Effect of Selenium-Yeast supplementation on gut microbiome, intestinal histomorphology and immunity in broilers

Selenium-yeast was mixed with the basal diet of treatment groups of birds at the dose rate of 0.15 mg/kg diet in treatment-I (T-I), 0.225 mg/kg in treatment-II (T-II) and 0.30 mg/kg in treatment-III (T-III) group. The body weight and weekly weight gain increased and, feed conversion ratio improved significantly (p<0.05) in the T-III group as compared to control group. The number of goblet cells, lymphatic nodules in the intestine was more in treated birds. The villi height and crypts depth and their ratio were significantly highest in T-III group. Bursal morphology significantly improved and the number of Hassall’s corpuscle, macrophages and plasma cells were more frequent in treated birds. Among the lymphoglandular organs, the maximum IL-10 positive cells were found in the caecum and Bursa of Fabricius (BF). Among the groups, T-III group showed weak to nil IL-10 reaction. When compared between the days, BF and spleen showed better IL-10 positive reaction on 7th day but in caecal tonsil and thymus, the better reaction was on 35th day. The expression of IL-10 in the lymphoglandular organs was significantly (p<0.05) lower and T-III had minimum expression. T-II and T-III groups were better than T-I and control groups for enhancing humoral immunity against Newcastle disease virus, reduction of gut Escherichia coli, Salmonella and increased gut Bifidobacterium spp. In conclusion, supplementation of Se-yeast to the poultry diet at the dose rate 0.30 mg/kg improved the growth performance and guts microbial health, and enhanced the immunity of poultry birds.

Effects of Dietary Selenium Yeast Supplementation on Lactation Performance, Antioxidant Status, and Immune Responses in Lactating Donkeys.

Donkey milk is a traditional medicinal food with various biological activities. However, its production is very low, and lactating donkeys often experience oxidative stress, leading to a further decline in milk yield. In this study, we supplemented the diets of lactating donkeys with yeast selenium (SY) to investigate its effects on lactation performance, antioxidant status, and immune responses, and we expected to determine the optimum additive level of SY in the diet. For this study, 28 healthy lactating Dezhou donkeys with days in milk (DIM, 39.93 ± 7.02 d), estimated milk yield (EMY, 3.60 ± 0.84 kg/d), and parity (2.82 ± 0.48) were selected and randomly divided into 4 groups of 7 donkeys in each: Group SY-0 (control), Group SY-0.15, Group SY-0.3, and Group SY-0.5, with selenium supplementation of 0, 0.15, 0.3, and 0.5 mg of Se/kg DM (in form of SY) to the basal diet, respectively. The results showed a dose-dependent increase in milk yield, milk component yield, milk protein production efficiency, milk production efficiency, the activities of glutathione peroxidases (GSH-Px), catalase (CAT), and total antioxidant capacity (T-AOC), as well as the content of serum interleukin-10 (IL-10), white blood cells (WBC), lymphocytes (LYM), red blood cells (RBC), hematocrit, plasma selenium, and milk selenium. Conversely, it presented a dose-dependent decrease in the activity of nitric oxide synthase (iNOS) and the contents of malondialdehyde (MDA), reactive oxygen species (ROS), nitric oxide (NO), interleukin-1β (IL-1β), interleukin-6 (IL-6), and interferon-γ (IFN-γ). In conclusion, the results confirmed that dietary supplementation with SY can improve lactation performance, antioxidant status, and immune responses in lactating donkeys, and the recommended dose of SY was 0.3 mg/kg.

Effects of Dietary Selenium Yeast Supplementation in Pregnant Cashmere Goats on the Development of Offspring Hair Follicles

The objective of this study was to investigate the effects of maternal dietary selenium yeast (SY) supplementation during pregnancy on the hair follicle development of kids. Sixty pregnant Hanshan white cashmere goats were randomly divided into the con group (fed with a basal diet) and the SY group (fed with a basal diet with 0.4 mg/kg SY). SY was supplemented during the pregnancy until the birth of the kids. The growth performance, cashmere performance, hair follicle characteristics, and serum antioxidant capacity of the kids were periodically determined. The results showed that the birth weight of the kids in the SY group was significantly higher than that in the con group (p &lt; 0.05), and the average weight at 15 days, 1 month, 3 months, and 5 months of age increased by 13.60%, 8.77%, 8.86%, and 3.90%, respectively (p &gt; 0.05). The cashmere fineness at early birth was dramatically reduced with SY supplementation (p &lt; 0.001), whereas cashmere length and production were significantly increased at 5 months of age (p &lt; 0.05). Histology assays indicated that the primary hair follicles were fully developed at birth, and there was no significant difference in the number of primary hair follicles between the two groups (p &gt; 0.05). The number of secondary hair follicles and the number and density of active secondary hair follicles in the SY group at 15 days were significantly higher than those in the con group (p &lt; 0.05) and were increased by 11.18%, 6.18%, and 22.55% at 5 months of age, respectively (p &gt; 0.05). The serum antioxidant capacity analysis revealed that the SY group had higher levels of T-AOC, SOD, CAT, and GSH-Px activities and lower levels of MDA (p &gt; 0.05). These results reveal that the maternal dietary supplementation of SY in gestation can promote the morphogenesis and maturation of secondary hair follicles and increase the number and density of secondary hair follicles by enhancing the body’s antioxidant capacity, contributing to the improvement of cashmere quality and yield.

