Bacterial Community Structure of Typical Lake Sediments in Yinchuan City and Its Response to Heavy Metals

Lake wetlands are extremely important and special ecosystems, which are important for regional water resource storage, environmental protection, and biodiversity maintenance. Sediment bacteria are an important component of lake ecosystems and are a major driver of biogeochemical cycling in lakes. In order to investigate the community structure of bacteria in typical lake sediments in Yinchuan City and their influencing factors, three typical lakes in Yinchuan City (Yuehai Lake, Mingcui Lake, and Xiniu Lake) were selected for the study and surface sediments were collected in January, April, July, and October 2021. The composition of the sediment bacterial community was examined using 16S rDNA high-throughput sequencing technology, and the response relationships between them and heavy metals were explored. The results showed that the ecological hazard coefficient for heavy metals in the sediments of three typical lakes in Yinchuan City was far less than 40, and the ecological hazard index was far less than 150, all of which indicated a minor ecological hazard. There were no significant differences in bacterial community diversity among the three lakes, but there were significant variations in diversity among the lakes in different seasons and significant differences in community composition. The dominant phyla (top three in terms of relative abundance) in Yuehai Lake, Mingcui Lake, and Xiniu Lake were Proteobacteria, Bacteroidetes, and Chloroflexi. The dominant lower orders were Gammaproteobacteria, Alphaproteobacteria, and Deltaproteobacteria. The main divergent species that occurred at the phylum level in typical lakes in Yinchuan were Proteobacteria, Bacteroidetes, Euryarchaeota, Firmicutes, Actinobacteria, and Acidobacteria. The sediment bacterial community structure of Yuehai Lake was significantly correlated with Cu, Fe, Mn, Zn, As, and Pb; the sediment bacterial community structure of Lake Mingcui was significantly correlated with Fe, Pb, and Cr; and the sediment bacterial community structure of Xiniu Lake was not significantly correlated with heavy metals. The types and contents of sediment heavy metals had a significant effect on the bacterial community structure of sediments in Yinchuan Yuehai Lake and Mingcui Lake and were important environmental factors that caused changes in the bacterial community structure of lake sediments.

Cyanobacterial bloom affects structural and functional succession of microbial communities in urban wetland sediments in the Qinghai Plateau

Abstract Algal blooms significantly affect microbial communities in wetland ecosystems. However, little is known about the succession of sediment microbial communities during algal blooms. This study aimed to investigate the temporal patterns of sediment bacterial community structure and function succession during algal blooms (March to May 2022) with high-throughput sequencing technology. To this end, algal blooms were divided into the bloom stage (BS), decomposition stage (DS) and end stage (ES). The results showed that: (i) The algal blooms were dominated by Microcystis species within Cyanobacteria. Both phytoplankton abundance and biomass reached the peak in the BS, with 45.78 × 105 cells/L and 5.97 mg/L, respectively. (ii) The alpha diversity indices of the sediment bacterial community showed a monotonically increasing trend, with the lowest value in the BS. Cyanobacterial blooms reduced the relative abundance of Actinobacteria and Chloroflexi, and raised that of Firmicutes, Proteobacteria and Bacteroidetes. (iii) Cyanobacterial blooms remarkably decreased the complexity of the co-occurrence network of the sediment bacterial community, which resisted extreme environmental conditions in the BS and DS by increasing positive relationships. (iv) Mantel test showed that phytoplankton biomass, T, dissolved oxygen and organic carbon had significant effects on the sediment bacterial community. The results of this study are important for the ecological management of algal blooms through microorganisms.

Seasonality in land-ocean connectivity and local processes control sediment bacterial community structure and function in a High Arctic tidal flat.

