A new strain of Saccharomyces cerevisiae in diets of lactating Holstein cows improved feed efficiency and lactation performance

Abstract This study compared the effects of feeding a new strain of Saccharomyces cerevisiae HSA2020 with a commercial strain on in vitro rumen fermentation and production performance of dairy cows. Permeate was used as a substrate for the laboratory production of the new strain of S. cerevisiae after the hydrolysis by β–galactosidase (5000 µ/mL at 37°C). Two experiments were conducted: in Experiment 1, the effects of three levels (1, 2 and 3 g/kg dry matter) of S. cerevisiae on in vitro ruminal fermentation kinetics were evaluated. In Experiment 2, for 60 days, sixty multiparous Holstein cows (639±24.8 kg BW, 3±1 parity, 7±1 days in milk, with a previous milk production of 23±2.0 kg/d) during the previous lactation, were randomly assigned to 3 treatments in a completely randomized design. Cows were fed without any additives (control treatment) or supplemented with 2 g/kg feed daily of laboratory produced (PY)or commercial (CY) S. cerevisiae. In Experiment 1, inclusion of PY and CY increased (P<0.05) gas production, propionate, and nutrient disappearance, while decreased (P<0.05) methane production and protozoal count. Moreover, in Experiment 2, PY followed by CY increased (P<0.01) nutrient digestibility, and serum concentrations of total protein, albumin, and glucose (P<0.05). Higher daily milk yield, and milk energy output were observed with PY and CY without affecting concentrations of milk components or milk fatty acid profile. Compared to control, increased feed efficiency was observed with PY and CY. Compared to PY, CY increased serum concentrations of urea-N and decreased triglycerides, while PY decreased serum aspartate transaminase and increased concentration of conjugated linoleic acids in milk. In early lactating cow diets, both strains of S. cerevisiae improved production performance at 2 g/kg improved production performance, and minimal differences between strains were found.

PSIV-A-11 Potential of black soldier fly (Hermetia illucens) larvae frass as a functional ingredient for nursery pigs

Abstract Frass is the leftover material from the production of black soldier fly larvae (BSFL), which includes chitin-containing exoskeleton shedding, small, processed larvae, leftover dietary substrate, and larvae excrement. A total of 120 nursery pigs (initial body weight of 6.85 ± 0.10 kg) weaned at 21 d of age were used in a 6-wk study to evaluate the effect of increasing dietary levels of frass on growth performance, diarrhea score, and serological indices. Pigs were randomly assigned within sex and body weight blocks to 1 of 3 dietary treatments using 30 pens (4 pigs/pen; 10 replicates/treatment). Diets were prepared from a common basal and fed in 3 dietary phases of 2 wk each and included: 1) a control diet based primarily on corn and soybean meal; 2) the control with 3% added frass; and 3) the control with 6% added frass. On d 4, 7, and 14, individual fecal samples were obtained using fecal loops and were evaluated on a scale of 1 to 5 representing hard, normal, soft, watery soft, and watery liquid feces, respectively. Blood samples were collected on d 42 from 1 median pig per pen for the analysis of serum chemistry. Data were analyzed using the mixed model procedures as a randomized complete block design using linear and quadratic contrast comparisons to determine the effects of frass. Supplementation of frass linearly increased average daily gain (ADG) from d 7 to 14 (P = 0.044; 347.9, 378.8, and 396.8 g/d for 0, 3, and 6% frass, respectively), and tended to linearly increase ADG during Phase 1 (P = 0.079; 265.1, 284.8, and 298.6 g/d). No differences were observed for final body weight (P ≥ 0.146; 29.90, 31.48, and 30.70 kg for 0, 3, and 6% frass, respectively), and overall ADG, average daily feed intake (ADFI), or gain to feed ratio (G:F). Diarrhea scores increased (P < 0.001) from d 4 (2.96) to d 7 (4.20) and then decreased (P < 0.001) on d 14 (3.69). Inclusion of frass at 6% had a decreased diarrhea score compared with 3% frass on d 7 (P = 0.029), but pigs fed 6% frass had a greater diarrhea score (P = 0.038) than pigs fed the control diet on d 14. However, scores were indicative of diarrhea for all treatments on d 7 and 14. Serum concentrations of total protein, albumin, globulin, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, γ-glutamyltranspeptidase, urea N, creatinine, glucose, Ca, P, Mg, K, Na, Cl, triglycerides, amylase, cholesterol, and creatine phosphokinase measured on d 42 were not different (P > 0.093) between treatments. All serological analytical results were generally within normal expected ranges. In conclusion, these findings suggest that dietary inclusion of frass may improve growth performance of nursery pigs, especially during the early stages of the nursery period.

