73 Effects of blend of organic acids and monoglycerides on immune responses of weaned pigs experimentally infected with enterotoxigenic Escherichia coli F18

Abstract Our previous research implies that dietary supplementation of organic acids blend, monoglycerides blend, and the combination of both can mitigate diarrhea of weaned pigs infected with Escherichia coli (E. coli) F18. The objective of this study was to investigate the effects of dietary supplementation with a blend of organic acids, a blend of monoglycerides, or the combination of both on immune responses of weaned pigs experimentally infected with E. coli F18. Forty pigs [average body weight (BW) = 7.81 ± 0.84 kg] were individually housed and randomly assigned to one of four treatment groups with 10 replicate pigs per treatment. The four dietary treatments were: () Control: nursery basal diet, 2) Acids: basal diet supplemented with a blend of organic acids (formic acid, lactic acid, and sodium formate) at 0.3%, 3) Monoglycerides: basal diet supplemented with a blend of monoglycerides of fatty acids at 0.3%, or 4) Combination of acids and monoglycerides: basal diet added with the acids blend at 0.2% and the monoglycerides blend at 0.2%. The experiment lasted 28 d, with 7 d before and 21 d after the first inoculation (d 0). All pigs were orally inoculated with E. coli F18 (1010 CFU/3 mL) for 3 consecutive days from d 0. Serum samples were collected on d 0, 2, 5, and 14 post-inoculation (PI) to measure the concentration of C-reactive protein (CRP) and haptoglobin using ELISA. Porcine Cytokine/Chemokine13-Plex Discovery Assay (Eve Technology, Canada) was also conducted with serum samples from d 0, 5, and 14 PI. Data were analyzed by ANOVA using PROC MIXED of SAS with a randomized complete block design. The concentrations of serum CRP and haptoglobin were increased (P < 0.0001) on d 2 PI and then gradually decreased on d 5 and 14 PI. However, no differences were observed in serum CRP and haptoglobin concentrations among all treatment groups throughout the experiment. Pigs fed with monoglycerides tended (P < 0.10) to have decreased serum granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin 1-alpha (IL-1α), and tumor necrosis factor-alpha (TNF-α) concentrations on d 0 PI compared with other treatments. Pigs supplemented with monoglycerides also tended (P < 0.10) to have decreased GM-CSF on d 5 PI and less IL-1α on d 14 PI among all treatments. Supplementation of the combination tended (P < 0.10) to have greater GM-CSF concentration on d 5 PI compared with other treatments. In conclusion, dietary supplementation of monoglycerides blend or the combination of organic acids and monoglycerides alleviates systemic inflammation of weaned pigs during E. coli F18 infection by modulating related cytokines. However, additional research is needed to understand the modulation mechanisms.

PSIV-17 Effects of butyrin-based additive supplementation on nursery pig performance, fecal dry matter, serum cytokines, short chain fatty acid production, and serum chemistry

Abstract Two experiments were conducted to determine the effects of butyrin-based feed additives on nursery pig performance, fecal dry matter (DM), serum chemistry, and short chain fatty acid (SCFA) production. In Exp. 1, 250 pigs (initially body weight = 5.6 kg) were randomly allotted to 1 of 5 treatments. There were 5 pigs/pen and 10 replications/treatment. Treatments consisted of a negative control, a positive control, and three diets containing various butyrin-based feed additives. The negative control was a corn-soybean meal-based diet with no antibiotics or pharmacological content of Zn or Cu. The positive control contained 3,000 and 2,000 mg/kg of Zn from ZnO in phase 1 (d 0 to 10) and 2 (d 10 to 24), respectively, 250 mg/kg of Cu from CuSO4 in phase 3 (d 24 to 38), and 55 mg/kg of carbadox (Mecadox; Phibro Animal Health Corp., Teaneck, NJ) across all phases. The butyrin-based feed additives were added at 0.1% to the negative control diet and consisted of monobutyrin, mono-tributyrin, or tributyrin. In Exp. 2, 1,492 pigs (initially 5.1 kg) were randomly allotted to 1 of 4 treatments. There were 26 or 27 pigs/pen and 14 replications/treatment. Treatments were fed over 3 phases consisting of a control diet and 3 diets containing different mono-tributyrin blends. In phase 1 and 2, the control diet contained 55 mg/kg of carbadox and 3,000 and 2,000 mg/kg of Zn from ZnO, respectively. Each feed additive was added at 0.3% to the control diet in phase 1 and 2, and 0.1% in phase 3. Phases 1 and 2 were fed by feed budget of 2.3 and 5.4 kg/pig, respectively, and phase 3 until the completion of the study. Overall, for Exp. 1, pigs fed the positive control had increased (P < 0.05) average daily gain (ADG) and average daily feed intake (ADFI) compared with all other treatments (Table). Pigs fed monobutyrin had decreased (P < 0.05) gain to feed ratio (G:F) compared with all other treatments. On d 24, pigs fed the positive control and mono-tributyrin had increased (P< 0.05) fecal DM compared with pigs fed monobutyrin and tributyrin with the negative control intermediate. On d 38, pigs fed mono-tributyrin had increased (P < 0.05) serum haptoglobin and cecal butyrate concentration compared with pigs fed the positive and negative control, respectively, with other treatments intermediate. For Exp. 2, there were no differences in growth performance throughout the study. On d 10, pigs fed mono-tributyrin blend 3 had increased (P< 0.05) fecal DM compared with mono-tributyrin blend 2 with other treatments intermediate. On d 24 and 42, pigs fed mono-tributyrin blend 1 had the highest (P ≤ 0.012) IL-12 concentration, without significant mean separation on d 24. In summary, butyrin-based feed additives had limited effects on growth performance. However, supplementing butyrin-based feed additives led to alterations in fecal consistency, SCFA production, and serum chemistry.

