Reducing Self-Assembly by Increasing Net Charge: Effect on Biological Activity of Mastoparan C.

The ability of amphipathic peptides to arrange themselves in aqueous solutions, known as self-assembly, has been found to reduce the effectiveness of these peptides in interacting with cell membranes. Therefore, minimizing their tendency to self-assemble could be a potential strategy for enhancing the pharmacological properties of antimicrobial peptides (AMPs). To explore this idea, this study prepared a series of natural peptides mastoparan C (MPC) with increased net charge and hydrophilicity via alanine-to-lysine substitution and investigated the impact on the biological activity. The preliminary data suggested the influence of both the overall positive charge and the position of a lysine residue on the self-assembly of MPC and its derivatives. Besides, the analogue MPC-A5K,A8K displayed higher anticancer activity and comparable antimicrobial activity with significantly lower hemolysis than MPC. Hence, reducing self-assembly by expanding the cationic area could be a promising approach for developing potent and selective AMPs.

Structural and biological characterization of shortened derivatives of the cathelicidin PMAP-36

Cathelicidins, a family of host defence peptides in vertebrates, play an important role in the innate immune response, exhibiting antimicrobial activity against many bacteria, as well as viruses and fungi. This work describes the design and synthesis of shortened analogues of porcine cathelicidin PMAP-36, which contain structural changes to improve the pharmacokinetic properties. In particular, 20-mers based on PMAP-36 (residues 12-31) and 13-mers (residues 12-24) with modification of amino acid residues at critical positions and introduction of lipid moieties of different lengths were studied to identify the physical parameters, including hydrophobicity, charge, and helical structure, required to optimise their antibacterial activity. Extensive conformational analysis, performed by CD and NMR, revealed that the substitution of Pro25-Pro26 with Ala25-Lys26 increased the α-helix content of the 20-mer peptides, resulting in broad-spectrum antibacterial activity against Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa and Staphylococcus epidermidis strains. Interestingly, shortening to just 13 residues resulted in only a slight decrease in antibacterial activity. Furthermore, two sequences, a 13-mer and a 20-mer, did not show cytotoxicity against HaCat cells up to 64 µM, indicating that both derivatives are not only effective but also selective antimicrobial peptides. In the short peptide, the introduction of the helicogenic α-aminoisobutyric acid forced the helix toward a prevailing 310 structure, allowing the antimicrobial activity to be maintained. Preliminary tests of resistance to Ser protease chymotrypsin indicated that this modification resulted in a peptide with an increased in vivo lifespan. Thus, some of the PMAP-36 derivatives studied in this work show a good balance between chain length, antibacterial activity, and selectivity, so they represent a good starting point for the development of even more effective and proteolysis-resistant active peptides.

Effect of Spacer Length Modification of the Cationic Side Chain on the Energetics of Antimicrobial Peptide Binding to Membrane-Mimetic Bilayers.