Metabolic response of endangered goats fed spineless cactus associated with Tifton-85 hay or Maniçoba hay

The aim of this study was to evaluate the effects of replacement of Tifton-85 hay by maniçoba hay, in spineless cactus-based diets, on markers of energy, protein, mineral metabolism and serum enzyme activity of Moxotó goats. Sixteen uncastrated male Moxotó breed goats, with 20 months old and initial body weight of 21.7 ± 3.92 kg were used in a randomized blocks design, using the initial weight as the criterion for the formation of blocks, with two treatments and eight replicates per treatment. The experimental treatments consisted of two diets: diets containing Miúda spineless cactus associated with Tifton-85 hay (1) or maniçoba hay (2), with roughage:concentrate ratio of 70:30. Blood samples were collected fortnightly (baseline, 15 days, 30 days and 45 days) during the feedlot period. There was a higher serum concentration of total protein and fructosamine (P < 0.05) in goats that received maniçoba hay. However, there was higher serum gamma-glutamyl transferase (GGT) enzyme activity in goats that ingested Tifton-85 hay (P < 0.05). Concerning to the profile of blood metabolites in the different fortnightly collections, it was possible to verify a significant effect for urea, fructosamine, GGT enzyme activity, K and Cl levels (P < 0.05). In conclusion, Tifton-85 hay and maniçoba hay combined with spineless cactus does not cause metabolic disorders in feedlot native goats.

Differential responses of rumen and fecal fermentation and microbiota of Liaoning cashmere goats after 2-hydroxy-4-(methylthio) butanoic acid isopropyl ester supplementation

The 2-hydroxy-4-(methylthio) butanoic acid isopropyl ester (HMBi), a rumen protective methionine, has been extensively studied in dairy cows and beef cattle and has been shown to regulate gastrointestinal microbiota and improve production performance. However, knowledge of the application of HMBi on cashmere goats and the simultaneous study of rumen and hindgut microbiota is still limited. In this study, HMBi supplementation increased the concentration of total serum protein, the production of microbial protein in the rumen and feces, as well as butyrate production in the feces. The results of PCoA and PERMANOVA showed no significant difference between the rumen microbiota, but there was a dramatic difference between the fecal microbiota of the two groups of Cashmere goats after the HMBi supplementation. Specifically, in the rumen, HMBi significantly increased the relative abundance of some fiber-degrading bacteria (such as Fibrobacter) compared with the CON group. In the feces, as well as a similar effect as in the rumen (increasing the relative abundance of some fiber-degrading bacteria, such as Lachnospiraceae FCS020 group and ASV32), HMBi diets also increased the proliferation of butyrate-producing bacteria (including Oscillospiraceae UCG-005 and Christensenellaceae R-7 group). Overall, these results demonstrated that HMBi could regulate the rumen and fecal microbial composition of Liaoning cashmere goats and benefit the host.

Evaluation of serum concentrations of total protein and gamma-globulin as an indicator of serum immunoglobulin G concentration in dairy calves

The aim of this study was to evaluate precision of estimating serum immunoglobulin G (IgG) concentration from total protein (TP) or gamma-globulin (γGLB) concentration as an alternative approach, and to compare morbidity of pre-weaned dairy calves differing in serum γGLB concentration. In trial 1, blood was sampled from 129 Holstein calves in the first week after birth, and serum concentrations of TP, γGLB, and IgG were measured. Spearman's correlation coefficient (rs) between serum IgG and TP concentrations was 0.89, and rs between serum IgG and γGLB concentrations was 0.96. Absolute residual (observed – predicted) serum IgG concentrations were smaller when estimated by serum γGLB concentration than by serum TP concentration, and differences in the absolute residuals were smaller for calves fed colostrum replacer (1.68 vs. 4.29 g/L) than those fed whole colostrum (2.41 vs. 3.48 g/L). In trial 2, blood was sampled from 740 Holstein heifer calves during the first week of age, and serum γGLB concentration was measured. The calves were divided into 4 categories based on their serum γGLB concentration; ≥ 1.0 g/dL (Excellent), 0.7 ≤ γGLB <1.0 g/dL (Good), 0.4 ≤ γGLB <0.7 g/dL (Fair) and <0.4 g/dL (Poor). Morbidity for diarrhea and respiratory disease in pre-weaned dairy calves was determined for the first 28 and 56 d of age, and compared among the 4 categories based on serum γGLB concentration. Calves with serum γGLB concentration higher than 0.7 g/dL (Good and Excellent) had less diarrhea during the first 28 d of age than those with lower serum γGLB concentration (Fair and Poor). Calves with serum γGLB concentration higher than 1.0 g/dL (Excellent) had less respiratory diseases for the first 56 d of age than those with lower serum γGLB concentration (Good, Fair, and Poor). These results suggest that serum IgG concentration can be estimated more precisely from concentration of γGLB than TP particularly for calves fed colostrum replacer, and that γGLB concentrations in the first week of age are associated with morbidity of calves. Transfer of passive immunity in dairy calves can be assessed effectively by serum γGLB concentration.