The effects of a nonsteroidal anti-inflammatory drug on the behavioural and physiological parameters of beef cows and calves assisted at calving

Non-steroidal anti-inflammatory drugs (NSAIDs) may be administered to the cow or the calf as a pain mitigation strategy to combat the negative impacts of injury or trauma associated with a difficult calving. However, the effects of NSAIDs on beef cows’ and calves’ behaviour related to pain and cow-calf bonding after an assisted calving have not been studied. Therefore, the objective of this study was to investigate the effects of meloxicam administered to beef cows and calves after an assisted calving by measuring differences in behaviours associated with pain and cow-calf bonding, and physiological parameters associated with pain. Twenty-three cow-calf pairs assisted at calving were randomly allocated to two medication groups, meloxicam (Metacam®, 20 mg/ml, 0.5 mg/kg body weight, Boehringer Ingelheim, Ingelheim, Germany) or placebo (0.025 ml/kg). Cow blood samples for serum haptoglobin concentration and thermal images of the vulva were taken within 10 minutes after calving and 24±1 hours after calving. Calf blood samples for L‐lactate concentration and thermal images of the head were taken at birth and images repeated at 24±1 hours after birth. Blood samples for serum IgG concentration were taken at 24±1 hours after birth. After at-birth sample collection and treatment administration, the cow-calf pairs were placed in separate box stalls, and behaviours were video recorded for 24±1 hours. A human approach test and a separation test were performed after the 24±1 hour period. Descriptive statistics, multivariable linear and logistic regression models, and non-parametric tests were used to analyze the data. There were no significant differences between meloxicam and placebo groups for serum IgG concentration, haptoglobin concentration, or changes in temperature in the cows’ vulva or calves’ head. Additionally, there were no significant differences between meloxicam and placebo groups for the cows’ behaviours (i.e., time spent lying in sternal recumbency, standing, feeding, self-grooming, grooming the calf, and frequency of tail flicking, tail raising, and pressing against the box stall). Calves treated with meloxicam were more active (P=0.04) and played more (P=0.01) than placebo calves. No other behaviours (i.e., time spent lying in sternal recumbency, attempting to stand, standing, walking, being in a nursing position, investigating, and frequency of approaching the cow) were significantly different between calves treated with meloxicam or placebo. No significant difference (P=0.83) in bonding behaviours was identified for cows and calves treated with meloxicam or a placebo. The anti-inflammatory effects of meloxicam in calves after an assisted calving may improve calf welfare.

Association of clinical respiratory disease signs and lower respiratory tract bacterial pathogens with systemic inflammatory response in pre-weaned dairy calves

This study investigated the potential associations between 3 acute phase proteins (APPs) haptoglobin, serum amyloid A, and fibrinogen, clinical signs of respiratory disease, and the presence of bacterial pathogens in the lower respiratory tract (LRT) of pre-weaned dairy calves. This cross-sectional study included 150 pre-weaned calves (2-86 d old) from 15 large dairy herds in Estonia. Tracheobronchial lavage, blood, and fecal samples were collected from 5 calves showing clinical signs indicative of LRT disease, and samples from 5 calves without clinical signs of LRT disease per herd. All samples collected from these calves were analyzed for concentrations of systemic APPs, LRT bacteria, and intestinal pathogens. Heifer blood and bulk tank milk samples were collected for the detection of disease-specific antibodies against bovine herpesvirus 1, bovine viral diarrhea virus, bovine respiratory syncytial virus, and Mycoplasma bovis. Mixed-effects linear regression models were used to analyze the associations of clinical respiratory disease signs and LRT bacteria with APPs. Increased plasma fibrinogen concentrations in calves were associated with higher rectal temperature (>39.5°C), increased respiratory rate (>50 breaths/min), and coughing. Increased serum amyloid A concentrations were associated with higher rectal temperature (>39.5°C) and respiratory rate between 40 and 50 breaths/min. Calves with the presence of fecal Cryptosporidium spp. and rectal temperature of 39°C and above had increased serum haptoglobin concentrations. Increased fibrinogen concentrations were associated with the presence of Pasteurella multocida in the calf LRT, whereas increased concentrations of fibrinogen and serum amyloid A were associated with the presence of Trueperella pyogenes. In conclusion, APPs showed variable associations with clinical signs of respiratory disease and LRT bacteria. Plasma fibrinogen concentration could be used as a complementary calf-side test to assess systemic inflammation caused by LRT bacteria such as P. multocida and T. pyogenes in pre-weaned dairy calves.