Understanding the mechanism of action of the antimicrobial peptide (AMP) in terms of its structure and energetics is the key to designing new potent and selective AMPs. Recently, we reported a membranolytic 14-residue-long lysine-rich cationic antimicrobial peptide (LL-14: NH3+-LKWLKKLLKWLKKL-CONH2) against Pseudomonas aeruginosa, Klebsiella pneumoniae, and Staphylococcus aureus, which is limited by cytotoxicity and expected to undergo facile protease degradation. Aliphatic side-chain-length modification of the cationic amino-acid residues (Lys and Arg) is a popular strategy for designing protease-resistant AMPs. However, the effect of the peptide side-chain length modifications on the membrane binding affinity and its relation to the atomic structure remain an unsolved problem. We report computer simulations that quantitatively calculated the difference in peptide binding affinity to membrane-mimetic-bilayer models (bacterial: 1-palmitoyl-2-oleoyl-phosphatidylethanolamine (POPE)/1-palmitoyl-2-oleoyl-phosphatidylglycerol (POPG) bilayer and mammalian: 1-palmitoyl-2-oleoyl-phosphatidylcholine (POPC) bilayer) upon decreasing or increasing the spacer length of the cationic lysine residues of LL-14 (as well as their arginine analogues). We show that the peptide/bilayer interaction energetics varies drastically in response to spacer length modification. The strength of peptide discrimination depends strongly on the nature of the bilayer (bacterial or mammalian mimetic model). An increase in the lysine spacer length by one carbon (i.e., homolysine analogue of LL-14) is weakly/strongly disfavored by the bacterial/mammalian-membrane-mimetic bilayer. Recently, we have demonstrated an excellent correlation between the antimicrobial activity of the membranolytic cationic peptides and their binding affinity to membrane-mimetic-bilayer models. Thus, the homolysine analogue of LL-14 is a promising noncytotoxic AMP with conserved activity. On the other hand, homoarginine analogue (arginine spacer length increment by a single carbon) was preferred by both the bacteria and the mammalian mimetic bilayers and displayed the strongest affinity for the former among the peptides studied in this work. Thus, the promising most potent homoarginine analogue is likely to be cytotoxic. Shortening the Lys/Arg side chain to a three-carbon spacer (Dab/Agb) improves the binding affinity to bacterial and mammalian-membrane-mimetic bilayers. Arginine and arginine-derivative peptides exhibited stronger binding affinity to the bilayers relative to the lysine analogue. The results provide a plausible explanation to the previous experimental observations, viz., superior antimicrobial activity of the arginine peptides relative to Lys peptides and the improvement of antimicrobial activity upon substitution of Lys with Dab in the cationic peptides. The simulations revealed that the small change in the peptide hydrophobicity by Lys/Arg spacer length modification could drastically alter the energetics of peptide/bilayer binding by fine-tuning the electrostatic interactions. The energetics underlying the peptide selectivity by simple membrane-mimetic bilayer models may be beneficial for designing new selective and protease-resistant AMPs.

Effect of substituting glutamine with lysine on structural and biological properties of antimicrobial peptide Polybia-MP1.

The natural antimicrobial peptide Polybia-MP1 is a promising candidate for developing new treatment therapy for infection and cancer. It showed broad-spectrum antimicrobial and anticancer activity with high safety on healthy cells. However, previous sequence modification usually resulted in at least one of two consequences: a notable increase in hemolytic activity or a considerable decrease in activity against Gram-negative bacteria and cancer cells. Herein, a new approach was applied by replacing the amino acid Glutamine at position 12 with Lysine and generating the MP1-Q12K analog. Our preliminary data suggested an enhancement in antibacterial and antifungal activity, whereas the anticancer and hemolytic activity of the two peptides were comparable. Moreover, MP1-Q12K was found to be less self-assembly than Polybia-MP1, which further supports the enhancement of antimicrobial properties. Hence, this study provides new information regarding the structure-activity relationships of Polybia-MP1 and support for the development of potent, selective antimicrobial peptides.

Structural insights on the selective interaction of the histidine-rich piscidin antimicrobial peptide Of-Pis1 with membranes

Of-Pis1 is a potent piscidin antimicrobial peptide (AMP), recently isolated from rock bream (Oplegnathus fasciatus). This rich in histidines and glycines 24-amino acid peptide displays high and broad antimicrobial activity and no significant hemolytic toxicity against human erythrocytes, suggesting low toxicity. To better understand the mechanism of action of Of-Pis1 and its potential selectivity, using NMR and CD spectroscopies, we studied the interaction with eukaryotic and procaryotic membranes and membrane models. Anionic sodium dodecyl sulfate (SDS) and lipopolysaccharide (LPS) micelles were used to mimic procaryotic membranes, while zwitterionic dodecyl phosphocholine (DPC) was used as eukaryotic membrane surrogate. In an aqueous environment, Of-Pis1 adopts a flexible random coil conformation. In DPC and SDS instead, the N-terminal region of Of-Pis1 forms an amphipathic α-helix with the non-polar face in close contact with the micelles. Slower solvent exchange and higher pKas of the histidine residues in SDS than in DPC suggest that Of-Pis1 interacts more tightly with SDS. Of-Pis1 also binds tightly and structurally perturbs LPS micelles. Of-Pis1 interacts with both Escherichia coli and mammalian cell membranes, but only in the presence of Escherichia coli membranes it populates the helical conformation. Furthermore, ligand-based NMR experiments support a tighter and more specific interaction with bacterial than with eukaryotic membranes. Overall, these data clearly show the selective interaction of this broadly active AMP with bacterial over eukaryotic membranes. The conformational information is discussed in terms of Of-Pis1 amino acid sequence and composition to provide insights useful to design more potent and selective AMPs.

