Effects of Summer Heat on Adipose Tissue Activity in Periparturient Simmental Cows.

Hot climate is one of the major factors affecting the dairy industry. Heat stress could be responsible for decreased feed intake and consequently leads to alterations in energy metabolism, particularly during late pregnancy and early lactation. This study aimed to assess the effects of summer heat on adipose tissue activities during the periparturient period in Simmental cows. Two groups of cows were involved: heat-stressed cows (n = 12) that calved from June to August and thermoneutral cows (n = 12) that calved from October to December. Blood samples were taken from each cow during the periparturient period: 21 and 7 days before calving and 8, 16, 24, 32, and 40 days after calving. Glucose, beta-hydroxy butyrate (BHB), non-esterified fatty acids (NEFA), leptin (LP), and adiponectin (ADP) were measured in serum samples by commercial kits. Thermoneutral cows expressed higher degrees of lipomobilization syndrome than heat-stressed cows, indicated by significantly higher serum NEFA and BHB concentrations in the early lactation. Leptin levels were significantly decreased, while adiponectin was increased in heat-stressed cows compared to thermoneutral ones. The results indicated that heat-stressed cows during the periparturient period mobilized less fat from adipose tissue to reduce the heat generation by fatty acid oxidation.

Hydrogen-rich water supplementation improves metabolic profile during peripartum period in Gurcu goats and enhances the health and survival of kids

In this study, the effect of intaking hydrogen-rich water (HRW) on the metabolic profile of Gurcu goats during the peripartum period and the survival/growth performance of kids were evaluated. Twenty-three pregnant goats were divided into two groups 21–23 days before the due date. Group 1 (G1, n = 10) was given HRW from day 21 before delivery until day 21 after delivery. Group 2 (G2, n = 13) served as the control. Blood samples were weekly taken from 21 days before delivery until 21 days after delivery. Hydrogen-rich water increased serum glucose concentration on the delivery day more than in G2 (P = 0.016). Hydrogen-rich water decreased serum total cholesterol (P = 0.02) and creatinine (P = 0.05) concentration at delivery. Group effect and time effect were significant in triglyceride (P < 0.001, P = 0.001, respectively) and albumin (P < 0.001, P = 0.002, respectively) concentration. Aspartate transaminase decreased towards the delivery day in G1 (P < 0.05). Serum non-esterified fatty acids concentration was lower in G1 than in G2, but there was no significant differences (P > 0.05). Beta-hydroxybutyric acid concentration an increased in both groups during the prepartum period, although there was no significance (P > 0.05). Hydrogen-rich water did not affect the birth weight and growth performance of the kids (P > 0.05), but it increased their survival rates and overall health, although there was no significance (P > 0.05). In conclusion, HRW may have an impact on the metabolic profiles during the peripartum period and have a positive effect on lipid profiles. Additionally, intaking HRW to goats during the peripartum period may improve the health and survival of kids and reduce their mortality.

Effect of Myrtus communis L. Plant Extract as a Milk Supplement on the Performance, Selected Blood Parameters and Immune Response of Holstein Calves.

This research aimed to understand the effects of adding myrtle plant extract obtained from its leaves (MPEL) and roots (MPER) to the milk fed to suckling female Holstein calves, focusing on performance, reproduction, selected blood parameters and immune response. The 50 Holstein female calves, one week of age, were divided into five groups: one group received no plant extract (Control), while the others were supplemented with myrtle plant extracts at doses of 25 mL/day leaf extract (MPEL-25), 25 mL/day root extract (MPER-25), 50 mL/day leaf extract (MPEL-50) and 50 mL/day root extract (MPER-50) for each calf in each treatment group. The extracts were given along with the milk to the experimental groups for 60 days, and for an additional 12 days post-weaning. The results reveal that the feed consumption and live weights increased significantly. Significantly higher leukocyte counts were observed in the 50 mL/head × day myrtle groups, and a higher IgG concentration was also noted in the MPER-50 group compared to the other groups. The serum non-esterified fatty acid (NEFA) concentration significantly decreased in the MPEL-50 and MPER-50 groups, whereas the betahyrdoxy butyric acid (BHBA) concentration increased and the serum glucose concentration significantly decreased with myrtle supplementation. In conclusion, it was determined that the performance, immune system and negative energy balance compensation of female Holstein calves were positively affected by administering extracts obtained from the leaves and roots of the Myrtus communis L. plant at dose levels of 25 and 50 mL/head × day for 72 days, without causing any side effects.

Comparison of serum very low-density lipoprotein concentrations during transition in primiparous and multiparous cows.

