Evaluation of the in vitro toxicity and anti-inflammatory activity of the methanolic extract of the leaves of Pistacia lentiscus L. harvested from northwestern Algeria.

Medicinal and aromatic plants have been used for thousands of years for their therapeutic properties, to treat various ailments and maintain health. This study was carried out to test the in vitro toxicity and anti-inflammatory activity of the methanolic extract of the leaves of Pistacia lentiscus L., native to the north-western region of Algeria. Toxicity was studied in vitro by the hemolysis test and anti-hemolytic activity on human red blood cells, while anti-inflammatory activity was assessed in vitro by the protein denaturation method. The study of toxicity by hemolysis of red blood cells showed a high rate of hemolysis at a dose of 200 mg/mL, with a hemolytic percentage of around 88.64%, while a high rate of anti-hemolysis was observed at a dose of 25 mg/mL, with an inhibition percentage of around 83.17%. Evaluation of anti-inflammatory activity revealed a high percentage of inhibition of ovalbumin denaturation, reaching 63.53% at the 2000 ug/mL dose, while the lowest percentage was obtained at the 3500 ug\mL concentration with 10.16%. This extract has a toxic substance but does not exceed the toxicity threshold which achieves significant anti-inflammatory activity with depandant doses.

Formulation and characterization of Tea tree and Jojoba oils nano-emulgel for in-vivo wound healing assessment

Cutaneous wounds are the most common surgical affections among living organisms worldwide, and their healing may be interrupted by several factors. This study aimed to formulate and evaluate the antioxidant, anti-inflammatory, and antimicrobial activity of tea tree and jojoba oils nano-emulsions, additionally, investigating the cytotoxicity of the optimized formula was investigated on normal human lung fibroblast cells (WI-38) by MTT colorimetric assay, additionally its in-vivo wound healing. Nano-emulsions (NEs) were prepared using a high-energy method and characterized by Transmission electron microscopy (TEM), Zeta potential, droplet size, and poly dispersive index (PDI). Nano-emulgel (NEG) was formulated by mixing the standard NE with carbopol® 940. For in-vivo wound healing, thirty adult female albino rats were assigned into control, moist exposed burn ointment (Mebo), and NEG-treated groups. The healing was assessed by analysis of superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor-alpha (TNF-α), interleukin-1β (IL-1β), and histopathology in healed wound tissues. All formulations demonstrated antioxidant, anti-inflammatory, and antimicrobial activity against Bacillus Subtilis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Salmonella typhi, Enterococcus faecalis, and Candida albicans. The CC50 of the optimized formula was 453.82± 3.87µg/mL, with a mean droplet size of 105.4nm and a zeta potential of -39.2 ± 2.1mV. NEG enhanced wound closure compared to Mebo-treated and control groups. Also, MDA significantly decreased and SOD significantly increased in NEG and Mebo-treated groups compared to the control (p ˂ 0.05). TNF-α, and IL-1β significantly decreased in NEG and Mebo-treated compared to the control (p < 0.05). Histopathology revealed reduced inflammatory cell infiltration, rapid epithelization, and increased collagen deposition in NEG-treated wound tissues compared to the control and Mebo-treated wounds. In conclusion, the NEG containing tea tree and jojoba oils demonstrated significant antioxidant, anti-inflammatory, antimicrobial, and wound healing activities.

Synthesis and Biological Evaluation of Gastroprotective Polymer Conjugates of Aceclofenac

: Nonsteroidal anti-inflammatory drugs account for a sizeable fraction of the drugs prescribed worldwide. If consumed for a long time, they can cause ulcers and lifethreatening side effects. Since the acidic group present in NSAIDs is mainly responsible for these side effects, the scientists try to synthesize polymer drug conjugates that avoid these side effects but still retain the potency of the parent drug. Macromolecular drug conjugates of aceclofenac were prepared by employing pectin, β cyclodextrin, deacetylated chitin (chitosan) and albumin (egg and bovine serum). The prepared conjugates were characterized and tested for their anti-inflammatory, antinociceptive and antiulcerogenic activity. Further experimentation was undertaken to analyze the behavior of compounds and their stability in hydrolytic conditions. Test compound A1, a pectin conjugate of aceclofenac, exhibited significant antinociceptive and anti-inflammatory activity with a significant decrease in ulcer index (4.45±0.24) as against aceclofenac (8.61±0.40) or vehicle (12.39±0.44) treated group. A novel and safer polymer drug conjugate of aceclofenac has been synthesized and evaluated.