Antimicrobial peptide functionalized gold nanorods combining near-infrared photothermal therapy for effective wound healing

Photothermal therapy using laser activated gold nanorods (AuNRs) is a strategy for treatment of bacterial infections. Nevertheless, it also exerts cytotoxicity against human cells which leads to adverse effects in healthy human tissues and limits the applicable dose. Functionalization of AuNRs with a selective antimicrobial peptide (AMP) with higher selectivity for bacteria over human cells is a promising strategy for increasing the selectivity of the AuNRs for bacteria, hence increasing their cellular uptake by the bacteria in order to achieve stronger antimicrobial effects with lower doses of AuNRs without damaging the human cells. In this study, the surface of AuNRs was functionalized with a short AMP named C-At5 and the efficiency of the peptide functionalized AuNRs in killing gram-positive and gram-negative bacteria was evaluated in vitro as well as their potential for facilitating wound healing in a mouse model of wound infection with and without application of laser. The peptide-conjugated AuNRs exhibited higher antibacterial activity in vitro compared to the plain AuNRs both in the presence and absence of laser irradiation. Furthermore, AuNR@C-At5 had very low toxicity against human skin fibroblasts and human red blood cells indicating their higher biocompatibility compared to the plain AuNRs. Treatment of wounded mice with AuNR@C-At5 accelerated the wound healing process which was further enhanced by applying laser. The system developed in this study has great potential for customization for specific antimicrobial or antifungal therapy via conjugation of different types of AMPs with higher selectivity and can therefore serve as a guide for any future attempts in this regard.

Stable isotope analysis confirms substantial changes in the fatty acid composition of bacteria treated with antimicrobial random peptide mixtures (RPMs)

Resistance of plant-pathogenic bacteria to classic antibiotics has prompted the search for suitable alternative antimicrobial substances. One promising strategy could be the use of purposely synthesized random peptide mixtures (RPMs). Six plant-pathogenic bacteria were cultivated and treated with two RPMs previously found to show antimicrobial activity mainly by bacterial membrane disruption. Here, we show that bacteria treated with RPMs showed partly remarkable changes in the fatty acid pattern while those unaffected did not. Quantitative changes could be verified by compound specific isotope analysis of δ13C values (‰). This technique was employed due to the characteristic feature of stronger bonds between heavier isotopes in (bio)chemical reactions. As a proof of concept, the increase in abundance of a fatty acid group after RPM treatment was accompanied with a decrease in the 13C content and vice versa. We propose that our findings will help designing and synthesizing more selective antimicrobial peptides.