This study was carried out as an observational study in order to examine the difference of change in serum very low-density lipoprotein (VLDL) between primiparous and multiparous cows. Twenty-one clinically healthy cows (10 primiparous and 11 multiparous) were selected at 21 days prior to expected calving. Blood samples were collected in the morning (before feeding) on days -21, -7, 7, 21 and 56 days in milk (DIM). At 7 and 21 DIM, the serum non-esterified fatty acid concentration of multiparous cows was significantly higher than that of primiparous cows. The serum β-hydroxybutyrate concentration was also markedly higher in multiparous cows than in primiparous cows at 21 DIM. These results suggested that the degree of negative energy balance was greater in multiparous cows than in primiparous cows during this period. In both, serum VLDL concentrations decreased at over 7 DIM, increased at 21 DIM, and then decreased at 56 DIM. On the other hand, triglyceride and total protein concentrations of VLDL in multiparous cows were significantly lower than in primiparous cows at 21 DIM. This suggests that multiparous cows have poor triglyceride secretion from the liver and that they become more susceptible to hepatic lipidosis.

Influences of oxidative stress and energy balance on pregnancy rates after the transfer of embryos to repeat-breeder Japanese Black cattle.

The purpose of this study is to reassess our previously reported findings on the diminished pregnancy rate of embryo transfer (ET) in summer for repeat-breeder (RB) cows compared with other seasons, with a focus on oxidative stress and energy balance. The study involved Japanese Black cattle, including 224 heifers (aged <3years) and 278 (aged <9years) cows with one or more deliveries, defined as RB animals. Evaluation of the effects of season on pregnancy rates revealed significantly lower rates in RB cows during summer compared with spring and autumn. Moreover, serum malondialdehyde (MDA) concentration in RB cows during summer was significantly higher than in winter, with no difference in RB heifers. Seasonal effects on serum nonesterified fatty acid (NEFA) concentration in both RB heifers and RB cows showed no significant differences. However, median MDA and NEFA concentrations in RB cows were significantly elevated compared with RB heifers, suggesting that factors contributing to low fertility should consider parity. Furthermore, our study indicated that RB cows were under oxidative stress, as demonstrated by significantly elevated MDA concentrations compared with normally reproducing cows in summer. Further investigation is necessary to gain a better understanding of these observations in the future.

Use of milk mid-infrared spectra to predict serum non-esterified fatty acid concentrations in Chinese Holstein cows

Negative energy balance (NEB) in high-yielding cows during the peripartum period raises the risk of postpartum diseases. High-level concentration of non-esterified fatty acid (NEFA) is a good indicator of excessive NEB. The current low-cost and high-throughput mid-infrared (MIR) spectroscopy method is gradually applied to predict NEFA concentrations for NEB identification. The objective of this study was to compare different pre-processing methods and analysis models for optimal predictions of serum NEFA using milk MIR spectra. Four spectral pre-processing methods: standard normal variate, first-order derivative (FD), second-order derivative, and Savitzky-Golsy convolution smoothing, and four prediction models: partial least squares regression, ridge regression, lasso regression (LassoR), and random forest regression were investigated. In total, 366 collected serum and milk samples within the 1–7 weeks postpartum were randomly divided into the training (70%) and test (30%) sets for cross-validations. The results showed that the combined strategy of FD-LassoR model when parity and days in lactation information were considered resulted in the highest R2 = 0.643, RMSE = 0.153 mmol/L, and highest residual predictive deviation = 1.665 of predictions on the test set. In addition, R2 and RMSE values of FD-LassoR combined with other information were still higher than the other four prediction scenarios. Therefore, our study enables the optimal prediction of serum NEFA concentrations using milk MIR spectra in the further research and practical applications.

Inositol-requiring enzyme 1α and c-Jun N-terminal kinase axis activation contributes to intracellular lipid accumulation in calf hepatocytes

During the perinatal period, dairy cows undergo negative energy balance, resulting in elevated circulating levels of nonesterified fatty acids (NEFA). Although increased blood NEFA concentrations are a physiological adaptation of early lactation, excessive NEFA in dairy cows is a major cause of fatty liver. Aberrant lipid metabolism leads to hepatic lipid accumulation and subsequently the development of fatty liver. Both inositol-requiring enzyme 1α (IRE1α) and c-Jun N-terminal kinase (JNK) have been validated for their association with hepatic lipid accumulation, including their regulatory functions in calf hepatocyte insulin resistance, oxidative stress, and apoptosis. Meanwhile, both IRE1α and JNK are involved in lipid metabolism in nonruminants. Therefore, the aim of this study was to investigate how IRE1α and JNK regulate lipid metabolism in bovine hepatocytes. An experiment was conducted on randomly selected 10 healthy cows (hepatic triglyceride [TG] content <1%) and 10 cows with fatty liver (hepatic TG content >5%). Liver tissue and blood samples were collected from experimental cows. Serum concentrations of NEFA and β-hydroxybutyrate (BHB) were greater, whereas serum concentrations of glucose and milk production were lower in cows with fatty liver. The western blot results revealed that dairy cows with fatty liver had higher phosphorylation levels of JNK, c-Jun, and IRE1α in the liver tissue. Three in vitro experiments were conducted using primary calf hepatocytes isolated from 5 healthy calves (body weight: 30-40 kg; 1 d old). First, hepatocytes were treated with NEFA (1.2 mM) for 0.5, 1, 2, 3, 5, 7, 9, or 12 h, which showed that the phosphorylated levels of JNK, c-Jun, and IRE1α increased in both linear and quadratic effects. In the second experiment, hepatocytes were treated with high concentrations of NEFA (1.2 mM) for 12 h with or without SP600125, a canonical inhibitor of JNK. Western blot results showed that SP600125 treatment could decrease the expression of lipogenesis-associated proteins (PPARγ and SREBP-1c) and increase the expression of fatty acid oxidation (FAO)-associated proteins (CPT1A and PPARα) in NEFA-treated hepatocytes. The perturbed expression of lipogenesis-associated genes (FASN, ACACA, and CD36) and FAO-associated gene ACOX1 were also recovered by JNK inhibition, indicating that JNK reduced excessive NEFA-induced lipogenesis and FAO dysregulation in calf hepatocytes. Third, short hairpin RNA targeting IRE1α (sh-IRE1α) was transfected into calf hepatocytes to silence IRE1α, and KIRA6 was used to inhibit the kinase activity of IRE1α. The blockage of IRE1α could at least partially suppressed NEFA-induced JNK activation. Moreover, the blockage of IRE1α downregulated the expression of lipogenesis genes and upregulated the expression of FAO genes in NEFA-treated hepatocytes. In conclusion, these findings indicate that targeting the IRE1α-JNK axis can reduce NEFA-induced lipid accumulation in bovine hepatocytes by modulating lipogenesis and FAO. This may offer a prospective therapeutic target for fatty liver in dairy cows.