Climate change is altering patterns of precipitation, cryosphere thaw, and land-ocean influxes, affecting understudied Arctic estuarine tidal flats. These transitional zones between terrestrial and marine systems are hotspots for biogeochemical cycling, often driven by microbial processes. We investigated surface sediment bacterial community composition and function from May to September along a river-intertidal-subtidal-fjord gradient. We paired metabarcoding of in situ communities with in vitro carbon-source utilization assays. Bacterial communities differed in space and time, alongside varying environmental conditions driven by local seasonal processes and riverine inputs, with salinity emerging as the dominant structuring factor. Terrestrial and riverine taxa were found throughout the system, likely transported with runoff. In vitro assays revealed sediment bacteria utilized a broader range of organic matter substrates when incubated in fresh and brackish water compared to marine water. These results highlight the importance of salinity for ecosystem processes in these dynamic tidal flats, with the highest potential for utilization of terrestrially derived organic matter likely limited to tidal flat areas (and times) where sediments are permeated by freshwater. Our results demonstrate that intertidal flats must be included in future studies on impacts of increased riverine discharge and transport of terrestrial organic matter on coastal carbon cycling in a warming Arctic.

Immobilization of phosphorus in water-sediment system by iron-modified attapulgite, calcite, bentonite and dolomite under feed input condition: Efficiency, mechanism, application mode effect and response of microbial communities and iron mobilization

Four kinds of iron-based materials, i.e., iron-modified attapulgite, calcite, bentonite and dolomite (abbreviated as Fe-ATP, Fe-CA, Fe-BT and Fe-DOL, respectively) were prepared and used to immobilize the phosphorus in the system of overlying water (O-water) and sediment under the feed input condition, and their immobilization efficiencies and mechanisms were investigated. The influence of application mode on the immobilization of phosphorus in the water-sediment system by Fe-ATP, Fe-CA, Fe-BT and Fe-DOL was researched. The effects of Fe-ATP, Fe-CA, Fe-BT and Fe-DOL on the concentration of labile iron in the water-sediment system and the microbial communities in sediment were also studied. The results showed that the Fe-ATP, Fe-CA, Fe-BT and Fe-DOL addition all can effectively immobilize the soluble reactive phosphorus (SRP), dissolved total phosphorus (DTP) and diffusive gradients in thin-films (DGT)-labile phosphorus in O-water under the feed input condition, and also had the ability to inactivate the DGT-labile phosphorus in the top sediment. Although the change in the application mode from the one-time addition to the multiple addition reduced the inactivation efficiencies of SRP and DTP in O-water in the early period of application, it increased the immobilization efficiencies in the later period of application. Although Fe-ATP, Fe-CA, Fe-BT and Fe-DOL had a certain releasing risk of iron into the pore water, they had negligible risk of iron release into O-water. The addition of Fe-ATP, Fe-CA, Fe-BT or Fe-DOL reshaped the sediment bacterial community structure and can affect the microorganism-driven phosphorus cycle in the sediment. Results of this work suggest that Fe-ATP, Fe-CA, Fe-BT and Fe-DOL are promising phosphorus-inactivation materials to immobilize the phosphorus in the water-sediment system under the feed input condition.

Combined impacts of algae-induced variations in water soluble organic matter and heavy metals on bacterial community structure in sediment from Chaohu Lake, a eutrophic shallow lake

Many lakes are suffering from eutrophication and heavy metals-contamination. However, the combined impacts of algae bloom and its induced variations in heavy metals on microbial community in sediment from eutrophic lakes remain unclear. In this study, we performed field experiments to investigate how algae bloom impacted water soluble organic matter (WSOM) and heavy metals in sediment from Chaohu Lake, a eutrophic shallow lake, and probed their combined impacts on sediment bacterial community structure. The results showed that algae bloom increased WSOM quantity, in particular, the soluble microbial by-product-like (SMP) and fulvic acid-like (Fa-L) components markedly enhanced by 203.70 % and 70.17 %, respectively. We also found that algae bloom redistributed the spatial patterns of heavy metals and altered their chemical species in sediment, then promoted contamination degree and potential ecological risk of heavy metals in sediment. Moreover, sediment bacterial community richness and diversity obviously decreased after algae bloom, and the variance partitioning analysis (VPA) results showed that combined impacts of algae-induced changes in WSOM and heavy metals explained 66.56 % of the variations in bacterial community structure. These findings depicted how algae bloom influence sediment WSOM and heavy metals, and revealed the combined impacts of algae-induced variations on microbial community structure in shallow eutrophic lake.

