In-vitro Antioxidant, In-vitro and In-silico Ovarian Anticancer Activity (Ovarian Cancer Cells-PA1) and Phytochemical Analysis of Cissus quadrangularis L. Ethanolic Extract.

Cissus quadrangularis is a valuable natural source of traditional medicines. An in vitro investigation was performed to determine whether the ethanolic extract from the whole portions of C. quadrangularis had anticancer and free radical scavenging activities against ovarian cancer cells-PA1. C. quadrangularis is a herb collected from rural areas in Andhra Pradesh, India. C. quadrangularis was air-dried and crushed, and the powder and ethanol (0.5 kg) were used in a Soxhlet device for continuous extraction. Phytochemical analysis of the extracts was performed using a standard procedure. The antioxidant activity of the ethanolic extract of C. quadrangularis was evaluated using DPPH. An in vitro anticancer study used an ethanolic extract against the PA1 cell line. Apoptosis of ovarian cancer cells was studied using DAPI and carboxy-H2DCFDA staining. From LC-MS analysis, quercetin-3-O-alpha-Lrhamnopyranoside and erucic acid were docked with the threonine tyrosine kinase (TTK) enzyme using auto docking. The ethanolic extract of C. quadrangularis demonstrated significant dose-dependent antioxidant activity compared to ascorbic acid. The ethanolic extract of C. quadrangularis was found to have high anticancer activity against ovarian cancer cell lines (PA1), with an IC50 value of 482.057 ± 113.857 μg/ml. DAPI and carboxy-H2DCFDA staining confirmed that C. quadrangularis ethanolic extract induced apoptosis in ovarian cancer cells (p < .001). Molecular docking studies helped identify the binding affinities between the protein and ligand complexes, such as Quercetin-3-O-alpha-Lrhamnopyranoside binding sites of target proteins 5N7V (MET602, GLN672) and erucic acid 5N7V (GLY354). Quercetin-3-O-alpha-L-rhamnopyranoside was reported to bind with 5N7V by hydrogen bonding at MET602 and GLN672 amino acids with 2.02, 2.99 Å bonding length distance and binding affinity of -7.9 kcal/mol. Erucic acid was reported to bind with 5N7V by hydrogen bonding at GLY354 amino acid with 3.18, 2.93 Å bonding length (Å) distance and binding affinity of -4.3 kcal/mol. The current analysis showed that the ethanolic extracts of C. quadrangularis L. exhibited antioxidant and anticancer properties against ovarian PA1 cells. The experimental results confirmed that C. quadrangularis L. is a promising, safe chemotherapeutic plant for ovarian cancer PA1 cells. The docking results demonstrated that Quercetin-3-O-alpha-L-rhamnopyranoside strongly binds threonine tyrosine kinase at the MET602 and GLN672 positions. This study showed that the C. quadrangularis ethanolic extract has Quercetin-3-O-alpha-L-rhamnopyranoside, which can be used as an anticancer agent.

Morphological and Molecular Characterization of Spirogyra Species from Water Bodies in Chiang Rai Province, Thailand: Insights into Bioactivity and Antioxidant Potentialin

This study aimed to identify Spirogyra species collected from lentic (sampling site code CR01) and lotic (sampling site code CR02) water areas in Chiang Rai Province, Thailand. The diagnosis was proceeded by studying the morphological characteristics combined with molecular techniques with the chloroplast rbcL gene (ribulose bisphosphate carboxylase). Additionally, the bioactive properties of Spirogyra extracts were evaluated through antimicrobial and antioxidant assays. The extraction was performed with water and ethanol extraction. The results indicated that the morphological characteristics of Spirogyra CR01 resembled Spirogyra neglecta, but the data of rbcL gene for this species were not available in the database. Therefore, the Spirogyra CR01 sequence was added to the GenBank database, representing the report of rbcL gene of Spirogyra neglecta sequencing data. Spirogyra CR02 was robustly identified as S. fluviatilisthrough molecular analysis. The extracts from both Spirogyra samples showed that the ethanolic extract has more activity than the water extract. The antimicrobial ability of the ethanolic extract from Spirogyra CR02 was observed to inhibit the growth of Bacillus cereus and Staphylococcus aureus, while the ethanolic extract from Spirogyra CR01 only inhibited B. cereus. Moreover, the ethanolic extract of Spirogyra CR01 had statistically significant (p&lt;0.05) antioxidant activity, with the highest value recorded at 114.92±2.38 mg GAE/g extract. Overall, this study provides new sequencing information on the S. neglecta and their potential for use Spirogyra in functional food, functional ingredients and cosmeceutical products.

