Sustainable seedling pots: Development and characterisation of banana waste and natural fibre-reinforced composites for horticultural applications

Plastic pots used in horticultural nurseries generate substantial waste, causing environmental pollution. This study aimed to develop biodegradable composites from banana pseudo-stem reinforced with agricultural residues like pineapple leaves, taro and water hyacinth as eco-friendly substitutes. The aim of this study is to develop optimised banana biocomposite formulations with suitable reinforcements that balance mechanical durability, biodegradation, and seedling growth promotion properties to serve as viable eco-friendly alternatives to plastic seedling pots. This study was carried out by fabricating banana fibre mats through pulping, drying and hot pressing. Composite sheets were reinforced with 50 % pineapple, taro or water hyacinth fibres. The mechanical properties (tensile, yield strength, elongation, bursting strength), hydrophilicity (contact angle, water absorption), biodegradability (soil burial test), and seedling growth promotion were evaluated through appropriate testing methods. The results show that banana-taro composites exhibited suitable tensile strength (25 MPa), elongation (27 %), water uptake (41 %) and 82 % biodegradation in 60 days. It was observed that biodegradable seedling trays fabricated from banana-taro composite showed 95 % tomato seed germination and a 125 cm plant height increase in 30 days, superior to plastic trays. The finding shows that the study demonstrates the potential of banana-taro biocomposites as alternatives to plastic nursery pots, enabling healthy seedling growth while eliminating plastic waste pollution through biodegradation.

Comparative Effect of Seed Coating and Biopriming of Bacillus aryabhattai Z-48 on Seedling Growth, Growth Promotion, and Suppression of Fusarium Wilt Disease of Tomato Plants.

Beneficial plant microbes can enhance the growth and quality of field crops. However, the benefits of microbes using cheap and efficient inoculation methods are still uncommon. Seed coating with biocontrol agents can reduce the amount of inocula along with having the potential for large-scale application. Hence, in this research work, the comparative potential of tomato seed coating and biopriming with Bacillus aryabhattai Z-48, harboring multiple plant-beneficial traits, to suppress Fusarium wilt disease along with its beneficial effect on seedling and plant growth promotion was analyzed. Among two bacterial strains, B. aryabhattai Z-48 was able to antagonize the mycelial growth of Fusarium oxysporum f.sp. lycopersici in vitro and its application as a seed coating superiorly benefited seedling traits like the germination percentage, vigor index, and seedling growth index along with a reduced germination time. The seed coating with B. aryabhattai Z-48 resulted in significant increases in the shoot length, root length, dry biomass, and total chlorophyll contents when compared with the bioprimed seeds with the same bacterial strain and non-inoculated control plants. The seed coating with B. aryabhattai Z-48 significantly reduced the disease index (>60%) compared with the pathogen control during pot trials. Additionally, the seed coating with B. aryabhattai Z-48 resulted in a significantly higher production of total phenolics, peroxidase, polyphenol oxidase, and phenylalanine ammonia lyase enzyme in tomato plants. The GC/MS-based non-targeted metabolic profiling indicated that the seed coating with B. aryabhattai Z-48 could cause large-scale metabolite perturbations in sugars, sugar alcohols, amino acids, and organic acids to increase the fitness of tomato plants against biotic stress. Our study indicates that a tomato seed coating with B. aryabhattai Z-48 can improve tomato growth and suppress Fusarium wilt disease effectively under conventional agricultural systems.

Antifungal and Plant-Growth Promotion Effects of Bacillus velezensis When Applied to Coastal to Pine (Pinus thunbergii Parl.) Seedlings