Enhanced Antioxidative Capacity Transfer between Sow and Fetus via the Gut-Placenta Axis with Dietary Selenium Yeast and Glycerol Monolaurate Supplementation during Pregnancy.

This study aims to investigate the impact of dietary supplementation with selenium yeast (SeY) and glycerol monolaurate (GML) on the transfer of antioxidative capacity between the mother and fetus during pregnancy and its underlying mechanisms. A total of 160 sows with similar body weight and parity of 3-6 parity sows were randomly and uniformly allocated to four groups (n = 40) as follows: CON group, SeY group, GML group, and SG (SeY + GML) group. Animal feeding started from the 85th day of gestation and continued to the day of delivery. The supplementation of SeY and GML resulted in increased placental weight and reduced lipopolysaccharide (LPS) levels in sow plasma, placental tissues, and piglet plasma. Furthermore, the redox balance and inflammatory markers exhibited significant improvements in the plasma of sows fed with either SeY or GML, as well as in their offspring. Moreover, the addition of SeY and GML activated the Nrf2 signaling pathway, while downregulating the expression of pro-inflammatory genes and proteins associated with inflammatory pathways (MAPK and NF-κB). Vascular angiogenesis and nutrient transportation (amino acids, fatty acids, and glucose) were upregulated, whereas apoptosis signaling pathways within the placenta were downregulated with the supplementation of SeY and GML. The integrity of the intestinal and placental barriers significantly improved, as indicated by the increased expression of ZO-1, occludin, and claudin-1, along with reduced levels of DLA and DAO with dietary treatment. Moreover, supplementation of SeY and GML increased the abundance of Christensenellaceae_R-7_group, Clostridium_sensus_stricto_1, and Bacteroidota, while decreasing levels of gut microbiota metabolites LPS and trimethylamine N-oxide. Correlation analysis demonstrated a significant negative relationship between plasma LPS levels and placental weight, oxidative stress, and inflammation. In summary, dietary supplementation of SeY and GML enhanced the transfer of antioxidative capacity between maternal-fetal during pregnancy via gut-placenta axis through modulating sow microbiota composition.

Effects of Dietary Selenium Yeast Supplementation on Oxidative Biomarkers of the Brain and Blood in Goats.

The present study evaluated the effects of dietary selenium yeast (SY) on the brain, CSF, and blood of 30 crossbreed goats (5-6months of age) of both sexes. After the acclimatization of 2weeks, they were randomly separated into two groups (n = 15) named C and SY groups. The C group received only a basal diet, while SY received a basal diet along with 0.3mg/kg/diet of SY (Sel-Plex®) in total 0.035mg/kg/diet of SY for 10weeks. Se concentration (µg /g dry weight) in 15 different parts of the goat's brain was accessed, and results showed that the highest concentration was found in the occipital cerebrum (322.0 ± 6.146), whereas the lowest concentration was found in the midbrain (10.33 ± 0.232). Besides, the oxidative biomarkers including GSH (12.13 ± 0.191), GSH-Px (206.7 ± 2.362), GST (23.80 ± 0.279), CAT (14.80 ± 0.279), and SOD (152.5 ± 9.540) were increased in SY as compared to GSH (8.200 ± 0.144), GSH-Px (112.9 ± 1.183), GST (18.93 ± 0.284), CAT (12.53 ± 0.215), and SOD (109.0 ± 1.966) of C. The level of cholesterol was also significantly decreased in the serum of the SY group (84.87 ± 0.960) as compared to C (110.5 ± 0.592). In addition, the cholesterol level in CSF decreased significantly in SY (0.3567 ± 0.016) as compared to C (0.509 ± 0.009). The current research suggests that SY supplementation has improved the brain's antioxidant status, blood biochemistry, and cholesterol levels in both serum and CSF of goats.

