Olig2-enriched exosomes: A novel therapeutic approach for cuprizone-induced demyelination

The research aims to study the therapeutic impact of HEK293-XPack-Olig2 cell-derived exosomes on remyelination of the corpus callosum in a cuprizone-induced demyelinating disease model. A lentiviral vector expressing Olig2 was constructed using XPack technology. The highly abundant Olig2 exosomes (ExoOs) were isolated by centrifugation for subsequent experiments. Western blot, nanoparticle tracking analysis (NTA), and electron microscopy showed no significant difference in particle size and morphology between Exos and ExoOs, and a high level of Olig2 expression could be detected in ExoOs, indicating that exosome modification by XPack technology was successful. The Black Gold/Fluromyelin staining analysis showed that the ExoOs group significantly reduced the demyelination area in the corpus callosum compared to the PBS and Exos groups. Additionally, the PDGFRa/APC staining of the demyelinating region revealed an increase in APC+ oligodendrocytes and a decrease in PDGFRa+ oligodendrocyte progenitor cells (OPCs) in the ExoOs group. Furthermore, there was evident myelin regeneration in the demyelinated areas after ExoOs treatment, with better g-ratio and a higher number of intact myelin compared to the other treatment groups. The level of Sox10 expression in the brain tissue of the ExoOs group were higher compared to those of the PBS and Exos groups. The demyelination process can be significantly slowed down by the XPack-modified exosomes, the differentiation of OPCs promoted, and myelin regeneration accelerated under pathological conditions. This process is presumed to be achieved by changing the expression level of intracellular differentiation-related genes after exosomes transport Olig2 enriched into oligodendrocyte progenitors.

Process optimization for microfluidic preparation of liposomes using food-grade components

This study explores the potential for optimizing a sustainable manufacturing process that maintains the essential characteristics of conventional liposomes using food-grade solvents and components. The focus was comparing the physicochemical, morphological, and interfacial properties of liposomes produced with these food-grade ingredients to those made by conventional methods. It was found that there was no significant difference in particle size (195.87 ± 1.40 nm) and ζ-potential (−45.13 ± 0.65 mV) between liposomes made from food-grade and conventional materials. The manufacturing process for liposomes, utilizing food-grade solvents and components, was optimized through the application of Plackett-Burman design and response surface methodology. This approach helped identify key parameters (soy lecithin, β-sitosterol, W/O ratio) and their optimal values (3.17 g, 0.25 g, 1:2.59). These findings suggest that it is possible to enhance the use of liposomes as an effective and safe delivery system in the food industry, adhering to the strict guidelines set by regulatory agencies.

Enhancing Intrinsic Electrocatalytic Activity of Pt/C Nanoparticles for Oxygen Reduction Reaction in Acidic Media by Microwave-Assisted Synthesis