Antimicrobial Peptide-Membrane Interactions: Insights from Molecular Simulations

Antimicrobial peptides (AMPs) are found in the innate immune systems of most living organisms. These peptides exhibit cell selectivity, and activity against a broad spectrum of microorganisms, making them promising candidates as antimicrobial biomaterials. The AMPs are anchored with polymer tethers and biologically synthesized as functionalized biomaterials. We successfully prepared LL-37 conjugated biopolymer materials with antimicrobial activity, which switch to micelles at temperatures between 27-30 oC. Understanding the peptide-membrane interactions represents the basis for AMP's selectivity for bacterial cell membranes. We hypothesized that peptide insertion is assisted by membrane curvature. Models of a gram-negative bacterial outer membrane comprising POPE/DMPG/CL (90:5:5) and a membrane with the composition DMPC/DMPG/CL (90:5:5) were investigated using molecular dynamics simulations. The antimicrobial peptide LL-37 was arranged on the surface of the membranes as “carpets” of ordered peptides. We also performed all-atom molecular dynamics simulations using NAMD 2.13 to visualize the carpet-to-barrel or toroidal-pore transition. To determine the energetically favorable model, the non-bonded interactions between the carpet and pore models were compared using the NAMD energy. From our in-silico observations, the pore model is more favorable than the carpet model for the same peptide/lipid ratio. Critical values of the peptide/lipid ratio required for cell penetration were investigated to determine the rate of peptide insertion into the membrane with different LL-37 concentrations. From the simulation timescale (in microseconds), as the peptide/lipid ratio increased, partial insertion and membrane curvature were observed in the bacterial mimic model. Further, the C-terminal helix of LL-37 was observed to unfold when interacting with phosphate head groups of the lipids. The propensity for AMPs to insert into the lipid bilayer via the N-terminus or C-terminus was similar. These observations from molecular dynamics simulations provide a basis for designing more advanced functionalized antimicrobial-biomaterials.

Antimicrobial Peptide Mimetics Based on a Diphenylacetylene Scaffold: Synthesis, Conformational Analysis, and Activity.

Mimics of natural antimicrobial peptides are promising compounds to fight the rising threat of multi-drug resistant bacteria. Here we report the design, synthesis and conformational analysis of a new class of antimicrobial peptide mimetics incorporating a diphenylacetylene scaffold. Within a small set of compounds, we observe a correlation between amphiphilicity, the efficiency of partitioning into negatively charged membranes and antibacterial activity. The most amphiphilic compound, which contains four isoleucine residues and four lysine residues, displays species-selective antibacterial activity (most active against Bacillus subtills) and low haemolytic activity. Solution-phase conformational analysis of this compound indicates that a defined structure is adopted in the presence of negatively charged phospholipid membranes and aqueous 2,2,2-trifluoroethanol but not in water. A conformation model indicates that the cationic and hydrophobic functional groups are segregated. These results may inform the development of highly selective antimicrobial peptide mimetics for therapeutic applications.

Crown ether modified peptides: Length and crown ring size impact on membrane interactions

Antimicrobial peptides are widely studied as an alternative to traditional antibiotics. However, they are difficult to develop, as multiple factors influence their potency and selectivity toward bacterial cells. In this paper, we investigate three simplified model peptides that bear crown ethers, and the effects of simple structural modifications (peptide length and crown ether ring size) on their secondary structures and their permeabilizing activity on living cells and model membranes made with egg yolk phosphatidylcholine or 1-palmitoyl-2-oleoylphosphatidylglycerol. Circular dichroism studies show that the peptide length and the crown ether ring size do influence the conformation, but no trend could be determined from the results. Permeabilization studies with model membranes and with red blood cells demonstrated that from 13 residues to 16 residues, there is a gradual increase in activity as the peptides get longer. However, the shortest tested analogs, with 12 residues, also exhibited an increase in activity caused by the removal of one amino acid that was bearing a crown ether. Permeabilization assays showed that larger ring size analogs showed higher hemolytic activities. Altogether, the results reported help design new and more selective antimicrobial peptides.

Selectivity of Antimicrobial Peptides: A Complex Interplay of Multiple Equilibria.

Antimicrobial peptides (AMPs) attack bacterial membranes selectively, killing microbes at concentrations that cause no toxicity to the host cells. This selectivity is not due to interaction with specific receptors but is determined by the different lipid compositions of the membranes of the two cell types and by the peculiar physicochemical properties of AMPs, particularly their cationic and amphipathic character. However, the available data, including recent studies of peptide-cell association, indicate that this picture is excessively simplistic, because selectivity is modulated by a complex interplay of several interconnected phenomena. For instance, conformational transitions and self-assembly equilibria modulate the effective peptide hydrophobicity, the electrostatic and hydrophobic contributions to the membrane-binding driving force are nonadditive, and kinetic processes can play an important role in selective bacterial killing in the presence of host cells. All these phenomena and their bearing on the final activity and toxicity of AMPs must be considered in the definition of design principles to optimize peptide selectivity.

