stimuli including oxidative stress, ER stress, and DNA damage. Inspiration of high oxygen fraction is a palliative therapy to counteract acute lung injury (ALI). Our previous data indicates that ASK1 plays a critical role in this process. However, it is unclear whether or not deletion of ASK1 in vivo protects against HALI. We investigated whether ASK1 deletion would lead to protection against HALI.MethodsASK1 KO mice and control littermates were exposed to 100% oxygen for 48‐72 hours and euthanized to collect bronchoalveolar lavage (BAL) fluid for cell counts and cytokine measurements, and lung tissue collected for assessment of lung epithelial cell apoptosis.ResultsBAL fluid analysis revealed a significant decrease in IL‐1β level in KO mice. Differential cell count shows suppressed inflammatory cell infiltration in KO mice. Lung epithelial cell apoptosis was attenuated in ASK1 KO mice compared to WT mice as determined by TUNEL staining. Moreover, ASK1 KO mice exhibited significant increase in protein concentration in BAL fluid.ConclusionASK1 has a proinflammatory profile. Meanwhile, the permeability increase was observed in ASK1 KO mice which indicate that hyperoxia‐induced increase in permeability is independent of ASK1. Nucleolar Molecular Signature of Pluripotent Stem Cells

Induced pluripotent stem cells (iPSC) are generated by reprogramming somatic cells to the pluripotent state. Identification and quantitative characterization of changes in the molecular organization of the cell during the process of cellular reprogramming is valuable for stem cell research and advancement of its therapeutic applications. Here we employ quantitative Raman microspectroscopy and biomolecular component analysis (BCA) for a comparative analysis of the molecular composition of nucleoli in skin fibroblasts and iPSC derived from them. We report that the cultured fibroblasts obtained from different human subjects, share comparable concentrations of proteins, RNA, DNA, and lipids in the molecular composition of nucleoli. The nucleolar molecular environment is drastically changed in the corresponding iPSC. We measured that the transition from skin fibroblasts to iPSC is accompanied by a statistically significant increase in protein concentrations ~1.3-fold, RNA concentrations ~1.3-fold, and DNA concentrations ~1.4-fold, while no statistically significant difference was found for the lipid concentrations. The analysis of molecular vibrations associated with diverse aminoacids and protein conformations indicates that nucleoli of skin fibroblasts contain similar subsets of proteins, with prevalence of tyrosine. In iPSC, we observed a higher signal from tryptophan with an increase in the random coil and α helix protein conformations, indicating changes in the subset of nucleolar proteins during cell reprogramming. At the same time, the concentrations of major types of macromolecules and protein conformations in the nucleoli of iPSC and human embryonic stem cells (hESC) were found to be similar. We discuss these results in the context of nucleolar function and conclude that the nucleolar molecular content is correlated with the cellular differentiation status. The approach described here shows the potential for spectroscopically monitoring changes in macromolecular organization of the cell at different stages of reprogramming.

Effect of reduction of milking frequency and supplementation of vitamin E and selenium above requirements on milk yield and composition in Assaf ewes

The aim of this research was to study the effect of milking frequency and supplementation with a vitaminmineral complex above requirements on intake, body weight (BW), and milk yield and composition in highyielding Assaf ewes. Sixteen lactating Assaf ewes were used in this study, separated into 4 groups of 4 ewes each. Animals in 2 of the groups (control groups) did not receive any extra vitamin-mineral supplement, whereas animals in the other 2 groups (supplement groups) received daily an oral dose of 1g of vitamin E (1,000IU, dl-α-tocopherol acetate) and 0.4mg of selenium (sodium selenite anhydrous). The experiment consisted of 2 consecutive periods of 3 wk (twice-daily milking in both mammary glands) and 8 wk (once-daily milking in one mammary gland and twice-daily milking in the other gland). Intake, BW, and milk composition were controlled weekly, and milk production was recorded 3 times a week. Administration of the vitamin-mineral supplement had no effect on dry matter intake, BW, or milk production and composition. The reduction of milking from twice to once a day caused a decrease in milk production and lactose concentration and a significant increase in protein concentration, total solids, and somatic cell count, without affecting the fat content. Administration of a vitamin E and Se supplement at the doses used in the present study does not seem to exert, in the short term, a noticeable effect on the mammary gland when milking frequency is reduced.

