Immobilisation efficacy of conducted electrical weapons on captive white-tailed deer

Context Wildlife emergencies (e.g. injured animals) often require capture or humane dispatch of animals to resolve the situation. Conducted electrical weapons (CEWs) have the potential to immobilise white-tailed deer (Odocoileus virginianus) for a short duration to avoid potential risks of extended immobilisation (e.g. chemical immobilisation) and increase safety for managers and the public during humane killing by gunshot. Aims To test rapid immobilisation with minimal long-term physiological consequences of CEW immobilisation of white-tailed deer. Methods We arranged five treatment groups of captive deer, including (1) chemical immobilisation with 5-s CEW exposure (n = 5), (2) chemical immobilisation with 15-s CEW exposure (n = 5), (3) 5-s CEW exposure with no chemical immobilisation (n = 10), (4) 15-s CEW exposure with no chemical immobilisation (n = 10) and (5) a control group with no chemical immobilisation or CEW exposure (n = 10). We conducted treatments 1 and 2 to determine short-term physiological effects of CEWs on deer, related to overall welfare. In treatment groups 1 and 2, we sedated deer to collect blood immediately before CEW exposure, and 2 days and 5 days post-exposure for serum biochemical analysis (to measure physiological markers of organ and tissue damage). We observed deer before, during and after treatments to evaluate potential behavioural changes. Key results All deer showed signs of muscle paralysis immediately after exposure to CEW and regained muscle control immediately after the exposure ended. Serum biochemistry results for treatment group 1 and 2 revealed significant increases in creatine kinase (CK) and aspartate aminotransferase (AST) 2 days post-treatment, before returning to pre-exposure concentrations by day 5-post-exposure, suggesting temporary myopathy. By 15-days post-exposure, deer that were only exposed to the CEW had 27 of 39 (69%) remotely fired probes still attached. No long-term behavioural or physiological changes were observed post-treatment. Conclusions Our findings suggest that for immobilisation of white-tailed deer, short-term exposure to a CEW to is a potential alternative to typical capture techniques and can provide sufficient immobilisation to approach and humanely kill deer by gunshot if necessary in wildlife emergencies. Implications As with all capture techniques, trainings and protocols should be developed to ensure the safety of personnel and animals during CEW exposures.

Vernonia amygdalina leaf extract protects against carbon tetrachloride-induced hepatotoxicity and nephrotoxicity: a possible potential in the management of liver and kidney diseases

The rising prevalence of liver and kidney diseases is worrisome and constitutes a major threat to public health. The present study investigates the medicinal potentials of Vernonia amygdalina leaves in the management of liver and kidney diseases. Albino rats were randomly divided into five groups each containing 5 animals. In all groups, except group I (control), animals were exposed to 3 ml/kg bw of CCl4 and then administered further different treatments. Groups III, IV, and V each were treated with 50 mg/kg bw, 100 mg/kg bw of bitter leaf extract, and 100 mg/kg bw of silymarin, respectively. Group II animals were left untreated after exposure to toxicant. Activities of creatine kinase (CK), aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), as well as the level of urea, uric acid, and bilirubin were determined in the serum and tissue homogenates. Lipid profile as well as activities of superoxide dismutase (SOD) and catalase (CAT) were also determined. Exposure to CCl4 resulted in a significant increase in CK, AST, ALP, and ALT levels as well as bilirubin, urea, and uric acid when compared to the control. Lipid profile was disrupted, activities of SOD and CAT were markedly inhibited and the level of GSH was significantly depleted. However, treatment with V. amygdalina reversed the toxic trend in a dose-dependent manner comparable to animals treated with silymarin. In conclusion, V. amygdalina leaf extract restored deranged lipid profile, distorted histoarchitecture as well as liver and kidney function markers. Hence, the plant is a potential candidate for the management of liver and kidney diseases.