Comparison of behavioral and physiological changes in postpartum healthy and inflammatory diseased cows.

During parturition, cows often experience intense pain and stress, which increases the risk of inflammatory diseases. This study aimed to compare the postpartum health status between healthy cows and those diagnoses with inflammatory diseases by examining behavioral and heart rate (HR) variability (HRV) changes, to provide information before the onset of disease. Eight Holstein cows were used in this study. HR, parameters of HRV (low-frequency power: LF; high-frequency power: HF; LF/HF ratio, and total power) and time budget of individual maintenance behaviors (standing, recumbency, feeding, rumination while standing and lying, and sleep) were continuously recorded from 0 to 168 h postpartum. Milk and blood samples were collected daily. Cows were categorized as diseases based on the positive result of California mastitis test and/or serum haptoglobin concentration that exceeded 50 μg/ml after all blood samples have been collected. Compared to healthy individuals (n = 3), diseased cows (n = 5) exhibited higher HR, LF/HF, and lower total power (p < 0.05), suggesting the dominance of the sympathetic nervous system in cows with inflammatory diseases. Additionally, diseased cows showed an increased standing time budget and reduced recumbency (p < 0.05), which may be a behavioral strategy in response to discomfort from inflammation.

Evaluation of haptoglobin concentration in clinically healthy dairy cows: correlation between serum and milk levels

ABSTRACT The study aimed to analyse serum haptoglobin concentrations by calving number and lactation phase in clinically healthy dairy cattle and to determine the correlation between concentrations in serum and milk samples. For this purpose, serum samples were collected from 228 clinically healthy cows after milking from 14 randomly selected herds. Additionally, milk samples were collected from 80 out of the 228 cows from the milking prior to blood collection. Both serum and milk samples were analysed with a commercial ELISA test. The effect of the calving number and lactation phase on mean serum haptoglobin concentrations was examined by Kruskal–Wallis/Jonckheere–Terpstra tests. After that, in a multivariate approach, a mixed-effects linear model was also performed. In addition, intraclass correlation coefficients were used to evaluate the correlation between concentrations in serum and milk samples. Results indicated a downward trend in haptoglobin concentrations throughout lactation, that was statistically significant when comparing fresh cows with the remaining lactation phases. The higher haptoglobin concentrations in fresh cows could be the result of inflammatory illnesses without clinical disease, stress or variations within the physiological state. Although previous studies showed some discrepancies, a good correlation between serum and milk concentrations has been observed in the present paper.

Supplemental trace minerals as complexed or inorganic sources for beef cattle during the receiving period.

To investigate effects of inorganic or complexed trace mineral source (zinc, copper, manganese, and cobalt) on receiving period performance and morbidity, crossbred beef heifer calves (n = 287) arriving on three delivery dates were used in a 42-d receiving trial. Heifers were processed after arrival, stratified by day -1 body weights (BW) and allocated randomly to eight pens (11 to 13 heifers/pen, 24 pens total). Within truckload, pens were assigned randomly to dietary treatment (n = 12 pens/treatment). Heifers were housed on 0.42-ha grass paddocks, provided ad libitum bermudagrass hay and provided dietary treatments in grain supplements fed daily. Treatments consisted of supplemental zinc (360mg/d), copper (125mg/d), manganese (200mg/d), and cobalt (12mg/d) from complexed (Zinpro Availa 4, Zinpro Corp. Eden Prairie, MN) or inorganic sources (sulfates). Heifers were observed daily for clinical bovine respiratory disease (BRD). If presenting BRD symptoms and rectal temperature ≥ 40 °C, heifers were deemed morbid and treated with antibiotics. Six heifers/pen were bled to determine serum haptoglobin concentrations on days 0, 14, and 28. Liver biopsies were taken on day 5 ± 2 and 43 ± 1 from three calves selected randomly from each pen for mineral status comparisons. Statistical analyses were performed using the MIXED, GLIMMIX, and repeated measures procedures of SAS 9.4 with truckload as a random effect and pen within truckload specified as subject. There tended to be a treatment by day interaction for BW (P = 0.07). Heifer BW did not differ on day 0 (P = 0.82) and day 14 (P = 0.36), but heifers fed complexed trace minerals had greater BW on day 28 (P = 0.04) and day 42 (P = 0.05). Overall average daily gains were greater for heifers fed complexed trace minerals (P = 0.05; 0.78 vs. 0.70kg, SE = 0.03). Heifers supplemented with inorganic trace minerals had greater BRD incidence (P = 0.03; 58 vs. 46%, SE = 3.6). Haptoglobin concentrations decreased throughout the trial (P < 0.001), and heifers fed complexed trace minerals tended to have a decrease in haptoglobin concentrations (P = 0.07). The source of trace mineral supplementation had no effect (P ≥ 0.20) on liver mineral concentrations and there were no treatment × day interactions (P ≥ 0.35). In conclusion, supplementing diets for the first 42 d after arrival with complexed trace mineral sources improved heifer performance as compared to heifers supplemented with inorganic trace minerals.

Monitoring ventral tail base surface temperature for fever detection in calves.