Dietary supplementation with ellagic acid improves the growth performance, meat quality, and metabolomics profile of yellow-feathered broiler chickens

The aim of this research was to explore the effects of ellagic acid (EA) on growth performance, meat quality, and metabolomics profile of broiler chickens. 240 healthy yellow-feathered broilers were randomly divided into 4 groups (6 replicates/group and 10 broilers /replicate): 1) a standard diet (CON); 2) CON+0.01% EA; 3) CON+0.02% EA; 4) CON+0.04% EA. Compared with the CON group, dietary 0.02% EA increased linearly and quadratically the ADG and lowered F/G ratio from 29 to 56 d and from 1 to 56 d of age (P < 0.05). The EA groups had higher spleen index and showed linear and quadratic improve thymus index (P < 0.05). A total of 0.02% EA linearly and quadratically increased the leg muscle percentage and quadratically increased the breast muscle percentage (P < 0.05). Compared to the control diet, 0.02% EA decreased quadratically the L* and increased a* of breast muscle at 45 min postslaughter (P < 0.05), and quadratically decreased (P < 0.05) the b* and increased linearly and quadratically (P < 0.05) drip loss. Additionally, EA improved linearly and quadratically (P < 0.05) serum total protein concentration and reduced linearly and quadratically (P < 0.05) serum blood urea nitrogen concentration. A total of 0.02% EA quadratically increased catalase activity and decreased malondialdehyde concentration in breast muscle compared with the control diet (P < 0.05). 0.02% and 0.04% EA could linearly and quadratically increase (P < 0.05) the concentrations of histidine, leucine and essential amino acids (EAA), 0.02% EA could linearly and quadratically increase (P < 0.05) the concentrations of threonine, glutamate, and flavored amino acids in breast muscle. 0.02% EA linearly and quadratically improved the C20:3n6, C22:6n3, polyunsaturated fatty acid (PUFA) concentrations, and the ratio of PUFA to saturated fatty acids (SFA), but reduced the C16:0 and the SFA concentrations in breast muscle than the CON group (P < 0.05). The EA diet linearly increased (P = 0.035) and quadratically tended (P = 0.068) to regulate the C18:2n6c concentration of breast muscle. Metabolomics showed that alanine metabolism, aspartate and glutamate metabolism, arginine and proline metabolism, taurine and hypotaurine metabolism, and glycerophospholipid metabolism were the most differentially abundant. These results showed that EA supported moderate positive effects on growth performance, meat quality, and metabolomics profile of broilers.

Blended-protein changes body weight gain and intestinal tissue morphology in rats by regulating arachidonic acid metabolism and secondary bile acid biosynthesis induced by gut microbiota.

The impact of dietary nutrients on body growth performance and the composition of gut microbes and metabolites is well-established. In this study, we aimed to determine whether dietary protein can regulate the physiological indexes and changes the intestinal tissue morphology in rats, and if dietary protein was a crucial regulatory factor for the composition, function, and metabolic pathways of the gut microbiota. A total of thirty male Sprague Dawley (SD) rats (inbred strain, weighted 110 ± 10g) were randomly assigned to receive diets containing animal-based protein (whey protein, WP), plant-based protein (soybean protein, SP), or a blended protein (soybean-whey proteins, S-WP) for a duration of 8weeks. To investigate the effects of various protein supplement sources on gut microbiota and metabolites, we performed a high throughput 16S rDNA sequencing association study and fecal metabolomics profiling on the SD rats. Additionally, we performed analyses of growth indexes, serum biochemical indexes, and intestinal morphology. The rats in S-WP and WP group exhibited a significantly higher body weight and digestibility of dietary protein compared to the SP group (P < 0.05). The serum total protein content of rats in the WP and S-WP groups was significantly higher (P < 0.05) than that in SP group, and the SP group exhibited significantly lower (P < 0.05) serum blood glucose levels compared to the other two groups. The morphological data showed the rats in the S-WP group exhibited significantly longer villus height and shallower crypt depth (P < 0.05) than the SP group. The gut microbial diversity of the SP and S-WP groups exhibited a higher level than that of the WP group, and the microbiomes of the WP and S-WP groups are more similar compared to those of the SP group. The Arachidonic acid metabolism pathway is the most significant KEGG pathway when comparing the WP group and the SP group, as well as when comparing the SP group and the S-WP group. The type of dietary proteins exerted a significant impact on the physiological indices of SD rats. Intake of S-WP diet can enhance energy provision, improve the body's digestion and absorption of nutrients, as well as promote intestinal tissue morphology. In addition, dietary protein plays a crucial role in modulating fecal metabolites by regulating the composition of the gut microbiota. Metabolomics analysis revealed that the changes in the levels of arachidonic acid metabolites and secondary bile acid metabolite induced by Clostridium_sensu_stricto_1 and [Eubacterium]_coprostanoligenes_group maybe the primarily causes of intestinal morphological differences.