The Problem of Drug Interactions Between Rosuvastatin and Ticagrelor in the Aspect of the Risk of Rhabdomyolysis: Discussion of the Problem and Description of the Clinical Case

Background. Combination therapy with two antiplatelet agents (ticagrelor or clopidogrel plus acetylsalicylic acid) and a high dose statin is recommended in accordance with clinical guidelines for patients undergoing acute coronary syndrome and coronary intervention. Combined therapeutic regimens have drug-drug interaction potential. Rhabdomyolysis is a known side effect of statin therapy, and there is evidence that co-therapy with ticagrelor increases the risk of this complication.Case description. A 72-year-old female patient was hospitalized with typical signs of rhabdomyolysis: muscle pain, oliguria, weakness, significant increases in creatine kinase (CK), myoglobin and creatinine. One month before that, she was urgently hospitalized with acute recurrent ST-elevation myocardial infarction and underwent endovascular intervention on a critical stenosis of the left anterior descending artery with stent implantation. After that, rosuvastatin 40 mg per day and ticagrelor 90mg 2 times a day were added to her therapy. During the current hospitalization, rosuvastatin, ACE inhibitors and spirolactone were canceled, infusion therapy was carried out, which led to a rapid regression of symptoms, restoration of adequate diuresis, and normalization of CK, myoglobin and creatinine levels. Conclusions. The combined use of ticagrelor with rosuvastatin (especially at a high dose) increases the risk of rhabdomyolysis in elderly patients. Patients taking ticagrelor may require changes in statin therapy, dose adjustments, and possible drug changes to avoid pharmacological interactions and an increased risk of side effects.

Acute effects of a typical rhythmic gymnastic training session on physiological parameters in Olympic athletes

The aim of this study was to evaluate the effects of a day with two separate training sessions (morning and afternoon) of rhythmic gymnastics on erythrocytes, leukocytes, muscle damage, oxidative stress, and hydration of Brazilian team [age 17.7 (±1.1) years; body height 165 (±0.5) cm; body mass 49.7 (±4.2) kg]. Heart rate and session-ratings of perceived exertion were used to monitor training intensity. Blood samples were collected immediately before (M1) and after (M2) the training day for analyzing erythrocytes, leukocytes, plasma creatine kinase activity, lactate dehydrogenase, alanine aminotransferase, aspartate aminotransferase, ferric reducing ability plasma, thyroid-stimulating hormone, and free T4. Saliva was collected for cortisol analysis. After 24 hours rest (M3), blood collection was performed to analyze creatine kinase and lactate dehydrogenase. The moderate-intensity training day induced significant elevations of total leukocytes (5,163.3 to 9,617.8), lymphocytes (1,752.7 to 2,729.7), neutrophils (2,873.9 to 6,163.6), monocytes (255.7 to 519.1), platelets (280,000.0 to 300,666.7), aspartate aminotransferase (13.1 to 25.6), lactate dehydrogenase (102.5 to 249.1), thyroid-stimulating hormone (1.0 to 3.2), and ferric reducing ability plasma (136.8 to 165.4), as well as significant reductions in red cells (4,691,111.1 to 4,497,777.8), hematocrit (42.1 to 39.3), and hemoglobin (12.9 to 12.5) at M2. There were also significant increases in creatine kinase (144.2 to 519.3) and lactate dehydrogenase (102.5 to 538.2) at M3. The average dehydration rate was 1.3%. A moderate-intensity day of training in rhythmic gymnastics of 8h21min duration caused hemolysis, leukocytosis, muscle damage, redox status perturbations, and insufficient hydration status. These findings show that athletes are exposed to physiological vulnerabilities that can possibly harm their performance and health.

Eccentric Overload during Resistance Exercise: A Stimulus for Enhanced Satellite Cell Activation.

This study aimed to assess if one bout of concentric/eccentric exercise with damaging eccentric overload (CON/ECC+) provides a sufficient stimulus to induce SC activation, proliferation, and differentiation. Biopsies from the vastus lateralis muscle of recreationally active men were obtained in the rested condition and again from the contralateral leg 7 d after exhaustive concentric/eccentric (CON/ECC) (n = 15) or CON/ECC+ (n = 15) leg extension exercise and in a nonexercising control group (CG) (n = 10). Total SC number (Pax7+), activated (Pax7+/MyoD+), and differentiating (myogenin+) SCs, fiber type distribution, and myofibers expressing neonatal myosin heavy chain (MHCneo) were determined immunohistochemically. Creatine kinase and myoglobin were measured in venous blood. Isokinetic strength tests were repeatedly conducted. Significant increases in creatine kinase and myoglobin (P = 0.001) indicated myofiber damage, whereas maximal strength was not impaired. Only after CON/ECC+, SC content (P = 0.019) and SC related to type II fibers (P = 0.011) were significantly increased. A significant increase in the proportion of activated SCs occurred after CON/ECC+ only (P = 0.003), the increase being significantly (P < 0.05) different from the changes after CON/ECC and in CG. The number of differentiating SC and MHCneo remained unchanged. Eccentric overload during leg extension exercise induced significant SC activation, increases in SC content and in SC number related to type II myofibers. However, there were no signs of increased SC differentiation or formation of new myofibers.