In this study, we investigated whether monitoring the ventral tail base surface temperature (ST) using a wearable wireless sensor could be effective for fever detection in calves with experimentally induced pneumonia after inoculation with Histophilus somni strain 2336. We found a significant difference in the changes in ST values between the control and H. somni-inoculated groups after 24 h of inoculation and detected fever; however, the rectal temperature showed a significant difference between the groups after 12 h of inoculation. When a significant difference in the ST between the two groups was observed, serum haptoglobin concentration and exacerbation of clinical score increased in the H. somni-inoculated group compared with those in the control group. Pneumonia was observed in the H. somni-inoculated group at necropsy, indicating that the changes in ST may reflect fever with inflammation caused by H. somni infection. Our results demonstrated that monitoring ST using a sensor attached to the ventral tail base can detect fever in calves and may be a useful and labor-saving tool for the health management of calves.

ABSTRACT: This study evaluated the effect of black wattle (Acacia mearnsii) condensed tannin extract in simple and complex diets for weaned piglets on performance, faecal consistency and serum haptoglobin concentrations. Eighty-eight female and castrated male piglets were used, weaned at 28 ± 3 days old, with an initial body weight of 8.02 ± 1.21 kg, housed in same-sex pairs and distributed in four treatments in a 2 x 2 factorial experiment in a randomised complete block design. The experimental treatments were simple diet (SD), simple diet + tannin (SD+T), complex diet (CD) and complex diet + tannin (CD+T). The simple diets contained lower concentrations of dairy ingredients, inclusion of barley and no addition of zinc oxide, palatant, acidifier and yeast. The diets with tannin were supplemented with 1,850 mg kg-1 of black wattle condensed tannin extract. The feed programme consisted of three diets: Pre-starter I (0-7 days), Pre-starter II (8-21 days) and Starter (22-28 days). Feed and water were available ad libitum. Body weight and feed intake were measured weekly to calculate the average daily feed intake, daily weight gain and feed conversion ratio. The faecal consistency score was measured daily to calculate the diarrhoea occurrence percentage. Blood samples for serum haptoglobin concentration were collected at 7 and 14 days after weaning. The addition of condensed tannin had no effect (P &gt; 0.05) on evaluated performance variables for weaned piglets. The diet type also did not affect the piglets’ performance (P &gt; 0.05). The inclusion of black wattle (Acacia mearnsii) condensed tannin does not affect piglet performance, reduces the incidence of diarrhoea in the first week after weaning and decreases the haptoglobin inflammatory response.

Characterizing pregnancy losses in Bos indicus beef females receiving a fixed-timed artificial insemination protocol

This experiment evaluated pregnancy losses from day 30 of gestation to calving in Bos indicus females, and evaluated if serum haptoglobin concentrations during early gestation impacts subsequent pregnancy losses. A total of 4926 Nelore females were used, being 1802 nulliparous heifers inseminated as yearlings (precocious heifers), 1356 nulliparous heifers inseminated at 24 months of age (conventional heifers), 887 suckling primiparous cows, and 881 suckling multiparous cows. Cows were assigned to an ovulation synchronization + fixed-time artificial insemination (FTAI) protocol from day −11 to 0 of the experiment. Pregnancy status was verified using transrectal ultrasonography on days 30 and 60 after FTAI, via transrectal palpation on day 150 after FTAI, and according to calf birth. Blood samples were collected from all animals diagnosed pregnant on day 30 after FTAI, and analyzed for serum concentrations of haptoglobin and pregnancy associated glycoproteins (PAG). Pregnancy loss was greater (P < 0.01) from day 60–150 of gestation (10.2 %) compared with day 30–60 of gestation (6.0 %) and with day 150 of gestation to calving (7.4 %), and differed (P = 0.04) between these latter two periods. Pregnancy loss from day 30–60 of gestation did not differ (P = 0.26) among parities, whereas total pregnancy losses (day 30 to calving) were greater (P < 0.01) in precocious (28.4 %) and conventional (27.1 %) heifers compared with primiparous (16.4 %) and multiparous (13.0 %) cows. Serum PAG concentrations on day 30 after FTAI were less (P ≤ 0.03) in cows that lost the pregnancy (5.63 ng/mL) from day 30–60 of gestation, as well as those that lost the pregnancy from day 30 to calving (8.59 ng/mL) compared with cohorts that maintained the pregnancy (9.39 and 9.32 ng/mL, respectively). No differences in serum PAG concentrations on day 30 after FTAI were noted (P ≥ 0.23) according to pregnancy losses from day 60 to calving. Serum haptoglobin concentration on day 30 after FTAI also did not differ (P ≥ 0.48) between cows that maintained or lost the pregnancy. This experiment provides novel information about pregnancy losses after day 30 of gestation in B. indicus cattle, with most losses occurring as fetal mortality and not affected by systemic inflammation during early gestation. Pregnancy losses were nearly doubled in precocious and conventional heifers compared with parous cows, demonstrating the relevance of this reproductive failure to B. indicus replacement heifers.

