In this study, the protocol for enrichment and separation of flavonoids from <em>Cryptotaenia japonica</em> Hassk. was developed using macroporous resins. After comparison of six kinds of macroporous resins for their adsorption and desorption properties of total flavonoids, LS-46D was found to be the most powerful resin for total flavonoids isolation from the plant. Under optimal conditions, after one round enrichment with LS-46D, the total flavonoids content in the product increased by 3.5 times, with a recovery yield of 94.84%. The contents of the main active components such as luteolin and pigenin increased 8.35 and 6.29 times, respectively. These results demonstrated that the method has great potential for production of total flavonoids from <em>Cryptotaenia japonica</em> Hassk.