Fungal diseases such as root rot and leaf blight cause substantial losses in coastal pine (Pinus thunbergii Parl.) seedling production, which hinders afforestation/forest restoration programs. We isolated and identified Fusarium oxysporum and Alternaria alternata as the causal agents of root rot and needle blight diseases and investigated the biocontrol efficacy against the fungal pathogens and growth promotion of coastal pine seedlings using Bacillus velezensis CE 100. The bacterium produced the hydrolytic enzymes chitinase, β-1,3-glucanase, and protease enzymes, and the crude enzyme fraction of the biocontrol strain caused the deformation of the fungal cell wall and antagonized F. oxysporum and A. alternata, causing respective inhibition of spore germination by 91.0% and 85.9% and mycelial growth by 58.3% and 54.3%, at a concentration of 1000 µL/mL. Consequently, the bacterial treatment improved the survival rate of seedlings 1.9 times relative to the control group. The bacterium secreted indole-acetic acid (IAA) phytohormone and enhanced root growth and absorption of nutrients, which notably enhanced the biomass production of coastal pine seedlings. Therefore, these results provide evidence that B. velezensis CE 100 is an effective antifungal and growth-promoting bacterium that can facilitate the production of high-quality coastal pine seedlings for the restoration and establishment of coastal forests.

Photosynthetically active radiation is required for seedling growth promotion by volcanic dacitic tuff breccia (Azomite).

A plant's growth and development are shaped by its genome and the capacity to negotiate its environment for access to light, water, and nutrients. There is a vital need to understand the interactions between the plant, its physical environment, and the fertilizers used in agriculture. In this study, a commercially available volcanic ash fertilizer, Azomite®, characterized as dacitic (rhyolitic) tuff breccia, was tested for its effect on promoting early seedling vigor. Early growth and photomorphogenesis processes are well studied in Arabidopsis. Seedling assays under different light conditions were used to dissect the underlying mechanisms involved. These assays are well established and can be translated to agriculturally important crop plants. The volcanic ash fertilizer was tested at different concentrations on seedlings grown on basic media lacking sucrose either in continuous darkness (Dc), continuous Red (Rc), Far-Red (FRc), or White Light (WLc). Micronutrients in the volcanic ash significantly increased seedling growth under Rc and WLc, but not under Dc and FRc, indicating that photosynthetically active radiation was required for the observed growth increase. Furthermore, red-light photoreceptor mutant, phyB-9, lacked the growth response, and higher amount of fertilizer reduced growth in all conditions tested. These data suggest that light triggers the ability of the seedling to utilize micronutrients in volcanic ash in a dose-dependent manner. The methods described here can be used to establish mechanisms of activity of various nutrient inputs and, coupled with whole-genome expression profiling, can lead to better insights into optimizing nutrient field applications to improve crop production.

The Small Auxin-Up RNA SAUR10 Is Involved in the Promotion of Seedling Growth in Rice.

Small auxin-up-regulated RNAs (SAURs) are genes rapidly activated in response to auxin hormones, significantly affecting plant growth and development. However, there is limited information available about the specific functions of SAURs in rice due to the presence of extensive redundant genes. In this study, we found that OsSAUR10 contains a conserved downstream element in its 3' untranslated region that causes its transcripts to be unstable, ultimately leading to the immediate degradation of the mRNA in rice. In our investigation, we discovered that OsSAUR10 is located in the plasma membrane, and its expression is regulated in a tissue-specific, developmental, and hormone-dependent manner. Additionally, we created ossaur10 mutants using the CRISPR/Cas9 method, which resulted in various developmental defects such as dwarfism, narrow internodes, reduced tillers, and lower yield. Moreover, histological observation comparing wild-type and two ossaur10 mutants revealed that OsSAUR10 was responsible for cell elongation. However, overexpression of OsSAUR10 resulted in similar phenotypes to the wild-type. Our research also indicated that OsSAUR10 plays a role in regulating the expression of two groups of genes involved in auxin biosynthesis (OsYUCCAs) and auxin polar transport (OsPINs) in rice. Thus, our findings suggest that OsSAUR10 acts as a positive plant growth regulator by contributing to auxin biosynthesis and polar transport.

Bacillus Strains with Catalase Enzyme Improve the Physiology and Growth of Rice (Oryza sativa L.)