255 Effects of Dietary Valerate Glycerides on Serum Inflammatory Markers, Serum Metabolomic Profile, and Intestinal Mucosa Gene Expression in Weanling Pigs Infected with a Pathogenic Escherichia Coli

Abstract Our previous work showed that dietary valerate glycerides reduced frequency of diarrhea and β-hemolytic coliforms in fecal cultures of pigs infected with F18+ Enterotoxigenic Escherichia coli (ETEC). However, the impact of these feed additives on the immune response and intestinal physiology of newly weaned pigs experiencing ETEC-associated diarrhea has not been evaluated. The present experiment aimed to determine the effects of dietary valerate glycerides on intestinal mucosa gene expression, systemic inflammatory status, and serum metabolomic profile of weanling piglets infected with F18+ ETEC. Sixty weaned piglets (6.97 ± 0.75 kg body weight; 21 days old) were randomly assigned to one of four dietary treatments: nursery basal diet (control), 0.075% or 0.1% monovalerin, or 0.1% trivalerin added to control. After a 7-day adaptation, all piglets were orally inoculated with F18+ ETEC (1010 CFU/3 mL) on d 0, d 1, and d 2 post-inoculation (PI). Serum samples were collected on d 0 prior to ETEC inoculation, d 3, d 6, and d 21 PI to analyze concentration of tumor necrosis factor-α (TNF-α), C-reactive protein (CRP), and haptoglobin. Serum samples from d 6 PI were submitted for untargeted metabolomics analysis at the West Coast Metabolomics Center. Six replicate pigs per treatment group were euthanized on d 7 PI to collect jejunal and ileal mucosa for gene expression analysis. Data were analyzed in RStudio by ANOVA in a linear mixed model with treatment as fixed effect and pig as random effect. Metabolomics analysis was performed using the online platform, MetaboAnalyst. Pigs fed trivalerin had reduced (P &amp;lt; 0.05) serum TNF-α on d 0 and d 6 PI compared with pigs in control. Whereas pigs supplemented with 0.1% monovalerin tended (P ≤ 0.10) to have decreased serum TNF-α on d 0 and d 3 PI, and less (P &amp;lt; 0.05) serum CRP on d 3 PI compared with pigs in control. No differences in serum concentrations of haptoglobin were detected among dietary treatments. Preliminary results for gene expression analysis indicate a tendency (P = 0.078) for increased mRNA expression of tight junction protein, ZO-1 in pigs fed 0.1% trivalerin compared with control. Among all valerate derivative treatments, significant fold changes [FC &amp;gt; 2, log2(FC) &amp;gt; 1] for individual metabolites were detected, where levels of adenosine, putrescine, and shikimic acid were decreased compared with the control group. No differential metabolomic profiles in serum were identified on d 6 PI as indicated by a lack of cluster separation using principal component analysis (PCA). In conclusion, valerate glycerides could regulate gene expression of the intestinal epithelium and systemic immune response of weanling piglets infected with F18+ ETEC.

221 Increasing Functional Amino Acid Ratios Does Not Improve Growth Performance in Pigs Raised Under Poor Sanitary Conditions When Fed with Individual Precision Feeding Or Conventional Feeding Systems

Abstract Immune system activation redirects dietary nutrients to defense mechanisms, thereby impairing growth performance of pigs. Supplementing pigs with functional amino acids (AA) such as Met, Thr and Trp was shown to support the immune system and reduce the negative effects on growth. However, pigs respond differently to immune stressors which increase herd variability. In this context, individual precision feeding (IPF) might be an alternative approach to reduce herd variability and increase nutrient efficiency in immune challenged pigs. This study aimed to evaluate growth performance, body composition, nitrogen (N) efficiency and haptoglobin and triglycerides serum concentration in growing pigs raised in poor sanitary conditions (PoorSC). The PoorSC was characterized by oral inoculation with 2 × 109 cfu of Salmonella typhimurium and manure spreading on the pen floor. Sixty gilts (23 ± 2.8 kg BW) were distributed in BW blocks according to a 2 dietary (D) × 2 feeding system (FS) factorial arrangement composed of to 2 diets [control (CN): INRA ideal AA ratio or supplemented (AA+) 120% of the ideal ratio for Met, Thr and Trp] and 2 feeding systems [conventional (GP) or IPF]. GP pigs were fed with a unique diet that matched the SID Lys requirement of the 80th-percentile pig at the beginning of the trial whereas the IPF pigs received a diet tailored daily to each pig requirement. All pigs were housed in the same pen and fed ad libitum during 28 experimental days with automatic feeding stations able to provide to each pig the assigned diet. Body composition was obtained by dual X-ray absorptiometry at days 0 and 28 and blood samples were collected at d 28. Data were analyzed with D and FS and their interaction as main fixed factors and body weight (BW) blocks as a random effect (Table 1). Haptoglobin serum concentration increased from d 0 to 28 regardless of FS and D (P &amp;lt; 0.10). IPF pigs fed with CN diet had the lightest BW at the end of the trial (P ≤ 0.10; FS × D), whereas BW was similar for the other treatments. However, across diets, IPF pigs had final BW, and protein, ADG, G:F, protein and lipid deposition, N intake and excretion and circulating triglycerides that were less than GP pigs. N utilization efficiency was similar across treatments. Additionally, increasing the provision of the studied functional AA did not improve pig performance. Thus, showing no benefits in increasing AA ratios for growth and immune response. The increased triglycerides concentration and low protein and lipid deposition suggests that IPF pigs prioritized the immune system to the expense of protein and lipid accretion. In conclusion, AA supplementation and IPF do not improve growth performance and N efficiency of pigs raised in poor sanitary conditions.

