Integrated Computational and Experimental Framework for Inverse Screening of Candidate Antibacterial Nanomedicine.

Nanomedicine is promising for disease prevention and treatment, but there are still many challenges that hinder its rapid development. A major challenge is to efficiently seek candidates with the desired therapeutic functions from tremendously available materials. Here, we report an integrated computational and experimental framework to seek alloy nanoparticles from the Materials Project library for antibacterial applications, aiming to learn the inverse screening concept from traditional medicine for nanomedicine. Because strong peroxidase-like catalytic activity and weak toxicity to normal cells are the desired material properties for antibacterial usage, computational screening implementing theoretical prediction models of catalytic activity and cytotoxicity is first conducted to select the candidates. Then, experimental screening based on scanning probe block copolymer lithography is used to verify and refine the computational screening results. Finally, the best candidate AuCu3 is synthesized in solution and its antibacterial performance over other nanoparticles against S. aureus and E. coli. is experimentally confirmed. The results show the power of inverse screening in accelerating the research and development of antibacterial nanomedicine, which may inspire similar strategies for other nanomedicines in the future.

Morphology Engineering in Multicomponent Hollow Metal Chalcogenide Nanoparticles.

Hollow metal chalcogenide nanoparticles are widely applicable in environmental and energy-related processes. Herein, we synthesized such particles with large compositional and morphological diversity by combining scanning probe block copolymer lithography with a Kirkendall effect-based sulfidation process. We explored the influence of temperature-dependent diffusion kinetics, elemental composition and miscibility, and phase boundaries on the resulting particle morphologies. Specifically, CoNi alloys form single-shell sulfides for the synthetic conditions explored because Co and Ni exhibit similar diffusion rates, while CuNi alloys form sulfides with various types of morphologies (yolk-shell, double-shell, and single-shell) because Cu and Ni have different diffusion rates. In contrast, Co-Cu heterodimers form hollow heterostructured sulfides with varying void numbers and locations depending on synthesis temperature and phase boundary. At higher temperatures, the increased miscibility of CoS2 and CuS makes it energetically favorable for the heterostructure to adopt a single alloy shell morphology, which is rationalized using density functional theory-based calculations.

Intermetallic Nanocrystal Discovery through Modulation of Atom Stacking Hierarchy.

A library of compositionally and structurally well-defined Au-Cu alloy nanocrystals has been prepared via scanning probe block copolymer lithography. These libraries not only allow one to map compositional and structure space but also the conditions (e.g., cooling rate) required to access specific structures. This approach enabled the realization of a previously unobserved architecture, an intermetallic nanoprism, that is a consequence of hierarchical atom stacking. These structures exhibit distinctive diffraction patterns characterized by non-integer-index, forbidden spots, which serve as a diagnostic indicator of such structures. Inspection of the library's pseudospherical particles reveals a high-strain cubic-tetragonal interfacial configuration in the outer regions of the intermetallic nanocrystals. Since it is costly and time-consuming to explore the nanomaterials phase space via conventional wet-chemistry, this parallel kinetic-control approach, which relies on substrate- and positionally isolated particles, may lead to the rapid discovery of complex nanocrystals that may prove useful in applications spanning catalysis and plasmonic sensing.

Regioselective Deposition of Metals on Seeds within a Polymer Matrix

We use scanning probe block copolymer lithography in a two-step sequential manner to explore the deposition of secondary metals on nanoparticle seeds. When single element nanoparticles (Au, Ag, Cu, Co, or Ni) were used as seeds, both heterogeneous and homogeneous growth occurred, as rationalized using the thermodynamic concepts of bond strength and lattice mismatch. Specifically, heterogeneous growth occurs when the heterobond strength between the seed and growth atoms is stronger than the homobond strength between the growth atoms. Moreover, the resulting nanoparticle structure depends on the degree of lattice mismatch between the seed and growth metals. Specifically, a large lattice mismatch (e.g., 13.82% for Au and Ni) typically resulted in heterodimers, whereas a small lattice mismatch (e.g., 0.19% for Au and Ag) resulted in core-shell structures. Interestingly, when heterodimer nanoparticles were used as seeds, the secondary metals deposited asymmetrically on one side of the seed. By programming the deposition conditions of Ag and Cu on AuNi heterodimer seeds, two distinct nanostructures were synthesized with (1) Ag and Cu on the Au domain and (2) Ag on the Au domain and Cu on the Ni domain, illustrating how this technique can be used to predictively synthesize structurally complex, multimetallic nanostructures.