Antioxidant and immunomodulatory potential of cell wall polysaccharides from Harungana madagascariensis Lam

Numerous plant polysaccharides have demonstrated immunomodulatory and antioxidant potentials, and can be investigated as novel molecules with biological properties that may enhance immune function. The aim of this study was to assess the antioxidant and immunomodulatory properties of cell wall polysaccharides fractions isolated from Harungana madagascariensis (HM). Pectin fraction (PF) and hemicellulose fraction (HCF) were extracted and tested for their antioxidant properties using reducing power, 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2′-azino-bis-3-éthylbenzylthiazoline6-sulphonic acid (ABTS) radical scavenging assays, chelating activity and inhibition of lipid peroxidation. Their immunomodulatory activities were carried out on peritoneal macrophages targeting NADH oxidase and lysosomal enzyme activities as well as the production of nitric oxide (NO) and hydroperoxides (H2O2) during phagocytosis of Saccharomyces cereviceae. The polysaccharides fractions showed significant antioxidant and immunostimulatory activities via stimulation of NADH oxidase and lysosomal enzyme of peritoneal macrophages and production of NO and H2O2. Taken together, the results of this study indicate the potential of polysaccharides from H. madagascariensis as antioxidant and immunomodulator agents.

Investigation of in-vitro antioxidant activity and heavy metals present in Ficus mollis leaves

The folklore medicinal plant leaves of Ficus mollis were selected for the study. We were performed for elemental analysis, i.e., macronutrients and toxic and heavy metal contents. All the values are within the safe limits. The plant specimens were successively extracted with solvents such as petroleum ether, Chloroform, ethyl acetate, and Ethanol. The preliminary phytochemical studies of FML confirmed the presence of flavonoids, tannins, steroids, glycosides, alkaloids, and triterpenoids. The present study also investigated FML antioxidant activity using in vitro methods. The EEFML showed significant antioxidant activity in all models studied compared with ascorbic acid. It possesses good activity in scavenging DPPH radical, Hydroxyl radical, Nitric oxide radical, and scavenging H2O2.Further investigation of individual Phyto compounds, their isolation, identification, and their role as free radical scavengers is needed.

Antioxidative Potential and Ameliorative Effects of Rice Bran Fermented with Lactobacillus against High-Fat Diet-Induced Oxidative Stress in Mice

Rice bran is an important byproduct of the rice polishing process, rich in nutrients, but it is underutilized and often used as feed or discarded, resulting in a huge amount of waste. In this study, rice bran was fermented by Lactobacillus fermentum MF423 to obtain a product with high antioxidant activity. First, a reliable and efficient method for assessing the antioxidant capacity of the fermentation products was established using high-performance liquid chromatography (HPLC), which ensured the consistency of the batch fermentation. The fermented rice bran product (FLRB) exhibited significant antioxidant activity in cells, C. elegans, and hyperlipidemic mice. Transcriptome analysis of mouse livers showed that the expression of plin5 was upregulated in diabetic mice administered FLRB, thereby preventing the excessive production of free fatty acids (FFAs) and the subsequent generation of large amounts of reactive oxygen species (ROS). These studies lay the foundation for the application of rice bran fermentation products.