The time course of recovery of indirect markers of exercise-induced muscle damage induced by multi- and single-joint exercises

Strength training is performed using multi-joint (MJ) or single-joint (SJ) exercises; however, it is not clear whether different time of recovery is necessary between the two types of exercise. The aim of the present study was to compare the time course of recovery of indirect markers of exercise-induced muscle damage (EIMD) in the elbow flexors after performing MJ and SJ exercises. Twenty-four (n = 24) untrained men were randomized in to the MJ (n = 12) or SJ group (n = 12). Exercise protocol to induce muscle damage consisted of four sets of ten repetitions at 80% of one repetition maximum (1RM) in front pull-down (MJ exercise) or biceps curl (SJ exercise). Maximal voluntary isometric contraction, muscle soreness during elbow extension, ultrasound imaging (muscle thickness and echo intensity) and creatine kinase (CK) were measured before and up to 96 h after exercise. Significant effect of time (p < 0.05) at all times after exercise was observed for isometric strength, muscle soreness, muscle thickness and at 48, 72 and 96 h for echo intensity, with no time × group interaction. However, significant time × group interaction (p = 0.03) was observed only for CK activity at 96 h (MJ = 3348 ± 2911 IU/l vs. SJ = 890 ± 1426 IU/l; p < 0.05). In addition, there was a significant increase in CK after MJ at 48 h, 72 h and 96 h after exercise (p < 0.05), while SJ increased only at 48 h after exercise. Despite a dissimilar time course of CK response, MJ and SJ exercises induced a similar recovery pattern for muscle strength, thickness, echo intensity and soreness.

A Randomized Controlled Trial Investigating the Effects of Undulatory, Staggered, and Linear Load Manipulations in Aerobic Training on Oxygen Supply, Muscle Injury, and Metabolism in Male Recreational Runners

BackgroundTaper is considered as a strategy to avoid overtraining and increase peak performance in athletes. Because aerobic exercise increases the level and duration of independence during the lifespan, the participation of recreational athletes has increased in running events around the world. However, the effects of using load reduction in their training and aerobic performance are still not well known.ObjectivesOur goal was to compare load manipulations, using tapering-like periods in the end of recreational athletes training evaluating alterations in oxygen supply, muscle injury, and metabolism markers.MethodsHealthy males (n = 88, 20–35 years old) were randomly distributed in groups using a combination of two mesocycles of 4 weeks, undulatory and staggered, with two daily microcycles, undulatory and linear. Undulatory-undulatory (Und-Und) and undulatory-linear (Und-Lin) groups had load reduction in the final week of training while the staggered-undulatory (Sta-Und) and staggered-linear (Sta-Lin) groups did not. Cardiorespiratory capacity (V̇O2max), body mass index (BMI), and body fat % were evaluated. Blood samples were also collected to analyze hematocrit (Ht), mean corpuscular hemoglobin (MCHC), circulating levels of hemoglobin (Hb), creatine kinase (CK), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea (U), cortisol (C), free testosterone (FT), and free T/C ratio.ResultsAfter 8 weeks of training, Und-Und had the highest trend to increase V̇O2max. Both Und-Und and Sta-Lin reduced body fat %. Und-Und showed a significant increase in MCHC, T and Free T/C ratio, a positive trend to increase Ht% and Hb levels, and a trend to decrease CK, LDH, and C circulating levels. Sta-Lin presented a trend to decrease Ht% and Hb levels, a significant increase in CK, LDH, AST, ALT circulating levels, and a decrease in Free T/C ratio.ConclusionThe use of daily undulatory training load provides better gains for aerobic fitness while the use of staggered load, mainly associated with linear load, promotes inadequate recovery in recreational runners.

No Effect of Tart Cherry Juice or Pomegranate Juice on Recovery from Exercise-Induced Muscle Damage in Non-Resistance Trained Men.