Higher circulating levels of non-esterified fatty acids are associated with faster kidney function decline in post-menopausal women with type 2 diabetes: a pilot prospective study.

Currently, there is little and inconsistent evidence regarding the possible adverse effects of circulating levels of non-esterified fatty acids (NEFA) on kidney function decline in patients with type 2 diabetes mellitus (T2DM). We followed for a median of 4.6 years 85 post-menopausal women with non-insulin-treated T2DM and preserved kidney function at baseline. Serum NEFA concentrations were measured using an enzymatic colorimetric method. Glomerular filtration rate (eGFR) was estimated using the Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equation. Enrolled patients had a baseline mean eGFRCKD-EPI of 83 ± 12mL/min/1.73m2 and a median serum NEFA concentration of 662uEq/L (interquartile range 524-842uEq/L). During the follow-up period, 13 patients developed kidney function decline at follow-up (defined as an eGFRCKD-EPI decline ≥ 30% from baseline). In Cox proportional hazards regression analyses, higher serum NEFA levels were significantly associated with an increased risk of developing kidney function decline (adjusted-hazard ratio 3.67, 95% CI 1.64-8.22, p < 0.001; for each 1-SD increment, i.e., 262uEq/L), even after adjustment for waist circumference, hemoglobin A1c, C-reactive protein, HOMA-estimated insulin resistance, hypertension, dyslipidemia, microalbuminuria, baseline eGFRCKD-EPI, as well as temporal changes in HbA1c levels or the use of renin-angiotensin system inhibitors over the follow-up. The findings of this exploratory prospective study show that in post-menopausal women with T2DM and preserved kidney function at baseline, higher circulating levels of NEFA were strongly associated with a faster kidney function decline, even after adjustment for established renal risk factors and potential confounders.

Does the peroral chromium administration in young Hariana calves reduce the risk of calf diarrhea by ameliorating insulin response, lactose intolerance, antioxidant status, and immune response?

BackgroundThe reduction in insulin sensitivity during rumen development may predispose dairy calves towards lactose intolerance, which could be the reason behind neonatal calf diarrhea (NCD). Chromium (Cr) results in a range of effects when fed to ruminants, but most studies have shown improved insulin sensitivity. The aim of this study was to determine the effect of Cr supplementation on insulin sensitivity, lactose intolerance, diarrhoea and antioxidant, and immune response in young Hariana calves. MethodsA total of 20 milk-fed Hariana calves were randomly assigned to 1 of 2 treatments, each consisted of 10 calves: (1) a control group without supplemental Cr and (2) a 0.15 mg Cr as Cr-picolinate (CrPic)/kg BW0.75 supplemented group (Cr0.15). ResultsA more rapid glucose disappearance with unaltered insulin kinetics during intravenous glucose tolerance test (IVGTT) and oral lactose tolerance test (OLTT) indicates greater insulin sensitivity in Cr supplemented calves. Better insulin sensitivity in Cr supplemented calves was further confirmed by higher values of the quantitative insulin sensitivity check index (QUICKI), revised quantitative insulin sensitivity check index (RQUICKI) and insulin receptor substrate-1 (IRS-1) and lower (P < 0.05) values of homeostasis model assessment-insulin resistance (HOMA-IR) and glucose-to-insulin ratio in Cr supplemented calves during IVGTT. Cr supplementation resulted in a lower (P < 0.05) serum cortisol concentration, whereas serum non-esterified fatty acid (NEFA) concentrations during IVGTT did not differ among the groups. The rise in serum glucose concentrations within 2 h post lactose infusion during OLTT peaked at more than twice the basal glucose concentration, therefore calves were not considered as lactose intolerant. Within monthly blood samples, concentrations of serum insulin were similar among treatments, whereas the Cr supplemented group had lower (P < 0.05) serum glucose concentration and glucose-to-insulin ratio compared with the control group. No treatment differences were detected in the biomarkers of antioxidant status and immunity. Serum Cr concentrations were higher (P < 0.05) in Cr supplemented calves while concentrations of other studied minerals were remained unaltered. The incidence, duration of diarrhea, and faecal score were better (P < 0.05) in calves fed on Cr supplemented diet whereas, no treatment effect was observed on average daily gain (ADG). During the study period, no calves died, and no calves were found to have pneumonia, navel or joint disease. ConclusionFeeding a Cr-supplemented diet improved insulin sensitivity and reduced the risk of diarrhoea in milk-fed young calves, but had no or minimal effects on lactose intolerance, antioxidant status, immune response, and growth performance.