Custom-made holey graphene via scanning probe block co-polymer lithography.

Oxidative chemical etching of metal nanoparticles (NPs) to produce holey graphene (hG) suffers from the presence of aggregated NPs on the graphene surface triggering heterogeneous etching rates and thereby producing irregular sized holes. To encounter such a challenge, we investigated the use of scanning probe block co-polymer lithography (SPBCL) to fabricate precisely positioned silver nanoparticles (AgNPs) on graphene surfaces with exquisite control over the NP size to prevent their aggregation and consequently produce uniformly distributed holes after oxidative chemical etching. SPBCL experiments were carried out via printing an ink suspension consisting of poly(ethylene oxide-b-2-vinylpyridine) and silver nitrate on a graphene surface in a selected pattern under controlled environmental and instrumental parameters followed by thermal annealing in a gaseous environment to fabricate AgNPs. Scanning electron microscopy revealed the uniform size distribution of AgNPs on the graphene surface with minimal to no aggregation. Four main sizes of AgNPs were obtained (37 ± 3, 45 ± 3, 54 ± 2, and 64 ± 3 nm) via controlling the printing force, z-piezo extension, and dwell time. Energy dispersive X-ray spectroscopy analysis validated the existence of elemental Ag on the graphene surface. Subsequent chemical etching of AgNPs using nitric acid (HNO3) with the aid of sonication and mechanical agitation produced holes of uniform size distribution generating hG. The obtained ID/IG ratios ≤ 0.96 measured by Raman spectroscopy were lower than those commonly reported for GO (ID/IG > 1), indicating the removal of more defective C atoms during the etching process to produce hG while preserving the remaining C atoms in ordered or crystalline structures. Indeed, SPBCL could be utilized to fabricate uniformly distributed AgNPs of controlled sizes on graphene surfaces to ultimately produce hG of uniform hole size distribution.

Synthesis of Metal-Capped Semiconductor Nanowires from Heterodimer Nanoparticle Catalysts.

Semiconductor nanowires (NWs) capped with metal nanoparticles (NPs) show multifunctional and synergistic properties, which are important for applications in the fields of catalysis, photonics, and electronics. Conventional colloidal syntheses of this class of hybrid structures require complex sequential seeded growth, where each section requires its own set of growth conditions, and methods for preparing such wires are not universal. Here, we report a new and general method for synthesizing metal-semiconductor nanohybrids based on particle catalysts, prepared by scanning probe block copolymer lithography, and chemical vapor deposition. In this process, metallic heterodimer NPs were used as catalysts for NW growth to form semiconductor NWs capped with metallic particles (Au, Ag, Co, Ni). Interestingly, the growth processes for NWs on NPs are regioselective and controlled by the chemical composition of the metallic heterodimer used. Using a systematic experimental approach, paired with density functional theory calculations, we were able to postulate three different growth modes, one without precedent.

Evolution of Dip-Pen Nanolithography (DPN): From Molecular Patterning to Materials Discovery.

Dip-pen nanolithography (DPN) is a nanofabrication technique that can be used to directly write molecular patterns on substrates with high resolution and registration. Over the past two decades, DPN has evolved in its ability to transport molecular and material "inks" (e.g., alkanethiols, biological molecules like DNA, viruses, and proteins, polymers, and nanoparticles) to many surfaces in a high-throughput fashion, enabling the synthesis and study of complex chemical and biological structures. In addition, DPN has laid the foundation for a series of related scanning probe methodologies, for example, polymer pen lithography (PPL), scanning probe block copolymer lithography (SPBCL), and beam-pen lithography (BPL), which do not rely on cantilever tips. Structures prepared with these methodologies have been used to understand the consequences of miniaturization and open a door to new capabilities in catalysis, optics, biomedicine, and chemical synthesis, where, in sum, a process originally intended to compete with tools used by the semiconductor industry for rapid prototyping has transcended that application to advanced materials discovery. This review outlines the major DPN advances, the subsequent methods based on the technique, and the opportunities for future fundamental and technological exploration. Most importantly, it commemorates the 20th anniversary of the discovery of DPN.