Phenolic Bisabolane Sesquiterpene Derivatives from an Arctic Marine-derived Fungus Aspergillus sydowii MNP-2

Background:: Filamentous fungi in the genus Aspergillus are well known for their important roles in production of bioactive secondary metabolites with diversely chemical structures and potential application in pharmaceutical industry. Objective:: The present study aimed to investigate the phenolic bisabolane sesquiterpene (PBS) derivatives from an Arctic marine-derived fungus Aspergillus sydowii MNP-2. Methods:: In this study, antimicrobial activities were carried out according to the broth microdilution assay, nitric oxide (NO) production in mouse macrophages (RAW264.7) and BV2 microglial cells was used to detect the inhibitory effect of compounds in inflammatory reactions, and in vitro inhibitory cell proliferation activity was determined by the cell counting kit-8 (CCK-8) assay. Results:: In this work, chemical investigation of an Arctic marine-derived strain A. sydowii MNP-2 led to the isolation of 11 PBSs (1-11) using various chromatographic methods. Their chemical structures were unambiguously determined by 1H NMR spectroscopy and mass spectrometry analyses as well as comparison with literature data. It is noteworthy that compounds 1, 7 and 11 were firstly obtained from A. sydowii. Antimicrobial assay showed that these chemicals had no potent inhibitory effect on Staphylococcus aureus, Escherichia coli, and Candida albicans with MIC values &gt; 16 μg/mL. Additionally, the inhibition of nitric oxide (NO) production in lipopolysaccharide (LPS)- induced inflammation in mouse macrophages (RAW264.7) and BV2 microglial cells were all below 10% for compounds 4-6 and 8, indicating almost negligible anti-inflammatory efficacy. Among the tested compounds 4-6 and 8 for tumor-cell proliferation inhibition activities, compound 5 demonstrated the strongest inhibitory effect against human acute promyelocytic leukemia cells (HL-6) with a 44.76% inhibition rate. Conclusion:: In the present study, 11 PBS derivatives were purified and characterized from the solidand liquid-state fermentations of the Arctic marine-derived fungus A. sydowii MNP-2. Unfortunately, none of these metabolites had significant antimicrobial, anti-inflammatory, or tumor-cell proliferation inhibition activities.

Almond polysaccharides inhibit DSS-induced inflammatory response in ulcerative colitis mice through NF-κB pathway

Ulcerative colitis (UC), a type of inflammatory bowel disease (IBD), is a chronic recurrent inflammatory disease of the colon. Our previous findings demonstrated that almond polysaccharide (AP-1) exhibits significant anti-inflammatory activity in vitro. Therefore, this study aimed to explore the ameliorative effect of AP-1 on dextrose sodium sulfate (DSS)-induced UC mice and to elucidate its possible mechanism of action. By observing changes in body weight, fecal viscosity, stool blood, disease activity index, and colon length, we found that AP-1 attenuated inflammation. It inhibited TNF-α, IL-1β, and IL-6 while boosting anti-inflammatory IL-10 levels. Histomorphologically, AP-1 protected against DSS-induced colonic tissue damage by reducing inflammatory cell infiltration and mucosal injury. It also lowered myeloperoxidase (MPO) and NO while increasing total superoxide dismutase (T-SOD) and glutathione peroxidase (GSH-Px) in colonic tissues. Moreover, using the Western blot technique, AP-1 was shown to inhibit the phosphorylation of p65 and IκB-α proteins in the NF-κB/iNOS/COX2 signaling pathway and down-regulate the expression of inflammation-associated proteins COX2 and iNOS, thus slowing down and ameliorating inflammatory processes. Therefore, the safe and effective beneficial effects of AP-1 make it a promising therapeutic strategy for relieving IBD, especially UC.