Phenolic Compound Characterization and Biological Activities of Mistletoe (Viscum album L.) Ethanol Macerates Used in Herbal Spirit Production

Biska, a traditional Istrian herbal spirit, is produced by macerating mistletoe leaves and twigs in a water–ethanol base, usually grape marc spirit. The aim of this study was to determine the optimal mistletoe maceration parameters (ethanol content, concentration of plant material, and duration) for the production of biska, which have not yet been the subject of scientific research. The prepared mistletoe macerates and the collected biska samples were subjected to phenolic compound characterization, including total phenolic content (TPC), total flavonoid content (TFC), and polyphenolic profile, by ultra-high-performance liquid chromatography coupled with mass spectrometry (UPLC-ESI-MS2). In addition, the macerates and the biska samples were analyzed for their biological properties: antioxidant and antiproliferative activity. The results showed that both the macerates and the biska samples contain phenolic compounds of mistletoe such as phenolic acids, flavonoids, and coumarins, which contribute to the observed biological activities—significant antioxidant activity and antiproliferative effect. By applying the maceration parameters, which yielded the highest content of phenolic compounds as well as volatile aromatic compounds (40–80 g/L of mistletoe, 40–55% v/v ethanol, and 21 days’ maceration time), the production process can be improved to produce biska with better aromatic and biological properties.

Phytochemical profiling, antioxidant, and tyrosinase inhibitory potential of the Acacia cyclops trunk bark: in vitro combined with in silico approach.

The aim of this study was to analyze the phytochemical profile of Acacia cyclops trunk bark ethyl acetate extract using LC-tandem mass spectrometry for the first time, along with evaluating its antioxidant and anti-tyrosinase properties. Consequently, we determined the total phenolic and flavonoid contents of the extract under investigation and identified and quantified 19 compounds, including phenolic acids and flavonoids. In addition to assessing their antioxidant potential against DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2'-azino-bis-[3-ethylbenzothiazoline-6-sulfonic] acid) assays, in vitro and in silico studies were conducted to evaluate the tyrosinase inhibitory properties of the A. cyclops extract. The ethyl acetate trunk bark extract exhibited a substantial total phenolic content and demonstrated significant antioxidant activity in terms of free radical scavenging, as well as notable tyrosinase inhibitory action (half-maximal inhibitory concentration [IC50] = 14.08 ± 1.10 μg/mL). The substantial anti-tyrosinase activity of the examined extract was revealed through molecular docking analysis and druglikeness prediction of the main selected compounds. The findings suggest that A. cyclops extract holds promise as a potential treatment for skin hyperpigmentation disorders.

GC-MS profiling and biological investigations of Cissampelos pareira L. & Lantana camara L.

The objectives of the study were to conduct phytochemical screening of crude extracts of Cissampelos pareira L. and Lantana camara L. and assessing their biological potentials against protein kinase enzymes, followed by the evaluation of antioxidant and antimicrobial capacities. The n-hexane (A-І, B-І) and ethyl extracts (A-ІІ, B-ІІ) displayed moderate to good antioxidant activity, while the methanol fractions (A-ІІІ, B-ІІІ) showed significant antioxidant activity. Among all the three crude extracts, the ethyl acetate extract (B-II) of L. camara exhibited significant protein kinase inhibitions with remarkable antioxidant potential and was therefore purified through column chromatography. Fraction B-ІІ-2 showed the highest flavonoid contents (735.43), with the most promising protein kinase inhibition exerted by the subtractions B-ІІ-2, B-ІІ-3, B-ІІ-4, and B-ІІ-7. Hence, active fractions were examined through GC-MS for the identification of active constituents, revealing 17 active compounds. In the case of Lantana camara, the ethyl acetate crude extract along with fractions led to the identification of more than thirty compounds, which may be further explored to find a potential lead for drug development.