Association of passive immunity and genetic composition, health, and performance of tropical dairy calves

This retrospective cohort study aimed to identify risk factors for the failure of transfer of passive immunity (FPI) in preweaned dairy calves, explore its associations with morbidity, mortality, genetics, and determine a standardized cut-off point for FPI. Analyzing data from 6011 calves, factors such as sire predicted transmitted ability for milk (PTA), birth season, retained placenta (RP), total serum protein concentration (TSP), morbidity (neonatal calf diarrhea - NCD, bovine respiratory disease - BRD, tick-borne disease - TBD), mortality, and average daily weight gain (ADG) were considered. The calves were categorized into predominantly Gyr (PG) and predominantly Holstein (PH) genetic composition groups. Multivariate mixed logistic regression revealed optimal TSP cut-off points for predicting morbidity (7.6 g/dL) and mortality (6.9 g/dL). PH calves exhibited 1.35 times higher odds of FPI and 1.48 times greater odds of disease. Calves from multiparous cows and those born to dams with RP had increased FPI odds. Disease prevalence was 53%, with 41% NCD, 18% BRD, and 10% TBD. Season, parity, PTA, and birth weight were associated with disease odds, though FPI was not a reliable predictor. The mortality percentage was 6%, with PH calves and those with a positive PTA having higher odds. The ADG was 0.64 kg, and FPI-affected calves gained less weight. The study compared and identified various risk factors that potentially impact calf immunity. However, the use of a standardized cut-off point for FPI assessment was not effective in predicting morbidity and mortality at this specific farm.

The effect of donation frequency on donor health in blood donors donating plasma by plasmapheresis: study protocol for a randomized controlled trial

BackgroundThe demand for plasma products is growing, necessitating an increase in plasma collection by plasmapheresis. While the 20th edition of the European Guidelines permits plasma donors in Europe to donate with 96-h donation intervals, the potential short- and long-term consequences of high-frequency plasma donations on donor health remain unknown. This study aims to measure the effect of plasma donation frequency on plasma protein composition, including total serum protein (TSP) and immunoglobulin G (IgG), in Norwegian male blood donors.MethodsThis randomized controlled trial (RCT) included 120 male blood donors who were randomized into two intervention groups and one control group: high-frequency plasma donors (HFPDs) who donated 650 mL of plasma 3 times every 2 weeks, whereas regular-frequency plasma donors (RFPDs) who donated 650 mL of plasma 1 time every 2 weeks. The control group consisted of whole blood donors. The primary outcomes are the concentrations of TSP and IgG.DiscussionThe findings from this study may have implications for recommendations related to donor health and plasma donation frequencies and may contribute to supporting the strategic independence of plasma products in Norway and Europe without compromising donor health.Trial registrationClinicalTrials.gov: NCT05179200. Enrichment of medium-quality colostrum by adding colostrum replacer, combined or not with transition milk in the feeding of dairy calves

Fifty Holstein calves were allocated in randomized blocks and distributed in a 2 × 2 factorial arrangement; (A) two sources of Ig: (1) Control: bovine colostrum (25% Brix); (2) Enriched colostrum: mid-quality bovine colostrum (20% Brix) enriched with colostrum replacer to 25% Brix; and (B) two transition feeding diets: (1) Whole milk (WM): supply of 4 L/day of whole milk for 3 days after the colostrum feeding; and (2) Formulated transition milk (FTM): supply 4 L/day of whole milk enriched with 70 g/L of colostrum replacer for 3 days after the colostrum feeding. Blood samples were collected at 0, 24, 48, and 72 h of age to determine total serum protein (TSP), glucose, non-esterified fatty acids (NEFA), erythrocyte and leukocyte concentrations. IgG was measured at 48 h. During the preweaning period, calves received 6 L/day of whole milk. Blood samples were collected weekly to determine TSP, glucose, and lactate. The colostrum protocols were equally efficient for transfer of passive immunity with IgG concentration at 48 h ≥ 49.6 g/L. Colostrum or transition feeding program did not influence the erythrocyte and leukocyte concentrations. The TSP concentration measured until 72 h was higher for calves fed maternal colostrum. Calves fed milk in the transition period had higher glucose concentrations. Calves receiving bovine colostrum and FTM had higher glucose concentrations in the preweaning period, while the enriched colostrum decreased plasma lactate concentrations. In summary, enrichment of mid-quality colostrum is an alternative in situations of a shortage of high-quality colostrum; however, feeding 4 L/day of FTM only for 3 days after colostrum feeding does not show additional benefits.

Diet with optimal glutathione supplement improves growth, nonspecific immunity, intestinal microbiota, and antioxidant ability in Micropterus salmoides.