Catalase can decompose hydrogen peroxide in plants under controlled and stressed conditions. In a stressed environment, an increase in hydrogen peroxide production makes catalase activity a major defense system for plant growth. The current study was conducted to evaluate the catalase activity of the pre-isolated and identified bacterial strains Bacillus aryabhattai (AN30), Bacillus megaterium (AN24), Bacillus megaterium (AN31) and Bacillus megaterium (AN35) and their potential for rice seedling growth promotion. These strains were characterized for quantitative catalase, urease, siderophore and exopolysaccharide production using LB media. Subsequently, the effectiveness of these strains was checked by quantifying the catalase activity in the rhizosphere, roots and shoots of rice seedlings. The secretion of organic and phenolic compounds produced by the tested strains in liquid culture was also investigated. Plant growth parameters were also studied in a growth room trial. Our results showed that the strain AN24 showed the maximum catalase activity (1.36 mol cm−1), urease activity (1.35 mol cm−1) and exopolysaccharide (4.20 µg mL−1) and siderophore (2.32%) production in LB media. All tested strains showed significantly higher catalase activity in soil compared to the control. Among sole applications, strain AN24 showed better results; however, the consortium application of strains AN24 + AN30 + AN35 + AN31 showed the maximum improvement in dry biomass, shoot and root length, and increase in catalase activity of rice seedlings. The results showed that a consortium of these Bacillus strains with catalase activity has greater potential to enhance the antioxidant defense system and growth promotion of rice seedlings. However, further experimentation under natural conditions is required before using these strains as potential bioinoculants for improving rice growth and yield.

Assessing the functional diversity of rhizobacteria from cacao by partitioning root and shoot biomasses.

Plant-microbe interactions are critical for the sustainability of agricultural production. In this study, our aims were to characterize the genetic and functional diversity of the culturable bacterial community associated with the cacao rhizosphere and access their potential for growth promotion of cacao seedling. Culture-dependent and molecular methods were used to characterize the population densities and diversity of bacterial communities from soil and cacao plants at two locations and two plant ages. A total of 63 strains were identified through hsp60 sequencing. Pseudomonas and Enterobacter were the most abundant genera in association with the cacao rhizosphere, whereas Bacillus was more numerous in soil. Parameters of seedling growth promotion were evaluated 60days after inoculation of seeds, with partition of the assessments into root and shoot weight. Each isolate showed beneficial, neutral or deleterious effects on plant growth, depending on the isolate and on the parts of plant assessed. Interestingly, although an apparent overall decrease in total biomass of seedlings (roots + shoots dry matters) was observed for the majority of isolates (89%), 94% of all isolates, in fact, revealed an increase in plant roots/shoots dry biomass ratio. Despite that part of the isolates (35%) appeared to significantly decrease plant height, and that 65% did not influence plant height (neutral effect), 18 had significantly increased root dry biomass; nevertheless, seven of these root growth-increasing isolates simultaneously decreased shoots-related growth parameters. The results of this study evidentiated the functional diversity of culturable cacao rhizobacteria and how the partitioning of roots and shoots in the assessment of plant growth parameters could reveal the biotechnological potential of these isolates for promoting growth of clones for rehabilitation of commercial cacao plantations. New insight on tomato seed priming with Anabaena minutissima phycobiliproteins in relation to Rhizoctonia solani root rot resistance and seedling growth promotion

AbstractCyanobacteria phycobiliproteins (PBPs) are already exploited in the food industries and for biotechnological applications but not in the agricultural field. Different concentrations (0.6 – 4.8 mg/mL) of Anabaena minutissima PBPs were applied to tomato seed to study their priming effect against the soil-borne fungal pathogen Rhizoctonia solani and in promoting plant growth. PBPs increased seedling emergence and vigour, showed activity against root rot disease (67%), and enhanced plant dry weight, length, and height. Generally, no dose effect has been observed except for dry weight (55% at 4.8 mg/mL). Seed treatment primed seeds and seedlings by leading to the activation of defence responses raising phenol (26% in hypocotyls) and flavonoid (26 and 45% in hypocotyls and epicotyls, respectively) contents and chitinase (4-fold at 2.4 and 4.8 mg/mL in hypocotyls) and β-1,3-D-glucanase (up to about 2-fold at all doses in epicotyls) activities. Micro-Attenuated Total Reflection Fourier Transform Infrared revealed changes in functional groups of primed seeds, hypocotyls and exudates released into the agar because of treatment. Protein extract from PBP-primed seedlings inhibited mycelial growth (67% for epicotyl proteins) and caused morphological alterations in hyphae. This research emphasizes the potential priming role of PBPs applied by seed treatment against soil-borne pathogens.