Tart cherry juice (TC) and pomegranate juice (POM) have been demonstrated to reduce symptoms of exercise-induced muscle damage (EIMD), but their effectiveness has not been compared. This randomized, double-blind, parallel study compared the effects of TC and POM on markers of EIMD. Thirty-six non-resistance trained men (age 24.0 (Interquartile Range (IQR) 22.0, 33.0) years, body mass index (BMI) 25.6 ± 4.0 kg·m−2) were randomly allocated to consume 2 × 250 mL of: TC, POM, or an energy-matched fruit-flavored placebo drink twice daily for nine days. On day 5, participants undertook eccentric exercise of the elbow flexors of their non-dominant arm. Pre-exercise, immediately post-exercise, and at 24 h, 48 h, 72 h and 96 h post-exercise, maximal isometric voluntary contraction (MIVC), delayed onset muscle soreness (DOMS), creatine kinase (CK), and range of motion (ROM) were measured. The exercise protocol induced significant decreases in MIVC (p < 0.001; max decrease of 26.8%, 24 h post-exercise) and ROM (p = 0.001; max decrease of 6.8%, 72 h post-exercise) and significant increases in CK (p = 0.03; max increase 1385 U·L−1, 96 h post-exercise) and DOMS (p < 0.001; max increase of 26.9 mm, 48 h post-exercise). However, there were no statistically significant differences between treatment groups (main effect of group p > 0.05 or group x time interaction p > 0.05). These data suggest that in non-resistance trained men, neither TC nor POM enhance recovery from high-force eccentric exercise of the elbow flexors.

Triglycerides: A Potential Hematological Marker in Animal Models of Cognitive Impairment

Mild Cognitive Impairment (MCI) is a clinical disorder characterized as an intermediate stage of cognitive impairment between normal cognitive aging and more advanced cognitive impairment associated with dementia. MCI causes measurable decline in cognitive abilities including memory and reasoning. Occurrence of MCI has been linked to altered physiological functions, co‐morbid conditions, and adverse events related to therapeutic drugs or toxins. MCI is commonly diagnosed by a physician's clinical judgement based on the following parameters: complete medical history, assessment of independent function and daily activities, input from family members, assessment of mental status (i.e. Mini Mental Status Exam), neurological examinations, and laboratory tests including blood tests and imaging of the brain's structure. The testing required is extensive and often subjective in nature. Currently, there are few biochemical markers thought to correlate with the onset of early‐stage MCI and those that do exist require expensive or invasive techniques to measure accurately. Therefore, using valid animal models representative of cognitive impairment, this study aims to determine the validity of using hematological markers as predictors of early‐stage MCI development.In this study, we measured the hematological markers in animal models of cognitive impairment compared to their respective control group. Cognitive impairment was observed in a chemotherapeutic (CT) rodent group dosed with doxorubicin (2 mg/kg, IP) and cyclophosphamide (50 mg/kg, IP) once weekly for 4 weeks. Cognitive impairment was also observed in a hyperglycemia rodent group dosed with Streptozotocin (STZ) 55 mg/kg, once, IP. Blood samples were withdrawn, and hematological markers analyzed. There was a significant increase in creatine kinase and lactate dehydrogenase (LDH) in the CT model versus control. Glucose levels were significantly increased in the STZ model versus control. With regard to BUN, there was a significant increase in the STZ model compared to their control group, however there was a decrease in the CT model compared to their control group. Interestingly, triglycerides were significantly elevated in both the STZ and CT treated animal models. Previous studies with human subjects have shown a potential link between the increase in triglyceride levels influencing cognitive impairment. Likewise, our data indicates a possible correlation with an increase in triglycerides to cognitive impairment in animal models. This suggests elevated levels of triglycerides may prove to be a potential, minimally invasive, hematological marker for the increased risk of cognitive deficit in these animal models. However, further studies are necessary to determine the causal relationship between elevated triglyceride levels and early‐stage MCI.This abstract is from the Experimental Biology 2018 Meeting. Psychophysiological Response and Fine Motor Skills in High-Altitude Parachute Jumps.