Multimetallic High-Index Faceted Heterostructured Nanoparticles.

Multimetallic heterostructured nanoparticles with high-index facets potentially represent an important class of highly efficient catalysts. However, due to their complexity, they are often difficult to synthesize. Herein, a library of heterostructured, multimetallic (Pt, Pd, Rh, and Au) tetrahexahedral nanoparticles was synthesized through alloying/dealloying with Bi in a tube furnace at 900-1000 °C. Electron microscopy and selected area diffraction measurements show that the domains of the heterostructured nanoparticles are epitaxially aligned. Although nanoparticles formed from Au alone exhibit low-index facets, Pt and Au form PtAu heterostructured nanoparticles with high-index facets, including domains that are primarily made of Au. Furthermore, the alloying/dealloying of Bi occurs at different rates and under different conditions within the heterostructured nanoparticles. This influences the types of architectures observed en route to the final high-index state, a phenomenon clearly observable in the case of PdRhAu nanoparticles. Finally, scanning probe block copolymer lithography was used in combination with this synthetic strategy to control nanoparticle composition in the context of PtAu nanoparticles (1:4 to 4:1 ratio range) and size (15 to 45 nm range).

Windowless Observation of Evaporation-Induced Coarsening of Au-Pt Nanoparticles in Polymer Nanoreactors.

The interactions between nanoparticles and solvents play a critical role in the formation of complex, metastable nanostructures. However, direct observation of such interactions with high spatial and temporal resolution is challenging with conventional liquid-cell transmission electron microscopy (TEM) experiments. Here, a windowless system consisting of polymer nanoreactors deposited via scanning probe block copolymer lithography (SPBCL) on an amorphous carbon film is used to investigate the coarsening of ultrafine (1-3 nm) Au-Pt bimetallic nanoparticles as a function of solvent evaporation. In such reactors, homogeneous Au-Pt nanoparticles are synthesized from metal-ion precursors in situ under electron irradiation. The nonuniform evaporation of the thin polymer film not only concentrates the nanoparticles but also accelerates the coalescence kinetics at the receding polymer edges. Qualitative analysis of the particle forces influencing coalescence suggests that capillary dragging by the polymer edges plays a significant role in accelerating this process. Taken together, this work (1) provides fundamental insight into the role of solvents in the chemistry and coarsening behavior of nanoparticles during the synthesis of polyelemental nanostructures, (2) provides insight into how particles form via the SPBCL process, and (3) shows how SPBCL-generated domes, instead of liquid cells, can be used to study nanoparticle formation. More generally, it shows why conventional models of particle coarsening, which do not take into account solvent evaporation, cannot be used to describe what is occurring in thin film, liquid-based syntheses of nanostructures.

Catalyst design by scanning probe block copolymer lithography

Scanning probe block copolymer lithography (SPBCL), in combination with density-functional theory (DFT), has been used to design and synthesize hydrogen evolution catalysts. DFT was used to calculate the hydrogen adsorption energy on a series of single-element, bimetallic, and trimetallic (Au, Pt, Ni, and Cu) substrates to provide leads that could be synthesized in the form of alloy or phase-separated particles via SPBCL. PtAuCu (18 nm, ∼1:1:1 stoichiometry) has been identified as a homogeneous alloy phase that behaves as an effective hydrogen evolution catalyst in acidic aqueous media, even when it is made in bulk form via solution phase methods. Significantly, the bulk-prepared PtAuCu/C nanocatalyst discovered via this process exhibits an activity seven times higher than that of the state-of-the-art commercial Pt/C catalyst (based upon Pt content). The advantage of using SPBCL in the discovery process is that one can uniformly make particles, each consisting of a uniform phase combination (e.g., all alloy or all phase-segregated species) at a fixed elemental ratio, an important consideration when working with polyelemental species where multiple phases may exist.

Structural Evolution of Three-Component Nanoparticles in Polymer Nanoreactors.