Bacillus-Loaded Biochar as Soil Amendment for Improved Germination of Maize Seeds.

Biochar is considered one of the most promising long-term solutions for soil quality improvement, representing an ideal environment for microorganisms' immobilization. Hence there is a possibility to design microbial products formulated using biochar as a solid carrier. The present study was aimed at development and characterization of Bacillus-loaded biochar to be applied as a soil amendment. The producing microorganism Bacillus sp. BioSol021 was evaluated in terms of plant growth promotion traits, indicating significant potential for production of hydrolytic enzymes, indole acetic acid (IAA) and surfactin and positive tests for ammonia and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase production. Soybean biochar was characterised in terms of physicochemical properties to evaluate its suitability for agricultural applications. The experimental plan for Bacillus sp. BioSol021 immobilisation to biochar included variation of biochar concentration in cultivation broth and adhesion time, while the soil amendment effectiveness was evaluated during maize germination. The best results in terms of maize seed germination and seedling growth promotion were achieved by applying 5% of biochar during the 48 h immobilisation procedure. Germination percentage, root and shoot length and seed vigour index were significantly improved when using Bacillus-biochar soil amendment compared to separate treatments including biochar and Bacillus sp. BioSol021 cultivation broth. The results indicated the synergistic effect of producing microorganism and biochar on maize seed germination and seedling growth promotion, pointing out the promising potential of this proposed multi-beneficial solution for application in agricultural practices.

Isolation and Characterization of Plant Growth promoting Rhizobacteria from Bamboo Rhizosphere and Their Role in Plant Growth Promotion

Plant growth-promoting rhizobacteria (PGPR) are a group of root-associated bacteria that intimately interact with the plant roots and consequently enhance growth by extemporising nutrient retrieval or phytohormone production. We isolated and screened indigenous phosphate solubilising and auxinproducing PGPR from bamboo rhizosphere. 66 soil samples from bamboo (Bambusa nutans subsp. cupulata, B. balcooa and B. tulda) rhizospheres were collected from Dhanusha, Mahottari and Sarlahi districts, Nepal. 120 isolates of PGPR were obtained by serial dilution method in (PVK) agar and Luria Bertani agar. 92 out of 120 isolates of PGPR with the ability to solubilise phosphate were selected based on the halo colony ratio in PVK agar medium and auxin production in Luria Bertani agar. Among them, six isolates having high phosphate solubilising index and high production capacity of indole-3-acetic acid were further screened. Biochemical analysis revealed that these isolates belonged to the genus Pseudomonas. Phosphate solubilising index and indole-3- acetic acid production by six isolates ranged from 4.19±0.8 to 7.65±1.3, and IAA production ranged from 72.93±0.2 to 82.48±0.9µg/ml respectively. These isolates significantly increased shoot length (13.26±0.56cm), shoot fresh weight (16.26±1.02mg), shoot dry weight (10.56±0.09mg), root length (4.9±0.5cm), root fresh weight (7.56±1.05mg), root dry weight (3.21±0.01mg), and chlorophyll ‘a’ and chlrophyll‘b’ and carotenoid (2.16±0.01mg/g, 1.19±0.06mg/g and 0.92±0.01mg/g respectively) of B. nutans subsp. cupulata seedlings. This study suggests that PGPR isolated from bamboo rhizosphere demonstrated outstanding contribution to the growth promotion of seedlings of B. nutans subsp. cupulata as compared to negative control.