Ovarian activity, metabolic and physiological parameters of canindé goats submitted to short-term supplementation with licuri oil

This study aimed to evaluate the estrous behavior, ovarian activity, feed intake, metabolic and physiological parameters of Canindé goats supplemented for nine days with Licuri oil (Syagrus coronata). Thirty-six adult female goats were distributed randomly into two groups: control group (n = 18) without Licuri oil and the oil group (n = 18) receiving a supplement containing 3% Licuri oil. The short-term food supplementation was associated with an estrus synchronization protocol. The number of ovulations and follicle diameter were evaluated by ultrasonography. Dry matter intake (DMI) and nutrient intake, water ingestion, and physiological parameters such as rectal temperature (RT), respiratory rate (RR), heart rate (HR), and serum concentrations of cholesterol and non-esterified fatty acids (NEFA) were also determined. As concern statistical analysis, ANOVA followed by Fisher’s LSD was used and the differences were considered significant when P < 0.05. The control group showed higher HR and RR in the afternoon shift compared to the Oil group. The Oil group showed lower DMI but higher ether extract intake, and serum cholesterol and NEFA concentration than control group. Treatments did not affect estrus response and ovulation rate, but the Licuri oil increased the number of small follicles. The supplementation of Licuri oil improved thermal comfort during the dry season and a marginal increase in the ovarian activity was observed.

Feeding colostrum and transition milk facilitates digestive tract functionality recovery from feed restriction and fasting of dairy calves

The objective of this study was to evaluate the digestive tract recovery and metabolism of feeding either bovine colostrum (BC), transition milk (TM), or milk replacer (MR) after an episode of feed restriction and fasting (FRF) in dairy calves. Thirty-five Holstein male calves (22 ± 4.8 d old) were involved in a 50-d study. After 3 d of feeding 2 L of rehydration solution twice daily and 19 h of fasting (d 1 of study), calves were randomly assigned to one of the 5 feeding treatments (n = 7): calves were offered either pooled BC during 4 (C4) or 10 (C10) days, pooled TM during 4 (TM4) or 10 (TM10) days, or MR for 10 d (CTRL) at the rate of 720 g/d DM content. Then, all calves were fed the same feeding program, gradually decreasing MR from 3 L twice daily to 2 L once daily at 12.5% DM until weaning (d 42), and concentrate feed, water, and straw were offered ad libitum until d 50. Citrulline, Cr-EDTA, β-hydroxybutyrate (BHB), and nonesterified fatty acids (NEFA) in serum and complete blood count (CBC) were determined on d -3, 1, 2, 5, and 11 relative to FRF, except BHB and NEFA at d -3. Volatile fatty acids (VFA), lactoferrin (LTF), IgA, and microbiota (Firmicutes to Bacteroidetes ratio and Fecalis prausnitzii) were analyzed in feces on d 5 and 11 before the morning feeding. Health scores were recorded daily from d -3 to d 14 as well as d 23 and 30. Feed concentrate, MR, and straw intake were recorded daily, and body weight on d -3, 1, 2, 5, and 11 and weekly afterward. Calf performance, intake, serum Cr-EDTA, CBC, fecal LTF concentrations and microbiota parameters were similar among treatments throughout the study. Serum NEFA concentrations were greater in TM4, TM10 and C10 calves compared with the CTRL ones from d 2 to 11, and after the FRF, serum concentrations of BHB were lower in CTRL calves than in the other treatments, and on d 11, serum BHB concentrations in the long treatments (C10 and TM10) remained greater than those in the shorter ones (C4 and TM4) and CTRL. Serum citrulline concentrations were similar on d -3 and 1 in all treatments, but they were greater in C4, C10, TM4, and TM10 on d 2 and 5, and on d 11 they were only greater in C10 and TM10 than in CTRL calves. Fecal IgA concentrations tended to be greater in C10 than in CTRL, TM4, and TM10 calves, and in C4 and TM10 than in CTRL animals. Fecal propionate proportion was lesser in C10 than in CTRL, TM4, and TM10 calves, while butyrate was greater in C4 and C10 than in TM4 and CTRL calves. The proportion of non-normal fecal scores of C10 fed calves was greater than TM4 and TM10 calves. Results showed that TM and BC may help to recover intestinal functionality, provide gut immune protection, and increase liver fatty acid oxidation in calves after a FRF episode.