Recent developments in scanning probe block copolymer lithography (SPBCL) enable the confinement of multiple metal precursors in a polymer nanoreactor and their subsequent transformation into a single multimetallic heterostructured nanoparticle through thermal annealing. However, the process by which multimetallic nanoparticles form in SPBCL-patterned nanoreactors remains unclear. Here, we utilize the combination of PEO-b-P2VP and Au, Ag, and Cu salts as a model three-component system to investigate this process. The data suggest that the formation of single-component Au, Ag, or Cu nanoparticles within polymer nanoreactors consists of two stages: (I) nucleation, growth, and coarsening of the particles to yield a single particle in each reactor; (II) continued particle growth by depletion of the remaining precursor in the reactor until the particle reaches a stable size. Also, different aggregation rates are observed for single-component particle formation (Au > Ag > Cu). This behavior is also observed for two-component systems, where nucleation sites have greater Au content than the other metals. This information can be used to trap nanoparticles with kinetic structures. High-temperature treatment ultimately facilitates the structural evolution of the kinetic particle into a particle with a fixed structure. Therefore, with multicomponent systems, a third stage that involves elemental redistribution within the particle must be part of the description of the synthetic process. This work not only provides a glimpse at the mechanism underlying multicomponent nanoparticle formation in SPBCL-generated nanoreactors but also illustrates, for the first time, the utility of SPBCL as a platform for controlling the architectural evolution of multimetallic nanoparticles in general.

Using Scanning-Probe Block Copolymer Lithography and Electron Microscopy To Track Shape Evolution in Multimetallic Nanoclusters.

Here we describe a general method for synthesizing multimetallic core-shell nanoclusters on surfaces. By patterning seeds at predesignated locations using scanning-probe block copolymer lithography, we can track shape evolution in nanoclusters and elucidate their growth pathways using electron microscopy. The growth of core-shell nanostructures on surface-bound seeds is a highly anisotropic process and often results in multimetallic anisotropic nanostructures. The shell grows at specific edge and corner sites of the patterned seeds and propagates predominately from the top hemisphere of the seeds.

Tip-Directed Synthesis of Multimetallic Nanoparticles.

Alloy nanoparticles are important in many fields, including catalysis, plasmonics, and electronics, due to the chemical and physical properties that arise from the interactions between their components. Typically, alloy nanoparticles are made by solution-based synthesis; however, scanning-probe-based methods offer the ability to make and position such structures on surfaces with nanometer-scale resolution. In particular, scanning probe block copolymer lithography (SPBCL), which combines elements of block copolymer lithography with scanning probe techniques, allows one to synthesize nanoparticles with control over particle diameter in the 2-50 nm range. Thus far, single-element structures have been studied in detail, but, in principle, one could make a wide variety of multicomponent systems by controlling the composition of the polymer ink, polymer feature size, and metal precursor concentrations. Indeed, it is possible to use this approach to synthesize alloy nanoparticles comprised of combinations of Au, Ag, Pd, Ni, Co, and Pt. Here, such structures have been made with diameters deliberately tailored in the 10-20 nm range and characterized by STEM and EDS for structural and elemental composition. The catalytic activity of one class of AuPd alloy nanoparticles made via this method was evaluated with respect to the reduction of 4-nitrophenol with NaBH4. In addition to being the first catalytic studies of particles made by SPBCL, these proof-of-concept experiments demonstrate the potential for SPBCL as a new method for studying the fundamental science and potential applications of alloy nanoparticles in areas such as heterogeneous catalysis.

Delineating the pathways for the site-directed synthesis of individual nanoparticles on surfaces

Although nanoparticles with exquisite properties have been synthesized for a variety of applications, their incorporation into functional devices is challenging owing to the difficulty in positioning them at specified sites on surfaces. In contrast with the conventional synthesis-then-assembly paradigm, scanning probe block copolymer lithography can pattern precursor materials embedded in a polymer matrix and synthesize desired nanoparticles on site, offering great promise for incorporating nanoparticles into devices. This technique, however, is extremely limited from a materials standpoint. To develop a materials-general method for synthesizing nanoparticles on surfaces for broader applications, a mechanistic understanding of polymer-mediated nanoparticle formation is crucial. Here, we design a four-step synthetic process that enables independent study of the two most critical steps for synthesizing single nanoparticles on surfaces: phase separation of precursors and particle formation. Using this process, we elucidate the importance of the polymer matrix in the diffusion of metal precursors to form a single nanoparticle and the three pathways that the precursors undergo to form nanoparticles. Based on this mechanistic understanding, the synthetic process is generalized to create metal (Au, Ag, Pt, and Pd), metal oxide (Fe(2)O(3), Co(2)O(3), NiO, and CuO), and alloy (AuAg) nanoparticles. This mechanistic understanding and resulting process represent a major advance in scanning probe lithography as a tool to generate patterns of tailored nanoparticles for integration with solid-state devices.