Investigation of Antioxidant Potential of Black Beans Due to Phytochemical and Globulin Content for Nutraceutical Application

The study aims to elucidate the antioxidant and anti-inflammatory properties of black beans and identify the key contributors within the beans. Black beans boast high protein and phytochemical content, suggesting potential antioxidant activity. This work delves into this possibility, focusing on the role of globulins (a protein fraction) and various phytochemicals in black bean’s bioactivity. Globulins were isolated from black beans by salt precipitation. Ethanolic and aqueous extracts were prepared, followed by phytochemical screening for phenolics, flavonoids, alkaloids, etc. Quantification of phenolics and flavonoids was performed by colorimetric assays. SDS-PAGE and mass spectrometry identified the isolated globulin. Ferric reducing potential and total antioxidant capacity tests assessed antioxidant capacity, while erythrocyte membrane stabilisation inhibition evaluated anti-inflammatory potential. Black beans exhibited a diverse range of phytochemicals. The ethanolic extract displayed the highest flavonoid content (11.18±0.11mg QE/gram extract), while the aqueous extract was richest in phenolics (0.71±0.14 mg GAE/gram extract). The aqueous extract displayed the strongest antioxidant activity (2.87 ± 0.98 mg AAE/gram extract), followed by the globulin fraction (1.23± 0.43 mg AAE/gram extract). The ethanolic extract demonstrated superior anti-inflammatory activity. Mass spectrometry identified globulin as a 68.7 kDa legumin protein from Phaseolus vulgaris. Black beans display significant antioxidant and anti-inflammatory activity due to globulins and diverse phytochemicals. By informing dietary practices and facilitating the development of nutraceutical interventions, this work has the potential to contribute to a healthier population.

Comparison of the difference in the anti-inflammatory activity of two different color types of Farfarae Flos based on in vitro, in vivo experiments and untargeted metabolomics.

Due to its remarkable anti-inflammatory pharmacological activity, Farfarae Flos has gained extensive usage in the treatment of various inflammatory diseases such as bronchitis, pneumonia, prostatitis and colitis. And Farfarae Flos come in two color types depending on the color of the flowers: yellowish-white (YW), and purplish-red (PR). However, the difference in anti-inflammatory activity and metabolic profiles between the two flower colors remains unexplored. This study aims to explore the difference in the anti-inflammatory potential between YW and PR variants of Farfarae Flos and unravel the mechanisms responsible for the observed differences in anti-inflammatory activity through an integrated approach encompassing untargeted metabolomics and in vivo/vitro experimental studies. Initially, we verified the contrasting effects of YW and PR on the inhibition of the inflammatory factors interleukin-6 (IL-6) and nitric oxide (NO) by utilizing an in vitro RAW 264.7 cell inflammation model. Subsequently, a comprehensive evaluation of the systemic inhibitory capacity of YW and PR on IL-6, Interleukin-10 (IL-10), and tumor necrosis factor-α (TNF-α) was conducted using a validated whole-body mouse model, followed by the analysis of inflammatory factors and histological examination of collected serum, liver, and spleen after 7days. Furthermore, non-targeted metabolomics profiling was employed to analyze the metabolite profiles of Farfarae Flos with different colors, and quantitative analysis was conducted to identify differential metabolites between YW and PR. The correlation between the anti-inflammatory activities of differentially accumulated metabolites (DAMs) and Farfarae Flos was investigated, resulting in the identification of 48 compounds exhibiting significant anti-inflammatory activity. Additionally, KEGG pathway enrichment analysis was performed to elucidate the underlying mechanisms. Our findings demonstrate that both YW and PR possess anti-inflammatory abilities, with PR exhibiting significantly superior efficacy. The integration of in vivo/vitro experiments and non-targeted metabolomics confirmed the exceptional anti-inflammatory potential of PR and solidified its classification as the "purplish-red better" of Farfarae Flos. This study provides valuable insights into the breeding and medical transformation of Farfarae Flos varieties, along with a scientific basis for the establishment of quality standards and the development of new drugs utilizing Farfarae Flos.