Inhibition of cluster antigen 36 protects against fatty acid-induced lipid accumulation, oxidative stress, and inflammation in bovine hepatocytes

When ketosis occurs, supraphysiological concentrations of nonesterified fatty acids (NEFA) display lipotoxicity and are closely related to the occurrence of hepatic lipid accumulation, oxidative stress, and inflammation, resulting in hepatic damage and exacerbating the progression of ketosis. However, the mechanism of these lipotoxic effects caused by high concentrations of NEFA in ketosis is still unclear. Cluster antigen 36 (CD36), a fatty acid transporter, plays a vital role in the development of hepatic pathological injury in nonruminants. Thus, the aim of this study was to investigate whether CD36 plays a role in NEFA-induced hepatic lipotoxicity in dairy cows with clinical ketosis. Liver tissue and blood samples were collected from healthy (n = 10) and clinically ketotic (n = 10) cows at 3 to 15 d in milk. In addition, hepatocytes isolated from healthy calves were treated with 0, 0.6, 1.2, or 2.4 mM NEFA for 12 h; or infected with CD36 expressing adenovirus or CD36 silencing small interfering RNA for 48 h and then treated with 1.2 mM NEFA for 12 h. Compared with healthy cows, clinically ketotic cows had greater concentrations of serum NEFA and β-hydroxybutyrate and activities of aspartate aminotransferase and alanine aminotransferase but lower serum glucose. In addition, dairy cows with clinical ketosis displayed excessive hepatic lipid accumulation. More importantly, these alterations were accompanied by an increased abundance of hepatic CD36. In the cell culture model, exogenous NEFA (0, 0.6, 1.2, or 2.4 mM) treatment could dose-dependently increase the abundance of CD36. Meanwhile, NEFA (1.2 mM) increased the content of triacylglycerol, reactive oxygen species and malondialdehyde, and decreased the activities of glutathione peroxidase and superoxide dismutase. Moreover, NEFA upregulated phosphorylation levels of nuclear factor κB (NF-κB) and the inhibitor of NF-κB (IκB) α, along with the upregulation of protein abundance of NLR family pyrin domain containing 3 (NLRP3) and caspase-1, and mRNA abundance of IL1B, IL6, and tumor necrosis factor α (TNFA). These alterations induced by NEFA in bovine hepatocytes were associated with increased lipid accumulation, oxidative stress and inflammation, which could be further aggravated by CD36 overexpression. Conversely, silencing CD36 attenuated these NEFA-induced detriments. Overall, these data suggest that CD36 may be a potential therapeutic target for NEFA-induced hepatic lipid accumulation, oxidative stress, and inflammation in dairy cows.

Plasma Exosomes Aggravate Acute Pancreatitis by Promoting M1 Polarization of Adipose Tissue Macrophages in Obesity-Related Severe Acute Pancreatitis.

Obesity may be a risk factor for severe acute pancreatitis (SAP). However, its precise mechanism is not yet fully understood. We fed rats with a standard laboratory diet (SLD) and a high-fat diet (HFD). SAP model rats were established by retrograde injection of sodium taurocholate. Serum non-esterified fatty acids (NEFAs), lipase (LPS), and myeloperoxidase (MPO) were measured, as were adipose IL-1, IL-6, IL-10, and TNF-α levels. HE staining was performed to determine the severity of pancreatitis. Serum exosomes were extracted from rats with obesity-related SAP, verified by transmission electron microscopy (TEM) and western blot analysis, and co-cultured with THP-1 cells. Flow cytometry was used to analyze the M1 and M2 phenotypes of macrophages in adipose tissues and THP-1 cells. Q-PCR was used to analyze the levels of IL-1, IL-6, IL-10, and TNF-α in each group of cells. The body weight and serum NEFA concentrations of rats in the HFD group were significantly higher than those in the SLD group. Adipose tissue macrophages in the HFD group exhibited a higher percentage of the M1 type than those in the SLD group. The severity of pancreatitis were significantly increased in the HFD + SAP group. Pro-inflammatory macrophages and cytokines were significantly higher in the HFD + SAP group and THP-1 cells co-cultured with serum exosomes extracted from rats with obesity-related SAP. Obesity might worsen the severity of pancreatitis by amplifying the immune response and activating M1 polarization in adipose tissue macrophages via serum exosomes in rats of obesity-related SAP. In our study, we isolated exosomes from the serum of mice with obesity-related SAP. After inducing THP-1 cells to become M0-typed macrophages, we co-cultured the cells with exosomes and observed that exosomes from obesity-related SAP increased the proportion of M1-typed macrophages and promoted the release of pro-inflammatory factors such as IL-1, IL-6, and TNF. Therefore, obesity might worsen the severity of pancreatitis by amplifying the immune response and activating M1 polarization in adipose tissue macrophages via serum exosomes in rats of obesity-related SAP.