In this study, Micropterus salmoides were fed with dietary glutathione (GSH, 0, 100, 300, and 500 mg/kg) for 56 days to investigate its effects on growth performance, serum nonspecific immunity, liver antioxidant capacity, tissue morphology, and intestinal microbiota. The results showed that the survival rate, weight gain rate, and specific growth rate and condition factor increased, whereas the feed conversion ratio, hepato-somatic index, and viscerosomatic index decreased in the GSH groups. Compared with the control group, the serum total protein content significantly increased, whereas the triglyceride and total cholesterol significantly decreased in the 300-mg/kg dietary GSH group. The activities of lysozyme, alkaline phosphatase, and acid phosphatase were significantly higher in GSH-supplemented groups, peaking at 300-mg/kg GSH. GSH supplementation significantly increased total antioxidant capacity and decreased malondialdehyde content, with the most pronounced effects at 300-mg/kg GSH. Further antioxidant indicators showed that a dietary supplement of 300-mg/kg GSH significantly increased the activities of superoxide dismutase, glutathione transferase, endogenous glutathione, glutathione reductase, and catalase. At 300-mg/kg GSH, the liver exhibited improved characteristics with alleviated vacuolation and hepatocyte nuclear shift, and intestine showed enhanced structure with increased villus height and intestinal wall thickness. Additionally, a 300-mg/kg GSH supplementation improved the diversity of intestinal microbiota, increased the abundance of probiotics such as Bacillus, and inhibited the development of pathogenic bacteria such as Plesiomonas. Overall, the results suggest that the effect of GSH addition on improving growth performance, nonspecific immunity, antioxidant capacity, and intestinal microbiota of M. salmoides is best in the 300-mg/kg addition group. Based on second-degree polynomial regression analysis of weight gain, the optimum requirement of dietary GSH in M. salmoides is a 336.84-mg/kg diet.

Interaction effects of glycine equivalent and standardized ileal digestible threonine in low protein diets for broiler grower chickens.

This study aims to investigate the interactive effect of a glycine equivalent (Glyequi) and standardized ileal digestible threonine (SID Thr) levels in low crude protein diets on performance, blood biochemistry, pectoral muscular creatine content and oxidative stability of meat in broiler chickens from 21 to 42 days. A total of 1,500, twenty-one-day-old Cobb-Vantress male broiler chickens were distributed in a completely randomized 5×3 factorial arrangement of Glyequi×SID Thr with five replicates of 20 birds each. Fifteen dietary treatments of 16.5% CP were formulated to contain five levels of total Glyequi (1.16%, 1.26%, 1.36%, 1.46%, and 1.56%) and three levels of SID Thr (0.58%; 0.68% and 0.78%). Interaction effects (p<0.05) of Glyequi and SID Thr levels were observed for weight gain, carcass yield, pectoral muscular creatine content and serum uric acid. Higher levels of Glyequi increased (p = 0.040) weight gain in 0.58% and 0.68% SID Thr diets compare to the 0.78% SID Thr diet. The SID Thr level at 0.68% improved (p = 0.040) feed conversion compared to other SID Thr diets. Levels of Glyequi equal to or above 1.26% in diets with 0.78% SID Thr resulted in birds with higher (p = 0.033) pectoral muscular creatine content. The breast meat yield observed in the 0.68% SID Thr diet was higher (p = 0.05) compared to the 0.58% SID Thr diet. There was a quadratic effect of Glyequi levels for pectoral pectoral muscular creatine content (p = 0.008), breast meat yield (p = 0.030), and serum total protein concentrations (p = 0.040), and the optimal levels were estimated to be 1.47%, 1.35%, and 1.40% Glyequi, respectively. The lowest (p = 0.050) concentration of malondialdehyde in the breast meat was found in 0.68% SID Thr diets at 1.36% Glyequi. The minimum dietary level of Glyequi needed to improve performance in low crude protein diets is 1.26% with adequate SID Thr levels for broiler chickens.

Star anise extract supplementation improved growth performance, hepatic-antioxidant enzyme status, hematology, serum biochemistry, and survival against crowding stress in Catla catla

This study was conducted to evaluate the effectiveness of star anise extract (SAE) (prepared using ethanol as the solvent) on growth, proximate composition, hepatic-antioxidant enzyme status, blood-serum indices, and survival against crowding stress in Catla calta. Healthy fingerlings (N = 900) with an average initial weight of 14.16 ± 0.11 g were acclimatized under laboratory conditions and then placed in 5 hapas (60 fish in each) in triplicate. SAE was supplemented in basal feed at 0.0, 0.5, 1.0, 2.0 and 4.0 g/kg and fed to the experimental groups up to satiation twice daily for 90 days. After completion of the feeding trial, 40 fish from each treatment were subjected to crowding stress (60 kg/m3) in triplicate. In the feeding trial, orthogonal polynomial contrast analysis revealed significant enhancement in final weight, weight gain and specific growth in linear and quadratic trends with weight gain percentage, feed conversion ratio and protein efficiency in a quadratic trend observed in response to SAE supplementation. The optimum supplementation level of SAE based on weight gain % was estimated to be 0.93 g/kg using broken line regression analysis (R2 = 0.97) for C. catla. No significant differences in survival rate or proximate composition were observed during the feeding trial. Dietary supplementation with SAE improved the activities of antioxidant enzymes (catalase, superoxide dismutase and glutathione peroxidase) in linear and quadratic trends. The activities of hepatic enzymes, such as alanine transferase, alkaline phosphatase and aspartate transaminase, were also enhanced linearly and quadratically in response to SAE supplementation. Furthermore, hematological indices (red blood cells, white blood cells, hematocrit, hemoglobin and mean corpuscular hemoglobin concentration) were also significantly improved with SAE supplementation. The serum total protein, albumin and globulin concentrations increased linearly and quadratically. Furthermore, stress markers (glucose, cortisol) and triglycerides were significantly reduced linearly and quadratically in response to SAE supplementation. Lysozyme activities also increased linearly and quadratically, indicating a better immune response. After 7 days of crowding stress, the lowest cumulative mortality was observed for the fish fed a diet supplemented with 1.0 g/kg SAE. In conclusion, dietary SAE supplementation significantly improved growth, hepatic-antioxidant enzyme status, blood-serum indices and survival against crowding stress in C. calta.