Characterization of the Sediment Bacterial Community Structure and Composition in Najafgarh Lake and Adjoining Dhansa Barrage.

This study was undertaken to assess the changes in the community structure, diversity, and composition of sediment bacteria in a shallow lake, Najafgarh Lake (NL), that receives untreated sewage effluent through drains connected to it. These changes were analyzed by comparing the sediment bacterial community structure of NL to the sediment bacterial community structure of Dhansa Barrage (DB), which receives no such effluents. 16S rRNA amplicon was used for bacterial community analysis. Water and sediment samples were also analyzed and compared revealing high conductivity, ammonia, nitrite content, and low dissolved oxygen in NL. The organic matter content is also higher in the sediments of NL. Proteobacteria, Firmicutes, Bacteroidetes, and Actinobacteria are the predominant phyla in both sites and account for 91% of total bacterial abundance in DB and only 77% in the case of NL. Proteobacteria have the highest relative abundance, accounting for around 42% of the total bacterial population in the case of DB and Firmicutes has the highest relative abundance in Najafgarh at 30%. The diversity analysis found significant differences in the community structure at the two sites. The variation in the bacterial communities in the two wetlands is significantly associated with two water parameters (conductivity and temperature) and two sediment parameters (Sediment Nitrogen and Sediment Organic Matter). Correlation Analysis showed that high ammonia, nitrite, and conductance in NL resulted in bacterial communities shifting towards phyla abundant in degraded ecosystems like Acidobacteria, Choloroflexi, Caldiserica, Aminicenantes, Thaumarchaeota, and Planctomycetes.

Mitigation of Eutrophication in a Shallow Lake: The Influences of Submerged Macrophytes on Phosphorus and Bacterial Community Structure in Sediments

Remediating water eutrophication is critical for maintaining healthy and sustainable development of lakes. The aim of this study was to explore the seasonal variation in phosphorus (P) speciation and bacterial community structure in sediments of Qin Lake (Taizhou, Jiangsu Province, China) associated with the growth of submerged macrophyte Vallisneria natans. The differences in sediment bacterial diversity and community structure between V. natans growing and control areas were analyzed over a period of one year. The results showed that V. natans growth reduced the total P and organic matter contents of the sediments and increased the bioavailable iron (Fe) and Fe-bound P contents. The α-diversity of sediment bacteria was significantly higher in the presence of V. natans than in the controls during the vigorous plant growth stage. In the presence of V. natans, there was a higher relative abundance of Proteobacteria and lower relative abundances of Chloroflexi and Acidobacteria. The Fe(II) content in the sediment had a larger influence on the spatial distribution of bacterial communities than sediment Fe-bound P, organic matter, and Fe(II) contents. V. natans growth could reshape sediment bacterial community structure in the shallow lake, which, in turn, enhanced P immobilization in the sediments and thereby improved the water quality.

Addition of organic acids to acid mine drainage polluted wetland sediment leads to microbial community structure and functional changes and improved water quality

Acid mine drainage (AMD) is a serious environmental problem worldwide that requires efficient and sustainable remediation technologies including the use of biological mechanisms. A key challenge for AMD bioremediation is to provide optimal conditions for microbial-mediated immobilisation of trace metals. Although organic carbon and oxygen can enhance treatment efficiency, the effect on microbial communities is unclear. In this study, surface sediments from a natural wetland with proven efficiency for AMD bioremediation were artificially exposed to oxygen (by aeration) and/or organic carbon (in the form of mixed organic acids) and incubated under laboratory conditions. In addition to measuring changes in water chemistry, a metagenomics approach was used to determine changes in sediment bacterial, archaeal and fungal community structure, and functional gene abundance. The addition of organic carbon produced major changes in the abundance of microorganisms related to iron and sulfur metabolism (including Geobacter and Pelobacter) and increased levels of particulate metals via sulfate reduction. Aeration resulted in an increase in Sideroxydans abundance but no significant changes in metal chemistry were observed. The study concludes that the utilisation of organic carbon by microorganisms is more important for achieving efficient AMD treatment than the availability of oxygen, yet the combination of oxygen with organic carbon addition did not inhibit the improvements to water quality.