Soybean Oil and Selenium Yeast Supplementation in Quail's Diet: Productive Performance, Fatty Acid Profile, Enzyme Activity, and Oxidative Stability of Meat

AbstractThe objective of this study is to evaluate the influence of different soybean oil (SO) and selenium yeast (SeY) levels in the diet of meat quails on productive performance, physicochemical, quality measurements, fatty acid profile, enzyme activity estimation, and oxidative stability of meat. A total of 288 one‐day old commercial meat quails are allocated into four groups according to a completely randomized experimental design, with two levels of SO (1.5% and 3.0%) and SeY (0 and 0.15 mg kg−1) in the feed. The supplementation period is 42 days. The use of up to 3% supplementation of SO in the diet improves (p &lt; 0.05) the productive performance. The polyunsaturated acid content and thiobarbituric acid value in the meat are increased (p &lt; 0.05) as an effect of the SO increase in the diet. The use of SeY in quail diets associates with 3% inclusion of SO and promotes improvements in the qualitative parameters of the quail meat. Therefore, the inclusion of 0.15 mg kg−1 of SeY in the diets is a viable alternative for improving the oxidative stability of meat since it improves the qualitative parameters of quail meat in diets with a high inclusion of SO.Practical Applications: These results have practical applicability in the meat industry, since it is possible to change the lipid profile of this raw material, meeting the requirement of the consumer market for healthy foods, reducing the total cholesterol content, and nutritionally desirable, through enrichment with polyunsaturated fatty acids.

Effects of Selenium Yeast on Egg Quality, Plasma Antioxidants, Selenium Deposition and Eggshell Formation in Aged Laying Hens.

Internal egg and eggshell quality are often deteriorated in aging laying hens, which causes huge economic losses in the poultry industry. Selenium yeast (SY), as an organic food additive, is utilized to enhance laying performance and egg quality. To extend the egg production cycle, effects of selenium yeast supplementation on egg quality, plasma antioxidants and selenium deposition in aged laying hens were evaluated. In this study, five hundred and twenty-five 76-week-old Jing Hong laying hens were fed a selenium-deficient (SD) diet for 6 weeks. After Se depletion, the hens were randomly divided into seven treatments, which included an SD diet, and dietary supplementation of SY and sodium selenite (SS) at 0.15, 0.30, and 0.45 mg/kg to investigate the effect on egg quality, plasma antioxidant capacity, and selenium content in reproductive organs. After 12 weeks of feeding, dietary SY supplementation resulted in higher eggshell strength (SY0.45) (p < 0.05) and lower shell translucence. Moreover, organs Se levels and plasma antioxidant capacity (T-AOC, T-SOD, and GSH-Px activity) were significantly higher with Se supplementation (p < 0.05). Transcriptomic analysis identified some key candidate genes including cell migration inducing hyaluronidase 1 (CEMIP), ovalbumin (OVAL), solute carrier family 6 member 17 (SLC6A17), proopiomelanocortin (POMC), and proenkephalin (PENK), and potential molecular processes (eggshell mineralization, ion transport, and eggshell formation) involved in selenium yeast's effects on eggshell formation. In conclusion, SY has beneficial functions for eggshell and we recommend the supplementation of 0.45 mg/kg SY to alleviate the decrease in eggshell quality in aged laying hens.

Supplementation of Selenium-Yeast Enhances Fishmeal Replacement by Soy Protein Concentrate in Diets for Golden Pompano (Trachinotus ovatus)

An 8-week feeding trial was conducted to investigate the effect of selenium-yeast (Se-yeast) supplementation on replacing fishmeal with soy protein concentrate (SPC) in diets for golden pompano (Trachinotus ovatus). The control diet (C) contained 400 g/kg fishmeal, of which 40% and 80% of the fishmeal in diet C were substituted by SPC, with or without 1 g/kg Se-yeast supplementation (four diets, R40, R80, R40 + Se, and R80 + Se, were formulated). The weight gain of the fish-fed diet C showed no obvious difference from that of the fed-diet R40 + Se, although it was higher than that of the fish-fed diets R40, R80, and R80 + Se. Fish-fed diets R40 + Se and R80 + Se exhibited higher weight gain and nitrogen and phosphorus retention efficiencies, whereas they had relatively lower feed intake, feed conversion ratio, and nitrogen and phosphorus waste outputs than fish-fed diets R40 and R80. No statistical differences were found in condition factor and whole-body components either between fish-fed diets R40 and R40 + Se or between fish-fed diets R80 and R80 + Se. Our research suggests that it is feasible to reduce dietary fishmeal for golden pompano to 240 g/kg with SPC and 1 g/kg Se-yeast inclusion.