Orchid mycorrhiza: Isolation, culture, characterization and application

In nature all orchids develop a mutualistic association with specialized group of fungi called orchid mycorrhiza. In natural environment, orchids are exclusively reliant on mycorrhizal fungi for seed germination, establishment, growth and development. In the early developmental stages, all orchids are mycoheterotrophic, and in most cases the association subsists throughout their lifespan. The significance of orchid mycorrhizal fungi (OMF) for seed germination, nourishment, growth promotion, and survival has been established more than a century ago and tremendous advancements were made in recent decades. Till date, understanding the mode of interaction between orchids and mycorrhizal fungi and determining the architecture of interaction networks are major challenges in ecology and evolutionary biology. The development of molecular techniques has empowered us to unscramble the fungal diversity comprised in the symbiosis and has facilitated us to overcome the difficulties associated with the conventional identification of OMF and orchid-fungal interactions. In addition, the application of modern biotechnological approaches resolved many controversial issues of OMF. Recently, OMF are used for biological hardening and growth promotion of in vitro raised seedlings, early flowering and quality improvement of flowers, and diseases and pests management. The aim of this communication is to update and summarize orchid mycorrhizal research including isolation, culture, and characterization of OMF, understanding the mechanisms of orchid-fungus symbiosis, and application of OMF in seed germination, growth promotion, and survival of asymbiotically raised seedlings and conservation of orchids. This appraisal will convey valuable insights to orchidologists engaged in biology, propagation, and conservation of orchids.

Designing synthetic consortia of Trichoderma strains that improve antagonistic activities against pathogens and cucumber seedling growth

BackgroundTrichoderma spp. are important agricultural biocontrol microorganisms that are often used as effective components of microbial fungicides and microbial biofertilizers. However, most of these products are prepared by a single strain in monoculture, which significantly limits the biocontrol efficiency and stability of Trichoderma products. Therefore, the establishment of a design and screening approach for consortia with multi-Trichoderma strains for co-culture is of great importance to overcome the shortage of traditional Trichoderma biocontrol products.ResultsFirst, 15 Trichoderma strains were screened in terms of mycelium growth rate, antagonistic activity to a variety of pathogens, stress tolerance to high temperature and salt stress, and cucumber seedling growth promotion level. Then, the combinations of Trichoderma asperellum GDSF1009 (CGMCC NO. 9512), Trichoderma asperelloides Z4-1 (CGMCC NO. 40245), Trichoderma harzianum 10569 (CGMCC NO. 40246), and T. asperellum 10264 (CGMCC NO. 22404) were finally screened as an optimal consortium for co-culture underlying the levels of plant growth-promoting and antagonistic activity to Fusarium oxysporum and seed germination promotion relative to the monoculture of a single strain. Consortia with multiple co-cultured strains were found to generate larger amounts of free amino acids than those from the monoculture of a single strain, and a pot assay also indicated that metabolites of co-cultures were able to promote cucumber seedling growth superior to that with monoculture of a single strain, even though the promotion was better than from simply mixed cultures from each of the four Trichoderma strains. Taken together, the co-culture consortia composed of the four compatible interactive Trichoderma strains was a potential novel multiple strain biocontrol agent based on the combination of synthetic consortia design and co-culture. In the field experiment, we found that the growth-promoting effect of the co-culture fermentation filtrate was better than that of the single culture fermentation filtrate. Compared with T-Z4-1, T-1009, T-10264 and T-10569, the plant height of cucumber was increased by 22.99%, 42.06%, 24.18% and 30.09%, respectively, and the stem diameter was increased by 16.59%, 18.83%, 13.65% and 14.70%, respectively.ConclusionAn approach to designing and screening Trichoderma consortia for co-culture was established. The consortia co-culture presented a better performance in antagonistic activity and cucumber growth compared with a monoculture of a single strain. Thus, it is of great significance to lay the foundation for the creation of a novel Trichoderma biofungicide or biomanure to resist cucumber Fusarium wilt and promote cucumber growth.

Seedling growth promotion and potential biocontrol against phytopathogenic Fusarium by native rhizospheric Pseudomonas spp. strains from Amarillo Zamorano maize landrace