Abstract: This study is focused on the enhancement of the intrinsic electrocatalytic activity of Pt nano-particles supported on C (Pt/C NPs) towards Oxygen Reduction Reaction (ORR) in acidic media. The goal was to investigate the effect of microwave-assisted synthesis on the electrocatalytic performance of Pt/C NPs towards ORR. Thus, Pt/C NPs were synthesized using a microwave-assisted method and by a conventional heating method; structural and morphological characteristics were analyzed by X-ray dif-fraction (XRD) and transmission electron microscopy (TEM). Electrochemical studies were performed using the rotating disk electrode technique to evaluate the ORR performance. Microwave-assisted syn-thesis produced Pt/C NPs with a smaller particle size (6.3 ± 0.2 nm) than conventionally synthesized nanoparticles (8.6 ± 0.3 nm). Electrochemical analysis showed that the microwave-synthesized Pt/C NPs exhibited higher mass activity (4.6 ± 0.8 mA･g-1Pt) for ORR compared to conventionally synthesized nanoparticles (1.9 ± 0.4 mA･mA･g-1Pt). These results demonstrate that microwave-assisted synthesis en-hances the intrinsic electrocatalytic activity of Pt/C NPs for ORR in acidic media. These findings have important implications for the development of efficient electrocatalysts for fuel cell applications. Background: The synthesis and characterization of platinum nanoparticles on C are crucial for advancing electrocatalysis, particularly in the context of potential applications in fuel cells. This study builds on previous research, focusing on two distinct synthesis methods to enhance our understanding of their im-pact on nanoparticle properties and electrocatalytic performance. Objective: To investigate the synthesis efficiency, structural characteristics, and electrocatalytic activities of platinum nanoparticles on C using microwave-assisted heating and conventional synthesis reactor heating. The objective is to discern any significant differences in particle size, structure, and electrocata-lytic performance between the two synthesis methods. Methods: The synthesis involved a comparative analysis of platinum nanoparticles using microwave-assisted and conventional heating methods. Chemical composition analysis verified the synthesis efficiency, and structural and morphological characterizations were performed using X-ray Diffraction and Transmission Electron Microscopy. Electrochemical studies employed the rotating disk electrode technique, with activation and evaluation conducted through cyclic voltammetry, and the oxygen reduction reaction studied via linear sweep voltammetry in an acidic media (0.5 mol⋅L-1 H2SO4). Results: Well-supported platinum nanoparticles with a face-centered cubic structure were obtained on C using both synthesis methods. However, microwave-synthesized particles (6.3 ± 0.2 nm) exhibited a smaller size compared to conventionally synthesized particles (8.6 ± 0.3 nm). Electrochemical assess-ment revealed superior mass activity for microwave-synthesized material (4.6 ± 0.8 mA･g-1Pt), outper-forming commercial Pt nanoparticles (3.0 ± 0.3 mA･g-1Pt) and conventionally synthesized material (1.9 ± 0.4 mA･mA･g-1Pt). Conclusion: This study concludes that microwave-assisted synthesis yields platinum nanoparticles on C with enhanced electrocatalytic performance, as evidenced by the smaller particle size and superior mass activity compared to conventionally synthesized material and commercial Pt nanoparticles. These findings highlight the potential of microwave-synthesized Pt nanoparticles for applications in fuel cells.

Effects of cavitation jet combined with ultrasound, alkaline hydrogen peroxide and Bacillus subtilis treatment on the properties of dietary fiber

In this study, we compared the effects of three methods on the structural, physicochemical, and functional properties of insoluble dietary fiber (IDF) in okara: cavitation jet plus ultrasonic treatment (UC), cavitation jet plus alkaline hydrogen peroxide treatment (HC), and cavitation jet plus fermentation with Bacillus subtilis (FC). Compared to UC-IDF, the two types of co-modified IDF showed significant differences in particle size and specific surface area. Structural analyses showed that HC-IDF had a porous structure and complex morphological structure, with a significant increase in the absorption peak area at around 3420 cm−1 and 1051 cm−1, and a significant decrease in thermal stability and crystallinity. Of the three treatments, HC most effectively enhanced the physicochemical and adsorption properties of IDF, including the emulsion stability (84.49% ± 3.5%), water-holding capacity (16.04 ± 0.05 g/g), glucose absorption capacity (53.17 ± 3.44 g/g), cholesterol absorption capacity (32.87 ± 0.89 mg/g), and nitrite absorption capacity (211.7 ± 0.17 μg/g). These results demonstrate the potential of HC-modified okara IDF as a valuable functional food ingredient. Therefore, the cavitation jet combined with alkaline hydrogen peroxide method has a good potential for application in the study of dietary fiber modification.

Influence of Different Ratios of DSPE-PEG2k on Ester Prodrug Self-Assembly Nanoparticles for Cell Migration and Proliferation Suppression.