First report of antioxidant potential of peptide fraction derived from Colaconema formosanum (Rhodophyta) protein hydrolysates

Foods that contain natural antioxidants can improve people's health and performance and make them more resistant to environmental stressors like diseases, lousy living conditions, and heat stress. Marine algae are well known for their function as an antioxidant. Colaconema formosanum is a new rhodophyte discovered in Taiwan and its bioactive activities are rarely studied. This work aims to prepare the protein hydrolysate fraction with antioxidant properties in C. formosanum using single enzyme or complex enzyme hydrolysis. Afterward, a 3 kDa molecular weight cut-off (MWCO) was used for centrifugation, and then fractionation was carried out using Strong Cation Exchange (SCX) Chromatography. The quantification of antioxidant activity was performed using the DPPH test. The results showed that the thermolysin hydrolysate (IC50 1.48 ± 0.92 mg/mL) and the combination of trypsin-thermolysin hydrolysates (IC50 1.37 ± 0.84 mg/mL) had the highest antioxidant activity. The fractionation procedure with strong cation exchange (SCX) yielded fraction C from thermolysin hydrolysates, which exhibited the most significant antioxidant activity with an IC50 value of 0.47 ± 0.02 mg/mL. The fraction 4 derived from trypsin-thermolysin hydrolysates exhibited the most potent antioxidant activity (IC50: 0.34 ± 0.002 mg/mL). No previous reports have been made about the antioxidant properties of the hydrolysate fractions derived from C. formosanum proteins. Consequently, these findings could serve as a foundation for exploring peptides that have potential therapeutic sources for reducing oxidative stress.

PHYTOCHEMICALS SCREENING, ANTIOXIDANT AND ANTIMICROBIAL ACTIVITY OF CARICA PAPAYA LEAF EXTRACTS

Objective: The objective of the present study aimed at investigating the phytochemical antioxidant and anti-microbial properties of Carica papaya leaf extracts. Methods: As phytochemicals are biologically active compounds and a powerful group of plant chemicals believed to stimulate the immune system along with antioxidants, the molecules which hinder oxidation of other molecules by the process of inhibiting or by generating the oxidizing chain reactions and preventing diseases. The anti-microbial activity on various gram-positive and gram-negative bacteria were determined using zone of inhibition and antioxidant by the 2,2,1-diphenyl-1-picrylhydrazyl method. Results: Phytochemical screening, antioxidant and antibacterial potential were determined using different aqueous and organic solvents in addition to the determination of trace element in leaves of C. papaya. Antioxidant activity using DPPH free radical scavenging assay indicated leaves extracts leaves showed inhibition of per oxidation. The result showed that the ethanolic extract of C. papaya possessed a significant antioxidant activity as compared to methanol and aqueous extract. The antibacterial study showed leaves extract is the best to cope infectious action of bacteria. Conclusion: This study was conducted to test the medicinal profile of C. papaya by extracting secondary metabolites with organic and aqueous solvents. Ethanol was found to be the best solvents of choice to extract natural products to get maximum medicinal benefits and could be used to medicinal formulation against different infectious diseases.

Valorization of guabirobeira leaf extracts (Campomanesia xanthocarpa) through pressurized liquid extraction and aqueous infusion: assessment of phytochemical composition, antibacterial, and antioxidant activities

AbstractBACKGROUNDThe leaves of guabirobeira contain important phenolic and flavonoid compounds, known for their antioxidant, anti‐inflammatory, antibacterial, and antinociceptive activities. The purpose of this work was to obtain extracts from guabirobeira leaves through aqueous infusion and pressurized liquid extraction with water, ethanol, and a combination of water–ethanol (ratio 1:1, v/v; PLE‐W:E). Subsequently, we used liquid chromatography coupled with mass spectrometry to identify the chemical compounds, and to study the antioxidant activity in rat brain cells; thiobarbituric acid reactive substances (TBARS) and reactive oxygen species (ROS) antibacterial activity against Gram‐negative and Gram‐positive bacteria were also seen.RESULTSAccording to the findings, all four extracts contained a significant amount of phytochemical compounds. Among them, the PLE‐W:E extract showed a higher concentration of flavonoids, demonstrating significant antioxidant activity in in vitro assays of TBARS and ROS in rat brain cells. It also exhibited antibacterial activity against both Gram‐positive and Gram‐negative bacteria.CONCLUSIONGuabirobeira leaf extracts hold promise as a potential source of bioactive compounds. These extracts can be applied across various sectors, including cosmetics and skincare products, pharmaceuticals, food and beverages, natural preservatives, and functional materials. By exploring these potential applications, valuable contributions can be made to the field of sustainable chemistry and pharmacy, furthering the utilization of guabirobeira leaf extracts in diverse industries. Cadmium oxide nanoparticles from new organometallic Cd(II)-Schiff base complex and in vitro biological potentials: dual S. aureus and E. coli DNA gyrase inhibition by the precursors via in silico binding modes’ study