16S and 18S rRNA Gene Metabarcoding Provide Congruent Information on the Responses of Sediment Communities to Eutrophication

Metabarcoding analyses of bacterial and eukaryotic communities have been proposed as efficient tools for environmental impact assessment. It has been unclear, however, to which extent these analyses can provide similar or differing information on the ecological status of the environment. Here, we used 16S and 18S rRNA gene metabarcoding to compare eutrophication-induced shifts in sediment bacterial and eukaryotic community structure in relation to a range of porewater, sediment and bottom-water geochemical variables, using data obtained from six stations near a former rainbow trout farm in the Archipelago Sea (Baltic Sea). Shifts in the structure of both community types were correlated with a shared set of variables, including porewater ammonium concentrations and the sediment depth-integrated oxygen consumption rate. Distance-based redundancy analyses showed that variables typically employed in impact assessments, such as bottom water nutrient concentrations, explained less of the variance in community structure than alternative variables (e.g., porewater NH4+ inventories and sediment depth-integrated O2 consumption rates) selected due to their low collinearity (up to 40 vs. 58% of the variance explained, respectively). In monitoring surveys where analyses of both bacterial and eukaryotic communities may be impossible, either 16S or 18S rRNA gene metabarcoding can serve as reliable indicators of wider ecological impacts of eutrophication.

Attributes of Drying Define the Structure and Functioning of Microbial Communities in Temperate Riverbed Sediment.

Combined effects of climate change and increasing anthropogenic water demand have increased and extended dry period occurrences in rivers worldwide. Riverbed drying can significantly affect sediment microorganisms, crucial drivers of biogeochemical processes in lotic systems. In this study, we evaluated how sediment bacterial and fungal community structure and composition (based on 16S rRNA gene and ITS metabarcoding) and microbial functions (community respiration and extracellular enzymatic activities) respond to different riverbed drying intensities over 90 days. Riverbed sediment collected in a flowing reach of the Spree river in northeastern Germany was dried under different rates in outdoor mesocosms during the summer months of 2018. Our results demonstrate that drying attributes (duration and intensity) and sediment organic matter (OM) content play a crucial role in sediment microbial community assembly and functioning throughout drying. Milder drying surprisingly triggered a more rapid and drastic change in the microbial community composition and diversity. After 90 days of drying, Bacilli (Firmicutes) became the dominant bacterial class in most treatments, except in sediments with low OM content under the most severe drying treatment. Fungal amplicon sequence variants (ASVs) from Dothideomycetes (Ascomycota) had by far the highest relative abundance in all our treatments at the end of the drying experiment, making up 65.1% to 94.0% of the fungal reads. CO2 fluxes, a proxy for sediment community respiration, were rapidly and strongly affected by drying in all treatments. Our results imply that even short riverbed drying periods are likely to have significant consequences for the biogeochemical dynamics in recently formed non-perennial temperate rivers.

Spatial and seasonal variations of sediment bacterial communities in a river-bay system in South China.

River-bay systems are transitional areas that hold important roles in biogeochemical processes between continents and oceans. However, composition and structure of microbial communities shaped by such environments have not been clear yet. In this study, we used high-throughput sequencing of 16S rRNA genes to analyze the diversity and composition of sediment bacterial communities from the Shenzhen river-bay system during dry and wet seasons. The results showed that sediment bacterial community structure was varied according to habitats (river vs. estuary) and seasons (wet season vs. dry season). The alpha diversity of sediment bacterial community was significantly higher in the dry season than in the wet season, while no significant difference of alpha diversity was found between river and estuary. Neutral community model revealed a significant influence of stochastic processes on sediment bacterial community assembly, especially in the wet season. However, the beta nearest-taxon index indicated that deterministic processes were more responsible for the assembly of sediment bacterial community. Additionally, redundancy analysis suggested strong links between sediment bacterial communities and environmental factors in Shenzhen river-bay system, with the environmental factors explaining 63.5% of the bacterial community variation. Specifically, NH4+, pH, and salinity were the three most important contributing factors that shaped the sediment bacterial communities. Overall, this study provides a valuable reference to get insights into the spatiotemporal pattern of sediment bacterial communities in a typical river-bay system. Characteristics of bacterial community structure and function associated with nutrients and heavy metals in coastal aquaculture area.