Bufalin (BFL, an active anti-tumor compound derived from toad venom) is limited in its application due to high toxicity and rapid metabolism of the cardiotonic steroid. Ester prodrug self-assembly nanoparticles have shown significant improved effects in addressing the above-mentioned issues. An ester bond was formed between linoleic acid and bufalin to synthesize linoleic acid-bufalin prodrug (LeB). The self-assembly nanoparticles (LeB-PSNs) containing different mass ratios of DSPE-PEG2k and prodrug (6:4, 7:3, 8:2, 9:1 and 10:0) were prepared via co-precipitation method and defined as 6:4-PSNs, 7:3-PSNs, 8:2-PSNs, 9:1-PSNs and LeB-PSNs, respectively. Further, the characterization (particle size, zeta potential, surface morphology and stability) of the nanoparticles was carried out. Finally, we evaluated the impact of different ratios of DSPE-PEG2k on the hydrolysis rate, cytotoxicity, cellular uptake, cell migration and proliferation suppression potential of the prodrug nanoparticles. The linoleic acid-bufalin prodrug (LeB) was successfully synthesized. Upon the addition of DSPE-PEG2k at different weight ratios, both particle size and polydispersity index (PDI) significantly decreased, while the zeta potential increased remarkably. No significant differences in particle size, PDI and Zeta potential were observed among the 9:1, 8:2 and 7:3 PSNs. Notably, the 8:2 (w/w) DSPE-PEG2k nanoparticles exhibited superior stability, hydrolysis and cellular uptake rates, along with efficient cell cytotoxicity, cell migration and proliferation suppression. These findings indicate that DSPE-PEG2k could improve the performance of BFL prodrug nanoparticles, namely enhancing stability and achieving adaptive drug release by modulating the hydrolysis rate of esterase. This study therefore provides more opportunities for the development of BFL application.

Pressure fluctuations in a fluidized bed of binary particles with significant differences in particle size

Pressure fluctuation signals are investigated in a binary particle fluidized bed in which two types of particles have similar physical properties but significant differences in particle size (90 μm Vs. 1800 μm). The relationship of the measured pressure drop to gas velocity exhibits a distinct ‘‘step’’ profile, indicating that the incipient fluidization of binary particle bed can be characterized by different stages, i.e. the fixed bed, the fine particle fluidization, the coarse particle partial fluidization and the binary particle complete fluidization. And then the statistical method and scale decomposition method are used to quantify pressure fluctuation amplitude and frequency in each stage. The result shows that when binary particles are completely fluidized, the bubble size and frequency in binary particle systems are larger than that in sole fine particle systems. Additionally, binary particle systems need higher gas velocity to achieve turbulent fluidization. Furthermore, the local solids holdup measured with optical fiber probes is also employed to verify the result of pressure fluctuation analysis.

Quantification and characterization of microplastics in farmland soils of Jiangsu Province, East China.

The soil ecosystem plays a significant role in the dynamics of microplastics. Qualitative and quantitative analyses of microplastics in six farmland plots from the cities of Nanjing, Suzhou, and Xuzhou were conducted by using laser infrared imaging system (LDIR). The results showed that the abundance of microplastics in farmland soils in Jiangsu Province ranged from 667-9333 items/kg, and the difference between the abundance of microplastics in soils from open-field cultivation and greenhouse farming was not significant. The vertical distribution characteristics showed that the abundance of microplastic decreased significantly with the increase of soil depth. However, there was no significant difference in particle size between top soil (0-5cm) and deep soil (10-20cm). Fragments were the most common microplastic form in soil samples (94.6%), with polyvinyl chloride (PVC) being the main polymer type, followed by polyethylene terephthalate (PET). Compared with previous studies conducted in China, the abundance of microplastics in farmland soils in Jiangsu Province was at moderate level but was notably higher than other places within the Yangtze River Delta region. The conclusions drawn from this paper provided important reference data for future assessment of microplastic pollution in agricultural fields. Furthermore, they establish a fundamental groundwork for understanding the migration patterns of MPs in soil environments.

Emissions and Exposures Associated with the Use of an Inconel Powder during Directed Energy Deposition Additive Manufacturing.

Additive manufacturing (AM) has been linked to potential exposure-related health risks, however, there is a paucity of sufficient research. This study aimed to supply information regarding emissions and exposure during directed energy deposition (DED) AM using inconel 718, with the main constituents being nickel, chromium, and cobalt. By using standardized occupational hygiene methods, the measurement strategy consisted of a combined approach, including powder characterization, particle emission monitoring, and personal exposure monitoring of AM operators. Powder characterization of virgin and used powder indicated no significant difference in particle size, shape, or elemental composition. Particle number emissions ranged between 102 and 105 p/cm3 for submicron particles (<1 µm in size). There was no significant difference in the particle emission rate between the three phases of AM or the two types of DED machines (p > 0.05). The particle emission rate for submicron particles peaked at 2.8 × 109 p/min. Metals of concern to human health were detected during the AM process but were considerably lower than the relevant exposure limits. This study confirms particle emissions, predominantly in the submicron range, above the background concentration during DED AM and, although insignificant in terms of potential health effects, AM operators are exposed to detectable concentrations of the metal constituents of inconel.