AbstractAt the nanoscale level, several biological processes take place, owing to the potential that engineered nanomaterials might interrelate with bio-molecules and cellular procedures. This study aimed to synthesize cadmium oxide nanoparticles via a one-step calcination process of tetradentate Schiff base-Cd(II) complex at different temperature ranges. The as-synthesized compounds were carried out via a viz UV–visible, elemental analysis, 1H NMR, molar conductivity, transmission electron microscopy (TEM), FT-IR spectroscopy, and X-ray diffraction (PXRD). The band gap energy and average particle sizes of the CdO particles are respectively (2.69 eV, 3.54 eV), 26.88 nm for CdO@250, and (3.20 eV, 3.57 eV), 25.67 nm for CdO@300, while CdO@350 exhibited the 3.78 eV and 28.42 nm values. The antioxidant accomplishments of the test samples through the scavenging activity of DPPH radicals showed CdO@300 to possess (IC50 = 5.18 ± 0.56 µg/mL). Similarly, the as-synthesized CdO nanoparticles exhibited higher antibacterial activities against S. aureus and E. coli as compared to the corresponding Cd-HMB and ligand (HMB), while ciprofloxacin acted as a standard antibiotic. Furthermore, HMB and its complex Cd-HMB were docked against the DNA gyrase enzymes of S. aureus (PDB IDs: 5CDQ) and E. coli (PDB IDs: 6F86) as receptors. The binding sites docking results showed that the binding energies of HMB and Cd-HMB to 5CDQ ranged from − 3.44 to − 4.99 kcal/mol and from − 6.45 to − 6.64 kcal/mol, while the binding energies related to the target 6F86 are in the ranges of (− 3.64, − 4.76) kcal/mol and (− 6.08, − 6.09) kcal/mol respectively. Therefore, the significant antioxidant and antibacterial activities of the ligand (HMB), Cd-HMB, and CdO NPs review the broad application prospects of these compounds as therapeutic agents for wide-ranging biomedical applications.

Identification, structure elucidation, and isomer differentiation of phenolic compounds of Chinese pepper cultivars by UPLC-ESI-Q-TOF-MS and their antioxidant activity.

A total of 43 compounds, including phenolic acids, flavonoids, lignans, and diterpene, were identified and characterized using UPLC-ESI-Q-TOF-MS coupled with UNIFI software. The identified flavonoids were mostly isomers of luteolin, apigenin, and quercetin, which were elucidated and distinguished for the first time in pepper cultivars. The use of multivariate data analytics for sample discrimination revealed that luteolin derivatives played the most important role in differentiating pepper cultivars. The content of phenolic acids and flavonoids in immature green peppers was generally higher than that of mature red peppers. The pepper extracts possessed significant antioxidant activities, and the antioxidant activities correlated well with phenolic contents and their molecular structure. In conclusion, the findings expand our understanding of the phytochemical components of the Chinese pepper genotype at two maturity stages. Moreover, a UPLC-ESI-Q-TOF-MS in negative ionization mode rapid methods for characterization and isomers differentiation was described.