Coastal aquaculture area has become one of the critical zones that are more susceptible to the influence of human activity. Many aquaculture operations invariably result in the accumulation of nutrients and heavy metals in the coastal ecosystem. Our study investigated sediment bacterial community structure and function across 23 sites under the influence of nutrients and heavy metals in the coastal aquaculture area. The habitat environment of the sediment was described by analyzing physicochemical characteristics. Sediment bacterial community structure and diversity were investigated by 16S rRNA sequencing. The sequencing data presented that Proteobacteria, Bacteroidetes, Planctomycetes, Acidobacteria and Chloroflexi were predominant at phylum level. Variations in the bacterial community composition and diversity were significant (P<0.01) among different groups (according to the distance from the bank side) which indicated that specific environmental conditions had shaped distinct bacterial community. Specifically, bacterial diversity and composition were significantly influenced by the temperature, salinity, pH, dissolved oxygen (DO), TOC, TON, nitrite, nitrate and heavy metals (P<0.05). Results related to functional prediction demonstrated that carbon, nitrogen and sulfur metabolism were the dominant processes in the coastal aquaculture area. In the meantime, the potential pathogens such as Arcobacter was found in site S3, which indicated the possible threat to the cultured species in this area. Overall, variations in bacterial communities caused by nutrients and heavy metals can affect biogeochemical cycles, which may provide an indication for the protection of coastal aquaculture environments.

Effect of hydrological regime on the sediment bacterial diversity and community structure in Sultanpur Lake, Gurugram, Haryana

The present study focuses on the changes in sediment bacterial community structure and diversity of Sultanpur Lake due to changes in its hydrological regime. The assessment of the bacterial community structure was done by using the Next Generation Sequencing of 16S rRNA amplicon, a bacterial phylogenetic marker. Sultanpur Lake is located in Sultanpur National Park Gurugram, in the state of Haryana, India. This place harbors great plant and animal biodiversity and serves as an important feeding, roosting and breeding ground for both resident and migratory avian fauna. Increasing pollution level and scarcity of water during summer months impose a great threat to the lake. Sediment bacteria are considered as reliable recorders of nutrient cycling and active mediums of biogeochemical processes and thus were assessed in this study. Wet and dry sediment samples were taken and analyzed for their bacterial community structure and diversity. The amplicon sequencing generated 881,118 reads which corresponded to 93 known species in case of dry sediments and 8,71,102 reads corresponding to 44 species in case of wet sediments. In dry sediments the top ten phyla were Firmicutes (26%), Bacteroidetes (14%), Proteobacteria (15%), Cyanobacteria (12%), Parcubacteria (7%), Euryarcheota (6%), Actinobacteria (5%), Armatimonadetes (5%), Chloroflexi (2%) and Planctomycetes (2%). In case of wet sediments, the top ten phyla were Firmicutes (35%), Bacteroidetes (22%), Proteobacteria (22%), Actinobacteria (7%), Chloroflexi (5%) Parcubacteria (3%), Verrucomimicrobia (3%) and, Deinococcus-Thermus (3%). There was change in the community structure at species level too with Clostridium species as the most dominant species in case of wet sediments and Lactobacillus species as the predominant one in dry sediments. This study provides an insight into the changes in the bacterial communities due to changes in hydrology and how this change will affect the health of this wetland as irregular water supply is a major stressor of this wetland.