Comparing Aerosol Generation in Human and Animal Tissues During Common Otolaryngology Procedures.

To determine whether hamster and human tissues generate similar amounts of aerosolized particles using common otolaryngology surgical techniques. Quantitative experimental research. University research laboratory. Drilling, electrocautery, and coblation were performed on human and hamster tissues. Particle size and concentration were measured during the surgical procedures using a scanning mobility particle sizer and an aerosol particle sizer (SMPS-APS) and GRIMM aerosol particle spectrometer. SMPS-APS and GRIMM measurements detected at least 2-fold increases in aerosol concentrations compared to baseline during all procedures. Procedures performed on human and hamster tissues produced similar trends and order of magnitude of aerosol concentrations. Generally, hamster tissues produced higher aerosol concentrations compared to human tissues, and some of these differences were statistically significant. Mean particle sizes for all procedures were small (<200 nm), although statistically significant differences in particle size were identified between human and hamster tissues during coblation and drilling. Aerosol-generating procedures performed on human and hamster tissue produce similar trends in aerosol particle concentrations and sizes, although we observed some differences between the 2 tissue types. Further studies should be performed to understand the clinical significance of these differences.

Soil organic carbon transfer in aggregates subjected to afforestation in karst region as indicated by 13C natural abundance

The measurement of 13C natural abundance provides valuable information in studying carbon (C) incorporation in soil aggregates. Exploring the pathways of C between aggregates of different size classes are vital to further understand the stabilization of soil organic matter (SOM) and aggregate formation. However, little is known on the mechanism of C stabilization in soil, especially when subjected to afforestation effects in karst rocky desertification area. Soil in the 0–20 cm layer was collected from two afforestation types [natural secondary forest (NF) and artificial Zanthoxylum bungeanum forest (AF)] in the karst rocky desertification area, with cropland (CL) as the reference. Stable isotope fractionation was used to analyze the dynamic changes in organic carbon (OC) in aggregates, and the Δ13C (difference in δ13C between the bulk soil and aggregate fractions) was utilized to quantify the C flow in aggregates under different land use types and soil layers. Results showed that afforestation significantly affected the dynamics of soil aggregate OC. Afforestation on cropland increased the number of large macroaggregates (>2 mm, LMA) and decreased the proportions of small macroaggregates (0.25–2 mm, SMA), microaggregates (0.053–0.25 mm, MI), and silt + clay fraction (<0.053 mm, SC). With the increase in aggregate class size, the OC content in aggregates rose under CL and AF, but no obvious regularity was found in NF. The proportion of new SOC was higher in NF (27.0%) than in AF (20.6%) and in 0–10 cm soil layer (27.3%) than in 10–20 cm soil layer (20.3%) under different afforestation measures. No significant differences in particle size were found among aggregates (P > 0.05). The δ13C in bulk soil and aggregates was significantly decreased by afforestation and increased with the increment in soil depth. Stable isotope analysis results showed that the flow of OC was mainly from larger aggregates to smaller aggregates in CL and AF, which is consistent with the stabilization mechanism of SOM in aggregate hierarchy theory. However, the C flow occurred from smaller aggregates to larger aggregates in NF topsoil due to that newly input OC is first associated with MI and then incorporated into macroaggregates during soil aggregation process. The different human disturbance frequencies and the replenishment levels of plant biomass through litter input, root biomass, and exudates resulted in variation in aggregate turnover (formation and breakdown) among CL, AF and NF. These conclusions can provide sufficient reference for the formation and stabilization of aggregate-associated OC.