Growth, Nutrient Retention, Waste Output, and Antioxidant Capacity of Juvenile Triangular Bream (Megalobrama terminalis) in Response to Dietary Selenium Yeast Concentration.

An 8-week feeding trial was conducted to investigate the effects of different dietary selenium yeast levels on growth, nutrient retention, waste output, and antioxidant capacity of juvenile triangular bream (Megalobrama terminalis). Five isonitrogenous (320 g/kg crude protein) and isolipidic (65 g/kg crude lipid) diets were formulated, with supplementation of graded levels of selenium yeast at 0 (diet Se0), 1 (diet Se1), 3 (diet Se3), 9 (diet Se9), and 12 g/kg (diet Se12). No significant differences were found in initial body weight, condition factor, visceral somatic index, hepatosomatic index, and whole body contents of crude protein, ash, and phosphorus among fish fed different test diet. The highest final body weight and weight gain rate were found in fish fed diet Se3. The specific growth rate (SGR) is closely related to dietary selenium (Se) concentrations with a relationship described as SGR = -0.0043 Se2 + 0.1062 Se + 2.661. Higher feed conversion ratio was found, while lower retention efficiencies of nitrogen and phosphorus were found in fish fed diets Se1, Se3, and Se9 than in fish fed diet Se12. Contents of selenium in whole body, vertebra, and dorsal muscle increased with dietary supplementation of selenium yeast increased from 1 mg/kg to 9 mg/kg. Lower nitrogen and phosphorous waste was found in fish fed diets Se0, Se1, Se3, and Se9 than in fish fed diet Se12. Fish fed diet Se3 exhibited the highest activities of superoxide dismutase, glutathione peroxidase, and lysozyme while the lowest malonaldehyde content in both the liver and kidney. Our results showed that the optimal dietary selenium requirement for triangular bream should be 12.34 mg/kg based on the nonlinear regression on SGR, and fish fed diet Se3 in which selenium concentration (8.24 mg/kg) was close to the optimal requirement displayed the best growth performance, feed nutrient utilization, and antioxidant capacity.

Supranutritional Selenium-Yeast Supplementation of Beef Cows during the Last Trimester of Pregnancy Results in Higher Whole-Blood Selenium Concentrations in Their Calves at Weaning, but Not Enough to Improve Nasal Microbial Diversity.

Simple SummaryWe previously showed that feeding selenium-enriched alfalfa hay to weaned beef calves diversified the nasal microbiome and improved health and growth in the feedlot, resulting in greater carcass weight and quality at slaughter. The objective of the current study was to see whether selenium supplementation of dams during various pregnancy trimesters similarly improves health and performance. Maternal supranutritional selenium supplementation during pregnancy increases selenium concentrations in calves, but is more effective the shorter the time period between maternal supplementation and assessment. Thus, calves should be fed selenium-enriched forages again at weaning in order to see a benefit in the nasal microbiome abundance and diversity, and consequently, in carcass measurements at slaughter.We previously reported that feeding Se-biofortified alfalfa hay to weaned beef calves in a preconditioning program increases whole-blood Se (WB-Se) concentrations and nasal microbiome abundance and diversity during the preconditioning period, decreases morbidity and mortality during the feedlot period, and increases carcass weight and quality at slaughter. The objective of the current study was to see whether similar improvements can be achieved through Se supplementation of dams during various pregnancy trimesters. In a two-year experimental study, 80 Angus-cross cows received once-weekly Se-yeast boluses containing 105 mg of Se, during either the first (TR-1), second (TR-2), or third (TR-3) pregnancy trimester, or were not bolused (CTR). Whole-blood Se concentrations were higher from CTR, to TR-1, to TR-2, and to TR-3 in newborn calves (all p < 0.01). At weaning, only calves from TR-3 mothers had higher WB-Se concentrations compared with calves from CTR mothers (p = 0.02), and no significant differences in nasal microbiome abundance and diversity or nasal microbiota were observed. In the feedlot period, morbidity was low, and no differences were observed. At slaughter, no differences in carcass weight and quality were observed. In conclusion, Se supplementation of pregnant cows is effective for increasing WB-Se concentration of newborn calves, and the increase can be sustained until weaning for calves born to TR-3 dams. However, the increase in WB-Se concentrations is small and does not result in beneficial changes in the nasal microbiome. Thus, calves should be fed Se-biofortified forages again at weaning in a preconditioning program in order to diversify the nasal microbiome prior to entering the feedlot.