Immunity transfer in mule foals fed with good IgG quality colostrum

While the passive transfer of immunity in horse and donkey foals has been extensively studied, there is limited information for mule foals. Immunoglobulin type G (IgG) and serum total protein concentration (TP) were assessed at different sampling times to evaluate the correlation between serum radial immunodiffusion (SRID) with electrophoresis, refractometry, and dry chemistry analyzer (Biuret), and to estimate serum IgG concentrations using serum TP in mule foals. We analyzed a total of 30 samples collected at birth, and at 6, 12, 24, and 48 h of life from 6 mule foals by SRID, electrophoresis TP, biuret TP, and refractometry TP.The SRID IgG concentration significantly increased from birth until T6 (p < 0.001). Serum TP analyzed with refractometry revealed differences between T0 and T12, T24 and T48 (p < 0.05), while a significant difference was observed with the biuret method between T0 and all the other sampling times (p < 0.001).A strong correlation was found between IgG SRID and biuret TP (r = 0.69, p < 0.001), and a good correlation existed between IgG SRID, refractometry TP, and electrophoresis TP (r = 0.44, p < 0.01 and r = 0.39, p < 0.05, respectively).All methods can be used to estimate the passive transfer of immunity in mule foals. TP refractometry and biuret TP values can be used to determine serum IgG concentrations in the blood of mule foals on their first day of life through the application of a specific equation.

The American cockroach (Periplaneta americana) residue could partially replace the dietary fish meal in the juvenile Nile tilapia (Oreochromis niloticus)

The American cockroach (Periplaneta americana) residue is a by-product of P. americana meal after extracting active ingredients with ethanol, and it still contains high protein, lipid, and a certain amount of active ingredients, may be a potential fish meal alternative. Five isonitrogenous and isolipidic experimental diets were formulated to replace 0%, 25%, 50%, 75%, and 100% of dietary fish meal with P. americana residue, and defined as Diet 1, Diet 2, Diet 3, Diet 4, and Diet 5, respectively. The juvenile Nile tilapia (Oreochromis niloticus) with an initial body weight of 2.98 ± 0.01 g was fed with respective diets for 10 weeks, which was conducted to investigate the effects of dietary fish meal replacement by P. americana residue on the growth performance, proximate composition, amino acid profile, metabolism, antioxidant capacity, and immunity of juvenile O. niloticus. There were no significant differences in the final body weight, weight gain rate, and specific growth rate among five diet groups, while the highest feed conversion ratio and the lowest protein efficiency ratio were recorded in Diet 4. There were no significant differences in the contents of crude protein, total lipid, ash, and total amino acid in the whole fish body among all groups. The Diet 1 and Diet 2 had higher activities of trypsin, α-amylase, and chitinase in the intestine and α-amylase in the liver. The highest serum urea content was detected in Diet 4, while the lowest liver urea content and the highest serum total protein content were detected in Diet 3. A higher liver catalase, glutathione peroxidase, and serum lysozyme activities were detected in Diet 2, and the lowest serum malondialdehyde content was detected in Diet 2. In conclusion, dietary fish meal replacement by the P. americana residue had no negative effects on the growth performance and body composition of juvenile O. niloticus, and significantly improved the antioxidant capacity and immunity, while feed utilization rate was reduced when dietary fish meal replacement levels exceeded 75%.

Effects of dietary supplementation of postbiotic derived from Bacillus subtilis ACCC 11025 on growth performance, meat yield, meat quality, excreta bacteria, and excreta ammonia emission of broiler chicks