High Alt Med Biol 18:392-399, 2017.-We analyzed the psychophysiological response and specific fine motor skill of an experienced jumper in HALO (high altitude low opening) and HAHO (high altitude high opening) parachute jumps. Eight HALO and eight HAHO jumpers were analyzed. They jumped at 5500 m, HALO jumpers opened the parachute at 500 m and HAHO jumpers at 4300 m of altitude. Before and after the jumps, parameters of muscle strength, cortical arousal, blood creatine kinase (CK) and glucose, blood oxygen saturation, rate of perceived exertion (RPE), and specific fine motor skills of an experienced jumper were assessed; during the jump, heart rate (HR), HR variability, and speed were evaluated. HALO and HAHO jumps produced a significant increase in CK, lactate, and RPE, and a decrease in glucose. HAHO decreased cortical arousal, presented a higher sympathetic modulation, and a higher HR during the jump than HALO. HALO and HAHO produced an increase in the physiological, sympathetic modulation and muscle destruction, and a decrease in cortical arousal and a higher blood lactate concentration only in the HAHO jump. Also, somatic and cognitive anxiety correlated with higher strength manifestation and muscle destruction. This novel research could be used to improve actual training systems in both civil and military parachute jumpers.

Muscle-Tendon Unit Properties during Eccentric Exercise Correlate with the Creatine Kinase Response.

Aim: The aim of this paper was to determine whether; (1) patella tendon stiffness, (2) the magnitude of vastus lateralis fascicle lengthening, and (3) eccentric torque correlate with markers of exercise induced muscle damage.Method: Combining dynamometry and ultrasonography, patella tendon properties and vastus lateralis architectural properties were measured pre and during the first of six sets of 12 maximal voluntary eccentric knee extensions. Maximal isometric torque loss and creatine kinase activity were measured pre-damage (−48 h), 48, 96, and 168 h post-damage as markers of exercise-induced muscle damage.Results: A significant increase in creatine kinase (883 ± 667 UL) and a significant reduction in maximal isometric torque loss (21%) was reported post-eccentric contractions. Change in creatine kinase from pre to peak significantly correlated with the relative change in vastus lateralis fascicle length during eccentric contractions (r = 0.53, p = 0.02) and with eccentric torque (r = 0.50, p = 0.02). Additionally, creatine kinase tended to correlate with estimated patella tendon lengthening during eccentric contractions (p < 0.10). However, creatine kinase did not correlate with resting measures of patella tendon properties or vastus lateralis properties. Similarly, torque loss did not correlate with any patella tendon or vastus lateralis properties at rest or during eccentric contractions.Conclusion: The current study demonstrates that the extent of fascicle strain during eccentric contractions correlates with the magnitude of the creatine kinase response. Although at rest, there is no relationship between patella tendon properties and markers of muscle damage; during eccentric contractions however, the patella tendon may play a role in the creatine kinase response following EIMD.

Creatine Kinase and Lactate Dehydrogenase Responses after Different Resistance and Aerobic Exercise Protocols.

The aim of this study was to investigate the responses of creatine kinase (CK) and lactate dehydrogenase (LDH) after performing different resistance and aerobic exercise protocols. Twelve recreationally trained men (age, 23.2 ± 5.6 years; body mass, 84.3 ± 9.3 kg; body height, 178.9 ± 4.5 cm; and BMI, 26.3 ± 2.3 kg·m2) volunteered to participate in this study. All subjects were randomly assigned to four experimental protocols (crossover): (a) aerobic training at 60% of VO2max, (b) aerobic training at 80% of VO2max, (c) a resistance exercise (RE) session with a bi-set protocol, and (d) an RE session with a multiple sets protocol. Blood samples were collected before, immediately after and 24 hours following the experimental protocols. After 24 hours, there was a significant increase in CK for the 80% of VO2max protocol vs. the bi-set RE session (p = 0.016). Immediately after the protocols, we observed a significant increase in LDH among certain groups compared to others, as follows: multiple sets RE session vs. 60% of VO2max, bi-set RE session vs. 60% of VO2max, multiple sets RE session vs. 80% of VO2max, and bi-set RE session vs. 80% of VO2max (p = 0.008, p = 0.013; p = 0.002, p = 0.004, respectively). In conclusion, aerobic exercise performed at 80% of VO2max appears to elevate plasma CK levels more than bi-set RE sessions. However, the bi-set and multiple sets RE sessions appeared to trigger greater levels of blood LDH compared to aerobic protocols performed at 60% and 80% of VO2max.