Bacterial community response to different nitrogen gradients of swine wastewater in surface flow constructed wetlands

How sediment bacterial community structure and diversity responds to different gradients of nitrogen (N) in swine wastewater is poorly understood. Here, the effects of different total nitrogen (TN) concentrations in swine wastewater on the microbial diversity and community composition in surface flow constructed wetlands (SFCWs) were investigated. The five concentration gradients included 2, 250, 300, 350, and 400 mg L−1. Under high N concentrations (>300 mg L−1), the Ace and Chao1 indexes increased, however, the Shannon index declined with increasing N concentration. The relative abundance of Chloroflexi, Acidobacteria and Actinobacteria showed an increasing trend. In contrast, under relatively low N concentrations (≤300 mg L−1), Shannon index increased with increasing N concentration. The relative abundance of Bacteroidetes and Verrucomicrobia exhibited an increasing trend with increasing N concentration. TN, NH4+ and NO3− significantly influenced on the microbial community distribution and composition (P < 0.05). These findings provide evidence that N concentration of swine wastewater is powerful predictor of bacterial diversity and community composition in SFCWs.

The key environmental influencing factors for the change of sediment bacterial community and antibiotics resistance genes in a long-term polluted lake, China.

In order to investigate the key environmental influencing factors for the change of sediment bacterial community structure (BCS) and antibiotics resistance genes (ARGs) in a long-term Quinolone antibiotics (QNs) and heavy metals (HMs) polluted lake, 16S rRNA MiSeq High-throughput Sequencing and Phylogenetic Investigation of Communities by Reconstruction of Unobserved States (PICRUSt) analysis methods were applied. Baiyangdian lake was subdivided into three habitats: (1) Habitat 1: greatly influenced by municipal wastewater; (2) Habitat 2: mainly impacted by aquaculture sewage and domestic wastewater; and (3) Habitat 3: with the least human activities. Through One-way ANOVA analysis, the results showed that most of QNs and HMs showed significant difference among three habitats. Both the highest richness and diversity indices of bacterial community appeared in Habitat 3. The abundance of Multidrug, Phenicol, Aminoglycoside, Teracycline, and Quinolone ARGs exhibited the highest values in Habitat 1, while the abundance of Macrolide-Lincosamide-Streptogramin (MLS), Rifamycinm, and Sulfonamide ARGs appeared the highest values in Habitat 2. The result of redundancy analysis exhibited that 68.8% and 93.8% of the change in BCS and ARGs can be explained by environmental factors, respectively. Pb (explained 37.5% (p = 0.002)) and Fleroxacin (FLE) (explained 51.3% (p = 0.026)) were the most important factors for the variation of BCS and ARGs, respectively. Therefore, the effects of antibiotics and HMs on BCS and ARGs should be simultaneously paid more attention. Furthermore, the ARGs results by PICRUSt were similar to the results by metagenomic shotgun sequencing analysis, thus the PICRUSt analysis method can be used in the future research.

Sediment Bacterial Community Structure Under the Influence of Different Domestic Sewage Types.

Sediment bacterial communities are critical to the biogeochemical cycle in river ecosystems, but our understanding of the relationship between sediment bacterial communities and their specific input streams in rivers remains insufficient. In this study, we analyzed the sediment bacterial community structure in a local river receiving discharge of urban domestic sewage by applying Illumina MiSeq high-throughput sequencing. The results showed that the bacterial communities of sediments samples of different pollution types had similar dominant phyla, mainly Proteobacteria, Actinobacteria, Chloroflexi and Firmicutes, but their relative abundances were different. Moreover, there were great differences at the genus level. For example, the genus Bacillus showed statistically significant differences in the hotel site. The clustering of bacterial communities at various sites and the dominant families (i.e., Nocardioidaceae, and Sphingomonadaceae) observed in the residential quarter differed from other sites. This result suggested that environmentally induced species sorting greatly influenced the sediment bacterial community composition. The bacterial cooccurrence patterns showed that the river bacteria had a nonrandom modular structure. Microbial taxonomy from the same module had strong ecological links (such as the nitrogenium cycle and degradation of organic pollutants). Additionally, PICRUSt metabolic inference analysis showed the most important function of river bacterial communities under the influence of different types of domestic sewage was metabolism (e.g., genes related to xenobiotic degradation predominated in residential quarter samples). In general, our results emphasize that the adaptive changes and interactions in the bacterial community structure of river sediment represent responses to different exogenous pollution sources.