Peripartum factors associated with variation in voluntary postpartum hay intake in dairy cows

The objective of this research was to assess variation in postpartum hay intake when offered alongside total mixed ration (TMR) as free choice, and identify factors related to the hay intake. Twenty multiparous cows were fed a closeup TMR (21.5% starch, 39.1% neutral detergent fiber [NDF] on a dry matter [DM] basis). After calving, cows were offered free choice timothy hay (61.6% NDF, 9.6% crude protein) in addition to a fresh cow TMR (26.8% starch, 33.0% NDF) for the first 5 d postpartum. Cows were fed individually with separate mangers for TMR and hay, each offered ad libitum. Prepartum DM intake (DMI) was recorded, and baseline blood samples were collected after calving, but before the first postpartum feeding. Free choice hay intake ranged from 0 to 4.7 kg/d (DM basis) or 0 to 55.2% (% of total DMI). Cows that consumed more hay (% of total DMI) from d 1 through 5 postpartum had lower DMI 2 d before calving (r = −0.63), and greater baseline concentrations of plasma β-hydroxybutyrate (r = 0.60) and serum haptoglobin (r = 0.68). Additionally, hay intake (% of total DMI) from d 1 through 5 postpartum tended to be positively related to baseline plasma fatty acid concentration (r = 0.41). These findings suggest that cows with lower intake before calving and cows with greater ketone production and inflammation at calving may consume more hay, when offered separate from TMR.

Effects of offering free-choice hay for the first 5 days postpartum on productivity, serum inflammatory markers, gut permeability, and colon gene expression in fresh dairy cows

The objective of the present study was to evaluate the effects of offering free-choice hay to cows during the first 5 d immediately after calving on feed intake, milk yield, plasma metabolites, serum inflammatory markers, rumination, gut permeability, and colon gene expression. It was hypothesized that cows offered free-choice hay would have lower gut permeability, lower inflammation, and higher milk production, compared with cows not offered hay. Thirty-two multiparous cows were fed a closeup total mixed ration (TMR; 21.5% starch, 32.1% forage neutral detergent fiber [NDF] on a dry matter basis) until calving. In the postpartum period, all cows were fed a fresh cow TMR (26.8% starch and 23.4% forage NDF) from calving until 21 DIM, and were assigned randomly to receive 1 of 2 treatments as follows: (1) free-choice timothy hay (61.6% NDF; 9.6% crude protein), offered outside of the TMR in a separate manger, for the first 5 d postpartum (FCH; n = 20), or 2) no free-choice hay (NH; n = 12). The FCH cows tended to have lower serum haptoglobin concentration on d 3, compared with NH (0.95 vs. 1.52 mg/mL). Within the FCH group, cows with greater hay intake had a smaller increase in serum amyloid A from d 1 to 3 after calving (r = 0.37), and tended to have a smaller increase in serum haptoglobin concentration (r = 0.36). Cows in the FCH group had a lower ratio of starch intake (kg) to forage NDF intake (kg) on d 1 and 2, compared with NH (0.91 vs. 1.14 ± 0.03), and cows that had a lower starch:forage NDF ratio tended to have a smaller increase in serum haptoglobin concentration from d 1 to 3 after calving (r = 0.32). Cows in the FCH group had lower TMR dry matter intake (DMI; 15.0 vs. 17.1 ± 0.93 kg/d) and lower total DMI (TMR + hay DMI; 15.9 vs. 17.1 ± 0.87 kg/d), from d 1 to 5 when free-choice hay was offered, compared with NH. However, the hay treatment did not affect plasma energy metabolite concentration, gut permeability, colon gene expression, milk yield, rumination time, or change in body weight or body condition score. Overall, these findings suggest that offering free-choice hay for the first 5 d after calving may reduce serum inflammatory marker concentration, but milk yield may not increase, due to lower intake.

Effects of a phytogenic feed additive on weaned dairy heifer calves subjected to a diurnal heat stress bout