Changes in creatine kinase, lactate dehydrogenase and aspartate aminotransferase in saliva samples after an intense exercise: a pilot study.

The aim of this study was to evaluate changes in the enzymes creatine kinase (CK), lactate dehydrogenase (LDH) and aspartate aminotransferase (AST) in saliva before and after an intense exercise consisting of a futsal match. CK, LDH and AST were analyzed in saliva and serum samples of eleven, injury-free, amateur young men before and 30 minutes, 12 hours and 36 hours after a futsal match. A significant increase in CK, LDH and AST was observed after the game in serum samples. In saliva, although a high interindividual variability was found with some individuals no showing increases, significant increases in CK and LDH were observed after the game. No significant changes were observed in saliva AST after the game. Our study showed for first time that CK and LDH can increase in saliva after an intensive exercise consisting on a futsal match. Results suggest that measurements of CK and LDH in saliva could be potentially used to evaluate possible muscle stress or damage in cases of intensive exercise.

Fascicle Lengthening During Eccentric Exercise Determines The Magnitude Of Muscle Damage

The characteristics of the muscle-tendon interaction during eccentric contractions suggests that tendon properties and the magnitude of fascicle lengthening contribute to the extent and the variability of exercise-induced muscle damage (EIMD). The mechanical processes which predispose the severity of EIMD remains unclear. PURPOSE: To determine whether; 1) patella tendon stiffness, at rest and during eccentric contractions, 2) the magnitude of vastus lateralis (VL) fascicle lengthening during eccentric contractions, and/or 3) eccentric torque, are determinants of EIMD. METHODS: Combining dynamometry, electromyography and ultrasonography, patella tendon properties and VL architectural properties were measured pre and during the first of 6 sets of 12 maximal voluntary eccentric knee extensions (range of motion 20 - 90°, 0° = full extension). The eccentric phase of the contraction was performed at an isokinetic angular velocity of 30°s-1 and the concentric phase was performed passively at an angular velocity of 60°s-1. Maximal isometric torque loss and creatine kinase (CK) activity were measured pre-damage (-48 h), 48, 96 and 168 h post-damage as markers of EIMD. RESULTS: Compared to pre-damage, a significant reduction in maximal isometric torque (264 ± 35 Nm, 221.0 ± 48.4 Nm, p = 0.004) and a significant increase in CK (136 ± 114, 796 ± 723 UL, p = 0.014) was reported 48 and 96 hrs post EIMD respectively. Relative changes in CK correlated with the relative change in fascicle length during eccentric contractions (r = 0.53, p = 0.02) and eccentric torque (r = 0.50, p = 0.02). Additionally, the relative change in CK tended to correlate with estimated patella tendon lengthening during eccentric contractions (r = -0.41, p < 0.10). However, relative change in CK did not correlate with resting measures of patella tendon properties or VL properties. Similarly, torque loss did not correlate with any patella tendon or VL properties at rest or during eccentric contractions. CONCLUSION: The extent of fascicle lengthening during eccentric contractions is related to the magnitude of the CK response. Furthermore, at rest, patella tendon properties are not determinants of indices of EIMD. During eccentric contractions of the VL, the patella tendon might play a modulatory role during exercise, to the susceptibility of EIMD.

Does a resistance exercise session with continuous or intermittent blood flow restriction promote muscle damage and increase oxidative stress?

ABSTRACTThe aim of this study was to compare the effect of low-load resistance exercise (LLRE) with continuous and intermittent blood flow restriction (BFR) on the creatine kinase (CK), lactate dehydrogenase (LDH), protein carbonyl (PC), thiobarbituric acid-reactive substance (TBARS) and uric acid (UA) levels in military men. The study included 10 recreationally trained men aged 19 ± 0.82 years who underwent the following experimental protocols in random order on separate days (72–96 h): 4 LLRE sessions at a 20% 1RM (one-repetition maximum [1RM]) with continuous BFR (LLRE + CBFR); 4 LLRE sessions at 20% 1RM with intermittent BFR (LLRE + IBFR) and 4 high-intensity resistance exercise (HIRE) sessions at 80% 1RM. The CK and LDH (markers of muscle damage) levels were measured before exercise (BE), 24 h post-exercise and 48 h post-exercise, and the PC, TBARS and UA (markers of oxidative stress) levels were measured BE and immediately after each exercise session. There was a significant increase in CK in the HIRE 24 post-exercise samples compared with the LLRE + CBFR and LLRE + IBFR (P = 0.035, P = 0.036, respectively), as well as between HIRE 48 post-exercise and LLRE + CBFR (P = 0.049). Additionally, there was a significant increase in CK in the LLRE + CBFR samples BE and immediately after each exercise (Δ = 21.9%) and in the HIRE samples BE and immediately after each exercise, BE and 24 post-exercise, and BE and 48 post-exercise (Δ values of 35%, 177.6%, and 177.6%, respectively). However, there were no significant changes in LDH, PC, TBARS, and UA between the protocols (P > 0.05). Therefore, a physical exercise session with continuous or intermittent BFR did not promote muscle damage; moreover, neither protocol seemed to affect the oxidative stress markers.