Novel efficacy of pregnane and flavonoid glycosides from Desmidorchis flava in in vivo nociceptive and inflammatory paradigms and their target prediction by cheminformatics approach

Inflammation is associated with multiple life-threatening conditions. Desmidorchis flava is an edible plant and traditionally used for managing various diseases. Three novel molecules, namely desmiflavaside-C (1), nizwaside (2), and desmiflanoside (3) were isolated from Desmidorchis flava, and their structures were confirmed by mass spectrometry and through reported literature. These compounds were in vivo examined for antinociceptive (tonic visceral nociception) and anti-inflammatory (carrageenan induced paw edema) activities. Significant antinociceptive potential was demonstrated by compound 1 at 0.5 and 1 mg/kg doses followed by compounds 2 and 3. At similar doses, significant anti-inflammatory activity was noted for all the tested compounds. Their antinociceptive and anti-inflammatory activities were comparable to the reference standards. In silico predicted binding modes suggests that these compounds may target allosteric sites of COX-1 and COX-2 enzymes to elicit their anti-inflammatory activities. These isolated natural products may have therapeutic potential in conditions afflicted with pain and inflammation.

Experimental and Theoretical Studies for Noncovalent Interactions Analysis of 2-Phenyl Imidazole Derivative: Perspective for Anti-Inflammatory Activity

Synthesis of 2-Phenyl imidazole derivative has been carried out for the studies of quantitative non-covalent interactions and anti-inflammatory activity. Hirshfeld surface analysis, crystal structure, and non-covalent interaction computation have all been used to do quantitative investigations of non-covalent interactions. The intermolecular interactions calculations have been done with crystalexplorer 17. Further, anti-inflammatory activities of synthesized molecules were evaluated by molecular docking studies through autodock vina and in-vivo studies through carrageenan rat model. The molecular docking and in-vivo studies revealed that all molecules have shown significant anti-inflammatory activity, compared to standard drugs nimesulide.

Exploring the Potential of Novel 4-Thiazolidinone Derivatives as Dual Anti-inflammatory and Antioxidant Agents: Synthesis, Pharmacological Activity and Docking Analysis

A series of 4-thiazolidinone was synthesized and characterized by means of TLC, melting point, and spectral data like IR, 1H NMR, and Mass spectra. The anti-inflammatory activity of the synthesized compounds was determined via in vivo studies. The antioxidant properties of the synthesized compounds were determined by carrageenan-induced rat paw edema model. The synthesized compounds (A1-A14) showed significant anti-inflammatory and antioxidant activities. The most promising results for both antioxidant and antiinflammatory activity were exhibited by compound A8 which may emerge as a potent anti-inflammatory agent with potential free radical scavenging activity. Molecular docking studies were carried out to determine the interaction of compounds into the active site of COX-2 inhibitor (PDB ID: 3LN1), which suggested compound A8 to have the best docking score by showing interactions with ASP483 and LYS478.

Co-encapsulation of probiotic bacteria L. rhamnosus GG and β-carotene by a novel biphasic encapsulation technique: Stability and in vivo anti-inflammatory properties

In this study, β-carotene (βC) was encapsulated in liposomes and spray-dried with the probiotic bacteria Lactobacillus rhamnosus GG (LGG), to obtain a biphasic structure with two functional components. Initially, the liposomes loaded with βC resulted in large multivesicular vesicles (LMVV) with spherical morphology, mean size of 1191 nm and entrapment efficiency of 81.33%. Then, the spray-drying of the mixture βC-LMVV with the LGG resulted in biphasic dried microparticles (BDM) with a spherical shape, retention of 64,48% of βC, and LGG survivor above 90%. The BDM showed high storage stability for 90 days at room temperature and, at the dose of 2000 mg/kg of BDM did not cause any acute toxicity in Wistar rats. In addition, at the same dose, presented significant anti-inflammatory activity in carrageenan-induced paw edema and pleurisy. Thus, the produced BDM could be an innovative ingredient with functional properties and also an efficient encapsulation strategy for βC.

Plant Phenolics in the Prevention and Therapy of Acne: A Comprehensive Review.

Plants are a rich source of secondary metabolites, among which phenolics are the most abundant. To date, over 8000 various polyphenolic compounds have been identified in plant species, among which phenolic acids, flavonoids, coumarins, stilbenes and lignans are the most important ones. Acne is one of the most commonly treated dermatological diseases, among which acne vulgaris and rosacea are the most frequently diagnosed. In the scientific literature, there is a lack of a detailed scientific presentation and discussion on the importance of plant phenolics in the treatment of the most common specific skin diseases, e.g., acne. Therefore, the aim of this review is to gather, present and discuss the current state of knowledge on the activity of various plant phenolics towards the prevention and treatment of acne, including in vitro, in vivo and human studies. It was revealed that because of their significant antibacterial, anti-inflammatory and antioxidant activities, phenolic compounds may be used in the treatment of various types of acne, individually as well as in combination with commonly used drugs like clindamycin and benzoyl peroxide. Among the various phenolics that have been tested, EGCG, quercetin and nobiletin seem to be the most promising ones; however, more studies, especially clinical trials, are needed to fully evaluate their efficacy in treating acne.