Antibacterial isoamphipathic oligomers highlight the importance of multimeric lipid aggregation for antibacterial potency

Cationic charge and hydrophobicity have long been understood to drive the potency and selectivity of antimicrobial peptides (AMPs). However, these properties alone struggle to guide broad success in vivo, where AMPs must differentiate bacterial and mammalian cells, while avoiding complex barriers. New parameters describing the biophysical processes of membrane disruption could provide new opportunities for antimicrobial optimization. In this work, we utilize oligothioetheramides (oligoTEAs) to explore the membrane-targeting mechanism of oligomers, which have the same cationic charge and hydrophobicity, yet show a unique ~ 10-fold difference in antibacterial potency. Solution-phase characterization reveals little difference in structure and dynamics. However, fluorescence microscopy of oligomer-treated Staphylococcus aureus mimetic membranes shows multimeric lipid aggregation that correlates with biological activity and helps establish a framework for the kinetic mechanism of action. Surface plasmon resonance supports the kinetic framework and supports lipid aggregation as a driver of antimicrobial function.

Peptidoglycan potentiates the membrane disrupting effect of the carboxyamidated form of DMS-DA6, a Gram-positive selective antimicrobial peptide isolated from Pachymedusa dacnicolor skin

The occurrence of nosocomial infections has been on the rise for the past twenty years. Notably, infections caused by the Gram-positive bacteria Staphylococcus aureus represent a major clinical problem, as an increase in antibiotic multi-resistant strains has accompanied this rise. There is thus a crucial need to find and characterize new antibiotics against Gram-positive bacteria, and against antibiotic-resistant strains in general. We identified a new dermaseptin, DMS-DA6, produced by the skin of the Mexican frog Pachymedusa dacnicolor, with specific antibacterial activity against Gram-positive bacteria. This peptide is particularly effective against two multiple drug-resistant strains Enterococcus faecium BM4147 and Staphylococcus aureus DAR5829, and has no hemolytic activity. DMS-DA6 is naturally produced with the C-terminal carboxyl group in either the free or amide forms. By using Gram-positive model membranes and different experimental approaches, we showed that both forms of the peptide adopt an α-helical fold and have the same ability to insert into, and to disorganize a membrane composed of anionic lipids. However, the bactericidal capacity of DMS-DA6-NH2 was consistently more potent than that of DMS-DA6-OH. Remarkably, rather than resulting from the interaction with the negatively charged lipids of the membrane, or from a more stable conformation towards proteolysis, the increased capacity to permeabilize the membrane of Gram-positive bacteria of the carboxyamidated form of DMS-DA6 was found to result from its enhanced ability to interact with peptidoglycan.

Partial Purification Characterization and Application of Bacteriocin from Bacteria Isolated Parkia biglobosa Seeds

Bacteriocins are proteinaceous toxins produced by bacteria to inhibit the growth of similar or closely related bacterial strains. Fermented Parkia biglobosa seeds (African locust bean) were screened for bacteriocin-producing lactic acid bacteria (LAB) with the characterization of putative bacteriocins. Bacteriocin-producing lactic acid bacteria (LAB) were identified by 16s rDNA sequencing. Molecular sizes of the bacteriocins were determined using the tricine-sodium dodecyl sulphate-polyacrylamide gel electrophoresis (tricine-SDS–PAGE) and effects of enzymes, pH, detergents and temperature on bacteriocin activity investigated, using standard procedures. Bacteriocins production and activities were measured by spectrophotometric analysis. Statistical analysis was carried out using student t-test and Analyses of Variance. Bacteriocigenic LAB isolated were Lactobacillus plantarum Z1116, Enterococcus faecium AU02 and Leuconostoc lactis PKT0003. They inhibited the growth of both Gram-positive and Gram-negative bacteria. The sizes of bacteriocins Z1116, AU02 and PKT0003 were 3.2 kDa, 10 kDa and 10 kDa, respectively. The synergistic effects of characterized bacteriocins and rifampicin tested on organisms showed significant differences (P < 0.05), as compared with the effects of only one of the two. The antimicrobial activity of the three bacteriocins was deactivated after treatment of the cell-free supernatants with proteinase K, papain, pepsin and trypsin. Parkia biglobosa seeds are, therefore, rich in LAB bacteriocins which could be explored. The biosynthetic mechanisms of LAB bacteriocins could be employed in food safety and security, preservation, peptide design, infection control and pharmacotherapy. This should help in the control of undesirable bacteria and in designing more potent and selective antimicrobial peptides.