Inhalation Toxicity and Lung Toxicokinetics of C60 Fullerene Nanoparticles and Microparticles

While several recent reports have described the toxicity of water-soluble C60 fullerene nanoparticles, none have reported the toxicity resulting from the inhalation exposures to C60 fullerene nanoparticles or microparticles. To address this knowledge gap, we exposed male rats to C60 fullerene nanoparticles (2.22 mg/m3, 55 nm diameter) and microparticles (2.35 mg/m3, 0.93 microm diameter) for 3 h a day, for 10 consecutive days using a nose-only exposure system. Nanoparticles were created utilizing an aerosol vaporization and condensation process. Nanoparticles and microparticles were subjected to high-pressure liquid chromatography (HPLC), XRD, and scanning laser Raman spectroscopy, which cumulatively indicated no chemical modification of the C60 fullerenes occurred during the aerosol generation. At necropsy, no gross or microscopic lesions were observed in either group of C60 fullerene exposures rats. Hematology and serum chemistry results found statistically significant differences, although small in magnitude, in both exposure groups. Comparisons of bronchoalveolar (BAL) lavage fluid parameters identified a significant increase in protein concentration in rats exposed to C60 fullerene nanoparticles. BAL fluid macrophages from both exposure groups contained brown pigments, consistent with C60 fullerenes. C60 lung particle burdens were greater in nanoparticle-exposed rats than in microparticle-exposed rats. The calculated lung deposition rate and deposition fraction were 41 and 50% greater, respectively, in C60 fullerene nanoparticle-exposed group than the C60 fullerene microparticle-exposed group. Lung half-lives for C60 fullerene nanoparticles and microparticles were 26 and 29 days, respectively. In summary, this first in vivo assessment of the toxicity resulting from inhalation exposures to C60 fullerene nanoparticles and microparticles found minimal changes in the toxicological endpoints examined. Additional toxicological assessments involving longer duration inhalation exposures are needed to develop a better and more conclusive understanding of the potential toxicity of inhaled C60 fullerenes whether in nanoparticle or microparticle form.

Injections of leptin into rat ventromedial hypothalamus increase adipocyte apoptosis in peripheral fat and in bone marrow

The accumulation of fat cells (adipocytes) in bone marrow is now thought to be a factor contributing to age-related bone loss. Women with osteoporosis have higher numbers of marrow adipocytes than women with healthy bone, and bone formation rate is inversely correlated with adipocyte number in bone tissue biopsies from both men and women. Adipogenic differentiation of bone marrow stromal cells increases with age, but the factors regulating populations of mature adipocytes are not well understood. Leptin is thought to regulate adipose tissue mass via its receptors in the ventromedial hypothalamus (VMH). We have therefore tested the hypothesis that stimulation of leptin receptors in the VMH regulates adipocyte number in bone marrow. Results indicate that unilateral twice-daily injections of leptin into the rat VMH for only 4 or 5 days cause a significant reduction in the number of adipocytes in peripheral fat pads and bone marrow and indeed eliminate adipocytes almost entirely from bone marrow of the proximal tibia. Osteoblast surface is not affected with leptin treatment. Apoptosis assays performed on bone marrow samples from control and treated rats have revealed a significant increase in protein concentration of the apoptosis marker caspase-3 with leptin treatment. We conclude that stimulation of leptin receptors in the VMH significantly decreases the adipocyte population in bone marrow, primarily through apoptosis of marrow adipocytes. Elimination of marrow adipocytes via this central pathway may represent a useful strategy for the treatment and prevention of osteoporosis.

Effects on lipoprotein subclasses of combined expression of human hepatic lipase and human apoB in transgenic rabbits.

The effects of combined expression of human hepatic lipase (HL) and human apolipoprotein B (apoB) on low-density lipoprotein (LDL) subclasses were examined in rabbits, a species naturally deficient in HL activity. In apoB-transgenic rabbit plasma, >80% of the protein was found in the 1.006- to 1.050-g/mL fraction. Gradient gel electrophoresis (GGE) of this fraction revealed two distinct species, designated large and small LDL. A denser fraction (d=1.050 to 1.063 g/mL) contained small LDL as well as another discrete LDL subspecies, designated very small LDL. Expression of HL resulted in reductions in protein concentrations in the 1.006- to 1.050-g/mL density-gradient subfractions containing large (6.5+/-4.1 versus 32.6+/-12.0 mg/dL, P<0.005) and small LDL (59.6+/-17.4 versus 204.3+/-50.3 mg/dL, P<0.002). A concomitant small but not significant increase in protein concentration in the denser LDL fraction (48.0+/-28.2 versus 44.6+/-18.2 mg/dL) was due primarily to an increase in very small LDL (25.9+/-3.1 versus 9.6+/-5.4% of total LDL GGE densitometric area, P<0.002). These findings support a direct role for HL in regulating total plasma LDL concentrations as well as in the production of smaller, denser LDL from larger, more buoyant precursors.