Changes In Markers Of Recovery, Readiness, And Nutritional Status In Division I Female Soccer Players Over The First Half Of The Competitive Season

Athletes strive for a competitive edge through proper management and monitoring of training. Biomarkers are emerging as a potential tool for optimally assessing player health, recovery, and readiness. PURPOSE: To evaluate changes in various biomarkers associated with breakdown, recovery, and nutritional status in female collegiate soccer players in the first half of a competitive season. METHODS: Division I female collegiate soccer players (N=29; Mage= 20 ±1.3 yrs; Mweight= 64.7 ± 6.3 kg; Mheight= 166.8 ± 7.2 cm; M%BF= 22.0 ± 3.9 %) participated in blood draws at the start of preseason (T1) the first quarter of the season (T2) and at the midpoint (T3). The athletes arrived fasted and euhydrated the morning after a game. Hematocrit (HCT), IL-6, creatine kinase (CK), total cortisol (CORT), iron (Fe), Vit-D, omega-3-6 ratio (OMG36), and prolactin (PRL) were assessed. All players were monitored for training load using the Polar Team2 System. RESULTS: There were significant changes in HCT, Fe, OMG36, CK, CORT, and PRL (P< .05) over the first half of the season. HCT, Fe, and OMG36 each decreased significantly from T1 to T2 (ΔHCT = -1.4 ± 0.4 %; ΔFe=-45.9 ± 12.9 mcg/dL, ΔOMG36 = -1.8 ± 0.6) and remained significantly lowered at T3, P<.05. CK increased from T1 to T2 (ΔCK = 112.3 ± 41.8 U/L, P<.05) before returning towards baseline by T3, while CORT increased from T1 to T3 (ΔCORT = 6.9 ± 1.0 mg/dL, P<.05). Both markers were above clinical norms in-season. PRL significantly increase from T1 to T3 (5.8 ± 1.5 ng/ml, P<.05). There were no changes in daily training load, though accumulated training load significantly decreased (P< .05) from T2 (3691.9 ± 187.8) to T3 (2566.9 ± 133.9). CONCLUSION: The significant increases in CK, CORT, and PRL are consistent with overreaching and indicate incomplete recovery, despite typical training loads. While monitoring training load is a useful tool to account for the daily sport demands, the biomarkers appear to provide critical additional information to account for cumulative strain and recovery. Based on the changes in Fe and OMG36, nutritional insufficiencies may be partly responsible for impaired recovery. These results are consistent with subjective observations in team performance and provide support for the inclusion of biomarkers in comprehensive athlete monitoring. Supported by Quest Diagnostics

Duchenne myopathy is today the most frequently encountered progressive muscular dystrophy in children, with an inexorable, progressive development to death in the third decade. Improvement in survival is related to improvement in orthopaedic management, early screening of cardiac and respiratory complications, but no curative therapy can be applied today beyond recent pharmacogenetic advances. This diagnosis is raised with evidence of proximal muscular deficit beginning after an interval free of symptoms lasting from 1 to several years. Muscular dystrophy's mechanism is suggested by a significant increase in CK (creatine kinase) and confirmed by muscle biopsy. The clinical motor and cognitive heterogeneity of this disease and its natural history need to be well known because it conditions future therapeutic trials. Identification of outcome measures such as the 6-minute walk test, the MFM score, manual muscle testing musculaire, or biomarkers is indispensable for patient follow-up and collaborative studies.