Two Routes for Sonochemical Synthesis of Pt-Nanoparticles

To improve the performance of electrochemical hydrogen conversion devices such as electrolyzers and fuel cells, more efficient catalyst materials must be developed. As the available surface area is key to the performance of a catalyst, high surface area nanoparticles are required. The synthesis of such nanostructured catalysts is predominantly based on chemical reduction with a strong reducing agent like sodium borohydride. However, it is difficult to achieve the desired particle properties without the use of additives such as surfactants or stabilizers.In order to achieve more control over the nucleation and growth stage of the nanoparticles, we can replace the chemical reduction route by sonochemical reduction. In sonochemistry, high power ultrasound is used to generate reducing radicals which allow for controlled reduction of metal precursors. The radical generation rate is strongly linked to the nucleation and growth of the resulting nanoparticles and can easily be tuned by adjusting the ultrasonic frequency and power. Ultrasound therefore offers an easy and reproducible way of tuning the size of nanoparticles.In our work we have compared the size, shape, and morphology of Pt-nanoparticles synthesized sonochemically and with chemical reduction to see if the sonochemical route indeed offers better control during nucleation and growth. TEM-images along with X-ray diffractograms revealed that the sonochemical route gives smaller and more monodisperse particles compared to chemical reduction. In addition, the particle shape was found to be spherical with the sonochemical route, whereas chemical reduction resulted in many different shapes. The better and easier control over synthesis parameters exhibited by the sonochemical route can therefore be utilized to produce nanoparticles of very similar physical properties. We also showed that the catalytic properties of these nanoparticles towards hydrogen evolution were very similar further strengthening the sonochemical route when it comes to development of catalyst materials.A comparison between high frequency (408 kHz) and low frequency (20 kHz) ultrasound was also made to determine the impact of the higher mechanical effects expected at lower frequencies. No significant differences in particle size and morphology were found, but agglomerate sizes were found to decrease at lower frequencies. In addition, it was found that extensive probe erosion occurs at 20 kHz resulting in contamination of the nanoparticle solution. In fact, it was shown that the eroded particles act as nucleation sites for Pt-nanoparticles and increased reduction rates were observed. No such particle erosion happened at higher frequencies which led us to conclude that direct sonication at low ultrasonic frequencies in general must be avoided to prevent contamination of your system. This is important in all applications of low frequency (20 kHz) ultrasound, whether it be sonochemistry, dispersion of colloids, or ink preparation for electrochemical characterization.

Superparamagnetic Composite of Magnetite-CTAB as an Efficient Adsorbent for Methyl Orange

In this study, a superparamagnetic composite of magnetite-cetyltrimethylammonium bromide (CTAB) has been prepared by the coprecipitation method and then applied as a charge-selective adsorbent of anionic methyl orange (MO). The VSM (Vibrating Sample Magnetometer) measurement suggests the superparamagnetic property of MNPs (Magnetite Nano Particles) with a magnetic saturation of 49.2 emu g–1. The SEM image exhibits the significant difference in particle size from nanometers in uncoated magnetite to micrometers in magnetite-CTAB. Calculations with ImageJ software indicate that the diameter of the composite is in the range of 2–13 µm, with an average diameter of 6.56 µm, possibly consisting of hundreds to thousands of magnetite-CTAB micelles. The adsorption kinetics of MO over magnetite-CTAB follows the pseudo-second-order adsorption model of Ho and McKay with a rate constant (k2) of 3.54 × 103 g mol–1 min. The adsorption isotherm is well described by the Langmuir model with a Langmuir constant (KL) of 7.46 × 104 L mol and a maximum capacity (qm) of 27.9 mg g–1. The developed material is intriguing because it can be easily and quickly recovered using an external magnet after adsorption and selectively adsorbs anionic dyes.

Enhancement of Anti-Inflammatory Activity of Optimized Niosomal Colchicine Loaded into Jojoba Oil-Based Emulgel Using Response Surface Methodology.