Exploring non-cytotoxic, antioxidant, and anti-inflammatory properties of selenium nanoparticles synthesized from Gymnema sylvestre and Cinnamon cassia extracts for herbal nanomedicine

The increasing need for pharmaceutical agents that possess attributes such as safety, cost-effectiveness, environmental sustainability, and absence of side effects has driven the advancement of nanomedicine research, which lies at the convergence of nanotechnology and medicine. Aims and objectivesThe study aimed to synthesize non-toxic selenium nanoparticles (SeNPs) using Gymnema sylvestre (G. sylvestre) and Cinnamon cassia (C. cassia) extracts. It also sought to develop and evaluate versatile nanomedicine formulations i.e. selenium nanoparticles of G. sylvestre and C. cassia (SeNPs), drug (lupeol) loaded SeNPs (DLSeNPs), drug-loaded and coated (PEG) SeNPs (DLCSeNPs) without side effects. MethodsThe SeNPs formulations were hydrothermally synthesized, loaded with lupeol to improve efficacy, coated with polyethylene glycol (PEG) for targeted delivery, and characterized using UV–Vis spectrophotometry, Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), zeta potential analysis, size distribution analysis, and X-ray diffraction (XRD). Hemolytic cytotoxicity, 2,2-Diphenyl-1-picrylhydzayl (DPPH), total Reducing power, and total antioxidant capacity (TAC) antioxidant assays, carrageenan-induced paw edema, and histological studies were used to estimate the acute anti-inflammatory activity of the synthesized SeNPs. ResultsThe final form of PEGylated and drug (lupeol)-loaded selenium nanoparticles (DLCSeNPs) exhibited an average particle size ranging from 100 to 500 nm as evidenced by SEM, and Zeta potential results. These nanoparticles demonstrated no cytotoxic effects and displayed remarkable antioxidant (IC50 values 19.29) and anti-inflammatory capabilities. These results were fed into Graph-pad Prism 5 software and analyzed by one-way ANOVA, followed by Tukey's post hoc test (p < 0.001). All nano-formulations exhibited significant overall antioxidant activity, with IC50 values ≤ 386 (p < 0.05) as analyzed by ANOVA. The study's results suggest that G. sylvestre outperformed C. cassia in terms of reducing 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) free radical, potassium ferricyanide, and ammonium molybdate in respective antioxidant assays. As far as anti-inflammatory activities are concerned drug (lupeol)-loaded and PEG-coated G. sylvestre SeNPs exhibited the highest anti-inflammatory potential from all other nano-formulations including drug (lupeol)-loaded and PEG-coated C. cassia SeNPs, as exhibited to reduce the release of pro-inflammatory signals i.e. cytokines and NF-kB, making them innovative anti-inflammatory nanomedicine. ConclusionThe study synthesized lupeol-loaded and PEG-coated SeNPs, showcasing the potential for biocompatible, cost-effective anti-inflammatory nanomedicines. G. Sylvester's superior antioxidant and anti-inflammatory performance than Cinnamon cassia emphasizes medicinal plant versatility.

Evaluation of cytotoxicity, acute toxicity, genotoxicity and antioxidant and antigenotoxicity activities of the sarcotesta fraction of punica granatum L. rich in lectin (PgTel)

ABSTRACT Punica granatum, popularly known as pomegranate, is a fruit tree with wide worldwide distribution, containing numerous phytochemicals of great medicinal value. The aim of the present study was to determine the phytochemical profile and antioxidant potential of a protein fraction (PF) derived from P. granatum sarcotesta which is rich in lectin. In addition, the acute oral toxicity, genotoxicity and antigenotoxicity of this protein fraction (PF) from P. granatum sarcotesta was measured. The phytochemical profile of PF was determined using HPLC. The in vitro antioxidant effect was assessed using the methods of total antioxidant capacity (TAC) and DPPH and ABTS+ radical scavenging. Acute oral toxicity was determined in female Swiss mice administered a single dose of 2000 mg/kg. This PF was examined for genotoxicity and antigenotoxicity at doses of 500, 1000 and 2000 mg/kg, utilizing mouse peripheral blood cells. Phytochemical characterization detected a high content of ellagic acid and antioxidant capacity similar to that of ascorbic acid (positive control). PF was not toxic (LD50 >2000 mg/kg) and did not exert a genotoxic effect in mice. PF protected the DNA of peripheral blood cells against damage induced by cyclophosphamide. In conclusion, this PF fraction exhibited significant antioxidant activity without initiating toxic or genotoxic responses in mice.