Bioevaluation, pharmacokinetics and molecular docking study of terpenoid-rich rhizome essential oil of Zingiber officinale

The study focuses on the phytochemical composition, in vitro and in silico antioxidant and NO reduction potential of Zingiber Officinale rhizome essential oil (ZOREO). The GC–MS analysis has revealed the presence of 47 volatile constituents representing 94.35 % of the total area of EO. The major compounds identified were α-zingiberene (20.58 %), geranial (10 %), β-sesquiphellandrene (7.39 %), and neral (7.23 %). The EO was rich in sesquiterpene hydrocarbons (45.93 %) followed by monoterpene hydrocarbons (29.29 %). The EO showed moderate antioxidant activity compared to the standard (IC50 = 5.16 ± 0.4 mg/mL for DPPH assay and EC50 = 0.52 ± 0.6 mg/mL for FRAP assay). The cytotoxic analysis showed significant viability towards RAW 264.7 cell lines. Treatment with ginger EO at a concentration of 100 μg/mL showed significant anti-inflammatory activity by the significant reduction of LPS-induced nitric oxide (NO) production to 31.35 %, in comparison with the LPS-treated group. Further, the EO components were screened in order to provide mechanistic insights into Xanthine oxidase (XO) and iNOS inhibition. The docking results showed that the volatile constituents of ginger EO have substantial binding affinity with the active site residues of the receptor. The simulation conducted for the top compounds- geranyl acetate and β-eudesmol having the highest binding affinity towards both the proteins XO and iNOS respectively. Both the results showed the compounds steadily interacted with both of the proteins active site residues throughout the simulation. The current analysis showed the phytoconstituents showed better antioxidant activity than the EO. Hence these compounds could be further considered for in vitro and in vivo evaluation for the development of efficient phytopharmaceuticals.

Pleiotropically activation of azaphilone biosynthesis by overexpressing a pathway-specific transcription factor in marine-derived Aspergillus terreus RA2905

The genome sequencing of Aspergillus terreus reveals that the vast number of predicted biosynthetic gene clusters have not reflected by the metabolic profile observed under conventional culture conditions. In this study, a silent azaphilone biosynthetic gene cluster was activated by overexpressing a pathway-specific transcription factor gene2642 in marine-derived fungus A. terreus RA2905. Consequently, twenty azaphilone compounds were identified from the OE2642 mutant, including 11 new azaphilones and their precursors, azasperones C−J (1 − 5, 7 − 9) and preazasperones A−C (15 − 17). The structures of those new compounds were unambiguously determined on the basis of NMR and HRESIMS spectra analysis, and the absolute configurations were established depending on ECD calculations. Compounds 1 and 2 were the rarely reported naturally occurring azaphilones with 2-N coupled phenyl-derivative. The bioactivity assay revealed that compounds 18 − 20 exhibited significant anti-inflammatory activity. Based on the occurrence of diverse intermediates and the putative gene functions, a plausible biosynthetic pathway of these compounds was proposed. The above results demonstrated that overexpression of the pathway-specific transcription factor presents a promising approach for enriching fungal secondary metabolites and accelerating the targeted discovery of novel biosynthetic products.

Curcumol derivatives exhibit ameliorating effects on lipopolysaccharide-induced acute lung injury: Synthesis, biological evaluation, structure–activity relationship and action mechanism