Recent progression in investigational studies aiming to integrate natural products and plant oils in developing new dosage forms that would provide optimal therapeutic effect. Therefore, the aim of the present exploration was to inspect the influence of jojoba oil in boosting the anti-inflammatory effect of colchicine natural product. To our knowledge, there is no formulation comprising colchicine and jojoba oil together to form a niosomal emulgel preparation anticipated for topical application. Colchicine is a natural product extracted from Colchicum autumnale that has been evidenced to show respectable anti-inflammatory activity. Owing to its drawbacks and low therapeutic index, it was preferable to be formulated into topical dosage form. The current study inspected colchicine transdermal delivery by developing niosomal preparation as a potential nanocarrier included into emulgel prepared with jojoba oil. Box Behnken design was constructed to develop 17 niosomal emulgel formulations. The optimized colchicine niosomal emulgel was evaluated for its physical characteristics and in vitro release studies. The in vivo anti-inflammatory activity was estimated via carrageenan-induced rat hind paw edema method. The developed colchicine niosomal preparation revealed particle size of 220.7 nm with PDI value 0.22, entrapment efficiency 65.3%. The formulation was found to be stable showing no significant difference in particle size and entrapment efficiency up on storage at 4 °C and 25 °C for 3 months. The optimized colchicine niosomal emulgel exhibited a pH value 6.73, viscosity 4598 cP, and spreadability 38.3 mm. In vitro release study of colchicine from niosomal emulgel formulation was around 52.4% over 6 h. Apparently, the proficient anti-inflammatory activity of colchicine niosomal emulgel was confirmed via carrageenan-induced rat hind paw edema test. Overall, the results recommend the combination of niosomal preparation with jojoba oil-based emulgel that might signify a favorable delivery of anti-inflammatory drug such as colchicine.

Particle size and mineralogy distributions in respirable dust samples from 25 US underground coal mines

Detailed characterization of respirable coal mine dust is critical to understanding occupational health outcomes, as well as improving exposure monitoring and dust controls in mines. However, data on characteristics such as particle size and mineralogy are still scarce, and there are virtually no datasets available that allow direct comparisons across many mines. Following up on a previous effort to characterize dust from eight underground mines in the Appalachian region of the United States, the current study expands the dataset to cover a total of 25 mines across the country. A total of 171 respirable dust samples were collected in standard locations of each mine and analyzed by scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM-EDX). Results demonstrate that significant differences in particle size and mineralogy distributions exist both within and between mines based on sampling location, mine region and/or mining method–and characteristics can be indicative of dust sources. In locations nearby to production or roof bolting, the respirable dust was clearly sourced from the mine strata. Interestingly, in the production location rock-strata sourced dust appeared to be inordinately abundant relative to the actual coal and rock strata heights being mined during sampling. With respect to particle size, diesel particulates and coal dust were generally found to be finer than mineral dust; and mineral dust likely sourced from the rock strata in the mine was finer than that associated with rock dusting products. On average, when considering all particles analyzed between 100 and 10,000 nm, results indicate that about 75% are in the submicron range, however these particles are estimated to account for only about 6% of the mass.

Development of Mupirocin- Tinidazole solid- Lipid Nanoparticles Loaded Topical gel for the Management of Bacterial Wound Infections

Solid lipid nanoparticles (SLNs) are novel drug carrier system which consists of a solid matrix composed of a lipid being solid at both room and body temperatures with a mean Particle Size (PS) between 50 and 1000 nm Mupirocin -Tinidazole solid-lipid nanoparticles were prepared using hot homogenization technique using Glyceryl monosterate, Stearic acid, Tween 80 and Poloxamer 188 using hot homogenization technique. Size of the nanoparticles was in the range of 83 to 211 nm with the zeta potential values between -2.1 to -5.2. Atomic Force Microscopy (AFM) confirms the spherical shape of solid lipid nanoparticles. Entrapment efficiency was best in the F1 formulation. In vitro release of the pure drug was found to be 75% of mupirocin and 66.5% of tinidazole at the end of 1 hr. Drug release from SLNs dispersion followed Korsermeyrs peppas-model, indicating fickian diffusion drug release, while that from the gel followed non Fickian model drug release. Antibacterial activity of the SLNs was less but the SLNs based gel shows no significant difference in activity to that of standard drug gentamycin against aerobic bacteria. The SLNs dispersion exhibited physicochemical stability under refrigeration upto 45 days without significant difference in particle size. Best formulation was developed into a topical gel using sodium alginate and it was evaluated for pH, viscosity, spreadbility, extrudability, bloom strength, Minimum Inhibitory Concentration (MIC) and Methicillin resistant staphylococcus aureus (MRSA). Extrudability and spreadability parameters of the gel are similar to that of marketed Mupirocin 2% cream formulation