The study objective was to evaluate the effects of a phytogenic feed additive (PFA) on dry matter intake (DMI), average daily gain (ADG), inflammation, and oxidative stress markers of heifer calves exposed to a heat stress bout in the summer. A total of18 Holstein and 4 Jersey heifer calves (192 ± 5 kg of body weight at 162 ± 16 d of age) housed in indoor stalls were assigned to 1 of 2 dietary treatments (n = 11; 9 Holstein and 2 Jersey): (1) a basal total mixed ration (CTL), and (2) CTL top-dressed with 0.25 g/d of PFA. Following 7 d of acclimation, baseline measurements were made over 7 d under regular summer conditions [average temperature-humidity index (THI) = 79 from 0900 to 2000 h, and 75 from 2000 to 0900 h]. Calves were then subjected to a 7-d cyclic heat stress bout (HS) by turning on barn heaters and increasing the barn temperature to 33.0°C only during the daytime (the average THI = 85 from 0900 to 2000 h). The study continued for an extra 4-d period after HS ended (post-HS). The HS increased rectal temperature, skin temperature, and respiration rate from the baseline by 1.0°C, 4.0°C, and 49 breaths/min, respectively. The drinking water intake increased by 32% in response to HS, and calves continued to consume more water (44%) than the baseline consumption even after HS ended. The treatment × time interactions were not significant for feed intake, ADG, partial pressure of O2 in the blood, and blood concentrations of inflammation markers such as haptoglobin and lipopolysaccharide binding protein (LBP), and antioxidant markers such as protein carbonyl and thiobarbituric acid (TBARS). The PFA tended to increase daytime DMI (0.24 kg/d) compared with CTL throughout the experiment but did not affect ADG, which decreased from 1.12 kg/d to 0.26 kg/d in response to HS. Both DMI (13%) and ADG (85%) increased during post-HS relative to baseline, indicating compensatory performances that were not affected by the PFA. Serum haptoglobin and plasma LBP concentrations of PFA calves were 44% and 38% lower than that of CTL calves across all time points. The PFA decreased O2 pressure and tended to decrease protein carbonyl concentration in the blood across all time points. The PFA tended to decrease TBARS concentration on the first day of HS and increase and decrease the ratio of reduced to oxidized glutathione in the blood during the baseline and post-HS periods, respectively. Despite the lack of growth improvements, feeding PFA seems to increase O2 levels in the blood and alleviate oxidative stress and inflammation of heifer calve exposed to diurnal heat waves (∼7 d) in the summer.

A proposed model to evaluate how changes in body condition score and the fatty acid profile of a supplement affect physiology and metabolic responses of nonlactating females

Two experiments were designed to evaluate the effects of altering body condition score (BCS) and the profile of a fatty acid (FA) supplement on the metabolism of Bos indicus Nellore females. In experiment 1, 16 and 24 B. indicus heifers and nonlactating cows, respectively, were assigned to (1) maintenance diet (MNT-MNT; n = 10), (2) maintenance diet and BCS loss (MNT-LSS; n = 10), (3) maintenance diet supplemented with calcium salts of soybean oil for 30 d and BCS loss for 40 d (MNT+CFA-LSS; n = 10), and (4) maintenance diet for 30 d and BCS loss for 40 d with a diet containing calcium salts of soybean oil (MNT-LSS+CFA; n = 10). Following the BCS loss period, MNT-LSS, MNT+CFA-LSS, and MNT-LSS+CFA were fed a diet to promote the gain of BCS. In experiment 2, 40 Bos indicus nulliparous heifers were assigned to (1) maintenance diet (MNT-MNT; n = 10), (2) BCS loss followed by a BCS gain (LSS-REM; n = 10), (3) BCS loss followed by a BCS gain diet with CFA of palm oil (LSS-REM+PLM; n = 10), and (4) BCS loss followed by a BCS gain diet with CFA of soybean oil (LSS-REM+SOY; n = 10). Blood samples were obtained for serum haptoglobin and fecal samples for pH (experiment 2 only). In experiment 1, a treatment × day interaction was observed for BCS during the 60-d BCS loss and gain period. Animals assigned to MNT-MNT had a greater BCS than the other treatment groups on d 40 and 60 of the experiment, but no other differences were observed. Moreover, a treatment × day interaction was observed for serum haptoglobin, as on d 60, MNT-LSS had a greater mean serum haptoglobin concentration. In experiment 2, a treatment × day interaction was also observed for BCS. From d −4 to 0, LSS-REM and LSS-REM+SOY had a reduced BCS versus MNT-MNT, but also lower for LSS-REM versus MNT-MNT on d 1, and LSS-REM+PLM versus MNT-MNT on d −1 and 0. For serum haptoglobin, no treatment or treatment × day interaction was observed. A treatment × day interaction was observed for fecal pH. From d −10 to 0, MNT often had a lower fecal pH, but during realimentation, LSS-REM heifers had a reduced fecal pH on d 1, 4, and 10. In summary, we failed to demonstrate an increase in serum haptoglobin due to a BCS loss. Still, supplementation with calcium salts of FA alleviated the increase in haptoglobin and maintained fecal pH at more stable values during realimentation, regardless of the FA profile of the supplement.

Local and systemic inflammatory response to the intrauterine infusion of enzymes during estrus in water buffaloes with subclinical endometritis