Maize is one of the most important staple crops around the word. In Mexico, Los Altos de Jalisco is an agricultural region with the highest production of maize with rainfed irrigation, an agronomic management based on the use of no longer sustainable agrochemicals, and sowing of coated seeds of different commercial origin. Nowadays, the microbiota associated to the rhizosphere of native landraces is considered a reservoir of microbial resources with the potential to promote plant growth and control diseases. With the aim to find ecofriendly and cost-effective alternatives that can reduce the use of agrochemicals at seed germination and seedling growth of maize crop in Los Altos de Jalisco, this study focused on the search of fluorescent Pseudomonas strains as germination and seedlings growth promoters and biocontrol agents of fusariosis in maize, and highlight the importance of Mexican maize landraces as natural reservoirs of plant growth-promoting microorganisms. Samples of rhizospheric soil and seeds were collected from ten maize landraces and a pure line. Pseudomonas and Fusarium strains were isolated and selected by its antagonistic and pathogenic potential, respectively. Pseudomonas strains were phylogenetically identified by a 16S rRNA gene analysis, and Fusarium strains by the phylogenetic analysis of ITS region. The potential of Pseudomonas strains to promote seed germination and seedling/plant growth was explored through metabolic profiles and bioassays to evaluate bacteria-plant-fungus interactions. A total of 57 Fusarium spp. strains were grouped into four different Fusarium species complexes and 14 fluorescent Pseudomonas strains were classified into four Pseudomonas fluorescens subgroups. Pseudomonas strains displayed plant growth promotion and biocontrol capacity by increasing 38%–66% the dry weight of maize seedlings (p < 0.05), 23%–63% growth-inhibition of different Fusarium strains in dual cultures, and a range of 4.5–20 inhibition score based on a rate scale when inoculated in seeds. Pseudomonas spp. metabolic profiles were diverse, but all the strains proved to be innocuous, and Pseudomonas sp. 4L, 10L and 11L showed desirable enzymatic capacities from an agrobiotechnological approach. Additionally, Pseudomonas sp. 4L increased germination and seedling growth of maize, by increasing 20–51% the number of germinated seeds, 16–38% the length of seedling aerial part, 30–140% seedling length of roots, 15–32% seedling fresh weight, and 21–33% seedling dry weight, and reduced 75–96% the damage generated by Fusarium spp. on naturally infected seeds. This study revealed that Pseudomonas sp. 4L and 11L are regional promising ecofriendly and cost-effective alternatives as adjuvants in the nutritional and phytosanitary management at seed germination and seedling growth of maize crop in Los Altos de Jalisco region, and highlight the importance of the rhizosphere microbiome of Mexican maize landrace as natural reservoir of plant growth-promoting microorganisms.

Exploring the Potential Applications of Paecilomyceslilacinus 112

Microorganisms are widely used to obtain biostimulants that can facilitate the assimilation of nutrients, ensuring high crop yield and quality. A particular category of biostimulants are protein hydrolyzates (PH), obtained from microbial cultures grown on a nutrient medium. In the present study, Paecilomyces lilacinus 112, an endophytic fungus isolated from soil, was tested to determine its effect on the growth promotion of tomato seedlings in greenhouse conditions. Additionally, other beneficial features of the P.lilacinus isolate were evaluated via several tests: antagonism against plant pathogenic fungi, production of secondary useful metabolites, and solubilization of vital micronutrients. Out of the tested pathogens, P.lilacinus exhibited the highest antifungal activity against a Cladosporium isolate (inhibition of 66.3%), followed by Rhizoctonia. solani (52.53%), and Sclerotinia sclerotiorum (50.23%). Paecilomyceslilacinus 112 was able to secrete hydrolytic enzymes and siderophores, and solubilize zinc and phosphorus. In the tomato treatment, the application of PH obtained from fungal cultivation on a feather medium led to the following increases in plant growth parameters: 3.54-fold in plant biomass; 3.26-fold in plant height, 1.28-fold in plant diameter; 1.5-fold in the number of branches/plant; and 1.43-fold in the number of leaves/plant, as compared to water treatment. The application of this isolate can be of benefit to bioeconomy because keratin wastes are valorized and returned, in agriculture, contributing to renewable natural resources.

Evaluation and Genome Analysis of Bacillus subtilis YB-04 as a Potential Biocontrol Agent Against Fusarium Wilt and Growth Promotion Agent of Cucumber.