Ameliorative effect of selenium yeast\xa0supplementation on the physio-pathological impacts ofchronic exposure to glyphosate and or malathion in Oreochromis niloticus

BackgroundPesticide exposure is thought to be a major contributor to living organism health deterioration, as evidenced by its impact on both cultured fish species and human health. Commercial fish diets are typically deficient in selenium (Se); hence, supplementation may be necessary to meet requirements during stress. Therefore, this study was conducted to investigate the protective role of selenium yeast (SY) supplementation for 60 days against the deleterious effects of glyphosate and or malathion chronic toxicity at sublethal concentrations in Oreochromis niloticus .MethodsTwo hundred and ten fish were divided into seven groups (n = 30/group) as follows: G1 (negative control); G2 (2 mg L− 1 glyphosate); G3 (0.5 mg L− 1 malathion); G4 (glyphosate 1.6 mg L− 1 and malathion 0.3 mg L− 1); G5 (glyphosate 2 mg L− 1 and SY 3.3 mg kg− 1); G6 (malathion 0.5 mg L− 1 and SY 3.3 mg kg− 1); and G7 (glyphosate 1.6 mg L− 1; malathion 0.3 mg L− 1 and SY 3.3 mg kg− 1).ResultsResults revealed significant alteration in growth performance parameters including feed intake (FI), body weight (BW), body weight gain (BWG), specific growth rate (SGR), feed conversion ratio (FCR), and protein efficiency ratio (PER). G4 has the highest documented cumulative mortalities (40%), followed by G3 (30%). Additionally, the greatest impact was documented in G4, followed by G3 and then G2 as severe anemia with significant thrombocytopenia; leukocytosis; hypoproteinemia; increased Alanine aminotransferase (ALT) and Aspartate aminotransferase (AST), urea, and creatinine, as well as malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GPx). Considering the previously mentioned parameters, selenium yeast (Saccharomyces cerevisiae) (3.3 mg kg− 1 available selenium) mitigated the negative impact of both the agrochemicals, whether exposed singly or in combination, in addition to their antioxidative action.ConclusionsIn conclusion, our study found that organophosphorus agrochemicals, single or combined, had negative impacts on Oreochromis niloticus regarding growth performance, biochemical and hematological changes in the serum, as well as induced oxidative damage in liver and kidney tissues. Supplementation of SY at the rate of 3.3 mg kg− 1 diet (2.36 mg kg− 1 selenomethionine and 0.94 mg organic selenium) ameliorated the fish performance and health status adversely affected by organophosphorus agrochemical intoxication.

Dietary selenium requirement of triploid rainbow trout ( Oncorhynchus mykiss ) based on selenium yeast supplementation

Selenium(Se) is a biological essential trace element that affects the growth and health of fish. The present study aimed to explore the dietary Se requirement of triploid rainbow trout using Se-yeast supplement. The levels of Se added to the basal diet were 0(Se0), 0.1(Se0.1), 0.3(Se0.3), 0.5(Se0.5), 1.0(Se1.0), 4.0(Se4.0) and 16.0(Se16.0) mg Se/kg diet respectively. Results showed that the fish fed Se4.0 diet had significantly higher weight gain and lower oxidative stress compared with the fish fed Se16.0 and Se0 diets. The optimal dietary Se requirement of triploid rainbow trout was 3.53 mg/kg based on broken-line regression of growth performance. Dietary Se supplementation significantly increased the expression level of selenoprotein genes both in liver (GPX1a, GPX1b1, GPX1b2, SelK, SelP, SelT1, SelT2, SelWl, TrxR3a, SelU) and in muscle (GPX1a, GPX1b2, SelK, SelP, SelU, SelWl, TrxR3b, SelN, MsrB1Al). The minimum dietary Se level was 2.19 or 1.28 mg/kg when the mRNA level of all these selenoproteins reached a plateau in muscle or liver respectively. In conclusion, based on broken-line regression of growth performance and sigmoidal regression of selenoprotein genes expression in muscle and liver, dietary Se requirement of triploid rainbow trout was 3.53, 2.19 and 1.28 mg/kg respectively.

Microbiome and ileum transcriptome revealed the boosting effects of selenium yeast on egg production in aged laying hens.