Our objective was to determine the effects of intrauterine infusion of proteolytic enzymes in buffaloes with subclinical endometritis (SCE) at estrus on the resolution of endometrial inflammation and reproductive performance. Buffaloes at spontaneous estrus (E1) were screened for SCE by endometrial cytology to identify SCE (≥5% PMN, n = 22) and non-SCE (<5% PMNs, n = 14) animals. All buffaloes underwent uterine ultrasonographic examination, low volume uterine lavage (cytokines and acute phase proteins) and blood sampling (cytokines and acute-phase proteins) at E1. On the same day (E1), SCE buffaloes were randomly selected either for intrauterine infusion of proteolytic enzymes (ENY, n = 11) or saline (PC, n = 11). Buffaloes without SCE were kept as untreated control (NC; n = 14). All buffaloes were re-examined and re-sampled during subsequent estrus (E2), inseminated during the following estrus (E3), and assessed for fertility related outcomes. Proteolytic infusion resulted a reduction in uterine PMN (P < 0.01) in SCE buffaloes. The concentrations of interleukin (IL)-1β and tumor necrosis factor (TNF)-α in uterus, and TNF-α and IL-10 in serum were higher (P < 0.01) at E1 in buffaloes with SCE (PC and ENY) compared to NC. After treatment, uterine IL-1β and TNF-α (P = 0.02), and serum TNF-α and IL-10 were lower within the animals of ENY group (P < 0.01). Before treatment, buffaloes with SCE had higher concentrations (P < 0.01) of serum and uterine amyloid-A and haptoglobin, which decreased (P < 0.01) after treatment in the ENY group. None of the fertility outcomes differ between the treatment groups. In conclusion, intrauterine infusion of proteolytic enzymes reduced endometrial inflammation; however, did not improve reproductive outcomes.

Effect of long-term carnosine/anserine supplementation on iron regulation after a prolonged running session.

Exercise-induced hemolysis, which is caused by metabolic and/or mechanical stress during exercise, is considered a potential factor for upregulating hepcidin. Intramuscular carnosine has multiple effects including antioxidant activity. Therefore, this study aimed to determine whether long-term carnosine/anserine supplementation modulates exercise-induced hemolysis and subsequent hepcidin elevation. Seventeen healthy male participants were allocated to two different groups: participants consuming 1,500 mg/day of carnosine/anserine supplements (n = 9, C+A group) and participants consuming placebo powder supplements (n = 8, PLA group). The participants consumed carnosine/anserine or placebo supplements daily for 30.7 ± 0.4 days. They performed an 80-running session at 70% VO2peak pre-and post-supplementation. Iron regulation and inflammation in response to exercise were evaluated. Serum iron concentrations significantly increased after exercise (p &lt; 0.01) and serum haptoglobin concentrations decreased after exercise in both groups (p &lt; 0.01). No significant differences in these variables were observed between pre-and post-supplementation. Serum hepcidin concentration significantly increased 180 min after exercise in both groups (p &lt; 0.01). The integrated area under the curve of hepcidin significantly decreased after supplementation (p = 0.011) but did not vary between the C+A and PLA groups. Long-term carnosine/anserine supplementation does not affect iron metabolism after a single endurance exercise session.

Yeast hydrolysate attenuates lipopolysaccharide-induced inflammatory responses and intestinal barrier damage in weaned piglets

BackgroundIntestinal inflammation is the main risk factor causing intestinal barrier dysfunction and lipopolysaccharide (LPS) can trigger inflammatory responses in various eukaryotic species. Yeast hydrolysate (YH) possesses multi-biological effects and is received remarkable attention as a functional ingredient for improving growth performance and promoting health in animals. However, there is still inconclusive on the protective effects of dietary YH supplementation on intestinal barrier of piglets. This study was conducted to investigate the attenuate effects of YH supplementation on inflammatory responses and intestinal barrier injury in piglets challenged with LPS.MethodsTwenty-four piglets (with an average body weight of 7.42 ± 0.34 kg) weaned at 21 days of age were randomly assigned to one of two dietary treatments (12 replications with one pig per pen): a basal diet or a basal diet containing YH (5 g/kg). On the 22nd d, 6 piglets in each treatment were intraperitoneally injected with LPS at 150 μg/kg BW, and the others were injected with the same amount of sterile normal saline. Four hours later, blood samples of each piglet were collected and then piglets were euthanized.ResultsDietary YH supplementation increased average daily feed intake and average daily gain (P < 0.01), decreased the ratio of feed intake to gain of piglets (P = 0.048). Lipopolysaccharide (LPS) injection induced systemic inflammatory response, evidenced by the increase of serum concentrations of haptoglobin (HP), adrenocorticotropic hormone (ACTH), cortisol, and interleukin-1β (IL-1β). Furthermore, LPS challenge resulted in inflammatory intestinal damage, by up-regulation of the protein or mRNA abundances of tumor necrosis factor-α (TNF-α), IL-1β, toll-like receptors 4 (TLR4) and phosphor-nuclear factor-κB-p65 (p-NFκB-p65) (P < 0.01), and down-regulation of the jejunal villus height, the protein and mRNA abundances of zonula occludens-1 (ZO-1) and occludin (OCC; P < 0.05) in jejunal mucosa. Dietary YH supplementation decreased the impaired effects of ACTH, cortisol, HP, IL-1β and diamine oxidase in serum (P < 0.05). Moreover, YH supplementation also up-regulated the jejunal villus height, protein and mRNA abundances of ZO-1 and OCC (P < 0.05), down-regulated the mRNA expressions of TNF-α and IL-1β and the protein abundances of TNF-α, IL-1β, TLR4 and p-NFκB-p65 in jejunal mucosa in LPS-challenged pigs (P < 0.01).ConclusionYeast hydrolysate could attenuate inflammatory response and intestinal barrier injury in weaned piglets challenged with LPS, which was associated with the inhibition of TLR4/NF-κB signaling pathway activation.Graphical