Peptide consensus sequence determination for the enhancement of the antimicrobial activity and selectivity of antimicrobial peptides.

The rise of multidrug-resistant bacteria is causing a serious threat to the world’s human population. Recent reports have identified bacterial strains displaying pan drug resistance against antibiotics and generating fears among medical health specialists that humanity is on the dawn of entering a post-antibiotics era. Global research is currently focused on expanding the lifetime of current antibiotics and the development of new antimicrobial agents to tackle the problem of antimicrobial resistance. In the present study, we designed a novel consensus peptide named “Pepcon” through peptide consensus sequence determination among members of a highly homologous group of scorpion antimicrobial peptides. Members of this group were found to possess moderate antimicrobial activity with significant toxicity against mammalian cells. The aim of our design method was to generate a novel peptide with an enhanced antimicrobial potency and selectivity against microbial rather than mammalian cells. The results of our study revealed that the consensus peptide displayed potent antibacterial activities against a broad range of Gram-positive and Gram-negative bacteria. Our membrane permeation studies displayed that the peptide efficiently induced membrane damage and consequently led to cell death through the process of cell lysis. The microbial DNA binding assay of the peptide was found to be very weak suggesting that the peptide is not targeting the microbial DNA. Pepcon induced minimal cytotoxicity at the antimicrobial concentrations as the hemolytic activity was found to be zero at the minimal inhibitory concentrations (MICs). The results of our study demonstrate that the consensus peptide design strategy is efficient in generating peptides.

Mechanism of Four de Novo Designed Antimicrobial Peptides

As pathogenic bacteria become resistant to traditional antibiotics, alternate approaches such as designing and testing new potent selective antimicrobial peptides (AMP) are increasingly attractive. However, whereas much is known regarding the relationship between the AMP sequence and potency, less research has focused on developing links between AMP properties, such as design and structure, with mechanisms. Here we use four natural AMPs of varying known secondary structures and mechanisms of lipid bilayer disruption as controls to determine the mechanisms of four rationally designed AMPs with similar secondary structures and rearranged amino acid sequences. Using a Quartz Crystal Microbalance with Dissipation, we were able to differentiate between molecular models of AMP actions such as barrel-stave pore formation, toroidal pore formation, and peptide insertion mechanisms by quantifying differential frequencies throughout an oscillating supported lipid bilayer. Barrel-stave pores were identified by uniform frequency modulation, whereas toroidal pores possessed characteristic changes in oscillation frequency throughout the bilayer. The resulting modes of action demonstrate that rearrangement of an amino acid sequence of the AMP resulted in identical overall mechanisms, and that a given secondary structure did not necessarily predict mechanism. Also, increased mass addition to Gram-positive mimetic membranes from AMP disruption corresponded with lower minimum inhibitory concentrations against the Gram-positive Staphylococcus aureus.

Aspirin Triggered Lipoxin A4 regulates NF-κB and induces antimicrobial peptides expression in airway epithelial cells to limit mucosal inflammation to enhance host response in pneumonia