Shelf-stable milk is consumed worldwide, and this market is expected to continue growing. One quality challenge for UHT milk is age gelation during shelf life, which is in part caused by bacterial heat-stable proteases (HSP) synthesized during the raw milk storage period before heat processing. Some Pseudomonas spp. are HSP producers, and their ability to grow well at refrigeration temperature make them important spoilage organisms for UHT processors to control. Previous studies have shown that lactose oxidase (LO), a natural and commercially available enzyme that produces hydrogen peroxide and lactobionic acid from lactose, can control bacterial growth in raw milk. In this research, we investigated the ability of LO to control HSP producer outgrowth, and thus delay age gelation in UHT milk. Six strains of Pseudomonas spp. were selected based on their ability to synthesize HSP and used as a cocktail to inoculate both raw and sterile (UHT) milk at a level of 1 × 105 cfu/mL. Groups were treated with and without LO, stored for 4 d at 6°C, and monitored for cell count and pH. Additionally, a sample from each was tested for HSP activity via particle size analysis (average effective diameter at 90° angle and 658 nm wavelength) and visual inspection on each day of the storage period. The HSP activity results were contrasted using Tukey's HSD test, which showed that in UHT milk, a LO treatment (0.12 g/L) effectively prevented gelation as compared with the control. In raw milk, however, a concentration of 0.24 g/L of LO was needed to obtain a similar effect. This test was scaled up to 19-L pilot plant batches of raw milk where they were challenged with Pseudomonas cocktail, treated with LO for 3 d, and then UHT processed. Resulting UHT milk bottles were monitored for gelation. Significant differences in particle size between the LO-treated samples and the control were observed as early as 1 mo after processing, and gelation was not detected in the LO-treated samples through 6 mo of storage. These results demonstrated that LO can be used to delay age gelation in UHT milk induced by HSP-producing Pseudomonas spp., representing an opportunity to improve quality and reduce postproduction losses in the shelf-stable milk market sector.

Cotransport of heavy metals and SiO2 particles at different temperatures by seepage

The cotransport of heavy metals (HMs; i.e., Pb2+ and Cd2+) and SiO2 suspended particles (SPs) in a porous medium (quartz sand) at different temperatures was investigated by one-dimensional laboratory column experiments. Considering the significant differences in particle size and dielectric property between HMs and SPs, a theoretical model describing their cotransport was developed in this study. The results show that temperature and Darcy velocity have a negligible effect on the transport of individual HMs and that the recovery ratio of Cd2+ is higher than that of Pb2+, which can be attributed to the lower adsorption of Cd2+ on the solid matrix. The presence of SPs can facilitate the transport of HMs due to the strong adsorption of positively charged HMs on negatively charged SPs. However, this effect tends to decrease with the size of the SiO2 particles. An increase in temperature (e.g., T = 20 → 60 °C) results in a decrease in the recovery ratio of HMs because more HMs adsorbed on SPs can be deposited at high temperatures. The adsorption of HMs on SPs can also change the dielectric properties of the SPs, thus causing a reduction in the repulsion between the SPs and the solid matrix and a consequent decrease in the recovery ratio of SPs. In addition, the results show that the developed model fits well with the experimental results.

Exploring the impact of extrinsic lactose fines, a USP modified sampling device and modified centrifuge tube on the delivered dose uniformity and drug detachment performance of a fluticasone propionate dry powder inhaler

