Prenylflavonoids from the Twigs of Artocarpus nigrifolius.

Two new prenylated flavones, artocarnin A (2) and carpachromenol (12), together with 13 known prenylflavonoids (1, 3-11, 13-15) were isolated from the twigs of Artocarpus nigrifolius for the first time. Their structures were elucidated by high resolution-electrospray ionization (HR-ESI)-MS, NMR spectroscopic analysis, and in comparison with the reported data. Compounds 1-15 were evaluated for their antiproliferative effects against SiHa and SGC-7901 human cancer cell lines in vitro. The most active compound, eleocharin A (10), showed significant cytotoxicity on SiHa cells (IC50=0.7±0.1 µM) and inhibitory activity against SGC-7901 cells (IC50=8.3±0.2 µM) and could be considered as potential lead compound for further development of novel anti-tumor agents.

Cytotoxic lanostanoid triterpenes from Ganoderma lucidum

Two new lanostanoid triterpenes, 23S-hydroxy-3,7,11,15-tetraoxo-lanost-8,24E-diene-26-oic acid (1), and 12β-acetoxy-3β-hydroxy-7,11,15,23-tetraoxo-lanost-8,20E-diene-26-oic acid (16), together with 17 known compounds, were isolated from the fruit bodies of Ganoderma lucidum. Their structures were established by spectroscopic methods, especially 2D-NMR and MS analyses and by comparison with literature data. The cytotoxic assay of the above compounds against p388, Hela, BEL-7402, and SGC-7901 human cancer cell lines showed their cytotoxicity with the IC50 values in the range of 8–25 μm.

Synthesis and anticancer activity of lipophilic platinum(II) complexes of 3,5-diisopropylsalicylate

Novel lipophilic platinum(II) complexes (LSPt-1–3), containing 3,5-diisopropylsalicylate (DIPS) as a leaving group and 2NH 3 or 1 R,2 R-diaminocyclohexane or (4 R,5 R)-4,5-bis(aminomethyl)-2-isopropyl-1,3-dioxolane as the carrier, have been synthesized, characterized and evaluated in vitro and in vivo. The octanol/water distribution coefficient of the complexes has also been measured. The results showed that the complexes achieved a typical square planar and the octanol/water distribution coefficient log P was 4.27, 4.37 and 4.31. The complexes were tested by SRB method to be more cytotoxic than Carboplatin, Oxaliplatin and Eptaplatin against 3AO, A549, NCI-H460 and SGC-7901 human cancer cell lines. Among complexes, LSPt-2 was much more effective than Carboplatin and Oxaliplatin in treating the NCI-H460 non-small-cell lung tumor-bearing mice. Its optimal activity was 38.8% ( T/ C) at a dose of 30 mg/kg following i.p. administration. LD 50 for the complex was found to be 230.9 mg/kg. LSPt-2 exhibited great anticancer activity, good lipophilic ability and low toxicity and therefore, it is a promising candidate for effective and stable pharmaceutical liposomal platinum anticancer drug.