Abstract Bacteria induced pneumonia remains a major global threat to health. Even with effective antibiotic treatment, the host response can be exuberant, resulting in tissue damage and morbidity. Resolution of inflammation is an active process governed in part by specialized pro-resolving mediators (SPMs). We hypothesized that the SPM, aspirin triggered lipoxin A4 (ATL) engaged airway mucosal epithelial cells to enhance resolution. In a murine model of bacterial aspiration pneumonia for E. coli, ATL (100 ng, iv) decreased the lung bacterial burden in an additive manner with ciprofloxacin (0.2 mg/kg, ip). Further, ATL also significantly limited neutrophils recruitment and decreased pro-inflammatory cytokine (IL-1α) and chemokine production (KC). These ATL mediated protective responses were mediated in part via regulation of NF-kB activation. In response to LPS and E. coli in vitro, ATL selectively influenced airway epithelial cells expression of NF-kB regulators A20, SIGIRR and ST2. In an in vitro bacteria killing assay by airway epithelial cells, ATL directly limited bacterial growth by increasing select antimicrobial peptides expression (LL37 and BPI). mCRAMP deficient mice (human homolog of LL37) failed to limit lung bacterial growth in E. coli pneumonia and ATL treatment was significantly less efficient in promoting host defense in these mice. In conclusion, the SPM ATL can limit pathogen mediated mucosal inflammation and enhance host defense in pneumonia via the selective regulation of NF-κB and antimicrobial peptides in airway epithelial cells. These findings with ATL provide evidence for SPM targeted treatment of the host response to pneumonia that has the potential for therapeutic benefit in combination with antibiotics.

Selectivity of Antimicrobial Peptides: Association to Bacterial and Eukaryotic Cells and Cell-Density Dependence

Several questions limit the applicability of antimicrobial peptides (AMPs) as a new class of antibiotics against the problem of multidrug resistant bacteria.AMPs are screened for their direct in vitro bactericidal activity, mediated by association to and perturbation of bacterial membranes. However, they act also as immunomodulators, and it is debated which function is predominant at peptide and bacteria concentrations relevant for in vivo conditions.In vitro, AMPs are toxic to mammalian cells only at concentrations higher than those needed for bactericidal activity. This selectivity is presumably determined by the difference in lipid composition of membranes of the two cell types, as studies on liposomes show a higher affinity for bilayers mimicking bacterial membranes. However, it might be just an experimental artifact of the in vitro assay conditions that employ lower cell densities for bacteria than for red blood cells (RBCs), which are also larger.Recently, by fluorescence spectroscopy we characterized the association of the cathelicidin PMAP-23 to bacterial cells, and determined for the first time the number of membrane-bound peptide molecules necessary to kill a bacterial cell (106 peptides per E. Coli cell) [ACS Chem. Biol. 2014 9:2003]. We now measured the dependence of killing activity on bacterial cell density: as a consequence of the binding equilibrium, total micromolar peptide concentrations are needed even with extremely low bacterial counts. We also extended our method to measure peptide binding to RBCs, showing that also in this case a high coverage of the cell surface is needed to cause lysis. Finally, we investigated the cell-density dependence of hemolytic activity.Overall, these data clarify the complex dependence of AMP activity and selectivity on the density of bacterial and human cells, and open new questions for their behavior in vivo.

The Role of Thermodynamics in the Activity and Selectivity of Antimicrobial Peptides

Antimicrobial peptides (AMPs) are natural bactericidals and immunomodulators, and their action involves association to cell membranes and their perturbation. In vitro, they are selective for bacteria versus eukaryotic cells. Although AMPs are promising compounds to fight drug-resistant bacteria, many unanswered questions hinder their therapeutic application: is their main role bactericidal or immunomodulatory? What are the structural determinants of selectivity, other than their cationic charge? Is cell selectivity real or an artifact of the assay conditions?[1] By combining spectroscopic experiments on liposomes and cells and molecular dynamics simulations, we characterized how AMP activity and selectivity are regulated by multiple thermodynamic equilibria: membrane binding, aggregation, conformational transitions and orientation in the bilayer.[2] By measuring peptide association to bacteria and erythrocytes, we found that high coverage of the cell surface is needed to cause its death/lysis, showing that the carpet mechanism is relevant in real cells. Due to the binding equilibrium, the total peptide concentration in solution necessary to reach this threshold never decreases below micromolar values, even at low cell densities. These data indicate that immunomodulation might prevail where these concentrations are not reached, and provide a new insight in the cell density dependence of selectivity. In addition, we showed that peptide folding or aggregation in solution are ways to increase peptide selectivity, by shielding the hydrophobic residues and reducing the hydrophobic-driving force for binding to the neutral surface of eukaryotes.