The Effect of a Simulated Basketball Game on Players' Sprint and Jump Performance, Temperature and Muscle Damage.

Despite extensive data regarding the demands of playing basketball, the relative importance of factors that cause fatigue and muscle potentiation has been explored only tentatively and remains unclear. The aim of this experimental field study was to assess changes in leg muscle power and relate these changes to body temperature modifications and indices of exercise-induced muscle damage in response to a simulated basketball game. College-level male basketball players (n=10) were divided into two teams to play a simulated basketball game. Ten-meter sprint and vertical counter-movement jump tests, core body temperature and creatine-kinase activity were measured within 48 h after the game. The participants’ body temperatures increased after a warm-up (1.9%, p<0.05), continued to increase throughout the game, and reached 39.4 ± 0.4ºC after the fourth quarter (p<0.05). The increase in temperature during the warm-up was accompanied by an improvement in the 10-meter sprint time (5.5%, p<0.05) and jump height (3.8%, p<0.05). The players were able to maintain leg power up to the fourth quarter, i.e., during the major part of the basketball game. There was a significant increase in creatine-kinase at 24 h (>200%, p<0.05) and 48 h (>30%, p<0.05) after the game, indicating damage to the players’ muscles. The basketball players’ sprint and jump performance appear to be at least in part associated with body temperature changes, which might contribute to counteract fatigue during the larger part of a basketball game.

Combinational effect of resveratrol and atorvastatin on isoproterenol-induced cardiac hypertrophy in rats

Introduction:Resveratrol is a natural polyphenol present mainly in grapes. It has been shown to offer strong cardio protection in animal models due to its ability to correct lipid peroxidation and maintain antioxidants level. Atorvastatin, a HMG-CoA reductase inhibitor, lowers cholesterol level and is commonly prescribed to heart patients. Our aim in this study was to see the combination effect of these two drugs against Isoproterenol-induced cardiac hypertrophy in rats.Materials and Methods:Wister Albino rats were treated with resveratrol (20 mg/kg/day, p.o), atorvastatin (20 mg/kg/day, p.o) and in combination (resveratrol [10 mg/kg/day, p.o] + atorvastatin [10 mg/kg/day, p.o]) for a period of 25 days and from 15th till 25th day Isoproterenol (5 mg/kg/day, s.c) was co-administered to rats to induce cardiac hypertrophy.Results:A significant increase in creatine kinase, lactate dehydrogenase, aspartate transaminase and lipid peroxidation with the significant decrease in reduced glutathione, superoxide dismutase and catalase were observed in Isoproterenol treated rats. Resveratrol, atorvastatin and their combination significantly reversed the effect. The histopathological studies and myocardial infarct size evaluation also confirmed the protection.Conclusion:Comparing the data we came to this conclusion that atorvastatin although showed the protection along all the parameters, the extent of protection offered by resveratrol alone and in combination were more effective. Hence, it can be concluded that resveratrol, an herbal nutritional supplement, alone and in combination is better against cardiac hypertrophy.

Rhabdomyolysis as a Clinical Manifestation of Association With Ciprofibrate, Sirolimus, Cyclosporine, and Pegylated Interferon-α in Liver-Transplanted Patients: A Case Report and Literature Review

BackgroundRhabdomyolysis is a syndrome characterized by impaired metabolic integrity of myocytes, causing the release of intracellular constituents into the circulation, and can be a serious side effect of drug intake. Case ReportThis report describes a unique case of rabdomyolysis secondary in which ciprofibrate, sirolimus, cyclosporine, and pegylated interferon-α in a liver transplant patient was used. A 47-year-old male liver transplant recipient in 2009, who had hepatitis C and incidental hepatocellular carcinoma, underwent immunosuppressive therapy (cyclosporine and sirolimus). The patient is currently in treatment for viral recurrence with pegylated interferon-α and ribavirin; he had a history of hypertriglyceridemia treated with ciprofibrate. He had development of severe and generalized myalgia and fever after the eighth application of pegylated interferon-α and increasing doses of cyclosporine. Laboratorial tests showed acute renal failure and significant increase in creatine kinase. Rhabdomyolysis secondary to interaction of fibrate-cyclosporine–pegylated interferon-α was postulated. ConclusionsMedical professionals should be aware of possible drug interactions and should monitor patients receiving these drugs.