The declines in laying performance during the late production period have adverse effects on the length of the production cycle. Improving the nutrition of laying hens is a crucial measure to reverse this declination. This study investigated the effect of selenium yeast (SY) on egg production, ileal gene expression and microbiota, as well as elucidating their associations in aged laying hens. A total of 375 Jinghong laying hens at 76 weeks old were randomly assigned into 5 dietary treatments, which included a selenium-deficient basal diet based on corn-soybean meal, and dietary supplementation of SY at 0.15, 0.30 and 0.45 mg/kg, and sodium selenite at 0.45 mg/kg. The results showed that SY ameliorated the depression in aged laying performance in the 0.30 mg/kg group (P < 0.01). Selenium yeast significantly increased ileum selenium concentration (P < 0.05), and SY groups had higher selenium deposition efficiency than the sodium selenite group. Functional enrichment and Short Time-series Expression Miner (STEM) analysis indicated that SY activated metabolic progress (e.g., glycerolipid metabolism, glycerophospholipid metabolism, and fatty acid metabolism), immune response and oxidative stress response. Four hub genes including thioredoxin reductase 1 (TXNRD1), dihydrolipoamide dehydrogenase (DLD), integrin linked kinase (ILK) and leucine zipper tumor suppressor 2 (LZTS2) were involved in intestinal metabolism which was closely associated with selenium deposition/status. Moreover, the relative abundance of Veillonella, Turicibacter and Lactobacillus was significantly increased, but the relative abundance of Stenotrophomonas was significantly decreased by SY supplementation. Multi-omics data integration and Canonical correspondence analysis (CCA) showed that both the ileum selenium content and the laying rate were highly correlated with pathways and bacteria enriched in metabolism and immune response. Meanwhile, the “switched on” gene prostate stem cell antigen (PSCA) had a positive relationship with Veillonella and a negative relationship with the opportunistic pathogens Stenotrophomonas. Overall, our study offered insight for the further exploration of the role of SY on boosting egg production and balancing ileum intestinal flora in aged laying hens.

The Selenium Yeast vs Selenium Methionine on Cell Viability, Selenoprotein Profile and Redox Status via JNK/ P38 Pathway in Porcine Mammary Epithelial Cells.

Comprehensive studies have been conducted to compare the effect of organic and inorganic selenium previously, but there is still limited knowledge about the difference between organic selenium (Se) from varied sources despite the widely use of organic Se in both animal and human being nutrient additives. In the present study, we systemically compared the effect of two different types of organic Se including selenium yeast (SeY) and selenium methionine (Sel-Met) on cell viability, selenoprotein transcriptome, and antioxidant status in porcine mammary epithelial cells (PMECs) and the results indicated that appropriate addition of SeY and Sel-Met both significantly promoted cell viability and up-regulated the mRNA expression of most selenopreoteins including DIOs, GPXs, and TrxRs family et al. (P < 0.05). Besides, two different sources of Se supplementation both greatly improved redox status with higher levels of T-AOC, SOD, and CAT (P < 0.05), while less content of MDA (P < 0.05), and reduced protein expression of cleaved-caspase-3 (P < 0.05) to mitigate cell apoptosis. Furthermore, the key proteins related to p38/JNK pathway including p38, p-p38, JNK, and p-JNK were apparently reduced in the groups with both of SeY and Sel-Met (P < 0.05). Interestingly we found that the changes induced by SeY supplementation in cell viability, selenoprotein transcriptome, antioxidative capacity, and anti-apoptosis were comprehensively greater compared with same levels addition of Sel-Met in PEMCs (P < 0.05). In conclusion, both SeY and Sel-Met promoted cell viability and attenuated cell apoptosis by regulating the selenoprotein expression and antioxidative capacity via p38/JNK signaling pathway in PMEC, but SeY has more efficient benefits than that of Sel-Met.

Effects of dietary supplementation with different levels of selenium yeast on growth performance, carcass characteristics, antioxidant capacity, and meat quality of Tan sheep