Cucumber wilt caused by Fusarium oxysporum f.sp. cucumerinum (Foc) is a highly destructive disease that leads to reduced yield in cucumbers. In this study, strain YB-04 was isolated from wheat straw and identified as Bacillus subtilis. It displayed strong antagonistic activity against F. oxysporum f.sp. cucumerinum in dual culture and exhibited significant biocontrol of cucumber Fusarium wilt with a higher control effect than those of previously reported Bacillus strains and displayed pronounced growth promotion of cucumber seedlings. B. subtilis YB-04 could secrete extracellular protease, amylase, cellulose, and β-1,3-glucanase and be able to produce siderophores and indole acetic acid. Inoculation with B. subtilis YB-04 or Foc increased cucumber defense-related enzyme activities for PPO, SOD, CAT, PAL, and LOX. However, the greatest increase was with the combination of B. subtilis YB-04 and Foc. Sequencing the genome of B. subtilis YB-04 showed that it had genes for the biosynthesis of various secondary metabolites, carbohydrate-active enzymes, and assimilation of nitrogen, phosphorous, and potassium. B. subtilis YB-04 appears to be a promising biological control agent against the Fusarium wilt of cucumber and promotes cucumber growth by genomic, physiological, and phenotypic analysis.

Growth Promotion of Phaseolus vulgaris and Arabidopsis thaliana Seedlings by Streptomycetes Volatile Compounds

Streptomyces are recognized as antipathogenic agents and plant-growth-promoting rhizobacteria. The objective of this study was to evaluate the capacities of four antifungal Streptomyces strains to: produce the substances that are involved in plant growth; solubilize phosphates; and fix nitrogen. The effects of the volatile organic compounds (VOCs) that are emitted by these strains on the growth promotion of Arabidopsis thaliana and Phaseolus vulgaris L. (var. Pinto Saltillo) seedlings were also tested. All of the Streptomyces strains produced indole-3-acetic acid (IAA) (10.0 mg/L to 77.5 mg/L) and solubilized phosphates, but they did not fix nitrogen. In vitro assays showed that the VOCs from Streptomyces increased the shoot fresh weights (89–399%) and the root fresh weights (94–300%) in A. thaliana seedlings; however, these effects were less evident in P. vulgaris. In situ experiments showed that all the Streptomyces strains increased the shoot fresh weight (11.64–43.92%), the shoot length (11.39–29.01%), the root fresh weight (80.11–140.90%), the root length (40.06–59.01%), the hypocotyl diameter (up to 6.35%), and the chlorophyll content (up to 10.0%) in P. vulgaris seedlings. 3-Methyl-2-butanol had the highest effect among the ten pure VOCs on the growth promotion of A. thaliana seedlings. The tested Streptomyces strains favored biomass accumulation in A. thaliana and P. vulgaris seedlings.

Aspergillus niger as a Biological Input for Improving Vegetable Seedling Production.

This study evaluated the potential of Aspergillus niger as an inoculant for growth promotion of vegetable seedlings. Seven vegetable species were evaluated in independent experiments carried out in 22 + 1 factorial schemes, with two doses of conidia (102 and 106 per plant) applied in two inoculation methods (seed treatment and in-furrow granular application), plus an uninoculated control. Experiments were carried out in a greenhouse. Growth parameters evaluated were shoot length, stem diameter, root volume, total root length, shoot and root fresh mass, shoot and root dry mass, and total dry mass. Regardless of the dose and inoculation method, seedlings inoculated with A. niger showed higher growth than uninoculated ones for all crops. The highest relative increase promoted by the fungus was observed for aboveground parts, increasing the production of shoot fresh mass of lettuce (61%), kale (40%), scarlet eggplant (101%), watermelon (38%), melon (16%), pepper (92%), and tomato (42%). Aspergillus niger inoculation also increased seedling root growth of lettuce, pepper, scarlet eggplant, watermelon, and tomato. This research shows that A. niger boosts the growth of all analyzed vegetables, appearing as a promising bio-input for vegetable seedling production.

The enhanced phosphorus use efficiency in phosphate-deficient and mycorrhiza-inoculated barley seedlings involves activation of different sets of PHT1 transporters in roots.