Evaluation of in vitro antioxidant activities, total phenolic and elemental contents of common herbs and spices (Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves) in South-West Nigeria

Abstract Natural remedies are being widely utilized in some disease’s conditions including cancer, diabetes, neurodegenerative diseases, atherosclerosis hypertension and other cardiovascular diseases. The therapeutic intervention of medicinal plants and spices cannot be overemphasized in the management and control of diseases but the exact modes of action of these herbs and plants have not been fully elucidated. This research work was designed to study the expression of selected elements, zinc (Zn), iron (Fe), copper (Cu), selenium (Se), lead (Pb) and cadmium (Cd); the phytochemical and the in vitro antioxidant properties of ethanolic extracts of Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves for their potential involvement in the prevention and management of cardiovascular diseases and cancer through different chemical methods. In vitro antioxidant properties were assayed by investigating 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radicals scavenging potentials and Ferric Reducing Antioxidant Potential (FRAP); total phenolic content was determined by using Folin-Ciocalteu assay and the elemental contents of these extracts was investigated through the use of Inductively Coupled Plasma Optical Emission Spectroscopy (ICP-OES). All tests were run in triplicates and analysed. Our results revealed that the ethanolic extracts of M. oleifera leaves, A. sativum (Garlic) and M. charantia (ejinrin) leaves possess significant antioxidant activities and express important and beneficial elements (in marginal and trace amounts) and phytochemicals. These results suggest that the medicinal attributes of these plants may be linked to the radical scavenging abilities, beneficial elements and phytochemicals expression of their ethanolic extracts which may likely be a good direction in the area of drug discovery and development.

Box-Behnken Design-Based Optimization of Phytochemical Extraction from Diplazium esculentum (Retz.) Sw. Associated with Its Antioxidant and Anti-Alzheimer's Properties.

A previous study reported that the ethanolic extract of the edible fern, Diplazium esculentum (Retz.) Sw. (DE), obtained from a non-optimized extraction condition exhibited anti-Alzheimer's disease (AD) properties through the inhibition of a rate-limiting enzyme in amyloid peptide formation, β-secretase-1 (BACE-1). Nevertheless, a non-optimized or suboptimal extraction may lead to several issues, such as a reduction in extraction efficiency and increased time and plant materials. In this study, extraction of the DE was optimized to obtain appropriate BACE-1 inhibition using a Box-Behnken design (BBD) and response surface methodology (RSM). Data revealed that the optimal extraction condition was 70% (v/v) aqueous ethanol, 50 min extraction time, 30 °C extraction temperature, and 1:30 g/mL solid/liquid ratio, giving BACE-1 inhibition at 56.33%. In addition, the extract also exhibited significant antioxidant activities compared to the non-optimized extraction. Metabolomic phytochemical profiles and targeted phytochemical analyses showed that kaempferol, quercetin, and their derivatives as well as rosmarinic acid were abundant in the extract. The optimized DE extract also acted synergistically with donepezil, an AD drug suppressing BACE-1 activities. Data received from Drosophila-expressing human amyloid precursor proteins (APPs) and BACE-1, representing the amyloid hypothesis, showed that the optimized DE extract penetrated the fly brains, suppressed BACE-1 activities, and improved locomotor functions. The extract quenched the expression of glutathione S transferase D1 (GSTD1), inositol-requiring enzyme (IRE-1), and molecular chaperone-binding immunoglobulin (Bip), while donepezil suppressed these genes and other genes involved in antioxidant and endoplasmic reticulum (ER) stress response, including superoxide dismutase type 1 (SOD1), activating transcription factor 6 (ATF-6), and protein kinase R-like endoplasmic reticulum kinase (PERK). To sum up, the optimized extraction condition reduced extraction time while resulting in higher phytochemicals, antioxidants, and BACE-1 inhibitors.