Epidemiology of bovine colostrum production in New York Holstein herds: Prepartum nutrition and metabolic indicators

Colostrum yield and quality are influenced by prepartum nutrition and the metabolic status of the cow; however, data considering these associations on multiple dairy farms are limited. Our objective was to identify cow-level prepartum metabolic indicators, as well as farm-level nutritional strategies associated with colostrum yield and the indicator of colostrum quality, Brix %. A convenience sample of 19 New York Holstein dairies (median: 1,325 cows; range: 620 to 4,600 cows) were enrolled in this observational study. Records for individual colostrum yield and Brix % were collected by farm personnel between October 2019 and February 2021. Farms were visited 4 times, approximately 3 mo apart, to obtain feed samples of the prepartum diets, collect blood samples from 24 pre- and postpartum cows, respectively, and determine prepartum body condition score. Feed samples were submitted for analysis of chemical composition, and particle size was determined on-farm using a particle separator. Prepartum serum samples (n = 762) were analyzed for glucose and nonesterified fatty acid concentrations. Whole blood from postpartum cows was analyzed for herd prevalence of hyperketonemia (proportion of samples with β-hydroxybutyrate ≥1.2 mmol/L). A cohort of primiparous (PP; n = 1,337) and multiparous (MPS; n = 3,059) cows calving ± 14 d of each farm visit were included in the statistical analysis. Animals calving in this period were assigned results for the close-up diet composition and herd prevalence of hyperketonemia collected from the respective farm visit. Greatest colostrum yield from PP and MPS cows was associated with moderate starch [18.6-22.5% of dry matter (DM)] and a moderate herd prevalence of hyperketonemia (10.1-15.0%). Greatest colostrum yield from MPS cows was associated with moderate crude protein (13.6-15.5% of DM) and a less severe negative dietary cation-anion difference (DCAD; >-8 mEq/100 g), whereas greatest colostrum yield from PP cows was associated with low crude protein (≤13.5% of DM). In addition, a moderate proportion of the diet with particle length ≥19 mm (15.3-19.1%) was associated with lowest colostrum yield from PP and MPS cows. Highest colostrum Brix % was associated with prepartum dietary factors of low neutral detergent fiber (≤39.0% of DM) and high proportion of the diet with particle length ≥19 mm (>19.1%). In addition, low starch (≤18.5% of DM) and low and moderate DCAD level (≥-15.9 mEq/100 g) were associated with greatest Brix % from PP cows, whereas moderate DCAD (-15.9 to -8.0 mEq/100 g) was associated with greatest Brix % from MPS cows. Prepartum serum nonesterified fatty acid concentration ≥290 µEq/L was associated with increased colostrum yield, but prepartum serum glucose concentration and body condition score were not associated with colostrum yield or Brix %. These data provide nutritional and metabolic variables to consider when troubleshooting colostrum production on farms.

Association of precalving serum NEFA concentrations with postpartum diseases and reproductive performance in multiparous Holstein cows: Cut-off values.

High concentrations of NEFA relative to a defined reference or 'cut-point' values before calving can predict the risk of specific or collective periparturient disease events. A field-based cohort study was conducted to evaluate the value and critical points of serum nonesterified fatty acids (NEFA) at the precalving time to predict the occurrence of postpartum diseases and reproductive performance in dairy cows. Blood samples were taken from 521 high-yielding dairy cows at 1 week (±3days) before calving and NEFA levels were measured. Health and reproduction information of each cow includes dystocia, retained placenta, milk fever, metritis, mastitis, pregnancy in the first insemination and pregnancy in the first two inseminations, and culling in the first 60 days of lactation and milk production. Our results show that there are significant relationships between precalving NEFA with the probability of pregnancy at the first and the first two inseminations after calving. The cows that had NEFA concentrations less than 0.5mmol/L at the last week of pregnancy were 3.51 and 3.15 times more likely to be pregnant at first insemination and the first two inseminations, respectively. Also, our results showed that there are significant relationships between precalving NEFA concentration and the likelihood of dystocia and milk fever. The probability of dystocia and milk fever occurrence were 2.56 and 1.91 times greater in those cows that had NEFA concentrations more than 0.3mmol/L, respectively. The present results indicated that Increasing NEFA during the prepartum period could adversely affect the reproductive efficiency of dairy cows.

Lipid metabolites, interleukin-6 and oxidative stress markers in follicular fluid and their association with serum concentrations in mares.

The application of trans-vaginal ovum pick up (OPU) and intracytoplasmic sperm injection (ICSI) is well established for commercial in vitro embryo production in horses. These assisted reproductive techniques are especially applied during the non-breeding season of the mare. However, little is known about how the health of the oocyte donor may affect the biochemical composition of the follicular fluid (FF) in small and medium-sized follicles routinely aspirated during OPU. This study aimed to investigate associations between systemic and FF concentrations of interleukin-6 (IL-6), total cholesterol, triglycerides, non-esterified fatty acids (NEFA), reactive oxygen metabolites (d-ROMs), biological antioxidant potential (BAP), and oxidative stress index (OSI) during the non-breeding season in mares. At the slaughterhouse, serum and FF of small (5-10mm in diameter), medium (> 10-20mm in diameter), and large (> 20-30mm in diameter) follicles were sampled from 12 healthy mares. There was a strong positive association (P < 0.01) between the concentration of IL-6 in serum and those measured in small (r = 0.846), medium (r = 0.999), and large (r = 0.996) follicles. Serum concentrations of NEFA were positively correlated (P < 0.05) with those measured in small (r = 0.726), medium (r = 0.720), and large (r = 0.974) follicles. Values of total cholesterol and OSI in serum and medium follicles were significantly associated (r = 0.736 and r = 0.696, respectively). The serum concentrations of all lipid metabolites were markedly higher than those measured in FF of small- and medium-sized follicles. Values of IL-6 and OSI did not change significantly between serum and all follicle classes (P ≥ 0.05). To conclude, changes in the blood composition associated with inflammation, oxidative stress, and disturbed lipid metabolism of mares may lead to an inadequate oocyte microenvironment, which could affect oocyte quality and the success rate of OPU/ICSI programs. Further research should indicate whether these changes may ultimately affect in vitro oocyte developmental capacity and subsequent embryo quality.