This study was conducted to evaluate the influence of dietary supplementation with different levels of selenium yeast (SeY) on growth performance, carcass characteristics, antioxidant capacity and meat quality of Tan sheep. Sixty-four (n = 64) five-month-old male Tan sheep with an initial body weight of 32 ± 1.95 kg (BW ± SD) were selected and randomly assigned to four treatments with 16 replicates of one sheep each. The four groups received a basal diet (0.16 mg Se/kg) supplemented with 28.94, 105.46, 258.51, 564.60 mg/kg SeY to final dietary selenium (Se) concentrations of 0.25 (Se0.25), 0.5 (Se0.5), 1.0 (Se1.0), or 2.0 (Se2.0) mg/kg for 60 days, respectively. The feed-to-weight gain ratio showed a quadratic significant change of first increasing and then decreasing with the increase of dietary SeY supplemental level (p<0.01). Dietary Se at >0.25 mg/kg markedly decreased the GR value of the sheep (p<0.05). Thioredoxin reductase activity in sheep serum was markedly higher in the Se0.5 group (p<0.05) and glutathione peroxidase activity in sheep liver was significantly higher in the Se1.0 group (p<0.01), and both began to decline afterwards. Dietary Se at >0.25 mg/kg significantly enhanced muscle thioredoxin reductase activity (p<0.05) and decreased the content of thiobarbituric acid reactive substances in the liver and muscle tissues (p<0.01). Different supplementary levels of SeY had no significant effect on pH values at 45 min and 24 h postmortem (p>0.05). Moreover, final dietary Se at >0.25 mg/kg was associated with significant decreases in the lightness and yellowness values of muscle tissues at 45 min (p<0.01), but had no obvious effect on the redness value (p>0.05). Meat color of mutton had a less change over time in the Se0.5 group, and the color stability was better. Drip loss of muscle decreased significantly with an increase in dietary SeY supplementation (p<0.05). In general, dietary supplemented with SeY to a final Se concentration of 0.5–1.0 mg/kg can maintain antioxidant capacity at a high level and obtain better meat quality of Tan sheep. The results of this study pave the way for the rational application of Se in the feed of Tan sheep.

Milk selenium content and speciation in response to supranutritional selenium yeast supplementation in cows.

The effects of selenium (Se) yeast supplementation on performance, blood biochemical and antioxidant parameters, and milk Se content and speciation were evaluated. Thirty-six mid-lactation Holstein dairy cows were randomly assigned to 1 of 3 treatments: 1) control (basal diet containing Se at 0.11 mg/kg DM), 2) basal diet + 0.5 mg supplemental Se/kg DM (SY-0.5), and 3) basal diet + 5 mg supplemental Se/kg DM (SY-5). Selenium was supplemented as Se yeast. The trial consisted of a 1-week pretrial period and an 8-week experimental period. Milk somatic cell score decreased with SY-5 supplementation (P < 0.05), but other performance parameters were not affected (P > 0.05). The serum Se concentration increased with the increasing levels of Se yeast supplementation (P < 0.05), however, blood biochemical parameters showed few treatment effects. The antioxidant capacity of dairy cows was improved with Se yeast supplementation reflected in increased serum glutathione peroxidase activity (P < 0.05) and total antioxidant capacity (P = 0.08), and decreased malondialdehyde concentration (P < 0.05). Milk total Se concentration increased with Se dose (P < 0.05). Also, the selenomethionine concentration increased with Se dose from 13.0 ± 0.7 μg/kg in control to 33.1 ± 2.1 μg/kg in SY-0.5 and 530.4 ± 17.5 μg/kg in SY-5 cows (P < 0.05). Similarly, selenocystine concentration increased from 15.6 ± 0.9 μg/kg in control and 18.9 ± 1.1 μg/kg in SY-0.5 to 22.2 ± 1.5 μg/kg in SY-5 cows (P < 0.05). In conclusion, Se yeast is a good organic Se source to produce Se-enriched cow milk with increased Se species including selenomethionine and selenocystine. The results can provide useful information on milk Se species when a high dose Se yeast was supplemented in the cow diet.

THE EFFECT OF TWO DIFFERENT DOSES OF SELENIUM YEAST AND SODIUM SELENITE ON SELENIUM LEVEL IN BLOOD, COLOSTRUM, MILK AND METABOLIC PROFILE IN DAIRY COWS

The aim of the study was to compare effects of two levels of selenium yeast (SY) and sodium selenite (SS) supplementation on blood, colostrum and milk Se levels, blood chemistry parameters in dairy cows under field conditions. The study included 36 Holstein cows from 5 weeks prepartum to 12 weeks postpartum. The cows were divided into 4 treatments (n = 9): 1. Negative Control (NC) – no Se supplement ; 2. Positive Control (PC) – 0.3 ppm Se (from sodium selenite) ; 3. S1 – 0.3 ppm Se (from selenium yeast – SELSAF ® , Lesaffre Feed Additives, France ) ; 4 . S2 – 5 ppm Se ( SELSAF ® , Lesaffre Feed Additives, France ). At 5 and 1 weeks prepartum, on the calving day, and 4, 8, 12 weeks postpartum, blood samples were taken . Colostrum and milk were collected on the calving day and 4, 8 and 12 weeks postpartum and analysed for Se content. Initial Se status in all the cows was marginal. Negative Control cows showed suboptimal blood Se levels for the whole period under study. S1 and S2 showed significantly (P &lt; 0.01) higher values than NC and PC cows. Colostrum Se levels were significantly higher in S1 (128.17 ug/L) and S2 (155.58 ug/L) than in NC (94.68 ug/L) and PC (99.3 ug/L) cows. Overall mean milk Se levels in S1 (20.4 ug/L) and S2 (50.0 ug/L) were higher than in NC (10.9 ug/L) and PC (16.6 ug/L). Biochemical indicators of blood did not significantly differ between the treatments and were within the physiological ranges. Se levels in whole blood, colostrum and milk, and GSH-Px activity were significantly increased by selenium yeast supplementation. The response was dose dependent and consistent. Selenium from organic source was more bioavailable than selenium from sodium selenite. Dietary Se at a tolerance dose (5 ppm) had no detrimental effect on the blood chemistry parameters analysed.