Acute lung injury (ALI) is an intricate clinical disease marked by high mortality and a sudden start. Currently, although there are no specific therapeutics for ALI, the administration of anti-inflammatory drugs is a promising treatment strategy. Curcumol, a terpenoid natural product, has demonstrated significant anti-inflammatory activity. Herein, we designed and synthesised 42 curcumol derivatives using curcumol as the core scaffold. These derivatives underwent in vitro screening for anti-inflammatory activity, and their structure–activity relationship was assessed. Among them, derivative 2 exhibited potent anti-inflammatory potential, inhibiting the expression of inflammatory markers at the nanomolar level. In addition, its water solubility was considerably improved, thereby laying the foundation for enhanced druggability. Derivative 2 also ameliorated lipopolysaccharide (LPS)-induced ALI and reduced pulmonary inflammation at a dose of 5 mg/kg. Proteomics analysis revealed that the anti-inflammatory effect of this compound primarily involved the mTOR signalling pathway. Furthermore, molecular docking and cellular thermal shift assays indicated that GSK3β is a critical target of action of derivative 2, as verified via western blotting. These findings suggest that derivative 2 can be a lead therapeutic compound for ALI, with GSK3β emerging as a promising novel target for the development of specific anti-ALI drugs.

Sulfated polysaccharide from Apostichopus japonicus viscera exhibits anti-inflammatory properties in vitro and in vivo

Polysaccharides from sea cucumbers are known for their biological activities, but little is known about those from sea cucumber viscera. The present study isolated a sulfated polysaccharide (SCVP-2) from the viscera of Apostichopus japonicas, which had a molecular weight of 209.1 kDa. SCVP-2 comprised 66.3 % total sugars, 2.1 % uronic acid, 4.5 % proteins, and 25.5 % sulfate groups, containing glucosamine, galactosamine, glucose, galactose, and fucose. FT-IR and NMR analyses identified SCVP-2 as a fucoidan sulfate with sulfation patterns of the fucose branches as Fuc2S, Fuc4S, and Fuc0S. SEM and AFM analyses showed irregular clusters and linear conformations. SCVP-2 demonstrated strong anti-inflammatory properties both in vitro and in vivo. In lipopolysaccharide (LPS)-induced inflammation in macrophage RAW264.7 cells, SCVP-2 significantly reduced nitric oxide (NO) and cytokine secretion (IL-1β, IL-6, TNF-α). Additionally, it downregulated the expression of these cytokine genes. Furthermore, the anti-inflammatory mechanism of SCVP-2 was related to the inhibition of the MAPKs and NF-κB pathways. SCVP-2's anti-inflammatory capacity was confirmed in acute inflammation models, including xylene-induced ear swelling and acetic acid-induced peritoneal capillary permeability, and in high-fat diet-induced systemic low-grade chronic inflammation. In conclusion, SCVP-2 exhibits significant anti-inflammatory activity, suggesting its potential for development as a functional food ingredient or therapeutic agent for inflammation-related diseases.

METABOLIC SYNDROME: IN VITROANTIOXIDANT, ANTICANCER, ANTI-INFLAMMATORY AND ANTIMICROBIAL ACTIVITIES OF DISSOTIS ROTUNDIFOLIA(SM.) WITH PHYTOCHEMICAL AND NUTRITIONAL COMPONENTS

Background:Dissotis rotundifolia(Sm.) Triana isa prostrate herb of the family Melastomataceae. It is used for the treatment of obesity, hypertension, circulatory problems and metabolic syndrome (MS). This study evaluated the antioxidant, anticancer, anti-inflammatory.and antimicrobial activities of the whole plant of D. rotundifoliato validateits therapeutic potential in the management and treatment of MS. Materials and Methods: The plant was screened for proximate, mineral, vitamin and phytochemical components, as well as antioxidant activities using standard laboratory techniques. The anticancer activities of the ethanol plant extract were against kidney (VERO) and breast (MCF-7) cancer cells. The anti-inflammatory activity of the extract was determined using nitric oxide production inhibition assay and the antimicrobial activity was evaluated against five pathogenic organisms. All data were subjected to statistical analysis. Results: The plant was rich in carbohydrate (47.87%), protein (22.5%), crude fibre (9.57%), Na (206.67 mg/100 g), Ca (183.33mg/100 g), vitamin C (25.53 mg/100 g), flavonoids (685.00 mg/100 g), alkaloids (616.67 mg/100 g) and saponins (471.67 mg/100 g) contents. Theextract exhibited considerableand variedantioxidant activities.The plant extract was very active against VERO and MCF-7 cellsatlow concentrations(0.145 mg/ml and 0.141 mg/ml respectively) compared to doxorubicin. It also exhibited significant anti-inflammatory action against lipopolysaccharide (LPS)-stimulated RAW macrophage. The extract was active against all test organisms. Overall, D. rotundifolia displayed significant antioxidant, anticancer, anti-inflammatory and antimicrobial activities. Conclusion: The observed bioactivities of the plant could be attributed to its phytocontents and are indications of itspotential as an agent for the managementand treatment ofmetabolic syndrome.