The primary aim of this study was to explore the impact of dietary supplementation with a postbiotic derived from Bacillus subtilis ACCC 11025 on growth performance, meat yield, meat quality, excreta bacterial populations, and excreta ammonia emissions of broiler chicks. A total of 480 day-old Arbor Acre broiler chicks, initially weighing 52.83 ± 1.38 g, were randomly allocated into 4 distinct groups. Each group was housed in 6 separate cages, each containing 20 birds. The experimental phase spanned 42 d, divided into 2 periods (d 1-21 and d 22-42). Dietary interventions were based on a basal diet, with postbiotic supplementation at levels of 0.000, 0.015, 0.030, or 0.045%. Our findings indicate that dietary supplementation with postbiotic had a positive influence on body weight gain (BWG) and feed efficiency. The most substantial improvements in BWG and feed efficiency were observed in the group of broiler chicks fed a diet containing 0.015% postbiotic. Furthermore, the inclusion of postbiotic in the diet led to an increase in the yield of breast and leg muscles, with a significant difference in meat yields observed between the control group and the group receiving 0.015% postbiotic supplementation. It's noteworthy that dietary manipulation did not exert any discernible impact on the quality of breast and leg muscle samples. Concurrently, we observed an elevation in serum albumin and total protein contents corresponding to the increasing postbiotic dosage in the diet. Additionally, dietary supplementation with postbiotic effectively controlled the emission of ammonia from excreta and reduced the abundance of Salmonella in excreta while enhancing the presence of Lactobacillus bacteria. The group receiving 0.015% postbiotic supplementation displayed the lowest levels of ammonia emission and the highest counts of Lactobacillus bacteria in excreta. In light of these results, we conclude that dietary supplementation with 0.015% postbiotic represents an efficacious strategy for increasing BWG and meat yield of broiler chicks by enhancing feed efficiency as well as mitigating ammonia emissions from excreta by modulating the composition of excreta bacterial communities.

Effects of α-amylase and coated α-amylase supplementation on growth performance, nutrient digestion, and rumen fermentation in Holstein bulls.

This study evaluated the impacts of α-amylase (AM) and coated α-amylase (CAM) on bull performance, nutrient digestibility, and ruminal fermentation. This study randomized 60 Holstein bulls of 365 ± 11.5 days of age and 457.5 ± 9.35 kg body weight into three groups: without AM addition, adding AM 0.6 g/kg dry matter (DM), and adding CAM 0.6 g AM/kg DM, separately. This whole experimental period was 80 days, including a 20-day adaptation period and a 60-day data and sample acquisition period. In comparison with the unsupplemented control, dry matter intake (DMI) was unaltered; however, average daily gain (ADG) and feed efficiency (FE) were greater for AM or CAM addition. Bulls receiving AM or CAM supply had greater total-tract nutrient digestibility, ruminal total volatile fatty acids (VFA) content, propionate molar proportion, cellulolytic enzyme and AM activities, and the number of microorganisms. In addition, the activities of AM and trypsin in the jejunum and ileum and glucose, albumin, and total protein concentrations in serum were greater for AM or CAM addition compared to the control. When comparing the supplementation mode of AM, bulls receiving CAM addition had greater ADG and FE. The crude protein and starch digestibility and intestinal AM and trypsin activity were higher, while acid detergent fiber (ADF) digestibility was lower for CAM addition than for AM addition. The lower propionate molar proportion and cellobiase and carboxymethyl cellulase activities, together with Ruminococcus albus, Ruminococcus flavefaciens, and Fibrobacter succinogenes populations were observed for CAM addition compared with AM addition. However, there were greater glucose, albumin, and total protein concentrations in serum after adding CAM. According to the data, the supply of AM improved ADG, nutrient digestion, and rumen fermentation. Notably, the optimum supplementation mode was in the form of CAM in bulls.

Rehydration post-transport: duration of oral fluid therapy on behavior, biochemical measures of hydration, and health of neonatal dairy calves.

The objective of this study was to determine the impact of 0, 1, 2, or 3d of oral electrolyte solutions (OES) following transport on calf behavior, biochemical measures of hydration, and subsequent health. Two cohorts of 60 Holstein bull calves ~3 to 7d of age were included in this experiment. Calves (n = 30/treatment) were randomized to 1 of 4 treatments on arrival at a calf-raising facility: (1) 1-d OES, (2) 2-d OES, (3) 3-d OES, or (4) no OES (control). Calves were evaluated for signs of arthritis, depression, dehydration, diarrhea, fever, navel inflammation, and respiratory disease on arrival (day 0) and 1, 2, 3, and 7d after arrival. Blood was obtained following each examination to assess biochemical measures of hydration, including serum electrolytes, glucose, hematocrit, lactate, pH, and serum total protein (STP). Calves were fitted with 3D accelerometers attached to the right hind leg along the metatarsus bone to continuously measure lying time. Linear regression models with Gaussian or Poisson link functions were used to test differences between treatments in blood parameters, as well as disease frequency and behavior, respectively. Calves spent the most time lying immediately post-transport (day 0); however, there was no effect of OES treatment on lying time. On arrival, 90% (95% CI: 84.6% to 95.4%) of calves had mild to moderate dehydration, and there was no significant difference in hydration status across treatments. Mildly dehydrated calves that received 1- and 2-d OES had higher STP concentration compared to the control. Moderately dehydrated calves that received 3- vs. 1-d OES had higher (101.4 vs. 93.7mmol/L, respectively; P = 0.01) glucose concentration, and 3- vs. 2-d OES (101.4 vs. 96.2, respectively; P = 0.07) calves tended to have higher glucose concentration. The prevalence of failed transfer of passive immunity (STP < 5.1g/dL) and hypoglycemia (glucose < 4.95mmol/L) was 26.7% (18.6% to 34.7%) and 76.5% (68.7% to 84.2%), respectively. The frequency of moderate dehydration was lower for calves that received 2-d OES compared to the control (count ratio: 0.4; 0.2% to 0.8%; P = 0.009). These results underline the need for more research on rehydration strategies to help inform calf processing protocols. Future research should investigate preconditioning strategies to minimize the impact of marketing and transport on dairy calves. Continued nutritional-based research is also needed to better support calves' recovery post-transport.