An evaluation on the selenium yeast supplementation in the practical diets of early juvenile sea cucumber ( Apostichopus japonicus ): Growth performance, digestive enzyme activities, immune and antioxidant capacity, and body composition

A 45-day feeding trial was carried out to investigate the effects of dietary selenium yeast (Se-yeast) on the survival, growth performance, activities of digestive enzymes and antioxidant enzymes, and body composition of early juvenile Apostichopus japonicus. Five isoprotic (15.6%) and isolipic (1.5%) feeds with graded levels of Se-yeast (0, 0.5, 1.0, 1.5 and 2.0 mg/kg) were formulated and randomly allocated to the early juvenile sea cucumbers (initial weight: 0.11 ± 0.01 g). The results showed that the weight gain rate (WGR) and relative visceral weight ratio (RVW) significantly increased as the Se-yeast supplementation increased from 0 to 1.0 mg/kg, and then reached a plateau with further increase in Se-yeast, while survival rate (SR) increased as the supplementation level of Se-yeast increased, and the lipase and amylase activities first significantly increased as the Se-yeast increased from 0 to 1.0 mg/kg, and then significantly decreased with the continuous supplementation of Se-yeast. The antioxidant capacity and nonspecific immunity were significantly elevated by the moderate level (0.5 and 1.0 mg/kg) of Se-yeast. Besides, the activities of immune-related enzymes and transcription of antioxidation-related genes were significantly elevated by the supplementation of Se-yeast. However, malonaldehyde contents were significantly reduced in the treatments with 0.5 and 1.0 mg/kg Se-yeast. The selenium content in the body wall of the sea cucumber showed a markedly increasing trend with increasing Se-yeast supplementation levels. Results above indicated that a moderate level (0.5–1.0 mg/kg) of Se-yeast enhanced the growth performance, digestive enzyme activities, antioxidant capacity and nonspecific immunity of early juvenile A. japonicus.

Selenium source and level on performance, selenium retention and biochemical responses of young broiler chicks

BackgroundSelenium (Se) has been recognized as an essential micronutrient for nearly all forms of life. In recent decades, broiler responses to dietary Se supplemental levels and sources have received considerable attention. On environmental grounds, organic trace mineral utilization in practical broiler feeds has been defended due to its higher bioavailability. In such feeds, trace minerals are provided simultaneously in the same supplement as inorganic salts or organic chelates, a fact commonly ignored in assays conducted to validate organic trace mineral sources. The current assay aimed to investigate growth and biochemical responses, as well as Se retention of growing chicks fed diets supplemented with organic and inorganic Se levels and where the trace minerals (zinc, copper, manganese, and iron) were provided as organic chelates or inorganic salts according to Se source assessed. In so doing, a 2 × 5 factorial arrangement was used to investigate the effects of sodium selenite (SS) and selenium-yeast (SY) supplemented in feeds to provide the levels of 0, 0.08, 0.16, 0.24, and 0.32 mg Se/kg.ResultsChicks fed selenium-yeast diets had body weight (BW), and average daily gain (ADG) maximized at 0.133 and 0.130 mg Se/kg, respectively. Both Se sources linearly increased (P < 0.05) the glutathione peroxidase (GSH-Px) activity in chick blood but higher values were observed in sodium selenite fed chicks (P < 0.05). Both Se sources influenced thyroid hormone serum concentrations (P < 0.05). Chicks fed SY exhibited greater retention of Se in the feathers (P < 0.05). Relative bioavailability of selenium yeast compared with SS for the Se content in carcass, feathers, total and Se retention were, 126, 116, 125 and 125%, respectively. SY supplementation resulted in lower liver Se concentration as Se supplementation increased (P < 0.05).ConclusionsBased on performance traits, the supplemental level of organic Se as SY in organic trace minerals supplement to support the maximal growth of broiler chicks is 0.133 mg Se/kg.