The present study aimed to probe influences exerted by extrinsic lactose fines on drug detachment and delivered dose uniformity (DDU) of fluticasone propionate (FP) dry powder inhaler (DPI). Extrinsic lactose fines (0–35%) containing lactose pre-blends were formulated using a low shear 3D shaker mixer and systematically analyzed for various physicochemical properties. Moreover, formulated FP DPIs were assessed for drug detachment by force distribution concept and the DDU using two different sampling devices i.e. the Dosage Unit Sampling Apparatus (DUSA) and USP modified glass sampling device (modified DUSA) coupled with Aerolizer® device at 60 L/min. The physicochemical assessment of control and pre-blend lactose samples displayed significant differences in particle size, surface roughness, degree of surface irregularity and powder flow properties. The formulated FP DPIs displayed monolayer-type FP distribution across the lactose particles in laser Raman spectroscopy with acceptable content uniformity (~100%). Drug detachment study using a modified centrifuge tube showed significant differences in drug detachment profile. The FP DPI formulated using 20% of lactose fines showed the lowest drug detachment force (349.45 gf) as compared to the other lactose samples. Moreover, the FP DPI formulated using 20% of lactose fines demonstrated acceptable mean delivered dose (107.76 and 110.62 μg) with the lowest coefficient of variation using DUSA and modified DUSA, respectively. The present study is the first comprehensive summary of delivered dose uniformity using two different USP sampling devices. Accordingly, these observations may be of high significance for both intuitional and industrial formulators.

Comparison of the ionic conductivity properties of microporous and mesoporous MOFs infiltrated with a Na-ion containing IL mixture.

IL@MOF (IL: ionic liquid; MOF: metal-organic framework) materials have been proposed as a candidate for solid-state electrolytes, combining the inherent non-flammability and high thermal and chemical stability of the ionic liquid with the host-guest interactions of the MOF. In this work, we compare the structure and ionic conductivity of a sodium ion containing IL@MOF composite formed from a microcrystalline powder of the zeolitic imidazolate framework (ZIF), ZIF-8 with a hierarchically porous sample of ZIF-8 containing both micro- and mesopores from a sol-gel synthesis. Although the crystallographic structures were shown to be the same by X-ray diffraction, significant differences in particle size, packing and morphology were identified by electron microscopy techniques which highlight the origins of the hierarchical porosity. After incorporation of Na0.1EMIM0.9TFSI (abbreviated to NaIL; EMIM = 1-ethyl-3-methylimidazolium; TFSI = bis(trifluoromethylsulfonyl)imide), the hierarchically porous composite exhibited a 40% greater filling capacity than the purely microporous sample which was confirmed by elemental analysis and digestive proton NMR. Finally, the ionic conductivity properties of the composite materials were probed by electrochemical impedance spectroscopy. The results showed that despite the 40% increased loading of NaIL in the NaIL@ZIF-8micro sample, the ionic conductivities at 25 °C were 8.4 × 10-6 and 1.6 × 10-5 S cm-1 for NaIL@ZIF-8meso and NaIL@ZIF-8micro respectively. These results exemplify the importance of the long range, continuous ion pathways contributed by the microcrystalline pores, as well as the limited contribution from the discontinuous mesopores to the overall ionic conductivity.

Development of Mupirocin-Tinidazole solid-Lipid Nanoparticles Loaded Topical Gel for the Management of Bacterial Wound Infections

Solid lipid nanoparticles (SLNs) are novel drug carrier system which consists of a solid matrix composed of a lipid being solid at both room and body temperatures with a mean Particle Size (PS) between 50 and 1000 nm Mupirocin-Tinidazole solid-lipid nanoparticles were prepared using hot homogenization technique using Glyceryl monosterate, Stearic acid, Tween 80 and Poloxamer 188 using hot homogenization technique. Size of the nanoparticles was in the range of 83 to 211 nm with the zeta potential values between-2.1 to-5.2. Atomic Force Microscopy (AFM) confirms the spherical shape of solid lipid nanoparticles. Entrapment efficiency was best in the F1 formulation. In vitro release of the pure drug was found to be 75% of mupirocin and 66.5% of tinidazole at the end of 1 hr. Drug release from SLNs dispersion followed korsermeyrs peppas-model, indicating fickian diffusion drug release, while that from the gel followed non fickian model drug release. Antibacterial activity of the SLNs was less but the SLNs based gel shows no significant difference in activity to that of standard drug gentamycin against aerobic bacteria. The SLNs dispersion exhibited physicochemical stability under refrigeration up to 45 days without significant difference in particle size. Best formulation was developed into a topical gel using sodium alginate and it was evaluated for pH, viscosity, spreadbility, extrudability, bloom strength, Minimum Inhibitory Concentration (MIC) and Methicillin resistant staphylococcus aureus(MRSA). Extrudability and spreadability parameters of the gel are similar to that of marketed Mupirocin 2% cream formulation