Fate and effects of sediment-associated polycyclic musk HHCB in subtropical freshwater microcosms

Galaxolide (HHCB) is used as a fragrance ingredient in household and personal care products, and has been ubiquitously detected in the environment. Here we investigated the fate of HHCB in subtropical freshwater microcosms, and evaluated effects of sediment-associated HHCB on a biological community consisting of algae, Daphnia, benthic macroinvertebrates and bacteria. The concentrations of sediment-associated HHCB did not change significantly during a 28 days exposure period, but HHCB accumulated in worms with biota-sediment accumulation-factor (BSAF) values in the range of 0.29–0.66 for Branchiura sowerbyi and 0.94–2.11 for Limnodrilus hoffmeisteri. There was no significant effects of HHCB (30 μg/g dry weight (dw) sediment) on chlorophyll-a content, sediment bacterial community composition, and survival and growth of benthic macroinvertebrates. However, the presence of benthic macroinvertebrates altered the sediment bacterial community structure relative to microcosms without introduced organisms. The findings of this study suggest that a single high-dose of HHCB, over 28 days, at environmentally relevant concentrations would not impose direct toxicological risks to aquatic organisms such as benthic macroinvertebrates.

Pressure and temperature effects on deep-sea hydrocarbon-degrading microbial communities in subarctic sediments.

The Hatton–Rockall Basin (North‐East Atlantic) is an area with potential for deep‐sea (2,900 m) hydrocarbon exploration. Following the Deepwater Horizon oil spill, many investigations into the responses of sediment microbial communities to oil pollution have been undertaken. However, hydrostatic pressure is a parameter that is often omitted due to the technical difficulties associated with conducting experiments at high pressure (>10 MPa). In this study, sediments from 2,900 m in the Hatton–Rockall Basin, following a one‐week decompression period in a temperature‐controlled room at 5°C, were incubated in factorial combinations of 0.1 and 30 MPa, 5 and 20°C, and contamination with a hydrocarbon mixture or uncontaminated controls to evaluate the effect of these environmental variables on the bacterial community composition. Our results revealed varying effects of pressure, temperature, and oil contamination on the composition of the bacterial community within the sediment. Temperature was the strongest determinant of differences in the bacterial community structure between samples followed by pressure. Oil contamination did not exert a strong change in the sediment bacterial community structure when pressure and temperature conditions were held at in situ levels (30 MPa and 5°C). The γ‐proteobacteria Pseudomonas and Colwellia, and several Bacteroidetes dominated communities at 30 MPa. In contrast, hydrocarbon degraders such as Halomonas, Alcanivorax, and Marinobacter decreased in relative abundance at the same pressure. This study highlights the importance of considering hydrostatic pressure in ex situ investigations into hydrocarbon‐degrading deepwater microbial communities.

Acid mine drainage affects the diversity and metal resistance gene profile of sediment bacterial community along a river

Acid mine drainage (AMD) is one of the most hazardous byproducts of some types of mining. However, research on how AMD affects the bacterial community structure of downstream riverine ecosystems and the distribution of metal resistance genes (MRGs) along pollution gradient is limited. Comprehensive geochemical and high-throughput next-generation sequencing analyses can be integrated to characterize spatial distributions and MRG profiles of sediment bacteria communities along the AMD-contaminated Hengshi River. We found that (1) diversities of bacterial communities significantly and gradually increased along the river with decreasing contamination, suggesting community composition reflected changes in geochemical conditions; (2) relative abundances of phyla Proteobacteria and genus Halomonas and Planococcaceae that function in metal reduction decreased along the AMD gradient; (3) low levels of sediment salinity, sulfate, aquatic lead (Pb), and cadmium (Cd) were negatively correlated with bacterial diversity despite pH was in a positive manner with diversity; and (4) arsenic (As) and copper (Cu) resistance genes corresponded to sediment concentrations of As and Cu, respectively. Altogether, our findings offer initial insight into the distribution patterns of sediment bacterial community structure, diversity and MRGs along a lotic ecosystem contaminated by AMD, and the factors that affect them.