Eucalcemia during lipopolysaccharide challenge in postpartum dairy cows: I. Clinical, inflammatory, and metabolic response

Hypocalcemia induced by immune activation is a conserved response across mammalian species; however, administration of Ca is discouraged in other species as it is associated with increased morbidity and mortality. Early postpartum cows experience a decrease in circulating Ca concentration following acute inflammation. Corrective Ca therapy during the transition period, particularly in dairy cows experiencing acute disease, is common practice. However, the effect of Ca administration on the inflammatory response during acute immune activation is unknown. Our objective was to compare the clinical, inflammatory, and metabolic response to an intravenous (IV) lipopolysaccharide (LPS) challenge between postpartum cows infused, or not, with IV Ca to maintain eucalcemia. Cows (n = 14, 8 ± 1 d in milk) were enrolled in a matched-pair randomized controlled design to receive IV Ca (IVCa) or sterile 0.9% NaCl (CTRL) during an IV LPS challenge (0.040 or 0.045 µg of LPS/kg of body weight over 1 h). Ionized Ca (iCa) was monitored cow-side, and IV Ca infusion was adjusted in a eucalcemic clamp for 12 h following the start of LPS infusion. Cows were monitored during the 24 h following challenge and serial blood samples were collected to quantify concentrations of glucose, β-hydroxybutyrate, nonesterified fatty acids, urea nitrogen, cytokines, acute-phase proteins, and cortisol. Blood iCa concentration decreased to 0.87 ± 0.03 mM in CTRL during challenge, and by design, iCa concentration was maintained within 3% of baseline in IVCa. Body temperature, heart rate, and respiratory rate were monitored for 24 h following the start of challenge and did not differ between groups. A treatment × time interaction was identified such that serum cortisol concentrations increased in both groups at 2 h but decreased to a greater extent at 6 h in IVCa compared with CTRL. Rumination time (min/h) over the first 12 h following challenge was greater in IVCa, but total rumination time in the 24 h following challenge did not differ from CTRL. Serum glucose and nonesterified fatty acid concentrations decreased, and β-hydroxybutyrate and urea nitrogen concentrations increased over time, but did not differ between groups. Acute leukopenia occurred in both groups at 4 h before leukocytosis was observed at 24 h with total white blood cell counts returning to baseline within 72 h. Plasma concentrations of tumor necrosis factor (TNF) and interleukin-10 (IL-10) increased within 1 h following the start of challenge and did not differ between groups. Serum haptoglobin and serum amyloid A concentrations increased within the 24 h following challenge and were elevated through 72 h but did not differ between groups. Eucalcemia during the acute systemic inflammatory response did not alter the TNF or IL-10 cytokine response, or the acute-phase protein SAA and haptoglobin response in this LPS challenge model; however, eucalcemia was associated with a more rapid decline in cortisol response and greater rumination time in the first 12 h following challenge. We did not find evidence that eucalcemia exacerbated the inflammatory response in early postpartum cows, but Ca administration may alter the clinical response to acute systemic inflammation.

Dietary Macronutrients and Circulating Nonesterified Fatty Acids: A Secondary Analysis of the OMNI Heart Crossover Trial

Nonesterified fatty acids (NEFAs) play key roles in the pathophysiology of diabetes and cardiovascular disease. We sought to determine whether macronutrient content differences affect NEFA concentrations in a randomized crossover trial. Total NEFAs were measured from postintervention specimens of participants in the OMNI Heart trial (Optimal Macronutrient Intake Trial to Prevent Heart Disease). OMNI Heart compared 3 healthful diets to evaluate their effect on systolic blood pressure and serum LDL cholesterol: carbohydrate-rich diet (58% carbohydrate); protein-rich diet (25% protein), about half from plant sources; and a diet rich in unsaturated fatty acids (21% unsaturated fat), predominantly monounsaturated. The trial included 164 participants who consumed the 3 diets, each for 6 wk. Data were analyzed from the 156 participants with unthawed serum available from the week 6 visit for all diet periods. We used ANCOVA and generalized estimating equations (GEEs) to compare serum NEFA concentrations across the 3 diet periods. The mean ± SD age of study participants was 52.9 ± 10.6 y and mean BMI was 30.3 ± 6.1 kg/m2. Fifty-five percent of participants were women and 55% were African American. Comparisons of adjusted mean serum NEFA concentrations after each diet intervention identified no statistically significant differences (58% carbohydrate: 0.144±0.083mEq/L; 25% protein: 0.143±0.076mEq/L; 21% unsaturated fat: 0.143±0.084mEq/L; ANCOVA, P=0.99). Likewise, we observed no significant serum NEFA concentration difference by diet in adjusted GEE models. In adjusted models, serum NEFA concentrations were positively associated, as anticipated, with female sex and higher BMI. In this randomized crossover trial, we observed nearly identical serum NEFA concentrations after each of 3 healthful diets, regardless of macronutrient content.