Transcriptional activation of subfamily II PHT1 members in roots is associated with the enhanced phosphorus use efficiency and growth promotion of barley seedlings inoculated with Glomus species. The arbuscular mycorrhizal (AM) fungi symbiotic associations in cereal crops are known to regulate growth in cultivar-specific manner and induce phosphate (Pi) transporters (PHT1) in roots. In the present study, we observed that both AM colonization of roots by Glomus species and phosphate starvation enhanced phosphorus use efficiency (PUE) in barley seedlings. Our search for the full complement of PHT1 members in the recently sequenced barley genome identified six additional genes, totaling their number to 17. Both AM colonization and Pi starvation triggered activation of common as well as different PHT1s. Pi starvation led to the robust upregulation of HvPHT1;6.2/6.3 at 7d and weak activation of HvPHT1;1 in shoots at 3d time-point. In roots, only HvPHT1;1, HvPHT1;6.2/6.3, HvPHT1;7, HvPHT1;8, HvPHT1;11.2 and HvPHT12 were induced at least one of the time-points. AM colonization specifically upregulated HvPHT1;11, HvPHT1;11.2, HvPHT1;12 and HvPHT1;13.1/13.2, members belonging to subfamily II, in roots. Sucrose availability seems to be obligatory for the robust activation of HvPHT1;1 as unavailability of this metabolite generally weakened its upregulation under Pi starvation. Intriguingly, lack of sucrose supply also led to induction of HvPHT1;5, HvPHT1;8, and HvPHT1;11.2 in either roots or shoot or both. The mRNA levels of HvPHT1;5 and HvPHT1;11.2 were not severely affected under combined deficiency of Pi and sucrose. Taken together, this study not only identify additional PHT1 members in barley, but also ascertain their AM, Pi and sucrose-specific transcript accumulation. The beneficial role of AM fungi in the promotion of PUE and barley seedlings' growth is also demonstrated.

Rice Seedling Growth Promotion by Biochar Varies With Genotypes and Application Dosages.

While biochar use in agriculture is widely advocated, how the effect of biochar on plant growth varies with biochar forms and crop genotypes is poorly addressed. The role of dissolvable organic matter (DOM) in plant growth has been increasingly addressed for crop production with biochar. In this study, a hydroponic culture of rice seedling growth of two cultivars was treated with bulk mass (DOM-containing), water extract (DOM only), and extracted residue (DOM-free) of maize residue biochar, at a volumetric dosage of 0.01, 0.05, and 0.1%, respectively. On seedling root growth of the two cultivars, bulk biochar exerted a generally negative effect, while the biochar extract had a consistently positive effect across the application dosages. Differently, the extracted biochar showed a contrasting effect between genotypes. In another hydroponic culture with Wuyunjing 7 treated with biochar extract at sequential dosages, seedling growth was promoted by 95% at 0.01% dosage but by 26% at 0.1% dosage, explained with the great promotion of secondary roots rather than of primary roots. Such effects were likely explained by low molecular weight organic acids and nanoparticles contained in the biochar DOM. This study highlights the importance of biochar DOM and crop genotype when evaluating the effect of biochar on plants. The use of low dosage of biochar DOM could help farmers to adopt biochar technology as a solution for agricultural sustainability.

24-epibrassinolide-induced growth promotion of wheat seedlings is associated with changes in the proteome and tyrosine phosphoproteome.

Brassinosteroids (BRs) represent a unique class of steroidal plant hormones that display pronounced growth-promoting activity at very low concentrations. Although many efforts have been made to characterize the molecular basis of BR action, little is known about the mechanisms behind the growth-promoting effect of BRs at protein level. Proteomic analysis of response to the steroid plant hormone 24-epibrassinolide (EBR) in wheat seedling shoots (Triticum aestivum L.) was performed using two-dimensional electrophoresis (2-DE) and immunoblotting with highly specific antibodies (PY20) to phosphotyrosine. EBR-modulated proteins and phosphotyrosine polypeptides were identified using MALDI-TOF mass spectrometry. The study revealed that EBR-stimulated growth of wheat seedlings was accompanied by changes in the content of multiple proteins as well as in tyrosine phosphorylation of numerous polypeptides. Among them, 22 differentially accumulated proteins and 13 phosphotyrosine proteins were identified. Based on their performed functions, the identified proteins are involved in physiological processes (photosynthesis, growth, energy and amino acid metabolism) closely associated with intensification of plant metabolism. The EBR-induced changes in protein abundance and tyrosine phosphorylation profile may contribute to growth stimulation of wheat seedlings under the action of EBR. The obtained data suggest an important role for EBR in the activation of protein metabolism underlying fundamental physiological processes, including growth promotion.