Chemical composition and bioactivity of Codium dwarkense collected from the earthquake crack region of Ras Muhammad National Park, Red Sea, Egypt

Ras Muhammad National Park, located at the southern tip of the Sinai Peninsula, Egypt, is well-known for its unique marine biodiversity. One interesting region of the park is the Earthquake Crack, a tidal-influenced inland cave formed by a 1968 earthquake, that known for its rich seaweed community, with dominance of Codium dwarkense. This study evaluates the bioactive compounds and bioactivities of C. dwarkense from the crack region. The sample were collected, processed, and assessed for hydrographic parameters, nutritional composition, minerals, bioactive compounds, and bioactivity. Results showed high levels of carbohydrates and significant mineral content, with potassium and calcium being predominant. C. dwarkense also showed considerable amounts of vitamin C (32.4 mg/100g), vitamin E (400 mg/100g), phenolics (250.08 mg GAE/g), and flavonoids (85.4 mg rutin/g). The HPLC profile was dominated with quercetin, apigenin, pyrogallol, zeaxanthin, and benzoic acid. The GC-MS analyses identified several fatty acids, which were dominated with Hexadecanoic acid and Oleic acid. Bioactivity assays demonstrated significant antimicrobial, antioxidant, antidiabetic, anti-inflammatory, and anticancer activities. This study highlights the influence of the unique features of the Earthquake Crack region on C. dwarkense ability to produce valuable bioactive compounds with diverse health benefits. These findings contribute to the recognition of C. dwarkense as a functional food and nutraceutical resource.

Anti-Inflammatory, Antinociceptive, and LC-MS Metabolic Profile from Pseudotrimezia juncifolia (Klatt) Lovo & A. Gil.

Pseudotrimezia juncifolia (Klatt) Lovo & A. Gil (Iridaceae) is a popularly known species with primarily ornamental economic interest. It has traditional uses as purgative, in conditions related to the menstrual cycle, for blood purification, as wound healing, and as anti-inflammatory. The anti-inflammatory and antinociceptive activities of the decoction from its aerial stems, corms, and stamens are described here with dereplication studies on LC-MS/MS supported by the GNPS platform, where phenolic compounds were annotated and correlated with its biological activity. The decoction was evaluated in chemical (formalin and capsaicin) and thermal (hot plate) induced nociception or carrageenan-induced inflammation in mice. Decoction (at 10, 30, or 100 mg/kg doses) significantly reduced formalin- or capsaicin-induced nociception. All doses also demonstrated an antinociceptive effect in the hot plate model increasing the time the animal spent in responding to thermal signal. Naloxone partially reversed the antinociceptive effect. An anti-inflammatory effect was observed since a reduction in cell migration, protein extravasation interleukin-1, and tumor necrosis factor production induced by carrageenan in the subcutaneous air pouch was quantified. Metabolomic analyses showed a predominance of phenolic substances, mainly flavonoids and chlorogenic acids. The literature showed that these two groups have significant anti-inflammatory and analgesic activity, and chemical data corroborate the pharmacological results observed.

Metabolic profiling of triptophenolide in rat plasma, urine, bile and faeces after intragastric administration

Triptophenolide, a major diterpenoid extracted from Tripterygium wilfordii Hook. f., has been reported to possess significant anti-tumour, anti-androgen and anti-inflammatory activities. However, the metabolic fate of triptophenolide remains unknown. Therefore, this study focused on the metabolic profiling of triptophenolide in rat plasma, urine, bile and faeces following intragastric administration. An ultraperformance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry with combination of extracted ion chromatogram strategy based on 71 typical metabolic reactions was established to comprehensively profile the metabolites of triptophenolide. This strategy allowed for the identification of 17 metabolites from the biosamples. Reduction, oxidation, glucuronide conjugation, and hydroxylation were considered as its main metabolic pathways in vivo. The present study will be greatly helpful for the further pharmacological studies on triptophenolide and would provide valuable information for its clinical application.