162 Differential Late Gestation Metabolizable Energy Intake Alters Adipose-Specific Insulin Responsiveness in Antepartum Beef Cattle

Abstract We evaluated how metabolizable energy (ME) intake influences antepartum insulin sensitivity in beef cattle. Angus-Simmental cattle (primiparous: n = 17; multiparous: n = 21) were randomly assigned to treatments supplying 80 (LowME, n = 19) or 120% (HighME, n = 19) of predicted ME requirements for 53 d prepartum. At 7 d antepartum, intravenous glucose tolerance tests (IVGTT) were imposed by infusing 1.36 g glucose/kg BW0.75 and sampling from -3 to 90 min to measure plasma glucose and insulin and serum non-esterified fatty acids (NEFA) concentrations. Data were analyzed using PROC GLIMMIX with the fixed effects of treatment, parity, time (repeated), and treatment×time and considering cow(treatment) as random. Baseline glucose concentration increased (P = 0.03) and baseline insulin concentration tended to increase (P = 0.06) for HighME relative to LowME cattle irrespective (P ≥ 0.33) of parity, whereas baseline NEFA concentration was elevated (P &amp;lt; 0.01) for LowME vs. HighME. Plasma glucose-insulin concentrations during the IVGTT did not differ (P ≥ 0.25) by treatment, treatment×time, or parity. However, serum NEFA was elevated (P &amp;lt; 0.01) throughout the IVGTT for LowME vs. HighME cattle and tended to be affected (P = 0.09) by the treatment×time interaction as NEFA concentrations became similar at 90 min due to HighME cattle having a shallower (treatment×time: P &amp;lt; 0.01) decremental NEFA response than LowME cattle. Low ME cattle cleared NEFA more quickly (75% increase; P &amp;lt; 0.01) than HighME. The glucose positive incremental area-under-the-curve throughout the IVGTT tended to be lesser (P = 0.06) for heifers than cows but heifers had a blunted (P ≤ 0.05) glucose-stimulated reduction in NEFA and NEFA clearance rate compared to cows. These data demonstrate that ME provision may alter adipose insulin responsiveness in antepartum beef cattle and that heifers respond to excess glucose and NEFA differently than cows.

Grape seed extract supplementation in heat-stressed preweaning dairy calves: II. Effects on growth performance, blood metabolites, hormonal responses, and fecal fermentation parameters

The objective of this study was to evaluate the effect of grape seed extract (GSE) supplementation on growth performance, blood metabolites, hormone responses, and fecal volatile fatty acids (VFA) concentration in dairy calves during the summer months. Sixty Holstein female calves 3 d old and 40.6 ± 2.17 kg body weight (BW) were randomly divided into four groups (n = 15). GSE was given orally to the calves with milk daily at doses of 0 (CON), 25 (GSE1), 50 (GSE2), and 100 (GSE3) mg/kg BW. Calves were weighed at 15-day intervals and blood samples were taken on d 3, 33, and 63. Fecal samples were collected on d 33 and 63 of the study to evaluate VFA. Temperature humidity index (THI) values ranged from 73.2 to 87.4 throughout the study, and the study was terminated on d 63. Calves fed GSE2 had the highest (P < 0.01) average daily gain (ADG) between d 3–18 and 33–48 of the study, while the GSE3 group had the highest (P < 0.01) ADG between d 18–33 and 48–63. Moreover, calves fed GSE3 had the highest (P < 0.01) starter mixture intake over the entire study period. Serum non-esterified fatty acids (NEFA) concentrations decreased (P = 0.003) in calves fed GSE2 compared to the other groups. Plasma insulin concentration decreased (P < 0.001) in calves fed GSE3 compared to the other groups, however, increased (P < 0.01) in GSE1 and GSE2 groups until d 33. Plasma cortisol concentration decreased (P < 0.01) in GSE2 and GSE3 groups compared to the CON and GSE1 groups. Also, calves fed GSE2 and GSE3 had lower (P < 0.01) leptin concentration than the others (day 63). Calves fed GSE3 increased (P < 0.01) fecal total VFA, acetate, propionate, and valerate concentrations compared to other groups, whereas the lowest (P < 0.05) concentrations were observed in the GSE1 group. In summary, GSE supplementation caused a beneficial effect on energy metabolism, endocrine system, and colonic fermentation and thus improved growth performance in Holstein calves under heat stress.