Effectiveness of alfalfa in reducing fertilizer N input for optimum forage yield, protein concentration, returns and energy performance of bromegrass-alfalfa mixtures

Grasses, when grown in association with legumes, may utilize some N fixed by the legumes resulting in improved forage dry matter and protein yield. Field experiments were conducted at Lacombe and Eckville, Alberta, Canada to determine the effectiveness of alfalfa (Medicago sativaLeyss) in reducing fertilizer N requirements for optimum forage dry matter yield (DMY), protein concentration (PC), net margins (returns above N fertilization and forage harvesting costs) and non-renewable energy performance of bromegrass (Bromus inermis Leyss)-alfalfa mixtures. Ammonium nitrate was applied in early spring of 1993 to 1995 at 0, 50, 100, 150 and 200 kg N ha−1 to five bromegrass-alfalfa compositions (pure bromegrass; 2:1, 1:1 and 1:2 ratio of bromegrass:alfalfa; and pure alfalfa) seeded in the summer of 1992. In the zero-N treatment, DMY was lowest in pure bromegrass stands, and increased substantially when alfalfa was grown in association with bromegrass. There was a marked increase in DMY from the application of N fertilizer in pure bromegrass stands, but the increase was much less in the mixed stands. There was a significant increase in PC in forage when bromegrass was grown in a mixture with alfalfa compared to bromegrass alone. Net margins were much greater from mixed stands than from pure bromegrass. In pure bromegrass stands, net margins increased with increasing N rates up to 200 kg N ha−1, but equivalent net margins were usually attained without fertilizer N in bromegrass-alfalfa mixtures as low as 2:1. Energy performance of pure bromegrass stands was substantially improved by including alfalfa in the stands, whereas application of N fertilizer caused a strong and steady decline in energy use efficiency. Our findings indicate that seeding alfalfa in mixed stands with bromegrass can generate savings in N fertilizer (for pure bromegrass stands) equivalent to about 100 kg N ha−1 or more, without any detrimental effect on forage yield, forage quality or net earnings. However, the short-lived nature of alfalfa in bromegrass-alfalfa mixtures remains a cautionary concern. Thus, producers should also adopt management practices that enhance longevity of alfalfa to maximize long-term benefits of using grass-legume mixtures.

A report of Froin's syndrome in five ovine thoracolumbar epidural abscess cases

An epidural abscess involving the thoracolumbar spinal cord was considered the probable cause of pelvic limb paresis in those sheep where analysis of lumbar cerebrospinal fluid (CSF) samples revealed a significant increase in protein concentration (P less than 0.05) and some increase in total white blood cell concentration and neutrophil percentage. There was no significant increase in protein concentration in the corresponding cisternal samples. This phenomenon caused by blockage to CSF flow is not dissimilar to that of Froin's syndrome in man which has been reported as a result of localized spinal meningitis (Brain, 1985).

Long-lasting change in the membrane-associated protein kinase C activity in the hippocampal kindled rat

The effect of hippocampal kindling on protein kinase C (PKC) activity and protein concentration was investigated in rat amygdala/pyriform cortex (AM/PC) and right (contralateral) and left (ipsilateral) hippocampus (HIPP). There was no difference in cytosolic PKC activity between control and kindled groups in any part of the brain. The membrane-associated PKC activity was altered as follows. One week after the last seizure, it was significantly increased in both right (by 26%, P less than 0.05) and left HIPP (by 30%, P less than 0.02). Four weeks after the last seizure, it was significantly increased in the AM/PC (by 14%, P less than 0.02), right HIPP (by 37%, P less than 0.01) and left HIPP (by 24%, P less than 0.05). The protein concentrations in the crude cytosolic extracts prior to elution of PKC through DE-52 columns were significantly increased in the AM/PC (by 11%, P less than 0.05) and right HIPP (by 18%, P less than 0.02) 4 weeks after the last seizure. In the membrane extracts, there was a significant increase by 23% (P less than 0.02) in the left HIPP 1 week after the last seizure. In the fraction co-eluted with PKC, a significant increase in protein concentration of the cytosolic preparation was confirmed in the AM/PC (by 12%, P less than 0.05) as well as in the left HIPP (by 15%, P less than 0.05) 4 and 1 weeks respectively after the last seizure.(ABSTRACT TRUNCATED AT 250 WORDS)

EFFECT OF ABDOMINAL OPERATION UPON PROTEIN EXCRETION IN MAN

Abdominal operations in patients with normal renal function are associated with and followed by a highly significant increase in protein concentration and excretion in the urine. These changes occur in the absence of clinical evidence of impairment of renal function, tend to reach a peak on the third or fourth postoperative day, and return to within normal limits at the time of discharge from hospital. Protein excretion is higher in patients who undergo major operations.