Variation in protein metabolism biomarkers during the transition period and associations with health, colostrum quality, reproduction, and milk production traits in Holstein cows

The aims of this study were to assess a) the variation of protein metabolism biomarkers and of factors affecting them during the transition period, b) the association of each biomarker with skeletal muscle reserves and their changes, and c) the association of these biomarkers with postpartum health, colostrum quality, reproduction, and milk production. For this purpose, 238 multiparous Holstein cows from 6 herds were used in a prospective cohort study. Plasma concentrations of 3-methylhistidine (3-MH) and 1-methylhistidine (1-MH) and serum concentrations of total protein (TP), albumin (ALB), urea nitrogen (BUN) and creatinine (SCR) were determined for each cow at -21d; -7d; 7d; 21d and 28d relative to calving. Clinical diseases were recorded during the first 28 d post-calving, and presence of subclinical ketosis (scKET) was investigated at 7d and 21d. Colostrum quality was estimated by Brix refractometry. Reproduction data by 150 d-in-milk (DIM) and milk production records were also available. Linear mixed models including the fixed effects of time point, herd, parity, body condition score (-21d), duration of dry period and post-parturient diseases were fitted to assess the variation in each biomarker's concentration. The association between the biomarkers' concentration during the pre-partum period with the odds for each post-parturient disease and for a combined trait (CD_1-28), defined as the presence of at least one clinical condition during the first 28 d after calving, were assessed with separate binary logistic models for time points -21d and -7d. The relationship of each biomarker's concentration with longissimus dorsi thickness (LDT) and the changes in LDT (ΔLDT) was assessed with pairwise correlations. Separate general linear models were used to assess the association of each biomarker with colostrum Brix values and milk production traits. Finally, the associated hazard for 1st artificial insemination (AI) and for pregnancy by 150 DIM (PREG_150DIM) was assessed with Cox proportional hazard models, while odds for pregnancy to the 1st AI (PREG_1stAI) were assessed with binary logistic models. 3-methylhistidine was affected mainly by herd, time points and their interaction. Higher 3-MH was associated with increased odds for metritis and CD_1-28, increased hazard for PREG_150 DIM and with increased milk production. 1-methylhistidine was affected mainly by herd, scKET and occurrence of displaced abomasum. Higher 1-MH was associated with better colostrum quality, increased odds for scKET, increased hazard for 1st AI by 150 DIM and with decreased milk production. Both 3-MH and 1-MH were weakly to moderately negatively correlated with LDT and moderately to strongly negatively correlated to ΔLDT at the corresponding time-periods. Additionally, higher TP was associated with increased odds for metritis and CD_1-28 and increased milk production, while higher ALB was associated with increased odds for scKET and increased milk production. Moreover, higher BUN was associated with decreased odds for scKET, increased odds for PREG_1stAI and increased milk production. Higher SCR was associated with decreased odds for retained fetal membranes, metritis, and CD_1-28. Peri-parturient protein metabolism is significantly associated with postpartum health, colostrum quality, reproduction, and milk production; mechanisms involved require further investigation.

Antioxidant and hepatoprotective activities of Juniperus excelsa M. Bieb against bile duct ligation-induced cholestasis

Background and purpose: Cholestasis is caused by a malfunction of the biliary liver system. Oxidative stress plays an essential role in the progression of cholestasis. This study aimed to investigate the antioxidant and hepatoprotective effects of ethanolic extract of Juniperus excelsa M. Bieb (JE) fruits on hepatic impairment induced by bile duct ligation (BDL) in rats. Experimental approach: Forty male Wistar rats were randomly divided into 4 groups; sham control + vehicle (SC), BDL + vehicle (BDL), BDL + JE extract (BDL + JE), and SC + extract (SC + JE). One day after surgery, the animals were treated with vehicle or ethanolic extract of JE (500 mg/kg/day) for 7 days. Finally, the blood was taken for biochemical and oxidative stress analysis. Furthermore, the liver tissue of rats was removed for histological examination. Findings/Results: Treatment with the extract of JE decreased the ALP level, whereas it enhanced total protein content compared to the BDL group. Also, JE increased the activity of SOD and GPx, as well as FRAP content compared to the BDL group; while it did not significantly affect the levels of MDA and inflammation markers. However, JE could not improve BDL-induced histopathological alterations in hepatic tissue. Conclusion and implication: This study demonstrated that JE may be useful as an adjuvant therapy by attenuating ALP activity, increasing serum total protein and FRAP content, as well as improving the antioxidant enzymes activity of SOD and GPx. However, further research is warranted to explore the other underlying mechanisms of action.