Construction and characterization of a novel Myr-OSADS-CIn micelle for enhanced stability and bioavailability of myricetin

In this study, a novel self-assembled micelle (Myr-OSADS-CIn) based on OSA-debranched lotus root starch (OSADS) and cationic inulin (CIn) was successfully constructed to enhance the stability and bioavailability of myricetin (Myr). Firstly, lotus root starch and inulin were chemically modified. The results of Fourier transform infrared spectroscopy, X-ray diffraction, Nuclear magnetic resonance spectroscopy and Element analysis confirmed the successful synthesis of OSADS and CIn. When the optimal weight ratio of Myr/OSADS/CIn was 1:40:120, a stable Myr-OSADS-CIn composite micelle was successfully formed. The key characteristics of the micelle included a polydispersity index (PDI) of 0.321, a zeta-potential of +24.247 mV, and an encapsulation efficiency of 87.536%. Transmission electron microscopy analysis revealed unique core-shell structures within the Myr-OSADS-CIn micelle. The formation and stability of the composite micelle were primarily influenced by hydrogen bonds and electrostatic interactions. Notably, the Myr-OSADS-CIn micelle exhibited excellent biocompatibility, significant antioxidant activity and controlled release of Myr. Furthermore, in vivo bioavailability of Myr in the Myr-OSADS-CIn micelle was found to be 4.054 times higher compared to free Myr. In conclusion, the Myr-OSADS-CIn micelle shows promise as a delivery system for the effective use of Myr in the food industry.

Phytochemical Characterization, Antioxidant, and Antimicrobial Activity of the Vegetative Buds from Romanian Spruce, Picea abies (L.) H. Karst.

This study aims to investigate the vegetative buds from Picea abies (spruce), naturally found in a central region of Romania, through a comprehensive analysis of the chemical composition to identify bioactive compounds responsible for pharmacological properties. Using HPLC/derivatization technique of GC-MS and quantitative spectrophotometric assays, the phenolic profile, and main components of an ethanolic extract from the buds were investigated. The essential oil was characterized by GC-MS. Moreover, the antioxidant activity with the DPPH method, and the antimicrobial activity were tested. Heavy metal detection was performed by graphite furnace atomic absorption spectrometry. The main components of the alcoholic extract were astragalin, quercetin, kaempferol, shikimic acid, and quinic acid. A total content of 25.32 ± 2.65 mg gallic acid equivalent per gram of dry plant (mg GAE/g DW) and of 10.54 ± 0.083 mg rutin equivalents/g of dry plant (mg RE/g DW) were found. The essential oil had D-limonene, α-cadinol, δ-cadinene, 13-epimanool, and δ-3-carene as predominant components. The spruce vegetative buds exhibited significant antioxidant activity (IC50 of 53 μg/mL) and antimicrobial effects against Staphylococcus aureus. Furthermore, concentrations of heavy metals Pb and Cd were below detection limits, suggesting that the material was free from potentially harmful contaminants. The results confirmed the potential of this indigenous species to be used as a source of compounds with pharmacological utilities.

The impact of geographical location on the nutritional quality, chemical composition, and antioxidant activity of Cucurbita maxima (rouge vif d'Etampes) seed oil

This study investigates the impact of geographical location on the nutritional quality, chemical composition, and antioxidant activity of Cucurbita maxima (rouge vif d'Etampes) seed oil. In terms of lipidic fraction, variations in fatty acid composition were observed across regions, with Guercif exhibiting the highest polyunsaturated fatty acid levels (57.78%) and El Jadida the lowest (44.21%). Guercif also had the highest β-sitosterol content (107.98 mg/100g oil), while total sterols were highest in Khemisset (314.92 mg/100g oil). Additionally, Guercif displayed the highest γ-tocopherol content (568.51 mg/kg oil). Physicochemical analysis revealed higher acid and peroxide values in El Jadida, along with lower iodine values. El Jadida also exhibited higher levels of carotenoids and chlorophyll. In terms of alcoholic fraction, guercif showed higher polyphenol content and strong free radical scavenging activity, while Khemisset demonstrated significant antioxidant activity (EC50 of 0.17 mg/mL). These findings underscore the importance of considering regional variations in future studies on pumpkin seed products.