Positionally Defined, Binary Semiconductor Nanoparticles Synthesized by Scanning Probe Block Copolymer Lithography

We report the first method for synthesizing binary semiconductor materials by scanning probe block copolymer lithography (SPBCL) in desired locations on a surface. In this work, we utilize SPBCL to create polymer features containing a desired amount of Cd(2+), which is defined by the feature volume. When they are subsequently reacted in H(2)S in the vapor phase, a single CdS nanoparticle is formed in each block copolymer (BCP) feature. The CdS nanoparticles were shown to be both crystalline and luminescent. Importantly, the CdS nanoparticle sizes can be tuned since their diameters depend on the volume of the originally deposited BCP feature.

Nanoreactors for Studying Single Nanoparticle Coarsening

The ability to observe intermediate structures as part of coarsening processes that lead to the formation of single nanoparticles (NPs) is important in gaining fundamental insight pertaining to nanostructure growth. Here, we use scanning probe block copolymer lithography (SPBCL) to create "nanoreactors" having attoliter volumes, which confine Au NP nucleation and growth to features having diameters <150 nm on a substrate. With this technique, one can use in situ TEM to directly observe and study NP coarsening and differentiate Ostwald ripening from coalescence processes. Importantly, the number of metal atoms that can engage in coarsening can be controlled with this technique, and TEM "snapshots" of particle growth can be taken. The size of the resulting nanostructures can be controlled in the 2-10 nm regime.

Single-molecule protein arrays enabled by scanning probe block copolymer lithography

The ability to control the placement of individual protein molecules on surfaces could enable advances in a wide range of areas, from the development of nanoscale biomolecular devices to fundamental studies in cell biology. Such control, however, remains a challenge in nanobiotechnology due to the limitations of current lithographic techniques. Herein we report an approach that combines scanning probe block copolymer lithography with site-selective immobilization strategies to create arrays of proteins down to the single-molecule level with arbitrary pattern control. Scanning probe block copolymer lithography was used to synthesize individual sub-10-nm single crystal gold nanoparticles that can act as scaffolds for the adsorption of functionalized alkylthiol monolayers, which facilitate the immobilization of specific proteins. The number of protein molecules that adsorb onto the nanoparticles is dependent upon particle size; when the particle size approaches the dimensions of a protein molecule, each particle can support a single protein. This was demonstrated with both gold nanoparticle and quantum dot labeling coupled with transmission electron microscopy imaging experiments. The immobilized proteins remain bioactive, as evidenced by enzymatic assays and antigen-antibody binding experiments. Importantly, this approach to generate single-biomolecule arrays is, in principle, applicable to many parallelized cantilever and cantilever-free scanning probe molecular printing methods.

Scanning probe block copolymer lithography

Integration of individual nanoparticles into desired spatial arrangements over large areas is a prerequisite for exploiting their unique electrical, optical, and chemical properties. However, positioning single sub-10-nm nanoparticles in a specific location individually on a substrate remains challenging. Herein we have developed a unique approach, termed scanning probe block copolymer lithography, which enables one to control the growth and position of individual nanoparticles in situ. This technique relies on either dip-pen nanolithography (DPN) or polymer pen lithography (PPL) to transfer phase-separating block copolymer inks in the form of 100 or more nanometer features on an underlying substrate. Reduction of the metal ions via plasma results in the high-yield formation of single crystal nanoparticles per block copolymer feature. Because the size of each feature controls the number of metal atoms within it, the DPN or PPL step can be used to control precisely the size of each nanocrystal down to 4.8 ± 0.2 nm.
